Abstract
The ultimate structure and function relationship of any protein lies in its amino acid sequence. The order of the amino acid sequence of a protein is called the primary structure of the protein. Protein sequencing can be done by three methods: Edman degradation, tandem mass spectrometry, and the bioinformatics method in which the protein sequence is derived from cDNA or mRNA or a DNA sequence available in databases. In this chapter, protein sequencing methods such as Edman degradation and tandem mass spectrometry are discussed in detail.
