Effects of lactose content in milk replacer on apparent digestibility, growth, liver mRNA expression, and blood parameters related to metabolism of dairy calves

Even with the same energy supply, differences in energy sources may affect calf growth and metabolism. In this study, we evaluated the effects of feeding 3 different milk replacers (MRs) with different lactose supplies under isoenergetic conditions on calf digestibility, growth, and metabolism-related markers. Fifteen Holstein bull calves were randomly assigned to one of 3 MR-feeding treatments: low-lactose (L:38%), medium-lactose (M:41%), or high-lactose (H:46%). After birth, calves were provided colostrum and treatment MRs were fed from 1 d of age and gradually increased to a maximum feeding rate at 20 d of age (L:1.16 kg/d, M:1.21 kg/d, H:1.26 kg/d DM) without feeding solid feeds during the experimental period. Blood samples were temporarily collected weekly to assess blood concentrations of metabolites and hormones. From 30 to 36 d of age, the calves were euthanized and liver samples were collected to determine growth-related mRNA expression. L calves showed a greater body length than H calves and the highest GHR mRNA expression. Plasma concentrations of total cholesterol, urea nitrogen, total protein, albumin, insulin, and insulin-growth factor-1 were not different, but plasma concentrations of triglycerides were higher in order H, M, and L. These results showed that the difference in lactose content in the MR affected calf metabolism, and the L-MR was suggested to be more likely to enhance growth into the peripheral tissues.

Assessment of Haematological, Biochemical and Hormonal Changes in Mehsana buffaloes with Postpartum True Anestrus following Herbal Heat Inducer Treatment

The present study was designed to explore the effects of a new Herbal heat inducer in Mehsana buffaloes with postpartum true anestrus and consecutive changes in Blood haematological, Biochemical and Hormonal profile. All the buffaloes were orally supplemented with Herbal heat inducer mixture on (Jantana powder-10gm) 1st, (Metrali powder-20gm) 2nd and (Hitali-powder-500mg) 5th day for augmentation of oestrus. Out of total treated animals 10 animals expressed the signs of estrus within 30 days. Post-treatment analysis of the overall blood packed cell volume percent in conceiving buffaloes had a significant (p≤0.05) elevate. Overall blood plasma total cholesterol concentration (mg/dl) was found to be significantly (p≤0.001) elevated in all treated and both conceiving and non-conceiving buffaloes, respectively. Overall blood plasma progesterone concentration (ng/ml) was found to be significantly (p≤0.0001) and (p≤0.001) elevated in all treated and both conceived and non-conceived buffaloes, which indicate the positive favourable effect of all the drugs on postpartum true anestrus condition. However, Overall blood plasma tri-iodothyronine concentration (ng/ml) was found to be significantly (p≤0.001) and (p≤0.01) elevated in all treated and all conceived buffaloes. Findings of the present study revealed that strategic use of Indigenous herbal heat inducer in feed mixture may resolve the postpartum true anestrus problems in buffaloes.

Effect of feeding sodium butyrate to beef female cows during pre- and post-partum period on concentrations of glucagon-like peptides in plasma and colostrum.

The objective of this study was to evaluate the effect of feeding beef cows with sodium butyrate during the late pregnancy and early post-partum periods on concentrations of glucagon-like peptide (GLP)-1 and 2 in plasma, colostrum, and transition milk. Twelve Japanese Black female cows were fed concentrate feed without (CON; n = 6) or with (BUTY; n = 6) sodium butyrate supplementation at 1.1% of dietary dry matter from -60 d relative to the expected parturition date to 4d after parturition. Plasma total cholesterol concentration was higher for the BUTY than for the CON (P = 0.04). In addition, plasma GLP-1 concentration was higher for the BUTY than for the CON at 3d after calving (P < 0.05). This study showed for the first time that GLP-1 is present in the colostrum of Japanese Black cows at higher concentrations as compared to in plasma (P < 0.01). On the other hand, no treatment effect was observed for concentrations of metabolite and hormone in colostrum and transition milk. In summary, feeding beef cows with sodium butyrate during the late gestation and early post-partum period likely increases plasma GLP-1 concentrations post-partum without affecting the components of colostrum and transition milk.

Differential Effects of Two Isocaloric Healthy Diets on Postprandial Lipid Responses in Individuals with Type 2 Diabetes.

High blood concentrations of triglycerides (TG) in the postprandial period have been shown to be more closely associated with the risk of cardiovascular disease (CVD) than fasting values in individuals with type 2 diabetes (T2D). Dietary changes are the primary determinants of postprandial lipid responses. We investigated the effects of an isocaloric multifactorial diet, rich in n-3 PUFA, MUFA, fiber, polyphenols, and vitamins, compared to an isocaloric diet, containing the same amount of MUFA, on the postprandial lipid response in T2D individuals. Following a randomized, controlled, parallel group design, 43 (25 male/18 female) T2D individuals were assigned to an isocaloric multifactorial (n = 21) or a MUFA-rich diet (n = 22). At the beginning and after the 8 weeks of dietary intervention, the concentrations of plasma triglycerides, total cholesterol, HDL cholesterol, and non-HDL cholesterol were detected at fasting and over a 4-h test meal with the same composition as the prescribed diet. The concentrations of fasting plasma triglycerides, total cholesterol, HDL cholesterol, and non-HDL cholesterol did not change after both diets. Compared with the MUFA diet, the 8-week multifactorial diet significantly lowered the postprandial response, which was evaluated as the incremental area under the curve (iAUC), of triglycerides by 33% (64 ± 68 vs. 96 ± 50 mmol/L·240 min, mean ± SD, respectively, p = 0.018), total cholesterol by 105% (-51 ± 33 vs. -25 ± 29, p = 0.013), and non-HDL cholesterol by 206% (-39 ± 33 vs. -13 ± 23, p = 0.013). In T2D individuals, a multifactorial diet, characterized by several beneficial components, improved the postprandial lipid response compared to a MUFA diet, generally considered a healthy diet being reduced in saturated fat, and probably contributed to the reduction of cardiovascular risk.

Influence of Dietary Cation-Anion Differences (DCAD) on Nutrient Intake, Milk Yield and Metabolic Parameters of Transition Buffaloes

The study was divided into 2 phases. During 1st phase, 20 pre-partum buffaloes were divided into two groups, given -100 (high negative; HN) and -50 (low negative; LN) mEq/kg of dry matter (DM) diets. In the second phase, +200 (low positive; LP) and +400 (high positive; HP) mEq/kg of DM diets were formulated for postpartum buffaloes. Buffaloes that received LN DCAD were divided into 2 groups (LN-LP and LN-HP), and those that received HN DCAD were also divided into 2 groups (HN-LP and HN-HP). In the first phase, the group fed the HN DCAD diet had significantly lesser DM, nutrient intake, and urine pH (p&lt;0.05) but higher ME intake and ADF digestibility. Also, higher serum phosphorus levels (p&lt;0.05), fewer cases of hypocalcemia and ketosis, and a relatively smaller decrease in body weight were reported with the HN group. In the second phase, DM, organic matter, and NDF intakes were higher (p&lt;0.05) in HN-HP and LN-LP groups, whereas ME intake and nutrient digestibility were higher in HN-LP and LN-HP (p&lt;0.05). Daily milk, solid not fat (SNF), fat, protein, lactose yields, and plasma total protein, urea, and cholesterol concentrations were the highest in the HN-HP group (p&lt;0.05), but the highest fat content and fat yield were recorded in the HN-LP group (p&lt;0.05). Better efficiency of milk production and the lowest cases of hypocalcemia and ketosis were observed for both groups. In conclusion, for transition buffaloes, the HN and LN DCAD (-100 and +200 mEq/kg of DM, respectively) diets showed a beneficial impact.

Effect of chronic apple consumption (Malus domestica Borkh.) on the lipid profile of adults with dyslipidemia: a systematic review

Aim: Cardiovascular disease (CVD) are among the main causes of death worldwide and dyslipidemias account for one of the risk factors for these diseases. Habitual apple consumption appears to be inversely associated with reduced cardiovascular risk. Then, this systematic review aims to investigate the effect of chronic apple consumption on the lipid profile of adults with dyslipidemia. Methods: A systematic search was performed in electronic databases, including PubMed, Embase, Web of Science and Scopus, without restriction of year of publication. Inclusion criteria were randomized clinical trials in humans that investigated the effect of chronic consumption of whole fresh or dried apple, for a period longer than two weeks of intervention on the lipid profile. Results: Based on the methodology used and following the pre-established search strategies, 4,468 articles were found. After applying the inclusion and exclusion criteria, five articles were selected for qualitative evaluation, covering 522 adult participants of both sexes. Three randomized controlled trials included in this review demonstrated that there was a decrease in plasma total cholesterol (TC), triglyceride and low-density lipoprotein cholesterol (LDL-c) concentrations, in addition to an increase in high-density lipoprotein cholesterol (HDL-c) concentration. Two other studies found different results. Low risk of bias was identified in three studies. Conclusions: The analysis of the studies indicates that the consumption of fresh and/or dried apples with the peel has a beneficial effect on the lipid profile of adults, with a decrease in TC and LDL-c. These effects may be related to polyphenols and soluble fibers, among other functional compounds present in this fruit.

Plasma lipids in Pseudoxanthoma Elasticum (PXE) patients: A comparative study with population-based reference values and Non-PXE controls

Background and aims– Pseudoxanthoma elasticum (PXE) is a rare genetic disease caused by pathogenic mutations in the ABCC6 gene, resulting in low values of inorganic pyrophosphate (PPi). While low PPi is thought to contribute to arterial calcification, it remains unclear whether this fully explains premature calcification in PXE. It has been hypothesized that the ABCC6 gene could be related to dyslipidemia, which could contribute to vascular calcification seen in PXE. The aim of this study is to evaluate the relation between PXE and plasma lipid concentrations in a large cohort of PXE patients compared with reference values for the general population and compared with non-PXE controls. Methods– The plasma concentrations of total cholesterol, HDL-cholesterol, tiglycerides, and LDL-cholesterol of 312 PXE patients were compared to age- and sex-matched modeled data of the general Dutch population. Differences in median lipid levels were compared with Mann-Whitney-U test. Secondly, plasma lipid concentrations of 44 PXE patients were compared to 44 not-genetically related relatives (spouses or friends), with linear models adjusted for age, sex and BMI. Results– Total cholesterol in PXE patients was 5.6 [IQR 4.6–6.4] mmol/L versus 5.3 [IQR 4.7–6.0] mmol/L (p < 0.01) in the general population; triglycerides were 1.1 [IQR 0.9–1.7] mmol/L versus 1.0 [0.7–1.4] mmol/L (p < 0.01); HDL-c was 1.4 [IQR 1.2–1.7] mmol/L versus 1.5 [IQR 1.2–1.8] mmol/L (p = 0.03) and LDL-c was 3.3 [IQR 2.7–4.1] mmol/L versus 3.2 [IQR 2.7–3.8] mmol/L (p = 0.01). In the patient control analysis with 44 pairs and age, sex and BMI adjusted, comparison with the non-PXE controls only triglycerides were significantly different (mean difference: 0.38 (0.13–0.63)). Conclusion–The lipid profiles of PXE patients are marginally different from the general population or compared to a matched control group, but the differences are unlikely to be clinically relevant. It is therefore unlikely that plasma lipids contribute to the premature vascular calcifications in PXE patients.

Phytosterol intake and risk of coronary artery disease: Results from 3 prospective cohort studies

BackgroundPhytosterols are structurally similar to cholesterol and partially inhibit intestinal absorption of cholesterol, although their impact on coronary artery disease (CAD) risk remains to be elucidated. ObjectivesThis study aimed to prospectively assess the associations between total and individual phytosterol intake and CAD risk in United States health professionals. MethodsThe analysis included 213,992 participants from 3 prospective cohorts—the Nurses’ Health Study (NHS), NHSII, and Health Professionals Follow-Up Study—without cardiovascular disease or cancer at baseline. Diet was assessed using a validated food frequency questionnaire every 2–4 y since baseline. Associations between phytosterol intake and the risk of CAD, such as nonfatal myocardial infarction and fatal CAD, were evaluated using Cox proportional hazards regression models. ResultsMore than 5,517,993 person-years, 8725 cases with CAD were documented. Comparing extreme quintiles, pooled hazard ratios (95% CIs) of CAD were 0.93 (0.86, 1.01; P-trend = 0.16) for total phytosterols, 0.89 (0.82, 0.96; P-trend = 0.05) for campesterol, 0.95 (0.88, 1.02; P-trend = 0.10) for stigmasterol, and 0.92 (0.85, 1.00; P-trend = 0.09) for β-sitosterol. Nonlinear associations were observed for total phytosterols, campesterol, and β-sitosterol: the risk reduction plateaued at intakes above ∼180, 30, and 130 mg/d, respectively (P-nonlinearity < 0.001). In a subset of participants (N range between 11,983 and 22,039), phytosterol intake was inversely associated with plasma concentrations of total cholesterol, triglycerides, high-density lipoprotein cholesterol, and IL-6 and positively associated with adiponectin, whereas no significant associations were observed for low-density lipoprotein cholesterol or C-reactive protein concentrations. ConclusionsHigher long-term intake of total and major subtypes of phytosterols may be associated with a modest reduction in CAD risk, displaying a nonlinear relationship that plateau at moderate intake levels. The role of phytosterols in preventing CAD warrants further investigation.

Flavonoids from mulberry leaves inhibit fat production and improve fatty acid distribution in adipose tissue in finishing pigs

This study evaluated the effects of flavonoids from mulberry leaves (FML) on plasma biochemical indices, serum activities of lipid metabolism-related enzymes, fat morphology, fatty acid composition, and lipid metabolism in different adipose tissues of finishing pigs. We used 120 Chinese hybrid barrows of Berkshire and Bama mini-pigs with an average initial body weight of 45.11 ± 4.23 kg. The pigs were randomly assigned to five treatment groups and fed a control diet based on corn, soybean meal, and wheat bran or a control diet supplemented with 0.02%, 0.04%, 0.08%, or 0.16% FML. Each experimental group had six replicates (pens), with four pigs per pen. After a 7-d adaptation period, the feeding trial was conducted for 58 d. Blood and adipose tissue samples were collected from 30 pigs (one pig per pen) at the end of the test. The results showed that FML supplementation significantly decreased the feed intake to body gain ratio, the plasma concentrations of total cholesterol and free fatty acids, and the serum activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase (linear or quadratic effects, P < 0.05), and decreased the plasma triglyceride concentration (quadratic, P = 0.07). Increasing FML supplementation increased the average daily gain and serum activities of lipoprotein lipase (linear and quadratic effects, P < 0.05) and adipose triglyceride lipase (linear, P < 0.05). Dietary FML supplementation decreased the adipocyte area in the dorsal subcutaneous adipose (DSA) tissue of finishing pigs (linear, P = 0.05) and increased the adipocyte area in the visceral adipose tissue (quadratic, P < 0.01). Increasing FML supplementation decreased the C20:1 content in DSA, abdominal subcutaneous adipose, and visceral adipose tissues of finishing pigs (P < 0.05) and increased the C18:3n3 and n-3 PUFA contents (P < 0.05). The lipid metabolism genes were regulated by the PPARγ-LXRα-ABCA1 signaling pathway, and their expressions differed in different adipose tissues. These findings suggest that FML improved growth performance, regulated lipid metabolism, inhibited fat production, and improved fatty acid distribution in the adipose tissue of finishing pigs, thereby improving pig fat's nutritional quality and health value.

Chocolate Byproducts and Protected Fats Enriched with Chromium to Replace Corn in Diets of Early Lactation Holstein Cows: Feed Utilization, Blood Profile, and Lactational Performance

Abstract The present experiment aimed to evaluate the partial replacement of corn with chocolate byproducts or protected fats in the diet of lactating cows on feed utilization, blood profile, and lactation performance. Fifty multiparous Holstein cows (560±22 kg BW, 3±1 parity, 7±1 days in milk, and previous milk production of 34±3 kg/d), were randomly assigned to 5 treatments in a completely randomized design for 90 days. The control diet contained a concentrate feed mixture containing (per kg DM) 412 g concentrate feed mixture, 412 g corn silage, and 176 g berseem hay. The control diet contained 17.6% crushed corn (DM basis). In another treatment, the control diet was supplemented with 1 g chromium daily (Chromium diet). In the other diets, each kg of crushed corn grain was replaced with 600 g chocolate byproducts (CH diet), 400 g protected fats (PF diet), or 500 g of chocolate byproducts and protected fats mixture (1:1 DM basis) (CHPF diet), DM basis, and each of the diets was also supplemented with 1 g chromium daily. Both amounts of protected fats and chocolate byproducts had the same energy concentration as 1 kg of corn. Without affecting feed intake, the replacement increased (P&lt;0.01) milk production, milk fat concentration, and feed efficiency. The replacement (P&lt;0.05) increased the concentrations of plasma total proteins, albumin, glucose, cholesterol, triglyceride, and chromium in comparison with the control diet. Higher (P&lt;0.001) nutrient digestibility was observed with CH, PF, and CHPF treatments. It is concluded that chromium supplementation did not affect feed utilization or cows’ performance; however, partial replacement of corn with chromium supplemented protected fat, chocolate byproducts or their mixture improved milk production and feed efficiency.

Alanyl-Glutamine Dipeptide Attenuates Non-Alcoholic Fatty Liver Disease Induced by a High-Fat Diet in Mice by Improving Gut Microbiota Dysbiosis.

Non-alcoholic fatty liver disease (NAFLD) manifests as a persistent liver ailment marked by the excessive buildup of lipids within the hepatic organ accompanied by inflammatory responses and oxidative stress. Alanyl-glutamine (AG), a dipeptide comprising alanine and glutamine, is commonly employed as a nutritional supplement in clinical settings. This research aims to evaluate the impact of AG on NAFLD triggered by a high-fat diet (HFD), while concurrently delving into the potential mechanisms underlying its effects. The results presented herein demonstrate a notable reduction in the elevated body weight, liver mass, and liver index induced by a HFD upon AG administration. These alterations coincide with the amelioration of liver injury and the attenuation of hepatic histological advancement. Furthermore, AG treatment manifests a discernible diminution in oil-red-O-stained regions and triglyceride (TG) levels within the liver. Noteworthy alterations encompass lowered plasma total cholesterol (TC) and low-density lipoprotein cholesterol (LDLC) concentrations, coupled with elevated high-density lipoprotein cholesterol (HDLC) concentrations. The mitigation of hepatic lipid accumulation resultant from AG administration is aligned with the downregulation of ACC1, SCD1, PPAR-γ, and CD36 expression, in conjunction with the upregulation of FXR and SHP expression. Concomitantly, AG administration leads to a reduction in the accumulation of F4/80-positive macrophages within the liver, likely attributable to the downregulated expression of MCP-1. Furthermore, AG treatment yields a decline in hepatic MDA levels and a concurrent increase in the activities of SOD and GPX. A pivotal observation underscores the effect of AG in rectifying the imbalance of gut microbiota in HFD-fed mice. Consequently, this study sheds light on the protective attributes of AG against HFD-induced NAFLD through the modulation of gut microbiota composition.

EVALUATION OF CARDIOVASCULAR DISEASE RISK FACTORS IN DIABETIC RATS ADMINISTERED ETHANOL EXTRACT OF Cucumis sativus FRUIT.

It is estimated that by the year 2030, the number of persons with diabetes mellitus (DM) would increase to 366 million. Diabetes mellitus is primarily defined by the level of hyperglycemia which causes micro- and macro-vascular damage. This study evaluated cardiovascular disease risk factors in diabetic rats administered ethanol extract of Cucumis sativus fruit. Male Wistar rats (n = 25, mean weight = 215 ± 15 g) were used. The rats were randomly assigned to five groups (5 rats per group): normal control, diabetic control, metformin, 200 mg/kg body weight (BWT) extract and 300 mg/kg BWT extract groups. Diabetes mellitus was induced via intraperitoneal injection of streptozotocin (STZ, 50 mg/kg BWT). The diabetic rats were then treated for 21 days with metformin (50 mg/kg BWT) or the extract at doses of 200 and 300 mg/kg BWT, respectively. The results showed that STZ-induced DM significantly increased plasma concentrations of total cholesterol (TC), triacylglycerol (TG), very low-density lipoprotein cholesterol (VLDL-C), low-density lipoprotein cholesterol (LDL-C), atherogenic index of plasma (AIP), atherogenic coefficient (AC) and cardiac risk ratio (CRR), but it significantly reduced high-density lipoprotein cholesterol (HDL-C) (p &lt; 0.05). However, treatment of diabetic rats with ethanol extract of C. sativus fruit led to significant reductions in circulating levels of lipid profile (except HDL-C, which increased) as well as AIP, AC and CRR (p &lt; 0.05). These results suggest that the medicinal plant extract has the capacity to protect against cardiovascular events in diabetic rats.

Plasma total cholesterol concentration and risk of higher-grade prostate cancer: A nested case-control study and a dose-response meta-analysis.

Our previous publication found an increased risk of higher-grade (Gleason sum ≥7) prostate cancer for men with high total cholesterol concentration (≥200 mg/dl) in the Health Professionals Follow-up Study (HPFS). With additional 568 prostate cancer cases, we are now able to investigate this association in more detail. For the nested case-control study, we included 1260 men newly diagnosed with prostate cancer between 1993 and 2004, and 1328 controls. For the meta-analyses, 23 articles studied the relationship between total cholesterol level and prostate cancer incidence were included. Logistic regression models and dose-response meta-analysis were performed. An increased risk of higher-grade (Gleason sum ≥4 + 3) prostate cancer for high vs low quartile of total cholesterol level was observed in the HPFS (ORmultivariable = 1.56; 95% CI = 1.01-2.40). This finding was compatible with the association noted in the meta-analysis of highest vs lowest group of total cholesterol level, which suggested a moderately increased risk of higher-grade prostate cancer (Pooled RR =1.21; 95%CI: 1.11-1.32). Moreover, the dose-response meta-analysis indicated that an increased risk of higher-grade prostate cancer occurred primarily at total cholesterol levels ≥200 mg/dl, where the RR was 1.04 (95%CI: 1.01-1.08) per 20 mg/dl increase in total cholesterol level. However, total cholesterol concentration was not associated with the risk of prostate cancer overall either in the HPFS or in the meta-analysis. Our primary finding, as well as the result of the meta-analysis suggested a modest increased risk of higher-grade prostate cancer, at total cholesterol concentrations exceeding 200 mg/dl.

Effect of Dietary Starch-to-Fat Ratio on Lipid Metabolism, Inflammation, and Microbiota of Multiparous Sow and Newborn Piglets

This experiment aimed to evaluate the effects of dietary starch-to-fat ratio on reproductive performance and lipid metabolism of sows and newborn piglets. A total of 75 Landrace × Yorkshire multiparous sows at d 84 of gestation were selected and randomly divided into three groups based on body weight. From d 85 of gestation to farrowing, sows were fed one of three dietary starch-to-fat ratios (20:1, 10:1, and 5:1). Dietary high starch-to-fat ratio increased the birth weight of piglets (p &lt; 0.05). The apparent total digestibility of dry matter, organic matter, and gross energy of sows was improved by an increasing starch-to-fat ratio during gestation (p &lt; 0.05). Decreased dietary starch-to-fat ratio increased the concentration of plasma triglycerides, total cholesterol, and GSH-Px in sows (p &lt; 0.05). During parturition, sows had increased plasma interleukin (IL) -1β, IL-6, and tumor necrosis factor α in the low ratio group (p &lt; 0.05). The relative abundance of Streptococcaceae in the low ratio group was significantly higher (p &lt; 0.05). The medium dietary starch-to-fat ratio significantly increased the concentrations of short chain fatty acids. In conclusion, this study suggested that for sows a diet with ahigh starch to fat ratio could ameliorate lipid metabolism disorder and maternal inflammation during late gestation.

Supranutrition of microalgal docosahexaenoic acid and calcidiol improved growth performance, tissue lipid profiles, and tibia characteristics of broiler chickens

BackgroundDocosahexaenoic acid (DHA) and calcidiol could be enriched in chicken for improving public nutrition and health. It remains unclear if supranutritional levels of DHA and calcidiol impair growth performance or metabolism of broiler chickens. This study was to determine singular and combined effects of high levels of supplemental DHA-rich microalgal biomass or oil and calcidiol on growth performance, concentrations of triglycerides, cholesterol, and nonesterfied fatty acids in plasma, liver, breast, and thigh, and biophysical properties of tibia.MethodsIn Exp. 1, 144 day-old Cornish chicks were divided into 4 groups (6 cages/treatment, 6 birds/cage), and were fed a corn-soybean meal basal diet (BD), BD + 10,000 IU calcidiol/kg (BD + Cal), BD + 1% DHA-rich Aurantiochytrium (1.2 g DHA/kg; BD + DHA), and BD + Cal + DHA for 6 weeks. In Exp. 2, 180 day-old chicks were divided into 5 groups, and were fed: BD, BD + DHA (0.33% to 0.66% oil, 1.5 to 3.0 g DHA/kg), BD + DHA + EPA (1.9% to 3.8% eicosapentaenoic acid-rich Nannochloropsis sp. CO18, 0.3 to 0.6 g EPA/kg), BD + DHA + calcidiol (6000 to 12,000 IU/kg diet), and BD + DHA + EPA + Cal for 6 weeks.ResultsBirds fed BD + Cal diet in Exp. 1 and BD + DHA + EPA diet in Exp. 2 had higher (P < 0.05) body weight gain (10%–11%) and gain:feed ratio (7%), and lower (P < 0.05) total cholesterol and triglyceride concentrations in plasma (18%–54%), liver (8%–26%), breast (19%–26%), and thigh (10%–19%), respectively, over the controls. The two diets also improved (P < 0.05) tibial breaking strength (8%–24%), total bone volume (2%–13%), and (or) bone mineral density (3%–19%) of chickens.ConclusionSupranutrition of dietary calcidiol and DHA alone or together did not produce adverse effects, but led to moderate improvements of growth performance, lipid profiles of plasma and muscle, and bone properties of broiler chickens.

Epigenome-wide association study of plasma lipids in West Africans: the RODAM study.

DNA-methylation has been associated with plasma lipid concentration in populations of diverse ethnic backgrounds, but epigenome-wide association studies (EWAS) in West-Africans are lacking. The aim of this study was to identify DNA-methylation loci associated with plasma lipids in Ghanaians. We conducted an EWAS using Illumina 450k DNA-methylation array profiles of extracted DNA from 663 Ghanaian participants. Differentially methylated positions (DMPs) were examined for association with plasma total cholesterol (TC), LDL-cholesterol, HDL-cholesterol, and triglycerides concentrations using linear regression models adjusted for age, sex, body mass index, diabetes mellitus, and technical covariates. Findings were replicated in independent cohorts of different ethnicities. We identified one significantly associated DMP with triglycerides (cg19693031 annotated to TXNIP, regression coefficient beta-0.26, false discovery rate adjusted p-value 0.001), which replicated in-silico in South African Batswana, African American, and European populations. From the top five DMPs with the lowest nominal p-values, two additional DMPs for triglycerides (CPT1A, ABCG1), two DMPs for LDL-cholesterol (EPSTI1, cg13781819), and one for TC (TXNIP) replicated. With the exception of EPSTI1, these loci are involved in lipid transport/metabolism or are known GWAS-associated loci. The top 5 DMPs per lipid trait explained 9.5% in the variance of TC, 8.3% in LDL-cholesterol, 6.1% in HDL-cholesterol, and 11.0% in triglycerides. The top DMPs identified in this study are in loci that play a role in lipid metabolism across populations, including West-Africans. Future studies including larger sample size, longitudinal study design and translational research is needed to increase our understanding on the epigenetic regulation of lipid metabolism among West-African populations. Effect of Modified-Hen-Egg-Yolk-Lipid-Composition Diet on Plasma Lipids in Rats

Abstract This study aimed to investigate the effects of dietary supplementation with modified-hen-egg-yolk on plasma lipids and lipoprotein profiles in rats. During the four-week-experiment, 64 Wistar rats were divided into four groups of 16 (eight of both sexes), and fed commercial rat food (group C); food containing 70% commercial rat mixture and 30% freshly cooked egg yolk originating from laying hen eggs fed with 3% fish oil (group F); 3% palm olein (group P), or 3% lard (group L). The cooked egg yolk in the rat diet affected the concentrations of plasma total and LDL-cholesterol in males of the P and L groups. Cholesterol and total fat in the diet did not have a hypercholesterolemic effect on their own, but when in combination with fatty acid composition, they could contribute to an increase in plasma total and LDL cholesterol concentrations in rats. HDL-cholesterol was the most resilient plasma lipoprotein of rats to dietary treatments in our experiment. Compared to the control group, the addition of hen egg yolk to the rat diet regardless of its quality, adversely affected the values of HDL-C/TC and HDL-C/LDL-C in both males and females.

Angiopoietin-like proteins inhibitors: New horizons in the treatment of atherogenic dyslipidemia and familial hypercholesterolemia.

Angiopoietin-like proteins (ANGPTL) are involved in the regulation of numerous physiological and biochemical processes. ANGPTL3, 4 and 8, which are involved in the regulation of lipoprotein metabolism, are particularly important. ANGPTL3, 4 and 8 have been shown to regulate triglyceride availability depending on the nutritional status of the body. In addition, a deficiency of these proteins has been found to cause hypolipidemia (reduction of all lipid fractions). Increases in ANGPTL3, 4 and 8 appear to be associated with cardiovascular risk. Animal studies indicate that the use of ANGPTL3 (evinacumab) inhibitors significantly reduces plasma total cholesterol, triglycerides and low-density lipoprotein concentrations. The use of evinacumab in clinical trials also led to the normalization of plasma lipid concentrations in patients with atherogenic dyslipidemia and homozygous familial hypercholesterolemia. The results of these studies indicate that evinacumab may in the future be used in the treatment of lipid disorders, especially those with hypertriglyceridemia.

Apolipoprotein mRNA in Liver and Intestine of Rats is Affected by Dietary Beet Fiber or Cholestyramine

Rats were fed a cholesterol-free diet with no added fiber (fiber-free) or with 15 g/100 g beet fiber or 5 g/100 g cholestyramine for 14 d. Final plasma total cholesterol concentrations were significantly lower in rats fed beet fiber than in those fed fiber-free or cholestyramine diets. This difference was due mainly to lower HDL cholesterol concentrations. The group fed beet fiber also tended (P < 0.1) to have lower apolipoprotein A-I concentration in plasma. Northern blot analysis revealed that the relative concentrations of jejunal apolipoprotein A-I and A-IV mRNA were the same in all groups, whereas ileal apolipoprotein A-I and A-IV mRNA levels were significantly lower in rats fed beet fiber or cholestyramine than in those fed the fiber-free diet. Hepatic apolipoprotein E mRNA concentrations were the same in all groups, but apolipoprotein A-I mRNA levels were significantly lower in rats fed beet fiber than in those fed the other diets. Apolipoprotein A-IV mRNA tended (P < 0.1) to be lower in rats fed the beet fiber diet. These data suggest that the hypocholesterolemic effect of dietary beet fiber is associated with diminished expression of the hepatic apolipoprotein A-I gene.

Age-and sex-specific reference intervals of total cholesterol, LDL cholesterol, HDL cholesterol and non-HDL cholesterol. Comparison of two algorithms for the indirect estimation of reference intervals

Abstract Objectives Reference intervals of total cholesterol concentrations in plasma and of their fractions low-density lipoprotein (LDL)-, high-density lipoprotein (HDL)-and non-HDL concentrations are seldom studied with respect to the relevance of age and sex. Therefore, the effect of age and sex on the reference intervals was reinvestigated with 2 indirect procedures. Methods As an indirect approach, the truncated minimum chi-square method was applied. All analyses were performed by computer programs available. The script published on the homepage of the German Society of Clinical Chemistry and Laboratory Medicine (DGKL) allows to derive a continuous age dependency of reference intervals together with their confidence and equivalence limits. The results of this approach were compared with those obtained by an indirect method developed more recently, the refineR algorithm. Results In the present study, the upper reference limits of total cholesterol varied from 5.1 to 7.8 mmol/L (197–302 mg/dL) depending on various biological variables (as age, sex, inpatients versus outpatients). These upper limits increased with age. Differences between sexes can be neglected except for the age above 80 years. The pattern of reference limits of LDL cholesterol and non-HDL cholesterol paralleled those of total cholesterol. The reference limits of HDL cholesterol were higher in women than in men but were independent of age. Conclusions Reference limits for the concentrations of total cholesterol and their fractions LDL-, HDL-and non-HDL concentrations should be stratified for age and sex.
