Scientific Advances of Milk Enrichment with Conjugated Linoleic Acid to Produce Anti-Cancer Milk

Background: Conjugated linoleic acid (CLA) is an unsaturated fatty acid that after the discovery of its anti-cancerous properties numerous researches conducted to increase its concentration in the food products. The current study aimed to review researches in the field of milk enrichment with CLA to identify the main actors and highlight the achievements in the field; therefore, the effectiveness of these researches for producing anti-cancer milk was surveyed. Methods: The journal of dairy science was selected for data extraction and related papers were identified via review of all published papers during years 2003 - 2013 and then were subjected into content analysis. Statistical analysis was done with the MEANS procedure of SAS software (Version 9.1). Results: Among all collected papers, only 72 were eligible. Data analysis showed that the USA, Canada and France were the most productive countries in the field of milk enrichment with CLA. Surprisingly, Iran did not have any paper in this field in the journal of dairy science. The supplements that were added to the animal diets were enriched by linoleic or linolenic acids. Enhancing the milk cis-9, and trans-11 CLA content were the main objectives of researches. More than 70% of treatments led to the supply of the minimum amount of CLA to prevent cancer with serving milk twice a day. Conclusions: Considerably, milk enrichment with CLA mostly has being attracted by developed countries. Enriching milk with CLA is a suitable scientific strategy to fight against cancer. It is important to increase researches in increasing trans- 10, and cis- 12 CLA.

Dietary fish oil supplements depress milk fat yield and alter milk fatty acid composition in lactating cows fed grass silage-based diets

The potential of dietary fish oil (FO) supplements to increase milk 20:5n-3 and 22:6n-3 concentrations and the associated effects on milk fatty acid (FA) composition, intake, and milk production were examined. Four multiparous lactating cows offered a grass silage-based diet (forage:concentrate ratio 58:42, on a dry matter basis) supplemented with 0, 75, 150, or 300g of FO/d (FO0, FO75, FO150, and FO300, respectively) were used in a 4×4 Latin square with 28-d experimental periods. Milk FA composition was analyzed by complementary silver-ion thin-layer chromatography, gas chromatography-mass spectrometry, and silver-ion HPLC. Supplements of FO decreased linearly dry matter intake, yields of energy-corrected milk, milk fat and protein, and milk fat content. Compared with FO0, milk fat content and yield were decreased by 30.1 and 40.6%, respectively, on the FO300 treatment. Supplements of FO linearly increased milk 20:5n-3 and 22:6n-3 concentrations from 0.07 to 0.18 and 0.03 to 0.10g/100g of FA, respectively. Enrichment of 20:5n-3 and 22:6n-3 was accompanied by decreases in 4- to 18-carbon saturated FA and increases in total conjugated linoleic acid (CLA), trans FA, and polyunsaturated FA concentrations. Fish oil elevated milk fat cis-9,trans-11 CLA content in a quadratic manner, reaching a maximum on FO150 (from 0.61 to 2.15g/100g of FA), whereas further amounts of FO increased trans-10 18:1 with no change in trans-11 18:1 concentration. Supplements of FO also resulted in a dose-dependent appearance of 37 unique 20- and 22-carbon intermediates in milk fat. Concentrations of 16-, 18-, 20-, and 22-carbon trans FA were all increased by FO, with enrichment of trans 18:1 and trans 18:2 being quantitatively the most important. Decreases in milk fat yield to FO were not related to changes in milk trans-10,cis-12 CLA concentration or estimated milk fat melting point. Partial least square regression analysis indicated that FO-induced milk fat depression was associated with changes in the concentrations of multiple FA in milk. Even though a direct cause and effect could not be established, a decrease in 18:0 supply in combination with increased mammary uptake of cis-11 18:1, trans-10 18:1, and trans 20- and 22-carbon FA may contribute. In conclusion, dietary FO supplements enrich 20:5n-3 and 22:6n-3 in milk, but also elevate mono- and polyenoic trans FA concentrations, and in high amounts alter the distribution of individual trans FA isomers.

Cheddar Cheese from Cow Milk with Elevated Conjugated Linoleic Acid Levels

The aim of the study was to evaluate the effect of feed management on conjugated linoleic acid (CLA) level in milk and subsequently on fatty acid profile of cheese. Twenty cows were divided into 2 groups, fed on grass silage and pasture feed with sunflower supplementation for 14 days. Milk samples were collected and analyzed for composition and fatty acid profile using gas chromatograph. Cheddar cheese was manufactured from both milk types and ripened for 120 days. During ripening, it was analyzed for composition and fatty acid quantification. The supplemented diet significantly increased the CLA content in milk. Similarly the cis-9, trans-11 CLA content in Cheddar cheese were more than twofold in case of supplemented diet as compared to grass silage. During ripening, CLA concentration remained stable. The sunflower supplementation showed no effect on cheese composition. Hence, supplemented pasture resulted in elevated CLA content in milk compared to grass silage.

Effects of Lactation Stage and Individual Performance on Milk cis-9, trans-11 Conjugated Linoleic Acids Content in Dairy Cows.

The goal of this study was to evaluate the effects of lactation stage and individual performance on milk cis-9, trans-11 conjugated linoleic acid (CLA) content in dairy cows. In experiment 1, the milk cis-9, trans-11 CLA content from dairy cows in early (0.33±0.014%), middle (0.37±0.010%), and late stages (0.44±0.020%) showed significant differences (p<0.05); and the individual contents of the major fatty acids, especially cis-9, trans-11 CLA in cows of the same lactation were also variable. In the second experiment design as a validation test, our results once again showed that the individual contents of cis-9, trans-11 CLA were various, and a difference of about 2-fold (0.55% vs 0.95%) was observed, although the animals were offered same diet. These data demonstrated that lactation stage and individual performance have considerable effects on milk cis-9, trans-11 CLA contents.

Estimation of cis-9, trans-11 conjugated linoleic acid content in UK foods and assessment of dietary intake in a cohort of healthy adults

Dietary conjugated linoleic acid (CLA) from ruminant-derived foods may be potentially beneficial to health. The quantity of cis-9, trans-11 CLA and trans-10, cis-12 CLA in a range of UK foodstuffs (112 foods) was determined using triple-column silver ion HPLC. The cis-9, trans-11 CLA content ranged from 1.9 mg/g lipid (mild Cheddar) to 7.3 mg/g lipid (processed cheese) in cheeses, from 0.9 mg/g lipid (ice cream) to 3.7 mg/g lipid (double cream) in non-cheese dairy products, and from 2.9 mg/g lipid (Swedish meatballs) to 6.0 mg/g lipid (minced lamb) in meat products. cis-9, trans-11 CLA concentrations for chocolate and sweets ranged from 0.1 mg/g lipid (hot chocolate) to 4.8 mg/g lipid (buttermint). The trans-10, cis-12 CLA isomer was undetected or negligible in the food samples examined. To provide information about dietary cis-9, trans-11 CLA intakes in the UK, a study was performed to estimate the daily intake of CLA in a cohort of eighteen healthy volunteers (nine female and nine male; aged 21-60 years; mean BMI = 24.0 kg/m2 (sd 2.2)) with a 7-d weighed food record. This information combined with the CLA isomer contents of UK foodstuffs was used to estimate the daily intake of the cohort. The mean daily intake of cis-9, trans-11 CLA was estimated to be 97.5 (sd 73.3) mg/d. Due to its potential health benefits, it is important to determine the CLA content of food and dietary intake as these data will be useful in determining the role of CLA in health and disease.

Milk production and fatty acid profile of dairy cows supplemented with flaxseed oil, soybean oil, or extruded soybeans

The objective of the study was to investigate the effects of oil sources in the diet on milk yield, milk composition, and fatty acid (FA) profiles in mid-lactating dairy cows. Forty-eight Chinese Holstein dairy cows averaging 150 days in milk (DIM) at the start of the experiment (body weight = 596±19 kg; milk yield = 29.7±3.00 kg/d) were used in a completely randomized block design. The animals were assigned into four dietary treatments according to DIM and milk yield, and supplemented with no oil (control), 2% flaxseed oil (FSO), 2% soybean oil (SBO), and 2% oil from extruded soybeans (ESB). The experiment lasted nine weeks including the first week for adaptation. Milk yields, milk compositions (fat, protein, and lactose), and milk FA profiles were measured. Daily milk yield from cows fed with FSO, SBO, and ESB were higher than milk yield of the control cows (27.0, 27.0, and 26.5 vs. 25.4 kg/d). Milk fat percentage of the control cows was greater than those cows fed with oil-supplemented diets. However, increasing dietary fat content resulted with no change in fat-corrected milk yield. The FA profile of milk was changed by fat supplementation. Feeding oil reduced the proportion of both short-chain (C8:0 to C12:0) and medium-chain (C14:0 to C16:1) FAs, and increased the proportion of long-chain (≥C18:0) FAs in milk fat. Cis-9, trans-11 conjugated linoleic acid (CLA) in milk fat was increased from 0.38% for the control to 0.79, 1.51, and 1.56% of fat for the cows supplemented with FSO, SBO, and ESB, respectively. Feeding oils rich in linoleic acid (SBO and ESB) was more effective in enhancing cis-9, trans-11 CLA in milk fat than oils containing linolenic acid (FSO). There was a linear relationship between transvaccenic acid and cis-9, trans-11 CLA content in milk. Overall, feeding the FSO, SBO, and ESB diets increased monounsaturated and polyunsaturated fatty acids and decreased the saturated fatty acid in milk fat.

Effects of Soybean Oil or Whole Cotton Seed Addition on Accumulation of Conjugated Linoleic Acid in Beef of Fattening Brahman×Thai-Native Cattle

Effects of soybean oil or whole cotton seed addition on conjugated linoleic acid (CLA) and performance of fattening BrahmanxThai-Native cattle were studied. Eighteen fattening cattle averaging 241±24 kg body weight and approximately 1 year old were stratified by live weight into three groups and randomly assigned by group to one of three dietary treatments. The treatments were control (concentrated 14% crude protein), control and supplemented with 170 g/d soybean oil, control plus 170 g/d of oil from whole cotton seed. All animals were weighed before and after the experimental period and 4 cattle per treatment were randomly slaughtered then carcass measurements were obtained. There were no statistically significantly differences in the final body weight, average daily gain and dry matter intake among treatments. However, the crude protein intake was significantly decreased (p<0.01) when whole cotton seed was fed compared with control and soybean oil treatments. The carcass composition and carcass characteristics were not significantly different in Longissimus and Semimembranosus muscle by feeding soybean oil and whole cotton seed compared with the control treatment. Supplementation of soybean oil increased (p<0.01) cis-9, trans-11 CLA by 116% in Longissimus muscle and by 240% in Semimembranosus muscle. However, whole cotton seed did not increase cis-9, trans-11 CLA in both muscles. The present study successfully increased cis-9, trans-11 CLA content of muscle lipids by soybean oil but not by whole cotton seed.

The effect of dietary inclusion of olive tree leaves and grape marc on the content of conjugated linoleic acid and vaccenic acid in the milk of dairy sheep and goats

Olive tree leaves (OTL) and grape marc (GM) are by-products with high linolenic (LNA) and linoleic (LA) acid content, respectively, which can be used as dietary ingredients to increase the cis-9 trans-11 conjugated linoleic acid (CLA) content of milk fat in sheep and goats. An experiment was conducted with 16 Friesian ewes and 16 Alpine goats to study the effect of OTL and GM inclusion in sheep and goat diets on their milk fatty acid profile, with emphasis on cis-9, trans-11 CLA and vaccenic acid (VA). Ewes and the goats were fed the control (C) diets from parturition to the 90 days in milk (DIM) and then both groups were divided into two sub-groups (treatments). The control groups of both species continued to be fed the C diets, whereas the treated groups were gradually switched over a 2-week period (DIM=91-105) from the C diets to that of treatment 1, which contained air-dried OTL. These OTL diets were fed ad libitum for 1 month (DIM=106-135). After that period, the same treated groups, after 2 weeks of gradual adaptation (DIM=136-150), were switched to treatment 2, which contained air-dried GM. The GM diets were fed ad libitum for 1 month (151-180 DIM). Concentrations of polyunsaturated fatty acids (PUFA) and mono-unsaturated fatty acids increased significantly in milk fat of sheep fed OTL v. C. For goats, only the PUFA in milk fat was increased by feeding OTL compared with C. Relative to C, GM increased significantly the concentration of PUFA only in milk fat of sheep. OTL and GM diets increased the cis-9, trans-11 CLA and VA content in milk fat, compared with C, only in sheep. GM caused a sharp increase in 18:0 only in sheep milk fat, while the OTL diet increased significantly the 18:0 in milk fat of goats. GM and OTL diets also had opposite effects on the 18:1/18:0 ratio of sheep milk fat. In conclusion, OTL and GM, when included in sheep diets altered the milk fatty acid profile with a pronounced increase in cis-9, trans-11 CLA and VA contents. The results show that the response of sheep and goats to OTL and GM diets was different, suggesting a species difference that needs further investigation.

Milk Conjugated Linoleic Acid Response to Fish Oil and Linseed Oil Supplementation of Grazing Dairy Cows

The effect of supplementing the diet of grazing dairy cows with fish oil (FO) and linseed oil (LSO) on milk conjugated linoleic acid (CLA) was investigated. Sixteen Holstein cows (170±19 DIM) were assigned into two groups and fed a grain supplement (8.0 kg/d; DM basis) containing 800 g of saturated animal fat (CONT) or 200 g FO and 600 g LSO (FOLSO). All cows grazed together on Sudan grass pasture ad libitum and were fed the treatment diets for 3 wks. Cows were milked twice a day and milk samples were collected during the last three days of the trial. Milk production (24.89 and 22.45 kg/d), milk protein percentage (2.76 and 2.82) and milk protein yield (0.68 and 0.64 kg/d) for the CONT and FOLSO diets, respectively, were not affected (p>0.05) by treatment diets. Milk fat percentage (3.90 and 2.86) and milk fat yield (0.97 and 0.64 kg/d) were lower (p<0.05) with the FOLSO diet compared with the CONT diet. The concentration and yield of milk cis-9 trans- 11 CLA were higher (p<0.05) with the FOLSO diet (2.56% of total FA and 16.44 g/d, respectively) than the CONT diet (0.66% of total FA and 6.44 g/d, respectively). The concentrations of milk trans C18:1 and vaccenic acid (VA) were higher (p<0.05) with the FOLSO diet (13.53 and 7.48% of total FA, respectively) than the CONT diet (3.69 and 2.27% of total FA, respectively). In conclusion, supplementing the diet of grazing cows with FO and LSO increased milk cis-9 trans-11 CLA content but reduced milk fat content and yield.

Nutritional regulation of milk fatty acid composition

There is increasing evidence that nutrition plays an important role in the development of chronic diseases in the human population including cancer, cardiovascular disease, insulin resistance and obesity. Developing foods that enhance human health is central to dietary approaches for preventing and reducing the economic and social impact of chronic disease. Numerous studies in human subjects have implicated a high consumption of medium-chain (12:0-16:0) saturated fatty acids (SFA) and trans fatty acids (TFA) as risk factors for cardiovascular disease risk, with emerging evidence of a possible role in the development of insulin resistance and inflammation. Milk and dairy products are a major source of 12:0, 14:0, 16:0 and TFA in the human diet. However, developing public health policies promoting a decrease in milk, cheese and butter consumption ignores the value of these foods as a versatile source of nutrients and bioactive lipids, including 4:0, branch-chain fatty acids, cis-9, trans-11 conjugated linoleic acid (CLA), vitamins A and D, β-carotene and sphingomyelin. Therefore, altering milk fatty acid composition through sustainable, environmental and welfare acceptable means is an integral component of an overall strategy for preventing human chronic disease. Nutrition is the major environmental factor regulating milk fat composition. Strategies for enhancing the nutritional value of milk fat have been directed towards reducing the proportion of 12:0, 14:0 and 16:0, increasing cis monounsaturated fatty acids (MUFA) and polyunsaturated fatty acid (PUFA) content and/or enhancing the concentration of bioactive lipids. Formulation of diets to alter milk fat composition to meet these targets typically involves 1) inclusion of plant oils and oilseeds, 2) supplements of marine lipids, 3) increasing the proportion of dietary energy derived from fresh grass, 4) replacing ensiled grass, maize or whole-crop cereal with forage legumes or 5) inclusion of rumen-protected lipids in the diet. Nutritional strategies for reducing 12:0, 14:0 and 16:0 and enhancing cis-MUFA in milk fat are dependent on increasing the supply of C18 or longer-chain fatty acids to inhibit mammary de novo fatty acid synthesis. Due to incomplete metabolism of dietary unsaturated fatty acids in the rumen, inclusion of oils or oilseeds in the diet also increases milk fat TFA content. Both the concentration and distribution of TFA isomers in milk is dependent on the amount and type of lipid supplement fed, composition of the basal diet and interactions between these factors. Altering the diet of lactating cows can be used as a means to significantly enrich milk fat cis-9, trans-11 CLA content. Increases in the concentration of CLA in milk are reliant, in the most part, on enhancing ruminal trans-11 18:1 outflow for endogenous cis-9, trans-11 CLA synthesis in the mammary gland. The potential to increase n-3 fatty acids in milk is relatively limited. Inclusion of fresh or ensiled red clover in the ruminant diet significantly increases 18:3 n-3 content, while increases in milk 20:5 n-3 and 22:6 n-3 concentrations to marine lipid supplements is relatively limited due to extensive ruminal metabolism of n-3 PUFA. Greater enrichment of 20:5 n-3, 22:5 n-3 and 22:6 n-3 in milk can be achieved using rumen-protected fish oil supplements. Overall, recent research has highlighted the important role of nutrition in attempts to modify milk fat composition for improved long-term human health.

Milk Conjugated Linoleic Acid Response to Fish Oil and Sunflower Oil Supplementation to Dairy Cows Managed Under Two Feeding Systems

Earlier research showed that conjugated linoleic acid (CLA) content in milk fat is highest when cows’ diets are supplemented with a blend of fish oil (FO) and linoleic acid-rich oils. The objective of this study was to compare the effect of FO and sunflower oil (SFO) supplementation on milk cis-9, trans-11 CLA when dairy cows managed on pasture or in confinement. Fourteen Holstein cows were assigned into 2 treatment groups: cows grazed on alfalfa-grass pasture (PAS) or were fed corn silage-alfalfa hay mix ad libitum (LOT). Both groups were supplemented with a 8.2 kg/d grain supplement containing 640g of FO and SFO (1:3 wt/wt). Grain supplement was fed in 2 equal portions after each milking, for a period of 3 wk. Milk samples were collected during the last 3 d of the experimental period. Milk yield was greater with the LOT diet (23.1 kg/d) compared with the PAS diet (19.4 kg/d). Milk fat percentages (2.51 and 2.95 for the LOT and PAS, respectively) and yields (0.57 and 0.51 kg/d) were similar for the 2 diets. Milk protein percentages were not affected by diets (3.34 and 3.35 for the LOT and PAS diets, respectively), but protein yields were lower for the PAS diet (0.61 kg/d) compared with the LOT diet (0.75 kg/d). Treatment diets had no effect on milk trans C18:1 concentrations [10.64 and 9.82 g/100g of total fatty acids (FA) for the LOT and PAS, respectively] or yields (60.65 and 64.01 g/d), but did affect isomers distributions. Concentration (g/100g of total FA) of vaccenic acid was lower with the LOT diet (2.15) compared with the PAS diet (4.52), whereas concentration of trans-10 C18:1 was greater with the LOT diet (4.99) compared with the PAS diet (1.69). Milk cis-9, trans-11 CLA concentration was greater with the PAS diet (1.52) compared with the LOT diet (0.84). In conclusion, the increase in milk cis-9, trans-11 CLA content was greater when pasture-based diets were supplemented with FO and SFO. The lower cis-9, trans-11 CLA concentration in milk from the confinement-fed cows resulted from trans-10 C18:1 replacing vaccenic acid as the predominant trans C18:1 isomer.

Transfer of Absorbed cis-9, trans-11 Conjugated Linoleic Acid into Milk Is Biologically More Efficient than Endogenous Synthesis from Absorbed Vaccenic Acid in Lactating Cows ,3

Cis-9, trans-11, the major isomer of conjugated linoleic acid (CLA) in bovine milk fat, is derived from ruminal biohydrogenation of 18:2 (n-6) and endogenous conversion of trans-11 18:1 (vaccenic acid; VA) in the mammary gland. Most evidence to date suggests that endogenous synthesis is the major source of cis-9, trans-11 CLA, but the extent of VA desaturation is less well defined. Four lactating cows were used in consecutive 4 × 4 Latin squares to examine changes in milk fatty acid composition and secretion in response to abomasal infusions of lipid supplements enriched with cis-9, trans-11 CLA (88.8%) or VA (29.4%). Treatments were infused over 4-d, followed by a 3-d washout, during 7 d experimental periods and administered to deliver 0, 3, 6, and 12 g cis-9, trans-11 CLA/d (Expt. 1) or 0, 7.5, 15 and 30 g VA/d (Expt. 2). Infusions of cis-9, trans-11 CLA increased linearly milk cis-9, trans-11 CLA concentrations from 0.68 to 1.46 g/100 g fatty acids. Abomasal infusions of VA increased linearly milk VA and cis-9, trans-11 CLA content from 1.22 to 2.72 and 0.61 to 1.24 g/100 g fatty acids, respectively. Changes in milk fatty acid secretion indicated that 28.9% of VA was converted to cis-9, trans-11 CLA. Results provide evidence that conversion by ∆9-desaturase to cis-9, trans-11 CLA in the lactating cow is independent of postruminal VA supply. In conclusion, endogenous synthesis via VA was equivalent to ∼21% of the response to increases in cis-9, trans-11 CLA available for absorption.

Examination of the Persistency of Milk Fatty Acid Composition Responses to Fish Oil and Sunflower Oil in the Diet of Dairy Cows

Based on the potential benefits of cis-9, trans-11 conjugated linoleic acid (CLA) for human health, there is a need to develop effective strategies for enhancing milk fat CLA concentrations. Levels of cis-9, trans-11 CLA in milk can be increased by supplements of fish oil (FO) and sunflower oil (SO), but there is considerable variation in the response. Part of this variance may reflect time-dependent ruminal adaptations to high levels of lipid in the diet, which lead to alterations in the formation of specific biohydrogenation intermediates. To test this hypothesis, 16 late lactation Holstein-British Friesian cows were used in a repeated measures randomized block design to examine milk fatty acid composition responses to FO and SO in the diet over a 28-d period. Cows were allocated at random to corn silage-based rations (8 per treatment) containing 0 (control) or 45g of oil supplement/kg of dry matter consisting (1:2; wt/wt) of FO and SO (FSO), and milk composition was determined on alternate days from d 1. Compared with the control, the FSO diet decreased mean dry matter intake (21.1 vs. 17.9kg/d), milk fat (47.7 vs. 32.6g/kg), and protein content (36.1 vs. 33.3g/kg), but had no effect on milk yield (27.1 vs. 26.4kg/d). Reductions in milk fat content relative to the FSO diet were associated with increases in milk trans-10 18:1, trans-10, cis-12 CLA, and trans-9, cis-11 CLA concentrations (r2=0.74, 0.57, and 0.80, respectively). Compared with the control, the FSO diet reduced milk 4:0 to 18:0 and cis 18:1 content and increased trans 18:1, trans 18:2, cis-9, trans-11 CLA, 20:5 n-3, and 22:6 n-3 concentrations. The FSO diet caused a rapid elevation in milk cis-9, trans-11 CLA content, reaching a maximum of 5.37g/100g of fatty acids on d 5, but these increases were transient, declining to 2.35g/100g of fatty acids by d 15. They remained relatively constant thereafter. Even though concentrations of trans-11 18:1 followed the same pattern of temporal changes as cis-9, trans-11 CLA, the total trans 18:1 content of FSO milk was unchanged because of the concomitant increases in the concentration of other isomers (Δ4–10 and Δ12–15), predominantely trans-10 18:1. In conclusion, supplementing diets with FSO enhances milk fat cis-9, trans-11 CLA content, but the high level of enrichment declines because of changes in ruminal biohydrogenation that result in trans-10 replacing trans-11 as the major 18:1 biohydrogenation intermediate formed in the rumen.

Milk fatty acid composition of dairy cows grazing at two pasture allowances and supplemented with different levels and sources of concentrate

Milk fatty acid (FA) composition of dairy cows from two grazing studies was examined. In the first study, effects of concentrate supplementation and pasture allowance were evaluated using 20 multiparous Holstein cows in five 4 × 4 Latin squares. The four treatments resulted from the combination of two pasture allowances (i.e., low, 25 versus high, 40 kg dry matter/cow/day) and two concentrate supplementation levels (i.e., 0 versus 1 kg concentrate/4 kg milk). No interactions occurred between concentrate supplementation and pasture allowance for milk FA composition. Concentrate supplementation increased short-chain FA content, and reduced the content of long-chain FA, trans11 C18:1, and cis9, trans11 conjugated linoleic acid (CLA) (1.36 versus 1.18 g/100 g). Concentrate supplementation increased saturated FA (58.6 versus 54.0 g/100 g) and reduced unsaturated FA content (39.9 versus 43.8 g/100 g). Grazing at high pasture allowance increased short-, medium-, and long-chain FA content, without affecting cis9, trans11 CLA content. Saturated FA content was higher (57.1 versus 55.6 g/100 g), and unsaturated FA content was lower (41.3 versus 42.5 g/100 g), when cows grazed at high pasture allowance. Concentrate supplementation reduced unsaturated FA and cis9, trans11 CLA in milk of dairy cows grazing at two pasture allowances. In the second study, two experiments evaluated effects of different energy supplements in grazing dairy cows. In Experiment 1, 25 multiparous Holstein cows were assigned to a cracked corn (CC) or a steam flaked corn (SFC) supplement containing 667 g/kg of corn grain plus a pelleted protein/mineral supplement. In Experiment 2, 22 multiparous Holstein cows were assigned to a ground corn grain (GC) or a non-forage fiber (NFF) supplement. The GC supplement contained 850 g/kg of corn grain, and the NFF supplement contained 440 g/kg of non-forage fiber sources (i.e., beet pulp, soyhulls, wheat middlings) that partially replaced corn grain. Milk FA composition was not affected by corn processing or carbohydrate source. The content of short-, medium-, long-chain FA, and cis9, trans11 CLA (2.5 g/100 g in Experiment 1; 2.1 g/100 g in Experiment 2) was similar between supplements in both experiments. The type of supplement did not affect the content of saturated (62.6 g/100 g in Experiment 1; 65.4 g/100 g in Experiment 2) and unsaturated FA (37.5 g/100 g in Experiment 1; 34.6 g/100 g in Experiment 2). Supplementation with supplements differing in the rate and extent of ruminal carbohydrate digestion did not affect the milk FA composition of grazing dairy cows.

Effect of forage type and proportion of concentrate in the diet on milk fatty acid composition in cows given sunflower oil and fish oil

AbstractBased on the potential benefits ofcis-9,trans-11 conjugated linoleic acid (CLA) for human health there is a need to develop effective strategies for enhancing milk fat CLA concentrations. In this experiment, the effect of forage type and level of concentrate in the diet on milk fatty acid composition was examined in cows given a mixture of fish oil and sunflower oil. Four late lactation Holstein-British Friesian cows were used in a 4 × 4 Latin-square experiment with a 2 × 2 factorial arrangement of treatments and 21-day experimental periods. Treatments consisted of grass (G) or maize (M) silage supplemented with low (L) or high (H) levels of concentrates (65 : 35 and 35 : 65; forage : concentrate ratio, on a dry matter (DM) basis, respectively) offered as a total mixed ration at a restricted level of intake (20 kg DM per day). Lipid supplements (30 g/kg DM) containing fish oil and sunflower oil (2 : 3 w/w) were offered during the last 14 days of each experimental period. Treatments had no effect on total DM intake, milk yield, milk constituent output or milk fat content, but milk protein concentrations were lower (P&lt; 0.05) for G than M diets (mean 43.0 and 47.3 g/kg, respectively). Compared with grass silage, milk fat contained higher (P&lt; 0.05) amounts of C12:0, C14:0, trans C18:1and long chain ≥ C20 (n-3) polyunsaturated fatty acids (PUFA) and lower (P&lt; 0.05) levels of C18:0and trans C18:2when maize silage was offered. Increases in the proportion of concentrate in the diet elevated (P&lt; 0.05) C18:2(n-6) and long chain ≥ C20 (n-3) PUFA content, but reduced (P&lt; 0.05) the amount of C18:3(n-3). Concentrations oftrans-11 C18:1in milk were independent of forage type, but tended (P&lt; 0.10) to be lower for high concentrate diets (mean 7.2 and 4.0 g/100 g fatty acids, for L and H respectively). Concentrations oftrans-10 C18:1were higher (P&lt; 0.05) in milk from maize compared with grass silage (mean 10.3 and 4.1 g/100 g fatty acids, respectively) and increased in response to high levels of concentrates in the diet (mean 4.1 and 10.3 g/100 g fatty acids, for L and H, respectively). Forage type had no effect (P&gt; 0.05) on total milk conjugated linoleic acid (CLA) (2.7 and 2.8 g/100 g fatty acids, for M and G, respectively) orcis-9,trans-11 CLA content (2.2 and 2.4 g/100 g fatty acids). Feeding high concentrate diets tended (P&lt; 0.10) to decrease total CLA (3.3 and 2.2 g/100 g fatty acids, for L and H, respectively) andcis-9,trans-11 CLA (2.9 and 1.7 g/100 g fatty acids) concentrations and increase milktrans-9,cis-11 CLA andtrans-10,cis-12 CLA content. In conclusion, the basal diet is an important determinant of milk fatty acid composition when a supplement of fish oil and sunflower oil is given.

Effect of dietary fish oil on biohydrogenation of fatty acids and milk fatty acid content in cows

AbstractMechanisms underlying milk fat conjugated linoleic acid (CLA) responses to supplements of fish oil were investigated using five lactating cows each fitted with a rumen cannula in a simple experiment consisting of two consecutive 14-day experimental periods. During the first period cows were offered 18 kg dry matter (DM) per day of a basal (B) diet formulated from grass silage and a cereal based-concentrate (0·6 : 0·4; forage : concentrate ratio, on a DM basis) followed by the same diet supplemented with 250 g fish oil per day (FO) in the second period. The flow of non-esterified fatty acids leaving the rumen was measured using the omasal sampling technique in combination with a triple indigestible marker method based on Li-Co-EDTA, Yb-acetate and Cr-mordanted straw. Fish oil decreased DM intake and milk yield, but had no effect on milk constituent content. Milk fat trans-11 C18:1, total trans-C18 : 1, cis-9 trans-11 CLA, total CLA, C18 : 2(n-6) and total C18 : 2content were increased in response to fish oil from 1·80, 4·51, 0·39, 0·56, 0·90 and 1·41 to 9·39, 14·39, 1·66, 1·85, 1·25 and 4·00 g/100 g total fatty acids, respectively. Increases in the cis-9, trans-11 isomer accounted for proportionately 0·89 of the CLA response to fish oil. Furthermore, fish oil decreased the flow of C18 : 0(283 and 47 g/day for B and FO, respectively) and increased that of trans-C18 : 1fatty acids entering the omasal canal (38 and 182 g/day). Omasal flows of trans-C18 : 1acids with double bonds in positions from delta-4 to -15 inclusive were enhanced, but the effects were isomer dependent and primarily associated with an increase in trans-11 C18 : 1 leaving the rumen (17·1 and 121·1 g/day for B and FO, respectively). Fish oil had no effect on total (4·36 and 3·50 g/day) or cis-9, trans-11 CLA (2·86 and 2·08 g/day) entering the omasal canal. Flows of cis-9, trans-11 CLA were lower than the secretion of this isomer in milk. Comparison with the transfer of the trans-9, trans-11 isomer synthesized in the rumen suggested that proportionately 0·66 and 0·97 of cis-9, trans-11 CLA was derived from endogenous conversion of trans-11 C18 : 1in the mammary gland for B and FO, respectively. It is concluded that fish oil enhances milk fat cis-9, trans-11 CLA content in response to increased supply of trans-11 C18:1that arises from an inhibition of trans-C18 : 1reduction in the rumen.

Independent effects of dietary linoleic and linolenic fatty acids on the conjugated linoleic acid content of cows’ milk

AbstractIt may be desirable to increase the level of conjugated linoleic acid (CLA) in milk as a health benefit in human nutrition. The purpose of this work was to separate the effects of linoleic and linolenic acids on CLA production in dairy cows and to determine to what extent endogenous synthesis contributes to cis-9, trans-11 CLA concentration in milk fat. Eight lactating cows and four non-lactating duodenal fistulated cows were used in a 4 ✕ 4 Latin-square design. All cows received a basal diet of grass silage that was supplemented with one of four concentrates, which were designed to differ in their linoleic and linolenic acid contents. The oil components of the concentrates were produced from mixtures of olive, linseed, rape, soya and sunflower oils to produce the four treatments: low linoleic/ low linolenic acid (LL), low linoleic/high linolenic acid (LH), high linoleic/low linolenic acid (HL) and high linoleic/ high linolenic acid (HH). Milk cis-9, trans-11 CLA contents were 0·8, 0·9, 0·9 and 1·1 g/100 g fatty acid methyl esters (P &lt; 0·05) and yields were 5, 7, 7 and 8 g/day (P &lt; 0·05) for the LL, LH, HL and HH treatments, respectively. The yields of trans-C18:1fatty acids in milk were 19, 22, 21 and 23 g/day (P &lt; 0·05), respectively. Taking the data for the cis-9, trans-11 CLA content and flow of duodenal fluid from the fistulated cows and representing this in terms of dietary intake by the lactating animals, the amounts of cis-9, trans-11 CLA produced in the rumen were calculated to be 0·8, 0·9, 1·2 and 1·1 g/day (P &lt; 0·05) and for trans-C18:1fatty acids 58, 58, 66 and 69 g/day (P &lt; 0·05). Increasing linoleic and/or linolenic acids in the diet can increase the cis-9, trans-11 CLA content of cows’ milk. Only diets high in linoleic acid increased cis-9, trans-11 CLA production in the rumen. On all four diets, more than 80% of cis-9, trans-11 CLA in milk was produced endogenously by Δ9-desaturase from trans-11 C18:1in the mammary gland. Cows on the same diet have different milk fat cis-9, trans-11 CLA concentrations that may be partially explained by differences in Δ9-desaturase activity between cows. Increasing the activity of Δ9-desaturase in the mammary gland may offer greater potential for enhancing the cis-9, trans-11 CLA content of milk fat than increasing cis-9, trans-11 CLA production in the rumen.
