Synergistic effects of peracetic acid and free ammonia pretreatment on anaerobic fermentation of waste activated sludge to promote short-chain fatty acid production for polyhydroxyalkanoate biosynthesis: Mechanisms and optimization

Peracetic acid (PAA) combined with free ammonia (FA) pretreatment can be utilized to promote anaerobic fermentation (AF) of waste activated sludge (WAS) to produce short-chain fatty acids (SCFAs), and the resulting SCFAs are desirable carbon sources (C-sources) for polyhydroxyalkanoate (PHA) biosynthesis. This work aimed to determine the optimum conditions for PAA + FA pretreatment of sludge AF and the feasibility of using anaerobic fermentation liquor (AFL) for PHA production. To reveal the mechanisms of integrated pretreatment, the impacts of PAA + FA pretreatment on different stages of sludge AF and changes in the microbial community structure were explored. The experimental results showed that the maximum SCFA yield reached 491.35 ± 6.02 mg COD/g VSS on day 5 after pretreatment with 0.1 g PAA/g VSS +70 mg FA/L, which was significantly greater than that resulting from PAA or FA pretreatment alone. The mechanism analysis showed that PAA + FA pretreatment promoted sludge solubilization but strongly inhibited methanogenesis. According to the analysis of the microbial community, PAA + FA pretreatment changed the microbial community structure and promoted the enrichment of bacteria related to hydrolysis and acidification, and Proteiniclasticum, Macellibacteroides and Petrimonas became the dominant hydrolytic and acidifying bacteria. Finally, after alkali treatment, the AFL was utilized for batch-mode PHA production, and a maximum PHA yield of 55.05 wt% was achieved after five operation periods.

Acidogenic Fermentation of Food Waste for the Production of Short-Chain Fatty Acids: The Impact of Inoculum Type and Inoculum Heat Pretreatment

Acidogenic fermentation is an emerging biotechnology that allows for the utilization of food waste as a feedstock to produce high-value products such as short-chain fatty acids (SCFAs), effectively offering a tangible solution for food waste management as well as resource recovery. The objectives of the current study were to identify the ideal inoculum, waste-activated sludge (WAS) or anaerobic digester sludge (AD), for the acidogenic fermentation of food waste at room temperature, as well as to evaluate the impact of heat pretreatment of these inoculums on fermentation performance. The maximum hydrolysis yield of 399 g sCOD/kg VS added was obtained when untreated AD was used as the inoculum, whereas the pretreated AD inoculum provided the highest SCFA yield and conversion efficiency of 238 g sCODSCFA/kg VS added and 71%, respectively. Heat pretreatment had a detrimental impact on the WAS inoculum, leading to lower hydrolysis and SCFA yields, but exerted a positive influence on the AD inoculum. The microbial community showed that heat pretreatment negatively impacted the abundance of non-spore-forming hydrolytic and acidogenic microorganisms. Overall, this study demonstrates the critical role of inoculum type and heat pretreatment in optimizing the acidogenic fermentation process, laying the groundwork for future refinements in SCFA production from food waste through inoculum design.

Thiosulfate/FeCl3 pre-treatment enhances short-chain fatty acid production and mitigates H2S generation during anaerobic fermentation of waste activated sludge: Performance, microbial community and ecological analyses

Anaerobic fermentation (AF) has been identified as a promising method of transforming waste activated sludge (WAS) into high-value products (e.g., short-chain fatty acids (SCFAs)). This study developed thiosulfate/FeCl3 pre-treatment and investigated the effects of different thiosulfate/FeCl3 ratios (S:Fe = 3:1, 3:2, 1:1, 3:4 and 3:5) on SCFA production and sulfur transformation during the AF of WAS. At a S:Fe ratio of 1:1, the maximal SCFA yield (933.3 mg COD/L) and efficient H2S removal (96.5 %) were obtained. S:Fe ratios ≤ 1:1 not only benefited hydrolysis and acidification but largely mitigated H2S generation. These results were supported by the enriched acidogens and reduced sulfur-reducing bacteria (SRB). Molecular ecological network analysis further revealed that the keystone taxon (g_Saccharimonadales) was found in S:Fe = 1:1, together with reductions in associations among methanogens, acidogens and SRB. This work provides a strategy for enhancing high-value product recovery from WAS and minimising H2S emissions.

Soluble carbon source recovery using preconditioning coagulants for applicable short-term fermentation of waste activated sludge in WWTPs

A huge production of waste activated sludge (WAS) has been a burden for wastewater treatment plants (WWTPs) with high disposal cost and little benefit back to wastewater purification. The short-chain fatty acids (SCFAs) produced by a short-term acidogenic fermentation of WAS before methane production have been proven to be a high-quality carbon source available for microbial denitrification process. The dual purpose of full recovery of fermentation liquid products and facilitating disposal of residual solid waste necessitate an efficient solid-liquid separation process of short-term fermentation liquid. The transformation and loss of various soluble carbon sources between solid and liquid are very important issues for carbon recovery efficiency when combining short-term fermentation and sludge dewatering in WWTPs. Here we testified the three conventional preconditioning coagulants, Polyferric Sulfate (PFS), Poly Aluminum Chloride (PAC) and Polyacrylamide (PAM), to improve the efficiency of subsequent solid-liquid separation. The results show that conversion yield of SCFAs in the liquid phase of sludge after short-term fermentation was 195 mg COD/g VSS, when using the coagulants PFS, PAC, and PAM for recovery, the recovery ratio was 79.5%, 82.0%, and 85.9%, respectively, while the dewaterability could be improved after preconditioning short-term fermentation sludge. The complexation of Al3+/Fe3+ in metal coagulants with carboxyl groups of SCFA demonstrated by Density Functional Theory calculation led to small part of soluble carbons co-migration to the solid phase, mainly a loss of high molecular weight organic compounds (carbohydrate, proteins, humic acids), while the application of PAM had little impact on carbon recovery. Economic calculations further showed PAM preconditioning short-term fermentation liquid of WAS could achieve higher recovery benefits.

Improved Short-Chain Fatty Acids Production and Protein Degradation During the Anaerobic Fermentation of Waste-Activated Sludge via Alumina Slag-Modified Biochar.

As the by-product in the biological sewage treatment, waste-activated sludge (WAS) always suffers from the difficulty of disposal. Anaerobic fermentation to achieve valuable carbon sources is a feasible way for resource utilization of WAS, whereas the process is always restricted by its biochemical efficiency. Hence, the WAS was used as the feedstock in this study. Alumina slag-modified biochar (Al@BioC) respectively from pine wood (PW) or fresh vinegar residue (FVR) was employed to stimulate the process of short-chain fatty acids (SCFAs) production during the anaerobic treatment of WAS. The results indicate that the addition of Al@BioC could facilitate the distinct increase in SCFAs yield (42.66 g/L) by 14.09% and acetate yield (33.30 g/L) by 18.77%, respectively, when compared with that in regular fermentation without Al@BioC addition. Furthermore, protein degradation was also improved. With the Al@BioCPW added, the maximum concentration of soluble protein reached 867.68 mg/L and was 24.39% higher than the initial level, while the enhancement in the group with Al@BioCFVR and without biochar addition was 12.49% and 7.44%, respectively. According to the results of 16S rDNA sequencing, the relative abundance of acid-producing bacteria (Bacteroidota and Firmicutes) was enriched, enhancing the pathways of protein metabolisms and the ability to resist the harsh environment, respectively. Moreover, Proteiniphilum under Bacteroidota and Fastidiosipila under Firmicutes were the main microorganisms to metabolize protein. The above results might provide a novel material for harvesting the SCFAs production, which is conducive to harmless disposal and carbon resource recovery.

Co-pretreatment (calcium peroxide + freeze) to improve anaerobic fermentation of waste activated sludge: Method optimization and mechanism insights

The reduction, harmlessness, and recycling of waste-activated sludge (WAS) can be accomplished by anaerobic fermentation, where sludge disintegration is the key speed-limiting process. Co-pretreatment, as a derivative of pretreatment, has attracted more and more attention. However, obtaining optimal co-pretreatment conditions is difficult. Response surface methodology (RSM) can combine statistical analysis with mathematics to optimize the parameters. Therefore, in the current work, RSM optimization for CaO2-freezing co-pretreatment conditions to improve the production of short-chain fatty acids (SCFAs) from WAS was carried out using the Box-Behnken Design based on Design Expert software. We obtained the optimal values of three pretreatment parameters: −18 °C freezing temperature, 48.6 h freezing time, and 0.19 g CaO2/g VSS addition. After 5 days of fermentation, the maximum cumulative yield of SCFAs was 162.34 mg COD/g VSS. CaO2-freezing co-pretreatment promoted the solubilization and release of organic matter in WAS during anaerobic fermentation, which not only provided more organic matter for the subsequent production of SCFAs but also improved the methane production capacity in the anaerobic digestion stage. 16S rRNA genes sequencing revealed that the abundance of SCFAs-producing bacteria (i.e., Macellibacteroides and Proteiniclasticum) was significantly enriched in the fermentation tank after CaO2-freezing co-pretreatment. All in all, this study successfully optimized the co-pretreatment conditions based on RSM, providing new insights and effective solutions for the environmentally friendly and efficient realization of solid waste recycling.

Improved anaerobic sludge fermentation mediated by a tryptophan-degrading consortium: Effectiveness assessment and mechanism deciphering

The hydrolysis of extracellular polymeric substances (EPS) represents a critical bottleneck in the anaerobic fermentation of waste activated sludge (WAS), while tryptophan is identified as an underestimated constituent of EPS. Herein, we harnessed a tryptophan-degrading microbial consortium (TDC) to enhance the hydrolysis efficiency of WAS. At TDC dosages of 5%, 10%, and 20%, a notable increase in SCOD was observed by factors of 1.13, 1.39, and 1.88, respectively. The introduction of TDC improved both the yield and quality of short chain fatty acids (SCFAs), the maximum SCFA yield increased from 590.6 to 1820.2, 1957.9 and 2194.9 mg COD/L, whilst the acetate ratio within SCFAs was raised from 34.1% to 61.2–70.9%. Furthermore, as TDC dosage increased, the relative activity of protease exhibited significant increments, reaching 116.3%, 168.0%, and 266.1%, respectively. This enhancement facilitated WAS solubilization and the release of organic substances from bound EPS into soluble EPS. Microbial analysis identified Tetrasphaera and Soehngenia as key participants in WAS solubilization and the breakdown of protein fraction. Metabolic analysis revealed that TDC triggered the secretion of enzymes associated with amino acid metabolism and fatty acid biosynthesis, thereby fostering the decomposition of proteins and production of SCFAs.

Synergic effects of free ammonia and sodium percarbonate for enhancing short-chain fatty acid production during sludge fermentation: Effectiveness assessment and mechanism elucidation

The production of short-chain fatty acids (SCFAs) from waste activated sludge (WAS) via anaerobic fermentation is typically restricted by poor sludge decomposition capacity and low substrate-availability. Therefore, the free ammonia (FA)‑sodium percarbonate (SPC) technology was presented to successfully overcome the limitation while addressing unsatisfactory acidogenic fermentation pretreated with sole FA or SPC. It revealed that FA + SPC co-pretreatment could boost the SCFA concentration to 347.1 mg COD/g VSS at 180 mg/L FA and 0.15 g/g TSS SPC. In-depth studies demonstrated that FA + SPC pretreatment greatly improved sludge disintegration, biodegradability of substrates and acidification of hydrolysis products. Furthermore, FA + SPC co-pretreatment stimulated the activity of hydrolytic and acidogenic enzymes but inhibited methanogenic enzymes while changing the microbial structure and promoting the enrichment of fermentation microorganisms. The synergistic effect of FA and SPC in this work improves the yield of SCFAs from WAS and facilitates the study of WAS carbon resource recovery.

The dual role of potassium ferrate in promoting primary sludge hydrolysis and acidogenesis in anaerobic fermentation

Potassium ferrate (PF), a green disinfectant has been widely used in drinking water disinfection and has drawn immense attention for its application in sludge treatment. This research study has investigated the effects of PF on the hydrolysis and acidification of primary sludge (PS) to promote the short chain fatty acids (SCFAs) recovery, while achieving solid waste treatment and carbon emission reduction. The results showed that PF could effectively promote the accumulation of SCFAs, and maximum SCFAs yield of 4686.56 mg COD/L was achieved by PF assisted PS fermentation at a PF dosage of 0.4 g/g VSS, which was 2.05 times higher than the control. Substance transformation and enzyme activity analysis indicated that PF significantly promoted the release and hydrolysis of insoluble organic matters from PS through chemical oxidation and alkaline hydrolysis. Furthermore, high doses of PF could also inhibit the growth of methanogens and alter the dominant fermentation flora responsible for SCFAs production. Under the influence of high-dose PF, the foremost acidogenic bacteria were shifted into Trichococcus, Enterococcus, Streptococcus and Gallicola, which might be selectively enriched under the stress of residual Fe (Ⅲ) in the system. The PS fermentation liquid produced after PF pretreatment exhibited a higher C/N ratio and lower phosphorus concentration, which enhanced its suitability as carbon sources for denitrification in WWTPs. This study offered an environmentally friendly method for recovering SCFAs from PS and provided a viable approach to addressing the issue of carbon source shortage in wastewater treatment processes.

Bioavailability and Antioxidant Activity of Rambutan (Nephelium lappaceum) Peel Polyphenols during in Vitro Simulated Gastrointestinal Digestion, Caco-2 Monolayer Cell Model Application, and Colonic Fermentation.

The bioavailability of rambutan peel polyphenols (RPPs) was studied via in vitro simulated digestion, a Caco-2 monolayer cell model, and colonic fermentation. Total phenolic content of RPPs decreased with the progress of the simulated digestion. A total of 38 phenolic compounds were identified during the digestion and colonic fermentation, of which 12 new metabolites were found during colonic fermentation. The possible biotransformation pathways were inferred. Geraniin was transformed into corilagin, ellagic acid, and gallic acid during the digestion and colonic fermentation. Ellagic acid could be further transformed into urolithin under the action of intestinal microbiota. The transformation of ellagitannins could be beneficial to transport on Caco-2 monolayer cell. The antioxidant capacity of RPPs increased with the progress of gastrointestinal digestion. Furthermore, RPPs could increase the yield of short-chain fatty acids, decrease the pH value, promote the growth of beneficial bacteria, and inhibit the growth of pathogenic Escherichia coli/Shigella during colonic fermentation.

A novel solid-state submerged fermenter (3SF) for acidogenic fermentation of food waste at high volumetric loading: Effect of inoculum to substrate ratio, design optimization, and inoculum enrichment

This study reports the development and operation of a solid-state submerged fermenter (3SF) to produce short-chain fatty acids (SCFAs) from food waste at high volumetric loading of 55 gVS/Lreactor. To maximize food waste degradation and SCFA production in the 3SF, three factors were optimized: inoculum to substrate ratio (16%, 9%, 6%, and 4%), mesh pore size of the food waste holding chamber (4.5 mm, 2 mm, and 1 mm), and enrichment of the inoculum. The 3SF operation at higher inoculum-to-substrate ratio (ISR) of 16% resulted in 13–20% higher hydrolysis (600 ± 6.6 gCOD/kgVSadded) and SCFA yields (480 ± 8.7 gCODSCFA/kgVSadded) than those obtained at lower ISRs of 4–6%. Optimization of the mesh pore size (2 mm) further improved hydrolysis and SCFA yield to 628 ± 6.1 gCOD/kgVSadded and 517 ± 7.3 gCODSCFA/kgVSadded, respectively. Enrichment of the inoculum increased the hydrolysis and acidification rates resulting in significantly higher (P ≤ 0.05) hydrolysis and SCFA yields than the non-enriched reactor in just 8 days, implying a 43% reduction in the fermentation time. SCFA composition comprised of acetate, propionate, and butyrate for all reactors. However, an increase in ISR and the use of enriched inoculum resulted in a gradual increase in butyrate composition within the SCFA mix with a maximum butyrate composition of 46% on day 8. Microbial composition showed a high relative abundance of Enterococcus, Clostridium Sensu Stricto 1, and Bacteroides.

Enhancement of waste activated sludge hydrolysis and decomposition by combined free nitrous acid and sodium citrate pretreatment

Hydrolysis of waste activated sludge (WAS) is the limiting step in the anaerobic fermentation process. In view of this, this study evaluated the feasibility of a combination of free nitrous acid (FNA) and sodium citrate (SC) pretreatment to promote WAS hydrolysis and anaerobic fermentation. The results showed that the combined treatment of FNA and SC could promote the decomposition of WAS. Under the conditions of 2.504 mg/L FNA and 0.25 g/g total suspended solids of SC pretreatment for 5 h, the soluble chemical oxygen demand release rate (DDSCOD) was 1.86 and 1.42 times that of FNA and SC pretreatment alone. Meanwhile, the contribution rate of intracellular organic compounds reached 84.18 %. The maximum short-chain fatty acids yield was obtained at the 8th day of fermentation, which was 580.34 mg COD/g volatile suspended solids. The protein utilization rate of combined pretreatment was increased to 74.10 %, which was much higher than that of single treatment. Moreover, FNA-SC also changed the activities of key enzymes, increasing the activities of protease and α-glucosidase but inhibiting the activities of coenzyme F420. Microbial analysis showed that the combined pretreatment significantly changed the composition of microbial community and effectively enriched acid-producing bacteria (such as Petrimonas and Macellibacteroides).

Comparative study on prebiotic effects of different types of prebiotics in an in vitro fermentation by gut microbiota of shrimp (Litopenaeus vannamei)

Prebiotic fermentation by gut microbiota is a key foundation for prebiotics exhibiting physiological functions. However, the relationship among prebiotic type, gut microbiota and gut microbiota metabolism remains unclear. In this study, the gut microbiota of shrimp (Litopenaeus vannamei) was inoculated into medium containing 1% fructooligosaccharide (FOS), galactooligosaccharide (GOS), mannanoligosaccharide (MOS), inulin, β-glucan, isomaltooligosaccharide (IMO) and xylooligosaccharide (XOS). The pH value, digestive enzyme activity, short-chain fatty acid yield and gut microbiota in the fermentation liquid were evaluated. The results showed that after 12 h of in vitro fermentation, β-glucan exhibited the lowest pH value and highest amylase activity among the other types of prebiotics. β-glucan had a higher ability to produce short-chain fatty acids (SCFA) and inhibit the formation of branched-chain fatty acids (BCFA) than the other types of prebiotics. At the genus level, β-glucan markedly promoted the growth of Bacillus. These findings suggest that β-glucan was the most suitable type of prebiotic and had greater prebiotic effects than the other types of prebiotics on the basis of in vitro fermentation by the gut microbiota of shrimp (Litopenaeus vannamei).

Thiosulfate pretreatment enhancing short-chain fatty acids production from anaerobic fermentation of waste activated sludge: Performance, metabolic activity and microbial community

A novel strategy based on thiosulfate pretreatment for enhancing short-chain fatty acids (SCFAs) from anaerobic fermentation (AF) of waste activated sludge (WAS) was proposed in this study. The results showed that the maximal SCFA yield increased from 206.1 ± 4.7 to 1097.9 ± 17.2 mg COD/L with thiosulfate dosage increasing from 0 to 1000 mg S/L, and sulfur species contribution results revealed that thiosulfate was the leading contributor to improve SCFA yield. Mechanism exploration disclosed that thiosulfate addition largely improved WAS disintegration, due to thiosulfate serving as a cation binder for removing organic-binding cations, especially Ca2+ and Mg2+, dispersing the extracellular polymeric substance (EPS) structure and further entering into the intracellularly by stimulated carrier protein SoxYZ and subsequently caused cell lysis. Typical enzyme activities and related functional gene abundances indicated that both hydrolysis and acidogenesis were remarkably enhanced while methanogenesis was substantially suppressed, which were further strengthened by the enriched hydrolytic bacteria (e.g. C10-SB1A) and acidogenic bacteria (e.g. Aminicenantales) but severely reduced methanogens (e.g. Methanolates and Methanospirillum). Economic analysis confirmed that thiosulfate pretreatment was a cost-effective and efficient strategy. The findings obtained in this work provide a new thought for recovering resource through thiosulfate-assisted WAS AF for sustainable development.

Insights into the structural, morphological, and thermal property changes in simulated digestion and fermentability of four resistant starches from high amylose maize starch

Changes in the structural, morphological, and thermal properties of four types of resistant starch (RS), including native (RS2), retrograded (RS3), chemically modified with octenyl succinic anhydride (RS4), and complexed with stearic acid (RS5), were investigated and compared in salivary, salivary-gastric, and salivary-gastric-intestinal digestion. In the process of digestion, the crystal types of RS2, RS3, RS4, and RS5 remained unchanged, maintaining a B-type structure, except for the missing V-type structure of RS5. The total short-chain fatty acid yield of RS2 was significantly higher than that of RS3, RS4, and RS5 at 6 and 12 h of fermentation. RS2 and RS4 had a better proliferation effect on Bifidobacterium in late fermentation than RS3 and RS5, while RS3 could promote proliferation to a larger extent in early fermentation. RS2, RS3, and RS5 had more obvious proliferation effects on Lactobacillus in the late fermentation period. This work suggests that the type of RS plays an important role in influencing the structural and digestive properties of RS during gastrointestinal digestion and can be used to develop formulations of healthy starch-based products with desired functionalities.

Performance and mechanisms of citric acid improving biotransformation of waste activated sludge into short-chain fatty acids

Numerous effective chemical strategies have been explored for short-chain fatty acids (SCFAs) production from waste activated sludge (WAS), but many technologies have been questioned due to the chemical residues. This study proposed a citric acid (CA) treatment strategy for improving SCFAs production from WAS. The optimum SCFAs yield reached 384.4 mg COD/g VSS with 0.08 g CA/g TSS addition. Meanwhile, CA biodegradation occurred and its contribution to the yield of total SCFAs, especially acetic acid, cannot be ignored. Intensive exploration indicated the sludge decomposition, the biodegradability of fermentation substrates, as well as the abundance of fermenting microorganisms were definitely enhanced in the existence of CA. The optimization of SCFAs production techniques based on this study deserved further study. This study comprehensively revealed the performance and mechanisms of CA enhancing biotransformation of WAS into SCFAs and the findings promotes the research of carbon resource recovery from sludge.

Effects of free nitrous acid combined with alkyl polyglucoside on short-chain fatty acids production from waste activated sludge anaerobic fermentation and fermentation liquor for polyhydroxyalkanoates synthesis

Free nitrous acid (FNA) pretreatment and alkyl polyglucoside (APG) addition are environmentally-friendly methods for enhancing waste activated sludge (WAS) anaerobic fermentation (AF) performances. The short-chain fatty acids (SCFAs) produced are available substrates for polyhydroxyalkanoates (PHAs) synthesis to reduce the C-source cost. This study aims to investigate the optimal fermentation conditions of FNA + APG treatment and the performance of PHAs synthesis by the SCFAs from AF system. Through investigating the FNA + APG combined treatment effects, key enzyme activities, and microbial community variation, the AFL from optimal fermentation condition was utilized as a C-source for PHAs synthesis. Experimental results indicated that 0.653 mg/L FNA + 0.075 g/g VSS APG treatment could synergistically achieve the maximum total SCFAs yield of 324.94 mg COD/g VSS at day 5 of AF, which was much more than those of sole FNA or APG treatment. Mechanism analysis indicated that FNA + APG treatment could promote WAS solubilization, hydrolysis and acidogenesis, while severely inhibiting methanogenesis. Microbial community analysis results demonstrated that SCFA producers, including Enterococcus and Clostridium, became the dominant genera in the FNA + APG reactor. Finally, AF liquor after ammonia removal was applied for batch-mode PHAs synthesis, and PHA yields increased to a maximum of 44.44 % of biomass (w/w) after five operation periods.

Novel Insights into the Mechanisms of Periodate-Based Pretreatment in Enhancing Short-Chain Fatty Acids from Waste Activated Sludge

Resource recovery in the form of short-chain fatty acids (SCFAs) from waste activated sludge (WAS) is usually limited due to the quantity and quality of SCFAs. To mitigate these restrictions, this study provides a novel method by using periodate (PI) pretreatment. With an increase in the PI concentration from 0 to 50 mg/g total solids (TS), the SCFA yield increased from 1486 to 3109 mg COD/L and acetic acid production was enhanced by more than twice. Further increase of PI had an adverse effect on SCFA production. Electron spin resonance analysis confirmed that hydroxyl radicals (•OH) and superoxide radicals (•O2–) might play crucial roles in the pretreatment process. Further mechanism studies revealed that PI pretreatment improved the disintegration of WAS and decomposition of humus as well as lignocellulose, thereby providing more substrates for SCFA production. The model-based synthetic wastewater tests combined with enzymatic analyses revealed that the stronger suppression of SCFA consumers compared to SCFA generators causes SCFA accumulation; specifically, the inhibitory order was methanogenesis > hydrolysis > acetogenesis > acidogenesis. The suppressed gene expression of acetate-dependent (such as cdh [EC: 2.1.1.245]) and CO2-type methanogenesis (such as fwd [EC: 2.3.1.101] and mch [EC: 1.5.98.1]) further triggered SCFA accumulation with PI addition. Moreover, PI pretreatment caused the microbial community to shift toward the favorable hydrolytic and acidification-associated (e.g., Longilinea, Acinetobacter, and Macellibacteroides) microorganisms. This study is expected to broaden an unknown application of PI and provide an alternative strategy for resource recovery in some specific circumstances.

Sulfite altered permanganate effects on acetate-enriched short-chain fatty acids production during sludge anaerobic fermentation

Anaerobic fermentation is a promising method for waste activated sludge (WAS) treatment, but ineffective solubilization and hydrolysis limit its application. The current study examined the function of sodium sulfite (SDS) in potassium permanganate (PP)-conditioned WAS fermentation for short-chain fatty acids (SCFAs) biosynthesis. The presence of SDS in the PP system (PP/SDS) reduced the positive effects of PP on total SCFAs yield (2755 versus 3471 mg COD/L), while effectively increasing the proportion of acetate (from 41 to 81 %). Not only did SDS decrease the promoting effects of PP on WAS solubilization and hydrolysis efficiency by 5–42 %, it also shifted microbial metabolic pathways to favor acetate production. In addition, the amino acid metabolism with acetate as end product was enhanced. Moreover, PP/SDS inhibited methanogenesis, resulting in an accumulation of acetate in high quantities. Thus, the current study a provided insight and direction for effective WAS treatment with acetate-enriched SCFAs production.

Simultaneous recovery of short-chain fatty acids and phosphorus during lipid-rich anaerobic fermentation with sodium hydroxide conditioning

Anaerobic fermentation (AF) could achieve simultaneous recovery of short-chain fatty acids (SCFAs) and phosphorus (P) when waste activated sludge (WAS) and meat processing waste (MPW) act as co-substrate. However, long-chain fatty acids, the degradation intermediates of lipids, always inhibit anaerobic microbial activity. Therefore, sodium hydroxide (NaOH) conditioning was applied to improve the lipid-rich AF performance in this study. The results demonstrated that 96% WAS (v/v) with NaOH addition that remaining at pH 7.5 could achieve the maximum SCFAs yield (1180.05 mg/g VSfed) at 12 d, and ortho-P content in the AF liquor (AFL) was much more than that of without NaOH addition. Anaerovibrio and Aminobacterium, one kind of lipolytic and proteolytic bacteria, respectively, became the major genus in the lipid-rich AF system. 86% of P in the AFL from 96% WAS + pH 7.5 reactor was recovered through vivianite crystallization method, with 91% of SCFAs remaining in the post-AFL. Meanwhile, analysis results verified vivianite formation in the P precipitate products. Overall, this study provided a new idea to achieve SCFAs and P simultaneous recovery from WAS and MPW through AF with NaOH conditioning and vivianite crystallization.
