Solid-state Fermentation of Wheat Bran by Aspergillus niger Strains: Effect on the Nutritional Composition and In vitro Digestibility

This study aimed to investigate the effect of Aspergillus niger on the nutritional composition and in vitro dry matter digestibility of wheat bran (WB) in solid-state fermentation. Soluble dietary fibers from solid-state fermentation of wheat bran by the fungus Cordyceps cicadae and their effects on colitis mice.

Ulcerative colitis is a chronic inflammatory disease with a complex pathogenesis for which there is no definitive therapeutic agent. Fermentation, as a green and efficient bioprocessing technique, has been shown to enhance the biological activity of food ingredients. Soluble dietary fiber isolated from plants is thought to have the potential to prevent and alleviate ulcerative colitis. This work was designed to study the differences in the chemical properties of the soluble dietary fiber from wheat bran fermented by Isaria cicadae Miq. (FSDF) and the unfermented soluble dietary fiber from wheat bran (UFSDF) and their effects on colitis mice. The results showed that FSDF and UFSDF differed in molecular weight, monosaccharide compositions, and surface morphology. In addition, supplementation with UFSDF and FSDF ameliorated the symptoms of DSS-induced colitis in mice by attenuating body weight loss, decreasing the disease activity index and splenic index, shortening the length of the colon, and attenuating colonic tissue damage. UFSDF and FSDF also increased the production of the anti-inflammatory cytokine IL-10 and inhibited the expression of IL-6, IL-1β, and TNF-α. The results of gut flora and short-chain fatty acid analyses showed that UFSDF and FSDF improved the diversity of gut microbiota, up-regulated the abundance of some beneficial bacteria such as Akkermansia and Muribaculaceae, increased the levels of acetic acid, propionic acid, and butyric acid, and restored dextran sodium sulfate (DSS)-induced dysbiosis of the intestinal flora in mice. These findings provide guidance for the development of FSDF and UFSDF as functional foods for the relief of ulcerative colitis.

Valorising pasta industry wastes by the scale up and integration of solid-state and liquid-submerged fermentations

Pasta waste has previously been studied in a process to obtain lactic acid through a sequential hydrolysis and fermentation. The process was improved by using enzymes produced via solid-state fermentation of wheat bran in shake flasks. However, the scale-up of the solid-state fermentation is a complex task. In this study, amylase was produced in a home-designed tray bioreactor which allowed to carry out the hydrolysis and fermentation steps at the pilot scale. Due to the efficiency of the solid-state fermentation and the activity of the enzyme, only a small amount (100 g) of wheat bran was required to achieve high yields in a hydrolysis in a 72 L bioreactor (50 L working volume). Overall, the lactic acid yield was 0.68 gLA/gdS, and after the purification, the lactic acid recovered was 55 %, with a total ion concentration of 500 mg/L and an enantiomeric purity of 98.1 % L-LA.

Exploring the Potential of Potato Peels for Bioethanol Production through Various Pretreatment Strategies and an In-House-Produced Multi-Enzyme System

This study aimed to explore the viability of converting potato peel waste into bioethanol using a custom-produced multi-enzyme preparation. Various pretreatment approaches were employed on the potato peels, including thermal, chemical, and thermo-chemical methods. These methods involved boiling for 30 and 60 min, steaming at different pressures and durations, and applying different concentrations of chemicals, including H2SO4, HNO3, CH3COOH, HCl, NaOH, Ca(OH)2, KOH, NH3, and H2O2, either individually or in combination with steam treatment. The pretreated potato peels were subsequently subjected to enzymatic hydrolysis using a crude multi-enzyme cocktail obtained from solid-state fermentation of wheat bran by a naturally occurring strain of Aspergillus niger P-19. This enzyme cocktail consisted of cellulases, hemicellulases, pectinase, and amylases. The most effective pretreatment combination involved the use of 3% H2SO4 followed by steam treatment under pressure, and enzymatic hydrolysis utilizing the crude multi-enzyme preparation. This combination resulted in the highest concentration of reducing sugars (141.04 ± 12.31 g/L), with a carbohydrate conversion rate of 98.49% when a substrate loading of 20% was used. As a result, an ethanol yield of 43.2 ± 3.82 g/L, representing 21.6% based on dry matter, was achieved. Furthermore, supplementing the medium with peptone, (NH4)(H2PO4), and ZnSO4 at a concentration of 0.1% w/v each, along with solid loadings of 22% and 24%, respectively, achieved yield improvements of 51.67 g/L and 54.75 g/L. However, the maximum productivity of 23.4% was observed with a 22% loading, compared to a yield of 22.8% with a 24% solid loading, based on dry matter.

Feruloyl oligosaccharides, isolated from bacterial fermented wheat bran, exhibit antioxidant effects in IPEC-J2 cells and zebrafish model.

Feruloyl oligosaccharides (FOs) were produced by solid-state fermentation of wheat bran using Bacillus subtilis, Bacillus licheniformis, and Saccharomyces cerevisiae, and its antioxidant activity was investigated using IPEC-J2 cells and zebrafish embryo model. Preliminary structure analysis revealed that FOs has an average molecular weight of 11.81 kDa and consists of mannose, ribose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, xylose, arabinose, and fucose. The obtained FOs possess superior reducing power and DPPH and hydroxyl free radical scavenging activities. In IPEC-J2 cells, antioxidant enzymes activities and GSH level were significantly increased, while MDA level was reduced by FOs. Further studies showed that FOs achieved the aforementioned effects by activating Nrf2 signaling pathway. In zebrafish embryo, FOs effectively suppressed ROS production, lipid peroxidation, and cell death by increasing SOD and GSH-Px activities. Our findings suggested that FOs from solid-state fermented wheat bran with mixed bacteria can be used as an antioxidant food additive or drugs.

Solid-state fermentation by Rhizopus oryzae improves flavor of wheat bran for application in food

This study aimed to evaluate the effect of solid-state fermentation of Rhizopus oryzae on the flavor of wheat bran and the sensory properties of bran-containing cake. The results of comprehensive sensory evaluation and electronic nose analysis showed that the optimal time for solid-state fermentation was 36 h. GC-MS analysis showed that the relative levels of unpleasant flavor substances such as n-hexanal, heptanal, and benzaldehyde were decreased, while the relative contents of various aromatic components such as alcohols and esters were increased in fermented wheat bran compared to unfermented wheat bran. Moreover, the addition of fermented wheat bran significantly improved the sensory and nutritional properties of the bran-containing cake. The contents of protein and dietary fiber were significantly increased in the cake, while the carbohydrate content was obviously decreased. Fermented wheat bran not only improved the flavor and texture properties, but also reduced the in vitro starch digestibility. Compared to traditional cakes, the fast digestible starch decreased by 37.03 ± 1.54% and the hydrolysis rate of starch reduced approximately 23.89% in bran-containing cake. Results showed solid-state fermentation of wheat bran by Rhizopus oryzae is a feasible approach to expand the utilization of wheat bran in food processing.

Ethanol from rice byproduct using amylases secreted by Rhizopus microsporus var. oligosporus. Enzyme partial purification and characterization

A three-stage bioethanol bioprocess was developed. Firstly, amylases were obtained from Rhizopus microsporus var. oligosporus using wheat bran in solid-state fermentation. Secondly, amylases hydrolyzed a rice byproduct to make a glucose-rich solution, and this sugar was finally metabolized by Saccharomyces cerevisiae to produce bioethanol. Besides, the secreted enzymes were also partially purified and characterized. The amylase activity (AA) in the crude extract was 358 U/g substrate, and the partially purified enzyme showed the best activity in the 4.0–5.5 pH range. A wide pH stability range (3.5–8.5) was confirmed. The amylase was thermostable up to 60 °C. The ion Mn+2 (10 mM) improved by 60% the AA. There was a 54.9% yield in the conversion of rice residues into reducing sugars in 10 h. The glucose-rich solution was undergone fermentation by S. cerevisiae and showed high ethanol efficiency, 95.8% of the theoretical value. These results suggested a promising technology for bioethanol production.

Cellulases and xylanases are enzymes of industrial significance, particularly in the pulp, paper, textile, and animal feed industries. Moreover, their utilization in the food industry, among them, bakery, brewery, winery and fruit and vegetable juice production, cannot be underestimated. One of the potential sources of enzymes is the filamentous fungi, and hence bio-prospecting of this specific group of microorganisms with the highest levels of cellulase and xylanase secretions is being continuously undertaken. The specific aim of this study was to isolate and characterize cellulase- and xylanase-producing filamentous fungi from termite mounds. Termite mounds have long been established as very good sources of filamentous fungi with the ability to secrete high levels of lignocellulolytic enzymes, and hence an ideal target for the bio-prospecting of cellulases and xylanases. In this study, various groups of filamentous fungi were isolated through enrichment and repeated sub-culturing. This was followed by screening using the Congo red plate-based assay. Cellulase and xylanase activities during the solid-state fermentation of wheat bran were detected and analyzed through spectrophotometry via the 3,5-dinitrosalicylic acid detection system for reducing sugars. The obtained fungal isolates were then finally characterized through zymography, reaction kinetics and morphological studies. Overall, a total of eight different groups of fungi, capable of decomposing cellulose and hemicellulose, were isolated, and their tentative identities established as Fusarium, Didymostible, Penicillium, Phytophthora, Oedocephalum, Aspergillus, Monosporascus and Acremonium. Taken together, findings of this study conceivably showed that termite mounds are a good source of filamentous fungi that in turn are also a good source of cellulases and xylanases that arguably, can be recommended for use in industrial and commercial settings. Production of cellulases by Thermomucor indicae-seudaticae: characterization of a thermophilic β-glucosidase

The current study evaluated the production and characterization of β-glucosidase by the thermophilic fungus Thermomucor indicae-seudaticae in solid-state fermentation of wheat bran. Isolated fungi have significant amounts of β-glucosidase, an enzyme that may be applied to different industrial processes, such as the production of fuels, food, and other chemical compounds. Maximal enzyme activity occurred in pH 3.5–4.5 and at 70 °C. The enzyme exhibited high thermostability, for 1 h, up to 60 °C, and good tolerance to glucose (10 mM) and ethanol (10%). The optimization of fermentative parameters on the production of β-glucosidase was carried out by evaluating the best supplementary nutrient source, pH of nutrient solution, initial substrate moisture and fermentation temperature. The optimization of the above fermentation parameters increased enzyme activity by 120.0%. The highest enzymatic activity (164.0 U/g) occurred with wheat bran containing 70% initial moisture, supplemented with 1.0% (NH4)2SO4 solution at pH 5.5–6.0 and fungus incubated at 40 °C. A more detailed study of β-glucosidase suggested that Sulfur is an important component of the main amino acid present in this enzyme. The enhancer of the enzyme activity occurred when the fungus was grown on wheat bran supplemented with a sulfur-containing solution. In fact, increasing the concentration of sulfur in the solution increased its activity.

Enzymatic saccharification of seaweeds into fermentable sugars by xylanase from marine Bacillus sp. strain BT21.

Enzymatic hydrolysis of seaweed biomass was studied using xylanase produced from marine bacteria Bacillus sp. strain BT21 through solid-state fermentation of wheat bran. Three types of seaweeds, Ahnfeltia plicata, Padina tetrastromatica and Ulva lactuca, were selected as representatives of red, brown, and green seaweeds, respectively. Seaweed biomass was pretreated with hot water. The efficiency of pretreated biomass to release reducing sugar by the action of xylanase as well as the type of monosaccharide released during enzyme saccharification of seaweed biomass was studied. It was seen that pretreated biomass of seaweed A. plicata, U. lactuca, and P. tetrastroma, at 121°C for 45min, followed by incubation with 50IU xylanase released reducing sugars of 233±5.3, 100±6.1 and 73.3±4.1µg/mg of seaweed biomass, respectively. Gas chromatography analysis illustrated the release of xylose, glucose, and mannose during the treatment process. Hot water pre-treatment process enhanced enzymatic conversion of biomass into sugars. This study revealed the important role of xylanase in saccharification of seaweed, a promising feedstock for third-generation bioethanol production.

Antioxidant molecular targets of wheat bran fermented by white rot fungi and its potential modulation of antioxidative status in broiler chickens

ABSTRACT1. The study focused on antioxidant molecular targets of wheat bran fermented by white rot fungi (WRF) in poultry. After solid-state fermentation of wheat bran by WRF for 12 d, scanning electron microscopy found that the lignocellulose structure showed degradation and fragmentation.2. A total of 300 1-d-old broilers were evenly divided by gender and randomly allocated into the following treatments: (1) maize–soybean meal (control group), (2) 10% of wheat bran replacing maize (10% WB group) or (3) 10% of fermented wheat bran replacing maize (10% FWB group).3. The results indicated that the antioxidant gene expression, such as haem oxygenase-1 and glutathione-S-transferase of chicken peripheral blood mononuclear cells, of the 10% FWB group was significantly higher than that of the control group at d 35. For genes of Nicotinamide adenine dinucleotide phosphate oxygenase 1 and reactive oxygen species modulator protein 1, the expression of the 10% FWB group was lower than that of the control group at d 21 and 35.4. In conclusion, wheat bran fermented by WRF could increase lignocellulolytic enzyme activities and the levels of active components that further regulate the expression of antioxidant molecular targets in poultry.

In vitro free radicals scavenging activity and antioxidant capacity of solid-state fermented wheat bran and its potential modulation of antioxidative molecular targets in chicken PBMC

The purpose of this study was to demonstrate the in vitro free radical scavenging activity and antioxidant capacity in solid-state fermented wheat bran and its potential modulation of antioxidative molecular targets in chicken peripheral blood mononuclear cells (PBMC). After solid-state fermentation of wheat bran by white rot fungi for 12 d, the scavenging action of the fermented wheat bran extracts was 1, 1-diphenyl-2-picrylhydrazyl (DPPH), and the free radicals increased significantly, approximately 1.5-fold. Trolox equivalent antioxidant capacity of 1 mg/mL fermented wheat bran extracts was increased from 100 to 150 mM trolox antioxidant capacity after 12 d of fermentation. Moreover, the extracts exhibited 50% of the chelating capacity observed for ferrous iron (Fe2+) after fermenting for 12 d. In vitro, and under the stimulus of fermented wheat bran, the antioxidant gene expression (GST, HO-1, Nrf2, and GCLC genes) of PBMC was more than double that of the PBS, ascorbic acid, and unfermented wheat bran. The expression of fermented wheat bran was the lowest for the NOX1 and ROMO1 genes. Solid-state wheat bran fermented by white rot fungi can increase the scavenging action of DPPH, the trolox equivalent antioxidant capacity, and the chelating capacity of ferrous iron; in addition, in vitro, it can regulate the expression of antioxidant molecular targets in chicken PBMC.

Nigerloxin (2-amido-3-hydroxy-6-methoxy-5-methyle-4-(prop-1’enyl) benzoic acid) is a Lipoxygenase and Aldose reductase inhibitor produced by Aspergillus niger MTCC 5116. It is produced only under solid state fermentation (SSF) and the inoculum required for the inhibitor production is developed in submerged condition. As this is a newly discovered enzyme inhibitor with potential commercial success against diabetic complications, such as neuropathy and cataract, its complete fermentation process parametric study is not reported.  In this study, the role of physical parameters (spore suspen-sion, initial pH, incubation temperature and agitation) in spore germination and pellet size in inoculum development broth in submerged fermentation for the enhanced production of nigerloxin through SSF has been studied. It was concluded that 500 µl of spore suspension in the inoculum development broth at pH 7, incubated at 30°C and 200 rpm gave an ideal  pellet size 1.23 mm resulting in 6.0 mg of nigerloxin/g dry weight of wheat bran in SSF. Solid fermentation of wheat bran for hydrolytic enzymes production and saccharification content by a local isolate Bacillus megatherium.

Back groundFor enzyme production, the costs of solid state fermentation (SSF) techniques were lower and the production higher than submerged cultures. A large number of fungal species was known to grow well on moist substrates, whereas many bacteria were unable to grow under this condition. Therefore, the aim of this study was to isolate a highly efficient strain of Bacillus sp utilizing wheat bran in SSF and optimizing the enzyme production and soluble carbohydrates.ResultsA local strain Bacillus megatherium was isolated from dung sheep. The maximum production of pectinase, xylanase and α-amylase, and saccharification content (total soluble carbohydrates and reducing sugars) were obtained by application of the B. megatherium in SSF using wheat bran as compared to grasses, palm leaves and date seeds. All enzymes and saccharification content exhibited their maximum production during 12–24 h, at the range of 40–80% moisture content of wheat bran, temperature 37-45°C and pH 5–8. An ascending repression of pectinase production was observed by carbon supplements of lactose, glucose, maltose, sucrose and starch, respectively. All carbon supplements improved the production of xylanase and α-amylase, except of lactose decreased α-amylase production. A little increase in the yield of total reducing sugars was detected for all carbon supplements. Among the nitrogen sources, yeast extract induced a significant repression to all enzyme productivity. Sodium nitrate, urea and ammonium chloride enhanced the production of xylanase, α-amylase and pectinase, respectively. Yeast extract, urea, ammonium sulphate and ammonium chloride enhanced the productivity of reducing sugars.ConclusionsThe optimization of enzyme production and sccharification content by B. megatherium in SSF required only adjustment of incubation period and temperature, moisture content and initial pH. Wheat bran supplied enough nutrients without any need for addition of supplements of carbon and nitrogen sources.

Precursor Supply Strategy for Tetramethylpyrazine Production by Bacillus Subtilis on Solid-State Fermentation of Wheat Bran

Tetramethylpyrazine (TTMP) is a widely used flavoring additive with a nutty and roasted taste. Solid-state fermentation (SSF) of wheat bran for producing TTMP was studied with Bacillus subtilis CCTCC M208157, which was an exogenous precursor-independent TTMP-producing strain. Factors influencing endogenous precursor supply and TTMP formation in this strain were investigated. According to the findings, glucose and diammonium phosphate contributed to TTMP production but excess salts caused an inhibition on cell growth and TTMP formation. Then a two-step supply strategy was applied: 10% glucose was added at the beginning of the process to allow acetoin formation, which was the precursor of TTMP, while 3% diammonium phosphate was added only after acetoin accumulation reached its maximum. By applying this strategy, acetoin increased from 5.44 to 13.2g/kg dry substrate (kgds), and consequently the yield of TTMP increased by 6.8 folds from 0.44 to 3.01g/kgds. This was the first report of using a two-step supply strategy for TTMP production by SSF, which proved to be conducive to TTMP production in this strain.

Lignocellulolytic Enzyme Activity of New Corticoid and Poroid Basidiomycetes Isolated from Latvian Cultural Monuments

Eleven corticoid and three poroid fungi have been isolated from Latvian cultural monuments and evaluated for their potential to produce wood-degrading enzymes. In submerged fermentation of mandarin peels, notable intergeneric and intrageneric differences were revealed with regard to the extent of hydrolase and oxidase activity. The xylanase activity of the tested basidiomycetes varied from 0.1 to 2.4 U/mL depending on the fungus species, while that among the strains of Athelia neuhoffii varied from 0.1 to 1.3 U/mL. Five strains of white-rot fungi produced neither laccase nor manganese peroxidases. Cellulase and xylanase of Hyphoderma praetermissum and Tubulicrinis glebulosus appeared to be inducible enzymes. Lignocellulosic substrates strongly affected the enzyme production, because the laccase of H. praetermissum and T. glebulosus was secreted only in the submerged fermentation on mandarin peels and wheat bran, whereas the production of manganese peroxidase by Hypochnicium punctulatum and Phellinus chrisoloma was observed only in the solid-state fermentation of wheat bran and ethanol production residue.

Studies on the production of nigerloxin using agro-industrial residues by solid-state fermentation

Nigerloxin, a new and potent lipoxygenase inhibitor, was discovered in our laboratory through solid-state fermentation of wheat bran by Aspergillus niger V. Teigh (MTCC-5166). The aim of this study is to investigate the possibility of using different agro-industrial residues as nutritional supplements along with wheat bran to enhance the production of nigerloxin. Nigerloxin produced by SSF was quantified spectrophotometrically at 292 nm. The results indicate that the inhibitor production was influenced by the type of solid substrate supplemented, moisture content, pH and size of the inoculum. Individually optimized supplements were tested in different combinations to determine their effects on nigerloxin production. A twofold increase in the production of nigerloxin (4.9 +/- 0.3 mg gds(-1)) was achieved by supplementing wheat bran with 10% w/w sweet lemon peel and 5% v/w methanol at optimized process parameters, that is, an initial moisture content of 65% v/w and incubation period of 6 days with an initial inoculum size of 2 ml (8 x 10(5) spores gds(-1)). Nigerloxin production was stable between pH of 4 and 5.

Lignocellulose-degrading enzyme production by white-rot Basidiomycetes isolated from the forests of Georgia

The production of lignocellulolytic enzymes by eleven basidiomycetes species isolated from two ecosystems of Georgia was investigated for the first time under submerged (SF) and solid-state fermentation (SSF) of lignocellulosic by-products. Notable intergeneric and intrageneric differences were revealed with regard to the extent of hydrolase and oxidase activity. Several fungi produced laccase along with hydrolases in parallel with growth during the trophophase, showing that the synthesis of this enzyme is not connected with secondary metabolism. The lignocellulosic substrate type had the greatest impact on enzyme secretion. Some of the substrates significantly stimulated lignocellulolytic enzyme synthesis without supplementation of the culture medium with specific inducers. Exceptionally high carboxymethyl cellulase (CMCase, 122 U ml(-1)) and xylanase (195 U ml(-1)) activities were revealed in SF of mandarin peelings by Pseudotremella gibbosa IBB 22 and of residue after ethanol production (REP) by Fomes fomentarius IBB 38, respectively. The SSF of REP by T. pubescens IBB 11 ensured the highest level of laccase activity (24,690 U l(-1)), whereas the SSF of wheat bran and SF of mandarin peels provided the highest manganese peroxidase activity (570-620 U l(-1)) of Trichaptum biforme IBB 117. Moreover, the variation of lignocellulosic growth substrate provides an opportunity to obtain enzyme preparations containing different ratios of individual enzymes.

Protease Production by Different Thermophilic Fungi

A comparative study was carried out to evaluate protease production in solid-state fermentation (SSF) and submerged fermentation (SmF) by nine different thermophilic fungi--Thermoascus aurantiacus Miehe, Thermomyces lanuginosus, T. lanuginosus TO.03, Aspergillus flavus 1.2, Aspergillus sp. 13.33, Aspergillus sp. 13.34, Aspergillus sp. 13.35, Rhizomucor pusillus 13.36 and Rhizomucor sp. 13.37--using substrates containing proteins to induce enzyme secretion. Soybean extract (soybean milk), soybean flour, milk powder, rice, and wheat bran were tested. The most satisfactory results were obtained when using wheat bran in SSF. The fungi that stood out in SSF were T. lanuginosus, T. lanuginosus TO.03, Aspergillus sp. 13.34, Aspergillus sp. 13.35, and Rhizomucor sp. 13.37, and those in SmF were T. aurantiacus, T. lanuginosus TO.03, and 13.37. In both fermentation systems, A. flavus 1.2 and R. pusillus 13.36 presented the lowest levels of proteolytic activity.

Production of cellulases and hemicellulases by Penicillium echinulatum grown on pretreated sugar cane bagasse and wheat bran in solid-state fermentation

To evaluate the solid-state fermentation (SSF) production of cellulase and hemicellulases (xylanases), by Penicillium echinulatum 9A02S1, in experiments carried out with different concentrations of the pretreated sugar cane bagasse (PSCB) and wheat bran (WB). This study reports the production of xylanolytic and cellulolytic enzymes by P. echinulatum 9A02S1 using a cheap medium containing PSCB and WB under SSF. The highest amounts of filter paper activity (FPA) could be measured on mixtures of PSCB and WB (32.89 +/- 1.90 U gdm(-1)). The highest beta-glucosidase activity was 58.95 +/- 2.58 U gdm(-1) on the fourth day. The highest activity for endoglucanases was 282.36 +/- 1.23 U gdm(-1) on the fourth day, and for xylanases the activity was around 10 U gdm(-1) from the second to the fourth day. The present work has established the potential of P. echinulatum for FPA, endoglucanase, beta-glucosidase and xylanase productions in SSF, indicating that WB may be partially substituted by PSCB. The incorporation of cheap sources, such as sugar cane bagasse, into media for the production of lignocellulose enzymes should help decrease the production costs of enzymatic complexes that can hydrolyse lignocellulose residues for the formation of fermented syrups, thus contributing to the economic production of bioethanol.
