Abstract

Aim. The purpose of this study is to develop a methodology for preparing tissue samples from representatives of fish from reservoirs with suspected cases of Haff disease to optimize the conduct of confirmatory biological samples using laboratory animals. Materials and methods. The object of the study was representatives of fish species typical for the affected water bodies (silver carp, Carassius gibelio), as well as control samples of fish that are not a source of the etiological factor of the disease from water bodies of the Tyumen region. To optimize the preparation of material for biological samples, extracts of muscle homogenates and a complex of fish internal organs were obtained using various solvents. The design of the study was developed taking into account the possibility of extracting target chemical compounds from fish tissues that cause the symptoms of “Gaff disease”, as well as preventing the negative effects of the extractants themselves on experimental animals. When preparing the samples of extracts, the peculiarities of the diet and the time of development of the symptoms of the disease in experimental animals were taken into account. Results. Extracts suitable for biological testing were obtained from tissues of experimental and control fish samples using extractants that differ in their physicochemical characteristics and in the spectrum of chemical compounds soluble in them. A proprietary technique has been developed to ensure the most complete extraction of toxic compounds. Conclusion. The most convenient, both from a technological and chemical point of view, is an extract obtained from a homogenate of the muscle tissue of fish caught in affected reservoirs, obtained using a volatile weakly polar aprotic extractant. His further research is aimed at identifying chemical compounds that are the etiological factor that causes the symptoms of Haff disease.
