Caffeine quantification in dietary supplements using high-throughput on-line solid phase extraction coupled to Venturi easy ambient sonic-spray ionization mass spectrometry.

Caffeine is present in a large number of beverages and is an additive used in dietary supplements. Therefore, the concern about its quality and safety for consumers has been increasing and hence requires faster and simpler analytical methods to determine the caffeine amount. The high-throughput analysis is an appropriate solution to pharmaceuticals, bioanalysis, forensic and food laboratory routines. In this sense, Venturi easy ambient sonic-spray ionization mass spectrometry (V-EASI-MS), a specific ambient ionization source, is suitable to enable direct analysis of sample solutions in real time and is appropriate to be coupled to liquid chromatography (LC). The development of an on-line solid phase extraction system coupled to V-EASI-MS optimizes the advantages of LC-MS hyphenation by enhancing the figures of merit of the analytical method according to AOAC guidelines and simultaneously minimizing the runtime analysis to 1.5 min per sample, as well as sample preparation steps and solvent consumption, which is currently a challenge for quantitative applications of ambient ionization MS.

Liquid chromatography coupled to Venturi easy ambient sonic spray ionization mass spectrometry

Venturi easy ambient sonic spray ionization (V-EASI) is a soft ambient ionization (AI) source that has the advantages of being suitable to the analysis of samples in solution (differently from the majority of AI sources), performing self-pumping, voltage- and heat-free ionization, and requiring minimum or no sample preparation. Since this ionization technique has not been fully explored, the present study provides a proof of principle of the coupling of liquid chromatography to mass spectrometry (LC-MS) using V-EASI as the interface. In order to test the performance of the developed LC-V-EASI-MS system, a quantification method for bixin, a natural dye from annatto (Bixa Orellana L.), which is known to be sensitive to the high voltage applied for electrospray ionization mass spectrometry (ESI-MS) analysis, was validated according to FDA criteria and tested in real samples. The analytical method was successfully applied and met the validation criteria, providing a detectability 10 times better than methods already reported to the quantification of bixin and no matrix effect was observed. Therefore, this proof of principle contributes to the continuous development of AI sources that represents the last great technological advance in MS towards becoming a miniaturized technique able to analyze samples closer to their actual state.

Monitoring Enzymatic Reactions in Real Time Using Venturi Easy Ambient Sonic-Spray Ionization Mass Spectrometry.

We developed a technique to monitor spatially confined surface reactions with mass spectrometry under ambient conditions, without the need for voltage or organic solvents. Fused-silica capillaries immersed in an aqueous solution, positioned in close proximity to each other and the functionalized surface, created a laminar flow junction with a resulting reaction volume of ∼5 pL. The setup was operated with a syringe pump, delivering reagents to the surface through a fused-silica capillary. The other fused-silica capillary was connected to a Venturi easy ambient sonic-spray ionization source, sampling the resulting analytes at a slightly higher flow rate compared to the feeding capillary. The combined effects of the inflow and outflow maintains a chemical microenvironment, where the rate of advective transport overcomes diffusion. We show proof-of-concept where acetylcholinesterase was immobilized on an organosiloxane polymer through electrostatic interactions. The hydrolysis of acetylcholine by acetylcholinesterase into choline was monitored in real-time for a range of acetylcholine concentrations, fused-silica capillary geometries, and operating flow rates. Higher reaction rates and conversion yields were observed with increasing acetylcholine concentrations, as would be expected.

A simpler sampling interface of venturi easy ambient sonic-spray ionization mass spectrometry for high-throughput screening enzyme inhibitors

High-throughput screening (HTS) is often required in enzyme inhibitor drugs screening. Mass spectrometry (MS) provides a powerful method for high-throughput screening enzyme inhibitors because its high speed, sensitivity and property of lable free. However, most of the MS methods need complicated sampling interface system. Overall throughput was limited by sample loading in these cases. In this study, we develop a simple interface which coupled droplet segmented system to a venturi easy ambient sonic-spray ionization mass spectrometer. It is fabricated by using a single capillary to act as both sampling probe and the emitter, which simplifies the construction, reduces the cost and shorten the sampling time. Samples sucked by venturi effect are segmented to nanoliter plugs by air, then the plugs can be detected by MS directly. This system eliminated the need for flow injection which was popular used in classic scheme. The new system is applied to screen angiotensin converting enzyme inhibitors. High-throughput was achieved in analyzing 96 samples at 1.6 s per sample. The plugs formation was at 0.5s per sample. Carry-over between samples was less than 5%, the peak height RSD was 2.92% (n = 15). Dose-response curves of 3 known inhibitors were also measured to validate its potential in drug discovery. The calculated IC50 agreed well with reported values.

Monitoring binding affinity between drug and α1-acid glycoprotein in real time by Venturi easy ambient sonic-spray ionization mass spectrometry

A new approach for monitoring the binding affinity between drugs and alpha 1-acid glycoprotein in real time was developed based on a combination of drug–protein reaction followed by Venturi easy ambient sonic-spray ionization mass spectrometry determination of the free drug concentrations. A known basic drug, propranolol was used to validate the new built method. Binding constant values calculated by venturi easy ambient sonic-spray ionization mass spectrometry was in good accordance with a traditional ultrafiltration combined with high performance liquid chromatography method. Then six types of basic drugs were used as the samples to conduct the real time analysis. Upon injection of alpha 1-acid glycoprotein to the drug mixture, the ion chromatograms were extracted to show the changes in the free drug concentrations in real time. By observing the drop-out of six types of drugs during the whole binding reaction, the binding affinities of different drugs were distinguished. A volume shift validating experiment and an injection delay correcting experiment were also performed to eliminate extraneous factors and verify the reliability of our experiment. Therefore, the features of Venturi easy ambient sonic-spray ionization mass spectrometry (V-EASI-MS) and the experimental results indicate that our technique is likely to become a powerful tool for monitoring drug–AGP binding affinity in real time.

A compact 3D-printed interface for coupling open digital microchips with Venturi easy ambient sonic-spray ionization mass spectrometry.

Digital microfluidics (DMF) based on the electrowetting-on-dielectric phenomenon is a convenient way of handling microlitre-volume aliquots of solutions prior to analysis. Although it was shown to be compatible with on-line mass spectrometric detection, due to numerous technical obstacles, the implementation of DMF in conjunction with MS is still beyond the reach of many analytical laboratories. Here we present a facile method for coupling open DMF microchips to mass spectrometers using Venturi easy ambient sonic-spray ionization operated at atmospheric pressure. The proposed interface comprises a 3D-printed body that can easily be "clipped" at the inlet of a standard mass spectrometer. The accessory features all the necessary connections for an open-architecture DMF microchip with T-shaped electrode arrangement, thermostatting of the microchip, purification of air (to prevent accidental contamination of the microchip), a Venturi pump, and two microfluidic pumps to facilitate transfer of samples and reagents onto the microchip. The system also incorporates a touch-screen panel and remote control for user-friendly operation. It is based on the use of popular open-source electronic modules, and can readily be assembled at low expense.

Use of 3-nitrobenzonitrile as an additive for improved sensitivity in sonic-spray ionization mass spectrometry.

Sonic-spray ionization (SSI) has been shown to produce gas-phase ions for a wide range of compounds, without the application of voltage or a laser. However, it remains to be shown that it can also provide similar sensitivities to those obtained by electrospray ionization mass spectrometry (ESI-MS). Here we report on an attempt to further improve the sensitivity of SSI-MS, more specifically a version of SSI that is referred to as Venturi easy ambient sonic-spray ionization (V-EASI) MS, by adding a signal-enhancing additive to the sample solution. The additive used is 3-nitrobenzonitrile (3-NBN), which has recently been used with success in a new ionization approach named matrix-assisted ionization vacuum. In order to conduct this study we have analyzed a range of compounds, including peptides, metalloproteins, and some organometalloids. During the V-EASI-MS analyses molecular ion and protonated molecule signal intensities as well as their corresponding signal-to-noise (S/N) ratios, obtained in the presence and absence of the 3-NBN, were compared. The 3-NBN-assisted V-EASI-MS approach developed here provides significant improvement in sensitivity relative to conventional V-EASI-MS for almost all compounds tested. More specifically, for peptides a 1.6- to 4-fold enhancement was realized, for proteins the enhancements were from 2- to 5-fold, and for some metalloid species enhancements reached up to 10-fold. However, optimum additive concentration and ion transfer capillary temperature were found to be compound-dependent and thus require optimization in order for maximum enhancements to be achieved. In most cases the 3-NBN-assisted V-EASI-MS approach provides comparable sensitivities and S/N ratios to ESI-MS on the same ion trap mass spectrometer. The use of 3-NBN with V-EASI-MS gives rise to a novel 3-NBN-assisted MS technique, which has demonstrated considerable signal enhancement for most of the compounds analyzed, thus improving its competitiveness towards the well-established and dominating ESI-MS technique.

Real-time analysis of self-assembled nucleobases by Venturi easy ambient sonic-spray ionization mass spectrometry

In this study, the real-time analysis of self-assembled nucleobases was employed by Venturi easy ambient sonic-spray ionization mass spectrometry (V-EASI-MS). With the analysis of three nucleobases including 6-methyluracil (6MU), uracil (U) and thymine (T) as examples, different orders of clusters centered with different metal ions were recorded in both positive and negative modes. Compared with the results obtained by traditional electrospray ionization mass spectrometry (ESI-MS) under the same condition, more clusters with high orders, such as [6MU7+Na]+, [6MU15+2NH4]2+, [6MU10+Na]+, [T7+Na]+, and [T15+2NH4]2+ were detected by V-EASI-MS, which demonstrated the soft ionization ability of V-EASI for studying the non-covalent interaction in a self-assembly process. Furthermore, with the injection of K+ to the system by a syringe pumping, the real-time monitoring of the formation of nucleobases clusters was achieved by the direct extraction of samples from the system under the Venturi effect. Therefore, the effect of cations on the formation of clusters during self-assembly of nucleobases was demonstrated, which was in accordance with the reports. Free of high voltage, heating or radiation during the ionization, this technique is much soft and suitable for obtaining the real-time information of the self-assembly system, which also makes it quite convenient for extraction samples from the reaction system. This “easy and soft” ionization technique has provided a potential pathway for monitoring and controlling the self-assembly processes.

Venturi-Electrosonic Spray Ionization Cataluminescence Sensor Array for Saccharides Detection

In this article, a Venturi electrosonic spray ionization (V-ESSI) cataluminescence (CTL) sensor array was reported for discriminating saccharides in solution. Integrating electrosonic spray ionization (ESSI), a liquid system of Venturi self-pumping injection for the CTL reaction, was fabricated for enhancing CTL reactivity of aqueous samples. Comparing with simple Venturi injection by air and Venturi easy ambient sonic-spray ionization without electric assistance (V-EASI), the remarkable enhancement of CTL signals resulted from V-ESSI. This system showed higher cross-reactive CTL responses catalyzed by alkaline earth metal-nanomaterials than other catalysts, giving different signals for a given saccharide on different catalysts and different responses for different saccharides on the same catalyst. Then, a 4 × 2 CTL sensor array was used for obtaining "fingerprints" of distinct CTL response patterns. Analyzed by linear discriminant analysis (LDA), this V-ESSI CTL sensor array not only achieved the well discrimination of different saccharides (99.9% of total variation) but also discriminated four groups of urine sugar-level for urine samples from diabetic patients (98.1% of discrimination accuracy). It had good reproducibility and gave a linear range of 22.5-67558 μg/mL (R > 0.99) for xylose with a detection limit of 7.4 μg/mL on MgO. As a new artificial tongue, this system provided a simple, rapid, low cost, low energy consumption, and environmentally friendly pathway for aqueous sample discrimination. It has dramatically expanded applications of the CTL-based senor array and will be applicable to clinical diagnoses, environment monitoring, industrial controls, food industry, and various marine monitoring.

Bipolar Mass Spectrometry of Labile Coordination Complexes, Redox Active Inorganic Compounds, and Proteins Using a Glass Nebulizer for Sonic-Spray Ionization

In this study, we report on the development of a novel nebulizer configuration for sonic-spray ionization (SSI) mass spectrometry (MS), more specifically for a version of SSI that is referred to as Venturi easy ambient sonic-spray ionization (V-EASI) MS. The developed nebulizer configuration is based on a commercially available pneumatic glass nebulizer that has been used extensively for aerosol formation in atomic spectrometry. In the present study, the nebulizer was modified in order to achieve efficient V-EASI-MS operation. Upon evaluating this system, it has been demonstrated that V-EASI-MS offers some distinct advantages for the analysis of coordination compounds and redox active inorganic compounds over the predominantly used electrospray ionization (ESI) technique. Such advantages, for this type of compounds, are demonstrated here for the first time. More specifically, a series of labile heptanuclear heterometallic [Cu(II) 6Ln(III)] clusters held together with artificial amino acid ligands, in addition to easily oxidized inorganic oxyanions of selenium and arsenic, were analyzed. The observed advantages pertain to V-EASI appearing to be a "milder" ionization source than ESI, not requiring electrical potentials for gas phase ion formation, thus eliminating the possibility of unwanted redox transformations, allowing for the "simultaneous" detection of negative and positive ions (bipolar analysis) without the need to change source ionization conditions, and also not requiring the use of syringes and delivery pumps. Because of such features, especially because of the absence of ionization potentials, EASI can be operated with minimal requirements for source parameter optimization. We observed that source temperature and accelerating voltage do not seem to affect labile compounds to the extent they do in ESI-MS. In addition, bipolar analysis of proteins was demonstrated here by acquiring both positive and negative ion mass spectra from the same protein solutions, without the need to independently adjust solution and source conditions in each mode. Finally, the simple and efficient operation of a dual-nebulizer configuration was demonstrated for V-EASI-MS for the first time.

Blends of Soybean Biodiesel with Petrodiesel: Direct Quantitation via Mass Spectrometry

Quantitation and identification of blends of soybean biodiesel with petrodiesel were performed via mass spectrometry using two ionization techniques: electrospray ionization (ESI) and Venturi easy ambient sonic-spray ionization in its liquid mode (VL-EASI). Different soybean biodiesel/petrodiesel blends (from B0 to B100) were diluted and then directly infused and analyzed by both techniques. To investigate adulteration of Bn blends, different soybean oil/biodiesel and soybean oil/petrodiesel blends were analyzed. Analytical curves were obtained in three replicates. The two techniques were shown to provide reasonably accurate quantitation in the B1-B20 range. These techniques were also successfully used to detect contamination or adulteration of Bn blends with vegetable oils. ESI is a widely used and commercially available technique whereas a VL-EASI source can be easily mounted using common laboratory parts requiring no use of high voltages. Both techniques require no pre-separation or derivatization steps and offer, therefore, simple and fast methods for the quantitation of Bn blends. The comprehensive snapshots of the molecular composition also allow quality control and typification of the biodiesel and eventually of the vegetable oils in illegal admixtures.

Gasoline, Kerosene, and Diesel Fingerprinting via Polar Markers

Venturi easy ambient sonic-spray ionization mass spectrometry working in the liquid mode (VL-EASI-MS) is shown to provide rapid and reliable characterization of crude oil distillates. With no extraction or pretreatments, samples of gasoline, kerosene, diesel, and admixtures of gasoline/diesel and gasoline/kerosene were directly analyzed by VL-EASI(+)-MS. Homologous series of natural N-heteroaromatics were detected in their protonated forms providing typical profiles of polar markers. For gasoline, VL-EASI(+)-MS detects a homologous series of mainly C1–C5 alkyl pyridines. For kerosene, a typical series of alkylated tetrahydroquinolines is detected. For diesel, the V-EASI(+)-MS profile is much richer because of the detection of several classes of many N-heteroaromatics. VL-EASI(+)-MS is also shown to provide typical spectra for petrochemical gasolines detecting specific antioxidants. Admixtures of gasoline/kerosene and gasoline/diesel are also clearly characterized with limits of detection of 10 and 1% (v/v), respectively. Fourier transform-ion cyclotron resonance mass spectrometry (FT-ICR MS) has been used to confirm assignments of the polar markers detected by VL-EASI(+)-MS.

Easy dual-mode ambient mass spectrometry with Venturi self-pumping, canned air, disposable parts and voltage-free sonic-spray ionization

An exceptionally easy to assemble source for ambient mass spectrometry is described. Based on Venturi easy ambient sonic-spray ionization (V-EASI), the source was further simplified by the use of a can of compressed air which simultaneously provides solution or solvent Venturi self-pumping and continuous, stable and abundant low-noise ion signal via voltage-free sonic-spraying. Further simplification was also attained by the use of inexpensive and readily commercially available parts: a surgical 2-way catheter, an aerosol can of compressed air, a 30 cm long fused-silica capillary and a hypodermic needle. This "Spartan" V-EASI source seems to offer one of the easiest and cheapest ways to make ions for ambient desorption/ionization mass spectrometry analysis of both liquid and solid samples.

Real Time Analysis of Brain Tissue by Direct Combination of Ultrasonic Surgical Aspiration and Sonic Spray Mass Spectrometry

Direct combination of cavitron ultrasonic surgical aspirator (CUSA) and sonic spray ionization mass spectrometry is presented. A commercially available ultrasonic surgical device was coupled to a Venturi easy ambient sonic-spray ionization (V-EASI) source by directly introducing liquified tissue debris into the Venturi air jet pump. The Venturi air jet pump was found to efficiently nebulize the suspended tissue material for gas phase ion production. The ionization mechanism involving solely pneumatic spraying was associated with that of sonic spray ionization. Positive and negative ionization spectra were obtained from brain and liver samples reflecting the primary application areas of the surgical device. Mass spectra were found to feature predominantly complex lipid-type constituents of tissues in both ion polarity modes. Multiply charged peptide anions were also detected. The influence of instrumental settings was characterized in detail. Venturi pump geometry and flow parameters were found to be critically important in ionization efficiency. Standard solutions of phospholipids and peptides were analyzed in order to test the dynamic range, sensitivity, and suppression effects. The spectra of the intact tissue specimens were found to be highly specific to the histological tissue type. The principal component analysis (PCA) and linear discriminant analysis (LDA) based data analysis method was developed for real-time tissue identification in a surgical environment. The method has been successfully tested on post-mortem and ex vivo human samples including astrocytomas, meningeomas, metastatic brain tumors, and healthy brain tissue.

Venturi Easy Ambient Sonic-Spray Ionization

The development and illustrative applications of an ambient ionization technique termed Venturi easy ambient sonic-spray ionization (V-EASI) is described. Its dual mode of operation with Venturi self-pumping makes V-EASI applicable to the direct mass spectrometric analysis of both liquid (V(L)-EASI) and solid (V(S)-EASI) samples. V-EASI is simple and easy to assemble, operating solely via the assistance of a sonic stream of nitrogen or air. The sonic gas stream causes two beneficial and integrated effects: (a) the self-pumping of solutions via the Venturi effect and (b) sonic-spray ionization (SSI) of analytes either in solution or resting on solid surfaces. In its liquid mode, V(L)-EASI is applicable to analytes in solution, forming negatively and/or positively charged intact molecular species in a soft fashion with little or no fragmentation. In its solid mode, V(S)-EASI relies on Venturi self-pumping of a proper SSI solvent solution in combination with SSI to form a stream of bipolar charged droplets that bombard the sample surface, causing desorption and ionization of the analyte molecules. As for its precursor technique (EASI), V-EASI generates bipolar droplets with considerably lower average charging, which increases selectivity for ionization with high signal-to-noise ratios and clean spectra dominated by single molecular species with minimal solvent ions. V-EASI also operates in a voltage-, heat-, and radiation-free fashion and is therefore free of thermal, electrical, or discharge interferences.
