Abstract
Adenoviral mediated transduction of the tumor suppressor melanoma differentiation-associated gene-7 (Ad-mda7) induces apoptosis in a broad spectrum of cancer cells, but not in normal cells. Transgenic MDA-7 protein expression downregulates β-catenin and PI3K signaling in breast cancer cells. Breast cancers overexpressing Her-2/neu are resistant to conventional chemotherapeutic agents, resulting in significantly lower overall survival rates. Herceptin (Trastuzumab), a monoclonal antibody directed against Her-2/neu, improves the efficacy of chemotherapeutics against Her-2/neu-overexpressing breast cancer cells. In this study, we evaluated the impact of combination therapy of Ad-mda7 and Herceptin on Her-2/neu-positive and negative breast cancer cell lines.
