Abstract
Endogenous plant hormones are naturally occurring low molecular weight compounds that have various physiological functions in the regulation of plant growth and development. There are several groups or families of hormones each with different physical and chemical properties. The ability of sensitive, fast, accurate and quantitative analysis of plant hormones is very important to elucidate metabolic and signaling processes in plants. Different analytical methods, including immunoassays, electrochemistry, chromatography, and mass spectrometry, have been used over the years, to analyze plant hormones quantitatively. The coupling of liquid chromatography (LC) to fractionate compounds, together with tandem mass spectrometry (MS/MS) to identify compounds, has established LC-MS/MS as the most powerful tool for quantitative analysis of plant hormones due to its high sensitivity, selectivity, and ease of manipulation. In this chapter, we review and introduce the current advances of plant hormones quantitative analysis methods based on LC-MS/MS, including sample preparation procedures and LC-MS/MS detection. Easy-to-follow quantification methods for different plant hormones based on LC-MS/MS are also presented in this chapter to facilitate the plant hormone community.
