Phonological and Semantic Specialization in 9- to 10-Year-Old Children During Auditory Word Processing.

One of the core features of brain maturation is functional specialization. Previous research has found that 7- to 8-year-old children start to specialize in both the temporal and frontal lobes. However, as children continue to develop their phonological and semantic skills rapidly until approximately 10 years old, it remained unclear whether any changes in specialization later in childhood would be detected. Thus, the goal of the current study was to examine phonological and semantic specialization in 9- to 10-year-old children during auditory word processing. Sixty-one children were included in the analysis. They were asked to perform a sound judgment task and a meaning judgment task, each with both hard and easy conditions to examine parametric effects. Consistent with previous results from 7- to 8-year-old children, direct task comparisons revealed language specialization in both the temporal and frontal lobes in 9- to 10-year-old children. Specifically, the left dorsal inferior frontal gyrus showed greater activation for the sound than the meaning task whereas the left middle temporal gyrus showed greater activation for the meaning than the sound task. Interestingly, in contrast to the previously reported finding that 7- to 8-year-old children primarily engage a general control region during the harder condition for both tasks, we showed that 9- to 10-year-old children recruited language-specific regions to process the more difficult task conditions. Specifically, the left superior temporal gyrus showed greater activation for the phonological parametric manipulation whereas the left ventral inferior frontal gyrus showed greater activation for the semantic parametric manipulation.

Neural pathways of phonological and semantic processing and its relations to children's reading skills.

Behavioral research shows that children’s phonological ability is strongly associated with better word reading skills, whereas semantic knowledge is strongly related to better reading comprehension. However, most neuroscience research has investigated how brain activation during phonological and semantic processing is related to word reading skill. This study examines if connectivity during phonological processing in the dorsal inferior frontal gyrus (dIFG) to posterior superior temporal gyrus (pSTG) pathway is related to word reading skill, whereas connectivity during semantic processing in the ventral inferior frontal gyrus (vIFG) to posterior middle temporal gyrus (pMTG) pathway is related to reading comprehension skill. We used behavioral and functional magnetic resonance imaging (fMRI) data from a publicly accessible dataset on OpenNeuro.org. The research hypotheses and analytical plan were pre-registered on the Open Science Framework. Forty-six children ages 8–15 years old were included in the final analyses. Participants completed an in-scanner reading task tapping into phonology (i.e., word rhyming) and semantics (i.e., word meaning) as well as standardized measures of word reading and reading comprehension skill. In a series of registered and exploratory analyses, we correlated connectivity coefficients from generalized psychophysiological interactions (gPPI) with behavioral measures and used z-scores to test the equality of two correlation coefficients. Results from the preregistered and exploratory analyses indicated weak evidence that functional connectivity of dIFG to pSTG during phonological processing is positively correlated with better word reading skill, but no evidence that connectivity in the vIFG-pMTG pathway during semantic processing is related to better reading comprehension skill. Moreover, there was no evidence to support the differentiation between the dorsal pathway’s relation to word reading and the ventral pathway’s relation to reading comprehension skills. Our finding suggesting the importance of phonological processing to word reading is in line with prior behavioral and neurodevelopmental models.

Neuro-Cognitive Differences in Semantic Processing Between Native Speakers and Proficient Learners of Mandarin Chinese.

The present study aimed to investigate the neural mechanism underlying semantic processing in Mandarin Chinese adult learners, focusing on the learners who were Indo-European language speakers with advanced levels of proficiency in Mandarin Chinese. We used functional magnetic resonance imaging technique and a semantic judgment task to test 24 Mandarin Chinese adult learners (L2 group) and 26 Mandarin Chinese adult native speakers (L1 group) as a control group. In the task, participants were asked to indicate whether two-character pairs were related in meaning. Compared to the L1 group, the L2 group had greater activation in the bilateral occipital regions, including the fusiform gyrus and middle occipital gyrus, as well as the right superior parietal lobule. On the other hand, less activation in the bilateral temporal regions was found in the L2 group relative to the L1 group. Correlation analysis further revealed that, within the L2 group, increased activation in the left middle temporal gyrus/superior temporal gyrus (M/STG, BA 21) was correlated with higher accuracy in the semantic judgment task as well as better scores in the two vocabulary tests, the Assessment of Chinese character list for grade 3 to grade 9 (A39) and the Peabody Picture Vocabulary Test-Revised. In addition, functional connectivity analysis showed that connectivity strength between the left fusiform gyrus and left ventral inferior frontal gyrus (IFG, BA 47) was modulated by the accuracy in the semantic judgment task in the L1 group. By contrast, this modulation effect was weaker in the L2 group. Taken together, our study suggests that Mandarin Chinese adult learners rely on greater recruitment of the bilateral occipital regions to process orthographic information to access the meaning of Chinese characters. Also, our correlation results provide convergent evidence that the left M/STG (BA 21) plays a crucial role in the storage of semantic knowledge for readers to access to conceptual information. Moreover, the connectivity results indicate that the left ventral pathway (left fusiform gyrus-left ventral IFG) is associated with orthographic-semantic processing in Mandarin Chinese. However, this semantic-related ventral pathway might require more time and language experience to be developed, especially for the late adult learners of Mandarin Chinese.

Both frontal and temporal cortex exhibit phonological and semantic specialization during spoken language processing in 7- to 8-year-old children.

A previous functional magnetic resonance imaging (fMRI) study by Weiss et al. (Weiss et al., Human Brain Mapping, 2018, 39, 4334–4348) examined brain specialization for phonological and semantic processing of spoken words in young children who were 5 to 6 years old and found evidence for specialization in the temporal but not the frontal lobe. According to a prominent neurocognitive model of language development (Skeide & Friederici, Nature Reviews Neuroscience, 2016, 17, 323–332), the frontal lobe matures later than the temporal lobe. Thus, the current study aimed to examine if brain specialization in the frontal lobe can be observed in a slightly older cohort of children aged 7 to 8 years old using the same experimental and analytical approach as in Weiss et al. (Weiss et al., Human Brain Mapping, 2018, 39, 4334–4348). One hundred and ten typically developing children were recruited and were asked to perform a sound judgment task, tapping into phonological processing, and a meaning judgment task, tapping into semantic processing, while in the MRI scanner. Direct task comparisons showed that these children exhibited language specialization in both the temporal and the frontal lobes, with the left posterior dorsal inferior frontal gyrus (IFG) showing greater activation for the sound than the meaning judgment task, and the left anterior ventral IFG and the left posterior middle temporal gyrus (MTG) showing greater activation for the meaning than the sound judgment task. These findings demonstrate that 7‐ to 8‐year‐old children have already begun to develop a language‐related specialization in the frontal lobe, suggesting that early elementary schoolers rely on both specialized linguistic manipulation and representation mechanisms to perform language tasks.

Enhanced activations in syntax-related regions for multilinguals while acquiring a new language

The neuroscientific foundation of multilingualism, a unique cognitive capacity, necessitates further elucidation. We conducted an fMRI experiment to evaluate the acquisition of syntactic features in a new language (Kazakh) for multilinguals and bilinguals. Results showed that the multilinguals who were more proficient in their second/third languages needed fewer task trials to acquire Kazakh phonology. Regarding group differences, the reduction in response times during the initial exposure to Kazakh were significantly larger for the multilinguals than the bilinguals. For the multilinguals, activations in the bilateral frontal/temporal regions were maintained at a higher level than the initial level during subsequent new grammar conditions. For the bilinguals, activations in the basal ganglia/thalamus and cerebellum decreased to the initial level each time. Direct group comparisons showed significantly enhanced activations for the multilinguals in the left ventral inferior frontal gyrus. These results indicate that both syntax-related and domain-general brain networks were more enhanced for the multilinguals. We also unexpectedly observed significant activations in the visual areas for the multilinguals, implying the use of visual representation even when listening to speech sounds alone. Because the multilinguals were able to successfully utilize acquired knowledge in an accumulated manner, the results support the cumulative-enhancement model of language acquisition.

Flexible reconfiguration of functional brain networks as a potential neural mechanism of creativity

Creativity relies on the reorganizing of multimodal information and flexible switching between different modes of thinking, suggesting an association between creativity and the reconfiguration of functional brain networks. Here, we investigated global and regional brain dynamics in high-creative (HCG, N= 22) and a low-creative (LCG, N= 20) groups during a divergent creative thinking task. We found that during the creative thinking task, the HCG demonstrated higher global network flexibility, as compared to the LCG. In addition, creative thinking in the HCG was associated with significantly higher regional flexibility in the medial superior temporal gyrus, superior parietal lobule, precuneus, nucleus accumbens, and the ventral inferior frontal gyrus. Interestingly, the LCG demonstrated decreased regional flexibility in the medial superior temporal gyrus, superior parietal lobule, and the ventral inferior frontal gyrus. We also found that the changes in global and regional flexibility in the creative compared with the control tasks were good features allowing for distinguishing between the HCG and the LCG. Taken together, these findings provide evidence that divergent creative thinking is associated with flexible reconfiguration of brain networks involved in verbal, working memory, and reward processing.

Developmental Differences of Structural Connectivity and Effective Connectivity in Semantic Judgments of Chinese Characters.

Previous studies have investigated the developmental differences of semantic processing regarding brain activation between adults and children. However, little is known about whether the patterns of structural connectivity and effective connectivity differ between adults and children during semantic processing. Functional magnetic resonance imaging (fMRI), diffusion spectrum imaging (DSI), and dynamic causal modeling (DCM) were used to study the developmental differences of brain activation, structural connectivity, and effective connectivity during semantic judgments. Twenty-six children (8- to 12-year-olds) and 26 adults were asked to indicate if character pairs were related in meaning. Compared to children, adults showed greater activation in the left ventral inferior frontal gyrus (IFG) and left middle temporal gyrus (MTG). Also, adults had significantly greater structural connectivity in the left ventral pathway (inferior frontal occipital fasciculus, IFOF) than children. Moreover, adults showed significantly stronger bottom-up effects from left fusiform gyrus (FG) to ventral IFG than children in the related condition. In conclusion, our findings suggest that age-related increases in brain activation (ventral IFG and MTG), IFOF, and effective connectivity (from FG to ventral IFG) might be associated with the bottom-up influence of orthographic representations on retrieving semantic representations for processing Chinese characters.

Functional Dissociations of the Left Anterior and Posterior Occipitotemporal Cortex for Semantic and Non-semantic Phonological Access

Previous studies have identified the ventral and dorsal brain regions that respectively support semantic and non-semantic phonological access. Nevertheless, the specific role of the left occipitotemporal cortex (lOTC) in the two pathways of phonological access is ambiguous. To address that question, the present study compared word reading in Chinese (presumably relying on the semantic pathway) with that in English (presumably relying on the non-semantic pathway). Results revealed a clear dissociation in the involvement of the anterior and posterior lOTC in semantic and non-semantic phonological access. Specifically, the anterior lOTC showed greater activation for Chinese than for English, whereas the posterior lOTC showed greater activation for English than for Chinese. More importantly, both psychophysiological interaction analysis and resting-state functional connectivity analysis showed that the anterior lOTC was functionally connected to the ventral brain regions (e.g., left anterior fusiform gyrus, anterior temporal lobe, and ventral inferior frontal gyrus), whereas the posterior lOTC was functionally connected to the dorsal brain regions (e.g., left posterior superior temporal gyrus, supramarginal gyrus, and dorsal inferior frontal gyrus). These results suggest that the anterior and posterior lOTC are involved in semantic and non-semantic phonological access, respectively.

Automatic semantic influence on early visual word recognition in the ventral occipito-temporal cortex

The left ventral occipitotemporal cortex (vOT) is a critical region in reading. According to the interactive account of reading, the vOT is an interface between the lower-level visual regions and higher-level language areas. One prediction of the interactive account is that orthographic activation in vOT should be automatically influenced by semantics and phonology. In the current study, we used functional magnetic resonance imaging (fMRI) and a masked priming paradigm with a relatively short duration (150 ms) to examine whether language information automatically influences vOT during Chinese reading. Participants were asked to perform a lexical decision task on target characters. We separately tested the phonological and semantic influence on orthographic processing in vOT. Brain activation analyses showed that the activation of vOT was modulated by semantic information. In addition, a functional connectivity analysis showed stronger connectivity between vOT and the left ventral inferior frontal gyrus was modulated by semantic information. These findings provided converging evidence for the existence of an automatic semantic influence on vOT during reading, supporting the interactive account. Our study did not show a phonological effect either in the activation of or connectivity with vOT. Taken together, these results reflect the unique processes of Chinese reading, which relies more on the mapping between orthography and semantics, as compared to the orthographic to phonology mapping.

Differential cortical contribution of syntax and semantics: An fMRI study on two-word phrasal processing

Linguistic expressions consist of sequences of words combined together to form phrases and sentences. The neurocognitive process handling word combination is drawing increasing attention among the neuroscientific community, given that the underlying syntactic and semantic mechanisms of such basic combinations—although essential to the generation of more complex structures—still need to be consistently determined. The current experiment was conducted to disentangle the neural networks supporting syntactic and semantic processing at the level of two-word combinations. We manipulated the combinatorial load by using words of different grammatical classes within the phrase, such that determiner-noun combinations (this ship) were used to boost neural activity in syntax-related areas, while adjective-noun combinations (blue ship) were conversely used to measure neural response in semantic-related combinations. By means of functional magnetic resonance imaging (fMRI), we found that syntax-related processing mainly activates the most ventral part of the inferior frontal gyrus (IFG), along the frontal operculum (FOP) and anterior insula (aINS). Fine-grained analysis in BA44 confirmed that the most inferior-ventral portion is highly sensitive to syntactic computations driven by function words. Semantic-related processing on the contrary, rather engages the anterior dorsal part of the left IFG and the left angular gyrus (AG) that is two regions which appear to perform different functions within the semantic network. Our findings suggest that syntactic and semantic contribution to phrasal formation can be already differentiated at a very basic level, with each of these two processes comprising non-overlapping areas on the cerebral cortex. Specifically, they confirm the role of the ventral IFG for the construction of syntactically legal linguistic constructions, and the prominence of the more anterior IFG and the AG for conceptual semantics.

Individual variation in intentionality in the mind-wandering state is reflected in the integration of the default-mode, fronto-parietal, and limbic networks

Mind-wandering has a controversial relationship with cognitive control. Existing psychological evidence supports the hypothesis that episodes of mind-wandering reflect a failure to constrain thinking to task-relevant material, as well the apparently alternative view that control can facilitate the expression of self-generated mental content. We assessed whether this apparent contradiction arises because of a failure to consider differences in the types of thoughts that occur during mind-wandering, and in particular, the associated level of intentionality. Using multi-modal magnetic resonance imaging (MRI) analysis, we examined the cortical organisation that underlies inter-individual differences in descriptions of the spontaneous or deliberate nature of mind-wandering. Cortical thickness, as well as functional connectivity analyses, implicated regions relevant to cognitive control and regions of the default-mode network for individuals who reported high rates of deliberate mind-wandering. In contrast, higher reports of spontaneous mind-wandering were associated with cortical thinning in parietal and posterior temporal regions in the left hemisphere (which are important in the control of cognition and attention) as well as heightened connectivity between the intraparietal sulcus and a region that spanned limbic and default-mode regions in the ventral inferior frontal gyrus. Finally, we observed a dissociation in the thickness of the retrosplenial cortex/lingual gyrus, with higher reports of spontaneous mind-wandering being associated with thickening in the left hemisphere, and higher repots of deliberate mind-wandering with thinning in the right hemisphere. These results suggest that the intentionality of the mind-wandering state depends on integration between the control and default-mode networks, with more deliberation being associated with greater integration between these systems. We conclude that one reason why mind-wandering has a controversial relationship with control is because it depends on whether the thoughts emerge in a deliberate or spontaneous fashion.

Predictive Brain Mechanisms in Sound-to-Meaning Mapping during Speech Processing.

In daily communication via spoken language, one of the core processes is understanding the words being used. Effortless and efficient information exchange via speech relies not only on the identification of individual spoken words, but also on the contextual information giving rise to expected meanings. Despite the accumulating evidence for the bottom-up perception of auditory input, it is still not fully understood how the top-down modulation is achieved in the extensive frontotemporal cortical network. Here, we provide a comprehensive description of the neural substrates underlying sound-to-meaning mapping and demonstrate how the dual-stream model functions in the modulation of expectations, allowing for a better understanding of how the human brain accomplishes sound-to-meaning mapping in a predictive manner.

Organization of cortico-cortical pathways supporting memory retrieval across subregions of the left ventrolateral prefrontal cortex.

Functional magnetic resonance imaging (fMRI) evidence indicates that different subregions of ventrolateral prefrontal cortex (VLPFC) participate in distinct cortical networks. These networks have been shown to support separable cognitive functions: anterior VLPFC [inferior frontal gyrus (IFG) pars orbitalis] functionally correlates with a ventral fronto-temporal network associated with top-down influences on memory retrieval, while mid-VLPFC (IFG pars triangularis) functionally correlates with a dorsal fronto-parietal network associated with postretrieval control processes. However, it is not known to what extent subregional differences in network affiliation and function are driven by differences in the organization of underlying white matter pathways. We used high-angular-resolution diffusion spectrum imaging and functional connectivity analysis in unanesthetized humans to address whether the organization of white matter connectivity differs between subregions of VLPFC. Our results demonstrate a ventral-dorsal division within IFG. Ventral IFG as a whole connects broadly to lateral temporal cortex. Although several different individual white matter tracts form connections between ventral IFG and lateral temporal cortex, functional connectivity analysis of fMRI data indicates that these are part of the same ventral functional network. By contrast, across subdivisions, dorsal IFG was connected with the midfrontal gyrus and correlated as a separate dorsal functional network. These qualitative differences in white matter organization within larger macroanatomical subregions of VLPFC support prior functional distinctions among these regions observed in task-based and functional connectivity fMRI studies. These results are consistent with the proposal that anatomical connectivity is a crucial determinant of systems-level functional organization of frontal cortex and the brain in general.

Sentence processing and verbal working memory in a white-matter-disconnection patient

The Arcuate Fasciculus/Superior Longitudinal Fasciculus (AF/SLF) is the white-matter bundle that connects posterior superior temporal and inferior frontal cortex. Its causal functional role in sentence processing and verbal working memory is currently under debate. While impairments of sentence processing and verbal working memory often co-occur in patients suffering from AF/SLF damage, it is unclear whether these impairments result from shared white-matter damage to the verbal-working-memory network. The present study sought to specify the behavioral consequences of focal AF/SLF damage for sentence processing and verbal working memory, which were assessed in a single patient suffering from a cleft-like lesion spanning the deep left superior temporal gyrus, sparing most surrounding gray matter. While tractography suggests that the ventral fronto-temporal white-matter bundle is intact in this patient, the AF/SLF was not visible to tractography. In line with the hypothesis that the AF/SLF is causally involved in sentence processing, the patient׳s performance was selectively impaired on sentences that jointly involve both complex word orders and long word-storage intervals. However, the patient was unimpaired on sentences that only involved long word-storage intervals without involving complex word orders. On the contrary, the patient performed generally worse than a control group across standard verbal-working-memory tests. We conclude that the AF/SLF not only plays a causal role in sentence processing, linking regions of the left dorsal inferior frontal gyrus to the temporo-parietal region, but moreover plays a crucial role in verbal working memory, linking regions of the left ventral inferior frontal gyrus to the left temporo-parietal region. Together, the specific sentence-processing impairment and the more general verbal-working-memory impairment may imply that the AF/SLF subserves both sentence processing and verbal working memory, possibly pointing to the AF and SLF respectively supporting each.

Superior Temporal Sulcus Disconnectivity During Processing of Metaphoric Gestures in Schizophrenia

The left superior temporal sulcus (STS) plays an important role in integrating audiovisual information and is functionally connected to disparate regions of the brain. For the integration of gesture information in an abstract sentence context (metaphoric gestures), intact connectivity between the left STS and the inferior frontal gyrus (IFG) should be important. Patients with schizophrenia have problems with the processing of metaphors (concretism) and show aberrant structural connectivity of long fiber bundles. Thus, we tested the hypothesis that patients with schizophrenia differ in the functional connectivity of the left STS to the IFG for the processing of metaphoric gestures. During functional magnetic resonance imaging data acquisition, 16 patients with schizophrenia (P) and a healthy control group (C) were shown videos of an actor performing gestures in a concrete (iconic, IC) and abstract (metaphoric, MP) sentence context. A psychophysiological interaction analysis based on the seed region from a previous analysis in the left STS was performed. In both groups we found common positive connectivity for IC and MP of the STS seed region to the left middle temporal gyrus (MTG) and left ventral IFG. The interaction of group (C>P) and gesture condition (MP>IC) revealed effects in the connectivity to the bilateral IFG and the left MTG with patients exhibiting lower connectivity for the MP condition. In schizophrenia the left STS is misconnected to the IFG, particularly during the processing of MP gestures. Dysfunctional integration of gestures in an abstract sentence context might be the basis of certain interpersonal communication problems in the patients.

Functional and anatomical correlates of word-, sentence-, and discourse-level integration in sign language

In both vocal and sign languages, we can distinguish word-, sentence-, and discourse-level integration in terms of hierarchical processes, which integrate various elements into another higher level of constructs. In the present study, we used magnetic resonance imaging and voxel-based morphometry (VBM) to test three language tasks in Japanese Sign Language (JSL): word-level (Word), sentence-level (Sent), and discourse-level (Disc) decision tasks. We analyzed cortical activity and gray matter (GM) volumes of Deaf signers, and clarified three major points. First, we found that the activated regions in the frontal language areas gradually expanded in the dorso-ventral axis, corresponding to a difference in linguistic units for the three tasks. Moreover, the activations in each region of the frontal language areas were incrementally modulated with the level of linguistic integration. These dual mechanisms of the frontal language areas may reflect a basic organization principle of hierarchically integrating linguistic information. Secondly, activations in the lateral premotor cortex and inferior frontal gyrus were left-lateralized. Direct comparisons among the language tasks exhibited more focal activation in these regions, suggesting their functional localization. Thirdly, we found significantly positive correlations between individual task performances and GM volumes in localized regions, even when the ages of acquisition (AOAs) of JSL and Japanese were factored out. More specifically, correlations with the performances of the Word and Sent tasks were found in the left precentral/postcentral gyrus and insula, respectively, while correlations with those of the Disc task were found in the left ventral inferior frontal gyrus and precuneus. The unification of functional and anatomical studies would thus be fruitful for understanding human language systems from the aspects of both universality and individuality.

Optimally Efficient Neural Systems for Processing Spoken Language

Cognitive models claim that spoken words are recognized by an optimally efficient sequential analysis process. Evidence for this is the finding that nonwords are recognized as soon as they deviate from all real words ( Marslen-Wilson 1984), reflecting continuous evaluation of speech inputs against lexical representations. Here, we investigate the brain mechanisms supporting this core aspect of word recognition and examine the processes of competition and selection among multiple word candidates. Based on new behavioral support for optimal efficiency in lexical access from speech, a functional magnetic resonance imaging study showed that words with later nonword points generated increased activation in the left superior and middle temporal gyrus (Brodmann area [BA] 21/22), implicating these regions in dynamic sound-meaning mapping. We investigated competition and selection by manipulating the number of initially activated word candidates (competition) and their later drop-out rate (selection). Increased lexical competition enhanced activity in bilateral ventral inferior frontal gyrus (BA 47/45), while increased lexical selection demands activated bilateral dorsal inferior frontal gyrus (BA 44/45). These findings indicate functional differentiation of the fronto-temporal systems for processing spoken language, with left middle temporal gyrus (MTG) and superior temporal gyrus (STG) involved in mapping sounds to meaning, bilateral ventral inferior frontal gyrus (IFG) engaged in less constrained early competition processing, and bilateral dorsal IFG engaged in later, more fine-grained selection processes.

Same Modulation but Different Starting Points: Performance Modulates Age Differences in Inferior Frontal Cortex Activity during Word-Retrieval

The neural basis of word-retrieval deficits in normal aging has rarely been assessed and the few previous functional imaging studies found enhanced activity in right prefrontal areas in healthy older compared to younger adults. However, more pronounced right prefrontal recruitment has primarily been observed during challenging task conditions. Moreover, increased task difficulty may result in enhanced activity in the ventral inferior frontal gyrus (vIFG) bilaterally in younger participants as well. Thus, the question arises whether increased activity in older participants represents an age-related phenomenon or reflects task difficulty effects. In the present study, we manipulated task difficulty during overt semantic and phonemic word-generation and used functional magnetic resonance imaging to assess activity patterns in the vIFG in healthy younger and older adults (N = 16/group; mean age: 24 vs. 69 years). Both groups produced fewer correct responses during the more difficult task conditions. Overall, older participants produced fewer correct responses and showed more pronounced task-related activity in the right vIFG. However, increased activity during the more difficult conditions was found in both groups. Absolute degree of activity was correlated with performance across groups, tasks and difficulty levels. Activity modulation (difficult vs. easy conditions) was correlated with the respective drop in performance across groups and tasks. In conclusion, vIFG activity levels and modulation of activity were mediated by performance accuracy in a similar way in both groups. Group differences in the right vIFG activity were explained by performance accuracy which needs to be considered in future functional imaging studies of healthy and pathological aging.

Electrical Brain Stimulation Improves Cognitive Performance by Modulating Functional Connectivity and Task-Specific Activation

Excitatory anodal transcranial direct current stimulation (atDCS) can improve human cognitive functions, but neural underpinnings of its mode of action remain elusive. In a cross-over placebo ("sham") controlled study we used functional magnetic resonance imaging (fMRI) to investigate neurofunctional correlates of improved language functions induced by atDCS over a core language area, the left inferior frontal gyrus (IFG). Intrascanner transcranial direct current stimulation-induced changes in overt semantic word generation assessed behavioral modulation; task-related and task-independent (resting-state) fMRI characterized language network changes. Improved word-retrieval during atDCS was paralleled by selectively reduced task-related activation in the left ventral IFG, an area specifically implicated in semantic retrieval processes. Under atDCS, resting-state fMRI revealed increased connectivity of the left IFG and additional major hubs overlapping with the language network. In conclusion, atDCS modulates endogenous low-frequency oscillations in a distributed set of functionally connected brain areas, possibly inducing more efficient processing in critical task-relevant areas and improved behavioral performance.

Stochastic tractography study of Inferior Frontal Gyrus anatomical connectivity in schizophrenia

BackgroundAbnormalities within language-related anatomical structures have been associated with clinical symptoms and with language and memory deficits in schizophrenia. Recent studies suggest disruptions in functional connectivity within the Inferior Frontal Gyrus (IFG) network in schizophrenia. However, due to technical challenges, anatomical connectivity abnormalities within this network and their involvement in clinical and cognitive deficits have not been studied. Material and methodsDiffusion and anatomical scans were obtained from 23 chronic schizophrenia patients and 23 matched controls. The IFG was automatically segmented, and its white matter connections extracted and measured with newly-developed stochastic tractography tools. Correlations between anatomical structures and measures of semantic processing were also performed. ResultsWhite Matter connections between the IFG and posterior brain regions followed two distinct pathways: dorsal and ventral. Both demonstrated left lateralization, but ventral pathway abnormalities were only found in schizophrenia. IFG volumes also showed left lateralization and abnormalities in schizophrenia. Further, despite similar laterality and abnormality patterns, IFG volumes and white matter connectivity were not correlated with each other in either group. Interestingly, measures of semantic processing correlated with white matter connectivity in schizophrenia and with gray matter volumes in controls. Finally, hallucinations were best predicted by both gray matter and white matter measures together. ConclusionsOur results suggest abnormalities within the ventral IFG network in schizophrenia, with white matter abnormalities better predicting semantic deficits. The lack of a statistical relationship between coexisting gray and white matter deficits might suggest their different origin and the necessity for a multimodal approach in future schizophrenia studies.
