Abstract

This chapter describes methods of sequence analysis in proteins. Sequence analysis in proteins or peptides must be approached differently. Each protein or peptide presents a different problem which is treated individually. It is deemed that the only rational approach to sequence analysis lies in the study of the methods and techniques used previously by other people. It is essential for the peptide chemist to be flexible in his approach, to base his plan of attack on the experiences of others and adapt this plan as he goes along. The importance of their achievement to the whole of protein chemistry cannot be over-emphasized. Not only was this the first protein to have its primary structure determined, but it is the first real proof of the hypothesis that amino acids in a protein are joined in (X-peptide linkage in a specific sequence. Furthermore, it inspired workers in this field and gave them confidence to tackle other proteins.
