Xylem safety in relation to the stringency of plant water potential regulation of European beech, Norway spruce, and Douglas-fir trees during severe drought

Key messageNorway spruce operates with larger hydraulic safety margins (HSM) than beech and Douglas-fir despite the known drought sensitivity of spruce, questioning a pivotal role of HSM in drought tolerance.The exceptional 2018/2019 drought exposed Central Europe’s forests to severe stress, highlighting the need to better understand stomatal regulation strategies and their relationship to xylem safety under extreme drought. We studied diurnal, seasonal, and inter-annual variation in stomatal conductance (gs) and leaf water potential (ΨLeaf) in co-occurring European beech (F. sylvatica), Norway spruce (P. abies), and Douglas-fir (P. menziesii) trees in the two summers and related them to hydraulic traits characterizing drought resistance. In 2018, F. sylvatica exhibited a continuous ΨLeaf decline from June to September, as is characteristic for an anisohydric strategy, while P. abies closed stomata early and reached the least negative ΨLeaf-values at the end of summer. P. menziesii showed low ΨLeaf-values close to P12 (the xylem pressure at onset of embolism) already in July. Both conifers closed stomata when approaching P12 and maintained low gs-levels throughout summer, indicative for isohydric regulation. In 2019, all three species showed a linear decline in ΨLeaf, but F. sylvatica crossed P12 in contrast to the conifers. The three species exhibited similar water potentials at turgor loss point (− 2.44 to − 2.51 MPa) and branch P50 (xylem pressure at 50% loss of hydraulic conductance; − 3.3 to − 3.8 MPa). Yet, F. sylvatica and P. menziesii operated with smaller hydraulic safety margins (HSM means: 0.79 and 0.77 MPa) than P. abies (1.28 MPa). F. sylvatica reduced leaf size and specific leaf area in 2019 and increased Huber value. Our species comparison during extreme drought contradicts the general assumption that conifers operate with larger HSMs than angiosperm trees. Contrary to expectation, P. abies appeared as hydraulically less vulnerable than Douglas-fir.

Grass veins are leaky pipes: vessel widening in grass leaves explain variation in stomatal conductance and vessel diameter among species.

The widening of xylem vessels from tip to base of trees is an adaptation to minimize the hydraulic resistance of a long pathway. Given that parallel veins of monocot leaves do not branch hierarchically, vessels should also widen basipetally but, in addition to minimizing resistance, should also account for water volume lost to transpiration since they supply water to the lamina along their lengths, that is 'leakiness'. We measured photosynthesis, stomatal conductance, and vessel diameter at five locations along each leaf of five perennial grass species. We found that the rate of conduit widening in grass leaves was larger than the widening exponent required to minimize pathlength resistance (0.35 vs c. 0.22). Furthermore, variation in the widening exponent among species was positively correlated with maximal stomatal conductance (r2 = 0.20) and net CO2 assimilation (r2 = 0.45). These results suggest that faster rates of conduit widening (> 0.22) were associated with higher rates of water loss. Taken together, our results show that the widening exponent is linked to plant function in grass leaves and that natural selection has favored parallel vein networks that are constructed to meet transpiration requirements while minimizing hydraulic resistance within grass blades.

Granal thylakoid structure and function: explaining an enduring mystery of higher plants.

In higher plants, photosystems II and I are found in grana stacks and unstacked stroma lamellae, respectively. To connect them, electron carriers negotiate tortuous multi-media paths and are subject to macromolecular blocking. Why does evolution select an apparently unnecessary, inefficient bipartition? Here we systematically explain this perplexing phenomenon. We propose that grana stacks, acting like bellows in accordions, increase the degree of ultrastructural control on photosynthesis through thylakoid swelling/shrinking induced by osmotic water fluxes. This control coordinates with variations in stomatal conductance and the turgor of guard cells, which act like an accordion's air button. Thylakoid ultrastructural dynamics regulate macromolecular blocking/collision probability, direct diffusional pathlengths, division of function of Cytochrome b6 f complex between linear and cyclic electron transport, luminal pH via osmotic water fluxes, and the separation of pH dynamics between granal and lamellar lumens in response to environmental variations. With the two functionally asymmetrical photosystems located distantly from each other, the ultrastructural control, nonphotochemical quenching, and carbon-reaction feedbacks maximally cooperate to balance electron transport with gas exchange, provide homeostasis in fluctuating light environments, and protect photosystems in drought. Grana stacks represent a dry/high irradiance adaptation of photosynthetic machinery to improve fitness in challenging land environments. Our theory unifies many well-known but seemingly unconnected phenomena of thylakoid structure and function in higher plants.

Stomatal Limitation Is Able to Modulate Leaf Coloration Onset of Temperate Deciduous Tree

Autumn phenology, determined mainly by temperature and photoperiod, is essential for ecosystem carbon sequestration. Usually, the variations in the maximum rate of Rubisco (Vcmax) and the maximum rate of ribulose-bisphosphate regeneration (Jmax) are taken as the mechanism regulating the seasonal pattern of photosynthetic rates and autumn phenology. In this study, we used Quercus mongolicus seedlings as an example to examine the photosynthetically physiological mechanism of leaf coloration onset (LCO) responding to different warming and photoperiod treatments based on experimental data acquired from large artificial climate simulation chambers. The results indicated that: (1) LCO and the net CO2 assimilation rate (Pn), transpiration rate (Tr), stomatal conductance (Gs), Vcmax, and Jmax of Quercus mongolicus seedlings were significantly affected by the changes of photoperiod. (2) LCO was significantly correlated only with the Pn approach, supporting the view that leaf senescence is the result of a trade-off between nutrient resorption and reserves. (3) The major variation in stomatal conductance (Gs) is the mechanism by which photoperiod regulates the seasonal pattern of photosynthetic rates, implying that both limitations of stomatal and photosynthetical capacity (Vcmax and Jmax, non-stomatal limitation) are able to modulate LCO. Our study riches the knowledge of phenology and provides a reference for phenological modelling and ecosystem carbon estimation.

Physiological traits for evaluating heat‐tolerance of Australian spring wheat cultivars at elevated CO2

AbstractHigh temperatures and increasing CO2 concentrations are a major threat to global wheat (Triticum aestivum L.) production, demanding the development of heat‐tolerant wheat cultivars. Plant physiological traits are potential surrogates for evaluating genetic variation for crop stress tolerance. This research evaluated 23 Australian wheat cultivars and two breeding lines for heat‐tolerance by characterising the associated physiological traits. The interactive effects of heat‐stress and elevated CO2 were studied under controlled greenhouse conditions and delayed sowing in the field with exposure to higher temperature. Physiological changes, pollen viability and grain yield were assessed. A significant correlation was observed between physiological and agronomical traits under high temperature at 35°C and elevated CO2 at 800 µl L−1, providing evidence of adaptation to high temperatures in some genotypes. Variation in stomatal conductance, transpiration rate, canopy temperature, leaf chlorophyll and photosynthesis were associated with maintenance of pollen viability and grain yield at high temperatures. Of the genotypes assessed, seven were classified as heat tolerant at both ambient and elevated CO2 and nine were heat susceptible while the remainder showed a variable response to CO2 at high temperatures. Field data confirmed the heat responses of the most heat‐tolerant and susceptible genotypes. The heat tolerant genotypes identified are candidates for further breeding and selection to improve adaptation to a changing climate.

Resposta fotossintética de umbuzeiro ao deficit de pressão de vapor

Abstract The objective of this work was to evaluate the photosynthetic response to vapor pressure deficit (VPD) in umbu (Spondias tuberosa) tree accessions. The experiment was carried out in a completely randomized design in a 5×7×2 factorial arrangement (five umbu accessions – BRS-68, EPAMIG-05, BGU-61, BGU-75, and BGU-50 –, seven evaluation times, and two reading times – at 8 a.m. and 2 p.m.) in split-split plots in time. Leaf temperature increased with air temperature. The variation of stomatal conductance and transpiration with the VPD was not significant. Net photosynthesis rate, carboxylation efficiency, and instantaneous water-use efficiency decreased with increasing VPD in all accessions, except in BRS-68, whose rates remained positive.

Dynamic Changes in Plant Resource Use Efficiencies and Their Primary Influence Mechanisms in a Typical Desert Shrub Community

Understanding plant resource use efficiencies (RUEs) and their tradeoffs in a desert shrub community, particularly as it concerns the usage of water, light, and nitrogen, remains an ecological imperative. Plant RUEs have been widely used as indicators to understand plant acclimation processes to unfavorable environmental conditions. This study aimed to examine seasonal dynamics in RUEs in two widely distributed plant species in a typical desert shrub community (i.e., Artemisia ordosica and Leymus secalinus) based on in-situ measurements of leaf photosynthesis, specific leaf area (SLA), leaf nitrogen concentration (i.e., Nmass + Narea), and several site-related abiotic factors. Both species exhibited significant seasonal variation in RUEs, with a coefficient of variation (CV) >30% and seasonal divergence among the various RUEs. Seasonal divergence was largely controlled by variation in stomatal conductance (Gs), which was in turn influenced by variation in soil water content (SWC) and water vapor pressure deficit (VPD). RUEs between species converged, being positively correlated, yielding: (i) r2 = 0.40 and p < 0.01 for WUE; (ii) r2 = 0.18 and p < 0.01 for LUE; and (iii) r2 = 0.25 and p < 0.01 for NUE. RUEs for A. ordosica were mostly larger than those for L. secalinus, but less reactive to drought. This suggests A. ordosica was more conservative in its usage of available resources and was, therefore, better able to adapt to arid conditions. Resource use strategies between species differed in response to drought. Desert shrubs are projected to eventually replace grasses, as drought severity and duration increase with sustained regional climate change.

Isohydricity of Two Different Citrus Species under Deficit Irrigation and Reclaimed Water Conditions.

Citrus species are frequently subjected to water and saline stresses worldwide. We evaluated the effects of diurnal changes in the evaporative demands and soil water contents on the plant physiology of grapefruit and mandarin crops under saline reclaimed (RW) and transfer (TW) water conditions, combined with two irrigation strategies, fully irrigated (fI) and non-irrigated (nI). The physiological responses were different depending on the species. Grapefruit showed an isohydric pattern, which restricted the use of the leaf water potential (Ψl) as a plant water status indicator. Its water status was affected by salinity (RW) and water stress (nI), mainly as the combination of both stresses (RW-nI); however, mandarin turned out to be relatively more tolerant to salinity and more sensitive to water stress, mainly because of its low hydraulic conductance (K) levels, showing a critical drop in Ψl that led to severe losses of root–stem (Kroot–stem) and canopy (Kcanopy) hydraulic conductance in TW-nI. This behavior was not observed in RW-nI because a reduction in canopy volume as an adaptive characteristic was observed; thus, mandarin exhibited more anisohydric behavior compared to grapefruit, but isohydrodynamic since its hydrodynamic water potential gradient from roots to shoots (ΔΨplant) was relatively constant across variations in stomatal conductance (gs) and soil water potential. The gs was considered a good plant water status indicator for irrigation scheduling purposes in both species, and its responses to diurnal VPD rise and soil drought were strongly correlated with Kroot–stem. ABA did not show any effect on stomatal regulation, highlighting the fundamental role of plant hydraulics in driving stomatal closure.

Girdling changes root and shoot hormonal balance but does not alter drought-induced stomatal closure in soybean

To investigate the importance of shoot-to-root communication in modulating tissue phytohormone balance as the soil dries, root and leaf hormonal profiles were determined in soybean plants (Glycine max L. Merr. cv. Siverka) exposed to factorial girdling (achieved surgically on Day 0 by excising the phloem with a sharp razor blade thereby disrupting basipetal hormone transport) and soil drying treatments. Plants were randomized into 2 groups: girdled and intact plants, and each girdled group was further separated into two groups: well-watered and soil drying. Thus the experiment was a two (± girdling) x two (± soil drying) factorial experiment. Within 26 h while the soil was still moist, girdling increased root ACC (by 11-fold) and decreased root ABA concentrations (by 28 %), decreased foliar iP, tZ and GA3 concentrations (by up to 90 %), and increased foliar ABA and JA concentrations (by 1.6- and 4.9-fold respectively) compared to intact plants, but only leaf ABA concentration correlated with partial stomatal closure. Two days of soil drying increased root ABA and JA concentrations in intact plants, and both hormones significantly decreased in roots of girdled plants compared to intact plants. Soil drying increased foliar iP, ABA, JA and SA concentrations by 1.3-, 2.5-, 1.3- and 1.4-fold respectively compared to well-watered intact plants by the end of the experiment, but girdling prevented this foliar iP, ABA, JA accumulation compared to droughted intact plants. Thus, girdling affects the co-ordination of root and leaf phytohormone concentrations such as JA and ABA, which may transiently modulate stomatal closure. However, leaf ABA concentration varied consistently with root ABA concentration across all treatments, and explained more of the variation in stomatal conductance than any other hormone or leaf water potential.

Emerging satellite observations for diurnal cycling of ecosystem processes.

Diurnal cycling of plant carbon uptake and water use, and their responses to water and heat stresses, provide direct insight into assessing ecosystem productivity, agricultural production and management practices, carbon and water cycles, and feedbacks to the climate. Temperature, light, atmospheric water demand, soil moisture and leaf water potential vary over the course of the day, leading to diurnal variations in stomatal conductance, photosynthesis and transpiration. Earth observations from polar-orbiting satellites are incapable of studying these diurnal variations. Here, we review the emerging satellite observations that have the potential for studying how plant functioning and ecosystem processes vary over the course of the diurnal cycle. The recently launched ECOsystem Spaceborne Thermal Radiometer Experiment on Space Station (ECOSTRESS) and Orbiting Carbon Observatory-3 (OCO-3) provide land surface temperature, evapotranspiration (ET), gross primary production (GPP) and solar-induced chlorophyll fluorescence data at different times of day. New generation operational geostationary satellites such as Himawari-8 and the GOES-R series can provide continuous, high-frequency data of land surface temperature, solar radiation, GPP and ET. Future satellite missions such as GeoCarb, TEMPO and Sentinel-4 are also planned to have diurnal sampling capability of solar-induced chlorophyll fluorescence. We explore the unprecedented opportunities for characterizing and understanding how GPP, ET and water use efficiency vary over the course of the day in response to temperature and water stresses, and management practices. We also envision that these emerging observations will revolutionize studies of plant functioning and ecosystem processes in the context of climate change and that these observations and findings can inform agricultural and forest management and lead to improvements in Earth system models and climate projections.

Determination of The Variation in Stomatal Conductance and Sapflow in Olive Trees Under Deficit Irrigation

Su stresi zeytinde verimliği etkileyen önemli çevresel faktörlerden biridir. Çalışma, 2017 yılında, Bornova Zeytincilik Araştırma Enstitüsü Müdürlüğü’nde, zeytin (cv. Memecik) ağaçlarında yürütülmüştür. Çalışmada, kısıtlı sulama koşullarında zeytin ağaçlarında stoma iletkenliği (gs) ve bitki özsu akışı değişimleri izlenmiştir. Bu amaçla; konular, K1: Susuz (yağışa dayalı); K2: %100 ET, K3: %66 ET ve K4: %33 ET şeklinde oluşturulmuştur. 
 Çalışmada konulara uygulanan sulama suyu miktarı, 0 ile 912 mm arasında, bitki su tüketimi değerleri ise 91 ile 851 mm arasında değişiklik göstermiştir. Araştırmada, dönem boyunca hesaplanan ortalama stoma iletkenliği (gs) değerleri K1 konusu için 293.83 mmol m-2 sn-1, K2 konusu için 382.62 mmol m-2 sn-1, K3 konusu için 371.50 mmol m-2 sn-1, K4 konusu için 293.57 mmol m-2 sn-1 olarak elde edilmiştir. 
 Bitki özsu akış ölçümlerinin sezon ortalamaları ise K1, K2, K3 ve K4 konusu için sırasıyla 0.054, 0.091, 0.073 ve 0.065 ml cm-2 dak.-1 olarak elde edilmiştir. Zeytin ağaçlarında konulara ait özsu akış hızlarının kontrol konusuna (K2) ait özsu akış hızına oranını gösteren transpirasyon oranının sezon ortalaması ise K1, K2, K3 ve K4 için sırasıyla 0.59, 1, 0.80, 0.72 olarak bulunmuştur.
 Çalışmada elde edilen verilere göre stoma iletkenliği değerlerinde konular arasındaki fark istatistiksel olarak önemli bulunmuştur. Yine araştırmanın önemli bir bulgusu her iki parametre arasında pozitif doğrusal (r2 = 0.528) bir ilişkinin bulunmasıdır. Elde edilen veriler ışığında; zeytin ağaçlarında su stresinin belirlenmesinde, stoma iletkenliği (gs) ve bitki özsu akışı ölçüm değerlerinin kullanılabileceği ifade edilebilir.

Jasmonic acid priming of potato uses hypersensitive response-dependent defense and delays necrotrophic phase change against Phytophthora infestans

One of the most important diseases affecting potato is late blight, caused by the oomycete Phtytophthora infestans. The use of jasmonic acid has been reported to reduce the progression of the disease in potato, but the defense mechanisms involved in this response are unknown. In this study we described the effect of jasmonic acid as a priming agent over time in the defense response of potato against the invasion of P. infestans. We observed that the initial stimulus generated by the exogenous application of jasmonic acid had an effect on the stomatal conductance of the treated tissue and activated StMYC2 expression. Results reveal a priming effect in plants inoculated 11 days after treatment with jasmonic acid, evidenced by an increased transcriptional induction of defense-associated genes, decrease in the number of necrotic lesions and an evident reduction of lesion area (72.23%). Furthermore, in this study, we show that the tested concentration of jasmonic acid does not have an adverse effect at the physiological level in plants, since variation in stomatal conductance was transient, no change in chlorophyll a fluorescence and no early senescence in leaves was observed.

Biophysically Informed Imaging Acquisition of Plant Water Status

Vegetation controls carbon and water fluxes because of the fundamental tradeoff between carbon dioxide uptake and water loss occurring when stomata are open. Quantifying the rates of this exchange typically requires either intensive gas exchange or destructive harvesting of tissues and mass spectrometry analyses. Recent developments in high-throughput methods have enhanced our capacity to empirically test plant-environmental interactions. The vast integration characterizing satellite remote sensing methods masks organ-level physiological mechanisms limiting the predictive capability of current process models. Hence, more ground truth studies are necessary to determine the amount of mechanistic information needed to improve our understanding of forest, crop, and land management. Imaging methodologies, such as thermal and chlorophyll a fluorescence, are currently used to collect information for relevant traits such as water use, growth, and stress response. We tested these techniques during progressive drought across species with different susceptibility in controlled greenhouse conditions. We chose two highly represented tree species in North America: the gymnosperm Pinus ponderosa and the angiosperm Populus tremuloides. To better explore the whole drought response parameter space, we also tested a crop (Brassica rapa) and desert shrub (Artemisia tridentata). Thermal and fluorescence images of the canopy were coupled with leaf-level measurements as we performed three tests to predict drought response using 1) leaf temperature, 2) chlorophyll a fluorescence, and 3) the combination of the two. At five days of drought, leaf temperature increased 7 and 10%, accounting for 63% and 73% of the variation in stomatal conductance for both tree species, respectively. The fluorescence signal from images decreased ~12% and ~83% in moderately and severely droughted leaves respectively, reaching zero at mortality. Leaf water status was then predicted using a Bayesian approach that incorporated measurements’ uncertainty and parsimony in the analysis of the parameters. Changes in canopy temperature provided confident predictions for the reductions of daily evapotranspiration at the onset of drought. Empirically combining thermal and fluorescence measurements improved predictions (R2 = 0.81) of midday leaf water potential compared to univariate models. Our results represent an important step towards quantifying plant water status during drought using first principles that do not require species-specific information.

Circadian Regulation Does Not Optimize Stomatal Behaviour.

The circadian clock is a molecular timer of metabolism that affects the diurnal pattern of stomatal conductance (gs), amongst other processes, in a broad array of plant species. The function of circadian gs regulation remains unknown and here, we test whether circadian regulation helps to optimize diurnal variations in stomatal conductance. We subjected bean (Phaseolus vulgaris) and cotton (Gossypium hirsutum) canopies to fixed, continuous environmental conditions of photosynthetically active radiation, temperature, and vapour pressure deficit (free-running conditions) over 48 h. We modelled gs variations in free-running conditions to test for two possible optimizations of stomatal behaviour under circadian regulation: (i) that stomata operate to maintain constant marginal water use efficiency; or (ii) that stomata maximize C net gain minus the costs or risks of hydraulic damage. We observed that both optimization models predicted gs poorly under free-running conditions, indicating that circadian regulation does not directly lead to stomatal optimization. We also demonstrate that failure to account for circadian variation in gs could potentially lead to biased parameter estimates during calibrations of stomatal models. More broadly, our results add to the emerging field of plant circadian ecology, where circadian controls may partially explain leaf-level patterns observed in the field.

Morpho-physiological and molecular evaluation of drought tolerance in cassava (Manihot esculenta Crantz)

Abstract Understanding drought tolerance mechanisms of cassava is a pre-requisite to improve the performance of the crop in water-scarce regions. Several hypotheses have been formulated to suggest how cassava can withstand a prolonged period of drought. We performed field trials under drought conditions with a selection of 37 cassava genotypes to identify phenotypic and molecular patterns associated with drought tolerance. Plant morphologies varied significantly between cassava genotypes under drought conditions in Kenya, which indicates a strong genetic basis for phenotypic differences. Drought stress reduced yield by 59%, the number of edible storage roots by 43% and leaf retention by 50% on average. Over three years and in two experimental field sites, the most drought tolerant genotype bulked 7.1 (±2.1) t/ha yield while the most drought susceptible genotype yielded 3.3 (±1.4) t/ha under drought conditions. The significant positive correlation of yield under irrigated and non-irrigated conditions suggests that selection of genotypes with high yield performance under well-watered or control conditions should be prioritized to identify genotypes with superior performance under drought stress. The positive correlation between yield and leaf retention provided further evidence that leaf longevity positively contributes to yield in water-deficit conditions. Yield differences could be attributed in part to variation in stomatal conductance (gs) because selected drought tolerant genotypes maintained higher gs and delayed stomatal closure as compared to drought susceptible genotypes. Further analysis revealed that genetic or molecular differences for gs between drought tolerant and susceptible genotypes could be detected at early stages of water deficit. These differences likely involve both abscisic acid (ABA)-dependent and ABA-independent molecular pathways.

Day length regulates seasonal patterns of stomatal conductance in Quercus species.

Vapour pressure deficit is a major driver of seasonal changes in transpiration, but photoperiod also modulates leaf responses. Climate warming might enhance transpiration by increasing atmospheric water demand and the length of the growing season, but photoperiod-sensitive species could show dampened responses. Here, we document that day length is a significant driver of the seasonal variation in stomatal conductance. We performed weekly gas exchange measurements across a common garden experiment with 12 oak species from contrasting geographical origins, and we observed that the influence of day length was of similar strength to that of vapour pressure deficit in driving the seasonal pattern. We then examined the generality of our findings by incorporating day-length regulation into well-known stomatal models. For both angiosperm and gymnosperm species, the models improved significantly when adding day-length dependences. Photoperiod control over stomatal conductance could play a large yet underexplored role on the plant and ecosystem water balances.

Element content and expression of genes of interest in guard cells are connected to spatiotemporal variations in stomatal conductance.

Element content and expression of genes of interest on single cell types, such as stomata, provide valuable insights into their specific physiology, improving our understanding of leaf gas exchange regulation. We investigated how far differences in stomatal conductance (gs ) can be ascribed to changes in guard cells functioning in amphistomateous leaves. gs was measured during the day on both leaf sides, on well-watered and drought-stressed trees (two Populus euramericana Moench and two Populus nigra L. genotypes). In parallel, guard cells were dissected for element content and gene expressions analyses. Both were strongly arranged according to genotype, and drought had the lowest impact overall. Normalizing the data by genotype highlighted a structure on the basis of leaf sides and time of day both for element content and gene expression. Guard cells magnesium, phosphorus, and chlorine were the most abundant on the abaxial side in the morning, where gs was at the highest. In contrast, genes encoding H+ -ATPase and aquaporins were usually more abundant in the afternoon, whereas genes encoding Ca2+ -vacuolar antiporters, K+ channels, and ABA-related genes were in general more abundant on the adaxial side. Our work highlights the unique physiology of each leaf side and their analogous rhythmicity through the day.

Evaluation of the Phenotypic Repeatability of Canopy Temperature in Wheat Using Continuous-Terrestrial and Airborne Measurements.

Infrared canopy temperature (CT) is a well-established surrogate measure of stomatal conductance. There is ample evidence showing that genotypic variation in stomatal conductance is associated with grain yield in wheat. Our goal was to determine when CT repeatability is greatest (throughout the season and within the day) to guide CT deployment for research and wheat breeding. CT was measured continuously with ArduCrop wireless infrared thermometers from post-tillering to physiological maturity, and with airborne thermography on cloudless days from manned helicopter at multiple times before and after flowering. Our experiments in wheat, across two years contrasting for water availability, showed that repeatability for CT was greatest later in the season, during grain-filling, and usually in the afternoon. This was supported by the observation that repeatability for ArduCrop, and more so for airborne CT, was significantly associated (P < 0.0001) with calculated clear-sky solar radiation and to a lesser degree, vapor pressure deficit. Adding vapor pressure deficit to a model comprising either clear-sky solar radiation or its determinants, day-of-year and hour-of-day, made little to no improvement to the coefficient of determination. Phenotypic correlations for airborne CT afternoon sampling events were consistently high between events in the same year, more so for the year when soil water was plentiful (r = 0.7 to 0.9) than the year where soil water was limiting (r = 0.4 to 0.9). Phenotypic correlations for afternoon airborne CT were moderate between years contrasting in soil water availability (r = 0.1 to 0.5) and notably greater on two separate days following irrigation or rain in the drier year, ranging from r = 0.39 to 0.53 (P < 0.0001) for the midday events. For ArduCrop CT the pattern of phenotypic correlations, within a given year, was similar for both years: phenotypic correlations were higher during the grain-filling months of October and November and for hours-of-day from 11 onwards. The lowest correlations comprised events from hours-of-day 8 and 9 across all months. The capacity for the airborne method to instantaneously sample CT on hundreds of plots is more suited to large field experiments than the static ArduCrop sensors which measure CT continuously on a single experimental plot at any given time. Our findings provide promising support for the reliable deployment of CT phenotyping for research and wheat breeding, whereby the high repeatability and high phenotypic correlations between afternoon sampling events during grain-filling could enable reliable screening of germplasm from only one or two sampling events.

Abscisic acid signalling mediates biomass trade-off and allocation in poplar.

Abscisic acid (ABA) is a well known stress hormone regulating drought adaptation of plants. Here, we hypothesised that genetic engineering of genes involved in ABA stress signalling and photoperiodic regulation affected drought resistance by trade-off with biomass production in perennial poplar trees. We grew Populus tremula×tremuloides wild-type (T89) and various transgenic lines (two transformation events of 35S::abi1-1, 35S::RCAR, RCAR:RNAi, 35S::ABI3, 35S::AREB3, 35S::FDL1, FDL1:RNAi, 35S::FDL2 and FDL2:RNAi) outdoors and exposed them to drought in the second growth period. After the winter, the surviving lines showed a huge variation in stomatal conductance, leaf size, whole-plant leaf area, tree height, stem diameter, and biomass. Whole-plant leaf area was a strong predictor for woody biomass production. The 35S::AREB3 lines were compromised in biomass production under well irrigated conditions compared with wild-type poplars but were resilient to drought. ABA signalling regulated FDL1 and FDL2 expression under stress. Poplar lines overexpressing FDL1 or FDL2 were drought-sensitive; they shed leaves and lost root biomass, whereas the FDL RNAi lines showed higher biomass allocation to roots under drought. These results assign a new function in drought acclimation to FDL genes aside from photoperiodic regulation. Our results imply a critical role for ABA-mediated processes in balancing biomass production and climate adaptation.

Genotypic variation in whole-plant transpiration efficiency in sorghum only partly aligns with variation in stomatal conductance.

Water scarcity can limit sorghum (Sorghum bicolor (L.) Moench) production in dryland agriculture, but increased whole-plant transpiration efficiency (TEwp, biomass production per unit of water transpired) can enhance grain yield in such conditions. The objectives of this study were to quantify variation in TEwp for 27 sorghum genotypes and explore the linkages of this variation to responses of the underpinning leaf-level processes to environmental conditions. Individual plants were grown in large lysimeters in two well-watered experiments. Whole-plant transpiration per unit of green leaf area (TGLA) was monitored continuously and stomatal conductance and maximum photosynthetic capacity were measured during sunny conditions on recently expanded leaves. Leaf chlorophyll measurements of the upper five leaves of the main shoot were conducted during early grain filling. TEwp was determined at harvest. The results showed that diurnal patterns in TGLA were determined by vapour pressure deficit (VPD) and by the response of whole-plant conductance to radiation and VPD. Significant genotypic variation in the response of TGLA to VPD occurred and was related to genotypic differences in stomatal conductance. However, variation in TGLA explained only part of the variation in TEwp, with some of the residual variation explained by leaf chlorophyll readings, which were a reflection of photosynthetic capacity. Genotypes with different genetic background often differed in TEwp, TGLA and leaf chlorophyll, indicating potential differences in photosynthetic capacity among these groups. Observed differences in TEwp and its component traits can affect adaptation to drought stress.
