CYP720A1 function in roots is required for flowering time and systemic acquired resistance in the foliage of Arabidopsis

Systemic acquired resistance (SAR) is an inducible defense mechanism that systemically enhances resistance against pathogens in foliar tissues. SAR, which engages salicylic acid (SA) signaling, shares molecular components with the autonomous pathway, which is involved in controlling flowering time in Arabidopsis thaliana. FLOWERING LOCUS D (FLD) is one such autonomous pathway component that is required for flowering time and the systemic accumulation of SA during SAR. Here, we show that CYP720A1, a putative cytochrome P450 monoxygenase, controls FLD expression and is required for the timing of flowering and the manifestation of SAR. The delayed flowering time in the cyp720a1 mutant correlated with the elevated transcript level of the floral repressor FLC, while the SAR deficiency phenotype of the cyp720a1 mutant correlated with the inability to systemically accumulate SA. CYP720A1 transcript abundance in shoots is poor compared with roots. Reciprocal root-shoot grafting confirmed that CYP720A1 function in the roots is critical for flowering time and SAR. We therefore suggest that root to shoot communication involving a CYP720A1-dependent factor contributes to the timing of reproductive development and defense in the foliage.

Migratory strategy explains differences in timing of female reproductive development in seasonally sympatric songbirds

Abstract Divergent migratory strategies among populations can result in population‐level differences in timing of reproduction (allochrony) and local adaptation. However, the mechanisms underlying among‐population variation in timing are insufficiently understood, particularly in females. We studied differences in reproductive development and its related mechanisms along the hypothalamic–pituitary–gonadal axis (HPG) in closely related migratory and sedentary (i.e. resident) female dark‐eyed juncos (Junco hyemalis) living together in sympatry during early spring. Despite exposure to the same environmental cues in early spring, residents initiate breeding prior to the departure of migrants for their breeding grounds. We investigated whether residents would be more reproductively developed than migrants based on their behavioural differences. Alternatively, females could exhibit similar reproductive development in response to the same environmental cues despite differences in migratory behaviour. To compare their degree of reproductive development during seasonal sympatry and the underlying mechanisms of these differences, we collected ovarian and liver tissue in early spring prior to migration and compared transcript abundance of genes associated with reproduction using quantitative PCR. We also used stable hydrogen isotopes to infer relative breeding and wintering latitude of migrants. We found higher transcript abundance of luteinizing hormone receptor and aromatase in the ovary in addition to significantly heavier ovaries in residents than in migrants. Together, these results suggest greater sensitivity and response to upstream endocrine stimulation in resident females. Transcript abundance for other receptors in the ovary and liver associated with reproduction, however, did not differ between populations. When comparing ovarian development within migrants, females with lower hydrogen isotopes (indicating higher breeding latitudes) had smaller ovaries, suggesting that longer‐distance migrations may further delay reproductive development. Based on differences in ovary mass and transcript abundance, we conclude that females that differ in migratory strategy also differ in timing of reproductive development. These results support that divergent migratory behaviour drives allochrony and could enable reproductive isolation between populations; mechanistic differences at the level of gonadal stimulation can explain these differences in timing of reproductive development. Evolution of monogamy, paternal investment, and female life history in Peromyscus.

The timing of reproductive development and associated trade-offs in quantity versus quality of offspring produced across the life span are well documented in a wide range of species. The relation of these aspects of maternal life history to monogamy and paternal investment in offspring is not well studied in mammals, due in part to the rarity of the latter. By using five large, captive-bred populations of Peromyscus species that range from promiscuous mating with little paternal investment (P. maniculatus bairdii) to social and genetic monogamy with substantial paternal investment (P. californicus insignis), we modeled the interaction between monogamy and female life history. Monogamy and high paternal investment were associated with smaller litter size, delayed maternal reproduction that extended over a longer reproductive life span, and larger, higher quality offspring. The results suggest monogamy and paternal investment can alter the evolution of female life-history trajectories in mammals.

Chemical cues from kingsnakes do not cause inducible defenses in house mice

Abstract Many rodents exhibit inducible defenses when exposed to chemical cues from mammalian predators. These responses may include delays in sexual maturation, smaller adult body size and decreases in litter size and pup weight. We exposed the hybrid juvenile offspring of field-caught and lab-descended house mice Mus musculus to the chemical cues of mouse-fed or chick-fed kingsnakes, Lampropeltis getula, for 20 days after weaning, to examine the effects of ophidian predator cues on prey development. We hypothesized that these cues would elicit inducible defenses such as alteration of growth rates, and/or the timing of reproductive development in mice. Once mature, the reproductive effort of the mice might also be impacted by producing smaller litter sizes or lighter pups or not reproducing at all. We found no effect of kingsnake cues on any of the measures. These findings support the hypothesis that inducible defenses may have evolved as a strategy to deal with specific predators.

Expression of the Arabidopsis thaliana BBX32 Gene in Soybean Increases Grain Yield

Crop yield is a highly complex quantitative trait. Historically, successful breeding for improved grain yield has led to crop plants with improved source capacity, altered plant architecture, and increased resistance to abiotic and biotic stresses. To date, transgenic approaches towards improving crop grain yield have primarily focused on protecting plants from herbicide, insects, or disease. In contrast, we have focused on identifying genes that, when expressed in soybean, improve the intrinsic ability of the plant to yield more. Through the large scale screening of candidate genes in transgenic soybean, we identified an Arabidopsis thaliana B-box domain gene (AtBBX32) that significantly increases soybean grain yield year after year in multiple transgenic events in multi-location field trials. In order to understand the underlying physiological changes that are associated with increased yield in transgenic soybean, we examined phenotypic differences in two AtBBX32-expressing lines and found increases in plant height and node, flower, pod, and seed number. We propose that these phenotypic changes are likely the result of changes in the timing of reproductive development in transgenic soybean that lead to the increased duration of the pod and seed development period. Consistent with the role of BBX32 in A. thaliana in regulating light signaling, we show that the constitutive expression of AtBBX32 in soybean alters the abundance of a subset of gene transcripts in the early morning hours. In particular, AtBBX32 alters transcript levels of the soybean clock genes GmTOC1 and LHY-CCA1-like2 (GmLCL2). We propose that through the expression of AtBBX32 and modulation of the abundance of circadian clock genes during the transition from dark to light, the timing of critical phases of reproductive development are altered. These findings demonstrate a specific role for AtBBX32 in modulating soybean development, and demonstrate the validity of expressing single genes in crops to deliver increased agricultural productivity.

Ovarian developmental variation in the primitively eusocial waspRopalidia marginatasuggests a gateway to worker ontogeny and the evolution of sociality

Social insects are characterized by reproductive caste differentiation of colony members into one or a small number of fertile queens and a large number of sterile workers. The evolutionary origin and maintenance of such sterile workers remains an enduring puzzle in insect sociobiology. Here, we studied ovarian development in over 600 freshly eclosed, isolated, virgin female Ropalidia marginata wasps, maintained in the laboratory. The wasps differed greatly both in the time taken to develop their ovaries and in the magnitude of ovarian development despite having similar access to resources. All females started with no ovarian development at day zero, and the percentage of individuals with at least one oocyte at any stage of development increased gradually across age, reached 100% at 100 days and decreased slightly thereafter. Approximately 40% of the females failed to develop ovaries within the average ecological lifespan of the species. Age, body size and adult feeding rate, when considered together, were the most important factors governing ovarian development. We suggest that such flexibility and variation in the potential and timing of reproductive development may physiologically predispose females to accept worker roles and thus provide a gateway to worker ontogeny and the evolution of sociality.

The effect of environmental variation on the reproductive development time and output of the high-shore rockpool copepod Tigriopus brevicornis

The extreme environmental changes observed in high-shore rockpools occupied by Tigriopus brevicornis, provide a significant physiological challenge that may have implications for the energetics and hence the life history characteristics of this species. The present study was designed to assess the effect of this environmental variation on the reproductive development time and output of T. brevicornis in terms of duration of the various stages in ovary and egg sac development and maturation as well as naupliar development time to the first copepodite stage and total numbers of copepodites produced. There was a significant difference in reproductive development time and output in the three test salinities with only successful development to copepodite I stage seen in normal seawater (35 psu). There was also a successful release of nauplii seen at the low salinity (5 psu), but no subsequent survival to the copepodite I stage, whilst there was not even a development of egg sacs in the high salinity test medium (70 psu). There was also a significant difference in reproductive development times in the three test temperatures with fastest ovary development in the high temperature (23 °C) and slowest development time in the lowest temperature (5 °C). However, it should be noted that the highest number of nauplii produced was actually in the mid temperature (12 °C), not the high temperature. Exposure to hypoxic and anoxic conditions had a significantly detrimental effect on reproductive development with slower development time through the reproductive stages and no individuals surviving to copepodite stage I from the hypoxic and anoxic test conditions, but good development from the normoxic conditions. The present study indicates that animals living in high-shore rockpools may incur a considerable energetic cost, due to the environmental changes that are characteristic of this habitat and the importance of resource allocation in these different physiological conditions is paramount to the species' survival.

Improvement of Horticultural and Ornamental Crops Through Transgenic Manipulation of the Phytochrome Family of Plant Photoreceptors

Consumer demands drive continuous developments in the production of horticultural and ornamental crops. In addition to improvements in quality and nutritional content, crop producers must supply an increased variety of products to a year-round market. The availability of many horticultural and ornamental crop products is dependent on the timing of reproductive development. The time at which many plants initiate sexual or vegetative reproduction is governed by a number of interacting environmental factors such as daylength, light quality and temperature. Artificial manipulation of the growing environment is therefore frequently used to ensure production meets retailers' marketing programs, a strategy which can often result in high energy costs. An alternative approach involves the manipulation of genes encoding proteins responsible for perceiving and transducing environmental stimuli, in particular, the genes encoding the phytochrome family of plant photoreceptors. Alterations in the expression of genes encoding phytochromes can modulate not only the timing of reproductive development, but also plant architecture. Such approaches can therefore be used to modulate a variety of phenotypic traits such as height, lateral branching and harvest yield, while enabling growers to tailor crop reproduction to their marketing needs. In this review, we will discuss examples of crop improvement using transgenic manipulation of phytochrome expression, along with benefits and disadvantages of such approaches.

Sublethal effects on reproduction in native fauna: are females more vulnerable to biological invasion?

Although invasive species are a major threat to survivorship of native species, we know little about their sublethal effects. In soft-sediment marine systems, mat-forming invasive species often have positive effects, facilitating recruitment and enhancing the diversity and abundance of native invertebrates. However, because mat-forming invasive species change the habitat in which they invade, and benthic invertebrates are sensitive to environmental disturbance, important sublethal effects on native species may exist. Using a model marine system we show that the widespread mat-forming invasive alga Caulerpa taxifolia (Vahl) C. Agardh has strong negative effects on the reproductive traits of a native bivalve Anadara trapezia (Deshayes, 1840) (e.g. timing of reproductive development and spawning, and follicle and gamete production) even though the invader has positive effects on recruitment. Moreover, gender specific responses occurred and indicated that females were more susceptible to invasion than males. Our results indicate that sublethal effects of an invasive species on reproductive traits will have severe consequences for fitness of the native species.

Parenting and community background and variation in women's life-history development.

For a community sample of 623 low-income women (mean age = 26.9 years), the authors use life-history theory to explore relations between parenting and community background and timing of reproductive development and sexual behaviors. Among other patterns, reported levels of paternal involvement during childhood were related to delayed menarche and ages at 1st sexual intercourse and 1st childbirth. The relation between parental investment and women's reproductive delay varied with estimates of wealth in the community in which the participants grew up. The results suggest that parents increased their investment as the level of community wealth increased, and in ways that likely facilitated women's ability to compete for social status in adulthood. Implications for future research on the relation between parenting and child outcomes are discussed.

The Biology and Agronomy of Switchgrass for Biofuels

Switchgrass (Panicum virgatum L.)—a perennial, warm-season (C4) species—evolved across North America into multiple, divergent populations. The resulting natural variation within the species presents considerable morphological diversity and a wide range of adaptation. The species was adopted as a crop—initially as a forage—only in the last 50 yr. Its potential uses have recently been expanded to include biofuels. Management of switchgrass for biofuels is informed by an understanding of the plant's biology. Successful establishment requires attention to seed dormancy and weed control as well as proper depth and date of planting. The plant's growth rate is closely tied to temperature, but timing of reproductive development is linked to photoperiod. Accordingly, the period of vegetative growth can be extended by planting lower-latitude cultivars at higher latitudes. This strategy may provide a yield advantage, but cold tolerance can become limiting. Switchgrass is thrifty in its use of applied N; it appears able to obtain N from sources that other crops cannot tap. The N removed in harvested biomass is often greater than the amount of N applied. In areas with sufficient rainfall, sustainable yields of ∼15 Mg ha−1 yr−1 may be achievable by applying ∼50 kg N ha−1 yr−1. Harvesting biomass once per season—after plants have senesced and translocated N into perennial tissues—appears to allow plants to maintain an internal N reserve. Two harvests yr−1 may increase yields in some cultivars, but a single annual harvest maximizes yields in many cases. If two harvests are taken, more N must be applied to compensate for the N removed in the midseason harvest. Taking more than two harvests yr−1 often adversely affects long-term productivity and persistence. Switchgrass has potential as a renewable fuel source, but such use will likely require large infrastructural changes; and, even at maximum output, such systems could not provide the energy currently being derived from fossil fuels.

The highs and lows of plant life: temperature and light interactions in development

Plants must constantly respond to changes in the environment whilst maintaining developmental and growth processes if they are to survive into the next generation. A complex network of signals from temperature and light must correctly converge to achieve successful development, through vegetative to reproductive growth. Temperature can be thought of as an environmental factor that provides both 'inductive' and 'maintenance' signals in development. It can stimulate developmental processes such as seed dormancy release, germination and vernalization. However, when temperature is not regarded as inductive, an accommodating network of genes work in concert to ensure growth responses occur regardless of fluctuating microclimate conditions. Many of the temperature-regulated developmental pathways are intimately linked with light signaling. For example, light-temperature interactions are major determinants in the timing of reproductive development. Indeed, the ability to process and react to complex environmental cues is crucial for both normal and adaptive development in a changing environment. These responses are frequently mediated by manipulating the phytohormone network, which serves as a powerful, yet adaptable controller of development. This paper illustrates the influential role temperature perception plays throughout plant development and the close interaction between temperature, light and hormone signaling.

A pasture growth model for use in a whole-farm dairy production model

A generalised climate driven pasture growth model is described and evaluated by comparison to field observation. The model describes dry matter production and green-dead tissue flow dynamics for grazed temperate swards, especially perennial ryegrass ( Lolium perenne). The model includes a unique feature to account for light interception by non photosynthetic tissue. Extrapolation across environments occurs by the interaction of climate variables with three parameters that tune the model to a site. These parameters reflect the influence of soil fertility and sward species composition on production patterns. They are: (1) the efficiency of radiant energy use for photosynthesis, (2) the timing of reproductive development and (3) the relative efficiency of radiant energy use in vegetative compared to reproductive swards. Initial parameter settings were derived from data from a sheep grazing experiment in New Zealand. In this paper the ability to describe pasture production under dairy grazing at a different site is confirmed. When the three available parameters were calibrated for the dairy site, all but two of the 42 seasonal estimates of pasture production were within the 95% confidence interval for mean measured production. The model is being used as a component of a whole-farm dairy production model.

Effect of nutritional growth restriction on timing of reproductive development and plasma concentrations of insulin-like growth factor-1 and growth hormone in male red deer (Cervus elaphus) reared in constant photoperiod

AbstractThe effect of nutritional growth restriction on reproductive development in red deer stags reared in constant photoperiod was investigated and the correlation between reproductive status and circulating concentrations of growth hormone (GH) and insulin-like growth factor-1 (IGF-1) examined.Stags were reared from birth in constant photoperiod (12 h light: 12 h dark). They were offered a ‘complete diet’ ad libitum until they reached 52-5 kg live weight (LW) and were then maintained at this LWby restricting daily food dry-matter intake (DMI)for either a long (LR, 26 to 51 weeks of age, no. = 5) or short (SR, 21 to 33 weeks, no. = 6) period before being returned to ad libitum feeding.Relative to SR stags, antlers hardened later in LR stags (72·0 v. 57·5 weeks of age, P < 0·001); sustained elevations in plasma testosterone also began later (55·2 v. 38·5 weeks, P < 0·001) but at lower LW (54·0 v. 60·6 kg, P < 0·05). In both groups, the testosterone rise followed the return to ad libitum food by 4 to 5 weeks. Plasma IGF-1 was closely correlated with DMI (LR, r = 0·84, P < 0·001; SR, r = 0-93, P < 0·001) and with plasma testosterone (LR, r = 0·42, P < 0·001; SR, r = 0·38, P < 0·01). Also the increase in plasma IGF-1 preceded that of testosterone (by 2·8 (s.e. 0·94) weeks) and was associated with a transient plasma GH peak (P < 0·05) and elevated LH concentrations (SR, P < 0·01; LR, P < 0·05).The timing of reproductive development in growth-restricted male red deer in constant photoperiod may therefore be more sensitive to DMI than LW, and changes in plasma IGF-1 concentrations are consistent with a putative permissive role in relaying this information.

Influence of prenatal photoperiod on postnatal plasma concentrations of progesterone and prolactin in female red deer (Cervus elaphus) reared in constant equatorial photoperiod

Prenatal photoperiod influences postnatal prolactin secretion and the timing of reproductive development in male red deer reared from birth in a constant equatorial photoperiod (12:12 light:dark). The present trial investigated whether a similar phenomenon occurs in female red deer. Female deer whose mothers had been exposed for the last 14 weeks of gestation to long (group L, 18:6 light:dark) or short day length (group S, 6:18 light:dark) were kept from birth in constant equatorial day length with food available ad libitum. Both groups showed similar live-weight gain to 90-100 weeks of age. Blood samples taken once or twice weekly were analyzed for progesterone and prolactin. Progesterone concentrations indicated that there was no difference between the groups in the timing of the first incidence of ovarian (luteal) activity, which occurred at a normal or late age for natural puberty (67 weeks or older). Only one individual per group exhibited normal repeated luteal cyclicity since there was a high incidence of irregular or abnormal luteal function. Plasma prolactin concentrations at birth were higher in group L than group S (P < 0.001). Thereafter, although the mean and peak values did not differ significantly between the groups, there was a significant difference in the pattern of secretion; deer in group L showed significant clustering of prolactin peaks (P < 0.01) at a mean age of 48 weeks, whereas deer in group S showed a random distribution of peaks. Therefore, for female red deer raised in constant equatorial photoperiod, prenatal long day lengths did not advance timing of puberty.(ABSTRACT TRUNCATED AT 250 WORDS)

Limitations on Leaf Nitrate Reductase Activity during Flowering and Podfill in Soybean

The objective of this study was to identify factors which limit leaf nitrate reductase (NR) activity as decline occurs during flowering and beginning seed development in soybean (Glycine max [L.] Merr. cv Clark). Level of NR enzyme activity, level of reductant, and availability of NO(3) (-) as substrate were evaluated for field-grown soybean from flowering through leaf senescence. Timing of reproductive development was altered within one genotype by (a) exposure of Clark to an artificially short photoperiod to hasten flowering and podfill, and (b) the use of an early flowering isoline. Nitrogen (N) was soil-applied to selected plots at 500 kilograms per hectare as an additional variable. Stem NO(3) (-) concentration and in vivo leaf NR activity were significantly correlated (R(2) = 0.69 with nitrate in the assay medium and 0.74 without nitrate in the medium at P = 0.001) across six combinations of reproductive and soil N-treatment. The supply of NO(3) (-) from the root to the leaf tissue was the primary limitation to leaf NR activity during flowering and podfill. Levels of NR enzyme and reductant were not limiting to leaf NR activity during this period.
