Spatial pattern of genetic diversity in field populations of Fusarium incarnatum-equiseti species complex.

Fusarium is associated with a number of wilt, blight, scab, and rot diseases in a range of economically important staple food crops worldwide. An assessment of the genetic structure and population stratification of Fusarium incarnatum‐equiseti species complex (FIESC) pathogen populations is important to understand the evolutionary potential of such populations in adapting to environmental change. Based on intersimple sequence repeat polymerase chain reaction (ISSR‐PCR), it was found that the pathogen population was structured into three genetic clusters for which genetic differentiation was higher within than among populations. There was high intrapopulation genetic diversity for population 1 (94.63%) which consisted largely of isolates collected from North Trinidad. Populations 2 and 3 had a low level of admixture among the populations based on overall population differentiation. Population 1 accounted for the highest amount of genetic variation (95.82%) followed by populations 2 and 3. Population stratification was reflected in the dendrogram topology, which consisted of three main genetic clusters and which coincided with the outcome of Bayesian and PCoA analyses. The populations were isolated by distance, and Voronoi tessellations indicated physical or structural barriers to gene flow which contributed to restricted admixture between two of three populations. These findings suggest a high evolutionary potential for this FIESC pathogen population, the implications of which directly affect disease management strategies.

Genetic Diversity and Azole Fungicide Sensitivity in Pseudocercospora musae Field Populations in Brazil.

Pseudocercospora musae, causal agent of Sigatoka leaf spot, or yellow Sigatoka disease, is considered a major pathogen of banana (Musa spp.). Widely disseminated in Brazil, this study explored the genetic diversity in field populations of the pathogen from production areas in the Distrito Federal and the States of Bahia, Minas Gerais, and Rio Grande do Norte. Resistance to demethylation inhibitor (DMI) fungicides was also examined. For 162 isolates from 10 banana growing regions, analysis of mating type idiomorph frequency was conducted, together with estimation of genetic diversity at 15 microsatellite loci. A total of 149 haplotypes were identified across the examined populations, with an average genetic diversity of 4.06. In general, populations displayed 1:1 proportions of idiomorphs MAT1-1 and MAT1-2, providing evidence for sexual recombination. Multilocus linkage disequilibrium also indicated asexual reproduction contributing to the genetic structure of certain populations. AMOVA revealed that 86.3% of the genetic differentiation of the pathogen occurred among isolates within populations. Discriminant Analysis of Principal Components (DAPC) identified six most probable genetic groups, with no population structure associated with geographic origin or collection site. Although genetic similarity was observed among certain populations from different states, data revealed increasing genetic differentiation with increasing geographic distance, as validated by Mantel’s test (r = 0.19, P < 0.001). On the basis of DMI fungicide sensitivity testing and CYP51 gene sequence polymorphism, isolates from the Distrito Federal separated into two main groups, one with generally higher EC50 values against eight DMI fungicides. A clear phenotype-to-genotype relationship was observed for isolates carrying the CYP51 alteration Y461N. Conventionally adopted fungicides for control of Sigatoka leaf spot are likely to be overcome by combined sexual and asexual reproduction mechanisms in P. musae driving genetic variability. Continued analysis of pathogen genetic diversity and monitoring of DMI sensitivity profiles of Brazilian field populations is essential for the development of integrated control strategies based on host resistance breeding and rational design of fungicide regimes.

Non-target site SDHI resistance is present as standing genetic variation in field populations of Zymoseptoria tritici.

BACKGROUNDA new generation of more active succinate dehydrogenase (Sdh) inhibitors (SDHIs) is currently widely used to control Septoria leaf blotch in northwest Europe. Detailed studies were conducted on Zymoseptoria tritici field isolates with reduced sensitivity to fluopyram and isofetamid; SDHIs which have only just or not been introduced for cereal disease control, respectively.RESULTSStrong cross‐resistance between fluopyram and isofetamid, but not with other SDHIs, was confirmed through sensitivity tests using laboratory mutants and field isolates with and without Sdh mutations. The sensitivity profiles of most field isolates resistant to fluopyram and isofetamid were very similar to a lab mutant carrying SdhC‐A84V, but no alterations were found in SdhB, C and D. Inhibition of mitochondrial Sdh enzyme activity and control efficacy in planta for those isolates was severely impaired by fluopyram and isofetamid, but not by bixafen. Isolates with similar phenotypes were not only detected in northwest Europe but also in New Zealand before the widely use of SDHIs.CONCLUSIONThis is the first report of SDHI‐specific non‐target site resistance in Z. tritici. Monitoring studies show that this resistance mechanism is present and can be selected from standing genetic variation in field populations. © 2017 The Authors. Seasonal dynamics of insecticide resistance, multiple resistance, and morphometric variation in field populations of Culex pipiens

Resistance to insecticides that impairs nervous transmission has been widely investigated in mosquito populations as insecticides are crucial to effective insect control. The development of insecticide resistance is also of special interest to evolutionary biologists since it represents the opportunity to observe the genetic consequences of a well-characterized alteration in the environment. Although the frequencies of resistance alleles in Culex pipiens populations against different groups of insecticides have been reported, no detailed information is available on the relative change in these allele frequencies over time. In this study, we collected mosquitoes of the Cx. pipiens complex from six locations in three seasons in the Aegean region of Turkey and examined the i) seasonal variations in resistance to four different chemical classes of insecticides, ii) seasonal fluctuations in frequencies of resistance-associated target-site mutations of the three genes (ace-1, kdr, and Rdl), and iii) potential seasonal variations in wing morphometric characters that may be modified in resistant mosquitoes. Our bioassay results indicated the presence of different levels of resistance to all tested insecticides for all three seasons in all locations. The results of the PCR-based molecular analysis revealed low frequencies of mutations in ace-1 and Rdl that are associated with resistance to malathion, bendiocarb, and dieldrin and no obvious seasonal changes. In contrast, we detected high frequencies and striking seasonal changes for two kdr mutations associated with resistance to DDT and pyrethroids. In addition, the evaluation of the field populations from all seasons in terms of the combinations of polymorphisms at four resistance-associated mutations did not reveal the presence of insects that are resistant to all pesticides. Results from the morphological analysis displayed a similar pattern for both wings and did not show a clear separation among the samples from the three different seasons. The results of this study have advanced our knowledge of the potential dynamics of insecticide resistance among populations of the Cx. pipiens complex. The implications of these results to the understanding of the evolution of insecticide resistance and the management of resistance in mosquitoes are discussed.

High levels of genetic and genotypic diversity in field populations of the barley pathogen Ramularia collo-cygni

The ascomycete pathogen Ramularia collo-cygni causes Ramularia leaf spot (RLS) on barley. Although R. collo-cygni is considerd an emerging disease of barley, little is known about genetic diversity or population genetic structure of this pathogen. We applied a set of polymorphic AFLP (Amplified Fragment Length Polymorphism) markers to investigate population genetic structure in two Northern European populations of R. collo-cygni. The distribution of AFLP alleles revealed low levels of population subdivision and high levels of genetic diversity at both locations. Our analyses included 87 isolates and of these 84 showed a unique genotype pattern. The genetic structure of populations in Scotland and Denmark is highly similar and we find no evidence of population sub-division. An analysis of molecular variance was used to show that 86 % of the variance is attributable to within field genetic variance. In spite of the high levels of genetic and genotypic diversity in the R. collo-cygni populations, we find significant evidence of linkage disequilibrium among the AFLP alleles using a multilocus analysis. We propose that the high levels of genotypic diversity and the lack of population differentiation result from considerable levels of gene flow between populations most likely mediated by seed borne dispersal of inoculum.

Obviation of Wheat Resistance to the Hessian Fly Through Systemic Induced Susceptibility

Unlike most documented plant-insect interactions, Hessian fly-resistance [Mayetiola destructor (Say)] in wheat (Triticum aestivum L.) is initiated by a gene-for-gene recognition event in which plants carrying a specific R gene recognize salivary effectors encoded by a corresponding larval avirulence gene. However, dual infestation resulting from oviposition by virulent insects from 5 d before to 3 d after oviposition by avirulent insects on the same host plant, lead to systemic induced susceptibility, obviation of resistance, and ultimately the survival of both virulent and genetically avirulent progeny to adulthood. Simultaneous oviposition allowed greater survival of avirulent progeny than ovipositions separated by larger intervals. Because of the induction of plant resistance, hatch of avirulent larvae before virulent was more detrimental to rate of development than hatch of virulent before avirulent larvae. Obviation of resistance was not localized to the leaf being attacked by the virulent larvae, but also functioned across spatial distance into younger leaves. This research suggests that virulent Hessian fly larvae directly suppress the defense response of wheat, thus providing a refuge for avirulent genotypes, preserving diversity in field populations and increasing durability of deployed resistance genes.

High‐Resolution Melting technology: a new tool for studying the Wolbachia endosymbiont diversity in the field

Stable infections by maternally transmitted symbionts are frequently found in field populations, especially in arthropods. Many questions remain regarding their contribution to host biology and ecology, and especially on environmental adaptation of their host. Wolbachia is one of the most common endosymbiont of invertebrates. This cytoplasmically inherited endocellular bacterium induces number of reproductive alterations in its arthropod hosts and various fitness effects that allow it to spread in host populations. To better understand the influence of Wolbachia on host phenotypes and consequences of the manipulation of reproduction on the host genetic differentiation, it is crucial to be able to discriminate Wolbachia strains and determine their prevalence, which requires exhaustive screening. In the present report, we proposed the use of a new tool for the population studies, based on the high resolution melting (HRM) analysis, less expensive and faster than the 'classical' methods for large-scale studies. We investigated the effectiveness of HRM to explore and characterize the diversity of Wolbachia strains. Results obtained showed that HRM is a powerful tool to identify strains and detect polymorphism in singly infected hosts. When individuals harboured a mixture of Wolbachia strains (multiple infections), there is a risk of underestimation of the diversity if the proportions of the strains are highly different. However, the same limitations exist for the other techniques commonly used. Overall, this study demonstrated that HRM analysis is a rapid and reliable technique useful for studying, without a priori, Wolbachia strains diversity in field populations.

The Influence of Host Fruit and Temperature on the Body Size of AdultCeratitis capitata(Diptera: Tephritidae) Under Laboratory and Field Conditions

The adult body size of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), varies in natural conditions. Body size is an important fitness indicator in the Mediterranean fruit fly; larger individuals are more competitive at mating and have a greater dispersion capacity and fertility. Both temperature during larval development and host fruit quality have been cited as possible causes for this variation. We studied the influence of host fruit and temperature during larval development on adult body size (wing area) in the laboratory, and determined body size variation in field populations of the Mediterannean fruit fly in eastern Spain. Field flies measured had two origins: 1) flies periodically collected throughout the year in field traps from 32 citrus groves, during the period 2003-2007; and 2) flies evolved from different fruit species collected between June and December in 2003 and 2004. In the lab, wing area of male and female adults varied significantly with temperature during larval development, being larger at the lowest temperature. Adult size also was significantly different depending on the host fruit in which larvae developed. The size of the flies captured at the field, either from traps or from fruits, varied seasonally showing a gradual pattern of change along the year. The largest individuals were obtained during winter and early spring and the smallest during late summer. In field conditions, the size of the adult Mediterannean fruit fly seems apparently more related with air temperature than with host fruit. The implications of this adult size pattern on the biology of C. capitata and on the application of the sterile insect technique are discussed.

Direct and indirect effects of metal stress on physiology and life history variation in field populations of a lycosid spider

1. Under stress, life history theory predicts reduced growth rates and adult sizes, reduced reproductive allocation, production of larger offspring and postponed reproduction. Both direct and indirect effects of metals can explain these trends, mainly linked to energetic constraints. Metallothionein-like proteins (MTLP’s) are believed to be an important defense mechanism against the adverse effects of metals and other stressors. 2. We tested these predictions comparing six field populations of the wolf spider Pardosa saltans, three of which were on sites that are historically polluted with heavy metals. 3. As expected for life histories evolving under energetic constraints, adult size and condition correlated negatively and egg mass positively with Cd concentrations for a subset of four populations. In the population that showed the highest cadmium and zinc body burdens, reproductive output and allocation were lowest and reproduction was postponed. 4. Contrary to our expectation, for all six study populations MTLP concentrations did not increase in exposed populations, indicating that this defense mechanism cannot explain the observed variation in life histories. 5. We conclude that indirect and synergistic effects of metal pollution may be more important than physiological defense mechanisms in shaping life history traits in field populations.

GENETIC DIVERSITY OF Waitea circinata var. zeae IN SOUTH CAROLINA REVEALED BY AMPLIFIED FRAGMENT LENGTH POLYMORPHISM (AFLP)

Rhizoctonia zeae (Waitea circinata var. zeae) is pathogenic to rice (Oniki et al., 1985); corn (Sumner and Bell, 1982); onion (Erper et al., 2006); sugarbeet (Kuznia and Windels, 1994); wheat and barley (Ogoshi et al., 1990) and tall fescue (Martin and Lucas, 1983). Waitea circinata var. zeae also causes foliar lesions on bermuda grass, creeping bentgrass and annual bluegrass (Burpee and Martin, 1992; Hsiang and Dean, 2001) during midsummer. Diseases of turfgrass caused by these pathogens occur most frequently during the warm and humid season, at temperatures between 28 and 36°C, inciting leaf and sheath spot (Burpee and Martin, 1992; Smiley et al., 1992). Waitea circinata (Warcup and Talbot) classified into three varieties, W. circinata var. circinata, W. circinata var. oryzae and W. circinata var. zeae based on differences in the colony morphology of the vegetative state (Gunnell, 1986). Waitea circinata var. circinata forms orange to dark brown, globose sclerotia up to 2 mm in diameter; W. circinata var. oryzae forms orange to salmon, irregularly shaped sclerotia; and W. circinata var. zeae forms orange to brown, regularly shaped sclerotia up to 1 mm in diameter (Leiner and Carling, 1994). Rhizoctonia zeae was assigned to Waitea anastomosis group WAG-Z (Oniki et al., 1985). Previous studies have examined genetic variation of these three varieties at molecular level. Random amplified polymorphic DNA (RAPD) and restriction fragment length polymorphism (RFLP) of rDNA internal transcribed spacer (ITS) region revealed that isolates of W. circinata var. circinata, W. circinata var. oryzae and W. circinata var. zeae separated into individual clusters (Toda et al., 2005). These results confirmed by using sequence analysis of the internal transcribed spacer (ITS) region of rDNA (de la Cerda et al., 2007; Toda et al., 2007). Amplified fragment length polymorphism (AFLP) is a genetic mapping technique based on selective amplification of a subset of restriction enzyme-digested DNA fragments to create a unique fingerprint for a particular genome (Vos et al., 1995). It is highly reproducible and amenable to a wide range of applications and DNA sources. For these reasons, the method has steadily gained popularity in applications, including genetic mapping (Mueller and Wolfenbarger, 1999; Savelkoul et al., 1999), medical diagnostics (Klaassen et al., 2002; Borst et al., 2003; van den Braak et al., 2004), genetic diversity and phylogenetic studies (Tredway et al., 1999; Bakkeren et al., 2000; Doignon-Bourcier et al., 2000; Rademaker et al., 2000; Mougel et al., 2002; Lee et al.,2004) and environmental management studies (Lucchini, 2003). The similarities within each variety of W. circinata were very high, but similarities were significantly lower between varieties by using rDNA-ITS region (Toda et al., 2007). In this study, we used amplified fragment length polymorphism (AFLP) to examine the genetic diversity of field population of W. circinata var. zeae.

Genetic variation in field populations of baculoviruses: Mechanisms for generating variation and its potential role in baculovirus epizootiology

Baculoviridae is a family of insect-specific DNA viruses that have been used as biological control agents for insect pest control. In most cases these baculovirus control agents are natural field isolates that have been selected based on their infectivity and virulence. The advent of molecular tools such as restriction endonucleases, targeted polymerase chain reaction and new DNA sequencing strategies have allowed for efficient detection and characterization of genotypic variants within and among geographic and temporal isolates of baculovirus species. It has become evident that multiple genotypic variants occur even within individual infected larvae. Clonal strains of baculovirus species derived either by in vitro or in vivo approaches have been shown to vary with respect to infectivity and virulence. Many of the cell culture derived plague-purified strains have deletions that interrupt egt expression leading to virus strains that kill infected hosts more quickly. As well, in vitro clones often involve larger genomic deletions with the loss of pif gene function, resulting in strains deficient for oral infectivity. There are an increasing number of baculovirus species for which complete genome sequences are available for more than one strain or field isolate. Results of comparative analysis of these strains indicated that hr regions and bro genes often mark “hot spots” of genetic variability between strains and of potential recombination events. In addition, the degree of nucleotide polymorphisms between and within strains and their role in amino acid substitutions within ORFs and changes in promoter motifs is also beginning to be appreciated. In this short review the potential mechanisms that generate and maintain this genetic diversity within baculovirus populations is discussed, as is the potential role of genetic variation in host-pathogen interactions.

Genotypic diversity, a survival strategy for the apicomplexan parasite Theileria parva

The tick-borne protozoan parasite Theileria parva causes East Coast fever (ECF), a severe lymphoproliferative disease of cattle that is a major constraint to the improvement of livestock in eastern, central and southern Africa. Studies in cattle experimentally infected with T. parva have shown that the protective cytotoxic T lymphocyte (CTL) response is tightly focused, with individual animals recognizing only one or two dominant antigens, the identity of which varies with MHC class I phenotype. It is well known that cross-protection between T. parva stocks is limited, but precise evaluation of genetic diversity in field populations of the parasite has been hampered by a lack of molecular markers spanning the genome. A recently described panel of satellite markers has provided evidence for substantial genotypic diversity and recombination but does not provide cover for large segments of the genome. To address this deficiency, we undertook to identify additional polymorphic markers covering these regions and we report herein 42 newly identified PCR-RFLP markers distributed across the 4 T. parva chromosomes, as well as 19 new satellite markers for chromosomes 1 and 2. This brings the total number of available polymorphic markers to 141 for the 8.5Mb genome. We have used these markers to characterise existing parasite stabilates and have also shown that passage of the parasite through naïve cattle and ticks can lead to substantial changes of parasite populations in resulting stabilates. These markers have also been used to show that passage of mixed parasites through an immunised calf results in the removal of the immunising genotype from the parasite population produced by ticks fed on this animal.

HiddenWolbachiadiversity in field populations of the European cherry fruit fly,Rhagoletis cerasi(Diptera, Tephritidae)

The European cherry fruit fly Rhagoletis cerasi has been a field model for cytoplasmic incompatibility since the mid 1970s. Two Wolbachia strains were detected in this tephritid species and wCer2 was described as the CI inducing agent dividing European populations into two unidirectional incompatible groups, i.e. southern females produce viable offspring with northern males, whereas the reciprocal cross results in incompatibility. We detected three new Wolbachia strains by sequencing a multitude of plasmids derived from Wolbachia surface protein gene (wsp) polymerase chain reaction (PCR) products. Strain-specific primers were developed allowing individual diagnosis without need for cloning. Hybridization of specific PCR products with a wsp oligonucleotide enhanced the detection limit significantly and revealed the presence of low-titre infections in some strains, in different ontogenetic stages and in adults of different age. We then performed a survey of strain prevalence and infection frequency in eight European regions. wCer1 was fixed in all populations, whereas wCer2 was detected only in the South. wCer3 frequency was the lowest without a clear distribution pattern. The abundance of wCer4 was homogenous across Europe. Like wCer2, wCer5 showed significant differences in spatial distribution. Our new findings of previously undetected and recombinant Wolbachia strains in R. cerasi reveal a major caveat to the research community not to overlook hidden Wolbachia diversity in field populations. Low-titres and geographical variability in Wolbachia diversity are expected to influence the outcome of Wolbachia population dynamics and Wolbachia-based insect population control and may create invasion barriers for expanding and artificially introduced Wolbachia strains.

Spatial aggregation in Fusarium pseudograminearum populations from the Australian grain belt

Previous studies have evaluated the overall structure of populations of Fusarium pseudograminearum (teleomorph, Gibberella coronicola), causal agent of cereal crown rot, but there is no information available on spatial relationships of genetic variation in field populations. Three 1‐m‐row sections in crown‐rot‐affected wheat fields in the Australian grain belt were intensively sampled to estimate population genetic parameters and the spatial aggregation, or clustering, of disease aggregates and genotypes. Estimates of population genetic parameters based on amplified fragment length polymorphisms (AFLPs) indicated that the genetic diversity in isolates from the 1‐m‐row populations described a significant portion of the diversity recorded for corresponding field and regional populations. In point pattern analysis, there was physical clustering and aggregation of F. pseudograminearum isolates from two of the three sites. Analysis of the spatial distribution of clonal haplotypes (DICE similarity ≥ 97%) indicated significant aggregation of clones in all three 1‐m‐row populations. Based on matrix comparison tests, both mating types and genetic distances had significant spatial aggregation for at least two of the three 1‐m‐row populations. This is consistent with the presence of non‐random spatial genetic structure due to clonal aggregation. High levels of genetic diversity and spatial structuring of disease and genotypes in at least two of the three 1‐m‐row populations is consistent with the hypothesis that stubble is a primary inoculum source in no‐tillage farming systems, resulting in aggregated patterns of disease and allowing for haplotypes to be maintained in the field over a number of annual cropping cycles.

Marker stability throughout 400 days of in vitro hyphal growth in the filamentous ascomycete, Sclerotinia sclerotiorum

The stability of routinely used, population genetic markers through approximately 1 year of continuous laboratory growth was investigated in the common, plant pathogentic ascomycete Sclerotinia sclerotiorum. Given reports of accelerated mutation rates at higher temperatures, both a permissive temperature, 22°C, and a temperature at the high end of tolerance, 30°C, were employed. Because mycelial growth rate was tracked among mitotic lineages established for each strain, a subsidiary objective was addressed, testing the stability of a 30°C-competent phenotype. Twelve laboratory strains of S. sclerotiorum, including the genome sequence isolate, 1980, were propagated serially for up to 400 days at 22°C. Five of these strains were also propagated at 30°C. No mutations were observed in mycelial compatibility groupings (MCGs), DNA fingerprints, alleles at 7 microsatellite loci, or alleles at 56 AFLP loci. All of these markers show variation in field populations, which are likely much larger and influenced by different and more stochastic environmental processes. In S. sclerotiorum, population genetic markers were stable over time through serial transfer and growth of laboratory strains at both 22°C and 30°C. The strain isolated after extended drought and capable of infecting plants at 28°C demonstrated the stability of its high temperature-competent phenotype, in addition to its stable growth rate at 22°C. This observation has implications for modeling pathogen tolerance or adaptation under conditions of environmental stochasticity, including climate warming.

Estimation of phenotypic diversity in field populations of Magnaporthe grisea from two upland rice cultivars

The phenotypic diversity of Magnaporthe grisea was evaluated based on leaf samples with blast lesions collected from eight commercial fields of the upland rice cultivars 'BRS Primavera' and 'BRS Bonança', during the growing seasons of 2001/2002 and 2002/2003, in Goias State. The number of M. grisea isolates from each field utilized for virulence testing varied from 28 to 47. Three different indices were used based on reaction type in the eight standard international differentials and eight Brazilian differentials. The M. grisea subpopulations of ´Primavera' and 'Bonança', as measured by Simpson, Shannon and Gleason indices, showed similar phenotypic diversities. The Simpson index was more sensitive relation than those of Shannon and Gleason for pathotype number and standard deviation utilizing Brazilian differentials. However, the Gleason index was sensitive to standard deviation for international differentials. The sample size did not significantly influence the diversity index. The two sets of differential cultivars used in this study distinguished phenotypic diversity in different ways in all of the eight subpopulations analyzed. The phenotypic diversity determined based on eight differential Brazilian cultivars was lower in commercial rice fields of 'Primavera' than in the fields of 'Bonança,' independent of the diversity index utilized, year and location. Considering the Brazilian differentials, the four subpopulations of 'BRS Primavera' did not show evenness in distribution and only one pathotype dominated in the populations. The even distribution of pathotype was observed in three subpopulations of 'BRS Bonança'. The pathotype diversity of M. grisea was determined with more precision using Brazilian differentials and Simpson index.

Variation in growth and instar number in field and laboratory Manduca sexta

The tobacco hornworm Manduca sexta has been an important model system for understanding physiological control of growth, development and metamorphosis of insects for more than half a century. Like all Manduca, M. sexta typically has five larval instars, with developmental commitment to metamorphosis occurring early in the 5th (final) instar. Here we show that M. sexta from a field population in North Carolina (USA) shows substantial intraspecific variation in the number of larval instars when feeding on a modified artificial diet. Individuals with six instars consistently exhibited slower growth rates during early larval development than individuals with five instars. The frequency of individuals with six instars decreased with increased rearing temperature. In contrast, M. sexta from a laboratory colony consistently had five instars, and had more rapid larval growth rates than M. sexta from the field. We identify a threshold body size at the start of the 5th instar that predicts whether an individual will have five (greater than 600mg) or six instars (less than 600mg). Variation in field populations in Manduca provides an important resource for understanding physiological control, developmental plasticity and evolution of growth rate, body size and instar number.

Virulence Genes in Heterodera glycines: Allele Frequencies and Ror Gene Groups Among Field Isolates and Inbred Lines

ABSTRACT Genetic variation in field populations of Heterodera glycines is a key issue for both resistance gene deployment and basic understanding of virulence-gene flow in populations. In this study, we examined phenotypically defined genes for virulence under selection from host resistance. We separated the most common H. glycines genotypes in the United States into two virulence groups, based on their reproductive abilities on the resistant soybean plant introduction (PI) 88788. These groups correspond to previously identified virulence genes in the nematode, as follows: the dominant gene in H. glycines to PI88788, and the recessive genes to PI90763 and Pickett/Peking. Virulence allele frequencies and virulence genotype frequencies of selected field isolates were investigated by testing the host range of single-female-derived lines, which were developed through single-female inoculation on the standard susceptible soybean 'Lee 68'. By comparing virulence genotype frequencies between the original field isolates and their single-female-derived lines, we were able to determine allele frequencies in the field populations. The results suggest that tremendous variation in H. glycines virulence genes exists among field populations. Potential mechanisms of selection which could cause virulence genotype frequency increases are discussed as related to population genetics equilibrium theory.

Wild isolates of Plasmodium falciparum malaria show decreased sensitivity to in vitro inhibition of parasite growth mediated by autologous host antibodies.

Antigenic diversity in field populations of Plasmodium falciparum parasites may delay the acquisition of protective immunity to malaria, the development of which may thus require repeated exposure to infection over a prolonged period of time. In this study we show that P. falciparum parasites may vary in their sensitivity to antibody-mediated invasion/growth inhibition in vitro. Wild isolates of P. falciparum from children living in an endemic area of Burkina Faso were tested for their sensitivity to the growth inhibitory effects of antibodies originating from the same (autologous) and from other donors (heterologous). A significantly lower invasion inhibition activity was obtained when the isolates and antibodies were tested in autologous compared with heterologous combinations. The lower sensitivity to growth inhibition by autologous antibodies may be due to immune pressure in vivo, selecting from a heterogeneous parasite population those with a low expression of the antigens recognized by the host's antibodies. Alternatively, the parasites cultured from each child might represent expanding parasite populations, mainly constituting strains not earlier seen by the immune system of that specific host. The results reinforce the concern about Plasmodium antigenic diversity as a major obstacle towards the development of an effective malaria vaccine.

Revisiting the rDNA sequence diversity of a natural population of the arbuscular mycorrhizal fungus Acaulospora colossica.

In 1999, the diversity of a field population of the arbuscular mycorrhizal (AM) fungus Acaulospora colossica was characterized using DNA sequence data. Since 1999, AM fungal sequences have accumulated rapidly within public databases. Moreover, novel phylogenetic tools have been developed and can be used to interpret the data. A second analysis of those sequences collected in 1999 demonstrates that while the majority of the sequences are, in fact, sequences of A. colossica; a minority of the sequences still cannot be identified with confidence. Those sequences identified as A. colossica can be used to show that (1) the nuclear rDNA ITS regions are remarkably diverse, and (2) sequences isolated from different spores of the same site may be more closely related to each other than to sequences of other sites, so that the genetic diversity of an AM fungal field population may be spatially structured; however, identical sequences can also be recovered from different sites.
