Abstract
• We present a detailed 3-step protocol for live imaging of polarized primary RPE . • Step 1: a protocol for primary porcine retinal pigment epithelial culture. • This method results in highly polarized monolayers within 7 days. • Step 2: two gene delivery protocols that yield efficient transgene expression. • Step 3: High speed live imaging to visualize intracellular traffic in the RPE.
