Abstract

A new potential selective anxiolytic GB-115 (N-phenylhexanoylglycyl-L-tryptophan amide) has been created. An optimum method for GB-115 synthesis consists of five stages: (i) phenylhexanoyl chloride synthesis using thionyl chloride; (ii) glycine acylation with resulting chloride in alkaline conditions at 0 °С; (iii) L-tryptophan etherification using thionyl chloride at room temperature; (iv) N-phenylhexanoylglycyl-L-tryptophan ethyl ether synthesis by mixed anhydride method in dimethyl formamide using isobutyl chloroformiate; (v) ammonolysis of resulting dipeptide ether with gaseous ammonia in methanol. The activated ether and carbodiimide methods were also studied in addition to the mixed anhydride method with various solvents and different ways of product extraction.
