Exopolysaccharides from Enterococcus faecium and Streptococcus thermophilus: Bioactivities, gut microbiome effects, and fermented milk rheology

Exopolysaccharides (EPSs) are carbohydrate polymers that can be produced from probiotic bacteria. This study characterized the EPSs from Enterococcus faecium (EPS-LB13) and Streptococcus thermophilus (EPS-MLB10) and evaluated their biological and technological potential. The EPSs had high molecular weight and different monosaccharide compositions. The EPSs exhibited various biological activities at 250 mg/L, such as scavenging free radicals (10 % to 88.8 %), enhancing antioxidant capacity (714 to 2848 µg/mL), inhibiting pathogens (53 % to 74 %), and suppressing enzymes and cancer cells (2 % to 83 %), etc. The EPSs supported the growth of beneficial gut bacteria from Proteobacteria, Bacteroidetes, Firmicutes, and Acinetobacter in fecal fermentation with total Short-chain fatty acids production from 5548 to 6023 PPM. Moreover, the EPSs reduced the gelation time of fermented skimmed bovine milk by more than half. These results suggest that the EPSs from LB13 and MLB10 have promising applications in the dairy and pharmaceutical industries.

Effects of Lacticaseibacillus paracasei SNB-derived postbiotic components on intestinal barrier dysfunction and composition of gut microbiota

The bacterial surface components are considered as effector molecules and show the potential to support intestinal health, but the detailed mechanism of how the gut microbiota changes after the intervention of surface molecules is still unknown. In the present study, capsular polysaccharide (B-CPS) and surface layer protein (B-SLP) were extracted from Lacticaseibacillus paracasei S-NB. The protective effect of direct administration of B-CPS (100 μg/mL) and B-SLP (100 μg/mL) on intestinal epithelial barrier dysfunction was verified based on the LPS-induced Caco-2 cell model. Additionally, the B-CPS and B-SLP could be utilized as carbon source and nitrogen source for the growth of several Lactobacillus strains, respectively. The postbiotic potential of B-CPS and B-SLP was further evaluated by in vitro fermentation with fecal cultures. The B-CPS and a combination of B-CPS and B-SLP regulated the composition of gut microbiota by increasing the relative abundances of Bacteroides, Bifidobacterium, Phascolarctobacterium, Parabacteroides, Subdoligranulum and Collinsella and decreasing the abundance of pathogenic bacteria like Escherichia-Shigella, Blautia, Citrobacter and Fusobacterium. Meanwhile, the total short-chain fatty acid production markedly increased after fermentation with either B-CPS individually or in combination with B-SLP. These results provided an important basis for the application of B-CPS and B-SLP as postbiotics to improve human intestinal health.

Effect of ellagic and gallic acid on the mitigation of methane production and ammonia formation in an in vitro model of short-term rumen fermentation

Ruminant production is an important source of animal proteins for human nutrition. However, ruminants contribute to about 30% of anthropogenic methane (CH4) emissions worldwide. The reduction of CH4 emissions could represent an important strategy against climate warming. Tannins can play an important role in the mitigation of CH4 emissions from ruminants. However, their mode of action is not yet well known. Thus, the present study aimed to gain a better understanding of the effect of ellagic acid (EA) and gallic acid (GA) on rumen fermentation using a model of short-term in vitro rumen fermentation. The basal diet (hay) was supplemented with EA and GA in five treatments (mg/g dry matter): i) EA 75, ii) EA 150, iii) GA 75, iv) GA 150 and v) EA 75 + GA 75. After a 24 h incubation, pH, ammonia formation, gas production, short-chain fatty acids (SCFA), in vitro organic matter digestibility (IVOMD) and the microbial count were assessed. Total gas production and digestible organic matter (dOM) were decreased after all the treatments, except for GA 75. The treatments EA 150 and EA+GA significantly decreased CH4 production per unit of dietary DM, dOM, CO2 and SCFA. Ammonia production was significantly decreased by EA 150 and EA+GA. EA and GA differently affected the relative abundance of selected bacterial taxa in rumen microbiota. To conclude, EA 150 and EA+GA exerted a significant effect on the reduction of CH4 emissions and ammonia formation, but affecting also the rumen degradability of the diet and the total SCFA production, whereas EA 75 and GA 75 were not effective as EA 150 and EA+GA on CH4 and ammonia, but were less detrimental on feed degradability and SCFA. Further studies are needed to determine whether the beneficial and detrimental effects of tannins on rumen fermentation can be dissociated.

Effect of Pretreatments on Releasing of Bound Phenolics from Dried Bamboo Shoots during Simulated In Vitro Colonic Fermentation

Salt-dried bamboo shoots (SDBS), steamed dried bamboo shoots (TDBS), fumigation-dried bamboo shoots (UDBS), and fermentation-dried bamboo shoots (FDBS) are four popular types of dried bamboo shoots (DBS) enjoyed by Chinese people. In this work, we aimed at investigating the release pattern of bound phenolics from DBS prepared by different processes on simulated in vitro colonic fermentation. The results showed that nine phenolic compounds were identified from DBS, including seven phenolic acids and two flavonoids. Also, the contents of chlorogenic acid, p-coumaric acid, and ferulic acid were higher. Among the four types of DBS, FDBS exhibited the highest release of bound phenolics during colonic fermentation, with a value of 3.619 mg GAE/g DW. In addition, all the DBS produced four short-chain fatty acids (SCFAs), with the highest content of acetic acid at 7.721 mmol/L, followed by propionic acid and valeric acid, and the lowest content of butyric acid at only 0.023 mmol/L, along with a gradual decrease in their cellulose, hemicellulose, and lignin contents. Concurrently, confocal laser scanning microscopy (CLSM) observation images showed that reduced red fluorescence exhibited a decrease in bound phenolics content, while reduced green fluorescence indicated a decrease in fiber content. A significant positive correlation ( P < 0.05) was observed between the release of bound phenolics and total SCFA production during colonic fermentation of SDBS, TDBS, UDBS, and FDBS, with correlation coefficients of 0.632, 0.844, 0.717, and 0.916, respectively, and a significant negative correlation ( P < 0.05) with the IC50 values for the inhibition of α-amylase and α-glucosidase, with correlation coefficients of −0.643, −0.613, −0.823, and −0.839, respectively. The four DBS were ranked in descending order of bound phenolic release, SCFA production, dietary fiber reduction, and inhibitory activity associated with the metabolic syndrome: FDBS, TDBS, SDBS, and UDBS. DBS released large amounts of bound phenolics and produced SCFAs during colonic fermentation, which might be potentially beneficial for maintaining intestinal microbiota balance.

The Novel Synbiotic, AG1®, Increases Short-Chained Fatty Acid Production in the Simulator of Human Intestinal Microbial Ecosystem (SHIME) Model®

Recently, there is growing usage of prebiotics and probiotics as dietary supplements due to their purported health benefits. AG1® (AG1) is a novel foundational nutrition supplement which contains vitamins, minerals, phytonutrients, wholefood concentrates, adaptogens, and functional mushrooms. AG1 could be classified as a synbiotic because it contains traditional and non-traditional prebiotics (e.g., inulin and phytonutrients) as well as lactic-acid-producing probiotics. The purpose of this study was to employ the Simulator of Human Intestinal Microbial Ecosystem (SHIME®) model, which measures various aspects of gastrointestinal fermentation, to investigate the synbiotic effects of AG1. The SHIME experiment quantified gas production, changes in pH, and byproducts of carbohydrate and protein fermentation at baseline, 1, 24, and 48 h following the administration of AG1 or a blank control. The results indicated that AG1 significantly increased (p < 0.05; 41.9% increase) the production of total short-chain fatty acids (SCFAs) including acetate (p = 0.001; 49.0% increase) and propionate (p < 0.001; 70.8% increase). Regarding non-carbohydrate fermentation byproducts, AG1 produced a small but significant increase in ammonium production (p = 0.02; 5.1% increase) but did not promote significant branched-chain SCFA production. These data suggest fermentation occurred in a transplanted human colonic microbiota and these processes were enhanced by the AG1 nutritional supplement. Ultimately, AG1 showed preclinical evidence as a synbiotic given the significant increases in total SCFA production, acetate, propionate, and other metabolic byproducts of fermentation.

Fermentation patterns of prebiotics fructooligosaccharides-SCFA esters inoculated with fecal microbiota from ulcerative colitis patients.

Ulcerative colitis (UC) is believed to arise from an imbalance between the intestinal microbiota and mucosal immunity, leading to excessive intestinal inflammation. Modulating the gut microbial community through dietary components presents a valuable strategy in aiding the treatment of UC. In this study, esters formed by binding of well-known prebiotics, fructooligosaccharides (FOS), with short chain fatty acids (SCFAs) via both enzymatic and chemical methods were evaluated for their impact on the gut microbiota of UC patients. An in vitro human colonic fermentation model was employed to monitor changes in total carbohydrates and SCFAs production during the fermentation of these esters by microbiota from patients with active and remission UC. The results showed that pronounced abundance of [Ruminococcus]_gnavus_group, Escherichia_Shigella, Lachnoclostridium, Klebsiella and other potential pathogens were detected in the fecal samples from UC patients, with a milder condition observed during the remission phase. Significant higher levels of corresponding SCFA were observed in the groups with addition of FOS-SCFAs esters during fermentation. Butyrylated fructooligosaccharides (B-FOS) and propionylated fructooligosaccharides (P-FOS) by enzymatic synthesis successfully promoted the proliferation of Bifidobacterium and inhibited Clostridium_sensu_stricto_1 and Klebsiella. Overall, B-FOS and P-FOS exhibit promising potential for restoring intestinal homeostasis and alleviating intestinal inflammation in individuals with UC.

Pro-pre and PostbioticFermentation of the Dietetic Dairy Matrix with Prebiotic Sugar Replacers.

In this study, bacterial growth, postbiotic short-chain fatty acids(SCFAs) formation,and gelation properties of sugar-free probiotic milk gels produced with stevia and inulin as a sugar replacer and synbiotic interactions were investigated with regard to prebiotic/bio-therapeutic potential and consumer preference. Lactobacillus acidophilus and Bifidobacterium animalis subsp. lactis cultures were used in the manufacture of dietetic milk gels. The addition of stevia and inulin promoted the viability of bacteria and enhanced milk gel firmness throughout its shelf life. The activity of the probiotic bacteria was identified to be within the potential prebiotic effects (> 8.30 log10 cfu mL-1) ina food matrix. However, it was determined that especially stevia and stevia + inulin addition increased the survival rate of probiotic bacteria and in vitro total SCFA production with higher scores for consumers' preferences rather than with the addition of stevia alone. Yoghurts containing B. animalis subsp. lactis have improved the instrumental textural properties, whereas yoghurts containing L. acidophilus had higher scores for sensorial attributes.

Bioaccessibility and bioactivities of phenolic compounds from microalgae during in vitro digestion and colonic fermentation.

Microalgae are a developing novel source of carbohydrates, phenolic compounds, carotenoids and proteins. In this study, in vitro digestion and colonic fermentation were conducted to examine the total phenolic content and potential antioxidant activity of four microalgal species (Chlorella sp., Spirulina sp., Dunaliella sp., and Isochrysis sp.). The bioaccessibility of targeted phenolic compounds and the short-chain fatty acid (SCFA) production were also estimated. Particularly, Spirulina sp. exhibited the highest total phenolic content (TPC) and free radical scavenging (2,2'-diphenyl-1-picrylhydrazyl, DPPH) capacity after gastrointestinal digestion of 7.93 mg gallic acid equivalents (GAE) per g and 2.35 mg Trolox equivalents (TE) per g. Meanwhile, it had the highest total flavonoid content (TFC) of 1.07 quercetin equivalents (QE) per g after 8 h of colonic fermentation. Dunaliella sp. and Isochrysis sp. showed comparable ferric reducing antioxidant power (FRAP) of 4.96 and 4.45 mg QE per g after 4 h of faecal reaction, respectively. p-hydroxybenzoic and caffeic acid almost completely decomposed after the intestine and fermented in the colon with the gut microflora. In Dunaliella sp. and Isochrysis sp., these phenolic acids were found in the colonic fermented residual, probably due to the presence of dietary fibre and the interactions with other components. All four species reached the highest values of SCFA production after 16 h, except Spirulina sp., which displayed the most increased total SCFA production after 8 h of fermentation. It is proposed that Spirulina sp. could be more beneficial to gut health.

Mangiferin alleviates trimethylamine-N-oxide (TMAO)-induced atherogenesis and modulates gut microbiota in mice.

Trimethylamine-N-oxide (TMAO), originating from dietary trimethylamine-containing nutrients such as choline, has been recognized as a risk factor for atherosclerosis. Mangiferin is a bioactive xanthone initially extracted from mango (Mangifera indica). The present study aimed to investigate the effect of mangiferin on TMAO-induced atherogenesis in mice fed a high-choline diet. Female ApoE-/- mice were randomly divided into three groups and fed either a control diet, a high-choline diet with 1% free choline, or an experimental diet with 1% free choline plus 0.5% mangiferin for 15 weeks. Our results showed that a high-choline diet elevated plasma TMAO levels, accelerated atherogenesis, promoted cholesterol accumulation, and reduced the generation of short-chain fatty acids (SCFAs) by gut microbes. Mangiferin alleviated inflammation, and lowered plasma total cholesterol levels by facilitating the elimination of neutral and acidic sterols in feces, resulting in a 16.7-29.0% reduction in aortic atherosclerotic lesions. Notably, mangiferin could favorably remodel the composition of the gut microbiota by fostering the growth of the beneficial taxa Akkermansia, Parabacteroides, and Bifidobacteriaceae, while reducing the relative abundance of the pathogenic genus Helicobacter. This modulation led to a decrease in plasma lipopolysaccharide levels, enhanced the production of total SCFAs by gut microbes, and reduced susceptibility to atherosclerosis. In conclusion, mangiferin exhibited its ability to alleviate TMAO-induced atherosclerosis through its anti-inflammatory, cholesterol-lowering, and gut microbial modulatory activities.

In Vitro Digestion and Colonic Fermentation of UHT Treated Faba Protein Emulsions: Effects of Enzymatic Hydrolysis and Thermal Processing on Proteins and Phenolics.

Faba bean (Vicia faba L.) protein is a new plant protein alternative source with high nutrient content especially protein and phenolic compounds. The present study investigated physicochemical properties, phenolic content, antioxidant potential, and short chain fatty acids (SCFAs) production during in vitro digestion and colonic fermentation of faba bean hydrolysates and oil-in-water (O/W) emulsions. Results indicate that the enzymic hydrolysates of faba proteins exhibited higher protein solubility, increased electronegativity, and decreased surface hydrophobicity than native faba protein. O/W emulsions showed improved colloidal stability for the faba protein hydrolysates after ultra-high temperature processing (UHT). Furthermore, UHT processing preserved total phenolic content, DPPH and ABTS radical scavenging abilities while decreasing total flavonoid content and ferric reducing power. Besides, the release of phenolic compounds in faba bean hydrolysates (FBH) and emulsions (FBE) improved after intestinal digestion by 0.44 mg GAE/g and 0.55 mg GAE/g, respectively. For colonic fermentation, FBH demonstrated an approximately 10 mg TE/g higher ABTS value than FBE (106.45 mg TE/g). Total SCFAs production of both FBH and FBE was only 0.03 mM. The treatment of FBH with 30 min enzymatic hydrolysis displayed relatively higher antioxidant capacities and SCFAs production, indicating its potential to bring more benefits to gut health. Overall, this study showed that enzymic hydrolysis of faba proteins not only improved the colloidal emulsion stability, but also released antioxidant capacity during in vitro digestibility and colonic fermentation. Colonic fermentation metabolites (SCFAs) were related to the degree of hydrolysis for both FBH and FBE. Additional studies are required to further elucidate and differentiate the role of phenolics during faba protein processing and digestion stages in comparison to contributions of peptides, amino acids and microelements to digestion rates, antioxidant capacities and colonial SCFA production.

In Vitro Assessment of the Prebiotic Potential of Xylooligosaccharides from Barley Straw.

Barley straw was subjected to hydrothermal pretreatment (steam explosion) processing to evaluate its potential as a raw material to produce xylooligosaccharides (XOS) suitable for use as a prebiotic. The steam explosion pretreatment generated a liquid fraction containing solubilised hemicellulose. This fraction was purified using gel permeation chromatography to obtain a fraction rich in XOS DP2-DP6. The sample was characterised through analytical techniques such as HPAEC-PAD, FTIR and MALDI-TOF-MS. The prebiotic activity was evaluated using in vitro fermentation in human faecal cultures through the quantification of short-chain fatty acid (SCFA) and lactate production, the evolution of the pH and the consumption of carbon sources. The total SCFA production at the end of fermentation (30 h) was 90.1 mM. Positive significant differences between the amount of XOS from barley straw and fructooligosaccharides after incubation were observed.

Study on the interaction between β-carotene and gut microflora using an in vitro fermentation model

β-Carotene, a typical non-oxygenated carotenoid, is the most efficient source of retinol (VA). The low bio-availability of β-carotene lead to large accumulation in colon; however, the relationship between β-carotene and gut microflora remains unclear. This study intends to explore the interaction between β-carotene and gut microflora using an in vitro fermentation model. After 24 h fermentation, the degradation rate of β-carotene was (64.28 ± 6.23)%, which was 1.46 times that of the group without gut microflora. Meanwhile, the production of VA was nearly 2 times that of the group without gut microflora, indicating that the gut microflora can metabolize β-carotene into VA. β-Carotene also influences the production of short-chain fatty acids (SCFAs), the production of total SCFAs in 0.5 mg/mL β-carotene (BCM) group was (44.00 ± 1.16) mmol/L, which was 2.26 times that of the blank control (BLK) group. Among them, the production of acetic acid in BCM group was (19.06 ± 0.82) mmol/L, which was 2.64 time that of the BLK group. Furthermore, β-carotene significantly affected the structure and composition of gut microflora, increasing the abundance of Roseburia, Parasutterella and Lachnospiraceae, and decreasing the abundance of Dialister, Collinsella and Enterobacter (P < 0.05). This study provides a new way to understand how β-carotene works in human body with gut microflora.

Flavones interact with fiber to affect fecal bacterial communities in vitro

This study investigated the effects of the sugarcane flavones diosmin, diosmetin, luteolin, and tricin, and their interactions with sugarcane fiber on the modulation of gut microbiota using in vitro batch fermentation. The alteration of fecal fermentation bacterial profile was analyzed using 16S rRNA sequencing data, while the bioavailability of fiber was indicated by short-chain fatty acid (SCFA) production and metabolism of polyphenols was measured directly by phenolic metabolites. Application of diosmin, diosmetin, luteolin, and tricin without fiber had no significant effect on the overall microbiota profile after 24 h of fermentation. When fiber alone was added, total SCFA production increased, specifically that of propionic and valeric acids. However, when flavones were combined with fiber, synergistic effects on the modulation of relative abundances of different bacterial taxa was noted. In particular, the proportion of Prevotella spp. was significantly increased by the combinations of diosmin, luteolin, and tricin with fiber.

In Vitro Colonic Fermentation Profiles and Microbial Responses of Cellulose Derivatives with Different Colloidal States.

Although cellulose derivatives are widely applied in the food industry, the effects of their structural properties on colonic health is unknown. Here, four types of cellulose derivatives, including microcrystalline cellulose (MCC), TEMPO-oxidized nanofibrillated cellulose (TOCNF), TEMPO-oxidized nanocrystalline cellulose (TOCNC), and carboxymethyl cellulose (CMC) were selected to investigate their in vitro fermentation profiles. TOCNF exhibited the highest production of total short-chain fatty acids (SCFAs), followed by TOCNC. The results suggested that reduced particle size and increased aspect ratio improved the fermentability of insoluble cellulose derivatives. MCC and CMC were barely fermented with similar total SCFAs production as the blank. 16S rRNA sequencing revealed that the fermentation of cellulose derivatives resulted in divergent microbial community structures. Moreover, Bacteroides cellulosilyticus showed high specificity to utilize TOCNF and TOCNC. The findings demonstrated that the colloidal states of cellulose derivatives, such as size and solubility, were important factors governing microbial community composition and metabolites.

Assessment of the bioaccessibility of phenolics from Australian grown lettuces by in vitro simulated gastrointestinal digestion and colonic fermentation

Lettuce varieties are rich in phytochemicals with strong antioxidant properties, mainly due to the presence of many phenolic compounds. The health effects of these phenolic compounds are dependent on their bioaccessibility, bioavailability and bioactivities. This study aimed to evaluate the differences in the content and bioaccessibility of phenolic compounds in green lettuce (iceberg, baby cos, and green little gem) and red lettuce (red oak leaf, lollo rosso, and red little gem), their antioxidant properties and short chain fatty acids (SCFAs) production after in vitro gastrointestinal digestion and colonic fermentation. Lollo rosso showed the highest content of phenolic compounds (1.23 mg GAE/g) and flavonoids (0.36 mg QE/g), as well as the antioxidant capacities after gastrointestinal digestion. After colonic fermentation, only baby cos and red little gem showed significantly higher TPC (5.13 mg GAE/g) and TFC (1.95 mg GAE/g), respectively. Most phenolic compounds in lettuce were mostly bioaccessible after gastric and small intestinal digestion, except for the kaempferol which relied more on the colonic fermentation. However, high percentage of kaempferol still could be found in the colonic fermented residual, probably due to the presence of dietary fiber and the interactions with other components. All lettuce varieties achieved the peak production of SCFAs after 16 h, except iceberg which displayed the highest total SCFAs production just after 8 h fermentation. It could be inferred that the iceberg (green lettuce) could be more beneficial to gut health while lollo rosso (red lettuce) could contribute more antioxidant properties after consumption.

Starchy and fibrous feedstuffs differ in their in vitro digestibility and fermentation characteristics and differently modulate gut microbiota of swine

BackgroundAlternative feedstuffs may contribute to reducing feed costs of pig production. But these feedstuffs are typically rich in fiber and resistant starch (RS). Dietary fibers and RS are fermented in the gastrointestinal tract (GIT) and modulate the microbial community. Certain microbes in the GIT can promote host health, depending on the type of fermentation substrates available. In this study, six alternative feedstuffs (three starchy: Okinawan sweet potato, OSP; yam, and taro, and three fibrous: wheat millrun, WMR; barley brewers grain, BBG; and macadamia nut cake, MNC) were evaluated for their in vitro digestibility and fermentation characteristics and their effects on pig’s hindgut microbial profile. After 2 steps of enzymatic digestion assay, residues were fermented using fresh pig feces as microbial inoculum, and gas production was recorded periodically for 72 h and modeled for fermentation kinetics. After fermentation, the residual liquid phase was analyzed for short-chain fatty acid (SCFA), and the solid phase was used to determine the nutrient’s digestibility and microbial community.ResultsIn vitro ileal digestibility of dry matter and gross energy was higher in starchy than fibrous feedstuffs. Total gas and SCFA production were significantly higher (P < 0.001) in starchy feedstuffs than fibrous feedstuffs. Both acetate and propionate production was significantly higher (P < 0.001) in all starchy feedstuffs than BBG and MNC; WMR was in between. Overall alpha diversity was not significantly different within and between starchy and fibrous feedstuffs. Beta diversity (measured using bray Curtis dissimilarity distance) of starchy feedstuffs was significantly different (P < 0.005) than fibrous feedstuffs.ConclusionStarchy feedstuffs acted as a substrate to similar types of microbes, whereas fibrous feedstuffs resulted in a more diverse microbial population. Such alternative feedstuffs may exert comparable beneficial effects, thus may be included in swine diets to improve gut health.

Hermetia illucens fat affects the gastrointestinal tract selected microbial populations, their activity, and the immune status of broiler chickens

Abstract The present study investigated the effect of Hermetia illucens larvae (BSFL) fat, derived using supercritical CO2 extraction and added to broiler chickens’ diets as a partial (50%) or total replacement for commonly used soybean oil, on the gastrointestinal tract (GIT) microbial population, its activity, and selected physiological and immune traits. A total of 576 one-day-old female Ross 308 chicks were randomly assigned to 3 dietary treatments with 16 replicates each. The following treatments were applied: SO – 100% soybean oil, BSFL50 – a mixture of BSFL and soybean oils in a 50:50 ratio, and BSFL100 – 100% BSFL fat. Digesta samples from the crop, jejunum and caeca were collected for further analyses, i.e., pH measurements, fluorescent in situ hybridization, and short-chain fatty acid (SCFA) concentrations. Additionally, the selected plasma biochemical parameters and immunological traits were assessed. In general, the implementation of BSFL fat in broilers’ diets resulted in increased proliferation of potentially pathogenic bacterial populations in the crop, such as Enterobacteriaceae, Bacteroides–Prevotella cluster, and Clostridium perfringens. Furthermore, BSFL100 enhanced microbial activity via total SCFA production and lowered the pH in this segment. However, no detrimental effects were observed in terms of other GIT segments, i.e., the jejunal and cecal microecosystems. The strongest impact on reduction of select components of the microbial population in the cecum was observed with the BSFL50 treatment for potentially pathogenic bacteria such as Enterobacteriaceae, Bacteroides–Prevotella cluster, while commensal populations were also limited, i.e., Bacillus spp., C. leptum subgroup, and C. coccoides–Eubacterium rectale cluster. Additionally, BSFL100 reduced the cholesterol concentration in the blood, while both experimental treatments decreased the ALT level. In conclusion, due to the insufficient release of lauric acid from the BSFL fat in the crop, an adverse shift in the microbiota can be noted. However, a positive suppressive effect on the select components of the cecal microbiota, as well as improvement of liver health suggests implying the BSFL fat in broiler nutrition.

Bioaccessibility and bioactivities of phenolic compounds from roasted coffee beans during in vitro digestion and colonic fermentation

Bioaccessibility and bioactivity of phenolic compounds in coffee beans relate to roasting and digestion process. This study aimed to estimate phenolic content, antioxidant potential, bioaccessibility, and changes in short chain fatty acids (SCFAs) production during in vitro digestion and colonic fermentation of commercial roasted (light, medium and dark) coffee beans. There was no significant difference found among all three different roasting levels. TPC and DPPH were enhanced 15 mg GAE/g and 60 mg TE/g during gastrointestinal digestion, respectively. For colonic fermentation, the highest TPC and FRAP of all coffee beans was found at 2 and 4 h, respectively. The gastric bioaccessibility of most of the phenolic compounds were relatively higher due to thermal phenolic degradation. Total SCFAs production was only up to 0.02 mM because of thermal polysaccharide decomposition. Light roasted beans exhibited relatively higher phenolic bioaccessibility, antioxidant activities and SCFAs production, which would be more beneficial to gut health.

In vitro fecal fermentation characteristics of bamboo insoluble dietary fiber and its impacts on human gut microbiota

Bamboo contains abundant hemicellulose, cellulose, and lignin, which are a high-quality insoluble dietary fiber (IDF) raw material. To investigate IDF- induced changes in the production of short-chain fatty acids (SCFAs) and the composition of human gut microbiota, IDF was extracted by alkaline hydrogen peroxide (named BIDF), complex enzymatic hydrolysis method (named OIDF) from bamboo, and commercial bamboo fiber BF90. The in vitro fecal fermentation characteristics of BIDF, OIDF, BF90 and its impacts on human gut microbiota were studied for the first time. Results showed that BIDF, OIDF, and BF90 could promote the production of total SCFAs after 24 h fermentation. Additionally, BIDF could alter the composition and microbial diversity of gut microbiota, especially increase the relative abundance of Bacteroides and decrease the ratio of Firmicutes to Bacteroidetes (F/B) value. These results revealed that the IDF from bamboo could be partially utilized by specific bacteria in human intestines and provide a reference for the study of the effects of IDF fermentation on SCFAs production and microbial composition.

Sugarcane polyphenol and fiber to affect production of short-chain fatty acids and microbiota composition using in vitro digestion and pig faecal fermentation model

This study aimed to examine the effects of sugarcane polyphenol and fiber (Phytolin + Fiber) on gut microbiota, short-chain fatty acids (SCFAs) production and phenolic metabolites production using in vitro digestion and fermentation model. Microbial profiling by 16S rRNA sequencing was used to analyze the pig faecal microbiota profile. SCFAs were identified and quantified by GC-FID, and phenolic metabolites were characterized by LC-ESI-QTOF-MS/MS. The results showed that Phytolin + Fiber exert synergistic effects on the pig gut microbiota by increasing the relative abundances of Lactobacillus and Catenibacterium, and decreasing the relative abundances of Mogibacterium, Dialister, and Escherichia-Shigella. Phytolin + Fiber also significantly increased the total SCFAs production, particularly the propionic and butyric acids. Production of phenolic metabolites related to major polyphenols in Phytolin were tentatively identified. These results suggest that Phytolin + Fiber could be beneficial to human colon health given the similarities between pig and human intestine in terms of physiology and microbiome.
