Abstract

BackgroundThe incidence of type 2 diabetes mellitus (T2DM) is increasing worldwide. Many studies carried out in several populations exhibited an association of adiponectin receptor 1 gene (AdipoR1) polymorphisms with type 2 diabetes mellitus. MethodsA case-control study was conducted to verify the association of AdipoR1 polymorphisms (rs1342387 G>A and rs12733285C>T) with T2DM in Iraqi population. The study included 400 T2DM patients as well as 400 healthy control individuals. DNA was extracted from whole blood and genotyped for the analyzed genes by PCR-RFLP. The odds ratio for the risk of developing T2DM was calculated after adjustment for age, sex, and BMI. Multinomial logistic regression analysis was applied to compare the results. ResultsThe genotyping of rs1342387 G>A SNP in AdipoR1 highlighted an insignificant (OR = 1.53, CI 95% = 0.88–6.32 & P = 0.08) difference of the AA genotype in T2DM patients when compared with the control group. Similarly, the AG genotype was insignificantly (OR = 1.27, CI 95% = 0.93–1.74 & P = 0.14) changed among the two groups after sex and age adjustment. The minor allele frequency A was insignificantly (P = 0.13) differed in T2DM patients (29.88%) with respect to the control group (26.25%). The assessment of characteristics-genotype distribution relationship in T2DM patient group revealed significant elevations of BMI (P = 0.03) and HOMA-IR (P = 0.02) values in subgroups carried the A allele when compared with those of the reference alleles. The evaluation of the results of the genotype distribution, minor allele frequency and characteristics-genotype distribution relationship for the rs12733285C>T SNP failed to point out significant alteration. ConclusionAdipoR1polymorphisms (rs1342387 G>A & rs12733285C>T) do not associate directly with T2DM in Iraqi population. However, there may be an indirect impact via the directing of insulin resistance and BMI.
