Techno-economic analysis of biomass gasification for hydrogen production in three US-based case studies

Production of low-emissions hydrogen can improve the sustainability of certain hard-to-decarbonize sectors, like industrial and residential heating. Gasification of biomass has been identified as a promising production method for high-volume clean hydrogen, but uncertainty around its economic conditions have prevented it from being used in practice. This work evaluates the economics of three potential gasification facilities in the United States to identify the optimal conditions for a successful hydrogen plant. The locations were selected based on local policies that create a need for high volume clean hydrogen by regulating natural gas emissions. Local features, like biomass type, availability, and distance between the biomass and hydrogen market, are taken into account. A novel simulation is used to build upon previous models to determine the effects of biomass composition on hydrogen yield and utility usage. The environmental impact of each location is then assessed using GREET, Argonne's greenhouse gas emissions quantification tool, to determine the plant's qualifications for tax credits under the Inflation Reduction Act. Ultimately, the cost of transported hydrogen in a 40 metric ton/day facility was calculated to be $3.47/kg H2 in Klamath County, Oregon (which serves both the Oregon and California markets), $4.11/kg H2 in Park County, Colorado, and $3.63/kg H2 in Middlesex County, Massachusetts. A sensitivity analysis found transportation distance and biomass availability to be major factors in these costs. Variability in biomass composition had minimal effects on plant performance.

What CHO is made of: Variations in the biomass composition of Chinese hamster ovary cell lines

BackgroundCell line-specific, genome-scale metabolic models enable rigorous and systematic in silico investigation of cellular metabolism. Such models have recently become available for Chinese hamster ovary (CHO) cells. However, a key ingredient, namely an experimentally validated biomass function that summarizes the cellular composition, was so far missing. Here, we close this gap by providing extensive experimental data on the biomass composition of 13 parental and producer CHO cell lines under various conditions. ResultsWe report total protein, lipid, DNA, RNA and carbohydrate content, cell dry mass, and detailed protein and lipid composition. Furthermore, we present meticulous data on exchange rates between cells and environment and provide detailed experimental protocols on how to determine all of the above. The biomass composition is converted into cell line- and condition-specific biomass functions for use in cell line-specific, genome-scale metabolic models of CHO. Finally, flux balance analysis (FBA) is used to demonstrate consistency between in silico predictions and experimental analysis. ConclusionsOur study reveals a strong variability of the total protein content and cell dry mass across cell lines. However, the relative amino acid composition is independent of the cell line and condition and thus needs not be explicitly measured for each new cell line. In contrast, the lipid composition is strongly influenced by the growth media and thus will have to be determined in each case. These cell line-specific variations in biomass composition have a small impact on growth rate predictions with FBA, as inaccuracies in the predictions are rather dominated by inaccuracies in the exchange rate spectra. Cell-specific biomass variations only become important if the experimental errors in the exchange rate spectra drop below twenty percent.

Recovery of microalgal biomass and metabolites from homogenized, swirl flash-dried microalgae

Production of intracellular metabolites from microalgae involves various processing steps. Since algal drying and cell disruption are integral processes of these operations, effects of novel swirl flash-drying technique and cell cracking by high-pressure liquid shear method were tested. Variations in biomass composition (focusing on the fatty acid and pigment composition) of two microalgal species, Chlorella pyrenoidosa and Nannochloropsis salina, were studied in order to investigate effects of novel drying technique, including whether recovery of bioactive compounds is more efficient on dried or wet biomass. Applying novel swirl flash-drying technique showed no significant adverse effect on the fatty acid composition, including heat sensitive eicosapentaenoic acid (EPA), of tested microalgal species. Pigment and tocopherol composition of C. pyrenoidosa showed tendency to degrade after applying both cell cracking and drying treatment. Considering these data swirl flash-drying technique has a potential as a new drying technique for microalgae biomass.

Bioenergy from wastewater resources: Nutrient removal, productivity and settleability of indigenous algal-bacteria polyculture, and effect of biomass composition variability on methane production kinetics and anaerobic digestion energy balance

Algal-bacteria high-rate ponds represent an energy-efficient wastewater treatment approach and a source for affordable and sustainable biomass feedstock for production of renewable energy through anaerobic digestion (AD). However, there is still a need for more data on wastewater treatment efficiency, biomass productivity and settleability from outdoor treatment facilities, as well as on impact of variability in biomass composition and digestibility on methane yield and energy output. Hydraulic retention time (HRT) and wastewater quality fed into 30 m2 raceway ponds had a major effect on algal-bacteria polyculture productivity, settleability, phylogenetic and biochemical compositions, digestibility and methane yield. While Micractinium, Scenedesmus, Chlorella, and pennate diatoms were always among the key species observed, the gross productivity and 2-hour settleability during summer cultivation varied in the first-stage ponds treating primary wastewater from 29 ± 5 to 54 ± 12 gAFDW/m2/d and from 88 ± 8 to 94 ± 4% for HRT of 3 and 2 days, respectively. For these conditions, the effluent had csBOD of 3.4 or 3.3 mg/L and Ntotal (mostly NO3−-N) of 9.2 or 8.2 mg/L, respectively. The second-stage algal ponds (HRT 3 days) showed lower productivity of 16 ± 6 gAFDW/m2/d, settleability of 84 ± 11%, and effluent csBOD 3.7 mg/L and Ntotal 0.8 mg/L. Biomass composition from different ponds was 34–38% protein, 18–28% total lipids and 6–14% FAME. The methane yield varied about 30% with largest value of 0.34 ± 0.01 L/gVS and showed a positive correlation with biomass lipid content (R2 = 0.93). First-order and pseudo-parallel first-order rate kinetic models exhibited a better fit for methane production (most R2 > 0.993) than the modified Gompertz model. The variation in biomass composition led to significant differences in energy output (varied by about 60%), Net Energy Ratios (ranged from 1.6 to 2.2) and Net Energy Efficiency (from 60% to 70%) when projecting the energy balance for a large-scale continuous AD process with an optimal HRT of 20–30 days.

Life cycle assessment of ethanol production from miscanthus: A comparison of production pathways at two European sites

AbstractLignocellulosic ethanol represents a renewable alternative to petrol. Miscanthus, a perennial plant that grows on marginal land, is characterized by efficient use of resources and is considered a promising source of lignocellulosic biomass. A life cycle assessment (LCA) was performed to determine the environmental impacts of ethanol production from miscanthus grown on marginal land in Great Britain (Aberystwyth) and an average‐yield site in Germany (Stuttgart; functional unit: 1 GJ). As the conversion process has substantial influence on the overall environmental performance, the comparison examined three pretreatment options for miscanthus. Overall, results indicate lower impacts for the production in Stuttgart in comparison with the corresponding pathways in Aberystwyth across the analysed categories. Disparities between the sites were mainly attributed to differences in biomass yield. When comparing the conversion options, liquid hot water treatment resulted in the lowest impacts, followed by dilute sulphuric acid. Dilute sodium hydroxide pretreatment represented the least favourable option. Site‐dependent variation in biomass composition and degradability did not have substantial influence on the environmental performance of the analysed pathways. Additionally, implications of replacing petrol with miscanthus ethanol were examined. Ethanol derived from miscanthus resulted in lower impacts with respect to greenhouse gas emissions, fossil resource depletion, natural land transformation and ozone depletion. However, for other categories, including toxicity, eutrophication and agricultural land occupation, net scores were substantially higher than for the fossil reference. Nevertheless, the results indicate that miscanthus ethanol produced via dilute acid and liquid hot water treatment at the site in Stuttgart has the potential to comply with the requirements of the European Renewable energy directive for greenhouse gas emission reduction. For ethanol production at the marginal site, carbon sequestration needs to be considered in order to meet the requirements for greenhouse gas mitigation.

Emission characteristics of a pyrolysis-combustion system for the co-production of biochar and bioenergy from agricultural wastes

The co-production of biochar and bioenergy using pyrolysis-combustion processes can potentially minimize the emission problems associated with conventional methods of agricultural by-product disposal. This approach also provides significant added-value potential through biochar application to soil. Despite these advantages, variations in biomass composition, including sulfur, nitrogen, ash, and volatile matter (VM) content, may significantly influence both the biochar quality and the emissions of harmful particulate matter (PM) and gaseous pollutants (SO2, H2S, NO2, NO). Using a laboratory-scale continuous pyrolysis-combustion facility, the influence of biomass composition (rice husk and grape pruning) and volatile production (pyrolysis) temperature (400–800 °C) on the biochar properties and emissions during combustion of the raw pyrolysis volatiles were evaluated. Utilization of grape pruning resulted in higher energy-based yields of PM10 than the rice husk, the majority of which consisted of the PM1.1 fraction due to the elevated pyrogas content of the volatiles. The PM emissions were found to be independent of the feedstock ash content due to its retainment in the biochar. Greater volatilization of biomass sulfur and nitrogen during pyrolysis at higher temperatures resulted in higher yields of sulfurous and nitrogenous gaseous pollutants. The energy-based yields of NO and NO2 were found to increase by 16% and 50% for rice husk and 21% and 189% for grape pruning respectively between 400 and 800 °C. The same trend was also observed for the emissions of H2S and SO2 for both feedstocks.

Biomass composition of Arthrospira platensis during cultivation on industrial process water and harvesting

Microalgae have the ability to utilize nutrients from wastewater and use it for biomass production. The effluent from a biogas process was tested as a nutrient source for blue-green microalga Arthrospira platensis cultivation and compared with conventional synthetic medium. Cultivation was carried out in four different concentrations of industrial process water (25, 50, 75, and 100%). The biomass was then harvested by microfiltration, and centrifugation followed by freeze drying. Variations in biomass composition were studied, in order to investigate effects of industrial process water on A. platensis over 30 days of cultivation. Applied harvesting techniques were evaluated for their effect on physiochemical properties of the biomass. Arthrospira platensis was able to grow in all tested wastewater concentrations except 100%, however, increase of wastewater concentration in medium resulted in a decreased growth rate. Partial substitution of synthetic Zarrouk medium with 25% of wastewater showed no adverse effect on chemical composition of the biomass including high protein content (45–58% dry weight) and favorable fatty acid composition (42–45% PUFAs of total fatty acids). Evaluation by optical microscopy showed that microfiltration caused cell rupture at the moderate level while centrifugation had more severe effect on A. platensis. Effect of centrifugal forces and shear stress on A. platensis cells was confirmed by detecting lower lipid content in samples after applying both microfiltration and centrifugation due to cell content leakage.

Clostridium butyricum maximizes growth while minimizing enzyme usage and ATP production: metabolic flux distribution of a strain cultured in glycerol

BackgroundThe increase in glycerol obtained as a byproduct of biodiesel has encouraged the production of new industrial products, such as 1,3-propanediol (PDO), using biotechnological transformation via bacteria like Clostridium butyricum. However, despite the increasing role of Clostridium butyricum as a bio-production platform, its metabolism remains poorly modeled.ResultsWe reconstructed iCbu641, the first genome-scale metabolic (GSM) model of a PDO producer Clostridium strain, which included 641 genes, 365 enzymes, 891 reactions, and 701 metabolites. We found an enzyme expression prediction of nearly 84% after comparison of proteomic data with flux distribution estimation using flux balance analysis (FBA). The remaining 16% corresponded to enzymes directionally coupled to growth, according to flux coupling findings (FCF). The fermentation data validation also revealed different phenotype states that depended on culture media conditions; for example, Clostridium maximizes its biomass yield per enzyme usage under glycerol limitation. By contrast, under glycerol excess conditions, Clostridium grows sub-optimally, maximizing biomass yield while minimizing both enzyme usage and ATP production. We further evaluated perturbations in the GSM model through enzyme deletions and variations in biomass composition. The GSM predictions showed no significant increase in PDO production, suggesting a robustness to perturbations in the GSM model. We used the experimental results to predict that co-fermentation was a better alternative than iCbu641 perturbations for improving PDO yields.ConclusionsThe agreement between the predicted and experimental values allows the use of the GSM model constructed for the PDO-producing Clostridium strain to propose new scenarios for PDO production, such as dynamic simulations, thereby reducing the time and costs associated with experimentation.

Sources of Biomass Feedstock Variability and the Potential Impact on Biofuels Production

Terrestrial lignocellulosic biomass has the potential to be a carbon neutral and domestic source of fuels and chemicals. However, the innate variability of biomass resources, such as herbaceous and woody materials, and the inconsistency within a single resource due to disparate growth and harvesting conditions, presents challenges for downstream processes which often require materials that are physically and chemically consistent. Intrinsic biomass characteristics, including moisture content, carbohydrate and ash compositions, bulk density, and particle size/shape distributions are highly variable and can impact the economics of transforming biomass into value-added products. For instance, ash content increases by an order of magnitude between woody and herbaceous feedstocks (from ∼0.5 to 5 %, respectively) while lignin content drops by a factor of two (from ∼30 to 15 %, respectively). This increase in ash and reduction in lignin leads to biofuel conversion consequences, such as reduced pyrolysis oil yields for herbaceous products as compared to woody material. In this review, the sources of variability for key biomass characteristics are presented for multiple types of biomass. Additionally, this review investigates the major impacts of the variability in biomass composition on four conversion processes: fermentation, hydrothermal liquefaction, pyrolysis, and direct combustion. Finally, future research processes aimed at reducing the detrimental impacts of biomass variability on conversion to fuels and chemicals are proposed. © 2015 Battelle Energy Alliance, LLC, contract manager for Idaho National Laboratory.

Seasonal and spatial variation in biochemical composition of Saccharina latissima during a potential harvesting season for Western Sweden

Abstract This study monitored the biomass composition of Saccharina latissima during a potential harvesting season on the West coast of Sweden, in order to find suitable harvest times for biorefinery purposes. Specimens of S. latissima were sampled at three locations in June, August and October and the biomass was analysed for its macromolecular composition, as well as for the content of several specific compounds, e.g. sugars and fatty acids. PERMANOVA analyses showed that there was a significant difference in the biomass composition among time points. The total carbohydrate concentration was lowest in June and peaked at 360 mg g-1 dry weight in August, while the mannitol content was highest, 90 mg g-1, in June and decreased throughout the sampling period. Total protein and fatty acid concentrations were found to be approximately 80 and 3 mg g-1, respectively, with relatively little variation over time. Overall, there was little spatial variation in the macromolecular composition, although the concentration of some specific monosaccharides and fatty acids, as well as the total phenolic content, differed among localities. We discuss the implications of the observed variation in biomass composition of S. latissima for future biorefinery purposes.

Achieving Metabolic Flux Analysis for S. cerevisiae at a Genome-Scale: Challenges, Requirements, and Considerations.

Recent advances in 13C-Metabolic flux analysis (13C-MFA) have increased its capability to accurately resolve fluxes using a genome-scale model with narrow confidence intervals without pre-judging the activity or inactivity of alternate metabolic pathways. However, the necessary precautions, computational challenges, and minimum data requirements for successful analysis remain poorly established. This review aims to establish the necessary guidelines for performing 13C-MFA at the genome-scale for a compartmentalized eukaryotic system such as yeast in terms of model and data requirements, while addressing key issues such as statistical analysis and network complexity. We describe the various approaches used to simplify the genome-scale model in the absence of sufficient experimental flux measurements, the availability and generation of reaction atom mapping information, and the experimental flux and metabolite labeling distribution measurements to ensure statistical validity of the obtained flux distribution. Organism-specific challenges such as the impact of compartmentalization of metabolism, variability of biomass composition, and the cell-cycle dependence of metabolism are discussed. Identification of errors arising from incorrect gene annotation and suggested alternate routes using MFA are also highlighted.

Effect of Brassica napus cultivar on cellulosic ethanol yield.

BackgroundIntraspecific variations in biomass composition are likely to influence their suitability for biorefining. This may be particularly important in species such as Brassica napus, which contain many different crop types bred for different purposes. Here, straw derived from 17 B. napus cultivars, of varying crop types, were steam exploded, saccharified and fermented to establish differences in biomass composition relevant to cellulosic ethanol production.ResultsDespite being grown and processed in the same manner, straw from the various cultivars produced different saccharification and fermentation yields after processing. Fermentation inhibitor abundances released by steam explosion also varied between genotypes. Cultivars with glucan-rich straw did not necessarily produce higher saccharification or ethanol yields after processing. Instead, the compositions of non-cellulosic components were more reliable indicators of substrate quality. The abundance of pectins and arabinogalactans had the greatest influence on saccharification efficiency between straw genotypes.ConclusionsIn dicotyledonous species, such as B. napus, variations in the abundance of pectins between crop cultivars are likely to influence processing efficiency for bioethanol production. Knowledge of these genotypic variants provides targets for plant breeding and could aid in the development of improved cellulase cocktails.

Enzymatic saccharification of shrub willow genotypes with differing biomass composition for biofuel production

In the conversion of woody biomass feedstocks into liquid fuel ethanol, the pretreatment process is the most critical and costly step. Variations in biomass composition based on genetic differences or environmental effects have a significant impact on the degree of accessibility accomplished by pretreatment and subsequent sugar release by enzymatic hydrolysis. To evaluate this, biomass from 10 genetically diverse, genotypes of shrub willow (Salix spp.) was pretreated with a hot-water process at two levels of severity, hydrolyzed using a combination of two commercial enzyme cocktails, and the release of hexose and pentose monomers was quantified by high-performance liquid chromatography. Among the genotypes selected for analysis, cellulose content ranged from 39 to 45% (w/w) and lignin content ranged from 20 to 23% (w/w) at harvest. Differences in the effectiveness of the pretreatment process were observed among the various willow genotypes. Correlations were identified between total sugar release and % cellulose and % lignin content. There was a significant effect of pretreatment severity on polysaccharide accessibility, but the response to pretreatments was different among the genotypes. At the high severity pretreatment ‘SV1’ was the least recalcitrant with sugar release representing as much as 60% of total biomass. These results suggest that structural, as well as chemical characteristics of the biomass may influence pretreatment and hydrolytic efficiency.

Correlations of expression of cell wall biosynthesis genes with variation in biomass composition in shrub willow (Salix spp.) biomass crops

We have measured significant genetically determined variation in biomass composition among breeding populations of shrub willow, a biomass feedstock crop. This project was aimed to ask whether patterns of cell wall gene expression can be correlated with genetic variation in biomass composition at harvest, in order to develop assays of early differences in gene expression as indicators of harvestable biomass chemical composition and potentially reduce the time of selection for new willow genotypes. Previous studies have demonstrated that manipulation of expression of cell wall biosynthetic genes results in altered biomass chemical composition. We analyzed genes encoding enzymes involved in lignin biosynthesis and carbohydrate active enzymes selected based on their functional characterization and conservation in Populus trichocarpa and Arabidopsis thaliana. Fragments of 20 genes were cloned from young stem cDNA of Salix sachalinensis and Salix miyabeana. Expression profiling in willow stem apical tissue and developing stem tissue was performed for each isolated gene using probe-based quantitative real-time PCR. Two willow parental genotypes and six progeny within a hybrid family were selected for analysis, and significant differences in expression among the individuals and between tissue types were observed for most of the genes. Significant correlations between patterns of gene expression and variation in the biomass chemical composition of those genotypes provide insight into the genetic regulation of lignocellulosic deposition in this important bioenergy crop and could be utilized as a tool for early selection of new genotypes.
