Evaluation of pathogen spread risk from excavated landfill

Landfill is a huge pathogen reservoir and needs special attention. Herein, the distribution and spread risk of pathogen were assessed in excavated landfill scenario. The results show that landfill excavation will greatly increase the risk of environmental microbial contamination. The highest total concentration of culturable bacteria among landfill refuse, topsoil and plant leaves was found to be as high as 1010 CFU g−1. Total coliforms, Hemolytic bacteria, Staphylococcus aureus, Salmonella, Enterococci, and Fecal coliforms were detected in the landfill surrounding environment. Notably, pathogens were more likely to adhere to plant leaves, making it an important source of secondary pathogens. The culturable bacteria concentration in the air samples differed with the landfill zone with different operation status, and the highest culturable bacteria concentration was found in the excavated area of the landfill (3.3 × 104 CFU m−3), which was the main source of bioaerosol release. The distribution of bioaerosols in the downwind outside of the landfill showed a tendency of increasing and then decreasing, and the highest concentration of bioaerosols outside of the landfill (6.56 × 104 CFU m−3) was significantly higher than that in the excavated area of the landfill. The risk of respiratory inhalation was the main pathway leading to infection, whereas the HQin (population inhalation hazardous quotient) at 500 m downwind the excavation landfill was still higher than 1, indicating that the neighboring residents were exposed to airborne microbial pollutants. The results of the study provide evidence for bioaerosols control protective measures taken to reduce health risk from the excavated landfill.

MBR and GAC filtration followed by UV disinfection – implications for wastewater reuse at full scale

Abstract Influences of upstream wastewater treatment on the process combination of granular activated carbon (GAC) and ultraviolet (UV) disinfection were studied and the implications of this for wastewater reuse were assessed. GAC is an efficient chemical barrier but contributes little to the removal of indicator bacteria, and generally increases total bacteria concentrations, necessitating disinfection with UV radiation, for example, to ensure the safe reuse of wastewater. The efficiency of UV disinfection is impacted by factors such as particle concentration and UV absorbance of the water and is thus affected by upstream treatment processes. A full-scale wastewater treatment plant with a membrane bioreactor (MBR) followed by GAC filtration was compared to a treatment plant with a conventional activated sludge process and sand filtration, followed by GAC filtration. The removal of indicator bacteria was higher by the GAC filter that was preceded by an MBR. A UV fluence of 400 J/m2 was sufficient to reach irrigation water quality for both process combinations and to meet the criteria for microbial drinking water quality in the MBR + GAC effluent. One sample was selected for chemical analysis, comprising approximately 100 parameters, demonstrating that the MBR + GAC + UV (400 J/m2) effluent met all drinking water criteria except for nitrate levels.

Effect of Yogurt Acid Whey on the Quality of Maize Silage

The increasing popularity of Greek yogurt generates large amounts of acid whey worldwide. The use of yogurt acid whey in animal nutrition is limited. The aim of this study was to determine the effect of a yogurt acid whey powder (YAWP) addition to maize forage prior to ensiling on the nutritional, microbial and fermentation quality of maize silage. Depending on the addition level of the YAWP to maize forage, there were the following four experimental treatments: YAWP 0, 2.5, 5 and 10% w/w. An increasing YAWP inclusion level linearly increased the maize silage dry matter, crude protein and ash concentrations, whereas it reduced the crude fiber, neutral-detergent fiber and acid-detergent fiber concentrations. The silage pH decreased quadratically with the increasing YAWP level, with the lower plateau noted for the YAWP 5% addition. Concentrations of total bacteria in the silage and Lactobacillus spp. decreased linearly with the YAWP increase. The silage acetic acid content decreased linearly, whereas propionic acid, lactic acid and the ratio of lactic to acetic acid increased linearly with the increasing YAWP level. The ammonia-N content decreased linearly with the increasing YAWP level. In conclusion, the incorporation of the 5 and 10% YAWP addition in silage preparation improved the nutritional and fermentative quality of the produced silage.

Investigating Structural Defects in Extra Hard Cheese Produced from Low-Temperature Centrifugation of Milk.

The present study investigated some physico-chemical and microbiological traits of 20-month ripened hard cheeses produced from low-temperature high-speed centrifuged raw milk that developed a structural defect consisting of eyes or slits in the paste. Cheeses obtained using the same process and that did not develop the defect were used as controls. The colour, texture, moisture, water activity, proton molecular mobility, microstructure, extent of proteolysis, and viable microorganisms have been evaluated in all the cheese samples, and the significant differences between the defective and non-defective cheeses have been critically discussed. At a microstructural level, the defects caused fat coalescence and an unevenly organised protein matrix with small cracks in the proximity of the openings. The different fat organisation was correlated to a different transverse relaxation time of 1H population relaxing at higher times. The textural and colour features were not different from those of the control cheeses and were comparable with those reported in the literature for other long-ripened hard cheeses. On the other hand, the defective cheeses showed a higher moisture level and lower lactobacilli and total mesophilic bacteria concentrations, but the microbial origin of the defect remains an open hypothesis that deserves further investigation.

Assessment of indoor bioaerosol exposure using direct-reading versus traditional methods—potential application to home health care

Home healthcare workers (HHCWs) can be occupationally exposed to bioaerosols in their clients’ homes. However, choosing the appropriate method to measure bioaerosol exposures remains a challenge. Therefore, a systematic comparison of existing measurement approaches is essential. Bioaerosol measurements with a real-time, fluorescence-based Wideband Integrated Bioaerosol Sensor (WIBS) were compared to measurements with four traditional off-line methods (TOLMs). The TOLMS included optical microscopic counting of spore trap samples, microbial cultivation of impactor samples, qPCR, and next-generation sequencing (NGS) of filter samples. Measurements were conducted in an occupied apartment simulating the environments that HHCWs could encounter in their patients’ homes. Descriptive statistics and Spearman’s correlation test were computed to compare the real-time measurement with those of each TOLM. The results showed that the geometric mean number concentrations of the total fluorescent aerosol particles (TFAPs) detected with the WIBS were several orders of magnitude higher than those of total fungi or bacteria measured with the TOLMs. Among the TOLMs, concentrations obtained with qPCR and NGS were the closest to the WIBS detections. Correlations between the results obtained with the WIBS and TOLMs were not consistent. No correlation was found between the concentrations of fungi detected using microscopic counting and any of the WIBS fluorescent aerosol particle (FAP) types, either indoors or outdoors. In contrast, the total concentrations detected with microbial cultivation correlated with the WIBS TFAP results, both indoors and outdoors. Outdoors, the total concentration of culturable bacteria correlated with FAP-type AC. In addition, fungal and bacterial concentrations obtained with qPCR correlated with FAP types AB and AC. For a continuous, high-time resolution but broad scope, the real-time WIBS could be considered, whereas a TOLM would be the best choice for specific and more accurate microbial characterization. HHCWs’ activities tend to re-aerosolize bioaerosols causing wide temporal variation in bioparticle concentrations. Thus, the advantage of using the real-time instrument is to capture those variations. This study lays a foundation for future exposure assessment studies targeting HHCWs.

Identification Coliform Bacteria In Student Drinking Water In The Al-Mu'aawanah Boarding School, District Rajadesa, Ciamis District

Background & Objective: Drinking water is a very important human need. Drinking water that is suitable for consumption must meet established standards, among others, according to the Minister of Health of the Republic of Indonesia No.492/Menkes/IV/2010. This study aims to determine the presence or absence of coliform bacteria in the drinking water of students using the MPN method. Methods: This type of research is descriptive. The sampling was carried out at the Al-Mu'aawanah Islamic boarding school. The results showed that the samples of drinking water for male and female students for microbiological parameters showed that the samples were positive for coliform bacteria. The sample for female students was 22/100 mL, and the sample for male students was 2/100 mL. Based on Permenkes No.472/Menkes/IV/2010, the maximum total concentration of coliform bacteria is 0/100 mL, which means that it is not in accordance with the standards of Permenkes No.472/Menkes/IV/2010, and the growth of faecal coliform bacteria was found in EMB media in both samples. namely the metallic green Escherichia coli bacteria, which indicates that the sample does not meet the microbiological requirements of drinking water.

The benefits of flushing for mitigating Legionella spp. in non-chlorinated building plumbing systems

Flushing is a common corrective action recommended by Legionella management guidelines to remove stagnant water and replenish disinfectant. Due to water age and different local regulations, buildings may receive water with low or no residual disinfectant. In such situations, the evidence for flushing efficacy is often anecdotal, and the benefits are poorly quantified. Using a pilot-scale premise plumbing system, flushed shower outlets were evaluated against non-flushed outlets during simulated periods of both active and low water demand. Water and biofilm concentrations of total bacteria, Legionella spp., and Vermamoeba vermiformis were quantified using real-time quantitative PCR. Even after all outlets returned to active water demand, flushed shower outlets continued to have lower quantities of Legionella compared to non-flushed outlets for several days, but the decrease was of little practical benefit (<0.5 log10[copies/L]). During prolonged periods of low water demand, however, there was no apparent benefit to flushing. Total bacteria grew to stationary phase within 3 ± 1 days, while Legionella spp. took 12 ± 6 days, regardless of whether the pipes had recently been flushed or not. Flushing with hot or cold water had little to no effect on the concentrations of total bacteria and Legionella in pipe wall biofilms. Flushing with cold water (9.6 and 13.2°C) decreased total bacteria concentrations in the water by 0.96 and 1.00 log10[copies/L], respectively and by 1.27 and 1.74 log10[copies/L] with hot water (49 and 60°C, respectively) but the difference in Legionella spp. concentrations between flushing with cold (1.55 log10[copies/L] for both) and hot water (1.32 to 1.88 log10[copies/L], respectively) was negligible. This suggests that hot water flushing, even at 60°C, provides little or no added benefit for managing Legionella in non-chlorinated building plumbing systems versus cold water flushing. Finally, the immediate benefits of flushing in terms of reductions in total bacteria and Legionella in the water were comparable as well as the rates at which those populations recovered during post-flush or post-shower stagnation.

Effect of odor treatment systems on bioaerosol microbial concentration and diversity from wastewater treatment plants

Biofiltration, activated carbon and chemical scrubbing are technologies used for odor control in wastewater treatment plants. These systems may also influence the airborne microbial load in treated air. The study objectives were to 1) evaluate the capacity of three odor control system technologies to reduce the airborne concentration of total bacteria, Legionella, L. pneumophila, non-tuberculous mycobacteria (NTM) and Cladosporium in winter and summer seasons and 2) to describe the microbial ecology of the biofiltration system and evaluate its impact on treated air microbial diversity. A reduction of the total bacterial concentration up to 25 times was observed after odor treatment. Quantification by qPCR revealed the presence of Legionella spp. in all air samples ranging between 26 and 1140 GC/m3, while L. pneumophila was not detected except for three samples below the limit of quantification. A significant increase of up to 25-fold of Legionella spp. was noticed at the outlet of two of the three treatment systems. NTM were ubiquitously detected before air treatment (up to 2500 GC/m3) and were significantly reduced by all 3 systems (up to 13-fold). Cladosporium was measured at low concentrations for each system (< 190 GC/m3), with 68 % of the air samples below the limit of detection. Biodiversity results revealed that biofiltration system is an active process that adapts to air pollutants over time. Legionella spp. were detected in significant abundance in the air once treated in winter (up to 27 %). Nevertheless, the abundance of protozoan hosts is low and does not explain the multiplication of Legionella spp. The season remains the most influential factor shaping biodiversity. In summer only, air biofiltration caused a significant enrichment of the biodiversity. Although odor control technologies are not designed for bacterial mitigation, findings from this study suggest their potential to reduce the abundance of some genera harboring pathogenic species.

Fibre Composition and Maturity of Forage-Based Diets Affects the Fluid Balance, Faecal Water-Holding Capacity and Microbial Ecosystem in French Trotters.

Racing events challenge the fluid balance of athletic horses. The equine large intestine functions as a fluid reservoir, since the properties of dietary forage fibre affect the digesta water content and the milieu of this ecosystem. This study aimed to investigate the effect of grass maturity and legume forage on the faecal water-holding capacity (WHC) and microbial ecosystem, and the fluid balance and body weight (BW) of French trotters in race training. Six geldings were offered three diets with different fibre compositions: concentrate and late-harvested mature grass haylage (35:65 energy ratio) (CMGH); early-harvested grass haylage and mature grass haylage (80:20) (EGH); and lucerne and mature grass haylage (80:20) (LH), for 24 days in a Latin square design. Body weights were lower and faecal WHC higher when the horses were fed EGH compared to CMGH and LH (485 vs. 492 and 492 kg, p < 0.001; 12.6 vs. 11.1 and 11.4 g H2O/g dry faeces, p = 0.014, respectively). Total water intake and output did not differ between diets, but water excretion via faeces was lower and via urine was greater on EGH compared to CMGH and LH (13.1 vs. 18.8 and 17.6 kg, p = 0.001; 10.5 vs. 7.6 and 7.9 kg, p = 0.032, respectively). Total bacteria concentrations were higher on EGH than CMGH and LH (5.4 × 1011 vs. 2.8 × 1011 and 2.8 × 1011 CFU/mL, p = 0.018, respectively). Concentrations of butyrate were greater, and pH was lower when fed EGH compared to CMGH and LH (3.0 vs. 2.0 and 1.6 mmol/L, p = 0.034; 6.5 vs. 6.9 and 7.2, p = 0.005, respectively). In conclusion, forage harvested at an early stage of maturity could benefit athletic horses' fluid balance by providing a more available large intestine fluid reservoir without increasing BW.

Distribution of Bacterial Concentration and Viability in Atmospheric Bioaerosols Under Different Weather Conditions in the Coastal Region of Qingdao

Bacteria are the most abundant microorganisms in atmospheric bioaerosols, widely distributed in the environment. Bioaerosol samples were collected using the FA-1 impact sampler from October 2013 to January 2021 in the coastal city of Qingdao, and samples stained with a BacLightTM bacterial viability kit were used to measure the concentrations of viable and non-viable bacteria with an epifluorescence microscope. The viable and non-viable bacteria in bioaerosols were characterized during different seasons, with particular attention paid to the distribution characteristics of bacteria on foggy, hazy, and dust days. The results showed that the mean concentrations of total bacteria were (1.06±0.68)×105 cells·m-3 in Qingdao during the sampling period, and those of viable and non-viable bacteria were (8.20±4.88)×103 cells·m-3and (9.74±6.72)×104 cells·m-3, respectively. The seasonal concentrations of non-viable bacteria were the highest in spring and winter and the lowest in summer, whereas that of viable bacteria was highest in spring, followed by those in summer and autumn, and the lowest in winter. Atmospheric bacterial concentrations fluctuated with by month, and total bacteria presented a similar variation pattern with that of non-viable bacteria. The monthly average concentration of non-viable bacteria showed the highest value in March during the spring and the lowest in June during the summer in 2021, whereas the highest value for viable bacteria occurred in May during the spring in 2021 and the lowest in December during the winter in 2020. Viable bacteria concentrations were significantly positively correlated with temperature and significantly negatively correlated with NO2, SO2, and CO. Non-viable bacteria were significantly positively correlated with PM. The bacteria in bioaerosol particles showed bimodal, unimodal, and skewed size distributions, varying with season and month. Under different weather conditions, the concentration of non-viable bacteria on dusty days was significantly higher than that on sunny, foggy, or hazy days, but the bacteria viability was as low as 6.85% due to long-distance transport. Anthropogenic pollution resulted in the lowest viability of bacteria at 4.10% on hazy days, whereas the highest viability in bacteria was 16.26% on foggy days due to high humidity. The size distribution of bacteria in bioaerosol particles under different pollution days showed a bimodal distribution, and the peak size depended on the weather type.

Quantification of antibiotic resistance genes (ARGs) in clouds at a mountain site (puy de Dôme, central France)

Antibiotic resistance in bacteria is becoming a major sanitary concern worldwide. The extensive use of large quantities of antibiotics to sustain human activity has led to the rapid acquisition and maintenance of antibiotic resistant genes (ARGs) in bacteria and to their spread into the environment. Eventually, these can be disseminated over long distances by atmospheric transport. Here, we assessed the presence of ARGs in clouds as an indicator of long-distance travel potential of antibiotic resistance in the atmosphere. We hypothesized that a variety of ARGs can reach the altitude of clouds mainly located within the free troposphere. Once incorporated in the atmosphere, they are efficiently transported and their respective concentrations should differ depending on the sources and the geographical origin of the air masses. We deployed high-flow rate impingers and collected twelve clouds between September 2019 and October 2021 at the meteorological station of the puy de Dôme summit (1465 m a.s.l., France). Total airborne bacteria concentration was assessed by flow cytometry, and ARGs subtypes of the main families of antibiotic resistance (quinolone, sulfonamide, tetracycline; glycopeptide, aminoglycoside, β-lactamase, macrolide) including one mobile genetic element (transposase) were quantified by qPCR. Our results indicate the presence of 29 different ARGs' subtypes at concentrations ranging from 1.01 × 103 to 1.61 × 104 copies m−3 of air. Clear distinctions could be observed between clouds in air masses transported over marine areas (Atlantic Ocean) and clouds influenced by continental surfaces. Specifically, quinolones (mostly qepA) resistance genes were prevalent in marine clouds (54 % of the total ARGs on average), whereas higher contributions of sulfonamide, tetracycline; glycopeptide, β-lactamase and macrolide were found in continental clouds. This study constitutes the first evidence for the presence of microbial ARGs in clouds at concentrations comparable to other natural environments. This highlights the atmosphere as routes for the dissemination of ARGs at large scale.

Impact of hydraulic and physicochemical factors on spatiotemporal variations of particle-associated bacteria in a drinking water distribution system

Drinking water distribution systems are critical infrastructure that protect public health by ensuring safe water is transported from centralized treatment facilities to consumers. While growth of bacteria, such as opportunistic pathogens, in distribution system infrastructure is well established as a detriment to water quality, little is known about the role of sediment in conveying bacteria via biofilms throughout these systems. The objective of this study was to quantify the abundance of particle-associated bacteria in a rural drinking water distribution system with a chlorine disinfectant residual. The role of hydraulic and physicochemical factors in influencing the spatiotemporal loading of particle-associated bacteria in the system was also examined. The concentration of particle-associated bacteria averaged 1.28 log10 gene copies per mL, while total bacteria averaged 2.16 log10 gene copies per mL, demonstrating that biofilms formed on the surface of sediment represent a substantial portion of overall loading in the studied distribution system. Total suspended solids concentrations were correlated with particle-associated bacteria, but not total bacteria. Pipe diameter was found to be an important factor associated with the abundance of both total and particle-associated bacteria, as well as total chlorine concentration. Velocity, Reynold's number and the flow regime were also found to be important, as they were associated with both sediment and total bacteria, but not particle-associated bacteria. The results of this study indicate that particle-associated bacteria and total bacteria concentrations often followed disparate trends, demonstrating that their abundance is differentially influenced by a complex combination of physicochemical and hydraulic factors. These findings help to establish sediment as an important conduit for microbial loading in a chlorinated drinking water distribution system.

Nanofiltration as an Efficient Tertiary Wastewater Treatment: Elimination of Total Bacteria and Antibiotic Resistance Genes from the Discharged Effluent of a Full-Scale Wastewater Treatment Plant

Wastewater reuse for agricultural irrigation still raises important public health issues regarding its safety, due to the increasing presence of emerging contaminants, such as antibiotic resistant bacteria and genes, in the treated effluents. In this paper, the potential for a commercial Desal 5 DK nanofiltration membrane to be used as a tertiary treatment in the wastewater treatment plants for a more effective elimination of these pollutants from the produced effluents was assessed on laboratory scale, using a stainless steel cross-flow cell. The obtained results showed high concentrations of total bacteria and target carbapenem and (fluoro)quinolone resistance genes (blaKPC, blaOXA-48, blaNDM, blaIMP, blaVIM, qnrA, qnrB and qnrS) not only in the discharged, but also in the reused, effluent samples, which suggests that their use may not be entirely safe. Nevertheless, the applied nanofiltration treatment achieved removal rates superior to 98% for the total bacteria and 99.99% for all the target resistance genes present in both DNA and extracellular DNA fractions, with no significant differences for these microbiological parameters between the nanofiltered and the control tap water samples. Although additional studies are still needed to fully optimize the entire process, the use of nanofiltration membranes seems to be a promising solution to substantially increase the quality of the treated wastewater effluents.

Influence of Seasons and Sites on Bioaerosols in Indoor Wastewater Treatment Plants and Proposal for Air Quality Indicators

During wastewater treatment, bioaerosols are generated and, can either remain in suspension for several hours or settle on surfaces and workers may be exposed. The presence of pathogens in the air could contribute to an increased frequency of gastrointestinal or respiratory illness amongst workers. Due to harsh winter conditions in Eastern Canada, much of the steps in the wastewater treatment process occur indoors, leading to a greater risk of significant occupational exposure especially if there is inadequate ventilation or a lack of personal protection. This work has used stationary sampling at various indoor wastewater treatment steps in both winter and summer. Bioaerosols were evaluated using both culture and molecular methods along with ventilation characterization. Endotoxins were quantified, as well as total cultivable and Gram-negative bacteria and pathogen indicators using qPCR. This study highlights the presence of potential pathogens at all steps in the treatment process, which may represent a potential occupational hazard. Comparisons between summer and winter data suggest that water temperature is an important factor for microbial activity and suggest that increasing the rate of air changes per hour in summer would be beneficial to reduce the concentration of bioaerosols during this time of the year. The screening, grit/FOGs removal and biofiltration were the most bioaerosols loaded sites. Based on strong correlations, we suggest the reconsideration of exposure limits in WWTPs. Workers should be encouraged to use personal respiratory protection to limit the risk of health problems, especially during long-term work. Implication Statement The work presented herein showcases significant correlations between concentrations of endotoxins, cultivable bacteria, Gram-negative bacteria, and total bacteria by qPCR from air collected in indoor wastewater treatment plants. These correlations lead us to propose new Limit of Exposure values, revisited to fit the Endotoxin exposure limits recommendations. The results can serve as guidelines for future proposals for air quality indicators.

Quality improvement of grounded peanut using superheated steam treatment by the inhibition of microorganisms, lipase, and aflatoxin

There are many problems of peanuts, such as lipid instability, high microbial content, and aflatoxin. In this study, effect of superheated steam (SS) treatment on the security quality of grounded peanut (GP) was evaluated. Results indicated that concentrations of total bacteria, mold, and Bacillus spp. were maximum reduced 99.26, 100, and 98.57%, respectively. SS processing led to 70% lipase activity and 90% POD activity reduction at 215°C for 2 min, which induced lower AV and PV after 70 days of storage compared to untreated GP. The moisture content dramatically decreased, but color of GP was not affected significantly. SS processing (215°C) was first to apply on degradation of total aflatoxin and aflatoxin B1. Therefore, SS is a novel and effective way to remove the microorganisms and aflatoxin in GP, as well as inhibiting lipase and peroxidase activities so as to improve the lipid stability. Novelty impact statement Mold could be totally inactivated by SS treatment at 140°C for 4 min or over 160°C for 1 min. Compared to untreated GP, SS processing at 140, 160, 180, and 215°C for 4 min could decrease 24.73, 25.52, 30.57, and 35.25% of AV, respectively. SS processing at 215°C for 1 min reduced 77.6% of AFs and 75.7% of AFB1.

Analyzing the Factors Contributing to Bacterial Contamination of Domestic Water Sources in Estuarine Islands of Coastal Karnataka, India

Contamination of domestic water sources is a major concern in estuarine islands of coastal Karnataka. Awareness about practices for waste management and protection of water sources on these islands is poor. Using water having bacterial presence for domestic purposes can lead to various health risks in humans. The research investigates various factors leading to bacterial contamination of domestic water sources at Mudukudru island of Swarna river, in the Udupi district of Karnataka. Samples were collected during the Pre-monsoon (December–May) and Monsoon (June–September) seasons from 43 wells of individual houses on the island. The total concentration of bacteria, in the water samples was determined from microbial analysis. The multiple tube, most probable number (MPN), fermentation technique was adopted to determine the total coliform in the samples. Factors like the presence or absence of well lining, well housing, wellhead above or below ground level, the distance of well from sewage pits, and the distance of livestock from the wells were correlated. The results indicated bacterial contamination in 32 wells during the monsoon as compared to pre-monsoon data, with the total coliform count exceeding the standard of 500 MPN/100 mL. A significant relation between total coliform count in the water samples and the absence of well lining with sanitary protection is observed with p-value = .00 ( p ⩽ .05) and wells located near (&lt;10.0 m) to sewage pit with p-value ⩽ .05, were recorded respectively during both the seasons of sampling. The study highlights the major factors leading to bacterial contamination of wells on the island. Awareness about the planning of domestic wells through community-driven programs and hygiene education can be beneficial for the sustainable future of these islands.

The emergency discharge of sewage to the Bay of Gda\u0144sk as a source of bacterial enrichment in coastal air

The purpose of this research was to study the presence of potential pathogenic bacteria in the seawater and air in five coastal towns (Hel, Puck, Gdynia, Sopot, Gdańsk-Brzeźno) as well as the enrichment of bacteria from the seawater into the coastal air after an emergency discharge of sewage into the Bay of Gdańsk. A total of 594 samples of air and seawater were collected in the coastal zone between spring and summer (between 2014 and 2018). Air samples were collected using the impact method with a SAS Super ISO 100. The multivariate analysis, conducted using contingency tables, showed a statistically significant variation between the concentration of coliforms, psychrophilic and mesophilic bacteria in the seawater microlayer and air in 2018, after an emergency discharge of sewage into the Bay of Gdańsk, compared to 2014–2017. Moreover, we detected a marine aerosol enrichment in psychrophilic, mesophilic bacteria, coliforms and Escherichia coli. We also showed a statistically significant relationship between the total concentration of bacteria and humidity, air temperature, speed and wind direction. This increased concentration of bacteria in the seawater and coastal air, and the high factor of air enrichment with bacteria maybe associated with the emergency discharge of wastewater into the Bay of Gdańsk. Therefore, it is suggested that in the event of a malfunction of a sewage treatment plant, as well as after floods or sudden rainfall, the public should be informed about the sanitary and epidemiological status of the coastal waters and be recommended to limit their use of coastal leisure areas.

Disease surveillance of cultured marine fish in the North of Bali, Indonesia

A regular surveillance of marine fish diseases was conducted from March to November 2019 in order to determine the occurrence time of the diseases within the mariculture centre of the North of Bali, Indonesia. The monthly surveillance was conducted by collecting 15 fish samples from each of the three hatcheries in Gerokgak and Penyabangan villages and of the two floating net cages in Pegametan Bay, Sumberkima village. Bacterial concentrations were grouped into 4 categories including low, moderate, high and very high. Surveillance data were analyzed using bivariate descriptive statistical methods. The results showed some important findings. First, the results of the study showed that NNV infection was found during the transitional seasons in March to June and September to November. Parasite infection were more frequent observed in fish with high and very high bacterial population. Second, high concentration of total bacteria in fish-feces occurred throughout the year. The prevalence of NNV infection and bacterial populations at the high to very high concentration were mostly occurred in the cultured fish in hatcheries at the size of 1-10 g, while in cultured fish at the net cages were mostly occurred at the size of > 50 g.

Concentrations, Size Distribution, and Community Structure Characteristics of Culturable Airborne Antibiotic-Resistant Bacteria in Xinxiang, Central China

Antimicrobial resistance is considered an important threat to global health and has recently attracted significant attention from the public. In this study, the concentrations and size distribution characteristics of culturable airborne total bacteria (TB) and four antibiotic-resistant bacteria (tetracycline-resistant bacteria (TRB), ciprofloxacin-resistant bacteria (CRB), erythromycin-resistant bacteria (ERB), and ampicillin-resistant bacteria (ARB)) were investigated for approximately one year to explore their variations under different seasons, diurnal periods, and air quality levels. The concentrations of TB and four antibiotic-resistant bacteria in winter and night were higher than during other seasons and diurnal periods. Their maximum concentrations were detected from air under moderate pollution or heavy pollution. PM2.5, PM10, SO2, and NO2 were positively related to TB and four antibiotic-resistant bacteria (p &lt; 0.01), whereas O3 and wind speed were negatively related to them (p &lt; 0.05). The particle size of TB and four antibiotic-resistant bacteria were mainly distributed in stage V (1.1–2.2 µm). Bacillus was the dominant genus of ARB (75.97%) and CRB (25.67%). Staphylococcus and Macrococcus were the dominant genera of TRB (46.05%) and ERB (47.67%), respectively. The opportunistic pathogens of Micrococcus, Sphingomonas, Enterococcus, Rhodococcus, and Stenotrophomonas were also identified. This study provides important references for understanding the threat of bioaerosols to human health.

Effects of Differences in Fibre Composition and Maturity of Forage-Based Diets on the Microbial Ecosystem and Its Activity in Equine Caecum and Colon Digesta and Faeces

Simple SummaryHorses are herbivores and forage-based diets are a natural choice for them. Traditionally, horse diets have included a large portion of cereals and have been associated with different intestinal problems. Feeding more forage and less concentrate has been shown to promote both physical and mental health and performance in horses. However, the nutritional quality of forage can differ greatly. This study compared the effect of two different forage diets and the more conventional forage and concentrate diets, on the hindgut microorganisms and the environment. No differences were found between the three diets in the concentration of total bacteria, fungi and protozoa, of cellulose-utilising bacteria or in the concentration of short-chain fatty acids. It can be concluded that a forage diet which fulfils the energy and protein requirements without having to add starch rich concentrate can benefit hindgut health. In addition, further studies on plant-fibre and forage diets for horses are of great importance for horse feeding, for advisors, veterinarians and for the diet formulations industry.Fibrous feeds are essential for horses. When developing feeding regimens promoting health and performance, we need to understand the digestion of plant cell walls and the functioning of the hindgut microbial ecosystem. Our objective was to investigate the effect of grass fibre maturity and legume forage on the hindgut microbiota and its activity. Six caecum and colon fistulated geldings were fed three diets differing in fibre composition: concentrate and late harvested grass haylage (35:65 energy ratio) (C); early and late harvested grass haylage (80:20) (G); lucerne and late harvested grass haylage (80:20) (L) for 28 days in a Latin-square design. No differences were measured in total bacteria concentrations, fungi and protozoa numbers nor in cellulolytic bacteria concentrations between the diets. Short-chain fatty acid concentrations did not differ between diets, but a lower (acetate + butyrate)/propionate ratio when the horses were fed Diet C, compared to G and L, was observed, suggesting lower fibrolytic and higher amylolytic activity. The pH increased when the horses were fed Diet L and decreased when fed C and G from caecum to faeces. The buffering capacity (BC) of hindgut digesta was five to fifteen-fold higher than that of the feeds, suggesting a decreased effect of feed BC as digesta travelled through the digestive tract. In conclusion, an early harvested forage opens up the possibility for forage-only diets, providing high energy without the negative effects of concentrate.
