Abstract
A rapid and sensitive radiochemical microassay for UDP-N-acetylglucosamine pyrophosphorylase (EC 2.7.7.23) activity was developed in which the rate of production of UDP-N-acetylglucosamine was monitored. In this direction the assay was suitable for measuring the amount of pyrophosphorylase activity in cellular extracts in the presence of pyrophosphatase. The Km values for UTP and N-acetylglucosamine 1-phosphate in extracts prepared from Candida albicans are 0.8 and 2.1 mM, respectively.
