Abstract

Abstract Biotinylated single base extension (SBE) products that can fluoresce through the successful incorporation of fluorophore-labeled dideoxynucleoside triphosphate were immobilized on streptavidin-coated magnetic bead surfaces via the biotin–streptavidin interaction. The resultant beads were washed and packed into a capillary, followed by fluorescence imaging for detection. Using the above strategy, our bead-based SBE assay could clearly identify the allelic compositions at the targeted site on a tested DNA sequence.
