솔잎에 포함되어 있는 flavonoid들은 free radical 소거능이 탁월하다고 알려져 있다. 이들 중 특히 proanthocyanidin은 심장혈관 보호작용과 항산화 활성에 크게 기여하고 있다. 지리산 적송잎으로부터 열수, 에탄올, 헥산 및 아임계 장치를 이용하여 추출물을 제조하여 총 폴리페놀성 물질 중에 포함된 proanthocyanidin의 함량을 측정하였다. 그 결과, 총 폴리페놀성 물질 및 proanthocyanidin의 농도가 열수 추출물의 경우 가장 높게 나타났으며, 에탄을 추출물, 헥산 추출물, 아임계 추출물 순으로 낮아졌다. 한편, 각 추출물들의 총 폴리페놀성 물질의 농도에 대한 proanthocyanidin의 함량을 계산한 결과, 아임계 추출물의 경우 가장 높은 함량을 보였고, 헥산 추출물, 에탄을 추출물, 열수 추출물 순으로 낮아졌다. 이러한 결과로 적송잎을 열수 및 에탄올로 추출한 경우 높은 농도의 총 폴리페놀성 물질 및 proanthocyanidin이 포함된 추출물을 얻을 수 있다는 사실을 알 수 있었다. 또한, 아임계 추출물의 경우 비록 추출물에 포함된 총 폴리페놀성 물질의 농도는 낮지만 이들 대부분이 proanthocyanidin으로 이루어진 시료를 얻을 수 있다는 사실을 알 수 있었다 각 추출물에 대한 항산화 활성을 측정한 결과, 열수 추출물의 경우 가장 높은 활성을 보였으며 에탄올 추출물도 비교적 높은 활성을 타나냈다. 이상의 결과에서 솔잎으로부터 4가지 추출법에 의해 제조된 추출물을 비교 분석한 결과, 열수 추출물에서 가장 높은 기능성 성분 함량 및 항산화 활성을 나타냈으므로 솔잎을 이용한 기능성 식품의 추출 가공법 선택에 있어 좋은 자료로 활용될 수 있을 것으로 생각된다. Flavonoids in pine needles are known to be effective scavengers of free radical. Especially, proanthocyanidin, a kind of flavonoids possesses cardiovascular protection and antioxidative activities. Here, we evaluated proanthocyanidin contents in total polyphenolic compounds of pine needle extracts prepared by using hot water, ethanol, hexane or sub-supercritical <TEX>$CO_2$</TEX>. Analyses of total polyphenolic compounds and proanthocyanidin in each extracts indicated that hot water extract contained the highest concentrations, but sub-supercritical extract contained the lowest concentrations. On the other hand, evaluation of proanthocyanidin contents in total polyphenolic compounds in each extracts showed that sub-supercritical extract possessed the highest content, but hot water extract possessed the lowest content. These results indicate that extracts containing high concentrations of both total polyphenolic compounds and proanthocyanidin could be obtained by using hot water or ethanol extractions. Furthermore, extract containing high content of proanthocyanidin out of total polyphenolic compounds could be achieved by using sub-supercritical extraction. Measurement of antioxidative activities of extracts showed that hot water extract possessed the highest activity. In this study, we prepared extracts from pine needles by four different methods and evaluated the antioxidative compounds in extracts that could be used for effective components of functional food products.
