Studies on the retarded recrystallization of tungsten in CIMPLE-PSI exposed under very-high target temperature and long He+-fluence

Abstract Experiments are carried out in CIMPLE-PSI, to understand the recrystallization behavior of tungsten (W) exposed under very-high target temperature and ITER relevant long He+-fluence. The effect of helium bubbles on possible retardation of the recrystallization process is also studied. W samples are simultaneously exposed under He plasma and annealed by the plasma heat-load, in contrast to previously reported experiments in literature, which were carried out sequentially. Exposed samples are characterized by field emission scanning electron microscopy (FESEM), Vickers surface micro-hardness, nano-hardness and electron backscattered diffraction (EBSD). It is observed that the sample exposed to plasma under the highest temperature (1866 K) suffered acute retarded grain growth. This also contained small, unrecovered grains on the exposed surface. FESEM imaging of the cross-sections confirm relatively smaller helium bubbles still form even at the very high temperature conditions, which can impede the grain growth locally, whenever they are forming right on the grain boundaries. This results in an inhomogeneous mixture of surface grains with sizes from few micrometer to few tens of micrometer. EBSD estimates that the plasma exposed surface was only 34% recrystallized. The second sample exposed at a lower temperature (1699 K) but for three times higher fluence (ion fluence: 1.19×1027 m-2) was almost fully recrystallized, which shows retardation diminishes very fast with the duration of the exposure. Hardness measurements were undertaken to understand the variation with plasma exposure/annealing temperature and the extent of recrystallization, with three different probing length scales, spanning from few hundreds of nanometer to several micrometers. Both helium plasma exposed W samples are observed to undergo retarded softening up to a depth of few hundred nanometers from the surface, compared to when the metal may be recrystallized by simple heating, without any plasma exposure.

Evaluation of setting kinetics, mechanical strength, ion release, and cytotoxicity of high-strength glass ionomer cement contained elastomeric micelles

BackgroundLow mechanical properties are the main limitation of glass ionomer cements (GICs). The incorporation of elastomeric micelles is expected to enhance the strength of GICs without detrimentally affecting their physical properties and biocompatibility. This study compared the chemical and mechanical properties, as well as the cytotoxicity, of elastomeric micelles-containing glass ionomer cement (DeltaFil, DT) with commonly used materials, including EQUIA Forte Fil (EF), Fuji IX GP Extra (F9), and Ketac Molar (KT).MethodPowder particles of GICs were examined with SEM-EDX. Setting kinetics were assessed using ATR-FTIR. Biaxial flexural strength/modulus and Vickers surface microhardness were measured after immersion in water for 24 h and 4 weeks. The release of F, Al, Sr, and P in water over 8 weeks was analyzed using a fluoride-specific electrode and ICP-OES. The toxicity of the material extract on mouse fibroblasts was also evaluated.ResultsHigh fluoride levels in the powder were detected with EF and F9. DT demonstrated an initial delay followed by a faster acid reaction compared to other cements, suggesting an improved snap set. DT also exhibited superior flexural strength than other materials at both 24 h and 4 weeks but lower surface microhardness (p < 0.05). EF and F9 showed higher release of F, Al, and P than DT and KT. There was no statistically significant difference in fibroblast viability among the tested materials (p > 0.05).ConclusionsElastomeric micelles-containing glass ionomer cement (DT) exhibited satisfactory mechanical properties and cytocompatibility compared with other materials. DT could, therefore, potentially be considered an alternative high-strength GIC for load-bearing restorations.

Interplay Between Minerals in Bottled Water and Fluoride Toothpaste and Caries Lesion Remineralization.

he aim of this in vitro study was to evaluate the effects of some bottled waters on fluoride toothpaste efficacy in enhancing caries lesion remineralization. Early caries lesions were created in bovine enamel specimens and stratified into treatment groups based on Vickers surface microhardness (VHN). The present study followed a two (fluoride and fluoride-free toothpaste) by five (four bottled waters and tap water) factorial design. The treatment groups were bottled water with the following attributes: (a) 309.9 ppm Ca/1.20 ppm F; (b) 118.4 ppm Ca/0.16 ppm F; (c) 1.00 ppm Ca/1.01 ppm F; and (d) 0.1 ppm Ca/0.04 ppm F and tap water (48.7 ppm Ca/0.7 ppm F). The five water groups were paired either with 1100 ppm fluoride or fluoride-free toothpaste, yielding 10 groups. Specimens were pH-cycled for 10 days with the daily regimen comprised of twice daily toothpaste slurry, with four exposures to water in between. VHN was measured again and the difference was calculated (ΔVHN). Data were analyzed using two-way ANOVA at a 5% significance level. The two-way interaction between water and toothpaste was significant (p<0.001). All groups except fluoride-free toothpaste/bottled water with 0.1 ppm Ca/0.04 ppm F (p=0.411) had significant increases in VHN after pH cycling (p≤0.023). Fluoridated toothpaste resulted in a higher rate of remineralization compared to fluoride-free toothpaste (all p<0.001). Bottled water with 1.20 ppm F/309.9 ppm Ca exhibited the greatest extent of remineralization within fluoride toothpaste groups (p<0.001) and higher remineralization than lower fluoride water in fluoride-free toothpaste groups (p≤0.006). Within the fluoridated toothpaste group, tap water exhibited significantly less remineralization than all bottled waters (all p<0.001). Within the limitation of this study, bottled water with higher fluoride and calcium concentrations might improve fluoridated toothpaste efficacy by enhancing remineralization of early enamel caries-like lesions.

Physiochemical and mechanical characterisation of orthodontic 3D printed aligner material made of shape memory polymers (4D aligner material)

ObjectivesTo conduct a physiochemical and mechanical material analysis on 3D printed shape-memory aligners in comparison to thermoformed aligners. Materials and methodsFour materials were examined, including three thermoformed materials: CA Pro (CP), Zendura A (ZA), Zendura FLX (ZF), and one 3D printed material: Tera Harz (TC-85). Rectangular strips measuring 50 × 10 × 0.5 mm were produced from each material. Five tests were conducted, including differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), shape recovery tests, three-points bending (3 PB), and Vickers surface microhardness (VH). ResultsDSC recorded glass transition temperatures (Tg) at 79.9 °C for CP, 92.2 °C for ZA, 107.1 °C for ZF, and 42.3 °C for TC-85. In DMA analysis at 20–45 °C, a prominent decrease in storage modulus was observed, exclusively for TC-85, as the temperature increased. Notably, within the temperature range of 30–45 °C, TC-85 exhibited substantial shape recovery after 10 min, reaching up to 86.1 %, while thermoformed materials showed minimal recovery (1.5–2.9 %). In 3 PB test (at 30, 37, 45 °C), ZA demonstrated the highest force at 2 mm bending, while TC-85 exhibited the lowest. Regarding VH at room temperature, there was a significant decrease for both ZA and ZF after thermoforming. ZA had the highest hardness, followed by ZF and TC-85, with CP showing the lowest values. ConclusionsTC-85 demonstrates exceptional shape memory at oral temperature, improving adaptation, reducing force decay, and enabling, together with its higher flexibility, extensive tooth movement per step. Additionally, it maintains microhardness similar to thermoformed sheets, ensuring the durability and effectiveness of dental aligners. Clinical relevanceThe 3D printed aligner material with shape memory characteristics (4D aligner) has revolutionized the orthodontic aligner field. It showed mechanical properties more suitable for orthodontic treatment than thermoforming materials. Additionally, it offers enhanced control over aligner design and thickness, while optimizing the overall workflow. It also minimizes material wastage, and reduces production expenses.

Gastric acid challenge of lithium disilicate–reinforced glass–ceramics and zirconia-reinforced lithium silicate glass–ceramic after polishing and glazing—impact on surface properties

ObjectivesTo investigate the impact of simulated gastric acid on the surface properties of lithium disilicate–reinforced glass–ceramics and zirconia-reinforced lithium silicate glass–ceramic after certain polishing and glazing procedures.Materials and methodsFour different types of square-shaped specimens (10 × 10 × 2 mm3, n = 13) were manufactured: lithium disilicate–reinforced glass–ceramic milled and polished (LDS-P); milled, polished, and glazed (LDS-PG); milled, glazed, and no polishing (LDS-G); and milled and polished zirconia-reinforced lithium silicate glass–ceramic (ZR-LS). Specimens were immersed in hydrochloride acid (HCl 0.06 M, pH 1.2) to simulate gastric acid irritation and stored in the acid for 96 h in 37 °C. Specimen weight, surface gloss, Vickers surface microhardness and surface roughness (Ra, Rq, with optical profilometer), and surface roughness on nanometer level (Sq, Sal, Sq/Sal, Sdr, Sds with atomic force microscope) were measured before and after the acid immersion.ResultsZR-LS specimens lost significantly more weight after acid immersion (p = 0.001), also surface microhardness of ZR-LS was significantly reduced (p = 0.001). LDS-G and LDS-PG showed significantly lower surface roughness (Sa, Sq) values compared to LDS-P before (p ≤ 0.99) and after (p ≤ 0.99) acid immersion and ZR-LS after acid immersion (p ≤ 0.99).ConclusionsGastric acid challenge affects the surface properties of lithium disilicate–reinforced glass–ceramic and zirconia-reinforced lithium silicate glass–ceramic. Glazing layer provides lower surface roughness, and the glazed surface tends to smoothen after the gastric acid challenge.Clinical relevanceSurface finish of lithium disilicate–reinforced glass–ceramic and zirconia-reinforced lithium silicate glass–ceramic has a clear impact on material’s surface properties. Gastric acidic challenge changes surface properties but glazing seems to function as a protective barrier. Nevertheless, also glazing tends to smoothen after heavy gastric acid challenge. Glazing can be highly recommended to all glass–ceramic restorations but especially in patients with gastroesophageal reflux disease (GERD) and eating disorders like bulimia nervosa.

Detection of artificial enamel caries-like lesions with a blue hydroxyapatite-binding porosity probe

ObjectivesThis in vitro study investigated the ability of a blue protein-based hydroxyapatite porosity probe to selectively detect artificial enamel caries-like lesions of varying severities. MethodsArtificial caries-like lesions were formed in enamel specimens using a hydroxyethylcellulose-containing lactic acid gel for 4/12/24/72 or 168 h. One untreated group was used as a control. The probe was applied for 2 min and unbound probe rinsed off with deionized water. Surface color changes were determined spectrophotometrically (L*a*b* color space) and with digital photography. Lesions were characterized using quantitative light-induced fluorescence (QLF), Vickers surface microhardness, and transverse microradiography (TMR). Data were analyzed using one-way ANOVA. ResultsDigital photography did not reveal any discoloration in unaffected enamel. However, all lesions stained blue with color intensity positively correlated with demineralization times. The color data reflected similar trends: lesions became significantly darker (L* decreased) and bluer (b* decreased), while overall color differences (ΔE) increased significantly after probe application (4-h lesion, mean±standard deviation: ΔL*=-2.6 ± 4.1/Δb*=0.1 ± 0.8/ΔE=5.5 ± 1.3 vs. 168-h lesion: ΔL*=-17.3 ± 1.1/Δb*=-6.0 ± 0.6/ΔE=18.7 ± 1.1). TMR analysis revealed distinct differences in integrated mineral loss (ΔZ) and lesion depth (L) between demineralization times (4-h lesion: ΔZ=391±190 vol%min × µm/L = 18.1 ± 10.9 µm vs. 168-h lesion: ΔZ=3606±499 vol%min × µm/L = 111.9 ± 13.9 µm). QLF and microhardness were also able to differentiate between demineralization times. L and ΔZ strongly correlated (Pearson correlation coefficient [r]) with Δb* (L vs. Δb*: r=-0.90/ΔZ vs. Δb*: r=-0.90), ΔE (r = 0.85/r = 0.81), and ΔL* (r=-0.79/r=-0.73). ConclusionConsidering the limitations of this study, the blue protein-based hydroxyapatite-binding porosity probe appears to be sufficiently sensitive to distinguish between unaffected enamel and artificial caries-like lesions. Clinical significanceEarly detection of enamel caries lesions remains one of the most critical aspects in the diagnosis and management of dental caries. This study highlighted the potential of a novel porosity probe in detecting artificial caries-like demineralization by objective means.

Effect of Various Acid Solutions as an Aid in Removing the OrthoMTA-Based Root Canal Filling

The objectives of this study were to compare the effects of various acid solutions combined with ultrasonics as an aid to remove mineral trioxide aggregate (MTA)-based root canal filling and to assess their effect on the surface topography and microhardness of root canal dentin. Materials and Method: Fifty human permanent single rooted and single canaled freshly extracted teeth were decoronated and sectioned apically to prepare the middle third of root sections of 5 mm length. The canals were prepared in a step-back manner. OrthoMTA was packed throughout the prepared canals. These root sections were incubated for one week and subsequently randomly allocated to five groups (n = 10) according to the OrthoMTA removal method: No treatment (NT); 5% glycolic acid + ultrasonics (5% GA+U); 10% glycolic acid + ultrasonics (10% GA+U); 10% citric acid + ultrasonics (10% CA+U); Distilled water + ultrasonics (DW+U). A 1 mm deep well was created within the coronal end of the set OrthoMTA. Wells were filled with each respective test solution and left for 5 min. Thereafter, further removal of OrthoMTA used a specific ultrasonic tip. Finally, the canals were flushed using 1 mL of the respective test solutions and activated with a Controlled Memory ultrasonic tip for two cycles of 20 s each followed by flushing with 1 mL of distilled water and paper point drying of the canals. Then, specimens were longitudinally split into two halves and examined under a scanning electron microscope (1000×) to assess the residual OrthoMTA and surface topography of root canal dentin. The Vickers surface microhardness of treated radicular dentin was measured using the HMV-2 microhardness tester. Result: Data were analysed using one-way ANOVA followed by Tukey’s post hoc test. Significant differences for residual OrthoMTA were observed between (10% GA+U) with (5% GA+U), (10% CA+U), (DW+U) and (NT) (p value &lt; 0.01). In the context of microhardness, (5% GA+U) and (10% GA+U) showed statistically significant difference compared to (NT), (10% CA+U) and (DW+U) (p value &lt; 0.01). Conclusion: 10% GA+U was superior to other tested groups in removing OrthoMTA, but it substantially reduced dentin microhardness.

The anti-cariogenic effect of fluoride iontophoresis on sound enamel under different working conditions: an in vitro study

Objective. This study investigated the anti-cariogenic effect of fluoride iontophoresis on sound human enamel under different working conditions. Methods. One hundred and five enamel blocks were prepared and randomly assigned to seven different groups: Fluoride iontophoresis under different working conditions, including: different electric current intensities (0.2, 0.4, 0.6 mA) and durations of current application (3, 6, 9 min); No fluoride treatment (negative control); Fluoride immersion without iontophoresis (positive control). After 7 days of pH-cycling, morphology changes on enamel surfaces were examined by scanning electron microscopy (SEM), and Vickers surface microhardness (VH) values of enamel blocks before and after treatments were assessed using a Vickers microhardness tester. KOH-soluble fluoride (KOH-F) uptakes were measured by a fluoride ion-selective electrode. Results. Distinctly roughened enamel surfaces were showed in all treatment groups, with the greatest extent damaged surface in negative control group and the least in all the fluoride intophoresis groups. All the fluoride iontophoresis groups (except for the group under 0.2 mA) showed smaller reduction of VH values (p < 0.05) and greater KOH-F (p < 0.05) when compared with the negative and positive control groups. There was an increasing trend in the loss of VH and in fluoride uptakes with an increasing current intensity. Meanwhile, the reduction of VH values and KOH-F in Group I3 ( 0.6 mA) were significantly greater (p < 0.05) than those in Group I1 ( 0.2 mA). No significant differences of reduction of VH and KOH-F presented among the three groups applied with fluoride iontophoresis under different current durations. Conclusions. Fluoride iontophoresis resulted in a significantly increased KOH-F and a reduced loss of VH after the 7 days pH-cycling procedure, and its effectiveness was influenced by different electric current intensities but not influenced by different current durations.

Assessment of Physical/Mechanical Performance of Dental Resin Sealants Containing Sr-Bioactive Glass Nanoparticles and Calcium Phosphate.

The aim of this study was to assess the chemical/mechanical properties of ion-releasing dental sealants containing strontium-bioactive glass nanoparticles (Sr-BGNPs) and monocalcium phosphate monohydrate (MCPM). Two experimental sealants, TS1 (10 wt% Sr-BGNPs and 2 wt% MCPM) and TS2 (5 wt% Sr-BGNPs and 4 wt% MCPM), were prepared. Commercial controls were ClinproXT (CP) and BeautiSealant (BT). The monomer conversion (DC) was tested using ATR−FTIR (n = 5). The biaxial flexural strength (BFS) and modulus (BFM) were determined (n = 5) following 24 h and 7 days of immersion in water. The Vickers surface microhardness (SH) after 1 day in acetic acid (conc) versus water was tested (n = 5). The bulk and surface calcium phosphate precipitation in simulated body fluid was examined under SEM-EDX. The ion release at 4 weeks was analyzed using ICP-MS (n = 5). The DC after 40 s of light exposure of TS1 (43%) and TS2 (46%) was significantly lower than that of CP (58%) and BT (61%) (p < 0.05). The average BFS of TS1 (103 MPa), TS2 (123 MPa), and BT (94 MPa) were lower than that of CP (173 MPa). The average BFM and SH of TS1 (2.2 GPa, 19 VHN) and TS2 (2.0 GPa, 16 VHN) were higher than that of CP (1.6 GPa, 11 VHN) and BT (1.3 GPa, 12 VHN). TS1 showed higher Ca, P, and Sr release than TS2. Bulk calcium phosphate precipitation was detected on TS1 and TS2 suggesting some ion exchange. In conclusion, the DC of experimental sealants was lower than that of commercial materials, but their mechanical properties were within the acceptable ranges. The released ions may support remineralizing actions.

Biomimetic Enamel Remineralization: A Comparative in vitro Study of Self-Assembling Peptide, Fluoride Varnish, and their Combination at Different Times by Surface Microhardness Test

Aims: This study was aimed to evaluate the efficacy of self-assembling peptide, fluoride varnish, and their combination to remineralize the artificial carious lesion in vitro. Materials and methods: Sixty extracted sound premolars were randomly divided into four groups. The artificial carious lesion was created by immersion the specimens in the demineralizing solution. The enamel surface treated with: Group1: (n=15) artificial saliva alone, group2: (n=15) fluoride varnish, group3: (n=15) self-assembling peptide, group4: (n=15) self-assembling peptide + fluoride varnish. The enamel surface was assessed by Vickers surface microhardness at the baseline, after demineralization, 1-week, and 4-weeks after remineralization. Results: all four groups exhibited high statistically significant differences between the different times of test periods, In 1 and 4-weeks after remineralization, the greatest surface microhardness mean value was seen in the self-assembling peptide + fluoride varnish group followed by self-assembling peptide group then fluoride varnish group while the artificial saliva group showed the least surface microhardness mean value with a highly statistically significant difference between them. Conclusions: self-assembling peptide superior to fluoride varnish in remineralization ability and this potential is boosted when they combined together. Relating to surface microhardness, remineralization is a dynamic process that increases with time.

Longitudinal In Vitro Effects of Silver Diamine Fluoride on Early Enamel Caries Lesions.

This laboratory study evaluated the longitudinal surface microhardness changes in early, incipient, noncavitated, white-spot, enamel caries lesions treated with silver diamine fluoride (SDF). Five intervention groups (SDF, AgNO3, KF, 5% sodium fluoride varnish (FV), deionized water (DI)) × two-time intervals after intervention (immediate & delayed pH-cycling) resulted in 10 groups (n=18). Silver nitrate (AgNO3) and potassium fluoride (KF) groups served as controls to assist in evaluating if remineralization effects were due to the silver or fluoride component in SDF. Early, incipient, noncavitated, white-spot, enamel caries lesions were created in bovine enamel, the extent of demineralization was determined using Vickers surface microhardness (VHNlesion). Intervention treatments were applied. Half the specimens from each group underwent immediate 5-day pH-cycling, and half were stored in an incubator with artificial saliva for 2 weeks before undergoing 5-day pH-cycling. After pH-cycling, lesion hardness was evaluated using VHNpost. Specimens were then exposed to a second demineralization challenge, and lesion softening was evaluated (VHNsecdem). Hardness variables were calculated: ΔVHN = VHNpost - VHNlesion; ΔVHNsecdem = VHNsecdem - VHNpost. Data were analyzed using two-way ANOVA (α=0.05). Immediately cycled, SDF had significantly (p<0.0001) greater remineralization than DI, AgNO3, and FV. All delayed cycling groups had significantly greater remineralization than FV (p<0.0001). Significantly greater remineralization was noted in delayed AgNO3 (p≤0.0001), DI (p=0.0003), and FV (p=0.0006) compared to immediately cycled. After the second demineralization challenge, FV had significantly less surface softening than AgNO3 (p=0.0002), DI (p=0.0003), KF (p=0.0225), and SDF (p=0.0388) intervention groups. No significant difference was found between the pH-cycle timings (p=0.2710). Based the present findings, FV may be better suited than SDF to treat early, incipient, noncavitated, white-spot, enamel caries lesions.

The effect of silver diamine fluoride in preventing in vitro primary coronal caries under pH-cycling conditions

ObjectivesThis study investigated the ability of SDF, and its individual components, silver (Ag+) and fluoride (F−) ions, in preventing enamel demineralization under pH-cycling conditions in the presence or absence of twice-daily fluoride application. DesignPolished human enamel specimens were assigned to five treatment groups (n = 36 per group): SDF (38 %); SDF followed by application of a saturated solution of potassium iodide (SDF + KI); silver nitrate (AgNO3; silver control, 253,900 ppm Ag); potassium fluoride (KF; fluoride control, 44,800 ppm F); deionized water. Treatments were applied once. Specimens in each treatment group were divided into two subgroups (n = 18). During the subsequent 7-day pH-cycling phase, specimens were treated twice daily with either 275 ppm fluoride as sodium fluoride or deionized water, immediately before and after a 3-h cariogenic challenge with exposure to artificial saliva at all other times. Changes in color, Vickers surface microhardness (SMH), transverse microradiography (TMR) was calculated. Data were analyzed using two-way ANOVA. ResultsIn both models, SDF, SDF + KI and KF were superior in inhibiting demineralization compared to AgNO3 and deionized water (p < 0.0001). There was no statistically significant difference between SDF, SDF + KI and KF with twice daily fluoride treatments (p > 0.8). However, KF was more effective in preventing demineralization than SDF and SDF + KI in the absence of fluoride treatments (p = 0.0002). KI did not affect the ability of SDF to prevent demineralization (p > 0.4). ConclusionSDF and SDF + KI appears to be an effective option in preventing primary coronal caries.

Effectiveness of in vitro primary coronal caries prevention with silver diamine fluoride - Chemical vs biofilm models

ObjectivesThe main goal of this study was to investigate the effectiveness of SDF and its individual components, silver (Ag+) and fluoride (F−) ions, in preventing enamel demineralization using biofilm and chemical models. MethodesPolished human enamel specimens were assigned to five treatment groups (n = 18 per group): SDF (38 %); SDF followed by application of a saturated solution of potassium iodide (SDF + KI); silver nitrate (AgNO3; silver control, 253,900 ppm Ag+); potassium fluoride (KF; fluoride control, 44,800 ppm F); deionized water (DIW). Treatments were applied once to sound enamel. In the biofilm model, specimens were demineralized by aerobic overnight incubation using cariogenic bacteria isolated from human saliva in brain heart infusion supplemented with 0.2 % sucrose for three days. In the chemical model, enamel specimens were immersed in a demineralizing solution containing 0.1 M lactic acid, 4.1 mM CaCl2, 8.0 mM KH2PO4, 0.2 % Carbopol 907, pH adjusted to 5.0 for five days. Vickers surface microhardness was used to determine the extent of enamel demineralization. Data were analyzed using one-way ANOVA. ResultsIn the chemical model, there was no statistically significant difference between SDF and SDF + KI in preventing coronal caries (p < 0.0001). In the biofilm model, SDF + KI was significantly less effective in preventing demineralization than SDF (p < 0.0001). In both models, SDF and SDF + KI were superior in their ability to prevent caries lesion formation than AgNO3 and DIW. ConclusionKI application after SDF treatment appears to impair SDF’s ability to prevent biofilm-mediated but not chemically induced demineralization. Clinical significanceSDF may be a viable option in preventing primary coronal caries.

Calcium silicate cement interface with restorative materials through layering after different time intervals.

The aim was to evaluate the interfacial characteristics of Biodentine, CEM Cement, and ProRoot MTA when restored with different final restorative materials after different time intervals. Biodentine, CEM Cement and ProRoot MTA were layered with amalgam, composite resin or light cure glass ionomer cement. Layering was done either immediately, 24 or 72h after cement placement. The interface of cements with restorative materials was characterized using scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS) after separation. Vickers surface microhardness test was also performed on the interface. Statistical analysis included two-way Anova, Dunnett T3, and Tukey HSD. The significance level was set at P < 0.05. The highest microhardness values were seen when restorative materials were layered after 24h in the case of Biodentine, and after 72h in the case of CEM Cement and ProRoot MTA. In ProRoot MTA no significant difference was seen in the microhardness when layered with different restorative materials regardless of the time of layering. In immediate layering, Biodentine exhibited the highest microhardness values. Both immediate and delayed layering resulted in element transfer between calcium silicate cements (CSCs) and restorative materials. Deposition and depletion of element occurs subsequent to layering of restorative materials on CSCs. When immediate layering is necessary, Biodentine may be a better option compared to other CSCs evaluated.

The ability of dual whitening anti-caries mouthrinses to remove extrinsic staining and enhance caries lesion remineralization – An in vitro study

ObjectivesThis laboratory study investigated the ability of dual whitening anti-caries mouthrinses to remove extrinsic staining from artificially stained caries lesions and to enhance their remineralization and fluoridation. Materials and MethodsEarly caries lesions were created in bovine enamel specimens. The lesions were artificially stained and pH cycled for 10 days with the daily cycling regimen consisting of twice daily 60s-treatments with one of 11 mouthrinses, a 4-h demineralization period and artificial saliva treatments in between. Mouthrinses were eight commercially available products, all containing 100ppm fluoride but utilizing hydrogen peroxide, pyro-, tri- or hexametaphosphate salts and/or sodium bicarbonate. The three control mouthrinses were 100ppm fluoride, 30 % hydrogen peroxide and deionized water. Enamel color changes (ΔE) were determined spectrophotometrically. Vickers surface microhardness (VHN) was used to determine lesion remineralization. Enamel fluoride content (EFC) was determined using the microbiopsy technique. Data were analyzed using ANOVA. ResultsΔE was significantly different among groups (p=0.0045). Thirty percent hydrogen peroxide was superior to all other mouthrinses, while there were no differences between commercial mouthrinses and deionized water. There were small, directional but non-significant differences between commercial mouthrinses with those containing hydrogen peroxide providing better whitening. There were no significant differences between mouthrinses in their ability to remineralize caries lesions (p=0.2898). EFC differed among groups (p<0.0001), with the two mouthrinses containing pyrophosphate salts having lower EFC than all but the deionized water group. ConclusionsArtificially stained caries lesions show reduced susceptibility to fluoride remineralization and whitening effects of commercial whitening and anti-caries mouthrinses. Clinical RelevanceArtificially stained caries lesions appear to require stronger than over-the-counter interventions to successfully whiten and remineralizing them.

Influence of salivary conditioning and sucrose concentration on biofilm-mediated enamel demineralization.

The acquired pellicle formation is the first step in dental biofilm formation. It distinguishes dental biofilms from other biofilm types. Objective To explore the influence of salivary pellicle formation before biofilm formation on enamel demineralization.Methodology Saliva collection was approved by Indiana University IRB. Three donors provided wax–stimulated saliva as the microcosm bacterial inoculum source. Acquired pellicle was formed on bovine enamel samples. Two groups (0.5% and 1% sucrose–supplemented growth media) with three subgroups (surface conditioning using filtered/pasteurized saliva; filtered saliva; and deionized water (DIW)) were included (n=9/subgroup). Biofilm was then allowed to grow for 48 h using Brain Heart Infusion media supplemented with 5 g/l yeast extract, 1 mM CaCl2.2H2O, 5% vitamin K and hemin (v/v), and sucrose. Enamel samples were analyzed for Vickers surface microhardness change (VHNchange), and transverse microradiography measuring lesion depth (L) and mineral loss (∆Z). Data were analyzed using two-way ANOVA.Results The two-way interaction of sucrose concentration × surface conditioning was not significant for VHNchange (p=0.872), ∆Z (p=0.662) or L (p=0.436). Surface conditioning affected VHNchange (p=0.0079), while sucrose concentration impacted ∆Z (p<0.0001) and L (p<0.0001). Surface conditioning with filtered/pasteurized saliva resulted in the lowest VHNchange values for both sucrose concentrations. The differences between filtered/pasteurized subgroups and the two other surface conditionings were significant (filtered saliva p=0.006; DIW p=0.0075). Growing the biofilm in 1% sucrose resulted in lesions with higher ∆Z and L values when compared with 0.5% sucrose. The differences in ∆Z and L between sucrose concentration subgroups was significant, regardless of surface conditioning (both p<0.0001).Conclusion Within the study limitations, surface conditioning using human saliva does not influence biofilm–mediated enamel caries lesion formation as measured by transverse microradiography, while differences were observed using surface microhardness, indicating a complex interaction between pellicle proteins and biofilm–mediated demineralization of the enamel surface.

Effects of intracoronal bleaching agents on the surface properties of mineral trioxide aggregate.

Bleaching agents may affect surface properties of mineral trioxide aggregate (MTA) as a coronal barrier. The purpose of this study was to investigate surface properties of MTA after exposure to intracoronal bleaching agents. MTA was set in acrylic molds with a 4mm high central hole and a 6mm diameter. Specimens were divided into four groups (n = 10); three groups were exposed to bleaching agents three times on every fourth day (carbamide peroxide-CP, hydrogen peroxide-HP, sodium perborate-SP) and a control group-C. The surface roughness and Vickers surface microhardness were measured. Differences between groups were analyzed using a Kruskal-Wallis test and intergroup comparisons were assessed with a Mann-Whitney U test with a Bonferroni correction (p < 0.0001). The microstructure and elemental composition were observed using a scanning electron microscope (SEM) and an energy-dispersive X-ray microanalysis (EDX) system. In terms of microhardness, the decrease in the HP group was significantly greater than that of the CP and SP groups; CP group significantly greater than that of the SP group, however, there was no significant difference between the SP and C groups. Surface roughness values were compared between groups, and no significant differences were observed between the CP and HP groups, and they exhibited significantly higher roughness values than the SP and C groups. SEM/EDX showed that the bleaching agents affected the elemental distribution. Bleaching agents adversely affected the surface roughness, surface microhardness and elemental distribution of MTA, with exposure to SP causing fewer changes on the surface properties than CP or HP.

Effects of PVP-Iodine pH and Calcium Concentration on Fluoride Varnish Anti-Caries Efficacy In Vitro.

This laboratory study investigated the effects of PVP-iodine solutions with varying pH and calcium concentrations on enamel remineralization and fluoridation by subsequent treatment with fluoride varnish. Caries-like lesions were created in bovine enamel specimens (n = 15 per group) and characterized using Vickers surface microhardness (VHN). Specimens were treated with 10% PVP-iodine solutions varying in calcium concentration (0/10/100 mM) and pH (3.0/4.0/5.0), followed by 5% sodium fluoride varnish. A fluoride varnish-only control group was included. Specimens were then placed into artificial saliva for 16 h. The varnish was removed, hardness measured and enamel fluoride uptake (EFU) determined using the microbiopsy technique. Data were analyzed with one-way ANOVA. Groups receiving PVP-iodine pre-treatments exhibited directionally greater rehardening (range: p = 0.0001 - 0.7008) and EFU (p = 0.0001-0.2670) than the control group. The presence of calcium in the pre-treatment enhanced rehardening. the groups '10mM Ca/pH 3.0' (∆VHN = 10.5 ± 6.3), '100mM Ca/pH 3.0' (∆VHN = 9.7 ± 4.1) and '10mM Ca/pH 5.0' (∆VHN = 8.7 ± 7.0) displayed the highest numerical gain vs the control (∆VHN=3.6±2.2). Different pH values had a minor effect on rehardening and EFU. The calcium effect was more pronounced for EFU than for rehardening with all three '100 mM Ca' groups exhibiting higher EFU (7.0 - 7.2 µg F/cm2) than all other groups (6.1 - 6.9 µg F/cm2). PVP-iodine pre-treatments can be modified to enhance the rehardening and fluoridating effect of fluoride varnishes, thereby potentially improving their ability to prevent caries in vivo. Although numerical differences between groups were small, the addition of high concentrations of calcium paired with a low pH appears most favorable under the present conditions.

Setting Time and Surface Microhardness of Mineral Trioxide Aggregate and 1% and 5% Fluoride-Doped Mineral Trioxide Aggregate Mixed with Water and Gel-like Polymer.

Mineral trioxide aggregate (MTA) is extensively used in endodontics. However, it has limitations such as long setting time, low compressive strength and poor handling properties. Our study aimed to compare the setting time and surface microhardness of MTA and fluoride-doped MTA (FMTA) using gel-like polymer (GLP) or distilled water (DW) as liquid. An MTA-like cement was prepared by mixing Portland cement, bismuth oxide and gypsum (75%, 20% and 5%, respectively). FMTA (1% and 5%) was made by substituting 1% and 5% of MTA powder with fluoride. GLP, composed of methyl cellulose (MC) and propylene glycol (PG), was used as the hydrating liquid and compared with distilled water. Six experimental groups (n=10) were examined for each test. The samples were subjected to Vickers surface microhardness test after 4 and 28 days. Setting time was measured using ANSI/ADA standards. Data was analyzed using two-way and repeated measured ANOVA and the Tukey HSD tests. The MTA-like cement hydrated with GLP showed a significantly reduced setting time (P<0.05); 1% FMTA, mixed with GLP, had the shortest initial and final setting times. The microhardness values of all samples increased at different rates during 28 days (P<0.00001). The microhardness significantly decreased by addition of fluoride to MTA (P<0.001). The results of the present in vitro study suggested that using GLP as the hydrating liquid enhances the setting time with no adverse effect on the surface microhardness. However, adding fluoride can negatively affect the microhardness of MTA.

Caries lesion remineralization with fluoride toothpastes and chlorhexidine - effects of application timing and toothpaste surfactant

Habitual toothbrushing with fluoridated toothpaste followed by rinsing with antibacterial mouthwashes is a method to maintain good oral hygiene and to diminish the occurrence and severity of dental caries and periodontal disease. However, our understanding of how antimicrobial agents in mouthwashes affect fluoride-mediated caries lesion remineralization is still poor.Objective:The objectives of this in vitro study were a) to determine the effects of the waiting period of chlorhexidine (CHX) rinsing after fluoride toothpaste use and b) to further determine the effect of the type of toothpaste surfactant [sodium dodecyl sulfate (SDS) or cocamidopropyl betaine (CAPB)] on caries lesion remineralization associated with CHX rinsing.Material and Methods:Caries lesions were formed in bovine enamel specimens and assigned to 10 treatment groups (n=18) based on Vickers surface microhardness (VHN). Lesions were then pH-cycled for 10 days with daily regimen comprised of twice daily toothpaste slurry treatments (1150 ppm fluoride, with SDS or CAPB), followed by CHX solution treatments [0, 15, 30 or 60 minutes following slurry treatment or no CHX treatment (negative control)]. VHN was measured again and the extent of lesion remineralization calculated (∆VHN).Results:∆VHN with SDS-toothpaste was significantly lower than with CAPB-toothpaste, indicating more remineralization for the CAPB-toothpaste. ∆VHN with 0-minute waiting time was significantly lower than with 30-minute waiting time and with negative control.Conclusions:The absence of CHX as an adjunct to fluoride toothpastes led to greater remineralization of enamel lesions compared with the immediate use of CHX treatment for both SDS- and CAPB-toothpastes. CAPB-toothpastes indicated significantly greater remineralization than SDS-toothpastes, and can be suggested for patients at high risk of caries. A 30-minute waiting time for CHX treatment is recommended after brushing.
