We aimed to determine the herd and animal levels seroprevalence and associated factors for Toxoplasma gondii infections in cattle from the state of Paraíba, Northeastern Brazil. Herds (n = 434) and cows aged ≥ 24 months (n = 1,895) were randomly selected, and serum samples were tested with the immunofluorescence antibody test (IFAT) using as cutoff of 64. Of the 434 farms investigated, 197 had at least one seropositive cow (prevalence of 49.0%; 95% CI = 44.3%-53.8%), and the prevalence at animal level was 18.0% (95% CI = 5.3%-21.1%). The antibody titers ranged from 64 to 1024, with the most frequent titers being 64 (10.8%) and 128 (3.7%). The risk factors identified were property located in Sertão region (odds ratio [OR] = 3.07), property located in Agreste/Zona da Mata regions (OR = 2.00), animal purchasing (OR = 2.68), herd size of 34-111 animals (OR = 2.91) and herd size > 111 animals (OR = 6.97). The results suggest the wide distribution of T. gondii infections in cattle throughout the state of Paraíba, and the risk factors identified are not possible to correct.

Public Health Significance of Toxoplasma gondii Infections in Cattle: 2009-2020.

Toxoplasma gondii infections are common in humans and animals worldwide. The ingestion of food or water contaminated with oocysts excreted by infected cats or ingesting uncooked or undercooked meat containing tissue cysts of T. gondii are the 2 major modes of transmission of T. gondii. Viable T. gondii is more prevalent in pork and lamb than in beef. In the past decade, there have been many articles on the high seroprevalence in cattle, particularly from China. There is a report of an outbreak of acute toxoplasmosis in humans suspected to be linked to the ingestion of Artisan fresh cheese from cow's milk. There are conflicting reports concerning the rate of congenital transmission of T. gondii in cattle, especially from Brazil. In a report from Brazil, viable T. gondii was isolated from the blood of 1 of 60 pregnant cows slaughtered at an abattoir and from 1 fetus. The role of beef in the epidemiology of T. gondii infections is still not clear. Here, we review prevalence, persistence of infection, clinical disease, epidemiology, and public health risks of T. gondii infections in cattle from beef and cow's milk worldwide for the past decade.

Toxoplasma gondii seroprevalence in beef cattle raised in Italy: a multicenter study

Toxoplasmosis represents an important public health issue, with the consumption of raw or undercooked meat being a major way of human infection. The role of beef in the transmission of the parasite to humans is questioned due to lower quantity of tissue cysts compared with other meat-producing species. However, the habit of consuming raw beef is regionally diffused, and the risk posed by Toxoplasma gondii infection in cattle should not be overlooked. Therefore, to update information on T. gondii in cattle reared in Italy, a multicentric seroepidemiological survey was designed and implemented in four Northern regions (Liguria, Lombardy, Piedmont, and Trentino Alto Adige) and Sardinia. Overall, a convenience sampling was performed, collecting 1444 serum samples from 57 beef cattle herds. Thirteen beef breeds were sampled, besides cross-breed; bovines age varied from 3months to over 12years. Sera were tested with a commercial ELISA for the detection of anti-T. gondii antibodies. Individual and herd data were analyzed by binary logistic regression analysis. A T. gondii seroprevalence of 10.2% was recorded, with differences among regions and values ranging from 5.3% in Liguria to 18.6% in the Piedmont region (p value = 0.0001). Both young and adult animals and males and females tested positive, without any significant difference (age and gender: p value > 0.05). Lower seroprevalence values were recorded in cattle born in Italy (8.7%) if compared with animals imported from abroad (13.4%) (p value = 0.046). The spread of T. gondii in beef cattle destined to Italian consumers is confirmed, suggesting the need of continuous monitoring of the infection.

Serological survey and risk factors for Toxoplasma gondii infection in cattle from Amazonas, Brazil

Beef cattle farming in the Amazon region has expanded rapidly, but information on herd health is still scanty. The purpose of this study was to determine the presence of anti-Toxoplasma gondii antibodies in cattle, its spatial distribution and the risk factors associated with the infection in the state of Amazonas, Brazil. Blood samples were collected from 1073 animals on 47 farms, located in 33 municipalities in the four state subpopulations. Anti-T. gondii IgG antibodies were detected by means of the indirect fluorescent antibody test (IFAT). The overall prevalence was 30.9 % (332/1073), and seropositive animals were identified at 93.6 % farms (44/47). All the subpopulations studied in the state of Amazonas had cattle herds seropositive for T. gondii, with some areas showing higher prevalence rates. The risk factors identified in the logistic regression were number of animals (OR = 4.43) and presence of domestic cats (OR = 1.98). It is advisable to correct identified risk factors, particularly insofar as the definitive hosts of T. gondii are concerned. Attention should also focus on beef consumption, given the prevalence of T. gondii infection among cattle and the widespread clandestine slaughtering that occurs in this state.

Toxoplasma gondii in cattle in Brazil: a review.

Toxoplasma gondii is an apicomplexan protozoan that is frequently found in both humans and animals worldwide. The aim of this review was to list important aspects of Toxoplasma gondii infection in cattle in Brazil. The frequency of occurrence of T. gondii antibodies in Brazilian cattle ranges from 1 to 89.1%, depending on the region evaluated, based on data from 1978 to 2018. However, some characteristics of T. gondii infection in cattle remain uncertain, such as the role of meat intake in transmitting the parasite to humans. Most information regarding T. gondii infection among Brazilian cattle is limited to evaluations of the frequency of occurrence of antibodies. About 70% of the diagnoses of infection in these ruminants in Brazil are made via the indirect fluorescence antibody test (IFAT). Nevertheless, little is known about the population structure of this protozoan in cattle. It is necessary to expand the studies on toxoplasmosis in cattle, in order to better understand T. gondii infection in these animals and its implications for Brazilian public health.

Cross-Sectional Survey on Toxoplasma gondii Infection in Cattle, Sheep, and Goats in Algeria: Seroprevalence and Risk Factors.

A cross-sectional study aimed at assessing the seroprevalence and identifying the risk factors for Toxoplasma gondii infection in cattle, sheep, and goats in eight provinces located in two main Algerian agro-ecological zones was carried out from October 2015 to March 2018. Blood sera from 4074 animals of both sexes were tested for the presence of anti-T. gondii IgG antibodies, using the indirect, enzyme-linked, immunosorbent assay technique (ELISA). Moreover, to identify the potential risk factors of T. gondii infection, a survey through a breeders’ questionnaires was conducted. Nearly one-fourth of the total number of animals tested (1024/4074)—i.e., 25.1%—were seropositive. The seroprevalence in cattle, sheep, and goats was 28.7%, 25.6%, and 11.9%, respectively. The area, sex, age, and herd size were identified as risk factors for T. gondii infection. Higher seropositivity rates were recorded in cows and goats (odds ratio (OR) = 1.63 and 6.4), in old animals (cattle, OR = 2.1; sheep, OR = 1.9; and goat, OR = 3.9), and in small size herds (cattle, OR = 2.5; sheep, OR = 1.9; goat, OR = 2.2). In conclusion, there is widespread T. gondii infection in cattle, sheep, and goats in these two strategic agricultural areas. The identification of the risk factors determines the type of measures and strategies to be undertaken to reduce, control, and prevent T. gondii infection in domestic animals, and thereby reduce human infection.

Seroprevalence and risk factors for toxoplasmosis in large ruminants in northern Punjab, Pakistan.

Toxoplasmosis is a widespread zoonosis that causes significant economic losses due to abortions and other developmental disabilities in livestock animals. The objectives of the current study were to determine the prevalence and associated risk factors of Toxoplasma gondii infection in cattle and buffaloes in northern parts of Punjab, Pakistan, where no such work previously existed. Serum samples obtained from 400 cattle and 422 buffaloes present on different private and government-owned farms were tested for anti-Toxoplasma IgG and anti-Toxoplasma IgM antibodies by using enzyme linked immunosorbent assay (ELISA). Additional data, including water sources, hygienic status at the farm, management practices, size of the herd, and presence of cats in the vicinity of the farm were obtained using a questionnaire in surveys and interviews. The overall prevalence of infection was 19.75% (79/400) in cattle and 15.16% (64/422) in buffaloes. IgG antibodies were found in 75 (18.75%) cattle and 58 (13.74%) buffaloes, while IgM antibodies were found in 9 (2.25%) cattle and 10 (2.37%) buffaloes. Seroprevalence was significantly higher in females and older animals of both species. Seroprevalence was found to be associated with poor hygienic conditions, extensive management practices, and presence of cats in the surrounding areas. No difference of seroprevalence was observed with respect to different breeds, location, water source, and herd size. The present study found that T. gondii is prevalent in large ruminants in northern Punjab, and may have important implications for the livestock industry and public health.

Seroprevalence of Toxoplasma gondii infection in cattle, horses, pigs and chickens in Japan

The presence of antibodies to Toxoplasma gondii in livestock and poultry was investigated by latex agglutination tests; samples that agglutinated at dilutions of 1:64 or higher were regarded as positive. Sera were collected from fattening beef cattle (102 Japanese black, 105 crossbreeds and 114 castrated Holstein), culled dairy cattle (101 Holstein), 100 horses, 115 fattening pigs and 235 chickens (163 free-range and 72 broilers) at abattoirs in Gifu Prefecture, Japan, from August 2012 to August 2013. Antibodies to T. gondii were found in 7.3% (31/422) in cattle, 5.2% (8/155) in pigs, but not in horses or chickens. These results suggest that toxoplasmosis may be transmitted to humans via consumption of T. gondii-infected raw beef in Japan.

Prevalence and genotype of Toxoplasma gondii infection in cattle from Jilin Province, northeastern China.

Abstract The prevalence and genotype of Toxoplasma gondii infection in cattle in Jilin Province, northeastern China was investigated. A total of 1040 serum samples were collected from eight administrative regions from September to October, 2011, and antibodies to T. gondii were examined by enzyme-linked immunosorbent assay (ELISA). The results showed that the overall seroprevalence of T. gondii in cattle was 12.8% (95% confidence interval [CI], 10.8-14.8%), with a higher prevalence of 22.2% (95% CI 13.6-30.8%) in Siping. Sixty-six tissue samples were collected from Changchun, and T. gondii DNA was detected by a nested PCR. There were nine (13.6%; 95% CI 5.4-21.9%) positive samples, which were genotyped using 11 genetic markers for PCR restriction fragment length polymorphisms (RFLP). Only one sample could be completely genotyped, and all of the loci were grouped into clonal type I, except for type III at the GRA6 locus, implying that T. gondii in cattle in Changchun, Jilin Province was type I variant. This study is the first report on genotype of T. gondii infection in cattle in China.

Seroprevalence of Toxoplasma gondii infection in cattle, sheep, goats and pigs from the North of Portugal for human consumption

Prevalence of antibodies to Toxoplasma gondii and risk factors associated with infection were assessed in food animals from the North of Portugal for human consumption. Antibodies were assayed by means of the modified agglutination test with a cut-off titre of 100 for cattle, and 20 for sheep, goats, and pigs; 7.5% of 161 cattle, 33.6% of 119 sheep, 18.5% of 184 goats, and 9.8% of 254 pigs were seropositive. Among the risk factors examined animal age was an important risk factor for seropositivity to T. gondii. The consumption of raw or undercooked meat should be regarded as an important source of infection to people in the study area.

Risk factors associated with Toxoplasma gondii infection in dairy cattle, state of Rio de Janeiro

Toxoplasmosis is one of the most common parasitic zoonoses throughout the world. Infection in man and animals varies in different geographical areas influenced by many environmental conditions. Seroprevalence of Toxoplasma gondii infection in cattle in Brazil ranges from 1.03 to 71%. A cross-sectional survey was carried out in 58 out of 453 farms in the South Fluminense Paraiba Valley, State of Rio de Janeiro, Brazil. Over 3-year-old cattle (n=589) from dairy herds were selected for blood collection and detection of anti-T. gondii antibodies by indirect immunofluorescence reaction (IFA) with initial titration of 1:16; titers > 64 were considered positive. Univariate analysis of risk factors showed that cats in contact with cattle, cats in contact with drinking water, and number of cats were associated with T. gondii seroprevalence. Logistic regression revealed a two-fold increased risk for infection of cattle (p=0.0138) through larger number of cats (>3) compared with low numbers of cats (1-2) on the farm. In contrast, the presence of chickens was considered a protective factor (p=0.025).

Diagnosis of Sarcocystis cruzi, Neospora caninum, and Toxoplasma gondii infections in cattle

The aim of the study was to diagnose Sarcocystis sp. infections in cattle and to detect coinfections by Toxoplasma gondii and/or Neospora caninum. Blood, diaphragm, esophagus, and myocardium from 90 beef cattle from Argentina were collected. Histopathological, immunohistochemical, polymerase chain reaction assays, and direct microscopical examination were carried out. Sarcocysts from myocardium were measured and counted. Indirect fluorescent antibody test (IFAT) for the three protozoans was performed. Sarcocystis cruzi sarcocysts were found in 100% of myocardium samples. Sarcocysts per gram ranged from 8 to 380 with higher values found in adult cattle (p < 0.001). T. gondii and N. caninum were not detected by immunohistochemistry. T. gondii DNA was found in myocardium of 2/20 seropositive animals, while N. caninum DNA was not found. Antibodies against S. cruzi were detected in all samples, those against N. caninum in 73% and against T. gondii in 91% of the samples (IFAT titer > or =25). It is concluded that serology by IFAT is a suitable method to diagnose these protozoan infections due to its specific IgG detection; therefore, IFAT may be a useful tool to evaluate the impact of each protozoan infection in coinfected animals.

Cross-sectional survey on Toxoplasma gondii infection in cattle, sheep and pigs in Serbia: Seroprevalence and risk factors

Toxoplasmosis is a globally distributed zoonosis with a clinical impact in the unborn fetus and in the immunosuppressed individual. In Serbia, studies of risk factors for Toxoplasma gondii infection in humans have shown that the relatively high prevalence is associated mainly with consumption of undercooked meat and/or meat products. However, data on T. gondii infection in domestic animals mostly used for human consumption are scarce. We thus conducted a cross-sectional survey on the seroprevalence of T. gondii infection in a representative sample of cattle, sheep and pigs from different regions of Serbia between June 2002 and June 2003, and analyzed the main risk factors associated with the infection. Sera from 611 cattle (yearlings and adults of both sexes), 511 ewes, and 605 pigs (market-weight and sows), were examined for T. gondii antibodies by the modified agglutination test. The seroprevalences determined were 76.3% in cattle, 84.5% in sheep and 28.9% in pigs. The antibody levels ranged from 1:25 to 1:400 in cattle, and up to 1:25,600 in sheep and to 1:12,800 in pigs. Among the seropositive, the proportion of high antibody levels (≥1:1600), suggestive of acute infection, was 10% in sheep, and 4% in pigs. Possible association of the infection with biologically plausible risk factors including gender, age, herd size/farm type, type of housing, feeding practices and region, was analyzed by univariate analysis, and variables significant at P ≤ 0.1 were included in multivariate logistic regression models. The results showed that risk factors for cattle were small herd size (odds ratio, OR = 2.19, 95% confidence interval, CI = 1.28–3.75, P = 0.004) and farm location in Western Serbia (OR = 2.04, 95% CI = 1.10–3.79, P = 0.024), while housing in stables with access to outside pens was protective (OR = 0.37, 95% CI = 0.21–0.67, P = 0.001). In sheep, an increased risk of infection was found in ewes from state-owned flocks (OR = 4.18, 95% CI = 2.18–8.00, P < 0.001) vs. private flocks, and, interestingly, also in those from Western Serbia (OR = 4.66, 95% CI = 1.18–18.32, P = 0.028). In pigs, the risk of infection was highly increased in adult animals (OR = 3.87, 95% CI = 2.6–5.76, P < 0.001), as well as in those from finishing type farms (OR = 3.96, 95% CI = 1.97–7.94, P < 0.001). In addition to providing data on the current T. gondii seroprevalence in meat animals in Serbia, the results of this study show the main risk factors associated with infection, thereby pointing to the type of preventive measures to reduce T. gondii infection.

Seroprevalence of Toxoplasma gondii infection in cattle, swine and goats in four Mexican states

A serological survey for antibody activity against Toxoplasma gondii in cattle, swine and goats was carried out in four Mexican states. A total of 1203 pig sera from Cuernavaca, Morelos abattoir, 490 cattle sera on 12 ranches from four municipalities and 707 goat sera from nine farms were tested by the enzyme-linked immunosorbent assay (ELISA) test. Sera from 107 of the 1207 pigs (8.9%) were positive, and there was no significant difference ( P>0.05) due to the state of origin of the positive sera. Five of the nine goat farms had T. gondii seropositive goats and the average seroprevalence was 3.2%. Antibody activity to T. gondii was found in 47 of 397 cattle (11.9%) tested. Food animals in Morelos, faced two problems, the reproductive losses from toxoplasmosis and the public health implications associated with infected meat and milk.

A review of toxoplasmosis in cattle

Worldwide reports of natural and experimentally-induced Toxoplasma gondii infections in cattle are reviewed and tabulated. Serologic tests employed in most studies in the past are of suboptimal sensitivity for the diagnosis of T. gondii infection in cattle; therefore data need to be interpreted with caution. From the evidence available it is concluded that: (1) T. gondii is probably not important in causing abortion or clinical illness in cattle but further experimental studies are desirable; (2) T. gondii is eliminated quickly from bovine tissues; (3) Whether beef plays a part in the epidemiology of toxoplasmosis is unresolved; (4) Milk from T. gondii-infected cows is of negligible importance in the epidemiology of toxoplasmosis.
