Factors influencing subcanopy leaf and stolon exposure and associated absorption and translocation of herbicides in semidormant zoysiagrass

AbstractTurfgrass managers are concerned about zoysiagrass (Zoysia japonica Steud.) injury from nonselective herbicide treatment during winter dormancy. Research was conducted to assess factors affecting spray penetration into semidormant ‘Meyer’ zoysiagrass canopies and to evaluate absorption and translocation of [14C]glyphosate and [14C]glufosinate into green leaves and subtending stolons. Absorption of [14C]glyphosate and [14C]glufosinate was up to four times greater in stolons than in leaves. Zoysiagrass leaves treated with [14C]glufosinate had more rapid 14C absorption than those treated with [14C]glyphosate. More 14C translocated out of the treated area following [14C]glyphosate treatment compared with [14C]glufosinate and moved more readily from stolon to leaves than from leaves to stolon. When extended-range, flat-fan spray tips (XR) were positioned 61 cm above zoysiagrass, 73% and 11% of recovered colorant was extracted from dormant vegetation in the upper and lower canopy levels. Turbo TeeJet® spray tips (TTI) deposited fewer droplets into the upper canopy and more droplets into the middle and lower canopy regardless of position above the turf surface. Increasing pressure from 103 to 414 kPa increased droplet velocities from XR and TTI nozzles and decreased droplet diameters of XR nozzles. Droplet diameters were also substantially increased when using TTI nozzles compared with XR nozzles. Droplet diameter and associated mass were more determinant of turfgrass canopy penetration than droplet velocity. At 60 L ha−1 of carrier volume, 23% of colorant reached the lower canopy level, and this quantity increased by 2.3% per additional 100 L ha−1. When carrier volume was reduced from 584 to 60 L ha−1, 48% less colorant was delivered to the lower canopy level. Given that subcanopy stolons are always present and absorb more glyphosate and glufosinate than leaves, practices such as avoiding induction-type nozzles, raising spray height, and reducing spray volume can reduce herbicide delivery and potential injury to semidormant zoysiagrass.

First Case of Glyphosate Resistance in Bromus catharticus Vahl.: Examination of Endowing Resistance Mechanisms.

Bromus catharticus Vahl. has been used as a valuable forage crop, but it has also been noted as a weed of winter crops and an invader in several countries. In Argentina, a putative glyphosate-resistant population of B. catharticus was identified as a consequence of the lack of effective control with glyphosate in the pre-sowing of wheat. Plant survival and shikimate accumulation analysis demonstrated a lower glyphosate-sensitivity of this population in comparison to a susceptible B. catharticus population. The resistant population was 4-fold more resistant to glyphosate than its susceptible counterpart. There was no evidence of target-site mechanisms of glyphosate resistance or an enhanced capacity to metabolize glyphosate in the resistant population. However, the resistant plants showed a lower foliar retention of glyphosate (138.34 μl solution g−1 dry weight vs. 390.79 μl solution g−1 dry weight), a reduced absorption of 14C-glyphosate (54.18 vs. 73.56%) and lower translocation of 14C-glyphosate from the labeled leaf (27.70 vs. 62.36%). As a result, susceptible plants accumulated a 4.1-fold higher concentration of 14C-glyphosate in the roots compared to resistant plants. The current work describes the first worldwide case of glyphosate resistance in B. catharticus. A reduced foliar retention of herbicide, a differential rate of glyphosate entry into leaves and an altered glyphosate translocation pattern would be the most likely mechanisms of glyphosate exclusion.

Effect of Fertiactyl® on the absorption and translocation of 14C-glyphosate in young eucalyptus plants

Fertiactyl® is a foliar fertilizer with the potential to minimize the phytotoxicity effects caused by glyphosate drift in eucalyptus plants. As the interactions of the glyphosate and Fertiactyl® in tank mix on the plant behavior are not yet known, the objective was to evaluate the absorption and translocation of 14C-glyphosate, applied isolated and mixed in tank with Fertiactyl®, in young eucalyptus plants (clone I-144, Eucalyptus urophylla x E. grandis). The addition of Fertiactyl® to the mixture of 14C-glyphosate reduced the absorption by 94.3% in relation to the total absorbed at the end of the evaluation compared to plants treated only with 14C-glyphosate, i.e., Fertiactyl® protected the eucalyptus plants of the glyphosate intoxication by drift. The translocation rates from the treated leaves to the rest of the shoots and roots were low (<2% of the total recovered) in both treatments, suggest that restricted translocation is a mechanism of natural tolerance to glyphosate in plants of clone I-144. It is concluded that Fertiactyl®, mixed in the solution with glyphosate, protects young eucalyptus plants against glyphosate drift by reducing the amount of herbicide absorbed.

Fertiactyl® in mixture with glyphosate decreases herbicide absorption and translocation in coffee seedlings

The application of glyphosate to coffee crops can cause injuries to plants. Fertiactyl® foliar fertilizer reduces injuries when mixed with glyphosate; however, it is important to establish which mechanisms are responsible for this protective action. This study aimed to evaluate the absorption and translocation of glyphosate applied separately and in mixture with Fertiactyl® in coffee seedlings. Absorption and translocation were performed with 14C-glyphosate applied separately and in mixture with Fertiactyl® at 0, 6, 12, 24, 48, 96, and 144 hours after application (HAA). Most of the 14C-glyphosate applied to coffee seedlings was not absorbed. The 14C-glyphosate applied separately had a higher absorption by coffee seedlings (6.5%) than in a mixture with Fertiactyl® (2.7%) at 144 HAA. The maximum translocation of the 14C-glyphosate applied separately was 0.69% at 81.2 HAA and in mixture with Fertiactyl® was 0.41% at 41.2 HAA. The treated leaves retained a higher percentage of 14C-glyphosate when applied separately (5.6% at 144 HAA) than in a mixture with Fertiactyl® (2.2% at 144 HAA). Low translocation (<1%) for the rest of the plant shoots was observed both for the 14C-glyphosate applied separately and in combination with Fertiactyl®. Therefore, Fertiactyl® decreased the absorption and translocation of 14C-glyphosate in coffee seedlings.

Light availability interferes with absorption and translocation of 14C-glyphosate in Urochloa brizantha cv. Marandu plants

This research aimed to evaluate the absorption and translocation of 14C-glyphosate in Urochloa brizantha cv. Marandu subjected to different light availabilities. Six light conditions were studied: application followed by 24, 48 or 72 h of dark, 72 h of light, 24 h of dark + 48 h of light and daytime in different evaluation times: 0, 3, 6, 12, 24, 48, and 72 h after application of 14C-glyphosate (1.67 kBq/plant). The herbicide absorption was higher in plants with less light availability at 72 HAA (48 and 21% for 72 h of darkness and 72 h of light, respectively); however, there was a greater translocation of 14C-glyphosate to roots in the high availability of light (23 and 5% for 72 h of light and 72 h of dark, respectively). Thus, in U. brizantha plants, under conditions of greater light availability, 14C-glyphosate was less absorbed, but the product was more translocated. It is recommended that farmers to apply glyphosate at daytime since the light positively favored translocation, which is essential to the herbicidal activity.

Susceptible and Glyphosate-Resistant Palmer Amaranth (&amp;lt;i&amp;gt;Amaranthus palmeri&amp;lt;/i&amp;gt;) Response to Glyphosate Using C&amp;lt;sup&amp;gt;14&amp;lt;/sup&amp;gt; as a Tracer: Retention, Uptake, and Translocation

The foliar retention, absorption, translocation, and diffusion of glyphosate in glyphosate resistant-(R) and susceptible (S)-Palmer amaranth populations from seed collected in Georgia in 2007 were examined. The R population of Palmer amaranth had an elevated copy number of the EPSPS gene conferring the mechanism of resistance. When applications of 14C-glyphosate to a single leaf followed entire plant treatment with glyphosate, the distribution percentages were similar for R and S for the above and below treated leaves when harvested at 1, 6, 12, 24, and 48 hours after treatment (HAT). There were initially no differences between R and S at 1 HAT with an average of 8% absorption for both biotypes. However, data indicated that glyphosate absorption increased for R-Palmer amaranth reaching 41% within 6 HAT and was significantly different (P = 0.01) from the 28% absorbed by S-Palmer amaranth. Glyphosate resistant and susceptible Palmer amaranth averaged 44% 14C-glyphosate absorption by 24 HAT. There were no differences for 14C-glyphosate Bq/mg of plant tissue between R and S for the above the treated leaf and below the treated leaf portions of plants at 1, 6, 12, 24, or 48 HAT. However, root accumulation of 14C-glyphosate in plant tissue was significantly greater by 12 HAT for the roots of R (1.21 Bq/mg) than for S (0.51 Bq/mg). The treated leaf of the R-Palmer amaranth plants exhibited greater translocation of 14C-glyphosate in Bq/mg of tissue than the susceptible over time, indicating no detrimental effect or cost of fitness due to EPSPS gene amplification. Additionally, there were no differences in glyphosate retention in leaf discs assays between R and S biotypes. In spite of an average of 6.5 Bq efflux out of R and S leaf discs after 15 minute, only 0.4 Bq was retained after 150 minutes. Glyphosate was not retained over time in the leaf discs for R and S, and there were no biotype differences within bathing times. However, the rate of efflux (the slope of the curves) was greater for the R biotype. These data support the reported gene amplification non-target site glyphosate resistance mechanism in Palmer amaranth.

Investigating mechanism of glyphosate resistance in a common ragweed (Ambrosia artemisiifolia L.) biotype from Nebraska

Common ragweed is a weed in the midwestern United States and eastern Canada that is difficult to control due to the evolution of an important resistance to multiple herbicides including glyphosate. Recently, a common ragweed biotype with 19-fold glyphosate resistance was confirmed in Nebraska. The objective of this study was to determine the mechanism of glyphosate resistance in a common ragweed biotype from Nebraska. Both target site and non-target site based mechanisms of glyphosate resistance were investigated using glyphosate-resistant (GR) and known glyphosate-susceptible (GS) common ragweed biotypes. A lower amount of shikimate was accumulated in the GR (≤65 μg mL−1) compared with the GS (≥80 μg mL−1) biotype at all glyphosate concentrations tested. First Resistance Mechanisms Characterization in Glyphosate-Resistant Leptochloa virgata.

Leptochloa virgata (L.) P. Beauv. is an annual weed common in citrus groves in the states of Puebla and Veracruz, Mexico limiting their production. Since 2010, several L. virgata populations were identified as being resistant to glyphosate, but studies of their resistance mechanisms developed by this species have been conducted. In this work, three glyphosate-resistant populations (R8, R14, and R15) collected in citrus orchards from Mexico, were used to study their resistance mechanisms comparing them to one susceptible population (S). Dose-response and shikimic acid accumulation assays confirmed the glyphosate resistance of the three resistant populations. Higher doses of up to 720 g ae ha-1 (field dose) were needed to control by 50% plants of resistant populations. The S population absorbed between 7 and 13% more 14C-glyphosate than resistant ones, and translocated up to 32.2% of 14C-glyphosate to the roots at 96 h after treatment (HAT). The R8, R14, and R15 populations translocated only 24.5, 26.5, and 21.9%, respectively. The enzyme activity of 5-enolpyruvyl shikimate-3-phosphate synthase (EPSPS) was not different in the S, R8 and R14 populations. The R15 Population exhibited 165.9 times greater EPSPS activity. Additionally, this population showed a higher EPSPS basal activity and a substitution in the codon 106 from Proline to Serine in the EPSPS protein sequence. EPSPS gene expression in the R15 population was similar to that of S population. In conclusion, the three resistant L. virgata populations show reduced absorption and translocation of 14C-glyphosate. Moreover, a mutation and an enhanced EPSPS basal activity at target-site level confers higher resistance to glyphosate. These results describe for the first time the glyphosate resistance mechanisms developed by resistant L. virgata populations of citrus orchards from Mexico.

Physiology of Glyphosate-Resistant and Glyphosate-Susceptible Palmer Amaranth (Amaranthus palmeri) Biotypes Collected from North Carolina

Glyphosate-resistant (GR) biotypes of Palmer amaranth are now commonly found across the southern United States. Experiments were conducted to characterize physiological differences between a GR biotype and a glyphosate-susceptible (GS) biotype from North Carolina. The GR biotype had an 18-fold level of resistance based upon rates necessary to reduce shoot fresh weight 50%. Shikimate accumulated in both biotypes following glyphosate application, but greater concentrations were found in GS plants. Absorption and translocation of14C-glyphosate were studied in both biotypes with and without an overspray with commercial glyphosate potassium salt (840 g ae ha−1) immediately prior to14C-glyphosate application. Greater absorption was noted 6 h after treatment (HAT) in GS compared with GR plants, but no differences were observed at 12 to 72 HAT. Oversprayed plants absorbed 33 and 61% more14C by 48 and 72 HAT, respectively, than plants not oversprayed.14C distribution (above treated leaf, below treated leaf, roots) was similar in both biotypes. Together, these results suggest that resistance in this biotype is not due to an altered target enzyme or translocation but may be in part due to the rate of glyphosate absorption. These results also are consistent with resistance being due to increased gene copy number for the target enzyme.

Zinc Antagonizes Glyphosate Efficacy on Yellow Nutsedge (Cyperus esculentus)

Growers of glyphosate-resistant crops apply micronutrients tank-mixed with glyphosate to save time and production costs. Therefore, effect of zinc (Zn), as Zn sulfate, on absorption, translocation, and efficacy of glyphosate on yellow nutsedge was investigated. Glyphosate at 850 g ae ha−1provided 90% yellow nutsedge control at 5 wk after treatment (WAT). Presence of Zn at 1,000 ppmw in the glyphosate spray solution reduced yellow nutsedge control to 24 and 8%, 3 and 5 WAT, respectively. Yellow nutsedge control decreased with increasing Zn level (500 to 2,000 ppmw) in the spray solution. Yellow nutsedge treated with higher rates of Zn tank-mixed with glyphosate produced more tubers and tillers per plant than untreated plants. An abrupt decrease in absorption and translocation of14C–glyphosate occurred between 500 and 1,000 ppmw Zn. The antagonistic effect of Zn on glyphosate depended mainly on reduced absorption and translocation of14C–glyphosate within treated tissues. Less than 10% of applied14C–glyphosate was absorbed when glyphosate was mixed with 1,000, 2,000, or 4,000 ppmw Zn as compared with 85% absorption for glyphosate alone. These treatments inhibited &gt; 90% of14C–glyphosate translocation out of the treated leaf and &gt; 50% of tuber translocation relative to glyphosate alone. Results indicate that micronutrients containing Zn are not suitable for tank-mixing with glyphosate.

Translocation and Absorption of Glyphosate in Flowering Sicklepod (Senna obtusifolia)

Sicklepod is a competitive and prolific weed that emerges throughout the crop season. Glyphosate applications to flowering sicklepod greatly reduce seed production, but there is limited information on glyphosate translocation in flowering weeds. Therefore, a laboratory study was conducted to document the absorption and translocation of14C-glyhosate in flowering sicklepod. Flowering sicklepod plants were treated with a14C-glyphosate solution, and14C-glyphosate absorption and translocation was quantified by scintillation spectrometry. Absorption and translocation of14C-glyphosate in flowering sicklepod were similar for both 0.21 and 0.42 kg ae ha−1glyphosate. Although the treated leaflets retained approximately 50% of the recovered14C-glyphosate, movement of the herbicide was both acropetal and basipetal, with the highest14C-glyphosate concentrations in the shoot below the treated leaf and the roots. By 96 h after treatment, sicklepod buds and flowers had abscised, but analysis of the structures revealed accumulation of 2% of the recovered14C-glyphosate. Based on results of the study, effects of glyphosate accumulation in buds and flowers combined with plant stress associated with the primary and secondary effects of glyphosate result in bud and flower abscission, drastically reducing sicklepod seed production.

Absorption and Translocation of Glyphosate and Sucrose in Glyphosate-resistant Cotton

Studies were conducted to evaluate absorption and translocation of 14C-glyphosate in glyphosate-resistant (GR) cotton. Both commercial GR cotton events [glyphosate-resistant event 1, marketed as Roundup Ready®, released 1997 (GRE1), and glyphosate-resistant event 2, marketed as Roundup Ready Flex®, released 2006 (GRE2)] were evaluated at the four-leaf and eight-leaf growth stages. Plants were harvested at 1, 3, 5, and 7 d after treatment (DAT). Glyphosate absorption, as a percentage of applied, increased over time with 29 and 36% absorption at 7 DAT in four-leaf GRE1 and GRE2 cotton, respectively. In eight-leaf cotton, glyphosate absorption (33% at 7 DAT) was not different between events. Glyphosate translocation patterns were not different between events or harvest timings and exhibited a source–sink relation. Reduced Translocation Is the Cause of Antagonism of Glyphosate by MSMA in Browntop Millet (Brachiaria ramosa) and Palmer Amaranth (Amaranthus palmerii)

Studies were conducted in growth chambers to characterize absorption and translocation of14C-glyphosate applied alone or in mixture with MSMA in browntop millet and Palmer amaranth. MSMA antagonized activity of glyphosate in both weed species. Absorption of14C-glyphosate in Palmer amaranth was rapid and increased with time from 11.1% at 0.5 h after treatment to 68.1% at 168 HAT. Absorption of14C-glyphosate in browntop millet ranged from 1.6% at 0.5 HAT to 39.1% at 168 HAT. MSMA in mixture with glyphosate did not affect the absorption of glyphosate. In browntop millet, only 2.8% of the applied radioactivity translocated out of the treated leaf to the rest of the plant when glyphosate was applied in mixture with MSMA compared to 10.8% when glyphosate was applied alone at 72 HAT. Similarly, in Palmer amaranth, 3.2% of the applied radioactivity had translocated out of the treated leaf when glyphosate was applied in mixture with MSMA compared to 10.6% when glyphosate was applied alone. Reduced translocation appears to be the cause of the previously observed antagonism of glyphosate by MSMA.

Efeito de formulações na absorção e translocação do glyphosate em soja transgênica

Este trabalho teve como objetivo avaliar a absorção e translocação de glyphosate em diferentes formulações por plantas de soja (variedade CD 219RR). Para isso, aplicou-se o 14C-glyphosate misturado à calda em três formulações comerciais (Roundup Ready® e R. Transorb®, ambas contendo o sal de isopropilamina, e Zapp Qi®, formulado à base do sal potássico), quando as plantas apresentavam o segundo trifólio completamente expandido. Transcorridas 4, 16, 40 e 64 horas após a aplicação, as plantas foram coletadas e fracionadas, separando-se a folha de aplicação (trifólio), a parte aérea, as raízes e os nódulos radiculares. O 14C-glyphosate não-absorvido foi recuperado e contado por meio da lavagem da folha (metanol 80%). Entre as formulações foi observada variação na penetração e na translocação do 14C-glyphosate para as diferentes partes avaliadas. Todavia, em todas as formulações a maior absorção se deu nos intervalos posteriores a 16 horas da aplicação. Em relação ao total de herbicida encontrado nas plantas de soja, maior percentual na parte aérea foi observado quando se aplicou o Zapp Qi® (sal potássico) e, nas raízes, o R. Transorb® (sal de isopropilamina). Detectou-se a presença de 14C glyphosate nos nódulos radiculares das plantas em todos os tratamentos, sendo o maior percentual observado quando se utilizou R. Transorb®, 40 horas após a aplicação (0,13% do total medido ou 0,4% considerando somente o total presente na planta). Os resultados reforçam a hipótese de que o glyphosate pode prejudicar a simbiose entre rizóbio e soja, uma vez que o microssimbionte também apresenta em seu metabolismo a EPSPS, sensível a esse herbicida.

Comparison of efficacy, absorption and translocation of three glyphosate formulations on velvetleaf (Abutilon theophrasti)

Growth analysis, absorption and translocation studies were conducted to compare a 1-aminomethanamide dihydrogen tetraoxosulfate (GLY-A) formulation of glyphosate with two isopropylamine (GLY-IPA-1, GLY-IPA-2) formulations of glyphosate on velvetleaf. The two isopropylamine formulations differed by the presence of a surfactant in the formulation, GLY-IPA-1 containing surfactant whereas GLY-IPA-2 did not. Four- to six-leaf velvetleaf was treated with GLY-A and GLY-IPA-1 and GLY-IPA-2 (0, 50, 67, 89, 119, 158, 280, 420, 560 and 840 g AE ha(-1)) with and without ammonium sulfate (AMS; 20 g L(-1)). GLY-A and GLY-IPA-2 included a non-ionic surfactant (2.5 mL L(-1)) in the spray solution at all herbicide concentrations. No additional surfactant was added to GLY-IPA-1. The IC50 value for GLY-A was 88 g AE ha(-1) compared with 346 and 376 g AE ha(-1) for GLY-IPA-1 and GLY-IPA-2 respectively in the absence of AMS. When AMS (20 g L(-1)) was added to the spray solution, the estimated IC50 values were 143, 76 and 60 g AE ha(-1) for GLY-IPA-1, GLY-IPA-2 and GLY-A respectively. Absorption of 14C-glyphosate into the third leaf of five- to six-leaf velvetleaf was three- to sixfold greater 72 h after treatment (HAT) when applied as GLY-A compared with GLY-IPA-1 and GLY-IPA-2 respectively in the absence of AMS. AMS (20 g L(-1)) increased absorption of 14C-glyphosate in all glyphosate formulations two- to threefold, but differences among the formulations remained. Approximately three- and sixfold more 14C-glyphosate applied as GLY-A had translocated out of the treated leaf compared with GLY-IPA-1 and GLY-IPA-2 respectively by 72 HAT. Adding AMS (20 g L(-1)) increased translocation of 14C-glyphosate out of the treated leaf approximately 2.5-fold for all three formulations. The increased efficacy of GLY-A versus GLY-IPA-1 and GLY-IPA-2 on velvetleaf is due to the greater rate of absorption and subsequent translocation of glyphosate out of the treated leaf. AMS increased the efficacy of all three formulations by increasing absorption and translocation of glyphosate in the plant.

Glyphosate efficacy, absorption, and translocation in pitted morningglory (Ipomoea lacunosa)

Greenhouse and laboratory studies were conducted to examine the effects of site of plant exposure to glyphosate spray on efficacy, absorption, and translocation in pitted morningglory. Absorption of 14C-glyphosate in four-leaf pitted morningglory gradually increased with time from 19% at 1 h after treatment (HAT) to 44% at 192 HAT. The amount of 14C translocated with time ranged from 0.4% at 1 HAT to 25% at 192 HAT. Vining 1-m tall plants were controlled 75 to 100% when the top-, middle-, bottom one-third, or entire plant was treated with 1.38 or 2.76 kg ha−1 glyphosate, with control affected more by glyphosate rate than plant section exposed to glyphosate spray. Absorption of 14C-glyphosate at 96 HAT was similar whether it was applied to the top-, middle-, bottom one-third, or entire plant of 1-m tall pitted morningglory. The amount of 14C translocated out of the treated area (5 to 6%) did not differ whether it was applied to top-, middle-, or bottom one-third plant section. Results indicate that absorption and translocation of 14C-glyphosate in pitted morningglory was rapid and increased with time. Treating any one-third section of pitted morningglory plants was as effective as entire plant exposure, and control with glyphosate is more affected by rate than the degree of plant exposure to glyphosate.

Influence of formulation and glyphosate salt on absorption and translocation in three annual weeds

Absorption and translocation of three commercial formulations of glyphosate, the isopropylamine salt formulated as Roundup Ultra™ (IPA1) and Roundup UltraMax™ (IPA2) and the diammonium salt formulated as Touchdown™ IQ (DA), were compared in three- to five-leaf velvetleaf, common waterhemp, and pitted morningglory. Absorption of 14C-glyphosate in velvetleaf was not significantly different among the three formulations up to 50 h after treatment (HAT). More absorption of 14C-glyphosate occurred in the IPA1 (26.0%) vs. the IPA2 (17.7%) formulation over 74 h. Of the total 14C-glyphosate absorbed, 20 to 35% was translocated from the treated leaf to the rest of the plant. Initial absorption of 14C-glyphosate was rapid in common waterhemp with the IPA1 (42.7%) and IPA2 (30.7%) formulations; both were higher compared with absorption of the DA formulation (11.5%) by 2 HAT. These differences continued up to 26 HAT, but no differences were evident by 74 HAT. Up to 65% of the 14C-glyphosate absorbed was translocated out of the treated leaf by 74 HAT, with roots the primary sink. Initial absorption of 14C-glyphosate was slow in pitted morningglory compared with the other species. More foliar absorption occurred in plants treated with the DA (13.6%) vs. the IPA2 formulation (4.9%) by 6 HAT. Absorption beyond 26 HAT was not different among the three glyphosate formulations. Translocation of 14C-glyphosate to roots was 27% greater as the DA salt than IPA1 and IPA2 by 74 HAT. The distribution pattern of glyphosate was similar in all species; phosphorimages demonstrated movement both acropetal and basipetal, with accumulation in roots greater than in any other plant parts. An efficacy study parallel to the 14C study showed no difference among the three glyphosate formulations on the species investigated at both 74 HAT and 2 wk after treatment.

Droplet size affects glyphosate retention, absorption, and translocation in corn

Abstract The effect of droplet size on retention, absorption, and translocation of 14C-glyphosate was studied in glyphosate-resistant corn. Fine, medium, and coarse spray droplets were studied using a track-sprayer equipped with commercially available nozzles. Glyphosate-resistant corn was used to obtain measurements at field use rates in the absence of phytotoxicity. Spray retention on corn leaves was calculated based on recovered glyphosate per leaf area, and retention was higher with application of fine droplets (47%) than with application of coarse (38%) and medium (37%) droplets. Absorption in corn leaves was directly correlated with droplet size and reached a plateau 1 d after treatment (DAT) for all droplet sizes. Environmental factors affecting absorption and bio-efficacy of glyphosate in Florida beggarweed ( Desmodium tortuosum)

Experiments were conducted in controlled growth cabinets to examine the effect of air temperature (16°C, 22°C, 35°C) or relative humidity (RH, 45%, 70%, 95%) on 14C-glyphosate distributions in Desmodium tortuosum and bio-efficacy. Uptake and translocation of 14C-glyphosate were significantly higher at 22°C or 95% RH than 16°C and 35°C or 45% and 70% RH at all harvest times. These values were also significantly higher when influence of temperature or RH recorded when the effects across harvest times (6, 24, 48, 72 h) were averaged. Further, uptake and translocation were significantly higher at 48 h than the other harvest time when the effects across temperatures (16°C, 22°C, 35°C) were averaged. Maximum translocation was obtained at 72 h harvesting time when the effects of RH (45%, 70%, 95%) were averaged. When glyphosate was sprayed to beggarweed, the maximum foliar damage was observed 19% at 22°C or 35°C and 23% at 95% RH. Bioefficacy results were similar to 14C-glyphosate translocation at 22°C and 95% RH. The results indicated that temperature or RH influenced the absorption process of glyphosate and the optimum temperature and relative humidity for Desmodium control were 22°C and 95% RH.

Absorption and translocation of glyphosate in glyphosate-resistant cotton as influenced by application method and growth stage

The influence of herbicide placement and plant growth stage on the absorption and translocation patterns of 14C-glyphosate in glyphosate-resistant cotton was investigated. Plants at four growth stages were treated with 14C-glyphosate on a 5-cm2 section of the stem, which simulated a postemergence-directed spray (PDS) application, or on the newest mature leaf, which simulated a postemergence (POST) application. Plants were harvested 3 and 7 d after treatment and divided into the treated leaf or treated stem, mature leaves, immature leaves and buds, stems, roots, fruiting branches (including the foliage on the fruiting branch), squares, and bolls. The PDS versus POST application main effect on absorption was significant. Absorption of 14C-glyphosate applied to stem tissue was higher in PDS applications than in POST applications. Plants receiving PDS applications absorbed 35% of applied 14C-glyphosate, whereas those receiving POST applications absorbed 26%, averaged over growth stages at application. Absorption increased from the four-leaf growth stage to the eight-leaf stage in POST applications but reached a plateau at the eight-leaf stage. Plants with PDS applications showed an increase in absorption from the four- to eight- to twelve-leaf stages and reached a plateau at the 12-leaf stage. Translocation of 14C-glyphosate to roots was greater at all growth stages with PDS treatments than with POST treatments. Herbicide placement did not affect translocation of 14C-glyphosate to squares and bolls. Squares and bolls retained 0.2 to 3.7% of applied 14C-glyphosate, depending on growth stage. Separate studies were conducted to investigate the fate of foliar-applied 14C-glyphosate at the four- or eight-leaf growth stages when harvested at 8- or 10-leaf, 12-leaf, midbloom (8 to 10 nodes above white bloom), and cutout (five nodes above white bloom, physiological maturity) stages. Thirty to 37% of applied 14C-glyphosate remained in the plant at cutout in four- and eight-leaf treatment stages, respectively. The concentration of 14C-glyphosate in tissue (Bq g−1 dry weight basis) was greatest in mature leaves and immature leaves and buds in plants treated at the four-leaf stage. Plants treated at the eight-leaf stage and harvested at all growth stages except cutout showed a higher concentration of 14C-glyphosate in squares than in other plant tissue. Accumulation of 14C-glyphosate in squares reached a maximum of 43 Bq g−1 dry weight at harvest at the 12-leaf stage. This concentration corresponds to 5.7 times greater accumulation of 14C-glyphosate in squares than in roots, which may also be metabolic sinks. These data suggest that reproductive tissues such as bolls and squares can accumulate 14C-glyphosate at higher concentrations than other tissues, especially when the herbicide treatment is applied either POST or PDS during reproductive stages (eight-leaf stage and beyond).
