Fluorescence Enhancement of Nonemissive Monodeprotonated Luteolin in a Poly(vinyl alcohol) Film.

Solid polymer matrixes can modulate the electronic states of embedded chromophores and have been widely used in flexible optoelectronic and optical materials. Luteolin is one of the most common natural flavonoids, and its neutral and monodeprotonated forms are nonemissive in aqueous solution induced by ultrafast excited-state proton transfer (ESPT) followed by nonradiative relaxation. In this study, we have incorporated luteolin into poly(vinyl alcohol) (PVA) films and studied their fluorescence behaviors. Neutral and one monodeprotonated luteolin coexist in the PVA film. Weak steady-state fluorescence of neutral luteolin peaking at about 440 nm is observed for the first time. In addition, the monodeprotonated luteolin in PVA film exhibits obvious fluorescence peaking at 500 nm, with a fluorescence quantum yield of as high as 0.4 and a fluorescence lifetime of as long as 2.4 ns. Time-dependent density functional theory calculations have determined that the ESPT of neutral luteolin is barrierless but that of monodeprotonated luteolin needs to surmount a barrier, explaining their distinct emission properties. These results indicate the modulation ability of the PVA film in both ground-state deprotonation and ESPT, broadening the application areas of the solid polymer matrix.

Extending Non-Perturbative Simulation Techniques for Open-Quantum Systems to Excited-State Proton Transfer and Ultrafast Non-Adiabatic Dynamics.

Excited state proton transfer is an ubiquitous phenomenon in biology and chemistry, spanning from the ultrafast reactions of photobases and acids to light-driven, enzymatic catalysis and photosynthesis. However, the simulation of such dynamics involves multiple challenges, since high-dimensional, out-of-equilibrium vibronic states play a crucial role, while a fully quantum description of the proton's dissipative, real-space dynamics is also required. In this work, we extend the powerful matrix product state approach to open quantum systems (TEDOPA) to study these demanding dynamics, and also more general nonadiabatic processes that can appear in complex photochemistry subject to strong laser driving. As an illustration, we initially consider an open model of a four-level electronic system interacting with hundreds of intramolecular vibrations that drive ultrafast excited state proton transfer, as well as an explicit photonic environment that allows us to directly monitor the resulting dual fluorescence in this system. We then demonstrate how to include a continuous "reaction coordinate" of the proton transfer that allows numerically exact simulations that can be understood, visualized and interpreted in the familiar language of diabatic and adiabatic dynamics on potential surfaces, while also retaining an exact quantum treatment of dissipation and driving effects that could be used to study diverse problems in ultrafast photochemistry.

Substituent Effects in the Cationic Green Fluorescent Protein Chromophore: Ultrafast Excited‐State Proton Transfer or Twisting?

AbstractUnderstanding the structure‐function relationships of the green fluorescent protein (GFP) chromophore is important in rationally developing new molecular tools for biological imaging and beyond. Herein, we systematically modified the GFP chromophore structure with electron‐withdrawing and ‐donating groups (EWGs and EDGs) to investigate the substituent effects on the excited‐state proton transfer (ESPT) and twisting dynamics of the cationic chromophore in solution. With key insights gained from femtosecond transient absorption and stimulated Raman spectroscopy, we reveal that the EWG substitution by –F increases photoacidity in an additive manner and leads to an ultrafast barrierless ESPT by difluorination, while the EDG substitution by –OCH3 also results in ultrafast ESPT despite the weak photoacidity as estimated by the Förster equation. We ascribe the unusually fast kinetics in methoxylated derivatives to the occurrence of a pre‐existing chromophore‐solvent complex that sets up the acceptor site for ESPT. Furthermore, the kinetic competition between ESPT and twisting pathways is crucial for the observation of ESPT in action, particularly for molecules undergoing efficient nonradiative decay in the excited state through torsional motions. Such flexible and highly engineerable molecules can enable more versatile photoswitches and sensors.

Photochemical initiation of polariton-mediated exciton propagation

Abstract Placing a material inside an optical cavity can enhance transport of excitation energy by hybridizing excitons with confined light modes into polaritons, which have a dispersion that provides these light–matter quasi-particles with low effective masses and very high group velocities. While in experiments, polariton propagation is typically initiated with laser pulses, tuned to be resonant either with the polaritonic branches that are delocalized over many molecules, or with an uncoupled higher-energy electronic excited state that is localized on a single molecule, practical implementations of polariton-mediated exciton transport into devices would require operation under low-intensity incoherent light conditions. Here, we propose to initiate polaritonic exciton transport with a photo-acid, which upon absorption of a photon in a spectral range not strongly reflected by the cavity mirrors, undergoes ultra-fast excited-state proton transfer into a red-shifted excited-state photo-product that can couple collectively with a large number of suitable dye molecules to the modes of the cavity. By means of atomistic molecular dynamics simulations we demonstrate that cascading energy from a photo-excited donor into the strongly coupled acceptor-cavity states via a photo-chemical reaction can indeed induce long-range polariton-mediated exciton transport.

Quantifying the Ground‐State Hydrogen‐Bond Formation of a Super‐Photoacid by Inspecting Its Excited‐State Dynamics

AbstractThe identification and quantification of hydrogen (H)‐bonded complexes form the cornerstone of reaction‐mechanism analysis in ultrafast proton‐transfer reactions. Traditionally, the Benesi–Hildebrand method has been employed to obtain the formation constants of H‐bonded complexes, given that H‐bonding additives induce an alteration in spectral features exclusively through H‐bond formation. However, if the additive introduction impacts the bulk polarity of the solution, inducing a spectral shift, the spectroscopic method‘s accuracy in analyzing the H‐bond formation becomes compromised. In this study, we scrutinize H‐bond formation under the influence of an H‐bond accepting solute in an aprotic solvent. This is achieved by quantifying the fractions of two concurrent pathways involved in the excited‐state proton transfer (ESPT) of a super‐photoacid: the ultrafast ESPT of an H‐bonded complex versus the diffusion‐controlled ESPT of the free acid. Our method offers improved accuracy compared to conventional steady‐state spectroscopic techniques, by directly quantifying the H‐bonded complexes using the time‐resolved spectroscopic method, thereby circumventing the aforementioned limitation.

Targeting Ultrafast Spectroscopic Insights into Red Fluorescent Proteins.

Red fluorescent proteins (RFPs) represent an increasingly popular class of genetically encodable bioprobes and biomarkers that can advance next-generation breakthroughs across the imaging and life sciences. Since the rational design of RFPs with improved functions or enhanced versatility requires a mechanistic understanding of their working mechanisms, while fluorescence is intrinsically an ultrafast event, a suitable toolset involving steady-state and time-resolved spectroscopic techniques has become powerful in delineating key structural features and dynamic steps which govern irreversible photoconverting or reversible photoswitching RFPs, and large Stokes shift (LSS)RFPs. The pertinent cis-trans isomerization and protonation state change of RFP chromophores in their local environments, involving key residues in protein matrices, lead to rich and complicated spectral features across multiple timescales. In particular, ultrafast excited-state proton transfer in various LSSRFPs showcases the resolving power of wavelength-tunable femtosecond stimulated Raman spectroscopy (FSRS) in mapping a photocycle with crucial knowledge about the red-emitting species. Moreover, recent progress in noncanonical RFPs with a site-specifically modified chromophore provides an appealing route for efficient engineering of redder and brighter RFPs, highly desirable for bioimaging. Such an effective feedback loop involving physical chemists, protein engineers, and biomedical microscopists will enable future successes to expand fundamental knowledge and improve human health.

Ultrafast Excited-State Proton Transfer in 4-Nitrocatechol: Implications for the Photochemistry of Nitrophenols.

Nitrophenols are a class of environmental contaminants that exhibit strong absorption at atmospherically relevant wavelengths, prompting many studies of their photochemical degradation rates and mechanisms. Despite the importance of photochemical reactions of nitrophenols in the environment, the ultrafast processes in electronically excited nitrophenols are not well understood. Here, we present an experimental study of ultrafast electron dynamics in 4-nitrocatechol (4NC), a common product of biomass burning and fossil fuel combustion. The experiments are accompanied by time-dependent quantum mechanical calculations to help assign the observed transitions in static and transient absorption spectra and to estimate the rates of singlet-to-triplet intersystem crossing. Our results suggest that electronic triplet states are not efficiently populated upon 340 nm excitation, as efficient proton transfer occurs in the excited state on a time scale of a few picoseconds in water and tens of picoseconds in 2-propanol. This suggests that triplet states do not play a significant role in the photochemical reactions of 4NC in the environment and, by extension, in nitrophenols in general. Instead, consideration should be given to the idea that this class of molecules may serve as strong photoacids.

Theoretical insights into ultrafast excited state proton transfer coupled with twisted intramolecular charge transfer mechanism in 7-(2ʹ-pyridyl)indole: Effect of hydrogen bonding

The ultrafast proton transfer (PT) process in the excited state of 7-(2′-pyridyl)indole (abbreviated as 7PyIn) has been elucidated by on-the-fly dynamics simulations using the algebraic-diagrammatic construction scheme of second order (ADC(2)) method under the resolution of identity (RI) approximation. The tautomers of 7PyIn alone and associated with an acetonitrile molecule in the 7PyIn-CH3CN complex are most likely found after the excited state intramolecular proton transfer (ESIntraPT) within an ultrafast time scale between 40 and 60 fs. While a methanol molecule in the complex of 7PyIn-CH3OH could establish both ESIntraPT and excited-state intermolecular proton transfer (ESInterPT). ESInterPT of 7PyIn-CH3OH possibly occurs through a concerted process via the intermolecular hydrogen bonding bridge of the methanol molecule almost twice faster than that of ESIntraPT. This slower circumstance of the ESIntraPT process found in 7PyIn-CH3OH may be caused by the competition between formations of the intramolecular and intermolecular hydrogen bonds between 7PyIn and methanol. In addition, after the accomplishment of excited state proton transfer (ESPT), the initiation of conical intersection could be taking place by the structural twist of 7PyIn. The geometries, energies, proton probabilities, and provided mechanisms via the dynamics simulations of 7PyIn and its solvent clusters exhibiting ESIntraPT and ESInterPT are explored to provide the dynamic pictures of the ESPT processes at the molecular level.

Unraveling the Mechanistic Pathway for the Dual Fluorescence in Green Fluorescent Protein (GFP) Chromophore Analogue: A Detailed Theoretical Investigation.

The photophysical properties of the para-sulfonamide (p-TsABDI) analogue of the green fluorescent protein (GFP) chromophore with both proton donating and accepting sites have been exploited in polar solvents to understand the origin of the unusual dual fluorescence nature of the chromophore. In the polar solvents, the compound undergoes structural rearrangement upon photoexcitation, leading to the ultrafast excited-state intermolecular proton transfer (ESIPT) phenomenon at the S1 surface. In this work, we employed both the static electronic structure calculations and on-the-fly molecular dynamics simulation to unravel the underlying reason for this peculiar behavior of the p-TsABDI analogue in polar solvents. To represent this adequately and provide extensive information on the ESIPT mechanism mediated by the solvent molecules, we considered explicit solvent molecules using the integral equation formalism variant of polarizable continuum (IEFPCM) model. From the static calculation analysis, we can conclude that the dual emissive behavior of the compound is ascribed to the proton transfer (PT) phenomena in the excited-state. However, based on the static calculation exclusively, it is hard to ascertain the mechanistic pathway of the PT phenomena. Hence, to investigate the dynamics and reaction mechanism for the ESIPT process, we performed the on-the-fly dynamics simulation for p-TsABDI in solvent clusters. Dynamics simulation results reveal that, based on the time lag between all the proton transfer processes, the ESIPT mechanism occurs in a stepwise manner from the benzylidene moiety of the chromophore to its imidazolinone moiety. However, the nonexistence of crossings between the S1- and S0-states confirms the PT characteristics of the reactions.

Ultrafast Excited-State Proton Transfer of a Cationic Superphotoacid in a Nanoscopic Water Pool.

The excited-state proton transfer (ESPT) of a cationic superphotoacid, N-methyl-7-hydroxyquinolium, was studied within the water pool of an anionic aerosol-OT (AOT), bis(2-ethylhexyl) sulfosuccinate, reverse micelle (RM). Previously, we had found that the cationic photoacid residing at the anionic AOT interface was conducive to ESPT to the bound water having concentric heterogeneity on the time scale of hundreds of picoseconds to nanoseconds. In our present study, on the time scale of hundreds of femtoseconds to a few tens of picoseconds, the photoacid underwent an ultrafast ESPT influenced by mobile water constituting the core of the RM. The two subpopulations of the core water molecules that determine the ultrafast biphasic deprotonation of the photoacid on time scales differing by an order of magnitude were identified. The core water molecules solvating the counteranion of the photoacid showed a higher basicity than typical water clusters in bulk resulting in ESPT on a subpicosecond time scale. Bare water clusters sensed by the photoacid showed a slower ESPT, over several picoseconds, as typically limited by the rotational motion of water molecules for similar types of the photoacid.

Deprotonation-induced enhancement in fluorescence of 2-((2-hydroxybenzylidene)amino)phenol, a Schiff base

Excited state dynamics of neutral and anionic forms of 2-((2- hydroxybenzylidene)amino)phenol (HBAP), a Schiff base, has been elucidated using time resolved fluorescence spectroscopy. The neutral molecule exhibits keto-enol tautomerism. So, the fluorescent cis-keto excited state is formed by direct excitation as well as ultrafast excited state proton transfer (ESIPT) from the enolic excited state. It exhibits very feeble fluorescence, associated with a lifetime of tens of picosecond. The anion, however, exhibits significantly stronger fluorescence with lifetime of almost a nanosecond. The fluorescent excited state for the anion is formed by solvent relaxation around it and conformational relaxation of the molecule itself. Ion-dipole interaction in the ground state is manifested in the fluorescence properties. Comparison with very recent publications on other Schiff bases, it is established that enhancement of fluorescence, induced by deprotonation, is a general phenomenon for this class of molecules.

Structural Origin and Vibrational Fingerprints of the Ultrafast Excited State Proton Transfer of the Pyranine-Acetate Complex in Aqueous Solution.

The excited state proton transfer (ESPT) reaction from the photoacid 8-hydroxypyrene-1,3,6-trisulfonic acid (HPTS or pyranine) to an acetate molecule has been investigated in explicit aqueous solution via excited state ab initio molecular dynamics simulations based on hybrid quantum/molecular mechanics (QM/MM) potentials. In all the trajectories, the direct proton transfer has been observed in the excited state within 1 ps. We find that the initial structural configuration extracted from the ground state distribution strongly affects the ESPT kinetics. Indeed, the relative orientation of the proton donor-acceptor pair and the presence of a water molecule hydrogen bonded to the phenolic acid group of the pyranine are the key factors to facilitate the ESPT. Furthermore, we analyze the vibrational fingerprints of the ESPT reaction, reproducing the blue shift of the acetate CO stretching (COac), from 1666 to 1763 cm-1 testifying the transformation of acetate to acetic acid. Finally, our findings suggest that the acetate CC stretching (CCac) is also sensitive to the progress of the ESPT reaction. The CCac stretching is indeed ruled by the two vibrational modes (928 and 1426 cm-1), that in the excited state are alternately activated when the proton is shared or bound to the donor/acceptor, respectively.

Large Stokes shift fluorescence activation in an RNA aptamer by intermolecular proton transfer to guanine

Fluorogenic RNA aptamers are synthetic functional RNAs that specifically bind and activate conditional fluorophores. The Chili RNA aptamer mimics large Stokes shift fluorescent proteins and exhibits high affinity for 3,5-dimethoxy-4-hydroxybenzylidene imidazolone (DMHBI) derivatives to elicit green or red fluorescence emission. Here, we elucidate the structural and mechanistic basis of fluorescence activation by crystallography and time-resolved optical spectroscopy. Two co-crystal structures of the Chili RNA with positively charged DMHBO+ and DMHBI+ ligands revealed a G-quadruplex and a trans-sugar-sugar edge G:G base pair that immobilize the ligand by π-π stacking. A Watson-Crick G:C base pair in the fluorophore binding site establishes a short hydrogen bond between the N7 of guanine and the phenolic OH of the ligand. Ultrafast excited state proton transfer (ESPT) from the neutral chromophore to the RNA was found with a time constant of 130 fs and revealed the mode of action of the large Stokes shift fluorogenic RNA aptamer.

Controlling Light-Induced Proton Transfer from the GFP Chromophore.

Green Fluorescent Protein (GFP) is known to undergo excited-state proton transfer (ESPT). Formation of a short H-bond favors ultrafast ESPT in GFP-like proteins, such as the GFP S65T/H148D mutant, but the detailed mechanism and its quantum nature remain to be resolved. Here we study in vacuo, light-induced proton transfer from the GFP chromophore in hydrogen-bonded complexes with two anionic proton acceptors, I- and deprotonated trichloroacetic acid (TCA- ). We address the role of the strong H-bond and the quantum mechanical proton-density distribution in the excited state, which determines the proton-transfer probability. Our study shows that chemical modifications to the molecular network drastically change the proton-transfer probability and it can become strongly wavelength dependent. The proton-transfer branching ratio is found to be 60 % for the TCA complex and 10 % for the iodide complex, being highly dependent on the photon energy in the latter case. Using high-level ab initio calculations, we show that light-induced proton transfer takes place in S1 , revealing intrinsic photoacid properties of the isolated GFP chromophore in strongly bound H-bonded complexes. ESPT is found to be very sensitive to the topography of the highly anharmonic potential in S1 , depending on the quantum-density distribution upon vibrational excitation. We also show that the S1 potential-energy surface, and hence excited-state proton transfer, can be controlled by altering the chromophore microenvironment.

Switching between Ultrafast Pathways Enables a Green-Red Emission Ratiometric Fluorescent-Protein-Based Ca2+ Biosensor.

Ratiometric indicators with long emission wavelengths are highly preferred in modern bioimaging and life sciences. Herein, we elucidated the working mechanism of a standalone red fluorescent protein (FP)-based Ca2+ biosensor, REX-GECO1, using a series of spectroscopic and computational methods. Upon 480 nm photoexcitation, the Ca2+-free biosensor chromophore becomes trapped in an excited dark state. Binding with Ca2+ switches the route to ultrafast excited-state proton transfer through a short hydrogen bond to an adjacent Glu80 residue, which is key for the biosensor’s functionality. Inspired by the 2D-fluorescence map, REX-GECO1 for Ca2+ imaging in the ionomycin-treated human HeLa cells was achieved for the first time with a red/green emission ratio change (ΔR/R0) of ~300%, outperforming many FRET- and single FP-based indicators. These spectroscopy-driven discoveries enable targeted design for the next-generation biosensors with larger dynamic range and longer emission wavelengths.

Modeling Excited-State Proton Transfer to Solvent: A Dynamics Study of a Super Photoacid with a Hybrid Implicit/Explicit Solvent Model.

The rapid growth of time-resolved spectroscopies and the theoretical advances in ab initio molecular dynamics (AIMD) pave the way to look at the real-time molecular motion following the electronic excitation. Here, we exploited the capabilities of AIMD combined with a hybrid implicit/explicit model of solvation to investigate the ultrafast excited-state proton transfer (ESPT) reaction of a super photoacid, known as QCy9, in water solution. QCy9 transfers a proton to a water solvent molecule within 100 fs upon the electronic excitation in aqueous solution, and it is the strongest photoacid reported in the literature so far. Because of the ultrafast kinetics, it has been experimentally hypothesized that the ESPT escapes the solvent dynamics control (Huppert et al., J. Photochem. Photobiol. A2014,277, 90). The sampling of the solvent configuration space on the ground electronic state is the first key step toward the simulation of the ESPT event. Therefore, several configurations in the Franck–Condon region, describing an average solvation, were chosen as starting points for the excited-state dynamics. In all cases, the excited-state evolution spontaneously leads to the proton transfer event, whose rate is strongly dependent on the hydrogen bond network around the proton acceptor solvent molecule. Our study revealed that the explicit representation at least of three solvation shells around the proton acceptor molecule is necessary to stabilize the excess proton. Furthermore, the analysis of the solvent molecule motions in proximity of the reaction site suggested that even in the case of the strongest photoacid, the ESPT is actually assisted by the solvation dynamics of the first and second solvation shells of the water accepting molecule.

Ultrafast excited-state proton transfer dynamics in dihalogenated non-fluorescent and fluorescent GFP chromophores.

Green fluorescent protein (GFP) has enabled a myriad of bioimaging advances due to its photophysical and photochemical properties. To deepen the mechanistic understanding of such light-induced processes, novel derivatives of GFP chromophore p-HBDI were engineered by fluorination or bromination of the phenolic moiety into superphotoacids, which efficiently undergo excited-state proton transfer (ESPT) in aqueous solution within the short lifetime of the excited state, as opposed to p-HBDI where efficient ESPT is not observed. In addition, we tuned the excited-state lifetime from picoseconds to nanoseconds by conformational locking of the p-HBDI backbone, essentially transforming the nonfluorescent chromophores into highly fluorescent ones. The unlocked superphotoacids undergo a barrierless ESPT without much solvent activity, whereas the locked counterparts exhibit two distinct solvent-involved ESPT pathways. Comparative analysis of femtosecond transient absorption spectra of these unlocked and locked superphotoacids reveals that the ESPT rates adopt an "inverted" kinetic behavior as the thermodynamic driving force increases upon locking the backbone. Further experimental and theoretical investigations are expected to shed more light on the interplay between the modified electronic structure (mainly by dihalogenation) and nuclear motions (by conformational locking) of the functionalized GFP derivatives (e.g., fluorescence on and off).

Following Bimolecular Excited-State Proton Transfer between Hydroxycoumarin and Imidazole Derivatives.

The ultrafast dynamics of a bimolecular excited-state proton transfer (ESPT) reaction between the photoacid 7-hydroxy-4-(trifluoromethyl)-1-coumarin (CouOH) and 1-methylimidazole (MI) base in aprotic chloroform- d1 solution were investigated using ultrafast transient infrared (TRIR) and transient absorption (TA) spectroscopies. The excited-state lifetime of the photoacid in solution is relatively short (52 ps), which at the millimolar photoacid and base concentrations used in our study precludes any diffusion-controlled bimolecular ESPT reactions. This allows the prompt ESPT reaction between hydrogen-bonded CouOH and MI molecules to be studied in isolation and the "contact" ESPT dynamics to be unambiguously determined. Our time-resolved studies reveal that ultrafast ESPT from the CouOH moiety to hydrogen-bonded MI molecules occurs within ∼1 ps, tracked by unequivocal spectroscopic signatures of CouO-* photoproducts that are formed in tandem with HMI+. Some of the ESPT photoproducts subsequently π-stack to form exciplexes on a ∼35 ps time scale, minimizing the attractive Coulombic forces between the oppositely charged aromatic molecules. For the concentrations of CouOH and MI used in our study (up to 8 mM), we saw no evidence for excited-state tautomerization of coumarin anions.

Solvent Relaxation Accompanied Ultrafast Excited State Proton Transfer Dynamics Revealed in a Salicylideneaniline Derivative

AbstractIn this study, we have investigated the excited state intramolecular proton transfer (ESIPT) and subsequent dielectric relaxation of a newly synthesized Schiff base molecule in cyclohexane, methanol and SDS micelle through femtosecond fluorescence up‐conversion and broadband transient absorption spectroscopy. We found that the molecule showed ultrafast proton transfer with a timescale about 0.2 ps followed by solvent relaxation of the keto tautomer in methanol and SDS medium. In non‐polar cyclohexane, the solvation part was completely absent as expected. Computational study suggested the existence of a transition state in the proton transfer surface. Quantification of the system was done with two step model assigning the first rate constant to the relaxation of the excited state enol form and the second to the barrier crossing from the enol to the keto form.

Simulation and Analysis of the Spectroscopic Properties of Oxyluciferin and Its Analogues in Water.

Firefly bioluminescence is a quite efficient process largely used for numerous applications. However, some fundamental photochemical properties of the light emitter are still to be analyzed. Indeed, the light emitter, oxyluciferin, can be in six different forms due to interexchange reactions. In this work, we present the simulation of the absorption and emission spectra of the possible natural oxyluciferin forms in water and some of their analogues considering both the solvent/oxyluciferin interactions and the dynamical effects by using MD simulations and QM/MM methods. On the one hand, the absorption band shapes have been rationalized by analyzing the electronic nature of the transitions involved. On the other hand, the simulated and experimental emission spectra have been compared. In this case, an ultrafast excited state proton transfer (ESPT) occurs in oxyluciferin and its analogues, which impairs the detection of the emission from the protonated state by steady-state fluorescence spectroscopy. Transient absorption spectroscopy was used to evidence this ultrafast ESPT and rationalize the comparison between simulated and experimental steady-state emission spectra. Finally, this work shows the suitability of the studied oxyluciferin analogues to mimic the corresponding natural forms in water solution, as an elegant way to block the desired interexchange reactions allowing the study of each oxyluciferin form separately.
