Extraction and Characterization of Phenolic Compounds from Commiphora gileadensis Bark Using Ultrasonic-Assisted Extraction

Developing nations across the globe place significant importance on ensuring the complete protection, efficacy, and quality of herbal products and medicinal plants. Among these beneficial plants, Commiphora gileadensis stands out due to its diverse array of biological activities. The bark of this plant species offers numerous therapeutic advantages, including its potential for treating wounds, inflammatory conditions, and bacterial infections. To explore the process of extraction of biologically active components, this study employed ultrasonic-assisted extraction (UAE) on ethanolic extracts of C. gileadensis bark. Various variables of UAE process, such as ultrasonic frequency, extraction time, sample-to-solvent ratio, and ethanol concentration, were carefully investigated via the ‘’One-Factor-At-a-Time (OFAT)’’ method to understand their individual impacts on the extraction process. The obtained extracts underwent further analysis to identify the presence of different phytochemicals, employing techniques such as ‘’Gas Chromatography-Mass Spectroscopy (GC-MS) and Fourier Transform Infrared Spectroscopy (FTIR)’’. The experimental results demonstrated that the UAE process yielded promising outcomes, with the maximum extraction yields, total phenolic content (TPC), and total flavonoids content (TFC) of the ethanolic extract from C. gileadensis bark reaching 37.60 ± 0.31%, 149.75 ± 3.23mg GAE/g d.w., and 44.84 ± 3.02mg QE/g d.w., respectively. Additionally, this study successfully identified a total of 30 phenolic compounds present in the C. gileadensis bark extract for the first time, highlighting the efficacy of UAE as a suitable method for efficiently extracting various groups of phytochemicals from the bark of C. gileadensis.

Ultrasonic-assisted extraction, anti-biofilm activity, and mechanism of action of Ku Shen (Sophorae Flavescentis Radix) extracts against Vibrio parahaemolyticus.

The objective of this study is to optimize the ultrasonic-assisted extraction process of Ku Shen (Sophorae Flavescentis Radix) extracts (KSE) against Vibrio parahaemolyticus and explore their anti-biofilm activity and mechanism of action. The ultrasonic-assisted extraction process of KSE optimized by single factor experiment, Box-Behnken design and response surface methodology was as follows: 93% ethanol as solvent, liquid/material ratio of 30 mL/g, ultrasonic power of 500 W, extraction temperature of 80°C and time of 30 min. Under these conditions, the diameter of inhibition circle of KSE was 15.60 ± 0.17 mm, which had no significant difference with the predicted value. The yield of dried KSE is 32.32 ± 0.57% and the content of total flavonoids in KSE was 57.02 ± 5.54%. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of KSE against V. parahaemolyticus were 0.25 and 0.5 mg/mL, respectively. Crystal violet staining, Congo red plate, spectrophotometry, CCK-8 and scanning electron microscopy were used to investigate the activity and mechanism of action of KSE against V. parahaemolyticus biofilm. The results showed that the sub-MIC of KSE could significantly inhibit biofilm formation, reduce the synthesis of polysaccharide intercellular adhesin (PIA) and the secretion of extracellular DNA. In addition, the inhibition rate of biofilm formation and clearance rate of mature biofilm of 1.0 mg/mL KSE were 85.32 and 74.04%, and the reduction rate of metabolic activity of developing and mature biofilm were 77.98 and 74.46%, respectively. These results were confirmed by visual images obtained by scanning electron microscopy. Therefore, KSE has the potential to further isolate the anti-biofilm agent and evaluate it for the preservation process of aquatic products.

Establishment of HPLC method and optimization of ultrasonic-assisted extraction process for two lignan active substances in Asarum heterotropoides

In the present study, developed a dependable HPLC technique to quantify asarinin and sesamin in Asarum heterotropoides Fr. Schmidt var. mandshuricum (Maxim.) Kitag. The optimal conditions for the ultrasonic-assisted extraction were determined by conducting single-factor experiment, Plackett-Burman design experiment, and Box-Behnken response surface method. Based on actual operation factors and analysis, the optimal extraction parameters were methanol volume fraction of 80% (v/v), liquid-to-material ratio of 10:1 (mL/g), ultrasonic time of 40 min, ultrasonic temperature of 51°C, and ultrasonic power of 300 W. When subjected to these conditions, the average asarinin and sesamin content were found to be 13.40 mg/g and 2.39 mg/g, respectively, with a relative error of 1.04% and 3.46%, which was consistent with the theoretical value. This study can offer a theoretical reference for the extraction of asarinin and sesamin in Asarum.

Ultrasonic-assisted extraction to enhance the recovery of bioactive phenolic compounds from Commiphora gileadensis leaves

The “ultrasonic-assisted extraction (UAE)” method was utilized in this work to assess how different process parameters affected the yield and recovery of phenolic compounds from the leaf of Commiphora gileadensis, which is one of the medicinal plants with a variety of biological functions. Its leaf is used for a various of therapeutic applications, such as the treatment of bacterial infections, inflammation, and wound healing. The “One-Factor-At-a-Time (OFAT)” approach was employed to examine the impacts of various UAE process parameters on the process of extraction, which include time of extraction, sample/solvent ratio, ultrasonic frequency, and solvent (ethanol) concentration. The extracts were then investigated for the presence of several phytochemicals using analytical techniques such as “Gas Chromatography-Mass Spectroscopy (GC–MS)” and “Fourier Transform Infrared Spectroscopy (FTIR)” studies. The findings showed that the maximum extraction yield, the total phenolic content (TPC), and the total flavonoids content (TFC) of the ethanolic extract of the leaves of C. gileadensis using the UAE method were at 31.80 ± 0.41 %, 96.55 ± 2.81 mg GAE/g d.w. and 31.66 ± 2.01 mg QE/g d.w. accordingly under a procedure duration of 15 min, ultrasonic frequency of 20 kHz, solvent/sample ratio of 1:20 g/mL, and solvent concentration of 40 % v/v. The leaves extract of C. gileadensis included 25 phenolic compounds that were previously unreported, and GC–MS analysis confirmed their presence. Hence, it follows that the UAE technique can successfully extract the phytochemicals from C. gileadensis for a variety of therapeutic uses.

Green extraction of phenolics and terpenoids from passion fruit peels using natural deep eutectic solvents

AbstractThis work aimed to optimize the ultrasonic‐assisted extraction (UAE) and microwave‐assisted extraction (MAE) of phenolics and terpenoids from passion fruit peels using natural deep eutectic solvents (NADES). NADES formulated with choline chloride and polyols with organic acids were used to investigate the solubility of phenolics and flavonoids in the solvent. Among these formulations, those derived from lactic acid‐glycerine (LGLY) and citric acid‐glucose (CGLU) exhibited the highest efficiency in extracting phenolics and terpenoids, respectively, when combined with UAE. Then one‐factor experiments were conducted to examine the influence of the UAE conditions (liquid‐to‐solid ratios [LSR], water content in NADES, ultrasonic power, time, and temperature) and MAE conditions (LSR, water content in NADES, ultrasonic power, and time) on the extraction yield of phenolics and terpenoids. The Box‐Behnken design (BBD) model was employed to optimize the extraction conditions with conditional ranges determined based on the results of the one‐factor experiments. The optimal conditions for the CGLU‐based MAE of terpenoids were 387 W of microwave power, 30% water content, 2.4 min, and 20 mL/g of LSR. The optimized conditions for LGLY‐based MAE of phenolics were 299 W, 18.7%, 1.3 min, and 10 mL/g. Total terpenoid content acquired from NADES‐based UAE was higher than NADES‐based MAE, while total phenolic content obtained at the optimal conditions of NADES‐based UAE and MAE was statistically insignificant. The efficiency of the two extraction ways was compared to selecting the favorable extraction method for acquiring phenolics and flavonoids from passion fruit peels. The research proposed that NADES‐based UAE was a green and effective method for attaining phenolics and terpenoids from passion fruit peel.Practical applicationPassion fruit peels are a by‐product of beverage production, which causes environmental pollution and increases treatment costs in the factory. These peels contain various bioactive compounds which benefit human health. The traditional extraction method shows low efficiency and high energy consumption. Thus, the appropriate extraction method and green solvents are needed to improve the extraction efficiency of bioactive compounds and can be directly applied to food products. Ultrasonic‐assisted extraction (UAE) and microwave‐assisted extraction (MAE) combined with natural deep eutectic solvents can enhance the extraction efficiency of bioactive compounds. However, the detailed conditions of UAE and MAE are necessary to compare the performance of the two methods. In addition to showing the significant superiority of UAE with MAE, the research showed the optimized conditions of the UAE process, which can be applied on the industrial scale to extract bioactive compounds and directly add them to food products.

RSM and ANN approach for optimization of ultrasonic assisted extraction of pumpkin seed oil and their quality assessment

The current study focuses on optimization of process parameters for ultrasonic assisted extraction (UAE) of pumpkin seed oil (PSO) using response surface methodology (RSM) and artificial neural network (ANN). Three parameters of UAE process, amplitude, time and solvent to seed (S:S) ratio were selected in the range of 20–40 %, 15–45 min and 2–6 mL/g respectively and optimized on the basis of responses viz. yield, total phenolic compounds (TPC), squalene content and induction time of oil. The optimum process conditions of UAE were obtained as amplitude- 34.76 %, time- 34.37 min and the S:S ratio 6 mL/g. Under these optimum conditions, the oil showed good yield 39.05 %, TPC 45.02 mg GAE/g, squalene content 447.4 mg/100 g and induction time 5.27 h. The ANN model was developed using multilayer perceptron (MLP) and trained with back propagation algorithm utilizing training data set. The results indicate that the predicted values of responses by ANN model were in good agreement with the experimental values than the RSM model. The UAE-PSO showed better retention of bioactive compounds along with far better oil stability as compared to SE-PSO. The Scanning electron microscopy (SEM) analysis showed the random porous pores and rupture cell wall matrix in UAE treated seed powder.

Biological Activities of Citrus aurantium Leaf Extract by Optimized Ultrasound-Assisted Extraction

Several studies have explored the biological activities of Citrus aurantium flowers, fruits, and seeds, but the bioactivity of C. aurantium leaves, which are treated as waste, remains unclear. Thus, this study developed a pilot-scale ultrasonic-assisted extraction process using the Box–Behnken design (BBD) for the optimized extraction of active compounds from C. aurantium leaves, and their antityrosinase, antioxidant, antiaging, and antimicrobial activities were evaluated. Under optimal conditions in a 150× scaleup configuration (a 30 L ultrasonic machine) of a pilot plant, the total phenolic content was 69.09 mg gallic acid equivalent/g dry weight, which was slightly lower (3.17%) than the theoretical value. The half maximal inhibitory concentration of C. aurantium leaf extract (CALE) for 2,2-diphenyl-1-picrylhydrazyl–scavenging, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)–scavenging, antityrosinase, anticollagenase, antielastase and anti-matrix metalloprotein-1 activities were 123.5, 58.5, 181.3, 196.4, 216.3, and 326.4 mg/L, respectively. Moreover, the minimal inhibitory concentrations for bacteria and fungi were 150–350 and 500 mg/L, respectively. In total, 17 active compounds were detected in CALE—with linalool, linalyl acetate, limonene, and α-terpineol having the highest concentrations. Finally, the overall transdermal absorption and permeation efficiency of CALE was 95.9%. In conclusion, our CALE demonstrated potential whitening, antioxidant, antiaging, and antimicrobial activities; it was also nontoxic and easily absorbed into the skin as well as inexpensive to produce. Therefore, it has potential applications in various industries.

Extraction and Emulsification of Carotenoids from Carrot Pomaces Using Oleic Acid.

This study aimed to use oleic acid-based ultrasonic-assisted extraction (UAE) to recover carotenoids from carrot pomace and emulsify the enriched-carotenoid oleic acid using spontaneous and ultrasonic-assisted emulsification. The extraction performance of oleic acid was compared with traditional organic solvents, including hexane, acetone, and ethyl acetate. The one-factor experiments were employed to examine the impact of UAE conditions, including liquid-to-solid ratios, temperature, ultrasonic power, and time, on the extraction yield of carotenoids and to find the conditional ranges for the optimization process. The response surface methodology was employed to optimize the UAE process. The second-order extraction kinetic model was used to find the mechanism of oleic acid-based UAE. After that, the enriched-carotenoid oleic acid obtained at the optimal conditions of UAE was used to fabricate nanoemulsions using spontaneous emulsification (SE), ultrasonic-assisted emulsification (UE), and SE-UE. The effect of SE and UE conditions on the turbidity of nanoemulsion was determined. Then, the physiochemical attributes of the nanoemulsion from SE, UE, and spontaneous ultrasonic-assisted emulsification (SE-UE) were determined using the dynamic light scattering method. The extraction yield of carotenoids from carrot pomace by using sonication was the highest. The adjusted optimal conditions were 39 mL/g of LSR, 50 °C, 12.5 min, and 350 W of ultrasonic power. Under optimal conditions, the carotenoid content attained was approximately 163.43 ± 1.83 μg/g, with the anticipated value (166 μg/g). The particle sizes of nanoemulsion fabricated at the proper conditions of SE, UE, and SE-UE were 31.2 ± 0.83, 33.8 ± 0.52, and 109.7 ± 8.24 nm, respectively. The results showed that SE and UE are suitable methods for fabricating nanoemulsions. The research provided a green approach for extracting and emulsifying carotenoids from carrot pomace.

Green extraction of phenolics and flavonoids from black mulberry fruit using natural deep eutectic solvents: optimization and surface morphology

This study deployed ultrasonic-assisted extraction (UAE), combined with natural deep eutectic solvents (NADES), to extract phenolics and flavonoids from the black mulberry fruit, and the antioxidant activity was examined. The extraction yields of NADES-based UAE were assessed based on the yields of phenolics and flavonoids extracted from the black mulberry fruit. This study selected the molar ratios of hydrogen bond acceptors (HBA) and hydrogen bond donors HBD at 1:2 from previous studies. Choline chloride-lactic acid showed the highest solubility with phenolics and flavonoids among NADES systems. One-factor experiments evaluated the effect of UAE conditions (liquid-to-solid ratio (LSR), water content in NADES, temperature, and time) on TPC, TFC, and antioxidant activity. The suitable NADES-based UAE conditions for extracting phenolics and flavonoids from the black mulberry fruit were 60 ml/g of LSR, 40% water content, 70 °C, and 15 min. Response surface methodology with the Box-Behnken design model optimized the NADES-based UAE process based on response (TPC, TFC, ABTS, OH, and DPPH). The optimal conditions for the NADES-based UAE process were 70 ml/g of LSR, 38.9% water content in NADES, 67.9 °C, and 24.2 min of extraction time. The predicted values of the Box-Behnken design were compatible with the experimental results. Moreover, scanning electron microscopy (SEM) was used to survey the surface of black mulberry fruit with and without sonication. SEM can assist in demonstrating the destructive effect of NADES and ultrasonic waves on material surfaces. SEM findings indicated the high surface destruction capacity of NADES, which partially contributed to a superior extraction yield of NADES than conventional organic solvents. The study proposes an efficient and green method for extracting bioactive compounds from black mulberry fruits. The black mulberry fruit extracts can be applied to meat preservation and beverages with high antioxidants.

Optimization of ultrasonic-assisted extraction of eugenol-rich fraction from basil leaves: Characterization of extract for phenols, flavonoids and antioxidant activity

In this study, optimization of ultrasonic assisted extraction (UAE) of eugenol-rich fraction from O. basilicum was performed. Box-Behnken design of response surface methodology was applied with three independent variables viz. solvent concentration (25–75%), amplitude (70–90%) and sonication time (7–12 min). The effects of these three independent variables were studied on extract yield, total phenolic content, total flavonoid content, anti-oxidant activity and eugenol content present in O. basilicum. Significant (p≤0.05) effects of all the independent variables were recorded in all the product responses. Extract yield and total phenolic content ranged from 74.36–55.48% and 23.45–10.25 mgGAE/g, respectively while as total flavonoid content and anti-oxidant activity was 0.082–0.009 mg/g and 71.21–25.33% respectively and eugenol content of 0.846–0.207 mg/g was recorded for all the treatments. The optimum processing conditions obtained by numerical optimization for solvent concentration (A), amplitude (B) and sonication time (C) were 52.48%, 90% and 7.75 min, respectively. Eugenol content of 41.44% was further validated through GC-MS/MS in extract developed after following the optimal condition. Conclusively, RSM was found to be successful for optimization of UAE process of eugenol rich fraction from basil leaves. The results confirmed the success of using ethanol as a safe solvent for UAE of eugenol from basil leaves.

Optimization of Spirulina’s Phycocyanin Extraction Yield Using Response Surface Method

Besides cultivation, extraction is also a critical stage in enhancing the yield of phycocyanin production - a highly valuable compound from Spirulina biomass. In this study, the combined effect of three important variables in the ultrasonic-assisted extraction process on phycocyanin extraction yield, namely extraction temperature, sonication time, and solvent pH were investigated through a central composite design experiment. Furthermore, the response surface method was applied in order to define an optimal condition to achieve the highest extraction yield. The results showed that when temperature ranged from 35ºC to 45ºC, sonication time from 20 to 50 minutes, and solvent pH from 6 to 8, the average yield of 30.135±1.552 mg/g was obtained with an average purity of 0.871±0.043. A regression model was also successfully developed, which allowed a good prediction of extraction yield based on the three mentioned variables. On the other hand, an optimal condition for extraction was also proposed with sonication time = 43.57 minutes, extraction temperature = 37.6oC, and solvent pH = 6.7. These results were practically valuable for the improvement of phycocyanin extraction from Spirulina biomass.

Green extraction of total phenolic and flavonoid contents from mangosteen (Garcinia mangostana L) rind using natural deep eutectic solvents

This research combined ultrasonic-assisted extraction (UAE) and natural deep eutectic solvent (NADES) to recover phenolic and flavonoid components from mangosteen rind. The antioxidant activities were determined using DPPH, ABTS+, and hydroxyl assays. NADES prepared from lactic and 1,2–propanediol had the highest extraction efficiency based on the total flavonoid content (TFC) and phenolic contents (TPC). Single-factor experiments were employed to assess the influence of UAE conditions (liquid-to-solid ratio, temperature, water content in NADES, and time) on TFC, TPC, and antioxidant activities. NADES-based UAE conditions were optimized using response surface methodology with the Box-Behnken design model on five dependent responses (TPC, TFC, DPPH, ABTS, and OH). The optimal conditions for the lactic-1,2–Propanediol-based UAE process were 76.7 ml liquid/g solid with 30.3% of water content at 57.5 °C for 9.1 min. Scanning electron microscopy (SEM) was applied to examine the surface morphology of mangosteen rind before and after sonication. This study proposes an efficient, green, and practical approach for recovering phenolics and flavonoids from mangosteen rinds.

Ultrasound-assisted extraction of a condensed tannin and its application for removal dyes from water

The woody oil industry generates a large amount of seed shell waste during its production. Therefore, this paper employs a central composite design (CCD) methodology to optimize the ultrasonic-assisted extraction process, thus utilizing the byproduct. The infrared spectrum and thermogravimetric analysis is adopted to explore the chemical structure and thermal stability of tannin. Besides, the tannin resin (TF) is prepared to elaborate its adsorbing performance for dyes in water. The results verify the role of ultrasonic-assisted extraction method in increasing yield rate. The optimal conditions are as follows: 60% ethanol as extraction solvent, the solid-liquid ratio of 1:25 (g/mL), ultrasonic power of 160 W, and extracted at 80oC for 60min. The yield rate of tannin under such conditions stands at 32.04% (w/w). Temperature is vital for the extraction rate. Acer truncatum tannin as a condensed tannin, performs favorable thermal stability, and TF effectively absorbs cationic dye in water. The maximum adsorption capacity of methylene blue peaks at 176.13 mg/g. Acer truncatum shell serves as a resource of condensed tannin, and Acer truncatum tannin resin can be potentially developed into a new biomass adsorbent.

Quantification of microstructural changes in artichoke by-products by image analysis after high-power ultrasound-assisted extraction of bioactive compounds

Image analysis of optical and scanning electron micrographs was successfully used to quantify the microstructural effect of ultrasound application (at 200 and 335 W/L) in artichoke stem samples during the extraction of phenolic compounds and chlorogenic acid. Three temperatures were evaluated (25, 40, and 60 °C) and ultrasound-assisted extraction was compared with mechanical assisted extraction (100 rpm). The low extraction yields of phenolic compounds and chlorogenic acid at 25 °C with mechanical agitation (3.9% and 0.9%) increased up to 96.7% and 78.5%, respectively, at 60 °C and 335 W/L ultrasonic power density. Breakages in plant tissue increased from 4.3% after extraction by mechanical agitation at 25 °C to 10.7% and 16.7% at 25 °C when US at 200 W/L and 335 W/L were applied, respectively. The combined effect of temperature and ultrasound during extraction promoted cell shrinkage, and thus, an increase in cell number per area, reaching the highest increase (36%) when extraction was performed at 60 °C and 335 W/L. The intensified ultrasonic-assisted extraction process affected the microstructure of the solid, with the magnitude of the effects depending on both the extraction temperature and the ultrasonic power density, which explains the increase in extraction yields.

Screening of Endophytic Fungi in Locoweed Induced by Heavy-Ion Irradiation and Study on Swainsonine Biosynthesis Pathway.

Swainsonine (SW) is a substance with both animal neurotoxicity and natural anticancer activity produced by the metabolism of endophytic fungus Alternaria section Undifilum oxytropis of locoweed. This paper produced SW by fermentation of the endophytic fungus A. oxytropis of locoweed and obtained the optimal ultrasonic-assisted extraction process of SW by the response surface methodology. Meanwhile, four mutant strains with significant and stable SW-producing properties were screened out after the mutagenesis of A. oxytropis by heavy-ion irradiation. Of these, three were high-yielding stains and one was a low-yielding strain. In addition, through the analysis of metabolomics studies, it was speculated that the different SW production performance of the mutant might be related to the biosynthesis and utilization of L-lysine, L-2-aminoadipate-6-semialdehyde, etc. These results laid the foundation for the expansion of SW production, artificial construction of low-toxic locoweed and clarification of the SW biosynthesis pathway in A. oxytropis.

Comparative Content, Biological and Anticancer Activities of Heracleum humile Extracts Obtained by Ultrasound-Assisted Extraction Method.

As the safety and effectiveness of synthetic drugs remain in doubt, researchers are trying to develop natural medicines from medicinal plants. Herein, ethyl acetate, methanol and water extracts from the Heracleum humile plant were obtained by an ultrasonic-assisted extraction process and the aim was to evaluate some biological effects of the extracts due to the limited data on the pharmacological properties of Heracleum humile in the literature. Weak antibacterial activity was observed on tested bacterial species. The minimum inhibitory concentration and the minimum bactericidal concentration values ranged from 250 to 500 μg/mL. In addition, cytotoxic activity was determined using the MTT test. The strongest findings were determined for ethyl acetate extract on the MDA-MB-231 cell lines at the 48th hour (IC50 :97.94 μg/mL), followed by the MCF-7 cell lines at the 24th hour (IC50 :103.9 μg/mL). All extracts of Heracleum humile contained mainly flavonoids, phenolic acids and their derivatives, i. e., well-known compounds that possess numerous biological activities such as antioxidant, anti-inflammatory, anticancer, antimicrobial etc. The study results could provide important information that Heracleum humile could be a potential candidate as a natural enzyme inhibitor. It can be concluded that these extracts could be useful in the elementary step of improving novel plant-derived multifunctional pharmaceuticals.

Ultrasonic-assisted extraction and thermal stability of phytochemicals from fenugreek leaves

The current investigation aimed to determine the optimum ultrasonic-assisted extraction conditions of the phytochemicals from fenugreek leaves (Trigonella-foenum graecum L.). The effects of the solvent type, sample–solvent ratio, amplitude and extraction time on total phenolic compounds, total flavonoids, antioxidant activity and total saponin content were studied with a multi-objective perspective. Quadratic models were obtained by desirability function approach and a numerical optimization study was carried out to achieve the highest amount of phytochemicals in the extracts. Further, thermal stability of the extracts produced at the optimum conditions was determined in terms of the amount of the phytochemicals at different pH values (pH 3.0–9.0). The optimum extraction conditions of the ultrasonic-assisted extraction process were 100% of water, 35.02 g/L of sample–solvent ratio, 52.63% of amplitude, and 61.73 min of extraction time. Thermal stability of the extract obtained at the optimum extraction conditions was higher at pH 3.0 than that of pH 6.0 and pH 9.0, and its antioxidant properties were preserved by ~25% even after 10 h at 60 °C.

Optimization of the ultrasonic-assisted extraction process to obtain total phenolic and flavonoid compounds from watermelon (Citrullus lanatus) rind

This context presents the study of ultrasonic-assisted extraction (UAE) to obtain phenolic and flavonoid compounds from watermelon rind powder (WRP). The antioxidant activity of the extracts was investigated using DPPH and ABTS+ assays. One-factor experiments were conducted to examine the effect of each factor (solid-to-liquid ratio (SLR), acetone concentration (AC), temperature, and time) on the UAE of WRP. Box-Behnken Design (BDD) model was employed to optimize the UAE conditions based on total phenolic contents (TPC), total flavonoid content (TFC), and their antioxidant activities. The optimal conditions were 1:30.50 SLR, 70.71% AC, 29.78 °C, and 10.65 min extraction time. There were no significant differences between predicted and experimental results (less than 6.0%), recommending a feasible and innovative process of deploying UAE to extract phenolics and flavonoids effectively from watermelon rind.

An innovative ultrasonic-assisted extraction process for enhancing the patchoulol from Pogostemon cablin essential oil using hydrotropes

Pogostemon cablin (Patchouli) is an aromatic herb cultivated for its high-value essential oil. It is majorly used as a base oil in fragrance formulation due to its characteristics sesquiterpenoid alcohols. Patchoulol is an imperative and bioactive sesquiterpenoid in the patchouli essential oil, which is primarily isolated from the aerial part of the plant. The patchoulol was enriched in this essential oil by ultrasonic-assisted extraction techniques using different hydrotropes viz., sodium p-toluene sulphonate, resorcinol, sodium cumene sulphonate and sodium salicylate. The effect of various parameters such as extraction time (min), ultrasonic amplitude (%) and pulse interval (s) with 2 M concentration of hydrotropes was studied. Ultrasonic assisted-techniques has improved the percentage of patchoulol in patchouli essential oil. The highest composition of patchoulol i.e., 70.06 % was obtained for 5 min extraction time, 40 % ultrasonic amplitude, 30:30 s pulse interval for sodium salicylate at 2 M concentration.

Optimization of Extraction Conditions of Bioactive Compounds by Ultrasonic-Assisted Extraction from Artichoke Wastes

In this study, bioactive compounds were extracted by ultrasonic-assisted extraction and classical extraction processes using distilled water as solvent from artichoke leaves which are considered as agricultural wastes. Antioxidant capacity, total phenolic and total flavonoid content values of the obtained bioactive extracts were determined, and extraction yields and times were evaluated to compare the extraction processes. Also, the optimum extraction conditions of ultrasonic-assisted extraction (extraction time and ultrasonic power) which provide the highest extraction yield were determined using D-optimal design by 'desirability' function approach. According to the results, bioactive extracts having high antioxidant capacity were obtained at shorter times and higher extraction yields by ultrasonic-assisted extraction process than classical extraction. The highest extraction yield was estimated as 98.46% with an application of 20.05 minutes of extraction time and 65.02% of ultrasonic amplitude for the ultrasonic-assisted extraction process.
