Biotic and Abiotic Interactions in Freshwater Mesocosms Determine Fate and Toxicity of CuO Nanoparticles.

Transformation, dissolution, and sorption of copper oxide nanoparticles (CuO-NP) play an important role in freshwater ecosystems. We present the first mesocosm experiment on the fate of CuO-NP and the dynamics of the zooplankton community over a period of 12 months. Increasingly low (0.08-0.28 mg Cu L-1) and high (0.99-2.99 mg Cu L-1) concentrations of CuO-NP and CuSO4 (0.10-0.34 mg Cu L-1) were tested in a multiple dosing scenario. At the high applied concentration (CuO-NP_H) CuO-NP aggregated and sank onto the sediment layer, where we recovered 63% of Cu applied. For the low concentration (CuO-NP_L) only 41% of applied copper could be recovered in the sediment. In the water column, the percentage of initially applied Cu recovered was on average 3-fold higher for CuO-NP_L than for CuO-NP_H. Zooplankton abundance was substantially compromised in the treatments CuSO4 (p < 0.001) and CuO-NP_L (p < 0.001). Community analysis indicated that Cladocera were most affected (bk = -0.49), followed by Nematocera (bk = -0.32). The abundance of Cladocera over time and of Dixidae in summer was significantly reduced in the treatment CuO-NP_L (p < 0.001; p < 0.05) compared to the Control. Our results indicate a higher potential for negative impacts on the freshwater community when lower concentrations of CuO-NP (<0.1 mg Cu L-1) enter the ecosystem.

Ocean acidification enhances the embryotoxicity of CuO nanoparticles to Oryzias melastigma

Concerns are raised towards individual effects of ocean acidification (OA) and engineered nanoparticles (NPs) on marine organisms. However, there are scarce studies regarding nanotoxicity under OA conditions. We investigated the combined effects of OA (pHs, 7.70 and 7.40) and CuO NPs on the embryotoxicity of marine medaka Oryzias melastigma and the bioavailability of CuO NPs in embryos. The results showed that OA alleviated the aggregation of CuO NPs and promoted the dissolution of CuO NPs in seawater (increased by 0.010 and 0.029 mg/L under pHs 7.70 and 7.40, respectively). Synergistic effects of OA with CuO NPs on medaka embryos were observed as indicated by much higher mortality and oxidative damage. Importantly, the enhanced toxicity of CuO NPs to medaka embryos under OA conditions mainly originated from the higher bioavailability of particulate CuO (e.g., 30.28 mg/kg at pH 7.40) rather than their released Cu2+ ions (e.g. 3.04 mg/kg at pH 7.40). The weaker aggregation of NPs under OA conditions resulted in higher penetration of individual particles (or small aggregates) into embryos through the micropyle and chorionic pores, causing enhanced bioavailability of NPs. The obtained results provided underlying insights into understanding the risk of NPs to marine ecosystem under OA conditions.

Antagonism toxicity of CuO nanoparticles and mild ocean acidification to marine algae

The toxicity of CuO nanoparticles (NPs) to marine microalgae (Emiliania huxleyi) under ocean acidification (OA) conditions (pHs 8.10, 7.90, 7.50) was investigated. CuO NPs (5.0 mg/L) caused significant toxicity (e.g., 48-h growth inhibition, 20%) under normal pH (8.10), and severe OA (pH 7.50) increased the toxicity of CuO NPs (e.g., 48-h growth inhibition, 68%). However, toxicity antagonism was observed with a growth inhibition (48 h) decreased to 37% after co-exposure to CuO NPs and mild OA (pH 7.90), which was attributed to the released Cu2+ ions from CuO NPs. Based on biological responses as obtained from RNA-sequencing, the dissolved Cu2+ ions (0.078 mg/L) under mild OA were found to increase algae division (by 17%) and photosynthesis (by 28%) through accelerating photosynthetic electron transport and promoting ATP synthesis. In addition, mild OA enhanced EPS secretion by 41% and further increased bioavailable Cu2+ ions, thus mitigating OA-induced toxicity. In addition, excess Cu2+ ions could be transformed into less toxic Cu2S and Cu2O based on X-ray absorption near-edge spectroscopy (XANES) and high-resolution transmission electron microscopy (HR-TEM), which could additionally regulate the antagonism effect of CuO NPs and mild OA. The information advances our knowledge in nanotoxicity to marine organisms under global climate change.

Synthesis, biomedical applications, and toxicity of CuO nanoparticles.

Versatile nature of copper oxide nanoparticles (CuO NPs) has made them an imperative nanomaterial being employed in nanomedicine. Various physical, chemical, and biological methodologies are in use for the preparation of CuO NPs. The physicochemical and biological properties of CuO NPs are primarily affected by their method of fabrication; therefore, selectivity of a synthetic technique is immensely important that makes these NPs appropriate for a specific biomedical application. The deliberate use of CuO NPs in biomedicine questions their biocompatible nature. For this reason, the present review has been designed to focus on the approaches employed for the synthesis of CuO NPs; their biomedical applications highlighting antimicrobial, anticancer, and antioxidant studies; and most importantly, the in vitro and in vivo toxicity associated with these NPs. This comprehensive overview of CuO NPs is unique and novel as it emphasizes on biomedical applications of CuO NPs along with its toxicological assessments which would be useful in providing core knowledge to researchers working in these domains for planning and conducting futuristic studies. KEY POINTS: • The recent methods for fabrication of CuO nanoparticles have been discussed with emphasis on green synthesis methods for different biomedical approaches. • Antibacterial, antioxidant, anticancer, antiparasitic, antidiabetic, and antiviral properties of CuO nanoparticles have been explained. • In vitro and in vivo toxicological studies of CuO nanoparticles exploited along with their respective mechanisms.

OsCERK1 Contributes to Cupric Oxide Nanoparticles Induced Phytotoxicity and Basal Resistance against Blast by Regulating the Anti-Oxidant System in Rice.

CuO NPs (cupric oxide nanoparticles) are widely used in various fields due to their high electrical conductivity, electronic correlation effect, and special physical property. Notably, CuO NPs have good application prospects in agricultural production because of its antifungal activity to prevent crop diseases. However, the increasing release of CuO NPs into the environment has resulted in a serious threat to the ecosystem, including plants. Previous studies have reported the toxicity of CuO NPs on rice, but little is known about the underlying molecular mechanisms or specific genes involved in the response to CuO NPs. In this study, we found that the rice well-known receptor Chitin Elicitor Receptor Kinase 1 (OsCERK1), which is essential for basal resistance against pathogens, is involved in CuO NPs stress in rice. Knockout of OsCERK1 gene resulted in enhanced tolerance to CuO NPs stress. Furthermore, it was revealed that OsCERK1 reduces the tolerance to CuO NPs stress by regulating the anti-oxidant system and increasing the accumulation of H2O2 in rice. In addition, CuO NPs treatment significantly enhances the basal resistance against M. oryzae which is mediated by OsCERK1. In conclusion, this study demonstrated a dual role of OsCERK1 in response to CuO NPs stress and M. oryzae infection by modulating ROS accumulation, which expands our understanding about the crosstalk between abiotic and biotic stresses.

Ethylene accelerates copper oxide nanoparticle-induced toxicity at physiological, biochemical, and ultrastructural levels in rice seedlings.

The enormous use of metal-based nanoparticles (NPs) in different sectors may result in enhanced accumulation in agricultural soil, which could impose negative effects on crop productivity. Hence, strategies are needed to explore the mechanisms of copper oxide nanoparticle (CuO NP)-induced toxicity in crops. The present study aimed to investigate the involvement of ethylene in CuO NP-induced toxicity in rice seedlings. Here, our results indicate that 450mg L-1 of CuO NPs induced toxic effects in rice seedlings. Thus, it was evidenced by the reduced plant biomass accumulation, enhanced oxidative stress indicators, and cellular ultrastructural damages. More importantly, the exogenous supply of ethylene biosynthesis and signaling antagonists cobalt (Co) and silver (Ag) respectively provided tolerance and improved the defense system of rice seedlings against CuO NP toxicity. The ethylene antagonists could significantly reduce the extent of ultrastructural and stomatal damage by controlling the ROS accumulation in rice seedlings under CuO NP stress. Furthermore, Co and Ag augmented the antioxidant defense system against CuO NP-induced toxicity. Contrary to that, all oxidative damage attributes were further enhanced exogenous application of ethylene biosynthesis precursor [1-aminocyclopropane-1-carboxylic acid (ACC)] in the presence of CuO NPs. In addition, ACC could increase the CuO NP-induced stomatal and ultrastructural damages by reducing the ROS-scavenging ability in rice seedlings. Taken together, these results indicate the involvement of ethylene in CuO NP-induced toxicity in rice seedlings.

CuO nanoparticles in irrigation wastewater have no detrimental effect on rice growth but may pose human health risks

The possibility of metal-based nanoparticles (NPs) being released into agricultural soils via sewage systems has raised widespread concern about their negative effects on crop plants, soils, and potential risks to human health via the food chain. The objectives of this study were to (i) determine the effect of CuO NPs in irrigation water on plant growth and Cu accumulation in a rice–soil system using continuous sub-irrigation with treated wastewater (CSI), and (ii) assess the Cu exposure and potential health risk associated with rice consumption. CuO NPs were examined in treated municipal wastewater (TWW) at environmentally acceptable concentrations (0, 0.02, 0.2, and 2.0 mg Cu L−1), allowing for effluent discharge and/or crop irrigation reuse. Low CuO NP concentrations in TWW had no adverse effect on plant growth, yield, or grain quality. Cu accumulation significantly increased in various parts of rice plants and paddy soils at 2.0 mg Cu L−1. CuO NPs had no discernible effect on rice plants when compared to CuSO4 at 0.2 mg Cu L−1. The estimated daily intake of Cu derived from inadvertent consumption of Cu-contaminated rice (by CuO NPs in TWW) for young children aged 0–6 years exceeded the oral reference dose for toxicity. Overall, we found no acute toxicity of CuO NPs in TWW to rice plants, but significant Cu accumulation in grains. This implies that there is a high risk of human health problems associated with rice that was intensively irrigated with TWW containing CuO NPs.

Simultaneous exposure of wheat (Triticum aestivum L.) to CuO and S nanoparticles alleviates toxicity by reducing Cu accumulation and modulating antioxidant response

Widespread use of metal-based nanoparticles (NPs) may result in the increased accumulation of metals in agricultural soil, which could affect crop productivity and contaminate the food-chain. The effect of sulfur nanoparticles (S NPs, 200 mg/L) co-exposure on the toxicity of CuO nanoparticles (CuO NPs, 25 and 50 mg/L) to wheat seedlings was investigated in a hydroponic system. CuO NPs exposure significantly inhibited the growth of wheat seedlings, causing 43.6% and 54.1% decreases in the fresh biomass of plants and 82.8% and 83.1% decrease in the total chlorophyll contents at 25 and 50 mg/L (CuONP25 and CuONP50), respectively, as compared to controls. CuO NPs exposure at both concentrations increased the malondialdehyde (MDA) content in shoot and root tissues by 66.4–67.9% and 47.7–48.8%, respectively. Further, CuO NPs exposure elevated the activities POD, SOD, and CAT by 2.19–2.27, 5.82–6.09, and 1.44–1.95 times in roots, and by 45.2–67.8%, 86.7–154.5%, and 22.5–56.1% in shoot, respectively, in comparison to control. The addition of S NPs alone increased wheat biomass by 11.0% and total chlorophyll contents by 4.4%, compared to controls. Further, simultaneous exposure to S NPs (200 mg/L) and CuO NPs (25 or 50 mg/L) alleviated the CuO NPs toxicity; wheat biomass was 47.8% and 37.7% higher in CuONP25 + SNP and CuONP50 + SNP treatments, respectively, as compared to CuO NPs alone treated plants. Co-exposed plants showed reduced levels of total reactive oxygen species (ROS), O2·- and H2O2. Additionally, S NPs exposure reduced Cu uptake and accumulation in both root and shoot tissue by 32.2–54.4% and 38.3–57.5%, respectively. In summary, S NPs alleviated CuO NPs toxicity to wheat seedlings, most likely by reducing Cu bioavailability and accumulation of Cu in plant tissues, and also altered S nutrition and the modulation of antioxidant response in plants. These results showed that S NPs application has the potential to alleviate CuO NP toxicity and increase wheat productivity affected by metals toxicity.

Copper Oxide Nanoparticle Diameter Mediates Serum‐Sensitive Toxicity in BEAS‐2B Cells

Copper oxide (CuO) nanoparticles (NPs) are abundant in manufacturing processes, but they are an airway irritant. In vitro pulmonary toxicity of CuO NPs has been modeled using cell lines such as human bronchial epithelial cell line BEAS‐2B. In 2D in vitro culture, BEAS‐2B undergoes squamous differentiation due to the presence of serum. Differentiation is part of the repair process of lung cells in vivo that helps to preserve the epithelial lining of the respiratory tract. Herein, the effects of serum on the hydrodynamic diameter, cellular viability, cellular differentiation, and cellular uptake of 5 and 35 nm CuO NPs are investigated, and the mean cell area is used as the differentiation marker for BEAS‐2B cells. The results demonstrate that the hydrodynamic diameter decreases with the addition of serum to the culture medium. Serum also increases the mean cell area, and only affects dose‐dependent cytotoxicity of 35 nm CuO NPs, while simultaneously having no effect on intracellular Cu2+. This study presents evidence that both NP size and the presence of serum in culture media influence the relative viability of BEAS‐2B cells following CuO NP exposure and highlights a critical need for carefully designed experiments and accurately reported conditions.

Synergistic influence of CuO nanoparticles and spectral lights transforms biomass, antioxidative response, and antioxidants in Brassica nigra

Some nanoparticles (NPs) have photocatalytic and photodynamic properties that change on variation in wavelength of light carrying different energies. To access photo-dynamic toxicity of CuO NPs, Brassica nigra seeds were in vitro grown on CuO NPs supplemented media under the exposure of five different spectral lights (white, blue, red, yellow and green). Light wavelength significantly affected shoot and root length and fresh weight of B. nigra and CuO NPs exerted different level of toxicity under these lights. Noteworthy variations were observed in free radical scavenging activity, total antioxidantive response and total reducing power potential when plants were grown in presence of NPs and spectral lights. Significant variations in total phenolics and flavonoids contents were observed in shoots as compared with roots. HPLC analysis presented variation in phenolics in shoots extracts under synergistic influence of NPs and light regime. The study shows that nanoparticles exert different level of toxicity under different wavelengths of lights. Resultantly plants show variations in morphological and biochemical responses. Consequently secondary metabolites produce in plants at varying concentrations to mitigate nanotoxicity under photodynamic response. Phenolics and flavonoid production enhance in plant grown in presence of CuO NPs and different wavelength of striking light.

Exogenous Fe2+ alleviated the toxicity of CuO nanoparticles on Pseudomonas tolaasii Y-11 under different nitrogen sources.

Extensive use of CuO nanoparticles (CuO-NPs ) inevitably leads to their accumulation in wastewater and toxicity to microorganisms that effectively treat nitrogen pollution. Due to the effects of different mediums, the sources of CuO-NPs-induced toxicity to microorganisms and methods to mitigating the toxicity are still unclear. In this study, CuO-NPs were found to impact the nitrate reduction of Pseudomonas tolaasii Y-11 mainly through the action of NPs themselves while inhibiting the ammonium transformation of strain Y-11 through releasing Cu2+. As the content of CuO-NPs increased from 0 to 20 mg/L, the removal efficiency of NO3− and NH4+ decreased from 42.29% and 29.83% to 2.05% and 2.33%, respectively. Exogenous Fe2+ significantly promoted the aggregation of CuO-NPs, reduced the possibility of contact with bacteria, and slowed down the damage of CuO-NPs to strain Y-11. When 0.01 mol/L Fe2+ was added to 0, 1, 5, 10 and 20 mg/L CuO-NPs treatment, the removal efficiencies of NO3- were 69.77%, 88.93%, 80.51%, 36.17% and 2.47%, respectively; the removal efficiencies of NH4+ were 55.95%, 96.71%, 38.11%, 20.71% and 7.43%, respectively. This study provides a method for mitigating the toxicity of CuO-NPs on functional microorganisms.

Cytotoxicological evaluation of copper oxide nanoparticles on green algae, bacteria and crustacean systems.

Copper oxide (CuO) nanoparticles (NPs) have been utilized in several industries including textile, consumer products, medical, automobiles etc. The discharge of industrial effluents in environment increased the probability of CuO NPs contamination in the ecosystem. The present investigation used CuO NPs to determine the toxic effect on Lyngbya species, fresh water algae isolated from natural pond, bacterial species Pseudomonas aeruginosa and Staphylococcus aureus and a crustacean species Daphnia magna. The NPs average diameter and zeta potential was estimated to be 45 ± 3nm and 29 ± 1.78mV respectively. The results showed that 0.1µg/mL CuO NPs showed the growth inhibition of 47 ± 2% on Lyngbya sp. after 5 days of incubation. The CuO NPs also showed toxic effect to bacterial systems such as P. aeruginosa and S. aureus and crustacean system D. magna. Further, there was an increased lipid peroxidation and generation of reactive oxygen species (ROS) in algal cells observed up on NPs exposure. The exposure of NPs suppressed the antioxidant defense system. The amount of glutathione was reduced after the exposure of NPs. The study suggested the role of ROS in toxicity of algal and bacterial systems. The present study pointed out the potent toxicity of CuO NPs to the organisms present in the aquatic environment.

A global metabolomic insight into the oxidative stress and membrane damage of copper oxide nanoparticles and microparticles on microalga Chlorella vulgaris

To compare aquatic organisms’ responses to the toxicity of copper oxide (CuO) nanoparticles (NPs) with those of CuO microparticles (MPs) and copper (Cu) ions, a global metabolomics approach was employed to investigate the changes of both polar and nonpolar metabolites in microalga Chlorella vulgaris after 5-day exposure to CuO NPs and MPs (1 and 10 mg/L), as well as the corresponding dissolved Cu ions (0.08 and 0.8 mg/L). Unchanged growth, slight reactive oxygen species production, and significant membrane damage (at 10 mg/L CuO particles) in C. vulgaris were demonstrated. A total of 75 differentiated metabolites were identified. Most metabolic pathways perturbed after CuO NPs exposure were shared by those after CuO MPs and Cu ions exposure, including accumulation of chlorophyll intermediates (max. 2.4–5.2 fold), membrane lipids remodeling for membrane protection (decrease of phosphatidylethanolamines (min. 0.6 fold) and phosphatidylcholines (min. 0.2–0.7 fold), as well as increase of phosphatidic acids (max. 1.5–2.9 fold), phosphatidylglycerols (max. 2.2–2.3 fold), monogalactosyldiacylglycerols (max. 1.2–1.4 fold), digalactosylmonoacylglycerols (max. 1.9–3.8 fold), diacylglycerols (max. 1.4 fold), lysophospholipids (max. 1.8–3.0 fold), and fatty acids (max. 3.0–6.2 fold)), perturbation of glutathione metabolism induced by oxidative stress, and accumulation of osmoregulants (max. 1.3–2.6 fold) to counteract osmotic stress. The only difference between metabolic responses to particles and those to ions was the accumulation of fatty acids oxidation products: particles caused higher fold changes (particles/ions ratio 1.9–3.0) at 1 mg/L and lower fold changes (particles/ions ratio 0.4–0.7) at 10 mg/L compared with ions. Compared with microparticles, there was no nanoparticle-specific pathway perturbed. These results confirm the predominant role of dissolved Cu ions on the toxicity of CuO NPs and MPs, and also reveal particle-specific toxicity from a metabolomics perspective.

Assessment of toxic interaction of nano zinc oxide and nano copper oxide on germination of Raphanus sativus seeds.

Multiple applications of nanoparticles (NPs) could result in their potential release into agricultural systems and raised concerns about food safety. The NPs once released in the environment may interact with numerous pollutants, including other NPs. Present study assessed the impact of a single (CuO and ZnO NPs) and binary mixture (CuO+ZnO NPs) on the germination of Raphanus sativus seeds with a wide range of exposure concentration (0-1000 mg/L). Both the NPs have shown a deleterious effect on seeds at exposure concentration greater than 10 mg/L. Antagonistic interaction between effects of CuO and ZnO NPs on seeds was noticed for all the exposed concentrations. CuO NPs showed higher absorption capacity on the seedling surface than ZnO NPs. Internal uptake of Zn in ZnO NP-exposed seedlings was found to be greater than that due to CuO NP-exposed seedlings. Three different types of exposure adversely affected seed germination (reduction in root length, shoot length, and fresh weight). Reduction in growth parameters (length and weight) with concentration was compared using log-logistic dose-response model of "DRC" package of the "R" software, and EC50 was determined. As per EC50 values, the toxicity of CuO NPs was found to be maximum followed by CuO+ZnO NPs and then minimum for ZnO NPs. Seedlings accumulated Cu and Zn metals, and higher uptake was recorded for Zn (reported as mg/g seedling dry weight). The order of toxicity was found as CuO NPs > binary mixture (CuO+ZnO) NPs > ZnO NPs. Exposure concentration greater than 10 mg/L resulted in significant toxicity and uptake in germinated seedlings. These findings indicated that exposure of the mixture of NPs during germination might give different effects and thus, further attempts could prove quite beneficial to the literature.

Interaction of CuO nanoparticles with duckweed (Lemna minor. L): Uptake, distribution and ROS production sites

CuO engineered nanoparticles (NPs) are of increasing concern due to their extensive use in daily life and adverse effect on aquatic organisms. The investigations on the toxicity of CuO NPs to aquatic plants through uptake from roots versus fronds are limited. This paper discusses the interactions of CuO NPs with Lemna minor, a floating plant. After CuO NPs (150 μg L−1) exposure for 7 days, the frond number, frond surface area and dry weights of whole plants significantly decreased by 32%, 47% and 33%; the responses were dose-dependent. Microscopy imaging showed that the epidermis was severely damaged in fronds, edges were severely sloughed off and cell integrity was damaged in roots. Shrinkage of both chloroplast and starch grains were observed in the frond cells. Internalization of CuO NPs in root and frond cells during CuO NPs (1 mg L−1) exposure was confirmed with the root Cu levels of Lemna minor being three times higher than the fronds by using transmission electron microscopy and flame atomic absorption spectrophotometry. Reactive oxygen species, mainly H2O2 (increased by 56%) and ·OH (increased by 57%), accumulated in Lemna minor tissues in response to CuO NPs exposure. Moreover, chloroplasts were confirmed as a site of ROS production. These findings are helpful for better understanding the biological responses of aquatic plants upon NPs exposure.

Mechanisms of oxidative stress caused by CuO nanoparticles to membranes of the bacterium Streptomyces coelicolor M145

Toxic effects of widely used CuO nanoparticles (NPs) on the genus Streptomyces has been seldom studied. This work investigated toxicities of several sizes of CuO nanoparticles (NPs) to Streptomyces coelicolor M145 (S. coelicolor M145). Compared with NPs, toxicity of micrometer-sized CuO on M145 was trivial. In 0.9% NaCl, when the concentration of CuO NPs was 100 mg/L, survival of bacteria increased from 18.3% in 20 nm particles to 31.1% in 100 nm particles. With increasing concentrations of CuO, the level of ROS gradually increased and there were significant differences (p ＜ 0.05) in ROS exposed to 20, 40 and 100 nm (80 nm) CuO NPs. In TSBY medium, toxicity of CuO NPs was less and mainly attributed to release of Cu2+, analysis by confocal laser scanning microscope (CLSM) showed that size of the mycelium did not change although some individual bacteria died. This was likely due to Cu2+ released from NPs entering cells through the membrane, while in 0.9% NaCl, lesions on membranes was caused by NPs outside the bacteria. This research indicated that toxicity of CuO NPs to S. coelicolor, is related to both size of NPs and is dependent on characteristics of the medium. CapsuleThis is the first time to measure the toxicity of nano materials to Streptomyces, and toxic CuO NPs to Streptomyces have been shown to differ depending on medium.

Synthesis methods influence characteristics, behaviour and toxicity of bare CuO NPs compared to bulk CuO and ionic Cu after in vitro exposure of Ruditapes philippinarum hemocytes

Copper oxide (CuO) nanoparticles (NPs) are increasingly investigated, developed and produced for a wide range of industrial and consumer products. Notwithstanding their promising novel applications, concern has been raised that their increased use and disposal could consequently increase their release into marine systems and potentially affect species within. To date the understanding of factors and mechanisms of CuO (nano-) toxicity to marine invertebrates is still limited. Hence, we studied the characteristics and behaviour of two commercially available CuO NPs of similar size, but produced employing distinct synthesis methods, under various environmentally and experimentally relevant conditions. In addition, cell viability and DNA damage, as well as gene expression of detoxification, oxidative stress, inflammatory response, DNA damage repair and cell death mediator markers were studied in primary cultures of hemocytes from the marine clam Ruditapes philippinarum and, where applicable, compared to bulk CuO and ionic Cu (as CuSO4) behaviour and effects. We found that the synthesis method can influence particle characteristics and behaviour, as well as the toxicity of CuO NPs to Ruditapes philippinarum hemocytes. Our results further indicate that under the tested conditions aggregating behaviour influences the toxicity of CuO NPs by influencing their rate of extra- and intracellular dissolution. In addition, gene expression analysis identified similar transcriptional de-regulation for all tested copper treatments for the here measured suite of genes. Finally, our work highlights various differences in the aggregation and dissolution kinetics of CuO particles under environmental (marine) and cell culture exposure conditions that need consideration when extrapolating in vitro findings.

Effects of Copper Nanoparticles (CuO NPs) on Crop Plants: a Mini Review

Nanoparticles (NPs) received great attention due to their unique properties and beneficiary applications in various sectors. The rapid growth of NPs production and its abundant uses create additional risks on an anthropogenically modified ecosystem, and consequently on human beings. The main aim of this review article is to explore the possible threats imposed by CuO NPs on cultivated crop plants. We searched PubMed, Google Scholar, and Web of Science portals for the literature review to get latest updated information and developments in the field of toxicity of CuO NPs on cultivated plants. This review article clearly denoted the toxic effects of CuO NPs on cultivated crop plants by inhibiting seed germination, decreases in the shoot and root lengths, reduction in photosynthesis and respiration rate, and morphological as well enzymatic changes. The information is significant to researchers and policymakers to define limits and future prospectives.

Toxicity of Copper Oxide (CuO) Nanoparticles on Human Blood Lymphocytes.

CuO nanoparticles (CuO-NPs) serve several important functions in human life, particularly in the fields of medicine, engineering, and technology. These nanoparticles have been utilized as catalysts, semiconductors, sensors, gaseous and solid ceramic pigments, and magnet rotatable devices. Further use for CuO-NPs has been employed in the pharmaceutical industry especially in the production of anti-microbial fabric treatments or prevention of infections caused by Escherichia coli and methicillin-resistant Staphylococcus aureus. Two key potential routes of exposure to CuO-NPs exist through inhalation and skin exposure. Toxicity of these nanoparticles has been reported in various studies; however, no study as of yet has investigated the complete cellular mechanisms involved in CuO-NPs toxicity on human cells. The aim of this study was to determine the cytotoxicity of CuO-NPs on human blood lymphocytes. Blood lymphocytes were obtained from healthy male subjects through the use of Ficoll polysaccharide subsequently by gradient centrifugation. The following parameters were assayed in blood lymphocytes after a 6-h incubation with different concentrations of CuO-NPs: cell viability, reactive oxygen species (ROS) formation, lipid peroxidation, cellular glutathione levels, and mitochondrial and lysosomal damage. Our results demonstrate that CuO-NPs, in particular, decreased cell viability in a concentration-dependent manner and the IC50 determined was 382μM. CuO-NP cytotoxicity was associated with significant increase at intracellular ROS level and loss of mitochondrial membrane potential and lysosomal membrane leakiness. Hence, CuO-NPs are shown to effectively induce oxidative stress in addition to inflict damage on mitochondria and lysosomes in human blood lymphocytes.

Evaluation of the effect of test medium on total Cu body burden of nano CuO-exposed Daphnia magna: A TXRF spectroscopy study

Toxicity of Cu and Cu-based nanoparticles (NPs) to aquatic biota is usually mitigated in natural freshwater compared to organics-free artificial freshwater. The main aim of this study was to evaluate whether mitigated toxicity is accompanied by lower total copper body burden in the freshwater crustacean Daphnia magna and whether CuO NPs are more hazardous in this aspect than soluble Cu salts.Total copper body burden in different media (OECD202 artificial freshwater and two natural freshwaters) was measured by a relatively novel technique - total reflection X-ray fluorescence (TXRF) spectroscopy - which proved suitable for the analysis of individual juvenile daphnids. Mean copper body burden was 2.8–42 times higher in daphnids exposed to CuO NPs (0.05 mg Cu/L and 1 mg Cu/L) than in daphnids exposed to equal or equitoxic concentrations (0.025 mg Cu/L and 0.05 mg Cu/L) of CuSO4. Using natural freshwater instead of artificial one resulted in increased copper burden after exposure to CuO NPs but not after exposure to Cu salt. After 24 h post-exposure depuration in the presence of algae Raphidocelis subcapitata, total copper body burden in daphnids exposed to CuO NPs sharply decreased while in daphnids exposed to Cu salt it did not. Despite the CuO NP toxicity mitigating effect of natural freshwater, total copper body burden of aquatic crustaceans in natural waterbodies may be greater than could be predicted based on the results obtained using artificial freshwater as the test medium.
