Metabolomic Profile of the Healthy Eating Index-2015 in the Multiethnic Study of Atherosclerosis

BackgroundPoor diet quality is a risk factor for type 2 diabetes and cardiovascular disease. However, knowledge of metabolites marking adherence to Dietary Guidelines for Americans (2015 version) are limited. ObjectivesThe goal was to determine a pattern of metabolites associated with the Healthy Eating Index (HEI)-2015, which measures adherence to the Dietary Guidelines for Americans. MethodsThe analysis examined 3557 adult men and women from the longitudinal cohort Multiethnic Study of Atherosclerosis (MESA), without known cardiovascular disease and with complete dietary data. Fasting serum specimens and diet and demographic questionnaires were assessed at baseline. Untargeted 1H 1-dimensional nuclei magnetic resonance spectroscopy (600 MHz) was used to generate metabolomics and lipidomics. A metabolome-wide association study specified each spectral feature as outcomes, HEI-2015 score as predictor, adjusting for age, sex, race, and study site in linear regression analyses. Subsequently, hierarchical clustering defined the discrete groups of correlated nuclei magnetic resonance features associated with named metabolites, and the linear regression analysis assessed for associations with HEI-2015 total and component scores. ResultsThe sample included 50% women with an mean age of 63 years, with 40% identifying as White, 23% as Black, 24% as Hispanic, and 13% as Chinese American. The mean HEI-2015 score was 66. The metabolome-wide association study identified 179 spectral features significantly associated with HEI-2015 score. The cluster analysis identified 7 clusters representing 4 metabolites; HEI-2015 score was significantly associated with all. HEI-2015 score was associated with proline betaine [β = 0.12 (SE = 0.02); P = 4.70 × 10−13] and was inversely related to proline [β = −0.13 (SE = 0.02); P = 4.45 × 10−14], 1,5 anhydrosorbitol [β = −0.08 (SE = 0.02); P = 4.37 × 10−7] and unsaturated fatty acyl chains [β = 0.08 (SE = 0.02); P = 8.98 × 10−7]. Intake of total fruit, whole grains, and seafood and plant proteins was associated with proline betaine. ConclusionsDiet quality is significantly associated with unsaturated fatty acyl chains, proline betaine, and proline. Further analysis may clarify the link between diet quality, metabolites, and pathogenesis of cardiometabolic disease.

Divergent unsaturated fatty acid synthesis in two highly related model pseudomonads.

The genomes of the best-studied pseudomonads, Pseudomonas aeruginosa and Pseudomonas putida, which share 85% of the predicted coding regions, contain a fabA fabB operon (demonstrated in P. aeruginosa, putative in P. putida). The enzymes encoded by the fabA and fabB genes catalyze the introduction of a double bond into a 10-carbon precursor which is elongated to the 16:1Δ9 and 18:1Δ11 unsaturated fatty acyl chains required for functional membrane phospholipids. A detailed analysis of transcription of the P. putida fabA fabB gene cluster showed that fabA and fabB constitute an operon and disclosed an unexpected and essential fabB promoter located within the fabA coding sequence. Inactivation of the fabA fabB operon fails to halt the growth of P. aeruginosa PAO1 but blocks growth of P. putida F1 unless an exogenous unsaturated fatty acid is provided. We report that the asymmetry between these two species is due to the P. aeruginosa PAO1 desA gene which encodes a fatty acid desaturase that introduces double bonds into the 16-carbon acyl chains of membrane phospholipids. Although P. putida F1 encodes a putative DesA homolog that is 84% identical to the P. aeruginosa PAO1, the protein fails to provide sufficient unsaturated fatty acid synthesis for growth when the FabA FabB pathway is inactivated. We report that the P. putida F1 DesA homolog can functionally replace the P. aeruginosa DesA. Hence, the defect in P. putida F1 desaturation is not due to a defective P. putida F1 DesA protein but probably to a weakly active component of the electron transfer process.

Oxidative lipidomics to elucidate the non-volatile derivatives of four typical triglycerides in vegetable oils under simulated frying conditions

Vegetable oils with different saturations have varied composition of triglycerides (TGs) and produce different non-volatile derivatives during oxidation. Precise characterization of the non-volatile derivatives of TGs is essential for understanding the degradation of TGs and the production pattern of non-volatile derivatives. Oxidative lipidomics was combined with collision-induced dissociation and electron-activated dissociation to elucidate the precise structures of non-volatile derivatives produced under simulated frying conditions by 1,3-dipalmitoyl-2-oleoylglycerol (POP), triolein (OOO), trilinolein (LLL), and trilinolenin (LnLnLn). The results indicate that the unsaturated fatty acyl chains at the sn-2 position were more susceptible to oxidation compared with those at the sn-1/3 position. Species of non-volatile derivatives included epoxy-, hydroperoxy-, hydroxy-, and oxo-TGs, as well as degradation products. The potential reaction pathways of TGs and their non-volatile derivatives were also proposed. This study elucidated oxidative degradation mechanisms of the four typical TGs and provided a theoretical basis for changes of vegetable oils during frying.

Structure Analysis of Unsaturated Polymyxin E Components Based on High-Performance Liquid Chromatography - Quadrupole/ Time of Flight Tandem Mass Spectrometry and Photochemical Reaction

Background: Polymyxin E (PME), which is a complex of cationic cyclic lipodecapeptides, is used to treat multidrug-resistant gram-negative bacterial infections. Besides the main components PME1 and PME2, polymyxin containing unsaturated fatty acyl (FA) group with lower contents is hardly to determine the structure without chromatographic preparations and NMR. Introduction: The peptide sequences of PME components has been carried out based on high performance liquid chromatography-quadrupole / time-of-flight mass spectrometry (HPLC-Q/TOF-MS). However, the components with double bond on the FA, such as 2’, 3’-dehydro PME1, were difficult to be determined or easily misjudged by MS/MS. The transformation of such unsaturated components to be epoxidized components or di-hydroxylated components can promote the acquisition of more fragment ions in the MS/MS, so as to assist in judging the position of double bonds on FA. Methods: In this paper, the PME mixtures were dissolved in an equal proportion of 20% ACN aqueous solution and 2-acetylpyridine. The above PME solution was transferred to a quartz cuvette and irradiated with the ultraviolet lamp at 254 nm for 8h. The dehydro PME components were converted to be epoxy PMEs and dihydroxy PMEs. A fragmentation pathway of epoxidized components or di-hydroxylated components based on Q/TOF-MS/MS was proposed for the first time. Results: According to the characteristic ions of epoxidized components and di-hydroxylated components, 2’, 3’-epoxy PME1/E2 and 2’, 3’-dihydroxy PME1/E2 were confirmed. It can be inferred that the double bond is located at the 2’, 3’-position of FA. Conclusion: The structure of unsaturated PME component with double bond on the FA is elucidated by HPLC-Q/TOF-MS combined with photochemical reaction. This strategy is applicable to other lipopeptides containing unsaturated FA chain.

Effect of lipid saturation on the topology and oligomeric state of helical membrane polypeptides

Natural liquid crystalline membranes are made up of many different lipids carrying a mixture of saturated and unsaturated fatty acyl chains. Whereas in the past considerable attention has been paid to cholesterol content, the phospholipid head groups and the membrane surface charge the detailed fatty acyl composition was often considered less important. However, recent investigations indicate that the detailed fatty acyl chain composition has pronounced effects on the oligomerization of the transmembrane helical anchoring domains of the MHC II receptor or the membrane alignment of the cationic antimicrobial peptide PGLa. In contrast the antimicrobial peptides magainin 2 and alamethicin are less susceptible to lipid saturation. Using histidine-rich LAH4 designer peptides the high energetic contributions of lipid saturation in stabilizing transmembrane helical alignments are quantitatively evaluated. These observations can have important implications for the biological regulation of membrane proteins and should be taken into considerations during biophysical or structural experiments.

Positional Assignment of C-C Double Bonds in Fatty Acyl Chains of Intact Arsenosugar Phospholipids Occurring in Seaweed Extracts by Epoxidation Reactions.

Water-soluble diacyl arsenosugar phospholipids (As-PL) are natural products widespread in marine animals and algae, including the brown alga Undaria pinnatifida, also known as wakame. The systematic recognition of As-PL has been hampered by the lack of standard and of qualitative methods to establish the carbon-carbon double bond positions of unsaturated fatty acyl chains. Here, the epoxidation reaction of fatty acyl substituents of As-PL was carried out with high selectivity by meta-chloroperoxybenzoic acid and the C-C double bond localization was established by collision-induced dissociation of epoxidized species as deprotonated molecules, [epoM - H]-. Reversed-phase liquid chromatography (RPLC) separation and a sequential triple-stage MS (i.e., MS3) analysis of unsaturated and epoxidized As-PL were very helpful to characterize the carbon-carbon double bond locations of both sn-1 and sn-2 fatty acyl chains, starting from a diagnostic product ion pair with 16.0 Da mass difference. These results indicate that intact As-PL can be annotated in terms of fatty acyl chain composition and in terms of their C-C double bond position(s). Interestingly, hexadecenoic (16:1 Δ9) and octadecenoic (18:1 Δ9) along with octadecadienoic (18:2 Δ9,12) and octadecatrienoic (18:3 Δ9,12,15) were found to be the most abundant unsaturated fatty acyl chains of As-PL in the brown alga wakame, thus confirming it as a good source of essential fatty acids with a balanced ω6/ω3 ratio. Although the toxicity of As-including metabolites of algal As-PL is still a matter of debate and needs to be studied in more detail, the described approach can be exploited to assess if As-PL could contribute to the supply of essential fatty acids related to the use of algae as nutritious food.

Advances in the Structural Biology, Mechanism, and Physiology of Cyclopropane Fatty Acid Modifications of Bacterial Membranes.

Cyclopropane fatty acid (CFA) synthase catalyzes a remarkable reaction. The cis double bonds of unsaturated fatty acyl chains of phospholipid bilayers are converted to cyclopropane rings by transfer of a methylene moiety from S-adenosyl-L-methionine (SAM). The substrates of this modification are functioning membrane bilayer phospholipids. Indeed, in Escherichia coli the great bulk of phospholipid synthesis occurs during exponential growth phase, but most cyclopropyl synthesis occurs in early stationary phase. In vitro the only active methylene group acceptor substrate is phospholipid bilayers containing unsaturated fatty acyl chains.

The combination of RPLC-ESI-FTMS/MS and m-CPBA epoxidation for the location and geometry assignment of double bonds in unsaturated fatty acyl chains of drying oils

Abstract Oil painting is the process of painting with drying oils as binders. Such oils mainly consist of triacylglycerols characterized by high levels of unsaturated fatty acyl chains. For instance, poppy seed oils have received much attention for their use as oil paints since very ancient time. Upon exposure to oxygen, not only do drying oils harden to a densely cross-linked polymer network through radical reactions, but their fatty acids also interact with cations of some pigments forming the related carboxylate salts. These so-called metal soaps lead to severe degradation phenomena because they can migrate and aggregate, breaking through the surface of the paint. Since the fatty-acidome of a drying oil is involved in these processes, the current study is chiefly devoted to the identification of both free and total fatty acids in poppy seed oil by reversed-phase liquid chromatography coupled with high-resolution mass spectrometry by an electrospray ionization source (RPLC-ESI-FTMS). Total fatty acids (TFA) were investigated by enzymatic hydrolysis and comparisons were made between the free fatty acid (FFA) profile and the total one resulting from the digestion on the same poppy seed oil. A key component of our approach was the double bond (C=C) epoxidation reaction, which was carried out on unsaturated fatty acids by meta-chloroperoxybenzoic acid (m-CPBA). A chromatographic set up was developed to separate such oxidized lipids, that were identified as deprotonated molecules [epoM-H]− by accurate mass matching in full mass spectra. Their epoxide ring was subsequently cleaved upon tandem mass fragmentation to yield a pair of diagnostic product ions for C=C double bond localization. Interestingly, the double bond geometry (i.e., cis/trans) was also assigned based on the retention times of derivatized-FA. The present approach opens new avenues for the analysis of artworks whether affected by metal soap-related degradation phenomena.

Candidate Circulating Biomarkers of Spontaneous Miscarriage After IVF-ET Identified via Coupling Machine Learning and Serum Lipidomics Profiling.

Spontaneous miscarriage is a common pregnancy complication. Multiple etiologies have been proposed such as genetic aberrations, endocrinology disorder, and immunologic derangement; however, the relevance of circulating lipidomes to the specific condition remains unclear. In the present study, lipidomics profiling was examined on serum of women with spontaneous miscarriage after in vitro fertilization and embryo transfer (IVF-ET). Screening and analysis of differential lipid levels were conducted using a machine learning approach to verify the stability and validity of potential serum biomarkers. Seven lipid species presented significant differences between the abortion and term birth patients, including three types of sphingomyelins (SMs), two types of diglycerides (DGs), one phosphatidylcholine (PC), and one lysophosphatidylethanolamine (LPE). All the SMs presented with a fold change of > 1, while both the PC and LPE had a fold change of < 1. The DG containing two saturated fatty acyl chains was decreased, but that containing two unsaturated fatty acyl chains was increased in the miscarriage group compared to the control group. This study reveals the relevance of lipid profiles to spontaneous abortion after IVF-ET, providing potential biomarkers and therapeutic targets for the specific clinical scenario.

Farnesylation and lipid unsaturation are critical for the membrane binding of the C-terminal segment of G-Protein Receptor Kinase 1

Many proteins are modified by the covalent addition of different types of lipids, such as myristoylation, palmitoylation and prenylation. Lipidation is expected to promote membrane association of proteins. Visual phototransduction involves many lipid-modified proteins. The G-Protein-coupled receptor of rod photoreceptors, rhodopsin, is inactivated by G-Protein-coupled Receptor Kinase 1 (GRK1). The C-terminus of GRK1 is farnesylated and its truncation has been shown to result in a very high decrease of its enzymatic activity, most likely because of the loss of its membrane localization. Little information is available on the membrane binding of GRK1 as well as of most prenylated proteins. Measurements of the membrane binding of the non-farnesylated and farnesylated C-terminal segment of GRK1 were thus performed using lipids typical of those found in rod outer segment disk membranes. Their random coil secondary structure was determined using circular dichroism and infrared spectroscopy. The non-farnesylated C-terminal segment of GRK1 has no surface activity. In contrast, the farnesylated C-terminal segment of GRK1 shows a particularly strong binding to lipid monolayers bearing at least one unsaturated fatty acyl chain. No binding is observed in the presence of monolayers of saturated phospholipids, in agreement with the low affinity of farnesylated Ras proteins for lipids in the liquid-ordered state. Altogether, these data demonstrate that the farnesyl group of the C-terminal segment of GRK1 is mandatory for its membrane binding, which is favored by particular lipids or lipid mixtures. This information will also be useful for the understanding of the membrane binding of other prenylated proteins.

Reprogrammed lipid metabolism protects inner nuclear membrane against unsaturated fat.

SummaryThe cell nucleus is surrounded by a double membrane. The lipid packing and viscosity of membranes is critical for their function and is tightly controlled by lipid saturation. Circuits regulating the lipid saturation of the outer nuclear membrane (ONM) and contiguous endoplasmic reticulum (ER) are known. However, how lipid saturation is controlled in the inner nuclear membrane (INM) has remained enigmatic. Using INM biosensors and targeted genetic manipulations, we show that increased lipid unsaturation causes a reprogramming of lipid storage metabolism across the nuclear envelope (NE). Cells induce lipid droplet (LD) formation specifically from the distant ONM/ER, whereas LD formation at the INM is suppressed. In doing so, unsaturated fatty acids are shifted away from the INM. We identify the transcription circuits that topologically reprogram LD synthesis and identify seipin and phosphatidic acid as critical effectors. Our study suggests a detoxification mechanism protecting the INM from excess lipid unsaturation.

Mass Spectrometry Imaging of Lipids with Isomer Resolution Using High-Pressure Ozone-Induced Dissociation.

Mass spectrometry imaging (MSI) of lipids within tissues has significant potential for both biomolecular discovery and histopathological applications. Conventional MSI technologies are, however, challenged by the prevalence of phospholipid regioisomers that differ only in the location(s) of carbon–carbon double bonds and/or the relative position of fatty acyl attachment to the glycerol backbone (i.e., sn position). The inability to resolve isomeric lipids within imaging experiments masks underlying complexity, resulting in a critical loss of metabolic information. Herein, ozone-induced dissociation (OzID) is implemented on a mobility-enabled quadrupole time-of-flight (Q-TOF) mass spectrometer capable of matrix-assisted laser desorption/ionization (MALDI). Exploiting the ion mobility region in the Q-TOF, high number densities of ozone were accessed, leading to ∼1000-fold enhancement in the abundance of OzID product ions compared to earlier MALDI-OzID implementations. Translation of this uplift into imaging resulted in a 50-fold improvement in acquisition rate, facilitating large-area mapping with resolution of phospholipid isomers. Mapping isomer distributions across rat brain sections revealed distinct distributions of lipid isomer populations with region-specific associations of isomers differing in double bond and sn positions. Moreover, product ions arising from sequential ozone- and collision-induced dissociation enabled double bond assignments in unsaturated fatty acyl chains esterified at the noncanonical sn-1 position.

Correcting cis-trans-transgressions in macromolecular structure models.

Many macromolecular X-ray and cryo-EM structure models deposited in the PDB contain biologically relevant small molecule ligands with unsaturated fatty acid acyl chains, whose cis-trans stereochemistry is incorrect. The molecules are either not properly defined in their stereochemical restraint files, or the proper stereochemistry is neglected during model building. Often, the same molecules appear in deposited models in both isomeric configurations, one of which is almost always incorrect, and the use of the same moiety (HET) identifier and restraint files in model refinement is wrong. We present case studies of frequently occurring molecules and a compilation of identified cases of C-C=C-C cis-trans geometry in the deposited structure models. Full listings of cis/trans torsion angles are provided for models with commonly occurring molecules to assist identification and correction of cis-trans errors and prevent inadvertent use of incorrect models. Caveats for users, advice for modellers and suggestions for remediation efforts with a simple but effective restraint file modification are provided.

Studies on Three 1‐ O ‐acylglycerol‐3‐P Acyl transferases in Clostridium beijerinckii

The biosynthesis of phospholipids in bacteria begins with the acylation of sn-glycerol-3-P. The second step is the acylation of the resulting 1-O-acyl-glycerol-3-P catalyzed by PlsC to form phosphatidic acid. Clostridium beijerinckii has two genes annotated as plsC and a related third gene, plsD, of unknown function. We have used CRISPR-Cas9 to delete each of these genes and examined the effects on phospholipid chain compositions. Deletion of Cbei_5028 (plsC) had no effect on the composition of the acyl or alk-1-enyl chains of the lipids. Deletion of Cbei_1741 (plsC) resulted in polar lipids with fewer unsaturated chains in diacyl lipids and plasmalogens. Deletion of Cbei_0579 (plsD) resulted in longer chains in all the lipids examined. It appears that Cbei_1741 (PlsC) is selective for unsaturated fatty acyl chains and that Cbei_0579 (PlsD) is a 1-O-acyl-sn-glycerol-3-P acyltransferase selective for shorter acyl chains. Strain characterization experiments demonstrated that ∆Cbei_1741 mutant had slightly increased tolerance to butanol compared to the wild type, possibly due to the more saturated chains in diacyl lipids and plasmalogens in the cell membrane. More dramatically, ∆Cbei_1741 mutant grew much faster and produced 40% more butanol (with more glucose consumed) than the wild type in a regular batch fermentation (at 35 oC). Comparatively, both ∆Cbei_5028 and ∆Cbei_0579 mutant strains produced slightly lower butanol than wild type under this condition. Interestingly, ∆Cbei_0579 produced 40% more butanol than the wild type when the fermentation was conducted at 20 oC. While at higher temperature (40 oC), the butanol yield in ∆Cbei_0579 reached 0.32 g/g (33% higher than in wild type) although the final butanol titer was actually slightly lower than in wild type.

Lipidome-wide characterization of phosphatidylinositols and phosphatidylglycerols on C[dbnd]C location level

Phosphatidylglycerol (PG) and phosphatidylinositol (PI) are two essential classes of glycerophospholipids (GPs), playing versatile roles such as signalling messengers and lipid-protein interaction ligands in cell. Although a majority of PG and PI molecular species contain unsaturated fatty acyl chain(s), conventional tandem mass spectrometry (MS/MS) methods cannot discern isomers different in carbon-carbon double bond (CC) locations. In this work, we paired phosphate methylation with acetone Paternò–Büchi (PB) reaction, aiming to provide a solution for sensitive and structurally informative analysis of these two important classes of GPs down to the location of CC. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) workflow was established. Offline methylated PG or PI mixtures were subjected to hydrophilic interaction chromatographic separation, online acetone PB reaction, and MS/MS via collision-induced dissociation (CID) for CC location determination in positive ion mode. This method was sensitive, offering limit of identification at 5 nM for both PG and PI standards down to CC locations. On molecular species level, 49 PI and 31 PG were identified from bovine liver, while 61 PIs were identified from human plasma. This workflow also enabled ratiometric comparisons of CC location isomers (C18:1 Δ9 vs. Δ11) of a series of PIs from type 2 diabetes (T2D) plasma to that of normal plasma samples. PI 16:0_18:1 and PI 18:0_18:1 were found to exhibit significant changes in CC isomeric ratios between T2D and normal plasma samples. The above results demonstrate that the developed LC-PB-MS/MS workflow is applicable to different classes of lipids and compatible with other established lipid derivatization methods to achieve comprehensive lipid analysis.

Modifications of Membrane Phospholipids in Response to Extended Aging from Pork Loins

ObjectivesIt is well established that fresh meat shelf-life deteriorates during aging process. We hypothesize that part of the shelf-life reduction is due to membrane phospholipid deterioration through phospholipase activity and/or phospholipid oxidation during aging. Therefore, the objective of this study was to characterize the modifications/deterioration of phospholipid classes/species in pork loins from 3 different aging periods.Materials and MethodsLoins from 20 carcasses were collected at a commercial harvest facility in the Midwest 1 d postmortem from carcasses of Duroc sired crossbred pigs. Four chops from each carcass containing only the longissimus muscle were vacuum packaged and aged at 4°C for 1, 8, and 21 d. A sensitive approach based on electrospray ionization tandem mass spectrometry was used to comprehensively analyze phospholipid composition using the lipid extract from each sample at each aging period (n = 60). Unsaturation index (UI; measurement of the number of double bonds) for each phospholipid species class was also calculated to quantify fatty acyl chain unsaturation for each sample in each aging period.ResultsTotal phospholipid quantity in pork loins was not different between 1d and 8d aged chops but decreased significantly from 8d to 21d of aging (806.6 vs. 297.5 nmol phospholipid/mg lipid; P &lt; 0.01). On the other hand, the mol% data (distribution of each phospholipid species in relative % of total phospholipid) revealed that phosphatidylinositol (PI) and phosphatidylserine (PS) increased in mol% from 1d to 21d of aging in pork loins (P &lt; 0.01). This increase was mainly due to the increase of PI 38:4 (primarily 18:0/20:4) and PS 36:2 (primarily 18:0/18:2) between 1d and 21d samples (P &lt; 0.01). The results showed that phospholipid degradation products like lysophosphatidylcholine (LPC) mol% rose quickly after short term aging (8d) but remained constant through the rest of the 21d aging period (P &lt; 0.01). Conversely, lysophosphatidylethanolamine (LPE) was unaltered between 1d and 8d of aging but decreased between 8d and 21d aged pork loins (P &lt; 0.01). The mol% of phosphatidic acid (PA) also increased between 1d and 21d aged pork loins (P &lt; 0.05). Extended aging did not alter the mol% of total phosphatidylcholine (PC), ether-linked PC (ePC), sphingomyelin (SM), phosphatidylethanolamine (PE) or ether-linked PE (ePE; P &gt; 0.05). Surprisingly, UI revealed the exact opposite trend as the mol% data. The UI of PI and PS decreased (P &lt; 0.01) from 1d to 21 d of aging in pork loins due to the disappearance of many minor PI and PS species with very long chain fatty acids and multiple double bonds such as PI 42:10 and PS 44:10. There was also a slight increase of PC UI after 8 d of aging in pork loins (P &lt; 0.01). The UI for LPC, ePC, SM, LPE, PE, ePE and total phospholipid were not altered in any of the aging periods (P &gt; 0.05).ConclusionThese results confirmed our hypothesis that phospholipids undergo extensive degradation during aging. The data also indicated that the majority of phospholipids in pork loins may maintain integrity over short period aging (1–8d). Among the phospholipid classes, PI and PS were slightly more resistant to deterioration compared with the others due to their ability to modify fatty acyl chain saturation. Additional investigations are necessary to define the role of phospholipid modifications in fresh pork shelf-life and flavor.

Toward Complete Structure Elucidation of Glycerophospholipids in the Gas Phase through Charge Inversion Ion/Ion Chemistry.

Shotgun lipidomics has recently gained popularity for lipid analysis. Conventionally, shotgun analysis of glycerophospholipids via direct electrospray ionization tandem mass spectrometry (ESI-MS/MS) provides glycerophospholipid (GPL) class (i.e., headgroup composition) and fatty acyl composition. Reliant on low-energy collision-induced dissociation (CID), traditional ESI-MS/MS fails to define fatty acyl regiochemistry along the glycerol backbone or carbon-carbon double bond position(s) in unsaturated fatty acyl substituents. Therefore, isomeric GPLs are often unresolved, representing a significant challenge for shotgun-MS approaches. We developed a top-down shotgun-MS method utilizing gas-phase ion/ion charge inversion chemistry that provides near-complete GPL structural identification. First, in negative ion mode, CID of mass-selected GPL anions generates fatty acyl carboxylate anions via fragmentation of ester bonds linking the fatty acyl substituents at the sn-1 and sn-2 positions of the glycerol backbone. Product anions, including fatty acyl carboxylate ions, were then derivatized in the mass spectrometer via an ion/ion charge inversion reaction with tris-phenanthroline magnesium dications. Subsequent CID of charge-inverted fatty acyl complex cations yielded isomer-specific product ion spectra that permit (i) unambiguous assignment of carbon-carbon double bond position(s) and (ii) relative quantitation of isomeric fatty acyl substituents. The outlined strategy was applied to the analysis of targeted GPLs extracted from human plasma, including several proposed plasma biomarkers. A single experiment thus facilitates assignment of the GPL headgroup, fatty acyl composition, carbon-carbon double bond position(s) in unsaturated fatty acyl chains, and, in some cases, fatty acyl sn-position and relative abundances for isomeric fatty acyl substituents. Ultimately, this MSn platform paired with ion/ion chemistry permitted identification of major, and some minor, isomeric contributors that are unresolved using conventional ESI-MS/MS.

Stearoyl-CoA Desaturase 1 Protects Ovarian Cancer Cells from Ferroptotic Cell Death

Activation of ferroptosis, a recently described mechanism of regulated cell death, dramatically inhibits growth of ovarian cancer cells. Given the importance of lipid metabolism in ferroptosis and the key role of lipids in ovarian cancer, we examined the contribution to ferroptosis of stearoyl-CoA desaturase (SCD1, SCD), an enzyme that catalyzes the rate-limiting step in monounsaturated fatty acid synthesis in ovarian cancer cells. SCD1 was highly expressed in ovarian cancer tissue, cell lines, and a genetic model of ovarian cancer stem cells. Inhibition of SCD1 induced lipid oxidation and cell death. Conversely, overexpression of SCD or exogenous administration of its C16:1 and C18:1 products, palmitoleic acid or oleate, protected cells from death. Inhibition of SCD1 induced both ferroptosis and apoptosis. Inhibition of SCD1 decreased CoQ10, an endogenous membrane antioxidant whose depletion has been linked to ferroptosis, while concomitantly decreasing unsaturated fatty acyl chains in membrane phospholipids and increasing long-chain saturated ceramides, changes previously linked to apoptosis. Simultaneous triggering of two death pathways suggests SCD1 inhibition may be an effective component of antitumor therapy, because overcoming this dual mechanism of cell death may present a significant barrier to the emergence of drug resistance. Supporting this concept, we observed that inhibition of SCD1 significantly potentiated the antitumor effect of ferroptosis inducers in both ovarian cancer cell lines and a mouse orthotopic xenograft model. Our results suggest that the use of combined treatment with SCD1 inhibitors and ferroptosis inducers may provide a new therapeutic strategy for patients with ovarian cancer. SIGNIFICANCE: The combination of SCD1 inhibitors and ferroptosis inducers may provide a new therapeutic strategy for the treatment of ovarian cancer patients.See related commentary by Carbone and Melino, p. 5149.

Crystal structure of bacterial cyclopropane-fatty-acyl-phospholipid synthase with phospholipid.

The lipids containing cyclopropane-fatty-acid (CFA) protect bacteria from adverse conditions such as acidity, freeze-drying desiccation and exposure to pollutants. CFA is synthesized when cyclopropane-fatty-acyl-phospholipid synthase (CFA synthase, CFAS) transfers a methylene group from S-adenosylmethionine (SAM) across the cis double bonds of unsaturated fatty acyl chains. Here, we reported a 2.7-Å crystal structure of CFAS from Lactobacillus acidophilus. The enzyme is composed of N- and C-terminal domain, which belong to the sterol carrier protein and methyltransferase superfamily, respectively. A phospholipid in the substrate binding site and a bicarbonate ion (BCI) acting as a general base in the active site were discovered. To elucidate the mechanism, a docking experiment using CFAS from L. acidophilus and SAM was carried out. The analysis of this structure demonstrated that three groups, the carbons from the substrate, the BCI and the methyl of S(CHn)3 group, were close enough to form a cyclopropane ring with the help of amino acids in the active site. Therefore, the structure supports the hypothesis that CFAS from L. acidophilus catalyzes methyl transfer via a carbocation mechanism. These findings provide a structural basis to more deeply understand enzymatic cyclopropanation.

Modifications of Membrane Phospholipids in Response to Extended Aging from Pork Loins

ObjectivesIt is well established that fresh meat shelf-life deteriorates during aging process. We hypothesize that part of the shelf-life reduction is due to membrane phospholipid deterioration through phospholipase activity and/or phospholipid oxidation during aging. Therefore, the objective of this study was to characterize the modifications/deterioration of phospholipid classes/species in pork loins from 3 different aging periods.Materials and MethodsLoins from 20 carcasses were collected at a commercial harvest facility in the Midwest 1 d postmortem from carcasses of Duroc sired crossbred pigs. Four chops from each carcass containing only the longissimus muscle were vacuum packaged and aged at 4°C for 1, 8, and 21 d. A sensitive approach based on electrospray ionization tandem mass spectrometry was used to comprehensively analyze phospholipid composition using the lipid extract from each sample at each aging period (n = 60). Unsaturation index (UI; measurement of the number of double bonds) for each phospholipid species class was also calculated to quantify fatty acyl chain unsaturation for each sample in each aging period.ResultsTotal phospholipid quantity in pork loins was not different between 1d and 8d aged chops but decreased significantly from 8d to 21d of aging (806.6 vs. 297.5 nmol phospholipid/mg lipid; P < 0.01). On the other hand, the mol% data (distribution of each phospholipid species in relative % of total phospholipid) revealed that phosphatidylinositol (PI) and phosphatidylserine (PS) increased in mol% from 1d to 21d of aging in pork loins (P < 0.01). This increase was mainly due to the increase of PI 38:4 (primarily 18:0/20:4) and PS 36:2 (primarily 18:0/18:2) between 1d and 21d samples (P < 0.01). The results showed that phospholipid degradation products like lysophosphatidylcholine (LPC) mol% rose quickly after short term aging (8d) but remained constant through the rest of the 21d aging period (P < 0.01). Conversely, lysophosphatidylethanolamine (LPE) was unaltered between 1d and 8d of aging but decreased between 8d and 21d aged pork loins (P < 0.01). The mol% of phosphatidic acid (PA) also increased between 1d and 21d aged pork loins (P < 0.05). Extended aging did not alter the mol% of total phosphatidylcholine (PC), ether-linked PC (ePC), sphingomyelin (SM), phosphatidylethanolamine (PE) or ether-linked PE (ePE; P > 0.05). Surprisingly, UI revealed the exact opposite trend as the mol% data. The UI of PI and PS decreased (P < 0.01) from 1d to 21 d of aging in pork loins due to the disappearance of many minor PI and PS species with very long chain fatty acids and multiple double bonds such as PI 42:10 and PS 44:10. There was also a slight increase of PC UI after 8 d of aging in pork loins (P < 0.01). The UI for LPC, ePC, SM, LPE, PE, ePE and total phospholipid were not altered in any of the aging periods (P > 0.05).ConclusionThese results confirmed our hypothesis that phospholipids undergo extensive degradation during aging. The data also indicated that the majority of phospholipids in pork loins may maintain integrity over short period aging (1–8d). Among the phospholipid classes, PI and PS were slightly more resistant to deterioration compared with the others due to their ability to modify fatty acyl chain saturation. Additional investigations are necessary to define the role of phospholipid modifications in fresh pork shelf-life and flavor.
