Abstract

Nucleic acid (NA) can be utilized as a bio-marker for the detection of infectious disease. NA often presence in a low concentration in a matrix which is one of the major limitations for the utilization of NA. A preconcentration including column separation or centrifugation was used for the separation NA. Another method is to use surface-improved magnetic beads to bind to NA and then separate nucleic acids from their unused parts under an external magnetic field and using of magnetic beads is considered a fast and safe method. Therefore, we are interested to increase the concentration of NA in sample by using methacrylic acid and polyethyleneimine modified magnetic nanoparticles for nucleic acid extraction from samples to study the capturing efficiency of modified MNPs to nucleic acid.In this work we synthesize iron oxide nanoparticles (Fe3O4 NPs) in 4 sizes (11.56 ± 1.17 nm,9.40 ± 1.18 nm,14.07 ± 1.98 nm and 13.98 ± 3.07 nm) by a simple co-precipitation and coat with methacrylic acid on the largest size (14.07 ± 1.98 nm) by the classical radical before polyethyleneimine modification to increase positively charge on the Fe3O4 NPs surface for NA extraction from samples to reduce the process of measuring the infection.
