Cell based therapies using regenerative medicine techniques have emerged as an alternative method to organ transplantation. We have recently developed a novel method of generating acellular whole organ matrix as a scaffold for renal cell transplantation. In this study we investigated whether cells could be effectively distributed throughout the acellular kidney scaffold with the long-term goal of bioengineering a transplantable kidney. METHODS Porcine kidneys were obtained and resident cells were removed by perfusing the renal arteries with detergent and rinsing solutions. Acellular kidney scaffolds were statically seeded with murine MS1 endothelial cells and analyzed with MTS cell proliferation assay for biocompatibility/toxicity studies. Primary renal cells were seeded into the renal parenchyma, and MS1 cells were perfused through the renal artery. The whole organ scaffold was maintained in a perfusion bioreactor up to 72hrs for morphological analysis. RESULTS Whole porcine kidneys were successfully decellularized (removal of cells) within 7 days. Complete infiltration of media via a perfusion bioreactor confirmed the maintenance an intact vascular network. MS1 cells seeded on the scaffolds remained viable under culture conditions, indicating the biocompatibility of the scaffold. Cell attachment was observed with both injection and perfusion seeding of renal and endothelial cells, respectively, however, cell aggregation was observed in smaller vessels at all segments of the kidney. CONCLUSIONS These studies demonstrate that the use of acellular whole organ matrix as a scaffold for cell transplantation is feasible. The processed scaffolds are biocompatible and renal cells can be distributed throughout the scaffolds using optimized protocols.
