Abstract

멸종위기종인 남한산 상제나비의 28년 된 장기 건조표본을 이용하여 DNA 바코드 염기 서열을 최초로 분석하고, 이를 유라시아 10 지역 개체군 36개체들과 COI 특성을 비교해 보았다. 이들 개체군에서 총 5개의 haplotype을 확인하였고, haplotype I은 75%로 가장 높은 빈도를 나타내었으며, 유라시아 전 지역에 광범위하게 분포하고 있음을 확인하였다. 남한산 개체들은 모두 haplotype I에 속하고 있어 COI 유전자 상에서는 지역 고립성이 없는 것으로 밝혀졌다. 이 결과로 추후 남한산 상제나비 보전 및 복원은 타 지역 개체군 중 동일 haplotype 선별이 필수 요건으로 판단되나, 좀 더 정교한 평가를 위해서는 추가적인 마커를 이용한 분석이 필요할 것으로 제안한다. Aporia crataegi, an Korean endangered species, was first analyzed for DNA barcode sequences based on 28-year-old dried specimens and compared barcode characters with 36 individuals of ten geographical populations of Eurasia. They were revealed to consist of five different haplotypes. Among them, haplotype I was mostly extensive and high frequency with 75%. The south Korean individuals were confirmed to be belonging to haplotype I and have no genetic isolation on COI gene. By these results, we consider that selection of the identical haplotype from other geographical populations may be a requirement prior to performing for conservation and restoration of the Korean population. We also propose to analyse the additional genetic markers in order to understand a more accurate genetic structures between haplotypes of this species.
