Identification and functional analysis of cadmium-binding protein in the visceral mass of Crassostrea gigas

The Pacific oyster, Crassostrea gigas, is a traditional food worldwide. The soft body of the oyster can easily accumulate heavy metals such as cadmium (Cd). To clarify the molecular mechanism of Cd accumulation in the viscera of C. gigas, we identified Cd-binding proteins. 5,10,15,20-Tetraphenyl-21H,23H-porphinetetrasulfonic acid, disulfuric acid, tetrahydrate, and Cd-binding competition experiments using immobilized metal ion affinity chromatography revealed the binding of water-soluble high molecular weight proteins to Cd, including C. gigas protein disulfide isomerase (cgPDI). Liquid chromatography–tandem mass spectrometry (LC–MS/MS) analyses revealed two CGHC motifs in cgPDI. The binding between Cd and rcgPDI was confirmed through a Cd-binding experiment using the TPPS method. Isothermal titration calorimetry (ITC) revealed the binding of two Cd ions to one molecule of rcgPDI. Circular dichroism (CD) spectrum and tryptophan fluorescence analyses demonstrated that the rcgPDI bound to Cd. The binding markedly changed the two-dimensional or three-dimensional structures. The activity of rcgPDI measured by a PDI Activity Assay Kit was more affected by the addition of Cd than by human PDI. Immunological analyses indicated that C. gigas contained cgPDI at a concentration of 1.0 nmol/g (viscera wet weight). The combination of ITC and quantification results revealed that Cd-binding to cgPDI accounted for 20% of the total bound Cd in the visceral mass. The findings provide new insights into the defense mechanisms of invertebrates against Cd.

Negative charge at aspartate 151 is important for human lens αA-crystallin stability and chaperone function

Aggregation of lens protein is a major cause of senile cataract. Lens crystallins contain many kinds of modification that accumulate over lifespan. In particular, isomerization of Asp 151 in αA-crystallin has been found in aged lenses; however, its significance is unknown. The purpose of this study was to determine the effects of isomerization of Asp 151 in αA-crystallin. Trypsin digestion followed by liquid chromatography–mass spectrometry analysis of the water-soluble high molecular weight (HMW) fraction from human lens samples showed that isomerization of Asp 151 in αA-crystallin is age-independent, and that 50% of isomerization occurs shortly after birth. However, the extent of Asp 151 isomerization varied with the size of αA-crystallin oligomer species separated from the HMW fraction from aged lens. To evaluate the effects of modification, Asp 151 of αA-crystallin was replaced by glycine, alanine, isoleucine, asparagine, glutamate, or lysine by site-directed mutagenesis. All substitutions except for glutamate decreased heat stability and chaperone function as compared with wild-type αA-crystallin. In particular, abnormal hydrophobicity and alteration of the charge state at Asp 151 caused loss of stability and chaperone activity of αA-crystallin; these properties were recovered to some extent when the mutant protein was mixed 1:1 with wild-type αA-crystallin. The results suggest that, by itself, age-independent isomerization of Asp 151 in αA-crystallin may not contribute to cataract formation. However, the long-term deleterious effect of Asp 151 isomerization on the structure and function of αA-crystallin might cooperatively contribute to the loss of transparency of aged human lens.

Corona alternating current electrospinning: A combined approach for increasing the productivity of electrospinning

Corona alternating current electrospinning (C-ACES), a scaled-up productivity electrospinning method was developed by combining the intense forces of the alternating electrostatic field and a sharp-edged spinneret design with increased free surface. C-ACES reached two orders of magnitude higher productivity (up to 1200 mL/h) than the classical single needle direct current electrospinning (DCES) without any alteration of fiber properties. Polyvinylpyrrolidone K90 (PVPK90), a water soluble high molecular weight nonionic polymer was processed for the first time with single needle alternating current electrospinning (ACES) and C-ACES in order to prepare fast dissolving amorphous solid dispersions of spironolactone (SPIR), a poorly water-soluble antihypertensive model drug. The limited spinnability of PVPK90 with AC high voltage could only be resolved by optimizing the solution conductivity with organophilic salts such as sodium dodecyl sulfate (SDS) demonstrating the importance of conductivity during ACES. The effects of varied solution properties (composition and conductivity) and scaling-up were investigated by SEM imaging. Solid state analyses revealed that SPIR was dispersed in an amorphous form in the fibrous mats. In vitro dissolution tests showed ultrafast drug release in case of the amorphous formulations even when prepared with scaled-up C-ACES. Besides the enhancement of conductivity SDS also prevents SPIR from precipitation from the dissolution media due to its solubilization ability.

Post-translationally modified human lens crystallin fragments show aggregation in vitro

BackgroundCrystallin fragments are known to aggregate and cross-link that lead to cataract development. This study has been focused on determination of post-translational modifications (PTMs) of human lens crystallin fragments, and their aggregation properties. MethodsFour crystallin fragments-containing fractions (Fraction I [∼3.5kDa species], Fraction II [∼3.5–7kDa species], Fraction III [∼7–10kDa species] and Fraction IV [>10–18kDa species]), and water soluble high molecular weight (WS-HMW) protein fraction were isolated from water soluble (WS) protein fraction of human lenses of 50–70 year old-donors. The crystallin fragments of the Fractions I–IV were separated by two-dimensional (2D)-gel electrophoresis followed by analysis of their gel-spots by mass spectrometry. The Fractions I–IV were examined for their molecular mass, particle-diameters, amyloid fibril formation, and for their aggregation by themselves and with WS-HMW proteins. ResultsCrystallin fragments in Fractions I–IV were derived from α-, β- and γ-crystallins, and their 2D-gel separated spots contained multiple crystallins with PTMs such as oxidation, deamidation, methylation and acetylation. Crystallin fragments from all the four fractions exhibited self-aggregated complexes ranging in Mr from 5.5×105 to 1.0×108Da, with diameters of 10–28nm, and amyloid fibril-like formation, and aggregation with WS-HMW proteins. ConclusionThe crystallin fragments exhibited several PTMs, and were capable of forming aggregated species by themselves and with WS-HMW proteins, suggesting their potential role in aggregation process during cataract development. General significanceCrystallin fragments play a major role in human cataract development.

New dietary fibre content results for cereals in the Nordic countries using AOAC 2011.25 method

The definition of dietary fibre (DF) has been revised, and an analytical method AOAC 2011.25 is based on the updates. DF content and monosaccharide composition in composite samples from Swedish, Norwegian, Estonian and Finnish wholegrain wheat flour, rye flour and rolled oats were analysed and compared. Furthermore, DF amounts of 11 industrial and 6 commercial Finnish cereal products were analysed. New DF results are needed, since the national Food Composition Databases (FCDBs) currently contain DF values obtained using older methods underestimating some DF components, such as resistant starch and non-digestible oligosaccharides. The total DF amounts of rolled oats and rye flour were similar among countries, but variation was found in wholegrain wheat flour. The analysed cereal products contained 0.2–5.7g/100g oligosaccharides (SDFS), with the highest level in rye bran. Wheat and rye bran were found to be the best sources for total DF (46 and 32g/100g, respectively), wheat bran was a superior source for water-insoluble DF (IDF; 41g/100g), whereas oat bran was found to contain the most water-soluble high molecular weight DF (SDFP; 6g/100g). These new analytical results for total DF are higher than currently presented in FCDBs.

Degradation of an Old Human Protein

Long-lived proteins exist in a number of tissues in the human body; however, little is known about the reactions involved in their degradation over time. Lens proteins, which do not turn over, provide a useful system to examine such processes. Using a combination of Western blotting and proteomic methodology, age-related changes to a major protein, γS-crystallin, were studied. By teenage years, insoluble intact γS-crystallin was detected, indicative of protein denaturation. This was not the only change, however, because blots revealed evidence of significant cross-linking as well as cleavage of γS-crystallin in all adult lenses. Cleavage at a serine residue near the C terminus was a major reaction that caused the release of a 12-residue peptide, SPAVQSFRRIVE, which bound tightly to lens cell membranes. Several other crystallin-derived peptides with double basic residues also lodged in the cell membrane fraction. Model studies showed that once cleaved from γS-crystallin, SPAVQSFRRIVE adopts a markedly different shape from that in the intact protein. Further, the acquired helical conformation may explain why the peptide seems to affect water permeability. This observation may help explain the changes to cell membranes known to be associated with aging in human lenses. Age-related cleavage of long-lived proteins may therefore yield peptides with untoward biological activity.

Enhancement Potential of Sucrose Laurate (L-1695) on Intestinal Absorption of Water-Soluble High Molecular Weight Compounds

The potential of sucrose fatty acid esters (SEs) to enhance intestinal absorption was investigated in order to identify their utility for the intestinal absorption of water-soluble high molecular weight compounds. Fluorescein isothiocyanate-labeled dextran (FD) with a molecular weight (MW) of 4,000 (FD-4) was used as a model compound, and several SEs were tested as absorption enhancers. After FD-4 was administered intra-duodenally at 10 % (w/v) with the non-loop method in situ in rats in the absence or presence of SEs, the plasma concentration-time profiles of FD-4 were examined. As to sucrose laurate (L-1695), the relationship between concentration and enhancement effect was investigated. In addition, the enhancement effect after dosing into the different small intestinal regions, the effect on FDs with different MWs and the influence of N-acetyl-cysteine (NAC) co-existence were examined. Low water-soluble SEs exhibited slight and/or slow absorption enhancement effects, while L-1695, being highly water-soluble, had good potential to enhance the absorption rate and extent. The enhancement effect became greater as the concentration of L1695 increased. L-1695 displayed high enhancement potential in wide intestinal areas. The enhancement effect of L-1695 (10 %, w/v) depended on MWs of FDs; the mean values of the area under the plasma concentration curve from 0-120 min (AUC0-120 mins) increased by 14 and 8 times for FD-4 and FD-10 (MW 10,000), while it was hardly changed as for FD-70 (MW 70,000). The enhancement effect of L-1695 (10 %, w/v) was similar to that of sodium caprate (10 %, w/v), and was influenced to some extent by the co-existence of NAC (5 %, w/v). The absorption enhancement potential of SEs depended on their water-solubility. L-1695, being highly water-soluble, showed a good enhancement effect, and its absorption profiles were elucidated. This study proposes the possibility of SEs, in particular, L-1695, as intestinal absorption enhancers. As far as the present non-loop method is concerned, the intestinal damage was not observed macroscopically with the addition of L1695 at 2.5-20 % (w/v).

Proteoglycans from Boswellia serrata Roxb. and B. carteri Birdw. and identification of a proteolytic plant basic secretory protein

Water-soluble high molecular weight compounds were isolated in yields of 21-22% from the oleogum of Boswellia serrata and B. carteri. Using anion exchange chromatography and gel permeation chromatography, different proteoglycans were purified and characterized, leading to four principally different groups: (i) Hyp-/Ser-rich extensins with O-glycosidic attached arabinan side chains; (ii) Modified extensins, with arabinogalactosylated side chains containing GlA and 4-O-Me-GlcA; (iii) Glycoproteins with N-glycosidic side chains containing higher amounts of Fuc, Man and GluNH(2,) featuring a 200kD metalloproteinase that has been de novo sequenced and is described for the first time; (iv) Type II arabinogalactans-proteins. Significant differences between the gums from the two species were observed in the protein content (6% vs 22%), offering the possibility of a quick differentiation of gums from both species for analytical quality control. The data also offer an insight into the plant response towards wound-closing by the formation of extensin and AGP-containing gum.

Impact of lipid oxidation-derived aldehydes and ascorbic acid on the antioxidant activity of model melanoidins

As the heat-induced formation of antioxidants throughout the Maillard reaction is known, this study was undertaken to evaluate the impact of lipid oxidation-derived aldehydes and ascorbic acid in Maillard model systems on the resulting antioxidant activity. For this purpose, various fractions of melanoidin-like polycondensation products were obtained from mixtures of amino acids (glycine, lysine, arginine) and lipid oxidation-derived aldehydes (hexanal, (E)-2-hexenal), in the presence or absence of glucose or ascorbic acid. All fractions showed a significant radical scavenging capacity (DPPH assay) and ferric reducing power (FRAP assay). The activity varied according to the composition of the model system tested, although some similar trends were discovered in both assays applied. The presence of lipid oxidation products in the browning products augmented the antioxidant activity in specific cases. For instance, the combined presence of arginine, hexanal and glucose in heated model systems resulted in a significantly higher antioxidant capacity. With an exception of ascorbic acid-containing model systems, melanoidin-like polycondensation products possessed significantly stronger antioxidant activities than the corresponding unheated initial reactant mixtures. Water-soluble high molecular weight (>12kDa) and nonsoluble fractions comprised the major part of the antioxidants derived from amino acid/lipid oxidation product model systems, with or without glucose or ascorbic acid.

Transparent chitosan films reinforced with a high content of nanofibrillated cellulose

Abstract This paper reports the preparation and characterization of nanocomposite films based on different chitosan matrices and nanofibrillated cellulose (NFC) for the purpose of improving strength properties. The nanocomposite films were prepared by a simple procedure of casting a water-based suspension of chitosan and NFC, and were characterized by several techniques: namely SEM, X-ray diffraction, visible spectrophotometry, TGA, tensile and dynamic-mechanical analysis. The films obtained were shown to be highly transparent (transmittance varying between 90 and 20% depending on the type of chitosan and NFC content), flexible, displayed better mechanical properties, with a maximum increment on the Young's modulus of 78% and 150% for high molecular weight (HCH) and water-soluble high molecular weight (WSHCH) filled chitosans, respectively; and of 200% and 320% for low molecular weight (LCH) and water-soluble filled (WSLCH) chitosans, respectively. The filled films also showed increased thermal stability, with, for example, an increase in the initial degradation temperature (Td i ) from 227 °C in the unfilled LCH film up to 271 °C in filled LCHNFC50% nanocomposite films, and a maximum degradation temperature (Td 1 ) raising from 304 °C to 313 °C for the same materials.

Characterisation of Polycondensation Products from Amino Acids and Lipid Oxidation Products

The Maillard reaction, or nonenzymatic browning, includes a complex network of reactions initiated by the condensation reaction of a carbohydrate and an amino compound, and is of utmost importance for the formation of flavour and colour in thermally treated foods. Besides volatile flavour compounds, also brown-coloured polycondensation products, named melanoidins, are produced. In latest years, the interaction of lipid oxidation products in the classical Maillard reaction pathway and vice versa is of particular interest, since it has been shown that the course of both reactions can be modified by the reactants, intermediates and products of the other (ZAMORA & HIDALGO 2005). Therefore, in parallel with the formation of model melanoidins, polycondensation products resulting from the interaction of amino acids with lipid oxidation products were studied. For this purpose, various coloured water-soluble high molecular weight and water-nonsoluble reaction products were isolated from the model reactions of glycine or lysine with a lipid oxidation product (hexanal, (2E)-hexenal, (2E,4E)-decadienal) in the presence or absence of glucose. They were characterised by UV-visible absorbance measurements, elementary analysis, and thermal degradation followed by SPME-GC-MS analysis (ADAMSet al. 2003). The UV-visible absorbance spectra before and after dialysis indicated that the most important contributors to the formation of water-soluble coloured material were constituents of the low molecular fraction. Elementary analysis data showed that a higher amount of nitrogen was incorporated in the high molecular weight fractions as compared to the water-nonsoluble fractions, except for the water-nonsoluble reaction products from amino acid/(2E, 4E)-decadienal interactions, which demonstrated the lowest C/N ratio found. Volatile carbonyl compounds, furans, a liphatic compounds, pyridines, pyrroles and benzene derivatives were the main groups of compounds identified in the thermal degradation profile of each fraction tested. Aldol condensation reactions of the carbonyl compounds were very important in the initial reaction steps. Especially pyridines seem typical indicators of amino acid-lipid oxidation product interactions.

Factors in Various Fractions of Meat Homogenates That Affect the Oxidative Stability of Raw Chicken Breast and Beef Loin

The fractions of meat homogenates were analyzed to find the factors that determine the susceptibility of raw chicken breast and beef loin to lipid oxidation. The fractions used in this study were meat homogenate, precipitate, and supernatant of homogenate after centrifugation, and high and low molecular weight fractions from the supernatant. Chicken breast showed greater oxidative stability than beef loin during 10-d storage (P < 0.05). All fractions from chicken breast showed lower amounts of free ionic iron and myoglobin and higher total antioxidant capacity (TAC) than those from beef loin during storage. The TAC level of chicken breast maintained during storage. This suggested that the oxidative stability of chicken breast was ascribed to high, stable total antioxidant capacity with low level of catalysts for lipid oxidation. The water-soluble high molecular weight fraction, which contained myoglobin, was responsible for the high lipoxygenase-like activity and lipid oxidation potential (LOP) in beef loin. TAC in all fractions from beef loin decreased during storage. This suggested that high myoglobin content in beef loin caused the imbalance between pro- and antioxidant factors leading to the high susceptibility of beef loin to lipid oxidation. Myoglobin served a major source of catalysts, ferrylmyoglobin, hematin, and/or free ionic iron, for lipid oxidation.

Multi-crystallin complexes exist in the water-soluble high molecular weight protein fractions of aging normal and cataractous human lenses

The purpose of the study was to identify non-covalently held complexes that exist in the water-soluble high molecular weight (WS-HMW) protein fractions of normal human lenses of 20-year-old and 60- to 70-year-old, and in the age-matched 60- to 70-year-old cataractous lenses. The WS protein fractions were prepared from five pooled normal lenses of 20-year-old donors or five pooled lenses of 60- to 70-year-old donors or four pooled cataractous lenses (with nuclear opacity) of 60- to 70-year-old donors. Each WS protein fraction was subjected to size-exclusion chromatography using an Agarose A 5m column to recover the void volume WS-HMW protein fraction. A method known as blue-native polyacrylamide gel electrophoresis (BN-PAGE), which allows the isolation of large multi-protein complexes (MPCs) in their native state for further characterization, was used to separate such complexes from individual WS-HMW protein fractions. The protein species that existed as a complex were excised from a gel and trypsin-digested, and the amino acid sequences of the tryptic fragments analyzed by electrospray tandem mass spectrometry (ES-MS/MS). After the second-dimensional sodium dodecyl sulfate–PAGE during BN-PAGE, protein complexes containing a total of 16, 12, and 24 species with M r between 10 and 90 kDa were identified in the HMW protein fractions of normal lenses of 20-year-old, 60- to 70-year-old and cataractous lenses of 60- to 70-year-old donors, respectively. Based on the amino acid sequences of tryptic peptides of individual protein species in the complexes by the ES-MS/MS method, the presence of α-, β-, and γ-crystallin species along with beaded filament proteins (filensin and phakinin) was observed in the 20-year-old normal lenses. The 60- to 70-year-old normal lenses contained filensin and aldehyde dehydrogenase in addition to the above crystallins. Similarly, the age-matched cataractous lenses also contained the above crystallins and aldehyde dehydrogenase but lacked beaded filament proteins. Protein complexes, held mostly via non-covalent bonding, were seen in the WS-HMW proteins of 20-year-old normal, 60- to 70-year-old normal, and 60- to 70-year-old cataractous lenses. The complexes in the normal lenses were made of α-, β-, and γ-crystallin species, beaded filament proteins (filensin and/or phakinin), and aldehyde dehydrogenase. The complexes in the age-matched cataractous lenses also contained these crystallins, and aldehyde dehydrogenase, but not the beaded filament proteins. Further, the crystallin fragments were greater in number in the cataractous lenses compared to the age-matched normal lenses. During multi-angle light scattering (MALS), the HMW proteins from cataractous lenses exhibited species with lower molecular weight range than age-matched normal lenses. The HMW protein preparations from both normal and cataractous lenses showed spherical structures on electron microscopic analysis.

Inhibition of tumor growth and angiogenesis by water extract of Gac fruit (Momordica cochinchinensis Spreng)

The antitumor activity of the crude water extract from Gac fruit (Momordica cochinchinensis) was investigated in vivo and in vitro. A water extract prepared from 0.75 and 0.25 mg dry weight of Gac fruit per gram body weight was given daily to Balb/c mice (n=15/group). The water extract inhibited the growth of the colon 26-20 adenocarcinoma cell line, transplanted in Balb/c mice, reducing wet tumor weight by 23.6%. Histological and immunohistochemical results indicated that Gac water extract reduced the density of blood vessels around the carcinoma. The water extract also produced a marked suppression of cell proliferation in colon 26-20 and HepG2 cells. Cell cycle analysis demonstrated a significant accumulation of cells in the S phase by water extract. Immunoblotting showed that cyclin A, Cdk2, p27waf1/Kip1 were down-regulated, whereas the protein level of p21waf1/Cip1 was not decreased. Treatment of colon 26-20 cells with Gac extract induced necrosis rather than apoptosis. The antitumor component was confirmed as a protein with molecular weight of 35 kDa, retained in the water-soluble high molecular weight fraction. Thus, the bioactive antitumor compound in Gac extract is a protein, which is distinct from lycopene, another compound in Gac fruit with potential antitumor activity.

Genotoxicity and mutagenicity of melanoidins isolated from a roasted glucose–glycine model in human lymphocyte cultures, intestinal Caco-2 cells and in the Salmonella typhimurium strains TA98 and TA102 applying the AMES test

Melanoidins are formed during household cooking procedures and are part of our daily diet, but data on their toxicological potential are still scarce. Therefore, the mutagenic, cytotoxic and genotoxic activity of the water soluble total fraction (sol A), the water soluble high molecular weight fraction (HMW; Molecular weight > 12,400 Da) and the remaining water soluble low molecular weight fraction (LMW) isolated from a glucose–glycine model system roasted at 125 °C was comprehensively studied in human lymphocytes (genetic end point: sister chromatid exchange (SCE)), Caco-2 cells (SCE, cell viability, cell proliferation) and in the Salmonella typhimurium strains TA98 and TA102 (Ames test). Tests were performed in a dose- and time-dependent manner. The results indicate a significant increase in SCE formation in human lymphocytes after the exposure to 0.05% and 0.1% of the melanoidin fractions. In Caco-2 cells, only the exposure to LMW increased the SCE formation as a matter of concentration. Cell's proliferation and viability decreased significantly after exposure to melanoidins. In the AMES test, melanoidins did not show a mutagenic potential, neither using the TA98 nor the TA102 strain. These results show that melanoidins isolated from the glucose–glycine mixture exhibited modest but significant genotoxic effects in human lymphocytes and, in particular the LMW, in Caco-2 cells, but they induce neither in low nor in very high concentrations mutagenicity in bacteria strains.

Identification of 2,5-dimethyl-4-hydroxy-3(2 H)-furanone (DMHF) and 4-hydroxy-2(or 5)-ethyl-5(or 2)-methyl-3(2 H)-furanone (HEMF) with DNA breaking activity in soy sauce

Components with DNA breaking activity in soy sauce were investigated. It was found that there were water soluble high molecular weight DNA breaking components in soy sauce. Two DNA breaking components in the ethyl acetate extract of soy sauce were identified as fragrant components, 2,5-dimethyl-4-hydroxy-3(2 H)-furanone (DMHF) and 4-hydroxy-2(or 5)-ethyl-5(or 2)-methyl-3(2 H)-furanone (HEMF), in addition to the previously characterized DNA breaking fragrant component 4-hydroxy-5-methyl-3(2 H)-furanone (HMF) ( Hiramoto et al., 1996b). Characterization of DNA breaking activity of HEMF was performed, and the mechanisms for the breaking were considered. HEMF cleaved the single strands of supercoiled pBR 322 DNA at pH 7.4 dose dependently and time dependently. DNA breaking was inhibited by superoxide dismutase, catalase, hydroxyl radical scavengers, spin trapping agents and metal chelators, and enhanced by Fe(III) ion. Electron spin resonance–spin trapping technique revealed the generation of hydroxyl radical. Hence, active oxygen species derived from interaction of HEMF with metal ions and oxygen participated in the cleavage. HEMF exhibited mutagenicity to Salmonella typhimurium TA100 without metabolic activation and induced micronucleated mouse peripheral reticulocytes.

Preparation and characterization of water soluble high molecular weight β-cyclodextrin-epichlorohydrin polymers

Water soluble β-cyclodextrin polymers were prepared from β-cyclodextrin crosslinked with epichlorohydrin under basic conditions. The influence of preparation conditions on the molecular weight distribution has been investigated. The knowledge of this relationship allows for the choice of reaction parameters according to the desired weight distribution of the β-cyclodextrin polymer. A procedure was developed for the preparation of high molecular weight water soluble polymer ( Mw higher than 10 4).
