Polystyrene microplastic attenuated the toxic effects of florfenicol on rice (Oryza sativa L.) seedlings in hydroponics: From the perspective of oxidative response, phototoxicity and molecular metabolism

Antibiotics and microplastics (MPs) are two emerging pollutants in agroecosystems, however the effects of co-exposure to antibiotics and MPs remain unclear. The toxicity of florfenicol (FF) and polystyrene microplastics (PS-MPs) on rice seedlings was investigated. FF and PS-MPs caused colloidal agglomeration, which changed the environmental behavior of FF. FF inhibited rice growth and altered antioxidant enzyme (superoxide dismutase, peroxidase, and catalase) activities, leading to membrane lipid peroxidation; impaired photosynthetic systems, decreased photosynthetic pigments (Chlorophyll a, Chlorophyll b, and carotene), chlorophyll precursors (Proto IX, Mg-Proto IX, and Pchlide), photosynthetic and respiratory rates. The key photosynthesis related genes (PsaA, PsaB, PsbA, PsbB, PsbC, and PsbD) were significantly down-regulated. The ultrastructure of mesophyll cells was destroyed with chloroplast swelling, membrane surface blurring, irregular thylakoid lamellar structure, and number of peroxisomes increased. PS-MPs mitigated FF toxicity, and the IBR index values showed that 10 mg∙L−1 PS-MPs were more effective. Metabolomic analysis revealed that the abundance of metabolites and metabolic pathways were altered by FF, was greater than the combined "MPs-FF" contamination. The metabolism of amino acids, sugars, and organic acids were severely interfered. Among these, 15 metabolic pathways were significantly altered, with the most significant effects on phenylalanine metabolism and the citric acid cycle (p < 0.05).

Effect of Waterlogging Stress on Leaf Anatomical Structure and Ultrastructure of Phoebe sheareri Seedlings

Phoebe sheareri is an excellent roadside tree with a wide distribution range and high ornamental value. Excessive moisture can affect the external morphology, the microstructure, and the ultrastructure of the leaf. Little is known at present regarding the leaf structure of P. sheareri under waterlogging stress. In this paper, the external morphology of leaves, the microstructure of leaf epidermis, and the ultrastructure of mesophyll cells of P. sheareri seedlings under waterlogging stress and drainage were dynamically observed. Waterlogging stress contributed to the yellowing and wilting of P. sheareri seedling leaves, the gradual closing of leaf epidermal stomata, increasing density of leaf stomata, gradual loosening of the arrangement of leaf cell structure, and merging of leaf palisade tissue cells. Waterlogging stress forced the structure of the chloroplast membranes to blur, gradually causing swelling, and deformation, with plasmolysis occurring in severe cases. During waterlogging, the basal lamellae were disorganized, and the mitochondrial membrane structure was damaged. The damaged state of the leaves was not relieved after drainage. Waterlogging stress not only inhibited the growth of leaves, but also accelerated the closure of stomata, disordered the arrangement of palisade tissue and spongy tissue gradually, and damaged the internal organelles of mesophyll cells.

Within-leaf chloroplasts and nitrogen allocation to thylakoids in relation to photosynthesis during grain filling in maize

Nitrogen (N) is an important contributor to photosynthetic rate (Pn). However, during grain-filling stage in maize, some leaf N is remobilized to meet demands for grain protein accumulation rather than photosynthetic demands. Therefore, plants that can maintain a relatively high Pn during the N remobilization process would have the key to achieving both high grain yields (HGY) and high grain protein concentrations (HGPC). In this study, we investigated two high-yielding maize hybrids in photosynthetic apparatus and N allocation in a two-year field experiment. During grain filling, XY335 had a higher Pn and photosynthetic N-use efficiency than ZD958 had in the upper leaf, but not in the middle or lower leaves. In the upper leaf, the diameter and area of the bundle sheath (BS) were larger and the distance between bundle sheaths was greater in XY335 than in ZD958. XY335 had more bundle sheath cells (BSCs) and a larger BSC area, as well as a larger chloroplast area in the BSC, resulting in a higher total number and total area of chloroplasts in the BS. XY335 also had higher stomatal conductance (gs), intercellular CO2 concentration, and N allocation to the thylakoids. No genotypic differences were found in mesophyll cell ultrastructure, N content and starch content in the three types of leaves. Therefore, a trifecta of higher gs, greater N allocation to thylakoids for photo-phosphorylation and electron transport, and more and larger chloroplasts promoting CO2 assimilation in the BS confers a high Pn to simultaneously achieve HGY and high HGPC in maize.

Exogenous brassinosteroids alleviate calcium deficiency-induced tip-burn by maintaining cell wall structural stability and higher photosynthesis in mini Chinese Cabbage.

Tip-burn has seriously affected the yield, quality and commodity value of mini Chinese cabbage. Calcium (Ca2+) deficiency is the main cause of tip-burn. In order to investigate whether exogenous brassinosteroids (BRs) can alleviate tip-burn induced by calcium (Ca2+) deficiency and its mechanism, in this study, Ca2+ deficiency in nutrient solution was used to induced tip-burn, and then distilled water and BRs were sprayed on leaves to observe the tip-burn incidence of mini Chinese cabbage. The tip-burn incidence and disease index, leaf area, fluorescence parameters (Fv/Fm, NPQ, qP andφPSII) and gas exchange parameters (Tr, Pn, Gs and Ci), pigment contents, cell wall components, mesophyll cell ultrastructure and the expression of genes related to chlorophyll degradation were measured. The results showed that exogenous BRs reduced the tip-burn incidence rate and disease index of mini Chinese cabbage, and the tip-burn incidence rate reached the highest on the ninth day after treatment. Exogenous BRs increased the contents of cellulose, hemifiber, water-soluble pectin in Ca2+ deficiency treated leaves, maintaining the stability of cell wall structure. In addition, BRs increased photosynthetic rate by increasing the activities of Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and fructose 1,6-bisphosphatase (FBPase) related to Calvin cycle, maintaining relatively complete chloroplast structure and higher chlorophyll content via down-regulating the expression of BrPPH1 and BrPAO1 genes related to chlorophyll degradation. In conclusion, exogenous BRs alleviated calcium deficiency-induced tip-burn by maintaining cell wall structural stability and higher photosynthesis.

Effects of Drought Stress On Growth, Photosynthesis And Ultrstructure of Mesophyll Cell of Ulmus Szechuanica Fang Seedlings

Effects of drought stress on growth, photosynthesis and ultrastructure of mesophyll cells of Ulmus szechuanica Fang seedlings were studied. Results showed that under drought stress both leaf water content (LWC) and relative water content (RWC) decreased significantly and height growth (GH) and base diameter growth (GD) declined. Pn, Tr, Gs, Ci and Ls were significantly affected by water deficit. With the continuing of drought, Pn decreased significantly, the change of Gs, Ci, Tr was same. There existed significant effects of drought on stomatal density (SD), stomatal length (SL), stomatal width (SW), individual stomatal area (As) and percentage of individual stomatal area (At) too. Under drought condition, change of structure and chloroplast morphology of the leaves was evident, the number of chloroplasts and orophil granules increased, their proportion in cells amplified. Effects of arbuscular mycorrhizal fungi on redox homeostasis of rice under Cd stress

Arbuscular mycorrhizal fungi (AMF) can enhance cadmium (Cd) tolerance and decrease Cd uptake of rice plant. However, in these processes, the essential role of reactive oxygen species (ROS)-antioxidants interactions is so far not investigated. We inoculated AMF, Funneliformis mosseae (Fm) and Rhizophagus intraradices (Ri), to upland rice subjected to 0, 2 or 10 mg Cd kg−1 in soil. ROS, antioxidants, chlorophyll, mesophyll cell ultrastructure, Cd concentration and Cd transporters were measured. Results showed that the levels of ROS (i.e., superoxide, hydrogen peroxide and lipid peroxidation) were mitigated in +AMF + Cd treatments, especially in +Ri + Cd, compared with −AMF + Cd. For antioxidants, lower levels of catalase, peroxidase and superoxide dismutase, whereas higher levels of glutathione and glutathione peroxidase were observed in +AMF + Cd compared with −AMF + Cd. Chlorophyll content and mesophyll cell ultrastructure were improved by AMF. Cd concentrations in plants were significantly decreased in +AMF + Cd (10 mg Cd kg−1) treatments. AMF not only decreased Cd accumulation in rice but also regulated the ROS scavenging activities. The variations of ROS and antioxidants were restrained to smaller ranges by AMF. AMF may assist in Cd restriction and thus helping the host to release glutathione for ROS scavenging.

Ameliorative effects of Lanthanum(Ⅲ) on Copper(Ⅱ) stressed rice (Oryza sativa) and its molecular mechanism revealed by transcriptome profiling

Rare earth elements are known to alleviate heavy metal stress. However, the potential mechanisms of the alleviation remain unclear. This study compared the effects of La(NO3)3 and La(NO3)3-amino acid chelates (La (Ⅲ)-AA) on growth, oxidative stress, ultrastructure, bioaccumulation and gene expression in rice. Results demonstrated that 20 mg/L La (Ⅲ)-AA can effectively ameliorate CuSO4 (50 mg/L) stress in rice by reducing oxidative stress and increasing chlorophyll content, thus promoting growth. ICP and TEM revealed an antagonistic effect between La (Ⅲ) and Cu(Ⅱ). Exogenous La (Ⅲ)-AA decreased Cu(Ⅱ) content in rice leaves, stems and roots by 55.56%, 59.46% and 26.29%, and ameliorated Cu(Ⅱ) damage by maintaining the ultrastructure of mesophyll cells. RNA sequencing identified 7020 differentially expressed genes, and 8 were validated by qRT-PCR. Indole-3-acetic acid (IAA) concentration was detected by HPLC. Correlation analysis between OsGH3.4-IAA-Expansin revealed that IAA content is negatively correlated with OsGH3.4 (r = −0.82, P < 0.05), and positively correlated with Expansin (r = 0.78, P < 0.05). It's assumed that La (Ⅲ)-induced OsGH3.4 could inhibit IAA-dependent Expansin expression, thereby conferring resistance to Cu stress. This work provides novel insights into the molecular basis underlying La (Ⅲ)-induced Cu(Ⅱ) tolerance in rice.

The effects of exogenous organic acids on the growth, photosynthesis and cellular ultrastructure of Salix variegata Franch. Under Cd stress

We studied the effects of three organic acids (citric acid, tartaric acid and malic acid) on the biomass, photosynthetic pigment content and photosynthetic parameters of Salix variegata under Cd stress and observed the ultrastructure of mesophyll cells in each treatment. Cd stress significantly reduced photosynthesis by reducing the content of pigments and disrupting chloroplast structure, which consequently decreased the biomass. However, respective addition of three organic acids greatly increased the biomass of S. variegata under Cd stress. Among them, the effect of malic acid or tartaric acid on shoot and total biomass accumulation was greater than that of citric acid. The alleviation of biomass probably related with the photosynthetic process. Results revealed that treatment with each organic acid enhanced the net photosynthesis rate under Cd stress. Malic acid promoted plant growth and biomass by increasing the chlorophyll content and mitigating damage to the photosynthetic apparatus resulting from Cd stress. Tartaric acid had little impact on the photosynthetic pigment content, but it was important in mitigating the ultrastructural damage of plants caused by Cd. Addition of citric acid significantly increased the carotenoid as well as the number and volume of chloroplasts in mesophyll cells, while the mitigation of structural damage in the photosynthetic apparatus was weaker than that in tartaric acid or malic acid treatment. It is concluded that application of tartaric acid or malic acid is effective in increasing the growth potential of S. variegata under Cd stress and thus can be a promising approach for the phytoremediation of Cd-contaminated soil.

Increasing soil age drives shifts in plant-plant interactions from positive to negative and affects primary succession dynamics in a subalpine glacier forefield

The stress gradient hypothesis predicts that plant-plant interactions switch between facilitation (positive) and competition (negative) along environmental gradients, with facilitation being more common under high abiotic stress conditions relative to more moderate abiotic stress conditions. Our aim was to reveal, whether the interactions between Populus purdomii Rehder and Salix rehderiana Schneider switch from positive to negative during the early stages of primary succession in the Gongga Mountain glacier retreat region. We also investigated, whether soil age is a major driving factor for the transformation of interactions between neighboring plants. We analyzed differences between intraspecific interactions and interspecific interactions of Populus and Salix under 20- and 40-year-old soil conditions, including plant biomass accumulation and allocation, nutrient absorption and utilization, relative competition intensity, non-structural carbohydrates, foliar carbon and nitrogen isotope composition, mesophyll cell ultrastructure, soil microbial biomass and community structure, extracellular enzyme activities, and soil organic carbon (SOC), soil total nitrogen (TN), soil ammonium (NH4+-N), and soil nitrate (NO3−-N) contents. We found that P. purdomii and S. rehderiana growing under interspecific interactions had greater contents of aboveground dry matter, belowground dry matter and total dry matter compared to intraspecific interactions in 20-year-old soil. Furthermore, in 40-year-old soil conditions, the phospholipid fatty acid (PLFA) analysis showed that Populus and Salix exposed to interspecific interactions exhibited lower amounts of gram-positive bacteria, fungi (18,1 ω9c) and actinomycetes, and lower levels of total PLFAs than those growing under intraspecific interactions. The redundancy analysis (RDA) results demonstrated that soil N was the most important parameter contributing to the composition of microbial communities. In addition, the 15N stable isotope labeling method showed that Populus and Salix growing under interspecific interactions had higher foliage δ15N derived from NO3− (δ15N-NO3−) than those growing under intraspecific interactions in 20-year-old soil. In summary, our results demonstrated that Populus-Salix interactions exhibited positive effects on survival in 20-year-old soil. Conversely, under 40-year-old soil conditions, Populus-Salix interactions presented negative effects in relation to nutrients and elimination by neighboring plants. Moreover, soil age is a major driving factor for plant-plant interactions that shift from positive to negative with an increasing soil age in the Gongga Mountain glacier forefield. In all, our results support the stress gradient hypothesis. Our findings improve understanding of plant-plant interactions and plant-soil feedbacks during the early stages of soil development, and of the construction of vegetation communities.

Specific Features of the Ultrastructure and Biochemical Composition of Triticum spelta L. Leaf Mesophile Cells in the Initial Period of Stress Temperature Action

Under controlled conditions, the effect of high (40°C, 2 h) and positive low (4°C, 2 h) temperatures on the ultrastructure of mesophyll cells of the leaf and the content of photosynthetic pigments, phenols, and flavonoids in 2-week-old Triticum spelta plants was studied. The ultrastructure of the mesophyll cells of the leaf of the control plants was typical: a developed thylakoid system was clearly seen that was immersed in a fine-grained stroma in the chloroplasts of regular lenticular shape. Short-term hyperthermia caused a partial destruction of thylakoid membranes. Wave-shaped packing of grana thylakoids, significant expansion of lumen intervals, disturbance of the structural bond between the grana thylacoids and stroma thylakoids was noted. With hyperthermia, the mitochondria noticeably “swelled,” while the cristae membranes became less contrasting. The number of lipid droplets increased in the cytoplasm of cells. In the leaves, the content of chlorophylls and carotenoids decreased, however, the number of common phenols and flavonoids increased. Short-term hypothermia caused intense formation of plastoglobules, and an increase in the number and size of starch grains. Destruction of thylakoid membranes was not observed. Some of the mitochondria were rounded (40%), with their size being close to the control values, and some organelles were lenticular, “dumbbell,” and “cup-shaped.” Under hyper- and hypothermia, the T. spelta leaf mesophyll cells showed a tendency to increase the degree of chromatin condensation in the nucleus. Under hypothermia, the content and ratio of chlorophylls and carotenoids in leaves did not differ much from the control plants, and no significant quantitative changes in the total phenols and flavonoids were recorded.

Effects of exogenous calcium on mesophyll cell ultrastructure, gas exchange, and photosystem II in tobacco (Nicotiana tabacum Linn.) under drought stress

The effects of calcium chloride solution (10 mmol L–1) on mesophyll cell ultrastructure, gas exchange, chlorophyll and carotenoid content, and PSII in tobacco leaf were studied by simulating water deficit conditions via treatment with 25% PEG-6000 for 24 h. The results showed that under drought stress, the mesophyll cell structure and morphology were destroyed, photosynthesis and gas-exchange processes changed, photosynthetic pigment content decreased, and the electron transfer efficiency in PSII reduced. However, compared with the control treatment, under drought conditions, the addition of exogenous calcium could stabilize the structure and function of the chloroplasts, mitochondria, and endomembrane system in the mesophyll cells, maintain normal leaf net photosynthetic rate and gas exchange, alleviate the degree of photosynthetic pigment degradation, and increase the electron transfer energy in the leaves in PSII. As a means of ensuring normal photosynthesis under drought stress, we discovered that the application of exogenous calcium was more important for stabilization of the structure of the organelles, regulation of the osmotic balance, and increase of the photosynthetic pigment content, and proved to be less important for regulation of stomatal opening and closing.

Mesophyll cell ultrastructure of wheat leaves etiolated by lead and selenium

The ultrastructure of mesophyll cells was studied in leaves of the Triticum aestivum L. cv. “Trizo” seedlings after two weeks of growth on soil contaminated by Pb and/or Se. The soil treatments: control; (Pb1) 50mgkg−1; (Pb2) 100mgkg−1; (Se1) 0.4mgkg−1; (Se2) 0.8mgkg−1; (Pb1+Se1); (Pb1+Se2); (P2+Se1); and (Pb2+Se2) were used. Light and other conditions were optimal for plant growth. The (Se1)-plants showed enhanced growth and biomass production; (Pb1+Se1)-plants did not lag behind the controls, though O2 evolution decreased; chlorophyll content did not differ statistically in these treatments. Other treatments led to statistically significant growth suppression, chlorophyll content reduction, inhibition of photosynthesis, stress development tested by H2O2 and leaf etiolation at the end of 14-days experiment. The tops of etiolated leaves remained green, while the main leaf parts were visually white. Plastids in mesophyll cells of etiolated parts of leaves were mainly represented by etioplasts and an insignificant amount of degraded chloroplasts. Other cellular organelles remained intact in most mesophyll cells of the plants, except (Pb2+Se2)-plants. Ruptured tonoplast and etioplast envelope, swelled cytoplasm and mitochondria, and electron transparent matrix of gialoplasm were observed in the mesophyll cells at (Pb2+Se2)-treatment, that caused maximal inhibition of plant growth. The results indicate that Pb and Se effects on growth of wheat leaves are likely to target meristem in which the development of proplastids to chloroplasts under the light is determined by chlorophyll biosynthesis. Antagonistic effect of low concentration of Se and Pb in combination may retard etiolation process.

Three-dimensional intracellular structure of a whole rice mesophyll cell observed with FIB-SEM.

Ultrathin sections of rice leaf blades observed two-dimensionally using a transmission electron microscope (TEM) show that the chlorenchyma is composed of lobed mesophyll cells, with intricate cell boundaries, and lined with chloroplasts. The lobed cell shape and chloroplast positioning are believed to enhance the area available for the gas exchange surface for photosynthesis in rice leaves. However, a cell image revealing the three-dimensional (3-D) ultrastructure of rice mesophyll cells has not been visualized. In this study, a whole rice mesophyll cell was observed using a focused ion beam scanning electron microscope (FIB-SEM), which provides many serial sections automatically, rapidly and correctly, thereby enabling 3-D cell structure reconstruction. Rice leaf blades were fixed chemically using the method for conventional TEM observation, embedded in resin and subsequently set in the FIB-SEM chamber. Specimen blocks were sectioned transversely using the FIB, and block-face images were captured using the SEM. The sectioning and imaging were repeated overnight for 200-500 slices (each 50 nm thick). The resultant large-volume image stacks ( x = 25 μm, y = 25 μm, z = 10-25 μm) contained one or two whole mesophyll cells. The 3-D models of whole mesophyll cells were reconstructed using image processing software. The reconstructed cell models were discoid shaped with several lobes around the cell periphery. The cell shape increased the surface area, and the ratio of surface area to volume was twice that of a cylinder having the same volume. The chloroplasts occupied half the cell volume and spread as sheets along the cell lobes, covering most of the inner cell surface, with adjacent chloroplasts in close contact with each other. Cellular and sub-cellular ultrastructures of a whole mesophyll cell in a rice leaf blade are demonstrated three-dimensionally using a FIB-SEM. The 3-D models and numerical information support the hypothesis that rice mesophyll cells enhance their CO 2 absorption with increased cell surface and sheet-shaped chloroplasts.

Regulations of 6-Benzyladenine (6-BA) on Leaf Ultrastructure and Photosynthetic Characteristics of Waterlogged Summer Maize

A field experiment was performed to study the effects of spraying exogenous 6-benzyladenine (6-BA) after waterlogging for 6 days at the third leaf stage on the leaf mesophyll cell ultrastructure, chlorophyll content, gas exchange parameters, chlorophyll fluorescence, and malondialdehyde (MDA) content of summer maize (Zea mays L.) hybrids Denghai605 (DH605) and Zhengdan958 (ZD958), to determine the photosynthetic mechanism for the regulation of 6-BA to waterlogged summer maize. Results showed that exogenous application of 6-BA effectively alleviated the adverse effects of waterlogging on photosynthetic characteristics by increasing leaf area index and chlorophyll content. Treatment with spraying exogenous 6-BA resulted in significant and substantial (in the range of 9–37%) increases of the net photosynthetic rate compared to waterlogging treatments. In addition, spraying 6-BA effectively alleviated the adverse effects of waterlogging on leaf ultrastructure by increasing the numbers of grana and grana lamella, decreasing MDA content, and improving membrane structure and chloroplast structure. Visibly, spraying exogenous 6-BA effectively alleviated the adverse effects of waterlogging on leaf ultrastructure of summer maize led to the improvement of photosynthetic performance, resulting in significant and substantial (around 20%) increases in grain yield compared to those of the waterlogging treatments.

Effects of Waterlogging on Leaf Mesophyll Cell Ultrastructure and Photosynthetic Characteristics of Summer Maize.

A field experiment was performed to study the effects of waterlogging on the leaf mesophyll cell ultrastructure, chlorophyll content, gas exchange parameters, chlorophyll fluorescence, and malondialdehyde (MDA) content of summer maize (Zea mays L.) hybrids Denghai605 (DH605) and Zhengdan958 (ZD958). The waterlogging treatments were implemented for different durations (3 and 6 days) at the third leaf stage (V3), the sixth leaf stage (V6), and the 10th day after the tasseling stage (10VT). Leaf area index (LAI), chlorophyll content, photosynthetic rate (Pn), and actual photochemical efficiency (ΦPSII) were reduced after waterlogging, indicating that waterlogging significantly decreased photosynthetic capacity. The chloroplast shapes changed from long and oval to elliptical or circular after waterlogging. In addition, the internal structures of chloroplasts were degenerated after waterlogging. After waterlogging for 6 d at V3, the number of grana and grana lamellae of the third expanded leaf in DH605 were decreased by 26.83% and 55.95%, respectively, compared to the control (CK). Those in ZD958 were reduced by 30.08% and 31.94%, respectively. Waterlogging increased MDA content in both hybrids, suggesting an impact of waterlogging on membrane integrity and thus membrane deterioration. Waterlogging also damaged the biological membrane structure and mitochondria. Our results indicated that the physiological reactions to waterlogging were closely related to lower LAI, chlorophyll content, and Pn and to the destruction of chloroplast ultrastructure. These negative effects resulted in the decrease of grain yield in response to waterlogging. Summer maize was the most susceptible to damage when waterlogging occurred at V3, followed by V6 and 10VT, with damage increasing in the wake of waterlogging duration increasing.

Effects of shading on the photosynthetic characteristics and mesophyll cell ultrastructure of summer maize.

A field experiment was conducted to study the effects of shading on the photosynthetic characteristics and mesophyll cell ultrastructure of two summer maize hybrids Denghai605 (DH605) and Zhengdan958 (ZD958). The ambient sunlight treatment was used as control (CK) and shading treatments (40% of ambient sunlight) were applied at different growth stages from silking (R1) to physiological maturity (R6) (S1), from the sixth leaf stage (V6) to R1 (S2), and from seeding to R6 (S3), respectively. The net photosynthetic rate (P n) was significantly decreased after shading. The greatest reduction of P n was found at S3 treatment, followed by S1 and S2 treatments. P n of S3 was decreased by 59 and 48% for DH605, and 39 and 43% for ZD958 at tasseling and milk-ripe stages, respectively, compared to that of CK. Additionally, leaf area index (LAI) and chlorophyll content decreased after shading. In terms of mesophyll cell ultrastructure, chloroplast configuration of mesophyll cells dispersed, and part of chloroplast swelled and became circular. Meanwhile, the major characteristics of chloroplasts showed poorly developed thylakoid structure at the early growth stage, blurry lamellar structure, loose grana, and a large gap between slices and warping granum. Then, plasmolysis occurred in mesophyll cells and the endomembrane system was destroyed, which resulted in the dissolution of cell membrane, karyotheca, mitochondria, and some membrane structures. The damaged mesophyll cell ultrastructure led to the decrease of photosynthetic capacity, and thus resulted in significant yield reduction by 45, 11, and 84% in S1, S2, and S3 treatments, respectively, compared to that of CK.

Elevated CO2 can modify the response to a water status gradient in a steppe grass: from cell organelles to photosynthetic capacity to plant growth.

BackgroundThe atmospheric CO2 concentration is rising continuously, and abnormal precipitation may occur more frequently in the future. Although the effects of elevated CO2 and drought on plants have been well reported individually, little is known about their interaction, particularly over a water status gradient. Here, we aimed to characterize the effects of elevated CO2 and a water status gradient on the growth, photosynthetic capacity, and mesophyll cell ultrastructure of a dominant grass from a degraded grassland.ResultsElevated CO2 stimulated plant biomass to a greater extent under moderate changes in water status than under either extreme drought or over-watering conditions. Photosynthetic capacity and stomatal conductance were also enhanced by elevated CO2 under moderate drought, but inhibited with over-watering. Severe drought distorted mesophyll cell organelles, but CO2 enrichment partly alleviated this effect. Intrinsic water use efficiency (WUEi) and total biomass water use efficiency (WUEt) were increased by elevated CO2, regardless of water status. Plant structural traits were also found to be tightly associated with photosynthetic potentials.ConclusionThe results indicated that CO2 enrichment alleviated severe and moderate drought stress, and highlighted that CO2 fertilization’s dependency on water status should be considered when projecting key species’ responses to climate change in dry ecosystems.Electronic supplementary materialThe online version of this article (doi:10.1186/s12870-016-0846-9) contains supplementary material, which is available to authorized users.

Red and Blue Lights Significantly Affect Photosynthetic Properties and Ultrastructure of Mesophyll Cells in Senescing Grape Leaves

Light quality significantly affects photosynthetic efficiency in plants. The mechanisms for how light quality affects photosynthesis in grape is poorly understood. Therefore, to investigate the effects of different light qualities on chloroplast ultrastructure and photosynthesis efficiency, two grape cultivars ‘Italia’ (slower speed of leaf senescence) and ‘Centennial Seedless’ (faster speed of leaf senescence) grown under protected and delayed conditions were used. The three treatments, replicated three times, were control (no supplemental lighting), red light and blue light. Chlorophyll content, net photosynthetic rate, and the ratio of Fv/Fm significantly increased in red light relative to the control. The opposite trend was observed in blue light in the early phase of leaf senescence. At later stages, physiological indexes were gradually higher than that of control, resulting in a delay in leaf senescence. Compared to the control, red and blue light both significantly increased the chlorophyll a/b ratio. Electron microscopy showed that blue light caused severe damage to the fine structure of chloroplasts at early stages of leaf senescence, but effects at later stages of leaf senescence became less severe compared to the control. The degradation of chloroplast ultrastructure was apparently delayed in red light throughout the experimental timeframe compared to other treatments. In this experiment, ‘Italia’ showed higher chlorophyll content, net photosynthetic rate, ratios of Fv/Fm, chlorophyll a/b and better preserved chloroplast ultrastructure relative to ‘Centennial Seedless’, resulting in a slower rate of leaf senescence.

Structure, pigment content, and photosynthesis of Siberian larch needles in Northern and Sub-Arctic Urals

The structure and functional organization of the photosynthetic apparatus of Siberian larch growing on the western slope of Northern and Sub-Arctic Urals were studied. The needle morphostructure, the mesophyll cell ultrastructure, and the pigment complex of chloroplasts were quantitatively characterized. Active photosynthesis of larch was observed during warm days with a low deficit of humidity. Photoassimilation depression under hot and dry weather was followed by a decrease in the stomatal conductance of needles. Under extreme conditions of mountains, Siberian larch has low photosynthesis. It decreased gradually from Nothern to Sub-Arctic Urals.
