Interaction of the Erythrocyte Lactate Transporter (Monocarboxylate Transporter 1) with an Integral 70-kDa Membrane Glycoprotein of the Immunoglobulin Superfamily

Treatment of intact erythrocytes with 4,4'-diisothiocyanostilbene-2, 2'-disulfonate (DIDS) causes irreversible inhibition and chemical labeling of the lactate transporter, monocarboxylate transporter 1 (MCT1) (Poole, R. C., and Halestrap, A. P. (1992) Biochem. J. 283, 855-862). In rat erythrocytes DIDS also causes cross-linking of MCT1 to another protein in the membrane to give a product of 130 kDa on SDS-polyacrylamide gel electrophoresis. Cross-linking is markedly reduced by those compounds that protect against irreversible inhibition of lactate transport by DIDS and enhanced by imposition of a pH gradient across the plasma membrane to recruit the substrate binding site of MCT1 to an exofacial conformation. These data indicate that DIDS cross-linking is via the same site on MCT1 as is responsible for inhibition of transport. Antibodies raised against the cross-linked conjugate react with proteins of approximately 40 kDa (MCT1) and 70 kDa on Western blots of erythrocyte membranes and an additional band of 130 kDa after treatment of erythrocytes with 100 microM DIDS. The 70-kDa protein that is cross-linked to MCT1 was purified and shown to contain N-linked carbohydrate; the apparent core molecular mass is 40 kDa. Amino acid sequencing showed that the protein is the rat equivalent of the membrane-spanning mouse teratocarcinoma glycoprotein GP-70, a member of the immunoglobulin superfamily related to basigin (Ozawa, M., Huang, R. P., Furukawa, T. , and Muramatsu, T. (1988) J. Biol. Chem. 263, 3059-3062). Possible implications of the specific interaction between MCT1 and this protein are discussed.

Stimulatory effect of fatty acid treatment on glucose utilization in human erythrocytes

We previously reported that treatment of human erythrocytes with bee venom phospholipase A 2 increased the rate of lactate production from glucose. This increase was suggested to be mediated through liberation of free fatty acids from membrane phospholipids. So, in the present study we examined the mechanism of stimulation of glycolysis by fatty acids. Treatment of intact erythrocytes with most of the 15 fatty acids tested resulted in stimulation of lactate production from glucose. Among the fatty acids tested, myristoleic acid showed the highest stimulatory activity. The ratio of moles of lactate produced to those of glucose utilized was about 1.9 in both myristoleic acid-treated and untreated cells. Treatment of erythrocytes with myristoleic acid did not affect the amount of 2,3-bisphosphoglycerate. Lactate production from d-glyceraldehyde, which is thought to be phosphorylated to d-glyceraldehyde 3-phosphate and then metabolized in the glycolytic pathway, was not at all affected by treatment of cells with myristoleic acid. The cross-over plot of glycolytic intermediates suggested that the enhancement of glycolysis was induced by activation of the 6-phosphofructokinase (PFK) step. Fatty acids incorporated into erythrocytes were found to be present predominantly in the cytoplasm rather than in the plasma membrane. The PFK activity, but not the hexokinase activity, in hemolysates was clearly increased by a set of fatty acids, and myristoleic acid was again the most potent. However, partially purified human erythrocyte PFK was not activated by the acid. We conclude that fatty acids stimulate glycolysis through activation of PFK in cooperation with some other component(s) in erythrocytes.

Proteolytic digestion of band 3 at an external site alters the erythrocyte membrane organisation and may facilitate malarial invasion

Invasion of human erythrocytes by Plasmodium falciparum is inhibited by chymostatin. This suggests that digestion of erythrocyte surface proteins by a protease with chymotrypsin-like activity may be involved in the invasion process. We find that treatment of intact erythrocytes with chymotrypsin cleaves the integral membrane protein, band 3, generating a major fragment with an apparent molecular weight of 58 kDa. We have used measurements of the rotational mobility of band 3, labelled with the phosphorescence probe, eosin-5-maleimide, as a monitor of the changes in the molecular organisation of the erythrocyte membrane which accompany band 3 cleavage. We report that the chymotrypsin treatment increases the rotational freedom of band 3, possibly due to conformational changes which disrupt its interaction with the underlying peripheral membrane proteins. We also show that chymotrypsin-treated erythrocytes undergo extensive endocytosis upon incorporation of exogenous fluorescently labelled phospholipid. We suggest that during the invasion process, digestion of band 3 by a chymotrypsin-like protease may induce a localised disruption of the erythrocyte membrane. This destabilised region of membrane may represent the site for the insertion of parasite-derived phospholipid, thus allowing the formation of the parasitophorous vacuole membrane.

Human erythrocyte sialidase is linked to the plasma membrane by a glycosylphosphatidylinositol anchor and partly located on the outer surface

Treatment of human erythrocyte ghosts with phosphatidylinositol-phospholipase C (PIPLC) from Bacillus cereus liberated the ghost-linked sialidase. Maximal release of sialidase (about 70% of total) was achieved by incubating ghosts at 37 degrees C for 60 min, at pH 6.0, with PIPLC (PIPLC total units/ghost protein ratio, 4.5 each time) added at the beginning of incubation and every 15 min (four subsequent additions). Liberated sialidase was fully resistant to at least four cycles of rapid freezing and thawing and to storage at 4 degrees C for at least 48 h. The liberated enzyme had an optimal activity at pH 4.2, degraded ganglioside GD1a better than methylumbelliferyl N-acetylneuraminic acid (about fourfold), and gave a Km value of 2.56 x 10(-4) M and an apparent Vmax of 2.22 mU per mg protein on GD1a. Treatment of intact erythrocytes with PIPLC (PIPLC total units/erythrocyte protein ratio, 8), under conditions where haemolysis was practically negligible, caused liberation of 10-12% of membrane linked sialidase, indicating that the enzyme is, at least in part, located on the outer surface of the erythrocyte membrane. It is concluded that the erythrocyte membrane sialidase is anchored by a glycosylphosphatidylinositol structure sensitive to PIPLC action, and is partly located on the outer surface.

Localization of the C termini of the Rh (rhesus) polypeptides to the cytoplasmic face of the human erythrocyte membrane.

We have raised a rabbit antiserum to a synthetic peptide corresponding to the C terminus (residues 400-416) of the Rh30A polypeptide. The rabbit antiserum reacted with the Rh30B (D30) polypeptide in addition to the Rh30A (C/c and/or E/e) polypeptide(s), indicating that these proteins share homology at their C termini. The antiserum did not react with erythrocyte membranes from an individual with Rh(null) syndrome. The rabbit antiserum immunoprecipitated Rh polypeptides from erythrocyte membranes and alkali-stripped membranes, but not from intact erythrocytes. Treatment of intact red cells with carboxypeptidase Y did not affect the reactivity of the antiserum, whereas treatment of alkali-stripped and unsealed erythrocyte ghost membranes resulted in the loss of antibody binding. Carboxypeptidase A treatment of intact erythrocytes and alkali-stripped membranes had no effect on antibody binding, indicating that the C-terminal domains of the Rh polypeptides contain lysine, arginine, proline, or histidine residues. These results show that the C termini of the Rh polypeptides are located toward the cytoplasmic face of the erythrocyte membrane. Treatment of intact radioiodinated erythrocytes with bromelain followed by immunoprecipitation with monoclonal anti-D gave a band of M(r) 24,000-25,000, indicating that the Rh30B (D30) polypeptide is cleaved at an extracellular domain close to the N or C terminus, with loss of the major radioiodinated domain. Immunoblotting of bromelain treated D-positive erythrocyte membranes with the rabbit antiserum to the C-terminal peptide revealed a new band of M(r) 6000-6500, indicating that the extracellular bromelain cleavage site is located near the C terminus of the molecule. The band of M(r) 6000-6500 was not obtained in erythrocyte membranes derived from bromelain treated D-negative erythrocytes. Erythrocytes of the rare -D- phenotype appear to either totally lack, or have gross alterations in, the Cc/Ee polypeptide(s), since the bromelain treatment of these cells resulted in the total loss of staining in the M(r) 35,000-37,000 region and the concomitant appearance of the new band of M(r) 6000-6500.

Involvement of ATP-dependent aminophospholipid translocation in maintaining phospholipid asymmetry in diamide-treated human erythrocytes

Crosslinking of membrane skeletal proteins such as spectrin by oxidation of their SH-groups can be provoked by treatment of intact erythrocytes with diamide. Shortly after exposure of human erythrocytes to diamide and despite the transverse destabilization of the lipid bilayer that was observed in these cells (Franck, P.F.H., Op den Kamp, J.A.F., Roelofsen, B. and Van Deenen, L.L.M. (1986) Biochim. Biophys. Acta 857, 127-130), no abnormalities could be detected regarding the asymmetric distribution of the phospholipids when probed by either the prothrombinase assay or brief exposure of the cells to a modified phospholipase A2 with enhanced membrane penetrating capacity. This asymmetry appeared to undergo dramatic changes however, when the ATP content of the cytosol had decreased to less than 10% of its original level during prolonged incubation of the treated cells. These observations indicate that the initial maintenance of phospholipid asymmetry in diamide-treated erythrocytes can be solely ascribed to the action of the ATP-dependent aminophospholipid translocase. This view is supported by experiments involving radiolabeled phospholipids of which trace amounts had been inserted into the outer membrane leaflet of diamide-treated red cells and which still showed a preferential translocation of both aminophospholipids in favour of the inner monolayer, be it that the efficiency of the translocase was found to be impaired when compared to control cells.

Ecto-enzyme activity of human erythrocyte adenosine deaminase.

Adenosine deaminase is found primarily in the cytoplasm of many cell types. In the human erythrocyte, about 30 per cent of the total adenosine deaminase activity is membrane associated, and about two-thirds of this is inactivated by treatment of intact erythrocytes with the nonpenetrating reagent diazotized sulfanilic acid, without affecting lactate dehydrogenase, a soluble cytoplasmic enzyme. This indicates that within the cell membranes, the catalytic site of about two-thirds of the adenosine deaminase faces the external medium, i.e., ecto adenosine deaminase. Localization of adenosine deaminase activity at the cell membrane is demonstrated directly by electron microscopy by use of the substrate 6-Chloropurine ribonucleoside, which is dechlorinated by adenosine deaminase to produce Cl-, which is precipitated at its locus of formation by added Ag+, and the precipitated AgCl converted into the electron dense Ag0 upon exposure to light. From the Hydropathic Profile of the amino acid sequence of adenosine deaminase it is evident that there are two hydrophobic domains of sufficient length to span a biological membrane, and it is proposed that these domains could function to anchor the enzyme to the membrane. The importance of adenosine deaminase is indicated by the fatal immuno-deficiency which results from untreated genetic adenosine deaminase deficiency. It may be important to determine whether the amount of ecto adenosine deaminase activity is better suited to assess the clinical status of adenosine deaminase deficient patients that the currently used total cellular enzyme activity.

Immunoblotting of the solubilised ghosts with anti-Pta located the antigen on a band with an Mr of 31,600, about 1,100 higher than that of sialoglycoprotein gamma. Binding to the same band was also observed when cytoskeleton preparations from Pt(a+) erythrocytes were immunoblotted with the antibody. Haemagglutination and immunoblotting experiments were consistent in demonstrating that the Pta antigen is not inactivated by treatment of intact erythrocytes with neuraminidase or trypsin but is destroyed by treatment with alpha-chymotrypsin, papain or pronase. The data indicate that the Pta antigen is carried on a 'new' erythrocyte membrane protein.

Activation, desensitization, and recycling of frog erythrocyte beta-adrenergic receptors. Differential perturbation by in situ trypsinization.

We have utilized limited in situ trypsinization of the adenylate cyclase-coupled beta-adrenergic receptor of frog erythrocytes to probe the processes of receptor activation, desensitization, and recycling. Treatment of intact erythrocytes with trypsin (1 mg/ml) for 1 h at 20 degrees C converts all the receptor peptides (identified by photoaffinity labeling with p-azido-125I-benzylcarazolol) from a Mr approximately 58,000 to a Mr approximately 40,000 species. Nonetheless, the trypsinized beta-adrenergic receptors bind agonists and antagonists with unaltered affinity and with no change in the number of binding sites. Moreover, the ability of the proteolyzed receptors to interact with the nucleotide regulatory protein to form a high affinity guanine nucleotide-sensitive state and to activate adenylate cyclase were also unaltered. However, upon exposure of intact cells to the agonist isoproterenol, trypsinized beta-adrenergic receptors were more rapidly and more completely cleared from the plasma membranes ("down-regulated") than untrypsinized receptors. Whereas down-regulated receptors from nontrypsinized cells appear to recycle to the cell surface after removal of the agonist, internalized trypsinized beta-adrenergic receptors do not recycle to the plasma membrane and appear to be degraded within the cell. Moreover, when internalized receptors, recovered in a light vesicle fraction, were fused with a heterologous adenylate cyclase system, untreated but not trypsinized receptors reconstituted catecholamine stimulation of the enzyme. These data suggest that the beta-adrenergic receptor contains a trypsin-sensitive site which is exposed on the outer surface of the plasma membrane. Proteolysis at this site releases a fragment which though not critically involved in either ligand binding or "effector coupling" might be important for anchoring the receptors in the plasma membrane. These data also suggest that in situ proteolysis of the receptors might serve as a physiological trigger for their internalization and degradation.

Endoglycosidase F cleaves the oligosaccharides from the glucose transporter of the human erythrocyte

The glucose transporter from human erythrocytes is a heterogeneously glycosylated protein that runs as a very broad band of average apparent M r 55 000 upon sodium dodecyl sulfate polyacrylamide gel electrophoresis. When the purified preparation of transporter, solubilized in Triton X-100, was treated with endoglycosidase F, much of it ran as a sharp band of M r 46 000 upon electrophoresis. Moreover, endoglycosidase F released 80% of the radioactivity in a preparation of the transporter labeled in its oligosaccharides with galactose oxidase and tritiated borohydride, and almost none of the remaining radioactivity was located in the M r 46 000 band. These results suggest that endoglycosidase F can release virtually all of the carbohydrate linked to the transporter polypeptide. A quantitative analysis of the gels was complicated by partial aggregation of polypeptides that occurs due to prolonged incubation in Triton X-100, but at least 65% of the protein in the preparation of purified transporter is the 46 kDa polypeptide. The extracellular domain of the transporter is very resistant to proteolysis; no cleavage occurred upon treatment of intact erythrocytes with seven different proteases at high concentration.

Cation fluxes and Na+-K+-activated ATPase activity in erythrocytes of patients with essential hypertension.

Recently it has been claimed that the active potassium influx in erythrocytes of patients with essential hypertension would be increased. In view of the diagnostic and possibly therapeutic potential of this claim, we have determined the Na+-K+ activated ATPase activity and the affinity of the enzyme for Na+, K+, and ATP in membranes isolated from erythrocytes of hypertensive (with and without medication) and normotensive subjects. Subsequently, the active (ouabain-sensitive) sodium and potassium fluxes and their ratios have been determined after treatment of intact erythrocytes either with cold or with p-chloromercuribenzene-sulfonate (PCMBS). Finally, in view of a subsequent claim that the furosemide-sensitive, ouabain-insensitive cation fluxes would be greatly reduced in erythrocytes of patients with essential hypertension, we have determined these fluxes in choline chloride medium containing ouabain with and without furosemide. For none of these parameters has any significant difference between hypertensive and normotensive subjects been found except for a decrease in the ouabain-sensitive K+ influx after cold treatment in hypertensives. This is also true for the hypertensive subjects who had a known hypertensive parent. It is concluded that the results do not support a role of Na+-K+ activated ATPase or the furosemide-sensitive cation carrier in the pathogenesis of essential hypertension, and that ouabain-sensitive and furosemide-sensitive cation flux determinations in erythrocytes do not seem to be useful for the diagnosis of this condition.

Trinitrobenzenesulfonic acid and fluorodinitrobenzene: probes to study local anesthetic effects in cell membranes.

The interaction of local anesthetics with intact erythrocytes was studied by monitoring the extent of reaction of phospholipids with trinitrobenzenesulfonic acid and fluorodinitrobenzene. Incubating erythrocytes with local anesthetics increases the amount of phosphatidylethanolamine and phosphatidylserine available for reaction with trinitrobenzenesulfonic acid and fluorodinitrobenzene. The order of potency of the local anesthetics corresponded to that reported for blocking nerve conduction: dibucaine greater than tetracaine greater than butacaine greater than lidocaine greater than procaine. Treatment of intact erythrocytes with 1 mM tetracaine at 37 degrees C allows 4-5% more of the phosphatidylethanolamine to react with trinitrobenzenesulfonic acid as compared to control cells. Treatment with tetracaine has no effect at 0 degrees C, a temperature at which there is only limited partitioning of the anesthetic into the bilayer. Kinetic analysis of the reaction with trinitrobenzene sulfonic acid showed that the increased number of reactive phosphatidylethanolamine molecules are located mainly on the outer half of the erythrocyte membrane. Tetracaine also increases the number of phosphatidylserine and phosphatidylethanolamine molecules in the erythrocyte membrane which are available to react with the penetrating probe fluorodinitrobenzene. The reaction with PE is increased from 67 to 77% and the reaction of PS is increased from 44 to 57%. Thus tetracaine affects both halves of the lipid bilayer.

The Transmembrane Proteins in the Plasma Membrane of Normal Human Erythrocytes

Abstract The arrangement of proteins in the normal human erythrocyte membrane was studied using lactoperoxidase-catalyzed iodination. The outer membrane surface of both intact erythrocytes and resealed ghosts was labeled with externally added lactoperoxidase. The cytoplasmic membrane surface was probed by sealing lactoperoxidase inside the resealed ghosts. Following iodination, the membrane components were separated on sodium dodecyl sulfate acrylamide gels, and the distribution of radioactive iodide into the various molecular weight classes was determined. The same components appear to be labeled on the external surface of resealed ghosts as are labeled in intact erythrocytes: a 90,000 molecular weight component (Protein 3), the major glycoprotein (periodate fuchsin sulfite (PAS)I), and two minor glycoproteins with apparent molecular weights of 40,000 (PAS II) and 27,000 (PAS III). However, PAS II is more extensively labeled in resealed ghosts than in intact erythrocytes, suggesting that alteration of the membrane has occurred during the course of the preparation of membranes and their resealing. Iodination of the internal surface of resealed ghosts reveals that all of the components labeled on the exterior surface also appear to be labeled at the cytoplasmic surface of the membrane. In addition, spectrin also is labeled at the inner surface of resealed ghosts. Separation of the partially purified glycoproteins by sodium dodecyl sulfate gel electrophoresis indicates that the region of PAS II is actually composed of at least two components, designated PAS II' and PAS II. The slower moving component, PAS II', appears to be labeled at both surfaces of the membrane, whereas the faster moving component, PAS II, which also stains more intensely with periodate fuchsin sulfite, is labeled only at the cytoplasmic surface of the membrane. Treatment of intact erythrocytes with pronase hydrolyzes PAS II, indicating that this component also spans the membrane. The differences in labeling between the major glycoprotein, PAS I, and PAS II indicate that these components are not identical, as has been suggested by others. Thus all five components detectable on the external surface of the normal human erythrocyte span the membrane and can be labeled on the interior surface.

Relative susceptibilities to neuraminidase of the glycoproteins of the human erythrorcyte

Release of sialic acid from the glycoproteins of the normal human erythrocyte surface by neuraminidase was investigated. The glycoproteins of the membrane were separated by electrophoresis in sodium dodecylsulfate polyacrylamide gels. Sialic acid was determined in the sliced gel by a modification of the 2-thiobarbituric acid method, revealing three sialic acid-containing glycoproteins. Treatment of intact erythrocytes with neuraminidase to remove varying amounts of sialic acid indicates that all the glycoproteins are essentially equally accessible to the neuraminidase when 20%–60% of the sialic acid is removed. Similar but not quite identical results were obtained with isolated erythrocyte membranes. Treatment of intact cells with the lectins concanavalin A or phytohemagglutinin-P resulted in shielding of about 25% and 50%, respectively, of the sialic acid from neuraminidase. Concanavalin A blocked sialic acid release over long time periods and with high concentrations of neuraminidase. In contrast, the sialic acid shielding by phytohemagglutinin-P can be overcome by high concentrations of neuraminidase. Both lectins were found to shield the various glycoproteins selectively, with different patterns of shielding. Wheat germ agglutinin exhibited no detectable effect on the susceptibility of the erythrocyte sialic acid to neuraminidase.
