titative real-time polymerase chain reaction (PCR). The mRNA and protein expression level of endothelial nitric oxide synthase (eNOS) were detected by quantitative real-time PCR and western blot. MiR-92a functional assays were performed on rat aortic endothelial cells. RESULTS: Compared to normal control rats, the erectile function (the ratio of maximal intracavernous pressure to mean arterial blood pressure) was found to be dramatically decreased in diabetic rats. Twenty-eight miRNAs were identified significantly dysregulated in diabetic rat’s penile tissues. Among these altered miRNAs, miR-92a expression was dramatically up-regulated in diabetes-induced ED. The expression level of eNOS was significantly lower in diabetic rats’ penes than that in normal control rats. Overexpression of miR-92a significantly decreased the expression of eNOS mRNA and protein in rat aortic endothelial cells. On the contrary, inhibition of miR-92a significantly increased the expression of eNOS mRNA and protein in rat aortic endothelial cells. CONCLUSIONS: This is the first report demonstrating the altered microRNA expression signature in diabetes-associated ED. In vitro study showed that miR-92a was an important regulator of eNOS expression in endothelial cells, indicating upregulation of miR-92a might play an important role in diabetes-associated deterioration of erectile function.
