Impact of 5-Flurouracil loaded chitosan nanoparticle on A375 and A431 cell line for the therapy of skin cancer

Skin cancer is an uncontrolled proliferation of abnormal skin cells that is usually caused by damaging UV radiation. Skin cancer is classified into two types: non-melanoma skin cancer and melanoma skin cancer. The entire studies focus on cytotoxic effect of prepared 5-Flurouracil loaded chitosan nanoparticle using A375 and A431 skin cancer cell line. In-vitro cell cytotoxicity assay, DAPI test of prepared formulation was conducted to observe the cytotoxic effect. The cell cytotoxicity investigation revealed that nanoparticles had higher cytotoxicity, inducing greater apoptosis within 72 h. The experimental results show that the produced nanoparticle is a good candidate for use as a 5-FU carrier in the treatment of skin cancer.

Dermal Papilla Cells: From Basic Research to Translational Applications.

As an appendage of the skin, hair protects against ultraviolet radiation and mechanical damage and regulates body temperature. It also reflects an individual's health status and serves as an important method of expressing personality. Hair loss and graying are significant psychosocial burdens for many people. Hair is produced from hair follicles, which are exclusively controlled by the dermal papilla (DP) at their base. The dermal papilla cells (DPCs) comprise a cluster of specialized mesenchymal cells that induce the formation of hair follicles during early embryonic development through interaction with epithelial precursor cells. They continue to regulate the growth cycle, color, size, and type of hair after the hair follicle matures by secreting various factors. DPCs possess stem cell characteristics and can be cultured and expanded in vitro. DPCs express numerous stemness-related factors, enabling them to be reprogrammed into induced pluripotent stem cells (iPSCs) using only two, or even one, Yamanaka factor. DPCs are an important source of skin-derived precursors (SKPs). When combined with epithelial stem cells, they can reconstitute skin and hair follicles, participating in the regeneration of the dermis, including the DP and dermal sheath. When implanted between the epidermis and dermis, DPCs can induce the formation of new hair follicles on hairless skin. Subcutaneous injection of DPCs and their exosomes can promote hair growth. This review summarizes the in vivo functions of the DP; highlights the potential of DPCs in cell therapy, particularly for the treatment of hair loss; and discusses the challenges and recent advances in the field, from basic research to translational applications.

Potential for photosynthesis on Mars within snow and ice

On Earth, solar radiation can transmit down to multiple metres within ice, depending on its optical properties. Organisms within ice can harness energy from photosynthetically active radiation while being protected from damaging ultraviolet radiation. On Mars, the lack of an effective ozone shield allows ~30% more damaging ultraviolet radiation to reach the surface in comparison with Earth. However, our radiative transfer modelling shows that despite the intense surface ultraviolet radiation, there are radiatively habitable zones within exposed mid-latitude ice on Mars, at depths ranging from a few centimetres for ice with 0.01–0.1% dust, and up to a few metres within cleaner ice. Numerical models predict that dense dusty snow in the martian mid-latitudes can melt below the surface at present. Thus, if small amounts of liquid water are available at these depths, mid-latitude ice exposures could represent the most easily accessible locations to search for extant life on Mars.

Temperature and ultraviolet radiation on a high mountain daphnid: When do interactions become lethal to highly adapted populations?

ABSTRACT Aquatic organisms from high mountain lakes experience extreme influences of climate and solar radiation, especially ultraviolet radiation (UVR), manifested through alterations in physiology, life history, and phenology. Zooplankton, a pivotal component of lake ecosystems, is particularly sensitive to changes in environmental conditions because of their short life cycles and diminished mobility. In the high mountains of the tropical Andes, zooplankton populations are adapted to low temperatures and high UVR because most of the Andean lakes are >4000 m a.s.l. This study focuses on Daphnia pulicaria populations originating from high-mountain Andean lakes, investigating their responses to different temperatures and UVR exposure by means of lab experiments. Key findings indicate that temperature is the most important factor impacting population variables, with high temperatures and UVR exposure leading to unfavorable population outcomes; however, physiological variables (cell viability and pigmentation, measured as melanin) are similarly influenced by temperature and UVR exposure. Pigmentation increases with temperature and is accentuated by solar radiation, indicating an adaptive response to mitigate UVR damage. Conversely, cellular viability declines with elevated temperatures and UVR exposure, showing that higher temperatures may offset the protective effects of pigmentation. Overall, these findings underscore the vulnerability of daphnid populations in high-mountain Andean systems to anticipated climate change impacts, with potential consequences for ecological dynamics in these critical ecosystems. More importantly, they show the importance of studying temperature and UVR (and probably other environmental conditions) as interacting variables because the results will dramatically differ if each factor is considered separately.

Does pigmentation provide protection to bdelloid rotifers in a high ultraviolet B environment?

AbstractAquatic species found in habitats with limited shade and little dissolved organic carbon (DOC) have increased vulnerability to ultraviolet radiation (UVR) damage. Pigmentation is a common mechanism used by animals for protection from UVR. A pigmented bdelloid rotifer, Philodina, occurs in high densities in shallow rock pools in El Paso Co., TX, and is subject to repeated desiccation and high UVR. To understand the roles of DOC, pigmentation, and dormancy in reducing the effects of UVR exposure in these rotifers: (1) DOC levels in rock pools were measured before and after the summer monsoon season and (2) hydrated or dormant bdelloids (desiccated for 0, 1, 7, or 32 d) that differed in degree of pigmentation (highly, moderately, lightly, and none) were exposed to three intensities of UVB radiation (low, mid, or high) and monitored for survival after 48 h. Pigmented bdelloids were found in rock pools with lower DOC concentrations. Logistic regression analysis indicated that pigmentation level, desiccation time, and UVB intensity all affected survival. Bdelloids in the dormant form for 1 d were more resistant to UVB exposure at all pigmentation levels. However, as desiccation time increased, the odds of surviving decreased. Hydrated highly pigmented bdelloids were three times more likely to survive desiccation, UVB radiation, and their combined effects. Prolonged periods of drought due to the changing climate will alter DOC concentrations, causing photoprotection to become an increasingly important survival strategy for aquatic invertebrates, especially those inhabiting shallow waters.

The Skin's Sun Story: Risks and Precautions

Background: The skin, representing the largest organ in the human body, serves numerous funtions crucial for homeostasis, as well as acts as the primary physical barrier protecting the organism from external environmental factors such as microorganisms, mechanical damage, heat or cold exposure, toxins, as well as solar radiation, posing various risks to the skin, including sunburn, photoageing and skin malignancies. Objectives: This study aims to explore the effects of solar radiation on the skin and to discuss preventive measures against its negative influence on the skin. Methods: An exhaustive search of references related to sun exposure and skin published in PubMed was undertaken, using the search terms: "solar exposure and skin, ultraviolet radiation and skin, visible light and skin, sunscreen, photoprotection”. Conclusions: Solar radiation, particularly ultraviolet radiation (UVR), induces acute and chronic damage to the skin, leading to conditions such as sunburn, photoaging, and skin malignancies. However, adequate exposure to sunlight is crucial for the synthesis of vitamin D3, which plays essential roles in calcium homeostasis, immune function, and potentially other physiological processes. Strategies such as proper sunscreen use and other photoprotection methods can mitigate the adverse effects of solar radiation while allowing for sufficient vitamin D synthesis.

A review of plant phenolics and endozoochory.

Phenolic compounds (phenolics) are secondary metabolites ubiquitous across plants. The earliest phenolics are linked to plants' successful transition from an aquatic to a terrestrial environment, serving as protection against damaging ultraviolet (UV) radiation, and as antioxidants to reduce oxidative stress in an atmosphere with an increasingly high O2:CO2 ratio. In modern plants, phenolics are best known for the defense against fungal and bacterial pathogens and as antifeedants that deter herbivory. Phenolics also play a role in seed dormancy, delaying germination, and lengthening viability in the seed bank. Many plants' seeds are endozoochorous - dispersed by animals, like birds, who eat and later excrete the seeds. Plants send visual signals to attract birds with UV-sensitive (UVS) vision for pollination and seed dispersal. As fruits ripen, antioxidant activity and phenolic content decrease. The waxy cuticle of fruits increases in UV reflection as phenolic rings, which absorb UV light, degrade. The UV contrast that birds detect may act as an honest signal, indicating nutritional changes in the fruit. However, there is little evidence to support the evolution of UV coloration during ripening being driven by frugivore selection. Antioxidant properties of fruit phenolics may be dually adaptive in plants and avian frugivores.

Molecular mechanisms of the effects of mulberry (Morus Rubra) extracts on UV-radiation and oxidative stress-induced skin cell damage

Solar ultraviolet (UV) radiation, while beneficial for the homeostasis of human skin at proper doses, causes skin cell damage under extreme conditions, with a result of photoaging and skin cancer. With the depletion of the ozone layer, the prevalence of UV-induced skin cancer including melanoma has increased dramatically worldwide. Unfortunately, that poses a public health challenge for almost all countries. Over the past two decades, we have been working extensively on understanding the cellular and molecular mechanisms through which UV radiation induces skin aging and skin cancer. We have observed that UV radiation activates cell surface receptors, more pronouncedly EGFR, leading to a cascade of activation of MAP kinase pathways, with a result of MMP activation and in turn matrix degradation, and apoptosis. We have also found that UV radiation induces the downregulation of water channel aquaporins and skin cell dehydration. This novel discovery together with other reports has resulted in the emergence of numerous skin care products. Moreover, UV radiation induces the generation of reactive oxygen species, which exacerbates skin cell damage. With this important line of discovery, more skin care products with added antioxidants have been developed. Mulberries ( Morus Rubra) have been known to be rich in antioxidants and applied to cosmetic products in Asian countries with reported effcacy although the cellular and molecular mechanisms of its actions remain elusive. The objective of this study was to investigate these mechanisms from various perspectives. We hypothesize that the mulberry extracts affect cell signaling pathways that lead to protection against UV radiation or oxidative stressed-induced cell damage. First, we extracted the antioxidants with methanol from two types of mulberries, red and white. Human skin keratinocytes (HaCaT cells) were treated with UV radiation and hydrogen peroxide (H2O2), with and without mulberry extracts. Confocal microscopy and Western blot analysis were employed to measure various cell signaling components. The results showed that mulberry extracts protect against UV radiation and H2O2 induced cell death, with red extracts showing better protection than white extracts. Mulberry extracts attenuate H2O2-induced mitochondrial activity as tested by MitoTracker™ Red CMXRos. Our previous studies have shown that UV radiation induces mTOR pathway activation evidenced by elevation of phosphorylated S6. Interestingly, in this study, Western blot analysis data indicated that mulberry extracts potentiate UV-induced S6 phosphorylation and H2O2-induced MEK activation. Furthermore, confocal data demonstrated that mulberry extract pretreatment inhibits IL1β induced NFκB p65 translocation from the cytoplasm to the nucleus. Taken all together, our data suggest that mulberry extracts containing abundant antioxidants affect MAPK kinase and mTOR pathways and modulate cell signaling pathways associated with skin cell survival and inflammation under UV radiation and oxidative stress. Lapagyl mitigates UV-induced inflammation and immunosuppression via Foxp3+ Tregs and CCL pathway: A single-cell transcriptomics study

BackgroundAs the largest organ of the body, the skin is constantly subjected to ultraviolet radiation (UVR), leading to inflammations and changes that mirror those seen in chronological aging. Although various small molecule drugs have been explored for treating skin photoaging, they typically suffer from low stability and a high incidence of adverse reactions. Consequently, the continued investigation of photoaging treatments, particularly those utilizing herbal products, remains a critical clinical endeavor. One such herbal product, Lapagyl, is derived from the bark of the lapacho tree and possesses antioxidant efficacies that could be beneficial in combating skin photoaging. PurposeThis research aimed to evaluate the efficacy of the herbal product Lapagyl in combating UVR-induced skin photoaging. Additionally, it sought to unravel the mechanisms by which Lapagyl promotes the regeneration of the skin extracellular matrix. MethodsTo investigate whether Lapagyl can alleviate skin aging and damage, a UVR radiation model was established using SKH-1 hairless mice. The dorsal skins of these mice were evaluated for wrinkle formation, texture, moisture, transepidermal water loss (TEWL), and elasticity. Pathological assessments were conducted to determine Lapagyl's efficacy. Additionally, single-cell sequencing and spectrum analysis were employed to elucidate the working mechanisms and primary components of Lapagyl in addressing UVR-induced skin aging and injury. ResultsLapagyl markedly reduced UVR-induced wrinkles, moisture loss, and elasticity decrease in SKH-1 mice. Single-cell sequencing demonstrated that Lapagyl corrected the imbalance in cell proportions caused by UVR, decreased UVR-induced ROS expression, and protected basal and spinous cells from skin damage. Additionally, Lapagyl effectively prevented the entry of inflammatory cells into the skin by reducing CCL8 expression and curtailed the UVR-induced formation of Foxp3+ regulatory T cells (Tregs) in the skin. Both pathological assessments and ex vivo skin model results demonstrated that Lapagyl effectively reduced UVR-induced damage to collagen and elastin. Spectrum analysis identified Salidroside as the primary compound remaining in the skin following Lapagyl treatment. Taken together, our study elucidated the skin protection mechanism of the herbal product Lapagyl against UVR damage at the cellular level, revealing its immunomodulatory effects, with salidroside identified as the primary active compound for skin. ConclusionOur study provided a thorough evaluation of Lapagyl's protective effects on skin against UVR damage, delving into the mechanisms at the cellular level. We discovered that Lapagyl mitigates skin inflammation and immunosuppression by regulating Foxp3+ Tregs and the CCL pathway. These insights indicate that Lapagyl has potential as a novel therapeutic option for addressing skin photoaging.

Protein expression of nucleolar protein 12 in the retina and its implication in protection of retina from UV irradiation damage

Nucleolar protein 12 (NOL12), one of the nucleolar proteins which are primarily expressed in the nucleolus and play key roles in RNA metabolism, cell proliferation, cell cycle, and cell survival, is widely expressed in various species and multiple organs. Although it has been reported that the mRNA of Drosophila NOL12 homolog viriato is expressed in the eyes of Drosophila, the protein expression of NOL12 in mammalian eyes remains to be elucidated. In this study, we showed through immunohistochemistry that NOL12 was present in the rat retina, with predominant distribution in the cytoplasm of the retinal neuronal cells. In the human retinoblastoma cell line WERI-Rb1, we found that altering NOL12 expression led to a change in WERI-Rb1 cell viability. Knocking down NOL12 expression decreased cell viability. In contrast, overexpressing NOL12 increased cell viability. Furthermore, increasing NOL12 expression inhibited ultraviolet (UV)-induced apoptosis. These findings demonstrated that NOL12 may play an important protective role in retinal cells. In the WERI-Rb1 cells exposed to UV irradiation, we detected that NOL12 was degraded, but this degradation could be attenuated by a pan-Caspase inhibitor. Notably, the inhibitory effect of NOL12 against UV-induced apoptosis could be restrained by increasing the expression of ATR serine/threonine kinase (ATR), a kinase that, when activated by severe DNA damage, can result in apoptosis. We also found that upregulating NOL12 inhibited the activation of ATR caused by UV irradiation. Additionally, inhibiting ATR activity reduced apoptosis resulting from both silencing NOL12 expression and UV exposure. Thus, NOL12 may protect against UV irradiation-induced retinal damage by inhibiting ATR activity.

Ginseng Extract Attenuates the Injury from Ultraviolet Irradiation for Female Drosophila melanogaster through the Autophagy Signaling Pathway.

Ginseng is an ancient medicinal and edible plant with many health benefits, and can serve as a drug and dietary supplement, but there are few relevant studies on its use to ease ultraviolet (UV) irradiation damage. After 0.8 mg/mL ginseng extract (GE) was added to the medium of female Drosophila melanogaster subjected to UV irradiation, the lifespan, climbing ability, sex ratio, developmental cycle, and antioxidant capacity of flies were examined to evaluate the GE function. In addition, the underlying mechanism by which GE enhances the irradiation tolerance of D. melanogaster was explored. With GE supplementation, female flies subjected to UV irradiation exhibited an extension in their lifespan, enhancement in their climbing ability, improvement in their offspring sex ratio, and restoration of the normal development cycle by increasing their antioxidant activity. Finally, further experiments indicated that GE could enhance the irradiation tolerance of female D. melanogaster by upregulating the gene expressions of SOD, GCL, and components of the autophagy signaling pathway. Finally, the performance of r4-Gal4;UAS-AMPKRNAi flies confirmed the regulatory role of the autophagy signaling pathway in mitigating UV irradiation injury.

Retinomorphic Photonic Synapses for Mimicking Ultraviolet Radiation Sensing and Damage Imaging

AbstractPhotonic synapses integrate sensing and processing within a single device, making them promising candidates for emulating the visual perception of a biological retina. Most retinomorphic photonic synapses achieve image pre‐processing by differentially responding to red, green, and blue (RGB). However, the impact of invisible ultraviolet (UV) light on the retina remains insufficiently explored, hindering the perception of the full spectrum by retinomorphic devices. Herein, a photonic synapse is constructed with strong UV absorption of Poly[(9,9‐dioctylfluorenyl‐2,7‐diyl)‐co‐(4,4′‐(N‐(4‐sec‐butylphenyl) diphenylamine)] (TFB) as photosensitive layer. The device exhibits synaptic properties of long‐term memory (LTM) and short‐term memory (STM) at different UV radiation stages. Extracting the electrical conductance of the device under UV radiation generates patterns of different resolutions, facilitating the simulation of retinal damage's impact on object perception. Moreover, an electronic eye system, incorporating the photonic synapse under UV radiation, successfully replicates pupil constriction and eye‐closing behaviors akin to those observed in a real biological eye. This result underscores the authenticity of retinomorphic devices, expending a novel concept for the future development of visuomorphic computing.

Transcriptome analysis in mouse skin after exposure to ultraviolet radiation from a canopy sunbed.

Exposure to ultraviolet radiation (UV-R), from both natural and artificial tanning, heightens the risk of skin cancer by inducing molecular changes in cells and tissues. Despite established transcriptional alterations at a molecular level due to UV-R exposure, uncertainties persist regarding UV radiation characterization and subsequent genomic changes. Our study aimed to mechanistically explore dose- and time-dependent gene expression changes, that may drive short-term (e.g., sunburn) and long-term actinic (e.g., skin cancer) consequences. Using C57BL/6N mouse skin, we analyzed transcriptomic expression following exposure to five erythemally weighted UV-R doses (0, 5, 10, 20, and 40 mJ/cm2) emitted by a UV-R tanning device. At 96 h post-exposure, 5 mJ/cm2 induced 116 statistically significant differentially expressed genes (DEGs) associated with structural changes from UV-R damage. The highest number of significant gene expression changes occurred at 6 and 48 h post-exposure in the 20 and 40 mJ/cm2 dose groups. Notably, at 40 mJ/cm2, 13 DEGs related to skin barrier homeostasis were consistently perturbed across all timepoints. UV-R exposure activated pathways involving oxidative stress, P53 signaling, inflammation, biotransformation, skin barrier maintenance, and innate immunity. This invivo study's transcriptional data offers mechanistic insights into both short-term and potential non-threshold-dependent long-term health effects of UV-R tanning.

Surface analysis of different wood polymer composites exposed to artificial weathering

This study presents a superficial analysis of different wood polymer composites (WPC) comprising polystyrene (WPSC), poly(furfuryl alcohol) (WPFAC), and pine resin (WPRC) when exposed to artificial weathering. The study investigates the influence of resin type on the composites, focusing on their chemical, colorimetric, morphological, and thermal properties. The main findings indicate WPSC exhibits poor resistance to weathering, allowing ultraviolet radiation damage on the lignocellulosic component of the composite, leading to oxidized groups formation. Conversely, WPFAC and WPRC demonstrate good wood protection capabilities by preventing significant deterioration of holocellulose and acting as filters for radiation and moisture. Moreover, WPFAC exhibits the lowest decrease in carbonyl index and O/C ratio, with reductions of 2.6 and 11.6%, respectively. These findings are significant as they demonstrate that poly(furfuryl alcohol) and pine resin can be used to produce WPCs that delay and mitigate wood degradation by weathering, contributing to the increased potential utilization of bioresins.

Formulation and Evaluation of Herbal Sunscreen Stick from Pyrostegia venusta (Ker Gawl.) Miers leaf ethanolic extract: A Modern Approach for Plants as Photo protectants

Plants are considered as one of the most important sources of medicines. Plants are also used in the preparation of cosmetics. Sunscreens are more prevalent for the protection of the human skin from harmful UV rays of the sun. Medicinal plant can be used in the preparation of effective sunscreens which are helpful for the protection of our skin as they do not have any side effects and can be used for any skin type. Sunscreen in the stick form is currently gaining notice due to its capacity to only cover a very limited surface area with each application. It has excellent water resistance capability, is simple to use, and creates a cohesive, durable protective film. This study is based on studying the SPF of Pyrostegia venusta plant and to develop a formulation for photoprotection. The physical characteristics of the leaf were studied. Extraction of the leaf was performed. The phytochemical analysis of the ethanolic extract was carried out to check the phytoconstituents. From this ethanolic extract the SPF was studied by In¬ vitro method from Mansur equation. A formulation was developed from the extract which was a sunscreen stick. The results showed presence of alkaloids, tannins, and flavonoids in the ethanolic extract. The SPF of the leaf was reported to be 10.84075 ± 0.02. The sunscreen stick formulation also showed satisfactory photoprotective activity. The study's findings lead us to the conclusion that the formulation could shield our skin from the damaging ultraviolet radiation and protect various skin diseases.

Surface Passivation by Quantum Exclusion: On the Quantum Efficiency and Stability of Delta-Doped CCDs and CMOS Image Sensors in Space.

Radiation-induced damage and instabilities in back-illuminated silicon detectors have proved to be challenging in multiple NASA and commercial applications. In this paper, we develop a model of detector quantum efficiency (QE) as a function of Si-SiO2 interface and oxide trap densities to analyze the performance of silicon detectors and explore the requirements for stable, radiation-hardened surface passivation. By analyzing QE data acquired before, during, and after, exposure to damaging UV radiation, we explore the physical and chemical mechanisms underlying UV-induced surface damage, variable surface charge, QE, and stability in ion-implanted and delta-doped detectors. Delta-doped CCD and CMOS image sensors are shown to be uniquely hardened against surface damage caused by ionizing radiation, enabling the stability and photometric accuracy required by NASA for exoplanet science and time domain astronomy.

Mechanical and UV protection performances of polylactic acid spunlace nonwoven fabrics coated by eco-friendly lignin/water-borne polyurethane composite coatings

Lignin as a biopolymer, has drawn much attention owing to its renewable nature, biodegradability, easy availability, and excellent stability. Water-borne polyurethane (WPU)-based coatings have been contributing to sustainable and eco-friendly practices offer a straightforward avenue for the value-added utilization of lignin. With the alarming rise in death rates attributed to skin diseases due to ultraviolet (UV) radiation damage, researchers have been actively searching for sustainable coating processes that enhance the UV protective capabilities of fabrics. In this paper, a facile strategy was reported for fabricating an environment friendly, mechanically durable, and UV protective biodegradable polylactic acid (PLA) spunlace nonwoven fabrics (SNFs) using lignin/WPU composite coatings. Lignin/WPU coating pastes were prepared using five different recipes including different lignin concentrations (0.5, 1.0, 1.5, and 2.0 wt.%) and coated on PLA SNFs via an applicator and thermally cured. Mechanical properties (e.g. tensile strength and abrasion resistance) and UV protection factor (UPF) values of fabrics were investigated. The coating films were characterized by Fourier Transform-Infrared (FTIR) spectroscopy and scanning electron microscopy (SEM) analysis. Lignin/WPU coated SNFs showed excellent UV protection performance and UPF values were obtained above 100 for all formulations. Lignin/WPU coatings provided an improving effect on the tensile strength and abrasion resistance performances of coated SNFs compared to uncoated SNFs. This work provides a simple and efficient strategy for preparing environmentally friendly lignin/WPU coatings, which are promising for application in the textile industry.

Long-term effects of embryonic exposure to benzophenone-3 on neurotoxicity and behavior of adult zebrafish

Benzophenone-3 (BP-3) is the most widely used ultraviolet filter (UV filter) in industries to avoid UV radiation damage. BP-3 is added to most sunscreens to protect the skin, hair, and lips from sun rays. It results in continuous discharge into aquatic environments, leading to aquatic biota and human's continuous exposure. Consequences of BP-3 exposure on the physiology and behavior of aquatic animals, mainly zebrafish, have been investigated, including their neurotoxic effects. However, little is known about its consequences in long-term developmental endpoints. This study aimed to investigate the long-term effects of embryonic BP-3 exposure on biomarkers of neurotoxicity in zebrafish. For this, we exposed embryos to 5, 10, and 20 μg∙L−1 BP-3 concentration and let fish grow to adulthood (5mpf). We evaluated anxiety-like behavior, social preference, aggressiveness, and enzymatic activity of the antioxidant defenses system and neurotoxic biomarkers (Glutathione S-transferase -GST, catalase -CAT, and acetylcholinesterase -AChE) in adult zebrafish. Enzymatic activities were also investigated in larvae immediately after BP-3 exposure. Animals early exposed to BP-3 presented anxiety-like behaviors and decreased social preference, but aggressiveness was not altered. In general, exposure to BP-3 leads to altered enzymatic activity, which persists into adulthood. GST activity increased in embryos and adults, while CAT activity decreased in both life stages. AChE activity enhanced only at the larval stage (96 hpf). The long-term behavioral and biochemical effects of BP-3 highlight the need for abolishing or restricting the compound from personal care products, which are continually disposed into the environment and threaten the biota and human health.

Screening and evaluation of skin potential probiotic from high-altitude Tibetans to repair ultraviolet radiation damage.

Human skin microbes play critical roles in skin health and diseases. Microbes colonizing on the skin of Tibetans living in the high-altitude area for generations may have a stronger ability to resist the harsh environment, such as high ultraviolet radiation (UV). Isolation of a potential probiotic from Tibetans skin is beneficial for resistance of skin disease for humans in the world. In this study, the signature microbiota for Tibetan skin were characterized compared to low-altitude humans. Next, using culture-omics, 118 species were isolated. The culturability of high-altitude of Tibetan skin microbiome reached approximate 66.8%. Next, we found that one strain, Pantoea eucrina, had the greatest ability to repair UV damage to the skin as the lowest pathological score was observed in this group. Interestingly, another animal trial found this bacterium resisted UV rather than its metabolites. Using whole genome sequencing, this strain P. eucrina KBFS172 was confirmed, and its functions were annotated. It might involve in the metabolic pathway of carotenoid biosynthesis with anti-oxidative stress properties, which plays critical roles in UV-damage repair. In conclusion, we characterized the signature microbes of skin in high-altitude Tibetans, isolated a skin bacterium of Pantoea eucrina KBFS172 which could repair UV damage via involving the metabolic pathway of carotenoid biosynthesis. Our results provide a new potential skin probiotic for skin disease prevention or sunburn.
