This work considers the time-domain fluorescence diffuse optical tomography (FDOT). We recover the distribution of fluorophores in biological tissue by the boundary measurements. With the Laplace transform and the knowledge of complex analysis, we build the uniqueness theorem of this inverse problem. After that, the numerical inversions are considered. We introduce an iterative inversion algorithm under the framework of regularizing scheme, then give several numerical examples in three-dimensional space illustrating the performance of the proposed inversion schemes.

Mapping fluorescence resonance energy transfer parameters of a bifunctional agent using time-domain fluorescence diffuse optical tomography.

We present a method to map fluorescence resonance energy transfer (FRET) parameters of a bifunctional photodynamic therapy agent, (2-[1-hexyloxyethyl]-2-devinyl pyropheophorbide-a)-cyanine dye (HPPH-CD) conjugate, which consists of a photosensitizer (HPPH) and a fluorescent agent CD. We utilized time-domain fluorescence diffuse optical tomography, the normalized Born ratio model in the Fourier-domain, and an iterative algorithm to map depth-resolved spatial heterogeneities of FRET parameters. Our results exhibited depth-resolved changes of fluorophore's lifetime and the distance maps due to FRET between HPPH and CD. Our model suggests a potential approach of using FRET parameters to monitor efficacies of multifunctional photodynamic therapy agents in deep tissue.

Combined hemoglobin and fluorescence diffuse optical tomography for breast tumor diagnosis: a pilot study on time-domain methodology.

A combined time-domain fluorescence and hemoglobin diffuse optical tomography (DOT) system and the image reconstruction methods are proposed for enhancing the reliability of breast-dedicated optical measurement. The system equipped with two pulsed laser diodes at wavelengths of 780 nm and 830 nm that are specific to the peak excitation and emission of the FDA-approved ICG agent, and works with a 4-channel time-correlated single photon counting device to acquire the time-resolved distributions of the light re-emissions at 32 boundary sites of tissues in a tandem serial-to-parallel mode. The simultaneous reconstruction of the two optical (absorption and scattering) and two fluorescent (yield and lifetime) properties are achieved with the respective featured-data algorithms based on the generalized pulse spectrum technique. The performances of the methodology are experimentally assessed on breast-mimicking phantoms for hemoglobin- and fluorescence-DOT alone, as well as for fluorescence-guided hemoglobin-DOT. The results demonstrate the efficacy of improving the accuracy of hemoglobin-DOT based on a priori fluorescence localization.

Imaging a photodynamic therapy photosensitizerin vivowith a time-gated fluorescence tomography system

We report the tomographic imaging of a photodynamic therapy (PDT) photosensitizer, 2-(1-hexyloxyethyl)-2-devinyl pyropheophorbide-a (HPPH) in vivo with time-domain fluorescence diffuse optical tomography (TD-FDOT). Simultaneous reconstruction of fluorescence yield and lifetime of HPPH was performed before and after PDT. The methodology was validated in phantom experiments, and depth-resolved in vivo imaging was achieved through simultaneous three-dimensional (3-D) mappings of fluorescence yield and lifetime contrasts. The tomographic images of a human head-and-neck xenograft in a mouse confirmed the preferential uptake and retention of HPPH by the tumor 24-h post-injection. HPPH-mediated PDT induced significant changes in fluorescence yield and lifetime. This pilot study demonstrates that TD-FDOT may be a good imaging modality for assessing photosensitizer distributions in deep tissue during PDT monitoring.

Total light approach of time-domain fluorescence diffuse optical tomography

In this study, time-domain fluorescence diffuse optical tomography in biological tissue is numerically investigated using a total light approach. Total light is a summation of excitation light and zero-lifetime emission light divided by quantum yield. The zero-lifetime emission light is an emitted fluorescence light calculated by assuming that the fluorescence lifetime is zero. The zero-lifetime emission light is calculated by deconvolving the actually measured emission light with a lifetime function, an exponential function for fluorescence decay. The object for numerical simulation is a 2-D 10 mm-radius circle with the optical properties simulating biological tissues for near infrared light, and contains regions with fluorophore. The inverse problem of fluorescence diffuse optical tomography is solved using time-resolved simulated measurement data of the excitation and total lights for reconstructing the bsorption coefficient and fluorophore concentration simultaneously. The mean time of flight is used as the featured data-type extracted from the time-resolved data. The reconstructed images of fluorophore concentration show good quantitativeness and spatial reproducibility. By use of the total light approach, computation is performed much faster than the conventional ones.

A self-normalized, full time-resolved method for fluorescence diffuse optical tomography

A full time-resolved scheme that has been previously applied in diffuse optical tomography is extended to time-domain fluorescence diffuse optical tomography regime, based on a finite-element-finite-time-difference photon diffusion modeling and a Newton-Raphson inversion framework. The merits of using full time-resolved data are twofold: it helps evaluate the intrinsic performance of time-domain mode for improvement of image quality and set up a valuable reference to the assessment of computationally efficient featured-data-based algorithms, and provides a self-normalized implementation to preclude the necessity of the scaling-factor calibration and spectroscopic-feature assessments of the system as well as to overcome the adversity of system instability. We validate the proposed methodology using simulated data, and evaluate its performances of simultaneous recovery of the fluorescent yield and lifetime as well as its superiority to the featured-data one in the fidelity of image reconstruction.

We present a full three-dimensional, featured-data algorithm for time-domain fluorescence diffuse optical tomography that inverts the Laplace-transformed time-domain coupled diffusion equations and employs a pair of appropriate transform-factors to effectively separate the fluorescent yield and lifetime parameters. By use of a time-correlation single-photon counting system and the normalized Born formulation, we experimentally validate that the proposed scheme can achieve simultaneous reconstruction of the fluorescent yield and lifetime distributions with a reasonable accuracy.

Light propagation for time-domain fluorescence diffuse optical tomography by convolution using lifetime function

Time-domain light propagation in biological tissue is studied by solving the forward problem for fluorescence diffuse optical tomography using a convolution of the zero-lifetime emission light and the exponential function for a finite lifetime. We firstly formulate the fundamental equations in a time-domain assuming that the fluorescence lifetime is equal to zero, and then the solution including the lifetime is obtained by convolving the emission light and the lifetime function. The model is a two-dimensional (2-D) 10 mm-radius circle with the optical properties simulating biological tissue for the near infrared light, and contains some inclusions with fluorophores. Temporal and spatial profiles of excitation and emission light are calculated and discussed for several models with different inclusions. The results are physically reasonable and will be used for the inverse problem of fluorescence diffuse optical tomography.
