Publisher Summary This chapter discusses the procedures for the mixed lymphocyte reaction and cell-mediated lympholysis. The two types of systems that can be used for the production of the cultures in mixed lymphocyte culture period are (1) microplate cultures in which subsequent thymidine incorporation and cell-mediated lympholysis assays can be directly applied on individual cultures and (2) bulk mixed lymphocyte cultures, which are sampled for thymidine incorporation, and counted and redistributed in smaller cultures for the subsequent cell-mediated lympholysis. Mixed lymphocyte cultures led to the generation of cytotoxic T cells specific for the immunizing alloantigens, and showed that the addition of interferon during the mixed lymphocyte culture period suppressed the proliferative responses moderately, yet the killer cell product, measured by cell-mediated lympholysis against chromium-labeled lymphoblast targets, is enhanced. Microtiter plates for cell-mediated lympholysis and bulk cultures are left in the incubator for 6 days for standard cell-mediated lympholysis.
