Interpretable attention-based multi-encoder transformer based QSPR model for assessing toxicity and environmental impact of chemicals

The rising demand from consumer goods and pharmaceutical industry is driving a fast expansion of newly developed chemicals. The conventional toxicity testing of unknown chemicals is expensive, time-consuming, and raises ethical concerns. The quantitative structure–property relationship (QSPR) is an efficient computational method because it saves time, resources, and animal experimentation. Advances in machine learning have improved chemical analysis in QSPR studies, but the real-world application of machine learning-based QSPR studies was limited by the unexplainable ‘black box’ feature of the machine learnings. In this study, multi-encoder structure-to-toxicity (S2T)-transformer based QSPR model was developed to estimate the properties of polychlorinated biphenyls (PCBs) and endocrine disrupting chemicals (EDCs). Simplified molecular input line entry systems (SMILES) and molecular descriptors calculated by the Dragon 6 software, were simultaneously considered as input of QSPR model. Furthermore, an attention-based framework is proposed to describe the relationship between the molecular structure and toxicity of hazardous chemicals. The S2T-transformer model achieved the highest R2 scores of 0.918, 0.856, and 0.907 for logarithm of octanol-water partition coefficient (Log KOW), octanol-air partition coefficient (Log KOA), and bioconcentration factor (Log BCF) estimation of PCBs, respectively. Moreover, the attention weights were able to properly interpret the lateral (meta, para) chlorination associated with PCBs toxicity and environmental impact.

Polychlorinated biphenyls in bovine milk from a typical informal electronic waste recycling and related source regions in southern India before and after the COVID-19 pandemic outbreak

For more than a decade, Chennai city in southern India has been evidenced with informal electronic waste (e-waste) recycling and open burning practices as the potential sources for polychlorinated biphenyls (PCBs). PCBs can bioaccumulate in livestock particularly cows grazing on the contaminated soil. The outbreak of the COVID-19 pandemic led to additional challenges associated with waste management practices. Hence this study aims to elucidate twenty-five PCB congeners in bovine milk from the previously reported PCB source regions in Chennai and the suburbs before and after about three years of the pandemic outbreak along electronic waste recycling (EWR), open burning dumps (OBD), and residential (RES) transects. The geomean concentration of Ʃ25PCBs in ng/g lipid weight (lw) followed a decreasing trend of EWR (13 ng/g lw) > OBD (8 ng/g lw) > RES (4 ng/g lw). Over 80 % of PCBs stemmed from EWR and OBD transects before and after the pandemic. However, a significant surge in the level of PCB-52 was observed in the OBD transect after the pandemic outbreak. Most toxic PCB congeners, PCB-126 and -169 were significant contributors to TEQs in EWR and OBD transects and can be reasoned with the burning of waste materials and mixed plastics in these transects. The highest average daily dose (ADD) exposure risk was for children from EWR and was significantly higher (p < 0.05) than other transects. Mean ADD-induced TEQ (6.6 pg TEQ/kg-bw/day) from the cows grazing around Kodungaiyur dumpsite slightly exceeded the EU guideline of 5.5 pg TEQ/kg-bw/day after the outbreak of the pandemic due to PCB-126. However, none of the samples exceeded the US FDA (1.5μg/g milk fat) recommendation limits for PCBs in milk fat. Prolonged exposure to such persistent organic pollutants interlinked with the burning of mixed waste in the open dumps can be a public health concern.

Exposure to polychlorinated biphenyls selectively dysregulates endothelial circadian clock and endothelial toxicity

Polychlorinated biphenyls (PCBs) are lipophilic and persistent environmental toxicants, which pose health threats to the exposed population. Among several organs and cell types, vascular tissue and endothelial cells are especially prone to PCB-induced toxicity. Exposure to PCBs can exert detrimental impacts on biological pathways, expression of transcription factors, and tight junction proteins that are integral to the functionality of endothelial cells. Because biological and cellular processes are tightly regulated by circadian rhythms, and disruption of the circadian system may cause several diseases, we evaluated if exposure to PCBs can alter the expression of the major endothelial circadian regulators. In addition, we studied if dysregulation of circadian rhythms by silencing the brain and muscle ARNT-like 1 (Bmal1) gene can contribute to alterations of brain endothelial cells in response to PCB treatment. We demonstrated that diminished expression of Bmal1 enhances PCB-induced dysregulation of tight junction complexes, such as the expression of occludin, JAM-2, ZO-1, and ZO-2 especially at pathologically relevant longer PCB exposure times. Overall, the obtained results imply that dysregulation of the circadian clock is involved in endothelial toxicity of PCBs. The findings provide new insights for toxicological studies focused on the interactions between environmental pollutants and regulation of circadian rhythms.

Target Lipid Model and Empirical Organic Carbon Partition Coefficients Predict Sediment Toxicity of Polychlorinated Biphenyls to Benthic Invertebrates.

Quantifying causal exposure-response relationships for polychlorinated biphenyl (PCB) toxicity to benthic invertebrates can be an important component of contaminated sediment assessments, informing cleanup decisions and natural resource injury determinations. Building on prior analyses, we demonstrate that the target lipid model accurately predicts aquatic toxicity of PCBs to invertebrates, providing a means to account for effects of PCB mixture composition on the toxicity of bioavailable PCBs. We also incorporate updated data on PCB partitioning between particles and interstitial water in field-collected sediments, to better account for effects of PCB mixture composition on PCB bioavailability. To validate the resulting model, we compare its predictions with sediment toxicity data from spiked sediment toxicity tests and a variety of recent case studies from sites where PCBs are the primary sediment contaminant. The updated model should provide a useful tool for both screening-level and in-depth risk analyses for PCBs in sediment, and it should aid in diagnosing potential contributing factors at sites where sediment toxicity and benthic community impairment are observed. Nutritional profiling and contaminant levels of five underutilized fish species in Norway.

Exploring and making use of underutilized marine resources can be a sustainable approach to achieve future demands of fish consumption by the ever-growing population. Five species, namely European plaice (Pleuronectes platessa), European flounder (Platichthys flesus), lemon sole (Microstomus kitt), megrim (Lepidorhombus whiffiagonis), and thornback ray (Raja clavate), often captured as by-catch in Norway, were characterized for their nutritional value and potential accumulation of hazardous components. The proximate composition, protein profile, fatty acid profile as well as essential and toxic trace elements and polychlorinated biphenyls (PCBs) were analyzed. Digestible indispensable amino acid (DIAA) ratios and scores (DIAAS) and contributions of omega-3 fatty acids to the diet were calculated. Analysis on proximate composition revealed low fat contents of 0.74 to 1.25% and sufficient protein contents between 16.9 and 24% in the five species. Results of DIAA indicate a profitable distribution, with contributions exceeding the daily intake recommendations for an adult person related to a 200 g fillet. Moreover, findings on the distribution of eicosapentaenoic (EPA) and docosahexaenoic acid (DHA) showed remarkable results, considering that the investigated species are lean fish. All five investigated fish exceed the recommended average daily intake level (AI) of EPA + DHA in a 200 g portion. As to toxic trace elements and PCBs, no significantly elevated levels were found considering a portion size of 200 g. Consequently, the nutritional quality of the investigated fish can be regarded as profitable with overall low potential health risks.

2,3′,4,4′,5-Pentachlorobiphenyl induces mitochondria-dependent apoptosis mediated by AhR/Cyp1a1 in mouse germ cells

Polychlorinated biphenyls (PCBs) are environmental organic pollutants widely used in industry that can bioaccumulate and affect the reproductive systems of male animals of different species. 2,3′,4,4′,5-Pentachlorobiphenyl (PCB118) is a representative of the 209 toxic PCB congeners. In this study, male mice were exposed to PCB118 at 0, 50, and 500 μg/kg/day for 35 days beginning 3–4 weeks after birth. The results of the study showed that PCB118 exposure during puberty reduced testicular quality, caused tissue damage, decreased sperm motility and sperm count, and increased malformation and testicular cell apoptosis in mice. Moreover, PCB118 increased the oxidative stress levels in sperm and testicular tissue and the expression of aryl hydrocarbon receptor (AhR) and Cyp1a1 and siginificantly decreased the level of nuclear factor-erythroid 2-related factor 2 (Nrf2). The results indicate that PCB118 can activate the AhR/Cyp1a1 pathway and inhibit Nrf2 expression to aggravate testicular oxidative stress and induce cell apoptosis, resulting in testicular and sperm quality damage.

Polychlorinated Biphenyls in the Snow Cover of South-Eastern Kazakhstan

The presence of large sources of environmental pollution due to persistent organic pollutants (POPs), in particular, polychlorinated biphenyls (PCBs), in Kazakhstan necessitates the assessment of pollution as a result of these toxicants. For this purpose, we chose snow cover as an indicator for assessing pollution status in the study area. An assessment of the PCB accumulation level included in the list of POPs was carried out for a snow cover (SC) study in south-east Kazakhstan. The content of PCBs with a wide congener composition was determined using the chromatographic analysis method. During the winter periods of 2014, 2015, 2018–2020 and 2021, the SC pollution of the study area from up to 25 individual PCB congeners was identified. These congeners included highly toxic dioxin-like congener PCBs 105; 108; 114; 118 and “marker” PCBs 52; 101; 138; 153. These congeners were mainly found in snow samples with a wide range of PCB congener compositions. The main PCB pollution sources were indicated. The analysis of the obtained results and structure of the congener composition of PCBs show that the SC contamination in this territory occurs under the influence of local and regional sources.

Detection of Toxic Polychlorinated Biphenyls by Nanoporous Metal–Organic Frameworks

As a class of persistent organic pollutants (POPs), polychlorinated biphenyls (PCBs) are highly hazardous to the ecosystem, and their effective and facile detection is of high importance. Herein, we present two nanoporous metal–organic frameworks (MOFs), (Me2NH2)[In(CPTTA)]·(DMF)1.5(H2O)3.5 (BUT-174) and (Et2NH2)[In(CPTTA)]·(DEF)(H2O)3 (BUT-175), constructed by In3+ ions and the aromatic-rich tetratopic carboxylate ligands, 5′-(4-carboxylatophenyl)-[1,1′:3′,1″-terphenyl]-3,4′′,5-tricarboxylate (CPTTA4–), which exhibit highly efficient turn-down fluorescence response toward 12 PCBs, especially for three highly toxic non-ortho-substituted PCBs (PCB-77, -126, and -169) with the so-called “coplanar” molecular configuration in various solvents with the limits of detection (LODs) down to the ppb level owing to the inner filter effect between MOFs and PCBs. In addition, the MOFs show high sensing selectivity toward PCBs over many relevant interferents, including hexabromocyclododecane (HBCD), decabromodiphenyl oxide (DBDPO), perfluorobutanoic acid (PFBA), and perfluorooctanoic acid (PFOA), as well as good recyclability and stability. It is demonstrated by this work that structurally well-defined MOFs could be promising sensory materials for the detection of highly toxic PCBs in a facile way.

Chlorine substitution-dependent toxicities of polychlorinated biphenyls to the earthworm Eisenia fetida in soil

Polychlorinated biphenyls (PCBs) with different chlorine substitution patterns often coexist in e-waste-processing sites. However, the single and combined toxicity of PCBs to soil organisms and the influence of chlorine substitution patterns remain largely unknown. Herein, we evaluated the distinct in vivo toxicity of PCB28 (a trichlorinated PCB), PCB52 (a tetrachlorinated PCB), PCB101 (a pentachlorinated PCB), and their mixture to earthworm Eisenia fetida in soil, and looked into the underlining mechanisms in an in vitro test using coelomocytes. After a 28-days exposure, all PCBs (up to 10 mg/kg) were not fatal to earthworms, but could induce intestinal histopathological changes and microbial community alterations in the drilosphere system, along with a significant weight loss. Notably, pentachlorinated PCBs with a low bioaccumulation ability showed greater inhibitory effects on the growth of earthworm than lowly chlorinated PCBs, suggesting that bioaccumulation was not the main determinant of chlorine substitution-dependent toxicity. Furthermore, in vitro assays showed that the highly chlorinated PCBs induced a high-percentage apoptosis of eleocytes in the coelomocytes and significantly activated antioxidant enzymes, indicating that the distinct cellular vulnerability to lowly/highly chlorinated PCBs was the main contributor to the PCBs toxicity. These findings emphasize the specific advantage of using earthworms in the control of lowly chlorinated PCBs in soil due to their high tolerance and accumulation ability.

CYP1A1, VEGFA and Adipokine Responses of Human Adipocytes Co-exposed to PCB126 and Hypoxia.

It is increasingly recognized that hypoxia may develop in adipose tissue as its mass expands. Adipose tissue is also the main reservoir of lipophilic pollutants, including polychlorinated biphenyls (PCBs). Both hypoxia and PCBs have been shown to alter adipose tissue functions. The signaling pathways induced by hypoxia and pollutants may crosstalk, as they share a common transcription factor: aryl hydrocarbon receptor nuclear translocator (ARNT). Whether hypoxia and PCBs crosstalk and affect adipokine secretion in human adipocytes remains to be explored. Using primary human adipocytes acutely co-exposed to different levels of hypoxia (24 h) and PCB126 (48 h), we observed that hypoxia significantly inhibits the PCB126 induction of cytochrome P450 (CYP1A1) transcription in a dose-response manner, and that Acriflavine (ACF)—an HIF1α inhibitor—partially restores the PCB126 induction of CYP1A1 under hypoxia. On the other hand, exposure to PCB126 did not affect the transcription of the vascular endothelial growth factor-A (VEGFA) under hypoxia. Exposure to hypoxia increased leptin and interleukin-6 (IL-6), and decreased adiponectin levels dose-dependently, while PCB126 increased IL-6 and IL-8 secretion in a dose-dependent manner. Co-exposure to PCB126 and hypoxia did not alter the adipokine secretion pattern observed under hypoxia and PCB126 exposure alone. In conclusion, our results indicate that (1) hypoxia inhibits PCB126-induced CYP1A1 expression at least partly through ARNT-dependent means, suggesting that hypoxia could affect PCB metabolism and toxicity in adipose tissue, and (2) hypoxia and PCB126 affect leptin, adiponectin, IL-6 and IL-8 secretion differently, with no apparent crosstalk between the two factors.

Differences in Enantioselective Hydroxylation of 2,2',3,6-Tetrachlorobiphenyl (CB45) and 2,2',3,4',6-Pentachlorobiphenyl (CB91) by Human and Rat CYP2B Subfamilies.

Although polychlorinated biphenyls (PCBs) were commercially banned half a century ago, contamination of the environment and organisms by PCBs is still observed. PCBs show high persistence and bioaccumulation, resulting in toxicity. Among PCBs, chiral PCBs with more than three chlorine atoms at the ortho-position exhibit developmental and neurodevelopmental toxicity. Because toxicity is dependent on the atropisomer, atropisomer-specific metabolism is vital in determining toxicity. However, structural information on enantioselective metabolism remains elusive. Cytochrome P450 (CYP, P450) monooxygenases, particularly human CYP2B6 and rat CYP2B1, metabolize separated atropisomers of 2,2',3,6-tetrachlorobiphenyl (CB45) and 2,2',3,4',6-pentachlorobiphenyl (CB91) to dechlorinated and hydroxylated metabolites. Docking studies using human CYP2B6 predict 4'-hydroxy (OH)-CB45 from (aR)-CB45 as a major metabolite of CB45. Di-OH- and dechlorinated OH-metabolites from human CYP2B6 and rat CYP2B1 are also detected. Several hydroxylated metabolites are derived from CB91 by both P450s; 5-OH-CB91 is predicted as a major metabolite. CB91 dechlorination is also detected by identifying 3-OH-CB51. A stable conformation of PCBs in the substrate-binding cavity and close distance to P450 heme are responsible for high metabolizing activities. As hydroxylation and dechlorination change PCB toxicity, this approach helps understand the possible toxicity of chiral PCBs in mammals.

Polychlorinated biphenyls (PCBs) in soils from typical paddy fields of China: Occurrence, influencing factors and human health risks

The contamination of paddy soils is of great concern since it links to human health via food supply. Limited knowledge is available on PCB residue characteristics and the associated health risks in paddy soils under various environmental conditions. In this study, a soil sampling campaign was conducted in three typical paddy fields, i.e., Sanjiang Plain (SP), Taihu Plain (TP) and Hani Terrace (HT), crossing a transect of 4000 km in China. The concentrations of 29 quantified PCBs varied from 58.6 to 1930 pg/g in paddy soils, with samples at TP showing the highest burden. Tri-CBs were the major homologue group at SP and HT, whereas hexa-CBs at TP. Altitude, temperature, soil organic matter content and soil conductivity well explained the variations in PCB concentrations among sites. The homologue profiles of soil PCBs followed the fractionation theory. In addition, soil conductivity was found to be negatively correlated to low-chlorinated PCBs and positively to high-chlorinated congeners. Furthermore, the toxicities of soil PCBs and the exposure risks through rice intake were estimated. Higher toxicity equivalent quantities and hazard indexes were found at SP than TP and HT, with over one third of the samples displaying health risks. The results of this work highlight the necessity to better understand the occurrence characteristics and the associated health risks of PCBs in soils of rice-growing regions.

Polystyrene nanoplastics aggravated ecotoxicological effects of polychlorinated biphenyls in on zebrafish (Danio rerio) embryos

In view of the accumulation of nanoplastics (NPs) in the food chain of environment and animals, and the good adsorption properties of nano-plastics to toxic substances, it is necessary to explore the influence of NPs in living organisms. In this study, single and joint toxicological effects of polystyrene nanoplastics (PS-NPs, size 80 nm) and polychlorinated biphenyls (PCBs), were explored in freshwater aquatic animal model zebrafish (Danio rerio). Our study found that exposure to single PS-NPs induced mild acute toxicity, albeit the combined exposure of PS-NPs and polychlorinated biphenyls aggravated the toxicity of PCBs in a dose-dependent manner. Results from gene expression profiling showed that NPs exposure could activate detoxification process, resulting in a slight up-regulation of antioxidant genes (sod1, gstp1), bone development genes (bmp2, bmp4) and cardiac gene (tbx20); while PCBs suppressed the detoxification through down-regulation of these genes, and the addition of NPs will exacerbate the impact of PCBs on gene suppression. Importantly, the results of in vivo purification experiments found that NPs showed prolonged retention in liver, intestine and gills of zebrafish and they might have crossed biological barrier and accumulate in lipid-rich tissues and excretion does not appear as the significant pathway for their elimination. In conclusion, the toxic effects of polychlorinated biphenyls on chorionic protected embryos were not significant as zebrafish chorion plays an important role in resisting the invasion of pollutants; PCBs can seriously damage the bone and heart development of zebrafish, while the presence of NPs significantly enhanced the toxicity of PCBs in zebrafish, which is an alarming concern for growing NPs levels and ecological safety in aquatic environment.

Trace metals, polycyclic aromatic hydrocarbons and polychlorinated biphenyls in the surface sediments from Sanya River, China: Distribution, sources and ecological risk

The urban inland river ecosystems are now facing comprehensive pollution and governance pressures. Up to now, few works related to the multiple pollution assessment of trace metals, polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) for the urban inland river sediments have been reported in China. Our study investigated the spatial distribution, ecological risk and potential sources of trace metals, PAHs and PCBs in surface sediment collected from 20 sampling sites of Sanya River, Hainan Province, China. The pollution status and potential ecological risk of trace metals were evaluated using the contamination indexes including geoaccumulation index (Igeo), individual potential ecological risk (Eri), potential ecological risk index (RI) and pollution load index (PLI). Considering the carcinogenicity and toxicity of PAHs and PCBs to human health and the ecological environment, we also analyzed the distributions, sources and adverse biological effects of PAHs and PCBs according to the sediment quality guidelines (SQGs), principal component analysis (PCA) and other source analysis. This study revealed that the surface sediments in Sanya River were extremely slight pollution and showed a very low ecological risk according to Igeo, Eri, PLI and RI results for trace metals. Besides, PAHs and PCBs pollution detected may not pose considerable adverse biological effect to ecological environment in a foreseeable period on the basis of comprehensive research results. The overall surface sediments quality of the Sanya River not seem to pose a serious pollution and ecological risk based on the evaluation results of multiple pollution factors. The study provided detailed information on the multiple pollution status and location of surface sediments, one of the key environmental indicators of international tourism cities, in the Sanya River, which would be useful for the water quality improvement of Sanya River and the environmental remediation of the other coastal ecosystems from different regions.

Extraction of Chlorobenzenes and PCBs from Water by ZnO Nanoparticles

Metal oxide nanoparticles have great potential for selective adsorption and catalytic degradation of contaminants from aqueous solutions. In this study, we employ mass spectrometry and molecular dynamics simulations to better understand the chemical and physical mechanisms determining the affinity of chlorobenzenes and polychlorinated biphenyls (PCBs) for zinc oxide nanoparticles (ZnO NPs). The experiments and simulations both demonstrate that the adsorption coefficients for chlorobenzenes increase steadily with the number of chlorine atoms, while, for PCBs, the relation is more complex. The simulations link this complexity to chlorine atoms at ortho positions hindering coplanar conformations. For a given number of chlorine atoms, the simulations predict decreasing adsorption affinity with increasing numbers of ortho substitutions. Consequently, the simulations predict that some of the highest adsorption affinities for ZnO NPs are exhibited by dioxin-like PCBs, suggesting the possibility of selective sequestration of these most acutely toxic PCBs. Remarkably, the experiments show that the PCB adsorption coefficients of ZnO NPs with diameters ≤ 80 nm exceed those of a soil sample by 5–7 orders of magnitude, meaning that a single gram of ZnO NPs could sequester low levels of PCB contamination from as much as a ton of soil.

Inhibition of hydroxylated polychlorinated biphenyls (OH-PCBs) on sulfotransferases (SULTs)

Polychlorinated biphenyls (PCBs) have been demonstrated as a kind of the persistent organic pollutants (POPs) that could exert complicated influences towards metabolism in human bodies. Since hydroxylated polychlorinated biphenyls (OH-PCBs) are important metabolites of PCBs, our study focuses on investigating the potential inhibitory capability of OH-PCBs on four human sulfotransferase (SULT) isoforms. P-nitrophenol (PNP) was utilized as nonselective probe substrate for this study, and recombinant SULT isoforms were utilized as the enzyme resources. Ultra-performance liquid chromatography (UPLC)-UV detecting system was used to analyze PNP and its metabolite PNP-sulfate. As result, 100 μM of most tested OH-PCBs significantly inhibited the activity of four SULT isoforms. Concentration-dependent inhibition of OH-PCBs towards SULTs was found, and half inhibition concentration values (IC50) of some inhibition processes were determined. Inhibition kinetics (inhibition kinetic type and parameters) were determined using 4′–OH–PCB106 as the representative OH-PCB, SULT1B1 and SULT1E1 as representative SULT isoforms. The inhibition kinetic parameters (Ki) were 1.73 μM and 1.81 μM for the inhibition of 4′–OH–PCB106 towards SULT1B1 and SULT1E1, respectively. In silico docking simulation was utilized to analyze the inhibition capability of 2′–OH–PCB5, 4′–OH–PCB9, 2′–OH–PCB12 towards SULT1A3.All these results obtained in this study are helpful for further understanding the toxicity of PCBs.

The Aryl hydrocarbon receptor mediates reproductive toxicity of polychlorinated biphenyl congener 126 in rats

Polychlorinated biphenyls (PCBs) are endocrine disrupting chemicals with documented, though mechanistically ill-defined, reproductive toxicity. The toxicity of dioxin-like PCBs, such as PCB126, is mediated via the aryl hydrocarbon receptor (AHR) in non-ovarian tissues. The goal of this study was to examine the uterine and ovarian effects of PCB126 and test the hypothesis that the AHR is required for PCB126-induced reproductive toxicity. Female Holzman-Sprague Dawley wild type (n = 14; WT) and Ahr knock out (n = 11; AHR−/−) rats received a single intraperitoneal injection of either corn oil vehicle (5 ml/kg: WT_O and AHR−/−_O) or PCB126 (1.63 mg/kg in corn oil: WT_PCB and AHR−/−_PCB) at four weeks of age. The estrous cycle was synchronized and ovary and uterus were collected 28 days after exposure. In WT rats, PCB126 exposure reduced (P < 0.05) body and ovary weight, uterine gland number, uterine area, progesterone, 17β-estradiol and anti-Müllerian hormone level, secondary and antral follicle and corpora lutea number but follicle stimulating hormone level increased (P < 0.05). In AHR−/− rats, PCB126 exposure increased (P ≤ 0.05) circulating luteinizing hormone level. Ovarian or uterine mRNA abundance of biotransformation, and inflammation genes were altered (P < 0.05) in WT rats due to PCB126 exposure. In AHR−/− rats, the transcriptional effects of PCB126 were restricted to reductions (P < 0.05) in three inflammatory genes. These findings support a functional role for AHR in the female reproductive tract, illustrate AHR's requirement in PCB126-induced reprotoxicity, and highlight the potential risk of dioxin-like compounds on female reproduction.

Effects of Polychlorinated Biphenyls on Animal Reproductive Systems and Epigenetic Modifications.

Polychlorinated biphenyls (PCBs) are a group of highly toxic endocrine-disrupting chemicals comprising 209 homologs. PCBs are extensively found in the environment and can induce typical estrogenic and profound, long-lasting effects on animals. In this article, the introduction of PCB residues into the environment and the pathways of PCB enrichment in animals are described. PCBs are widely deposited and eventually accumulate in human tissues and body fluids through biomagnification. PCBs can significantly decrease animal fertility and interfere with endocrine processes, leading to the development of various diseases and even cancer. The effects of PCBs on the reproductive systems of animals can also be passed to their offspring, indicating that PCBs may affect the epigenetic modification process. There is currently no treatment to effectively inhibit the toxicity of PCBs in organisms; therefore, the severity of PCB toxicity needs to be widely recognized.

The Association of the Polychlorinated Biphenyl Class of Endocrine Disruptors With Polycystic Ovary Syndrome and Thyroid Dysfunction

Introduction:. Polychlorinated biphenyls (PCBs) are a class of endocrine disruptors with a long half-life in the body that are associated with irregular menses, growth and development delay, increased cancer risk, thyroid disorders and an increased risk of diabetes. Higher levels of PCBs have been related to polycystic ovary syndrome (PCOS). PCB toxicity depends on their structure, with coplanar PCBs being most toxic (akin to dioxins); therefore, PCB subtypes were determined to see if they differed in women with PCOS compared to normal controls. Methods:. PCB levels were compared in Caucasian women with (n=29) and without (n=30) PCOS and related to metabolic features. PCBs were fractionated then analysed by high-resolution gas chromatography-unit resolution mass spectrometry. Results:. The control and PCOS groups were age and BMI matched (p=ns); insulin resistance was not different (HOMA 1.7±1.0 v 2±1.6, p=ns) but free androgen index was increased in PCOS (p<0.004). PCB-118, 138, 153 and 180 were found in all subjects, whilst fewer subjects showed PCB-28(15/59), PCB-52(4/59) or PCB-101(26/59). There was no difference for PCB-188,138,153 and 180 between controls and PCOS, but all correlated with increasing age (p<0.01) and decreasing estimated glomerular filtration rate (p<0.05); no correlations with BMI, HOMA, testosterone, TSH or T3 were found; however, PCB-118 (the only coplanar PCB detected) associated with an increased T4/T3 ratio (p<0.01). Conclusion. Despite PCBs being banned over a decade ago, PCBs were detected, but did not differ between age and BMI matched women with and without PCOS. Thyroid dysfunction may be only associated with toxic coplanar PCBs, such as PCB-118 that was associated with a higher T4/T3 ratio.

Comparative study of single cultures and a consortium of white rot fungi for polychlorinated biphenyls treatment.

To evaluate the mycoremediation of polychlorinated biphenyls (PCBs) by either single cultures or binary consortia of Pleurotus pulmonarius LBM 105 and Trametes sanguinea LBM 023. PCBs tolerance, removal capacity, toxicity reduction and ligninolytic enzyme expression were assessed when growing single culture and binary consortium of fungus in 217mgl-1 of a technical mixture of Aroclor 1242, 1254 and 1260 in transformer oil. A decrease in tolerance and variation in ligninolytic enzyme secretion were observed in PCB-amended solid media. Pleurotus pulmonarius LBM 105 mono-culture was able to remove up to 95·4% of PCBs, whereas binary consortium and T. sanguinea LBM 023 could biodegrade about 55% after 24days. Significant detoxification levels were detected in all treatments by biosorption mechanism. Pleurotus pulmonarius LBM 105 in single culture had the best performance regarding PCBs biodegradation and toxicity reduction. Ligninolytic enzyme secretion changed in co-culture. The evaluation of PCBs bioremediation effectiveness of basidiomycetes consortium in terms of PCB removal, toxicity and ligninolytic enzyme production to unravel the differences between using individual cultures or consortium has not been reported. The results from this study enable the selection of P. pulmonarius LBM 105 mono-culture to bioremediate PCBs as it showed higher efficiency compared to binary consortium with T. sanguinea LBM 023 for potential decontamination of PCB-contaminated transformer oil.
