Abstract

This chapter addresses the harmful effects of drugs, both in the context of therapeutic use – so-called adverse drug reactions – and of deliberate or accidental overdose. We are concerned here with serious harm, sometimes life-threatening or irreversible, distinct from the minor side effects that virtually all drugs produce, as described throughout this book. The classification of adverse drug reactions is considered, followed by aspects of drug toxicity, namely toxicity testing in drug development, mechanisms of toxin-induced cell damage, mutagenesis and carcinogenicity, teratogenesis and allergic reactions.
