Temporal trends of plastic additive contents in sediment cores of three French rivers (Loire, Meuse and Moselle) over the last decades

Sediment cores from three major French watersheds (Loire, Meuse and Moselle) have been dated by 137Cs and 210Pbxs from 1910 (Loire), 1947 (Meuse) and 1930 (Moselle) until the present in order to reconstruct trajectories of plastic additive contaminants including nine phthalate esters (PAEs) and seven organophosphate esters (OPEs), measured by gas chromatography–mass spectrometer (GC–MS–MS). Historical levels of ∑PAEs were higher than those of ∑OPEs in the Loire and the Moselle sediments, while ∑PAEs and ∑OPEs contents were of the same order of magnitude in the Meuse sediments. Although increases in concentrations do not evolve linearly, our results clearly indicate an increase in OPEs and PAEs concentrations from the 1950–1970 period onwards, compared with the first half of the 20th century. Our results show that, ∑OPE contents increase gradually over time in the Loire and Meuse rivers but evolve more randomly in the Moselle River. Trajectories of ∑PAEs depend on the river and no generality can be established, suggesting sedimentary reworking and/or local contamination. Data from this study allowed comparisons of contents of ∑OPEs and ∑PAEs between rivers, with ∑OPE concentrations in the Moselle River > Meuse River > Loire River, and concentrations of ∑PAEs in the Loire River > Moselle River > Meuse River. Among all PAEs, di(2-ethylhexyl) phthalate (DEHP) was the most abundant in all sediment samples, followed by diisobutyl phthalate (DiBP). Tris (2-chloroisopropyl) phosphate (TCPP) was the most abundant OPE in sediments of the three rivers. In addition, strong positive Pearson correlations were observed between organic matter (OM) parameters and OPE concentrations, and to a lesser extent, between OM parameters and PAE concentrations. This is particularly true for the Moselle River and for the Loire River, but less so for the Meuse River.

Temporal evolution of plastic additive contents over the last decades in two major European rivers (Rhone and Rhine) from sediment cores analyses

Although global plastic distribution is at the heart of 21st century environmental concerns, little information is available concerning how organic plastic additives contaminate freshwater sediments, which are often subject to strong anthropogenic pressure. Here, sediment core samples were collected in the Rhone and the Rhine watersheds (France), dated using 137Cs and 210Pbxs methods and analysed for nine phthalates (PAEs) and seven organophosphate esters (OPEs). The distribution of these organic contaminants was used to establish a chronological archive of plastic additive pollution from 1860 (Rhine) and 1930 (Rhone) until today. Sediment grain size and parameters related to organic matter (OM) were also measured as potential factors that may affect the temporal distribution of OPEs and PAEs in sediments. Our results show that OPE and PAE levels increased continuously in Rhone and Rhine sediments since the first records. In both rivers, ∑PAEs levels (from 9.1 ± 1.7 to 487.3 ± 27.0 ng g−1 dry weight (dw) ± standard deviation and from 4.6 ± 1.3 to 65.2 ± 11.2 ng g−1 dw, for the Rhine and the Rhone rivers, respectively) were higher than ∑OPEs levels (from 0.1 ± 0.1 to 79.1 ± 13.7 ng g−1 dw and from 0.6 ± 0.1 to 17.8 ± 2.3 ng g−1 dw, for Rhine and Rhone rivers, respectively). In both rivers, di(2-ethylhexyl) phthalate (DEHP) was the most abundant PAE, followed by diisobutyl phthalate (DiBP), while tris (2-chloroisopropyl) phosphate (TCPP) was the most abundant OPE. No relationship was found between granulometry and additives concentrations, while organic matter helps explain the vertical distribution of PAEs and OPEs in the sediment cores. This study thus establishes a temporal trajectory of PAEs and OPEs contents over the last decades, leading to a better understanding of historical pollution in these two Western European rivers.

Determination of potential dermal exposure rates of phosphorus flame retardants via the direct contact with a car seat using artificial skin

Dermal exposure to phosphorus flame retardants (PFRs) has received much attention as a major alternative exposure route in recent years. However, the information regarding dermal exposure via direct contact with a product is limited. In addition, in the commonly used dermal permeability test, the target substance is dissolved in a solvent, which is unrealistic. In this study, a dermal permeability test of PFRs in three car seats was performed using artificial skin. The PFR concentrations in the car seats are 0.12 wt% tris(2-chloroethyl) phosphate (TCEP), 0.030–0.25 wt% tris(2-chloroisopropyl) phosphate (TCPP), 0.15 wt% triphenyl phosphate (TPhP), 0.89 wt% cresyl diphenyl phosphate (CsDPhP), 0.074 wt% tricresyl phosphate (TCsP), and 0.46–4.7 wt% diethylene glycol bis [di (2-chloroisopropyl) phosphate (DEG-BDCIPP). The mean skin permeation rates for a contact time of 24 h are 14 (TCEP), 5.4–160 (TCPP), 0.67 (CsDPhP), 0.38 (TPhP), and 3.3–58 ng cm−2 h−1 (DEG-BDCIPP). The concentrations of TCsP in receptor liquid were lower than the limit of quantification at the contact time of 24 h. The skin permeation rates were significantly affected by the type of car seat (e.g., fabric or non-fabric). The potential dermal TCPP exposure rate for an adult via direct contact with the car seat during the average daily contact time (1.3 h), which was the highest value assessed in this study, was estimated to be 16,000 ng kg−1 day−1, which is higher than that related to inhalation and dust ingestion reported as significant exposure route of PFRs in previous studies. These facts reveal that dermal exposure associated with direct contact with the product might be an important exposure pathway for PFRs.

Novel method for rapid monitoring of OPFRs by LLE and GC–MS as a tool for assessing biodegradation: validation and applicability

Organophosphate flame retardants (OPFRs) are high-production volume chemicals widely present in environmental compartments. The presence of water-soluble OPFRs (tri-n-butyl phosphate (TnBP), tris(2-butoxyethyl) phosphate (TBEP), tris(2-chloroethyl) phosphate (TCEP), tris(2-chloroisopropyl) phosphate (TCPP), and triethyl phosphate (TEP)) in water compartments evidences the struggle of conventional wastewater treatment plants (WWTPs) to effectively eliminate these toxic compounds. This study reports for the first time the use of white-rot fungi as a promising alternative for the removal of these OPFRs. To accomplish this, a simple and cost-efficient quantification method for rapid monitoring of these contaminants’ concentrations by GC–MS while accounting for matrix effects was developed. The method proved to be valid and reliable for all the tested parameters. Sample stability was examined under various storage conditions, showing the original samples to be stable after 60 days of freezing, while post-extraction storage techniques were also effective. Finally, a screening of fungal degraders while assessing the influence of the glucose regime on OPFR removal was performed. Longer chain organophosphate flame retardants, TBP and TBEP, could be easily and completely removed by the fungus Ganoderma lucidum after only 4 days. This fungus also stood out as the sole organism capable of partially degrading TCEP (35% removal). The other chlorinated compound, TCPP, was more easily degraded and 70% of its main isomer was removed by T. versicolor. However, chlorinated compounds were only partially degraded under nutrient-limiting conditions. TEP was either not degraded or poorly degraded, and it is likely that it is a transformation product from another OPFR’s degradation. These results suggest that degradation of chlorinated compounds is dependent on the concentration of the main carbon source and that more polar OPFRs are less susceptible to degradation, given that they are less accessible to radical removal by fungi. Overall, the findings of the present study pave the way for further planned research and a potential application for the degradation of these contaminants in real wastewaters.Graphical

N-acetylcysteine alleviated tris(2-chloroisopropyl) phosphate-induced sperm motility decline and functional dysfunction in mice through reversing oxidative stress and DNA damage

The decline in male fertility caused by environmental pollutants has attracted worldwide attention nowadays. Tris(2-chloroisopropyl) phosphate (TCPP) is a chlorine-containing organophosphorus flame retardant applied in many consumer products and has multiple side effects on health. However, whether TCPP impairs spermatogenesis remains unclear. In this study, we found that TCPP reduced the sperm motility and blastocyst formation, inhibited proliferation and induced apoptosis in mice testes and spermatocyte cell line GC-2. Moreover, TCPP induced imbalance of oxidant and anti-oxidant, DNA damage and mitochondrial dysfunction, thus induced abnormal spermatogenesis. In this process, p53 signaling pathway was activated and N-acetylcysteine treatment partially alleviated the side effects of TCPP, including decrease of sperm motility, activation of p53 signaling pathway and DNA damage. Finally, our study verified that TCPP elevated reactive oxygen species (ROS), decreased mitochondrial membrane potential and induced apoptosis in human semen samples. Overall, ROS mediated TCPP-induced germ cell proliferation inhibition and apoptosis, which finally led to the decline of sperm motility.

Unravelling bioaccumulation, depletion and metabolism of organophosphate triesters in laying hens: Insight of in vivo biotransformation assisted by diester metabolites

Organophosphate triesters (tri-OPEs) threaten human health through dietary exposure, but little is known about their feed-to-food transfer and in vivo behavior in farm animals. Herein 135 laying hens were fed with contaminated feed (control group, low-level group and high-level group) to elucidate the bioaccumulation, distribution, and metabolism of the six most commonly reported tri-OPEs. The storage (breast muscle), metabolism and mobilization (liver and blood) and non-invasive (feather) tissues were collected. The exposure-increase (D1∼14) and depuration-decrease (D15∼42) trends indicated that feed exposure caused tri-OPE accumulation in animal tissues. Tissue-specific and moiety-specific behavior was observed for tri-OPEs. The highest transfer factor (TF) and transfer rate (TR) were observed in liver (TF: 14.8%∼82.3%; TR: 4.40%∼24.5%), followed by feather, breast muscle, and blood. Tris(2-chloroisopropyl) phosphate (TCIPP) had the longest half-life in feather (72.2 days), while triphenyl phosphate (TPhP) showed the shortest half-life in liver (0.41 days). Tri-OPEs’ major metabolites (organophosphate diesters, di-OPEs) were simultaneously studied, which exhibited dose-dependent and time-dependent variations following administration. In breast muscle, the inclusion of di-OPEs resulted in TF increases of 735%, 1108%, 798%, and 286% than considering TCIPP, tributyl phosphate, tris(2-butoxyethyl) phosphate and tris(2-ethylhexyl) phosphate alone. Feather was more of a proxy of birds’ long-term exposure to tri-OPEs, while short-term exposure was better reflected by di-OPEs. Both experimental and in silico modeling methods validated aryl-functional group facilitated the initial accumulation and metabolism of TPhP in the avian liver compared to other moiety-substituted tri-OPEs.

Matrix solid-phase dispersion extraction of organophosphorus flame retardants in soil based on response surface methodology

Organophosphorus flame retardants (OPFRs) are widely used in commercial products owing to their exceptional flame-retarding and plasticizing properties. However, OPFRs are also well recognized as emerging persistent organic pollutants (POPs) because of their environmental persistence, biological concentration, and potential toxicity. Thus, the accurate detection of OPFRs in environmental media is critical for analyzing their fate, transport, and ecological risk. However, very few OPFR detection methods are currently available, and the types of OPFRs detected may vary from site to site. In this study, matrix solid-phase dispersion extraction (MSPD), a simple, rapid, and versatile technique for preparing solid, semisolid, liquid, and viscous samples, was combined for the first time with gas chromatography-tandem mass spectrometry (GC-MS/MS) to analyze 10 OPFRs in soil, namely, tripropyl phosphate (TPrP), tri-n-butyl phosphate (TnBP), tri-iso-butyl phosphate (TiBP), tris(2-chloroisopropyl) phosphate (TCIPP), tris(2-chloroethyl) phosphate (TCEP), tris(1,3-dichloro-2-propyl) phosphate (TDCPP), triphenyl phosphate (TPHP), 2-ethylhexyl diphenyl phosphate (EHDPP), triphenylphosphine oxide (TPPO), and trimethylphenyl phosphate (TCP). The GC-MS/MS system was equipped with a Bruker-5MS capillary column coupled with a triple quadrupole mass spectrometer operated in multiple reaction monitoring (MRM) mode. Prior to detection, a mixed standard solution was fortified with 10 ng of13C-PCB208 as an internal standard. The optimal conditions under which MSPD could achieve high selectivity for OPFRs were determined. In addition, single-factor analysis was used to examine the influence of the sorbent (i. e., C18, PSA, Florisil, GCB, and multiwalled carbon nanotubes (MWCNTs)) as well as the dosage, type, and volume of the eluent on the extraction efficiency of the method for the 10 OPFRs. When GCB and ethyl acetate were used as the adsorbent and solvent, respectively, during elution, high extraction recoveries for the OPFRs were achieved. Optimization via response surface methodology (RSM) was adopted to further analyze the impact of three key factors, namely, the adsorbent dosage, eluent volume, and grinding time, as well as their interactions, on OPFR recoveries. Under the optimal conditions of 0.3 g of GCB as the adsorbent, 10 mL of ethyl acetate as the eluent, and 5 min of grinding time, the relative average recovery of the OPFRs was 87.5%. Furthermore, the 10 OPFRs showed good linear relationships under five concentration gradients, with correlation coefficients greater than 0.998. The limits of detection (LODs) and quantification (LOQs) were calculated as signal-to-noise ratios (S/N) of 3 and 10, respectively, and found to be in the ranges of 0.006-0.161 and 0.020-0.531 ng/g, respectively. The performance of the proposed method was verified by determining the recoveries and relative standard deviations (RSDs) of the OPFRs in soils spiked at low, medium, and high levels (10, 20, and 100 ng/g, respectively). The recoveries of the OPFRs ranged from 70.4% to 115.4%, with RSDs ranging from 0.7% to 6.7%. Compared with the conventional accelerated solvent extraction (ASE) method, MSPD presents higher efficiency, simpler operation, and less solvent requirements. The developed method was applied to determine OPFRs in soil samples collected from different sites in Suzhou, including an electronics factory, an auto-repair factory, a paddy field, and a school field. The results revealed that the contents of OPFRs in the soils from the electronics and auto-repair factories were significantly higher than those in the soils from the paddy and school fields. The main pollutants in the soil samples collected from the electronics and auto-repair factories were TCIPP, TPPO, TCEP, and TDCPP. Moreover, the contents of these compounds were 5.30, 4.44, 4.54, and 4.20 ng/g, in soils from the electronics factory and 2.70, 3.93, 7.60, and 5.04 ng/g, in soils from the auto-repair factory. To the best of our knowledge, this study is the first to determine high concentrations of TPPO in industrial soils. Thus, the combination of MSPD and GC-MS/MS adopted in this study can provide useful insights into the detection of the 10 OPFRs in soil.

Occurrence, distribution, potential sources, and risks of organophosphate esters in fresh snow on Urumqi Glacier No. 1, eastern Tien Shan, China

Organophosphate esters (OPEs), extensively used as flame retardants, are widely detected in various regions and environments. The potential toxicity of OPEs has caused great concern in recent years. Based on the global distillation model, the Tien Shan glaciers, such as Urumqi Glacier No. 1, could be as a potential “sink” for OPEs. However, little is known about the concentration, distribution, potential sources, and ecological risks of OPEs in Tien Shan glaciers. In this study, fresh snow samples were collected at various altitudes on the Urumqi Glacier No. 1, eastern Tien Shan, China. The total concentrations of ten OPEs (Σ10OPEs) ranged from 116 to 152 ng/L. The most abundant OPE was tris-(2-chloroisopropyl) phosphate (TCIPP), contributing to 74 % of the total OPEs. Σ10OPEs, tri-n-butyl phosphate (TnBP), and TCIPP concentrations showed positive correlations with altitude, indicating the effect of cold condensation on OPEs deposition. Based on air mass back-trajectory analysis and principal component analysis, we found that emissions from both traffic and household products in indoor environment were the important sources, and OPEs on the Urumqi Glacier No. 1 might mainly originate from Europe. Our assessment also showed triphenyl phosphate (TPhP) posed a low ecological risk in snow. This is the first systematic study of OPEs on the Tien Shan glaciers.

The transport and distribution of novel brominated flame retardants (NBFRs) and organophosphate esters (OPEs) in soils and moss along mountain valleys in the Himalayas

Although the Himalayas act as a natural barrier, studies have demonstrated that certain traditional persistent organic pollutants (POPs) can be transported into the Tibetan Plateau (TP) through the mountain valleys. Herein, we selected five mountain valleys in the Himalayas to investigate novel flame retardants (NFRs), as representative novel POPs, their concentration, distribution, transport behavior, potential sources and ecological risk. The results revealed that total concentrations of 7 novel brominated flame retardants (NBFRs) ranged from 4.89 to 2853 pg/g dry weight (dw) in soil and from not detected (ND) to 4232 pg/g dw in moss. Additionally, total concentrations of 10 organophosphate esters (OPEs) ranged from ND to 84798 pg/g dw in soil. Among the NFRs, decabromodiphenylethane (DBDPE) and tri-phenyl phosphate (TPhP) were the predominant compounds. NBFRs and OPEs concentrations were slightly higher than those in the polar regions. The correlation between different compounds and altitude varies in different areas, indicating that the NFRs distribution in the mountain valleys result from a combination of long-range transport and local sources. The ecological risk assessment using risk quotient (RQs) revealed that TPhP and tris (2-chloroisopropyl) phosphate (TCIPP) exhibited medium or high risks at some sites. This study sheds light on the transport pathways and environmental behaviors of the NFRs in the valleys and highlights the need for increased attention to the ecological risks posed by OPEs in the TP.

Study of the Spatiotemporal Variations, Source Determination, and Potential Ecological Risk of Organophosphate Esters in Typical Coastal Tourist Resorts in China

Investigation of the environmental occurrence and behavior of organophosphate esters (OPEs) is very important and is becoming a hot topic in the academic community. In this study, 12 targeted OPEs in the water and sediment from 19 coastal tourist resorts in the Shandong Peninsula in China were analyzed to show their concentrations, spatial variations, and potential ecological risks. The results showed that the total concentrations of OPEs (ΣOPEs) were in the range of 18.52–3069.43 ng/L in the water and 3.20–568.76 ng/g in the sediment. The dominant OPEs in the water and sediment were tris (2-chloroisopropyl) phosphate (TCIPP), tris (2-chloroethyl) phosphate (TCEP), and triethyl phosphate (TEP). The OPE concentrations in the water were ranked as dry season > normal season > wet season. The sewage treatment plants near tourist resorts were important contributors to the level of OPEs in the water. Triphenyl phosphate (TPHP), tri-n-butyl phosphate (TNBP), and resorcinol-bis(diphenyl)phosphate (RDP) had a relatively higher ecological risk than other OPEs in the water samples. Industrial emissions might be the main source of OPEs in the coastal tourist resorts of Shandong Peninsula in China. The results of this study verified that OPEs occur in the water and sediment of coastal tourist resorts, and more attention should be given to the existence of OPEs and the safety of aquatic environments near coastal tourist resorts.

Screening of organophosphorus flame retardant residues in rice by QuEChERS-gas chromatography-quadrupole time-of-flight mass spectrometry

Organophosphorus flame retardants (OPFRs) have emerged as good alternatives to brominated flame retardants, the use of which is globally restricted. In this study, a screening method based on QuEChERS-gas chromatography-quadrupole time-of-flight mass spectrometry (GC-Q-TOF/MS) was established for the determination of 21 OPFRs in rice. First, full scan (scanning range, m/z 50-450) was performed with a mixed standard solution of the 21 OPFRs (0.1 μg/g) by GC-Q-TOF/MS. The fragmentation pathways of these OPFRs were then investigated to explore their cleavage fragments, the interrelationships among fragments, and the possible cleavage modes of alkylated, chlorinated, and aromatic OPFRs. The retention times, isotopic abundance ratios, and molecular formulas of the characteristic fragments as well as the exact mass of the compounds were obtained to establish a mass spectral library of the OPFRs. Rice samples were extracted and purified by the QuEChERS method, and 0.5% formate acetonitrile solution was used as the extraction solvent; 4 g of magnesium sulfate, 1 g of sodium chloride, 0.5 g of disodium hydrogen citrate, and 1 g of sodium citrate as the extraction-salt combination; and 50 mg of primary secondary amine (PSA), 50 mg of octadecylsilane (C18), and 150 mg of magnesium sulfate as the purification materials. The chromatographic separation of the 21 OPFRs was completed within 16 min under optimized temperature program conditions on the DB-5MS UI column. The screening parameters were optimized, and a full scan of the samples was performed under the following conditions: number of characteristic fragment ions ≥2; accurate mass window=±2×10-5 (±20 ppm); retention time deviation=±0.2 min, and ion abundance deviation<20%. The developed method was applied to the screening 21 OPFRs in the samples. The results indicated that the matrix interference was greatly reduced by decreasing the extraction accurate mass window, thereby improving the signal-to-noise ratio of the analytes. The targets were extracted from the matrix interference and background noise using deconvolution software, which improved the match between the target compounds and the mass spectral library. The detection rates of alkyl and aromatic OPFRs increased by 22% and 25%, respectively, when the spiking level was increased from 2 to 10 ng/g. Among the chlorinated OPFRs, only tris(2-chloroisopropyl) phosphate (TCIPP) was not detected at a spiking level of 2 ng/g, indicating that chlorinated OPFRs could be identified even at low concentrations. The characteristic ions of the detected compounds matched those of the home-made mass spectral library well, indicating that the practical application of the home-made mass spectral library. The established screening method was applied in the determination of OPFRs in rice samples from different regions in China. A total of 11 OPFRs were detected, among which trimethyl phosphate (TMP), tri-iso-butyl phosphate (TiBP), and tris(3,5-dimethylphenyl) phosphate (T35DMPP) had the highest detection rates. These results indicate that these three OPFRs are widely used and can easily come into contact with rice samples through various routes. Differences in the types of OPFRs detected in the actual samples may be related to the types of OPFRs produced in local factories. OPFRs can be detected in rice samples by the developed GC-Q-TOF/MS screening method, which is helpful for the identification of OPFRs in complex matrix samples.

High-resolution mass spectrometry-based screening and dietary intake assessment of organophosphate esters in foodstuffs from South China

Organophosphate esters (OPEs) are a group of emerging contaminants with widespread environmental occurrence, yet research on their occurrence in foodstuffs is limited. We collected 100 foodstuff samples in South China using a market basket method, and analyzed food extracts for the presence of OPEs and organophosphite antioxidants (OPAs) by suspect and nontarget screening through high-resolution mass spectrometry. Our analysis resulted in the identification of 30 OPEs, comprised of 25 OPEs with a confidence level (CL) of 1 (unequivocal identification using standards) and five OPEs with CL = 2b (probable structure based on diagnostic evidence). Interestingly, 11 of these identified OPEs had not been previously reported in food. No OPA was identified. The occurrence of identified OPEs within the food samples was further investigated. The highest median concentration of OPEs in all food samples was reached by tris(2-chloroisopropyl) phosphate (TCPP) (1.55 ng/g ww, range < 0.74–12.0 ng/g wet weight (ww)). Cereals demonstrated the highest median concentration of the cumulative 30 OPEs. Tris(2-chloroethyl) phosphate (TCEP), TCPP, and triethyl phosphate (TEP) predominantly contributed to OPEs contamination in most food categories. Eight OPEs, namely TEP, tris(2-ethylhexyl) phosphate (TEHP), TCEP, triphenyl phosphate (TPhP), 2-ethylhexyl diphenyl phosphate (EHDPP), bis(2-ethylhexyl) phenyl phosphate (BEHPP), resorcinol bis(diphenyl phosphate) (RDP), and methyl diphenyl phosphate (MDPP) exhibited significantly higher concentrations in the processed group as compared to non-processed group, suggesting that food processing may result in contamination of these OPEs. The median sum of estimated dietary intake (ΣEDI) of all OPEs was determined to be 161 ng/kg body weight/day. Cereals (38.5 %) and vegetables (23.5 %) were the predominant food categories contributing to ΣEDI, and TEP (29.0 %), TCEP (20.2 %), and TCPP (18.3 %) were three major OPEs contributing to ΣEDI. This study for the first time offered a comprehensive overview of OPE species and revealed their occurrence in foodstuffs from South China.

Developmental toxicity in early chicken embryos on exposure to an organophosphorus flame retardant, tris(2-chloroisopropyl) phosphate

Tris(2-chloroisopropyl) phosphate (TCIPP) is an organophosphate flame retardant detected in the environment and eggs, feathers, and livers. Early-developmental-stage avian embryos are vulnerable to the toxic effects of chemicals. However, studies on the specific effects of TCIPP on avian embryonic development are limited. We aimed to investigate the toxicity of TCIPP in early chicken embryos using a previously developed shell-less incubation system. Fertilized chicken (Gallus gallus domesticus) eggs (n = 220) were exposed to 50 or 500 nmol TCIPP/(g egg) or dimethyl sulfoxide (DMSO) as a vehicle control on Day 0 of incubation. Development of 198 embryos was monitored from Days 3–9 of incubation, and 22 embryos on Day 4 and 74 embryos on Day 9 were dissected. Messenger RNA expression levels for several genes were measured in embryos on Day 4. Both TCIPP-exposed groups showed a significant reduction in survival rate. Imaging analyses revealed significant decreases in body length, head and bill length, eye diameter, and forelimb and hindlimb length in both TCIPP-treated groups. TCIPP exposure significantly impaired the development of extraembryonic blood vessels and the production of red blood cells. A TCIPP-dose-dependent decreasing trend in heart rate was observed on Days 4–7. The somitic angle increased significantly on Days 4–6, and embryos with curved somites showed cleavage in the back and gaps between somites, resulting in asymmetrical somite formation. A significant correlation was found between the somitic angle and FGF8 expression levels, suggesting that TCIPP exposure affects somite formation through an altered FGF-signaling pathway. Embryos with somitic deformities in TCIPP-exposed groups had significantly reduced survival rates, indicating that abnormal segment formation directly increased mortality. Finally, eye weight and ocular luminosity values were significantly reduced, suggesting that TCIPP may also affect eye development. Overall, these findings highlight severe toxic effects of TCIPP on avian embryonic development, including in vascularization, cardiac function, and somite and ocular development.

Degradation of aqueous tris(2-chloroisopropyl) phosphate by electrochemical oxidation process: Mechanisms, toxicity and ecological risks of intermediate products

In this study, we investigated the electrochemical oxidation of the organophosphate flame retardant tris(2-chloroisopropyl) phosphate (TCPP). The results demonstrated the effective elimination of TCPP following pseudo-first order kinetics. The degradation efficiency of TCPP attained 98.4 % and the corresponding reaction rate constant (k) was 0.0502 min−1, with the initial concentration of 1 mg/L, current density of 10 mA/cm2, and Na2SO4 of 10 mM. Quenching experiments revealed that ·OH played a vital role, with a contribution rate of 91.3 %. Environmental indicators in water significantly influenced the degradation efficiency, where Cl− had an inhibitory effect, while humic acid (HA), NO3−, and HCO3− enhanced degradation at lower concentrations. A total of six intermediates (C6H13Cl2O4P, C3H8ClO4P, C9H18Cl3O5P, C9H17Cl2O6P, C6H12ClO6P, and C3H8ClO5P) were generated through oxidation, dichlorination and hydrolysis reactions. TCPP induced cell apoptosis of Escherichia coli (E. coli), with the proportions of apoptotic cells decreasing from 33.3 % to 1.6 % after 180 min of degradation. Furthermore, the ecological toxicity was predicted to decrease during the electrochemical oxidation using ECOSAR software. TCPP did not induce the microbial community composition at the phylum level, while the bacterial abundance at genus level significantly changed. The dominant genus Methylotenera, accounting for 15.6 % in the control, decreased to 6.8 % in the presence of TCPP, and subsequently increased to 9.3 % in the 120 min products exposure groups. Electrochemical oxidation process could effectively degrade and detoxify TCPP, as well as reduce the ecological risks.

Serum organophosphate flame retardants and plasticizers in Chinese females of childbearing age: Association with serum reproductive and thyroid hormones

Organophosphate flame retardants (OPFRs) are extensively used as flame retardants and plasticizers, but their endocrine disrupting potentials have raised concerns. However, the impacts of OPFR exposures on reproductive and thyroid hormones in females remains unclear. In this study, serum concentrations of OPFRs were investigated, and levels of reproductive and thyroid hormones, including follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol, anti-Müllerian hormone, prolactin (PRL), testosterone (T), and thyroid stimulating hormone, were analyzed in childbearing-age females undergoing in-vitro fertilization treatment from Tianjin, a coastal city in China (n = 319). Tris (2-chloroethyl) phosphate (TCEP) was the predominant OPFR, with a median concentration of 0.33 ng/mL and a detection frequency of 96.6%. In the whole population, tris(1,3-dichloro-2-propyl) phosphate (TDCIPP) and tris(2-chloroisopropyl) phosphate (TCIPP) were positively associated with T (p < 0.05), while triethyl phosphate (TEP) was negatively associated with LH (p < 0.05) and LH/FSH (p < 0.01). Particularly, TCIPP was negatively associated with PRL in the younger subgroup (age≤30, p < 0.05). Moreover, TCIPP was negatively associated with diagnostic antral follicle counting (AFC) in the mediation analysis by a dominating direct effect (p < 0.01). In conclusion, serum levels of OPFRs were significantly associated with reproductive and thyroid hormone levels and a risk of decreased ovarian reserve in childbearing-age females, with age and body mass index being significant influencing factors.

Concentrations, spatial and seasonal variations of Organophosphate esters in UK freshwater Sediment

This study provides the first report of organophosphate esters (OPEs) in UK freshwater sediments with a focus on the possible impact of factors such as spatial/seasonal variation and wastewater treatment plants (WWTPs) on the measured OPE concentrations in surficial sediment from 3 UK rivers and 1 canal. Detection frequencies of: (tris (chloroethyl) phosphate (TCEP), tris (2-chloroisopropyl) phosphate (TClPP), tris (1,3-dichloro-2 propyl) phosphate (TDClPP), tri-n-butyl phosphate (TNBP), tris (2-butoxyethyl) phosphate (TBOEP), 2-ethylhexyl diphenyl phosphate (EHDPP), tris (phenyl) phosphate (TPHP), and tri-m-tolyl phosphate (TmTP)) ranged from 58% to 100%. The concentration of Σ8OPEs ranged from 107 ng g−1 (dry weight - dw) (in the Worcester-Birmingham canal) to 52 ng g−1 (dw) in both Rivers Severn and Sowe). The highest ∑8OPE concentration for all study locations was recorded during the autumn months (September, October, and November), and concentrations of ∑8OPEs in sediment samples from the River Severn, River Tame, and River Sowe all varied inversely with the river level and flow rate. The average concentrations of each target OPE in each study location were lower than those reported elsewhere but fell broadly within the range of concentrations reported from other countries. The potential risk posed by target OPEs in all study locations was found to be low, except for a moderate risk identified for EHDPP in the Worcester - Birmingham canal.

Effects of TCPP and TCEP exposure on human corneal epithelial cells: Oxidative damage, cell cycle arrest, and pyroptosis

Tris(2-chloroisopropyl) phosphate (TCPP) and Tris(2-chloroethyl) phosphate (TCEP) are the widely used organophosphorus flame retardants indoors and easily accessible to the eyes as the common adhesive components of dust and particle matter, however, hardly any evidence has demonstrated their corneal toxicity. In this study, the adverse effects of TCPP, TCEP, and TCPP + TCEP exposure on human corneal epithelial cells (HCECs) were investigated. The cell viability and morphology, intracellular reactive oxygen species (ROS), cell cycle, and the expressions of cell cycle and pyroptosis-related genes were assessed to explain the underlying mechanisms. Compared to individual exposure, co-exposure to TCPP20+TCEP20 showed higher cytotoxicity with a sharp decrease of >30% in viability and more serious oxidative damage by increasing ROS production to 110.92% compared to the control group. Furthermore, the cell cycle arrested at the S phase (36.20%) was observed after combined treatment, evidenced by the upregulation of cyclin D1, CDK2, CDK4, CDK6, p21, and p27. Interestingly, pyroptosis-related genes GSDMD, Caspase-1, NLRP3, IL-1β, IL-18, NLRP1, and NLRC4 expressions were promoted with cell swelling and glowing morphology. Oxidative stress and cell cycle arrest probably acted as a key role in TCPP20+TCEP20-induced cytotoxicity and pyroptosis in HCECs. Our results suggested that TCPP20+TCEP20 co-exposure induced severer corneal damage, further illustrating its significance in estimating indoor health hazards to humans.

Human exposure to organophosphate esters in water and packed beverages

Drinks are an essential part of human diet, which makes them a source of human exposure to plasticizers such as organophosphate esters (OPEs). The current study provides new information about sixteen OPE levels in 75 different samples (tap water, packed water, cola drinks, juice, wine and hot drinks). Tap water mean levels (40.9 ng/L) were statistically higher than packed water mean levels (4.82 ng/L), mainly due to the contribution of tris(2-chloroisopropyl) phosphate (TCIPP) and tris(2-butoxyethyl) phosphate (TBOEP) that may come from PVC water pipes. Over 90% of samples presented at least one OPE, where regular cola drinks had the highest mean concentrations (2876 ng/L). There was a significantly higher presence of OPEs in added sugar beverages than sugar free drinks, especially for 2-ethylhexyl diphenyl phosphate (EHDPP), which might be related not only to packaging materials but to the added sugar content. Estimated daily intakes (EDIs) in normal and high-exposure scenarios were 2.52 ng/kg bw/day and 7.43 ng/kg bw/day, respectively. Human risk associated with beverages ingestion showed regular cola drinks, juice and tap water as the groups with the highest hazard quotients (HQs). Although OPE exposure was below to safety limits, it should be noted that EHDPP values for regular cola group must be cause of concern, and other routes of exposure such as food ingestion or air inhalation should be also considered.

Organophosphate flame retardants and plastics in soil from an abandoned e-waste recycling site: significant ecological risks derived from plastic debris

The distribution of 9 organophosphate flame retardants (OPFRs) was determined in plastic debris and soil samples separated from twenty soil samples collected from an abandoned e-waste recycling area. Tris-(chloroisopropyl) phosphate (TCPP) and triphenyl phosphate (TPhP) were the main chemicals, with median concentrations of 124-1930ng/g and 143-1170ng/g in soil, and 712-803ng/g and 600-953ng/g in plastics, respectively. Plastics contributed less than 10% of the total OPFR mass in bulk soil samples. No apparent OPFR distribution trend was observed in different sizes of plastics and soil. The ecological risks of plastics and OPFRs were estimated by the species sensitivity distributions (SSDs) method, which resulted in lower predicted no-effect concentrations (PNECs) of TPhP and decabromodiphenyl ether 209 (BDE 209) than the standard values derived from limited toxicity tests. In addition, the PNEC of polyethene (PE) was lower than the plastic concentration in the soil of a previous study. TPhP and BDE 209 had high ecological risks with risk quotients (RQs) > 0.1, and RQ of TPhP was among the highest values in literature.

Exposure to Organophosphate Ester Flame Retardants in the Occupational Environment

Rationale: Flame retardants (FR) are commonly used in consumer products, electronics, and safety equipment. Organophosphate esters (OPEs) are a sub-class of FRs and are widely used in various industries. Occupational risk of exposure is expected for many workers in various industries. However, the evidence of occupational exposure to OPE FRs is limited.&#x0D; Objectives: This study aimed to summarize evidence of occupational exposure to OPE FRs.&#x0D; Methods: A scoping review was conducted using Scopus (April-2022) and Google Scholar (January-2022). Search terms included ""occupational"", ""exposure"", ""flame retardants"", ""organophosphate"" and similar. Studies with quantitative measures of OPE exposure in workplaces were retained. Information on sample types, exposure levels, routes of exposure, and contextual information was extracted and summarized.&#x0D; Results: To-date, 15 studies have met the inclusion criteria. Groups most frequently assessed for OPE exposure include workers in e-waste recycling facilities and constructions. Published studies have described inhalation exposure, incidental ingestion, and dermal absorption routes. Sampling methods included active air sampling, passive air sampling, biological sampling, and dermal sampling. Triphenyl phosphate (TPhP), Tris(2-chloroisopropyl) phosphate (TCIPP), and Tris(2-chloroethyl) phosphate (TCEP) were the three most common sampled OPEs.&#x0D; Conclusions: There is evidence of OPE exposure in a limited number of industries and occupations at levels higher than the general population. The adverse health effects associated with OPE exposure included reproductive complications, endocrine disruption, and potential carcinogenicity. More research is needed to understand the prevalence and level of occupational exposure to OPE FRs and the potential work-related health impacts.
