Abstract

Aims and Objective: To analyze the difference in expression of different exosomal long non-coding RNA in subjects with DKD vs NDKD.Methodology: Second morning urine sample was collected from histologically confirmed 33 DKD and 30 NDKD patients. Exosomes were isolated using precipitation method and characterised by Nanoparticle Tracker Analysis (Nanosight NS 300) and Atomic Force Microscopy (Pico Plus 5500 ILM). RNA was isolated using trizole method. Expression of long Noncoding RNA (PVT1, MALAT1, NEAT1, HOTAIR, GAS5) were analysed by Real time PCR with respect to endogenous control (18s and B actin).Result: Expression of long non-coding RNA MALAT1 was 1.9 fold higher (p<0.005), NEAT (0.7 times) and PVT (0.9 times) in DKD and there was reduced expression of GAS5 and HOTAIR compared to NDKD.Conclusion: Exosomal long non-coding RNA MALAT1 expression is significantly higher and can serve as future target of research to differentiate DKD from NDKD.
