Abstract

A rapid procedure for the routine estimation of pregnanediol and allopregnanediol in nonpregnancy urine is described. Urine is hydrolyzed with hot hydrochloric acid in the presence of toluene. The toluene extract is washed with 25 per cent sodium chloride in normal sodium hydroxide and water and evaporated to dryness. After acetylation, a portion of the sample is chromatographed on 3 per cent QF-1 at 225°. Quantitation is by reference to an internal standard (androstenedione) added after hydrolysis. Control studies show that the use of an internal standard corrects for variable losses in the extraction procedure and allows an accurate estimation of pregnanediol to be made. Comparison studies with urines routinely assayed by the Klopper technique in another laboratory show that the gas chromatography method described is as good as the Klopper method for the evaluation of ovarian function, with the added advantage that the epimeric diols are measured separately.
