The genetic basis of salinity tolerance traits in Arctic charr (Salvelinus alpinus)

BackgroundThe capacity to maintain internal ion homeostasis amidst changing conditions is particularly important for teleost fishes whose reproductive cycle is dependent upon movement from freshwater to seawater. Although the physiology of seawater osmoregulation in mitochondria-rich cells of fish gill epithelium is well understood, less is known about the underlying causes of inter- and intraspecific variation in salinity tolerance. We used a genome-scan approach in Arctic charr (Salvelinus alpinus) to map quantitative trait loci (QTL) correlated with variation in four salinity tolerance performance traits and six body size traits. Comparative genomics approaches allowed us to infer whether allelic variation at candidate gene loci (e.g., ATP1α1b, NKCC1, CFTR, and cldn10e) could have underlain observed variation.ResultsCombined parental analyses yielded genome-wide significant QTL on linkage groups 8, 14 and 20 for salinity tolerance performance traits, and on 1, 19, 20 and 28 for body size traits. Several QTL exhibited chromosome-wide significance. Among the salinity tolerance performance QTL, trait co-localizations occurred on chromosomes 1, 4, 7, 18 and 20, while the greatest experimental variation was explained by QTL on chromosomes 20 (19.9%), 19 (14.2%), 4 (14.1%) and 12 (13.1%). Several QTL localized to linkage groups exhibiting homeologous affinities, and multiple QTL mapped to regions homologous with the positions of candidate gene loci in other teleosts. There was no gene × environment interaction among body size QTL and ambient salinity.ConclusionsVariation in salinity tolerance capacity can be mapped to a subset of Arctic charr genomic regions that significantly influence performance in a seawater environment. The detection of QTL on linkage group 12 was consistent with the hypothesis that variation in salinity tolerance may be affected by allelic variation at the ATP1α1b locus. IGF2 may also affect salinity tolerance capacity as suggested by a genome-wide QTL on linkage group 19. The detection of salinity tolerance QTL in homeologous regions suggests that candidate loci duplicated from the salmonid-specific whole-genome duplication may have retained their function on both sets of homeologous chromosomes. Homologous affinities suggest that loci affecting salinity tolerance in Arctic charr may coincide with QTL for smoltification and salinity tolerance traits in rainbow trout. The effects of body size QTL appear to be independent of changes in ambient salinity.

Mechanisms of cadmium toxicity and tolerance in Populus

Abstract Background and Methods: Cadmium (Cd), ranked the 7th most hazardous substance, is one of the most widespread pollutants of soil and water in industrialized nations. Its increased movement in soil-plant systems is posing a serious threat to human health. Cd, without any known functions in plants, was found to be toxic even at minute concentrations, leading to the development of symptoms such as leaf roll, chlorosis and root and shoot growth reduction. Phytoremediation is an emerging cost-effective and environment friendly technology that utilizes high biomass producing plants including Populus plants to remove, transform or stabilize contaminants in soils. The objectives of our study were to record phenotypic variation in a Populus pedigree to Cd exposure, to identify Cd tolerant and susceptible genotypes of Populus, to map QTLs (Quantitative Trait Loci - genomic regions responsible) for Cd tolerance and accumulation in Populus and to predict a mechanism for Cd toxicity and tolerance in susceptible and tolerant Populus genotypes.QTL mapping was accomplished by conducting a greenhouse hydroponic study in which 252 genotypes of a Populus pseudo-backcross progeny were grown for 40 days and treated with 25 µM Cd. Phenotypic variation in total dry weight was recorded on these genotypes and was used for identifying QTL for Cd tolerance. We identified genotypes with contrasting responses to the Cd treatment and conducted a microarray study to identify potential Cd tolerance mechanisms based on gene expression patterns. Results/Conclusions: Significant variation was observed among genotypes in response to Cd treatment based on changes in total dry weight. Cd tolerant and susceptible genotypes were identified based on the least square mean differences (Control-Cd treated for each genotype) among all the genotypes. Three QTLs were identified for Cd tolerance and they accounted for approximately 25% of the phenotypic variation in Cd tolerance measured as total dry weights.In the microarray study, the Cd-susceptible genotype had higher expression of Fe-transporters compared to the tolerant genotypes, and Cd and Fe levels were significantly different in foliage. Even the susceptible genotype controls had higher, though not significant, Fe levels than tolerant genotype controls. We therefore hypothesize that part of the mechanism for Cd tolerance is determined by the differences in the activity of Fe transporters in genotypes with differential Fe homeostasis.

Seedling salt tolerance in tomato

Soils with higher concentrations of salt are becoming more and more a constraint for many crops to obtain high yields. Wild tomato species, adapted to adverse environments, are a potential reservoir for genes underlying quantitative trait loci (QTL) related to salt tolerance in tomato. In this study two introgression line (IL) libraries derived from two different wild species, Solanum pennellii LA716 and Solanum lycopersicoides LA2951, were used to identify QTLs for salt tolerance in the seedling stage. In the S. pennellii IL library, four major QTLs were identified on chromosomes 6, 7 and 11. In the S. lycopersicoides IL library, six major QTLs were discovered which are located on chromosomes 4, 6, 9 and 12. Co-localization of QTLs on chromosome 6 in the two IL libraries and previously reports hinted that this locus might be conserved in the tomato crop. Three S. pennellii ILs (IL6-2, IL7-1 and IL7-5) harboring QTLs on chromosome 6 and 7 were crossed. Semi-dominance and dominance were shown for these three QTLs, and non-additive and epistatic interactions between them were observed.

Introgression of an intermediate VRNH1 allele in barley (Hordeum vulgare L.) leads to reduced vernalization requirement without affecting freezing tolerance

The process of vernalization is mainly controlled by two genes in winter barley (Hordeum vulgare L.), VRNH1 and VRNH2. A recessive allele at VRNH1 and a dominant allele at VRNH2 must be present to induce a vernalization requirement. In addition, this process is usually associated with greater low-temperature tolerance. Spanish barleys originated in areas with mild winters and display a reduced vernalization requirement compared with standard winter cultivars. The objective of this study was to investigate the genetic origin of this reduced vernalization requirement and its effect on frost tolerance. We introgressed the regions of a typical Spanish barley line that carry VRNH1 and VRNH2 into a winter cultivar, Plaisant, using marker-assisted backcrossing. We present the results of a set of 12 lines introgressed with all four possible combinations of VRNH1 and VRNH2, which were evaluated for vernalization requirement and frost tolerance. The reduced vernalization requirement of the Spanish parent was confirmed, and was found to be due completely to the effect of the VRNH1 region. The backcross lines showed no decline in frost tolerance compared with that of the recurrent parent unless they carried an extra segment of chromosome 5H. This extra segment, a carryover of the backcross process, apparently contained the well-known frost tolerance quantitative trait locus Fr-H2. We demonstrate that it is possible to manipulate the vernalization requirement with only minor effects on frost tolerance. This finding opens the path to creating new types of barley cultivars that are better suited to specific environments, especially in a climate-change scenario.

Boron toxicity tolerance in barley through reduced expression of the multifunctional aquaporin HvNIP2;1.

Boron (B) toxicity is a significant limitation to cereal crop production in a number of regions worldwide. Here we describe the cloning of a gene from barley (Hordeum vulgare), underlying the chromosome 6H B toxicity tolerance quantitative trait locus. It is the second B toxicity tolerance gene identified in barley. Previously, we identified the gene Bot1 that functions as an efflux transporter in B toxicity-tolerant barley to move B out of the plant. The gene identified in this work encodes HvNIP2;1, an aquaporin from the nodulin-26-like intrinsic protein (NIP) subfamily that was recently described as a silicon influx transporter in barley and rice (Oryza sativa). Here we show that a rice mutant for this gene also shows reduced B accumulation in leaf blades compared to wild type and that the mutant protein alters growth of yeast (Saccharomyces cerevisiae) under high B. HvNIP2;1 facilitates significant transport of B when expressed in Xenopus oocytes compared to controls and to another NIP (NOD26), and also in yeast plasma membranes that appear to have relatively high B permeability. We propose that tolerance to high soil B is mediated by reduced expression of HvNIP2;1 to limit B uptake, as well as by increased expression of Bot1 to remove B from roots and sensitive tissues. Together with Bot1, the multifunctional aquaporin HvNIP2;1 is an important determinant of B toxicity tolerance in barley.

Identification and fine mapping of a major quantitative trait locus originating from wild rice, controlling cold tolerance at the seedling stage

Cold tolerance at the seedling stage (CTSS) is an important trait affecting stable rice production in temperate climates and areas of high elevation. In this study, 331 single nucleotide polymorphism (SNP) markers were developed and used along with phenotypic evaluation to identify quantitative trait loci (QTLs) associated with CTSS from a mapping population of 184 F(2) plants derived from a cold tolerant wild rice, W1943 (Oryza rufipogon), and a sensitive indica cultivar, Guang-lu-ai 4 (GLA4). Three QTLs were detected on chromosomes 3, 10 and 11. A major locus, qCtss11 (QTL for cold tolerance at seedling stage), was located on the long arm of chromosome 11 explaining about 40% of the phenotypic variation. Introduction of the W1943 allele of qCtss11 to the GLA4 genetic background increased CTSS. Based on the phenotypic and genotypic assessment of advanced backcross progenies, qCtss11 was dissected as a single Mendelian factor. A high-resolution genetic map was constructed using 23 markers across the qCtss11 locus. As a result, qCtss11 was fine mapped to a 60-kb candidate region defined by marker AK24 and GP0030 on chromosome 11, in which six genes were annotated. Expression and resequence analyses of the six candidates supported the hypothesis that Os11g0615600 and/or Os11g0615900 are causal gene(s) of the CTSS.

Fine mapping a QTL qCTB7 for cold tolerance at the booting stage on rice chromosome 7 using a near-isogenic line

Low temperature at the booting stage is a serious abiotic stress in rice, and cold tolerance is a complex trait controlled by many quantitative trait loci (QTL). A QTL for cold tolerance at the booting stage in cold-tolerant near-isogenic rice line ZL1929-4 was analyzed. A total of 647 simple sequence repeat (SSR) markers distributed across 12 chromosomes were used to survey for polymorphisms between ZL1929-4 and the cold-sensitive japonica cultivar Towada, and nine were polymorphic. Single marker analysis revealed that markers on chromosome 7 were associated with cold tolerance. By interval mapping using an F(2) population from ZL1929-4 x Towada, a QTL for cold tolerance was detected on the long arm of chromosome 7. The QTL explained 9 and 21% of the phenotypic variances in the F(2) and F(3) generations, respectively. Recombinant plants were screened for two flanking markers, RM182 and RM1132, in an F(2) population with 2,810 plants. Two-step substitution mapping suggested that the QTL was located in a 92-kb interval between markers RI02905 and RM21862. This interval was present in BAC clone AP003804. We designated the QTL as qCTB7 (quantitative trait locus for cold tolerance at the booting stage on chromosome 7), and identified 12 putative candidate genes.

A major terminal drought tolerance QTL of pearl millet is also associated with reduced salt uptake and enhanced growth under salt stress

The performance of a major quantitative trait locus (QTL) of terminal drought tolerance (DT) of pearl millet was assessed under salt stress. The test-cross hybrids of the QTL donor parent (drought tolerant, PRLT 2/89-33), QTL recipient parent (drought sensitive, H 77/833-2), and a set of six near isogenic lines introgressed with a terminal DT-QTL (QTL-NILs) were evaluated for germination and seedling emergence at 7 days after sowing (DAS) in Petri plates at four salinity levels, and at vegetative (24 DAS) and maturity stages at three salinity and alkalinity levels. Na+ and K+ accumulation, their compartmentation in different plant parts, and their effects on growth and yield parameters were evaluated. The DT-QTL donor parent and QTL-NILs accumulated less Na+ in shoot parts at seedling, vegetative and maturity stages, and also partitioned the accumulated Na+ more into nodes and internodes and less into leaves than the drought-sensitive recurrent parent. The pattern of reduced salt accumulation in the drought-tolerant parent and QTL-NILs was consistently associated with better growth and productivity in saline and alkaline treatments. It is concluded that the DT-QTL contributed by PRLT 2/89-33 exerted favourable effects on growth and productivity traits under salt stress by limiting Na+ accumulation in leaves.

Identification of a major QTL allele from wild soybean (Glycine soja Sieb. & Zucc.) for increasing alkaline salt tolerance in soybean

Salt-affected soils are generally classified into two main categories, sodic (alkaline) and saline. Our previous studies showed that the wild soybean accession JWS156-1 (Glycine soja) from the Kinki area of Japan was tolerant to NaCl salt, and the quantitative trait locus (QTL) for NaCl salt tolerance was located on soybean linkage group N (chromosome 3). Further investigation revealed that the wild soybean accession JWS156-1 also had a higher tolerance to alkaline salt stress. In the present study, an F(6) recombinant inbred line mapping population (n = 112) and an F(2) population (n = 149) derived from crosses between a cultivated soybean cultivar Jackson and JWS156-1 were used to identify QTL for alkaline salt tolerance in soybean. Evaluation of soybean alkaline salt tolerance was carried out based on salt tolerance rating (STR) and leaf chlorophyll content (SPAD value) after treatment with 180 mM NaHCO(3) for about 3 weeks under greenhouse conditions. In both populations, a significant QTL for alkaline salt tolerance was detected on the molecular linkage group D2 (chromosome 17), which accounted for 50.2 and 13.0% of the total variation for STR in the F(6) and the F(2) populations, respectively. The wild soybean contributed to the tolerance allele in the progenies. Our results suggest that QTL for alkaline salt tolerance is different from the QTL for NaCl salt tolerance found previously in this wild soybean genotype. The DNA markers closely associated with the QTLs might be useful for marker-assisted selection to pyramid tolerance genes in soybean for both alkaline and saline stresses.

Drought and salt tolerances in wild relatives for wheat and barley improvement

Drought and salinity are the major abiotic stresses that dramatically threaten the food supply in the world. Tribe Triticeae, including wheat and barley, possesses tremendous potential for drought and salt tolerance that has been extensively and practically identified, tested, and transferred to wheat cultivars with proven expression of tolerance in experimental trials. Triticum dicoccoides and Hordeum spontaneum, the progenitors of cultivated wheat and barley, have adapted to a broad range of environments and developed rich genetic diversities for drought and salt tolerances. Drought- and salt-tolerant genes and quantitative trait loci (QTLs) have been identified in T. dicoccoides and H. spontaneum and have great potential in wheat and barley improvement. Advanced backcross QTL analysis, the introgression libraries based on wild wheat and wild barley as donors, and positional cloning of natural QTLs will play prevailing roles in elucidating the molecular control of drought and salt tolerance. Combining tolerant genes and QTLs in crop breeding programs aimed at improving tolerance to drought and salinity will be achieved within a multidisciplinary context. Wild genetic resistances to drought and salinity will be shifted in the future from field experiments to the farmer.

QTL associated with heat susceptibility index in wheat (Triticum aestivum L.) under short-term reproductive stage heat stress

Heat stress adversely affects wheat production in many regions of the world and is particularly detrimental during reproductive development and grain-filling. The objective of this study was to identify quantitative trait loci (QTL) associated with heat susceptibility index (HSI) of yield components in response to a short-term heat shock during early grain-filling in wheat. The HSI was used as an indicator of yield stability and a proxy for heat tolerance. A recombinant inbred line (RIL) population derived from the heat tolerant cultivar ‘Halberd’ and heat sensitive cultivar ‘Cutter’ was evaluated for heat tolerance over 2 years in a controlled environment. The RILs and parental lines were grown in the greenhouse and at 10 days after pollination (DAP) half the plants for each RIL received a three-day heat stress treatment at 38°C/18°C day/night, while half were kept at control conditions of 20°C/18°C day/night. At maturity, the main spike was harvested and used to determine yield components. A significant treatment effect was observed for most yield components and a HSI was calculated for individual components and used for QTL mapping. QTL analysis identified 15 and 12 QTL associated with HSI in 2005 and 2006, respectively. Five QTL regions were detected in both years, including QTL on chromosomes 1A, 2A, 2B, and 3B. These same regions were commonly associated with QTL for flag leaf length, width, and visual wax content, but not with days to flowering. Pleiotropic trade-offs between the maintenance of kernel number versus increasing single kernel weight under heat stress were present at some QTL regions. The results of this study validate the use of the main spike for detection of QTL for heat tolerance and identify genomic regions associated with improved heat tolerance that can be targeted for future studies.

Terminal drought-tolerant pearl millet [Pennisetum glaucum (L.) R. Br.] have high leaf ABA and limit transpiration at high vapour pressure deficit.

It was previously shown that pearl millet genotypes carrying a terminal drought tolerance quantitative trait locus (QTL) had a lower transpiration rate (Tr; g cm−2 d−1) under well-watered conditions than sensitive lines. Here experiments were carried out to test whether this relates to leaf abscisic acid (ABA) and Tr concentration at high vapour pressure deficit (VPD), and whether that leads to transpiration efficiency (TE) differences. These traits were measured in tolerant/sensitive pearl millet genotypes, including near-isogenic lines introgressed with a terminal drought tolerance QTL (NIL-QTLs). Most genotypic differences were found under well-watered conditions. ABA levels under well-watered conditions were higher in tolerant genotypes, including NIL-QTLs, than in sensitive genotypes, and ABA did not increase under water stress. Well-watered Tr was lower in tolerant than in sensitive genotypes at all VPD levels. Except for one line, Tr slowed down in tolerant lines above a breakpoint at 1.40–1.90 kPa, with the slope decreasing >50%, whereas sensitive lines showed no change in that Tr response across the whole VPD range. It is concluded that two water-saving (avoidance) mechanisms may operate under well-watered conditions in tolerant pearl millet: (i) a low Tr even at low VPD conditions, which may relate to leaf ABA; and (ii) a sensitivity to higher VPD that further restricts Tr, which suggests the involvement of hydraulic signals. Both traits, which did not lead to TE differences, could contribute to absolute water saving seen in part due to dry weight increase differences. This water saved would become critical for grain filling and deserves consideration in the breeding of terminal drought-tolerant lines.

Modeling and mapping QTL for senescence-related traits in winter wheat under high temperature

Senescence is a genetically programmed and environmentally influenced process resulting in the destruction of chlorophyll and remobilization of nutrients to younger or reproductive parts of plants. Delayed senescence, or stay-green, contributes to a long grain-filling period and stable yield under stress. To model senescence and identify quantitative trait loci (QTL) for the trait, a population of recombinant inbred lines (RIL) from a cross between winter wheat cultivars, ‘Ventnor’ and ‘Karl 92’ was evaluated for heat tolerance under optimum temperature of 20/15°C (day/night) and continuous heat stress of 30/25°C from 10 days after anthesis (DAA) until maturity. Ventnor is a heat-tolerant cultivar and Karl 92 is a relatively heat-susceptible cultivar. Green leaf area was measured and used to model percent greenness retained over the reproductive period. Chlorophyll content and chlorophyll fluorescence were recorded on flag leaves. Senescence was converted to a quantitative trait using the model. Based on the modeled parameters, the RILs were categorized into three groups. When senescence-related traits were evaluated, nine QTL for heat tolerance were found on chromosome 2A, two each on chromosomes 6A and 6B and one each on chromosome 3A, 3B, and 7A. Both parents contributed favorable alleles for most of the senescence-related traits. Microsatellite markers Xgwm356 and Xgwm5 prominently linked to the senescence-related traits may be useful in marker-assisted breeding. These and the linked AFLP (amplified fragment length polymorphism) markers XCGT.TGCG-349, XCGT.GTG-343, and XCGT.CTCG-406, if converted to STS (sequence tagged sites), can be used for further molecular dissection of the QTL for post-anthesis heat tolerance.

Identification and analysis of QTLs controlling cold tolerance at the reproductive stage and validation of effective QTLs in cold-tolerant genotypes of rice (Oryza sativa L.)

Low temperature or cold stress is one of the major constraints of rice production and productivity in temperate rice-growing countries and high-altitude areas in the tropics. Even though low temperature affects the rice plant in all stages of growth, the percent seed set is damaged severely by cold and this reduces the yield potential of cultivars significantly. In this study, a new source of cold-tolerant line, IR66160-121-4-4-2, was used as a donor parent with a cold-sensitive cultivar, Geumobyeo, to produce 153 F(8) recombinant inbred lines (RILs) for quantitative trait locus (QTL) analysis. QTL analysis with 175 polymorphic simple sequence repeat (SSR) markers and composite interval mapping identified three main-effect QTLs (qPSST-3, qPSST-7, and qPSST-9) on chromosomes 3, 7, and 9. The SSR markers RM569, RM1377, and RM24545 were linked to the identified QTLs for cold tolerance with respect to percent seed set using cold-water (18-19 degrees C) irrigation in the field and controlled air temperature (17 degrees C) in the greenhouse. The total phenotypic variation for cold tolerance contributed by the three QTLs was 27.4%. RILs with high percent seed set under cold stress were validated with linked DNA markers and by haplotype analysis that revealed the contribution of progenitor genomes from the tropical japonica cultivar Jimbrug (Javanica) and temperate japonica cultivar Shen-Nung89-366. Three QTLs contributed by the cold-tolerant parent were identified which showed additive effect on percent seed set under cold treatment. This study demonstrated the utility of a new phenotyping method as well as the identification of SSR markers associated with QTLs for selection of cold-tolerant genotypes to improve temperate rice production.

Meta-analysis of constitutive and adaptive QTL for drought tolerance in maize

The response of plants to drought stress is very complex and involves expression of a lot of genes and pathways for diverse mechanisms and interactions with environments. Many quantitative trait loci(QTL) mapping experiments have given heterogeneous results due to use of different genotypes and populations tested in various environments. Our purpose was to identify some important constitutive and adaptive QTL using meta-analysis and to find specific genes and their families for speculating on drought tolerance networks. A total of 239 QTL detected under water-stressed conditions and 160 detected under control conditions from 12 populations tested in 22 experiments were compiled and compared, resulting in identification of 39 consensus QTL under water stress, and 36 under control conditions. Of them, 32 consensus QTL were supposed to be adaptive while others were constitutive QTL. The consensus QTL on chromosomes 1, 2, 3, 5, 6 and 9 were highly overlapped with several different traits and could be identified under multiple environments, most of which were related to traits of high phenotypic variance. Moreover, 48 candidate genes related to stress tolerance were located in silico in these consensus QTL regions what should facilitate the construction of QTL networks and help to understand the mechanisms related to drought tolerance.

A QTL analysis of aluminium tolerance in barley, using gene-based markers

Barley is more sensitive than the other major cereal crops to aluminium (Al) toxicity. Here, a doubled haploid mapping population was exploited to study the inheritance of Al tolerance in barley. Quantitative trait locus (QTL) mapping was based on a genetic map constructed with genic markers. Al tolerance QTL were identified on chromosomes 2H, 3H and 4H. A sequence homology search was used to derive the putative function of the genes linked to the QTL, in order to identify potential candidate genes for Al tolerance. Some of these candidates are implicated in stress/defence responses, in particular, stress signal transduction, transcription regulation factors and cell metabolism.

Quantitative Trait Loci and Epistasis for Oat Winter‐Hardiness Component Traits

ABSTRACTWinter hardiness is a complex trait and poor winter hardiness limits commercial production of winter oat (Avena spp.). The objective of this study was to identify quantitative trait loci (QTL) for five winter‐hardiness component traits in a recombinant inbred line population derived from a cross between the winter‐tender cultivar Fulghum and the winter‐hardy cultivar Norline. Crown freezing tolerance, vernalization response, and photoperiod response were evaluated in controlled environment studies. Heading date and plant height were evaluated over two seasons in Kinston, NC, and winter field survival was evaluated in five environments over two seasons in the mountains of North Carolina and Virginia. A partial genetic linkage map of regions believed to affect winter hardiness was developed using restriction fragment length polymorphism and simple sequence repeat markers. Most QTL were located on linkage groups FN3, FN22, and FN24. Quantitative trait loci were identified for all traits except photoperiod response, and epistatic interactions were identified for winter field survival, crown freezing tolerance, vernalization response, and plant height. Major QTL for winter field survival (R 2 = 35%) and crown freezing tolerance (R 2 = 53%) were identified on linkage group FN3, which was associated with an intergenomic reciprocal translocation between chromosomes 7C and 17.

Constitutive water-conserving mechanisms are correlated with the terminal drought tolerance of pearl millet [Pennisetum glaucum (L.) R. Br.

Pearl millet, a key staple crop of the semi-arid tropics, is mostly grown in water-limited conditions, and improving its performance depends on how genotypes manage limited water resources. This study investigates whether the control of water loss under non-limiting water conditions is involved in the terminal drought tolerance of pearl millet. Two pairs of tolerant×sensitive pearl millet genotypes, PRLT 2/89-33–H77/833-2 and 863B-P2–ICMB 841-P3, and near-isogenic lines (NILs), introgressed with a terminal drought tolerance quantitative trait locus (QTL) from the donor parent PRLT 2/89-33 into H77/833-2 (NILs-QTL), were tested. Upon exposure to water deficit, transpiration began to decline at lower fractions of transpirable soil water (FTSW) in tolerant than in sensitive genotypes, and NILs-QTL followed the pattern of the tolerant parents. The transpiration rate (Tr, in g water loss cm−2 d−1) under well-watered conditions was lower in tolerant than in sensitive parental genotypes, and the Tr of NILs-QTL followed the pattern of the tolerant parents. In addition, Tr measured in detached leaves (g water loss cm−2 h−1) from field-grown plants of the parental lines showed lower Tr values in tolerant parents. Defoliation led to an increase in Tr that was higher in sensitive than in tolerant genotypes. The differences in Tr between genotypes was not related to the stomatal density. These results demonstrate that constitutive traits controlling leaf water loss under well-watered conditions correlate with the terminal drought tolerance of pearl millet. Such traits may lead to more water being available for grain filling under terminal drought.

Genetic and physiological basis of adaptive salt tolerance divergence between coastal and inland Mimulus guttatus

Local adaptation is a well-established phenomenon whereby habitat-mediated natural selection drives the differentiation of populations. However, little is known about how specific traits and loci combine to cause local adaptation. Here, we conducted a set of experiments to determine which physiological mechanisms contribute to locally adaptive divergence in salt tolerance between coastal perennial and inland annual ecotypes of Mimulus guttatus. Quantitative trait locus (QTL) mapping was used to discover loci involved in salt spray tolerance and leaf sodium (Na(+)) concentration. To determine whether these QTLs confer fitness in the field, we examined their effects in reciprocal transplant experiments using recombinant inbred lines (RILs). Coastal plants had constitutively higher leaf Na(+) concentrations and greater levels of tissue tolerance, but no difference in osmotic stress tolerance. Three QTLs contributed to salt spray tolerance and two QTLs to leaf Na(+) concentration. All three salt-spray tolerance QTLs had a significant fitness effects at the coastal field site but no effects inland. Leaf Na(+) QTLs had no detectable fitness effects in the field. * Physiological results are consistent with adaptation of coastal populations to salt spray and soil salinity. Field results suggest that there may not be trade-offs across habitats for alleles involved in local salt spray adaptations.
