Detection and Quantification of Carbohydrate-Deficient Transferrin by MALDI-Compatible Protein Chips Prepared by Ambient Ion Soft Landing.

Transferrin is synthetized in the liver and is the most important iron-transport carrier in the human body. Severe alcohol consumption leads to alterations in glycosylation of transferrin. Mass spectrometry can provide fast detection and quantification of transferrin isoforms because they have different molecular masses. In this study, we used antibody chips in combination with MALDI-TOF MS for the detection and quantification of transferrin isoforms. Protein chips were prepared by functionalization of indium tin oxide glass using ambient ion soft landing of electrosprayed antitransferrin antibody. Two microliters of patient serum was applied on the antibody-modified spots, and after incubation, washing, and matrix deposition, transferrin isoforms were detected by MALDI-TOF MS. Peak intensities of each transferrin form were used to calculate total carbohydrate-deficient transferrin (CDT). The CDT values obtained by the MALDI chip method were compared with the results obtained by a standard capillary electrophoresis (CE). The chip-based MALDI-TOF MS method was used for enrichment and detection of CDT from human serum. A sample cohort from 186 patients was analyzed. Of these samples, 44 were positively identified as belonging to alcoholic patients, whereas 142 were negative by the MALDI chip approach. The correlation of the data obtained by the CE and the chip-based MALDI was r = 0.986, 95% CI. Functionalized MALDI chips modified by antitransferrin antibody prepared by ambient ion soft landing were successfully used for detection and quantification of CDT from human sera.

A novel quantification strategy of transferrin and albumin in human serum by species-unspecific isotope dilution laser ablation inductively coupled plasma mass spectrometry (ICP-MS)

Species-specific (SS) isotope dilution analysis with gel electrophoresis (GE)-laser ablation (LA)-ICP-MS is a promising technique for the quantification of particular metal-binding proteins in biological samples. However, unavailable isotopically enriched spike and metal losses in GE separation are main limitations for SS-isotope dilution PAGE-LA-ICP-MS. In this study, we report for the first time the absolute quantification of transferrin (Tf) and albumin (Alb) in human serum by non-denaturing (native) GE combined with species-unspecific isotope dilution mass spectrometry (IDMS). In order to achieve a homogeneous distribution of both protein and isotope-enriched spike (simulated isotope equilibration), immersing the protein strips with 34S spike solution after gel electrophoresis was demonstrated to be an effective way of spike addition. Furthermore, effects of immersion time and 34S spike concentration were investigated to obtain optimal conditions of the post-electrophoresis isotope dilution method. The relative mass of spike and ablated sample (msp/msam) in IDMS equation was calculated by standard Tf and Alb proteins, which could be applied to the quantification of Tf and Alb in ERM-DA470k/IFCC for method confirmation. The results were in agreement with the certified value with good precision and small uncertainty (1.5–3%). In this method, species-specific spike protein is not necessary and the integrity of the heteroatom-protein could be maintained in sample preparation process. Moreover, the application of species-unspecific isotope dilution GE-LA-ICP-MS has the potential to offer reliable, direct and simultaneous quantification of proteins after conventional 1D and 2D gel electrophoretic separations.

Quantification of Transferrin in Human Serum Using Both QconCAT and Synthetic Internal Standards

Transferrin, an iron transport protein, is a clinically important biomarker in diseases such as iron-deficiency anemia. Current diagnostic methods for transferrin levels lack quantitative accuracy, suggesting the need for alternative approaches like LC-MS with isotope-labeled peptides as internal standards. Besides solid-phase synthesis, isotope-labeled peptides are also generated by a method called QconCAT where peptides are expressed from DNA in the presence of heavy isotope media. After evaluation of the expressed QconCAT, this study compares transferrin levels obtained by synthetic peptides versus QconCAT peptides as internal standards. Transferrin levels obtained by both internal standards give overlapping, or nearly overlapping, uncertainty values and are near ≈200 mg/dL of transferrin in human serum. Close agreement between the two methods suggests that the quantitative values are reasonable. Using QconCAT and synthetic peptides in parallel gives a refined focus on method development, and the resulting methods should be applicable to other clinically relevant proteins.

Comparison of surface plasmon resonance transferrin quantification with a common immunoturbidimetric method

Objectives The variation in the results from different clinical laboratories testing the same patient sample using classical methods for serum transferrin determination can affect interpretation. We developed a surface plasmon resonance (SPR) method for serum transferrin assay that may aid standardization. Design and methods Quantification of transferrin was performed by direct analysis on a Biacore system (GE Healthcare). We assessed analytical performance of this new assay and compared it to a common immunoturbidimetric method (Beckman Coulter). Results Intra-run coefficients of variation (CV) were up to 1.10% and inter-day CVs were up to 2.10%. The comparison of this new SPR assay (Biacore) with a classical immunoturbidimetric method (Beckman Coulter) showed a good correlation ( r = 0.959), and the Bland and Altman plot with no global significant difference. Conclusions According excellent achievement of SPR transferrin determination, we suggest its use as an independent method for comparison different immunoassays. This could be an important tool to help clinical laboratories to deliver standardized values, essential for the decision to investigate genetic hereditary hemochromatosis.

Absolute quantification of transferrin in blood samples of harbour seals using HPLC-ICP-MS

Harbour seals (Phoca vitulina) are bio-indicators for the assessment of their habitat and environmental changes. Besides population parameters and trends (survival, age structure, sex ratio), the individual health status represents a further important parameter for this assessment. The health status of seals is a complex and vague term, determined by a wide range of diagnostic parameters. Quantities of important blood proteins such as transferrin (Tf), as well as altered distribution patterns of its glycoforms, are frequently used as biomarkers in clinical diagnosis. Within this context Tf quantities and a varying pattern of its glycoforms are used as indicator for e.g. certain liver diseases, which also represents one of the most frequently observed pathological indication in harbour seals of the North Sea. Currently, most assay based quantification methods for Tf are limited since they often provide only information regarding the total Tf concentration rather than information of its different glycoforms. Due to a lack of suitable seal Tf antibodies also the application of more specific antibody based approaches is not possible. Within this background a new approach for the absolute quantification of the iron-transport protein Tf in the blood of harbour seals using its characteristic iron content and HPLC-ICP-MS detection is described. Method validation was performed using a certified human serum reference material (ERM-DA470K/IFCC). A Tf concentration of 2.33 ± 0.03 g L(-1) (sum of all quantified glycoforms) has been calculated, which is in good agreement with the certified total Tf concentration of 2.35 ± 0.08 g L(-1), confirming the accuracy of the proposed analytical method. Finally, different seal samples were analysed to demonstrate the suitability of the procedure for the quantification of Tf in real samples as well as to observe modified glycoform patterns. Compared to our previous studies for the first time it was possible to quantify the serum Tf baseline reference range for male (1.42-2.35 g L(-1)) and female German North Sea seals (1.93-2.74 g L(-1)) as well as a CDT level of 0.00-0.10 g L(-1), respectively, which provides valuable further diagnostic information regarding the health status of these specific marine mammals. Compared to assay based quantification approaches the proposed technique indicates great potential to obtain comparable and traceable absolute quantitative results, which are in particular important for long term investigations. This absolute quantification is based on an accurate, traceable element standard, while assay based approaches often show variations depending on the kit quality or changing activities of the used antibodies.

The Toca-1-N-WASP Complex Links Filopodial Formation to Endocytosis

The transducer of Cdc42-dependent actin assembly (Toca-1)-N-WASP complex was isolated as an essential cofactor for Cdc42-driven actin polymerization in vitro. Toca-1 consists of an N-terminal F-BAR domain, followed by a Cdc42 binding site (HR1 domain) and an SH3 domain, (the N-WASP interacting site). N-WASP is an activator of actin nucleation through the Arp2/3 complex. The aim of the present study was to investigate the cellular function of the Toca-1-N-WASP complex. We report that Toca-1 induces filopodia and neurites as does N-WASP in N1E115 neuroblastoma cells. Toca-1 requires the F-BAR domain, Cdc42 binding site, and SH3 domain to induce filopodia. Toca-1 and N-WASP both require each other to induce filopodia. The expression of Toca-1 and N-WASP affects the distribution, size, and number of Rab5 positive membranes. Toca-1 interacts directly with N-WASP in filopodia and Rab5 membrane as seen by Forster resonance energy transfer. Thus the Toca-1-N-WASP complex localizes to and induces the formation of filopodia and endocytic vesicles. Last, three inhibitors of endocytosis, Dynamin-K44A, Eps15Delta95/295, and clathrin heavy chain RNA interference, block Toca-1-induced filopodial formation. Taken together, these data suggest that the Toca-1-N-WASP complex can link filopodial formation to endocytosis.

Establishment of a miniaturized enzyme-linked immunosorbent assay for human transferrin quantification using an intelligent multifunctional analytical plate

A miniaturized enzyme-linked immunosorbent assay (ELISA) with a reaction volume of 5 μl for human transferrin quantification has successfully been developed using an intelligent multifunctional analytical plate (IMAPlate 5RC96), the first miniature analytical platform capable of manually performing parallel liquid transfer, reaction, and analysis. This is the first article to validate the platform for the ELISA application. The data obtained from the standards in this miniaturized ELISA can well be fitted by a one-site binding reaction mode, the coefficient of variation (CV) of the whole plate for an artificial sample (spiking a known concentration of human transferrin into the assay diluent) is 7.0%, and the mean recovery is between 94 and 114% ( n = 96), comparable to the values from conventional ELISA in a 96-well format plate. The IMAPlate 5RC96-based miniaturized ELISA not only can reduce sample and reagent consumption to 5% of the conventional ELISA but also can shorten the reaction time. Combined with the advantages brought by miniaturization, the easy-to-handle, parallel, and simultaneous liquid transfer features of the IMAPlate 5RC96 provide a completely new lab tool for manually performing high-throughput ELISA. Our results demonstrate that the IMAPlate 5RC96 is a convenient, robust, high-throughput lab device feasible for miniaturized ELISA in an ordinary laboratory.

Metabolismo do ferro em suínos recebendo dietas contendo fitase, níveis reduzidos de fósforo inorgânico e sem suplemento micromineral e vitamínico

O presente estudo foi desenvolvido com o objetivo de avaliar o metabolismo do ferro por meio da determinação do eritrograma, contagem de reticulócitos, dosagem de ferro sérico, ferritina sérica e transferrina sérica de suínos em fase de terminação alimentados com dietas contendo fitase, sem suplemento micromineral/vitamínico e redução dos níveis de fósforo inorgânico (Pi). Foram utilizadas 48 fêmeas suínas de linhagem comercial, com peso inicial de 60kg, distribuídas em seis tratamentos com oito animais em cada grupo. A colheita de sangue foi feita em um grupo de 24 animais com 100kg e em outro grupo de 24 animais com 120kg. Não foram observadas diferenças (P>0,05) nos valores obtidos do eritrograma, da contagem de reticulócitos, de ferro sérico e de transferrina para os animais nos tratamentos testados. Com relação à ferritina, verificou-se que os animais até os 100kg de peso vivo que receberam ração sem suplemento micromineral/vitamínico, sem fósforo inorgânico e com fitase apresentaram valores superiores (P<0,05) quando comparados aos alimentados com ração sem suplemento micromineral/vitamínico, com fósforo inorgânico e contendo fitase. A enzima, mesmo na ausência de suplementação, garantiu a manutenção de estoques de ferro do organismo. Essa diferença não foi detectada para os animais que foram alimentados com as mesmas rações até os 120kg de peso vivo. Os resultados encontrados neste experimento mostram que a redução do fósforo inorgânico, a retirada do suplemento vitamínico e mineral, bem como a adição da fitase, em dietas para suínos em terminação, não desencadeiam alterações significativas no metabolismo do ferro.

Design and Construction of Automated Amperometric Immunosensing System

An automated amperometric immunosensors for the quantification of transferrin was fabricated using two monoclonal IgG class anti-rat transferrin antibodies, 15C2H3 and 22A06D2. The former was immobilized on a gold electrode, while the latter was conjugated with the positively charged polymers containing ferrocenyl groups. The quantitative analysis of transferrin was investigated by measuring the electrooxidation current of ferrocene, which is proportional to the transferrin concentration for the equivalently observed sandwich formation. The deposition of electroconductive polymers, polypyrrole (poly-P) and poly⌈ferrocenyl-methyl-3-(pyrrol-l-yl)propyldimethylammonium bromide⌉ (poly-FP), were performed on the surface of antibody-immobilized gold electrode in order to increase the efficiency of the electron transfer between the antibody-cojugated ferrocence and the gold electrode, and to eliminate the nonspecific binding of ferrocene-labeled antibody by a mutual repulsion.

Interference of Transferrin Isoform Types with Carbohydrate-deficient Transferrin Quantification in the Identification of Alcohol Abuse

Isoforms of transferrin interfere with measurement of carbohydrate-deficient transferrin (CDT) as a marker of heavy alcohol consumption. We evaluated the rate of inaccurate CDT results by immunoassays. We studied 2360 consecutive sera (1614 individuals) submitted for CDT assay without clinical information as well as samples from 1 patient with a congenital disorder of glycosylation (CDG Ia) and from 6 healthy carriers of CDG Ia. The CDTect, %CDT-TIA, and new %CDT immunoassays were compared with HPLC (%CDT-HPLC). Transferrin isoform pattern were evaluated by isoelectric focusing (IEF). Transferrin BC and CD heterozygotes were found at frequencies of approximately 0.7% and approximately 0.2%, respectively. Another transferrin C subtype, where di- and trisialotransferrin partly coeluted (tentatively identified as C2C3), was observed in approximately 0.6%. Compared with the %CDT-HPLC method, the immunoassays often produced low results for transferrin BC and high results for transferrin CD and "C2C3". A very high trisialotransferrin value (frequency approximately 1%) often produced high CDT immunoassay results. In four of six healthy carriers of CDG Ia, a- and disialotransferrin were highly increased and the HPLC and IEF isoform patterns were indistinguishable from those in alcohol abuse. Rare transferrin isoform types and abnormal amounts of trisialotransferrin (total frequency approximately 2-3%) may cause incorrect determination of CDT with immunoassays. The observed variants were readily identified by HPLC and IEF, which can be recommended for verification of CDT immunoassay results in doubtful cases. In healthy carriers of CDG Ia, CDT is high by all assays.
