Dynamics of Infections in Cattle and Rhipicephalus microplus: A Preliminary Study.

Tick-borne pathogens (TBPs) pose a significant threat to livestock, including bovine species. This study aimed to investigate TBPs in cattle and ticks across four sampling points, utilizing real-time microfluidic PCR. The results revealed that Rhipicephalus microplus ticks were found infesting all animals. Among the detected TBPs in cattle, Anaplasma marginale was the most frequently identified, often as a single infection, although mixed infections involving Rickettsia felis, uncharacterized Rickettsia sp., and Anaplasma sp. were also observed. In ticks, A. marginale was predominant, along with R. felis, Rickettsia sp., and Ehrlichia sp. It is noteworthy that although A. marginale consistently infected all cattle during various sampling times, this pathogen was not detected in all ticks. This suggests a complex dynamic of pathogen acquisition by ticks. A phylogenetic analysis focused on the identification of Anaplasma species using amplified 16S rDNA gene fragments revealed the presence of A. marginale and Anaplasma platys strains in bovines. These findings underscore the presence of multiple TBPs in both cattle and ticks, with A. marginale being the most prevalent. Understanding the dynamics and phylogenetics of TBPs is crucial for developing effective control strategies to mitigate tick-borne diseases in livestock.

Unraveling the genetic mechanisms governing the host response to bovine anaplasmosis

Bovine anaplasmosis caused by Anaplasma marginale is a tick-borne disease of livestock with widespread prevalence and huge economic implications. In order to get new insights into modulation of host gene expression in response to natural infections of anaplasmosis, this study is the first attempt that compared the transcriptome profiles of peripheral blood mononuclear cells (PBMCs) of A. marginale infected and healthy crossbred cattle. Transcriptome analysis identified shared as well as unique functional pathways in the two groups. Translation and structural constituent of ribosome were the important terms for the genes abundantly expressed in the infected as well as healthy animals. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis of the differentially expressed genes revealed that immunity and signal transduction related terms were enriched for the up-regulated genes in the infected animals. The over-represented pathways were cytokine-cytokine receptor interaction and signaling pathways involving chemokines, Interleukin 17 (IL17), Tumour Necrosis Factor (TNF), Nuclear Factor Kappa B (NFKB) etc. Interestingly, many genes previously associated with parasite-borne diseases such as amoebiasis, trypanosomiasis, toxoplasmosis, and leishmaniasis were profusely expressed in the dataset of the diseased animals. High expression was also evident for the genes for acute phase response proteins, anti-microbial peptides and many inflammatory cytokines. Role of cytokines in mediating communication between immune cells was the most conspicuous gene network identified through the Ingenuity Pathway Analysis. This study provides comprehensive information about the crosstalk of genes involved in host defense as well as parasite persistence in the host upon infection with A. marginale.

Four Tick-Borne Microorganisms and Their Prevalence in Hyalomma Ticks Collected from Livestock in United Arab Emirates.

Ticks and associated tick-borne diseases in livestock remain a major threat to the health of animals and people worldwide. However, in the United Arab Emirates (UAE), very few studies have been conducted on tick-borne microorganisms thus far. The purpose of this cross-sectional DNA-based study was to assess the presence and prevalence of tick-borne Francisella sp., Rickettsia sp., and piroplasmids in ticks infesting livestock, and to estimate their infection rates. A total of 562 tick samples were collected from camels, cows, sheep, and goats in the Emirates of Abu Dhabi, Dubai, and Sharjah from 24 locations. DNA was extracted from ticks and PCR was conducted. We found that Hyalomma dromedarii ticks collected from camels had Francisella sp. (5.81%) and SFG Rickettsia (1.36%), which was 99% similar to Candidatus Rickettsia andeanae and uncultured Rickettsia sp. In addition, Hyalomma anatolicum ticks collected from cows were found to be positive for Theileria annulata (4.55%), whereas H. anatolicum collected from goats were positive for Theileria ovis (10%). The widespread abundance of Francisella of unknown pathogenicity and the presence of Rickettsia are a matter of concern. The discovery of T. ovis from relatively few samples from goats indicates the overall need for more surveillance. Increasing sampling efforts over a wider geographical range within the UAE could reveal the true extent of tick-borne diseases in livestock. Moreover, achieving successful tick-borne disease control requires more research and targeted studies evaluating the pathogenicity and infection rates of many microbial species.

Ticks and Tick-Borne Diseases of Livestock in the Middle East and North Africa: A Review.

Simple SummaryThe Middle East and North Africa represent a hyper arid region of the world. Humans in these regions have historically kept domestic livestock usually under harsh environmental circumstances. With recent human development, especially in the Middle East, the livestock industry has increased. Livestock is affected by ticks and tick-borne diseases on which there has been relatively few studies in this region. This review paper aims to (i) examine the diversity of ticks infesting livestock, (ii) assess the variety of pathogens in animals and humans, and (iii) to identify gaps in our understanding of tick biology and tick-borne disease transmission in the region. We found 55 tick species on livestock including camels, cows, goats and sheep, belonging to eight genera. Fifteen tick-borne pathogens were reported from livestock in the region. We highlight the magnitude of the tick problem in the region and evaluate the control efforts currently in place. We suggest that disease control and prevention could be achieved effectively through collaborative work among all stakeholders such as funding international research projects and establishing joint control programs to combat cross-border movement of ticks.Ticks are important vectors of an array of viral, bacterial and protozoan pathogens resulting in a wide range of animal and human diseases. There is limited information in the literature about tick species in the Middle East and North Africa (MENA) countries, even though they have suitable climate and vegetation for ticks and their hosts. We reviewed the occurrence of tick species and the pathogens they transmit from the MENA on published papers from 1901–2020. We found taxonomic records of 55 tick species infesting livestock representing the following eight genera: Ornithodoros, Otobius, Amblyomma, Dermacentor, Haemaphysalis, Hyalomma, Ixodes, and Rhipicephalus. In addition, 15 pathogens were recorded causing diseases of significance, with Crimean–Congo hemorrhagic fever, theileriosis, babesiosis and anaplasmosis being widely distributed diseases in the region. In recent decades, there has been increasing trends in disease occurrence and movement associated with global movement of humans and global trade of animals. We suggest that disease control and prevention could be achieved effectively through good integration between public health, veterinary medicine and animal management, and ecological approaches. We recommend further research in the areas of tick ecology and tick born-disease transmission. Furthermore, we suggest evaluation and improvement of disease control policies in the region.

Livestock owners\u2019 worry and fear of tick-borne diseases

BackgroundRecent global changes have led to an increase in distribution of ticks towards higher elevation and latitude in Europe and livestock are at increasing risk of contracting tick-borne diseases, but psychological aspects of how this affects human well-being are rarely assessed. Departing from the theory on emotional appraisal coming from psychology, this study investigates which factors that modulate worry and fear associated with the presence of ticks among livestock owners of sheep and/or cattle.MethodsSurvey data from 775 livestock owners in Norway were analysed by hierarchical multiple regression analysis with an index of fear of tick-borne diseases among livestock as the outcome variable.ResultsTwenty-nine per cent of the livestock owners reported worry and fear of tick-borne diseases among their livestock. The model explained 35% of the variance in worry and fear. There was a weak association between estimated incidences of tick-borne diseases in livestock and livestock owners’ worry and fear. Whereas previous personal experience of ticks and tick-borne diseases in livestock, and the livestock owners’ appraisals of the situation were more strongly associated with relatively stronger feelings of worry and fear.ConclusionsLivestock owners’ worry and fear of tick-borne diseases in livestock can partly be understood as their appraisals of perceived personal relevance of the presence of ticks, its potential negative implications for their daily life at large, and what potential they have to cope by different strategies to adapt or adjust to the situation.

Exploring the prevalence and diversity of bovine ticks in five agro-ecological zones of Pakistan using phenetic and genetic tools

Tick infestation is a leading cause of tick-worry and tick-borne diseases in livestock and associated economic losses in tropical and subtropical regions of the world. The cattle and buffalo populations in Pakistan are exposed to tick infestation throughout the year, but very little is known about the biology, diversity and distribution of tick species across different agro-ecological zones (AEZ) of the country. The present study aimed to investigate the prevalence (number of bovines infested with ticks out of the investigated population) and diversity of hard ticks infesting bovines in 30 villages located in five distinct AEZs (i.e. Arid, Indus delta, Northern irrigated plain, Sandy desert and Southern irrigated plain). We collected a total of 774 ticks (adult and nymphs) from cattle (n = 116) and water buffaloes (n = 88) on small-holder dairy farms (with <10 bovids per establishment) from September to November 2017. The overall tick prevalence was 46.1% (cattle: 47.9%; buffaloes: 44%), which varied significantly from 22.2% in the Indus delta to 70.5% in the Sandy desert. Tick prevalence was slightly higher in female (46.5%) than male animals (45%), and higher in calves (i.e. ≤ 1 year of age) (55%) than in young animals (i.e. up to 3 years of age) (39%) and adults (48%). Five tick species - Hyalomma anatolicum, Hyalomma hussaini, Hyalomma scupense, Rhipicephalus microplus and Rhipicephalus annulatus - were identified morphologically and then genetically. Genetic identification, achieved using the sequences of two mitochondrial (cytochrome c oxidase subunit 1 and 16S) and one nuclear ribosomal (second internal transcribed spacer) regions, was consistent with the morphological findings. Phylogenetic analyses of the DNA sequence data sets showed that the five species of tick identified here were closely related to the same species or closely related species from within and outside of Pakistan. Of five presently recognised taxa within the R. microplus complex, two were identified herein, including the R. microplus clade C and R. annulatus. This investigation provides the first genetic evidence of the occurrence of R. annulatus in Pakistan as well as Hy. hussaini and Hy. scupense in bovines specifically in the provinces of Sindh and Punjab, respectively. The present findings emphasise the importance of combining morphological and molecular approaches to study the diversity of ticks. Further longitudinal studies are required to establish seasonal variations in the prevalence and distribution of bovine ticks in different AEZs of Pakistan.

Molecular Detection and Phylogenetic Analysis of Anaplasma spp. in Korean Native Goats from Ulsan Metropolitan City, Korea.

Background: Tick-borne diseases (TBDs) can be fatal to humans as well as to animals causing severe economic losses globally to livestock industries. Many countries conduct regular surveillance of TBDs in livestock. Serological and molecular surveillance of TBDs in livestock and humans was carried out in the Republic of Korea. However, there are not a lot of data on analyses of anaplasmosis in Korean native goats and the correlation with rearing methods and seasons. Methods: In this study, goats in Ulsan city were tested for anaplasmosis by PCR and 16S rRNA sequencing. A total of 452 goat blood samples were collected from 20 farms in 2016. The goat farms in Ulsan city had three different types of rearing methods: conventional, confined, and mixed grazing-confined. Results: Forty-nine of the 452 goats (10.8%) were anaplasmosis positive. Sequence analysis of the PCR products from these 49 goats revealed that 39 of 452 goats (8.6%) were Anaplasma bovis positive, and 10 of 452 goats (2.2%) were infected with Anaplasma capra. The highest outbreaks of anaplasmosis occurred in mixed grazing-confined type of farms (27.1%, 33/122) (χ2 = 60.72, df = 2, p < 0.05), but there was no significant difference in the occurrence of anaplasmosis between spring, summer, and fall seasons. Conclusions: This study was the first detection of A. bovis in Korean native goats and its relationship with rearing methods and seasons. These findings suggested that Korean native goats were highly exposed to Anaplasma spp. during summers when the tick population is the highest and in farms employing mixed grazing-confined rearing methods.

Coxiella-like endosymbiont associated to the “Anatolian brown tick” Rhipicephalus bursa in Southern Italy

Several different ticks have been reported to harbor microbes related to Coxiella burnetii, the agent of the Q fever. Rhipicephalus bursa is an important vector of tick-borne diseases in livestock in Mediterranean area; it is also abundant in ovi-caprine farms with C. burnetii infection, in Southern Italy. 60 females of Rh. bursa (15 pools) and 40 their eggs (2 pools) were screened for C. burnetii by a conventional PCR targeting the insertion sequence IS1111 and by Loop mediated isothermal amplification assay (LAMP) targeting com1 gene. One of 15 tick pools (1/15) and both egg pools (2/2) were found positive by LAMP assay and negative by PCR targeting IS1111 gene. 16S rRNA gene was amplified by PCR from the LAMP-positive pools, amplicons were sequenced and found 95% similar to the corresponding sequences from C. burnetii. This let us to hypothesize the presence of a new Coxiella-like endosymbiont associated with Rh. bursa which could be vertically transmitted, described here for the first time. The lack of detection of IS1111 in Coxiella endosymbiont of Rh. bursa could be related to the possible absence of the Pathogenicity island of C. burnetii, to which IS1111s are associated.

Ethnoecological knowledge of ticks and treatment of tick-borne diseases among Maasai people in Northern Tanzania.

Aim:The aim of this study was to understand traditional knowledge of tick ecology and remedies for tick-borne diseases (TBDs) among the Maasai people in northern Tanzania.Materials and Methods:Semi-structured interviews were conducted among specific groups likely to be knowledgeable about tick ecology and TBDs in livestock among the Maasai people.Results:A total of 25 plant species belonging to 18 families were used to treat 8 different TBDs of livestock. Most of the plant species used were of Fabaceae and Burseraceae families. Aloe volkensii, Cissus grandifolia, and Terminalia brownii were the most commonly used plant species. The major plant growth form used was trees, while stems and bark were the main plant parts used. Most treatments were taken orally.Conclusion:Maasai people have substantial knowledge on tick ecology exemplified by their ability to differentiate between different tick species and the range of remedies for each of the TBDs. Because traditional ethnoveterinary remedies are frequently utilized, their effectiveness should be further investigated.

Tick-borne haemoparasites in African buffalo (Syncerus caffer) from two wildlife areas in Northern Botswana.

BackgroundThe African buffalo (Syncerus caffer) is a host for many pathogens known to cause economically important diseases and is often considered an important reservoir for livestock diseases. Theileriosis, heartwater, babesiosis and anaplasmosis are considered the most important tick-borne diseases of livestock in sub-Saharan Africa, resulting in extensive economic losses to livestock farmers in endemic areas. Information on the distribution of tick-borne diseases and ticks is scarce in Northern Botswana. Nevertheless, this data is necessary for targeting surveillance and control measures in livestock production at national level.MethodsIn order to address this gap, we analyzed 120 blood samples from buffalo herds for the presence of common tick-borne haemoparasites causing disease in livestock, collected in two of the main wildlife areas of Northern Botswana: the Chobe National Park (CNP, n = 64) and the Okavango Delta (OD, n = 56).ResultsAnalysis of the reverse line blot (RLB) hybridization assay results revealed the presence of Theileria, Babesia, Anaplasma and Ehrlichia species, either as single or mixed infections. Among the Theileria spp. present, T. parva (60%) and T. mutans (37%) were the most prevalent. Other species of interest were Anaplasma marginale subsp. centrale (30%), A. marginale (20%), Babesia occultans (23%) and Ehrlichia ruminantium (6%). The indirect fluorescent antibody test (IFAT) indicated 74% of samples to be positive for the presence of T. parva antibodies. Quantitative real-time PCR (qPCR) detected the highest level of animals infected with T. parva (81% of the samples). The level of agreement between the tests for detection of T. parva positive animals was higher between qPCR and IFAT (kappa = 0.56), than between qPCR and RLB (kappa = 0.26) or the latter and IFAT (kappa = 0.15).ConclusionsThis is the first report of tick-borne haemoparasites in African buffalo from northern Botswana, where animals from the CNP showed higher levels of infection than those from OD. Considering the absence of fences separating wildlife and livestock in the CNP and the higher levels of some parasite species in buffalo from that area, surveillance of tick-borne diseases in livestock at the interface in the CNP should be prioritized.

Occurrence of Ticks and Tick-Borne Diseases of Livestock in Zambezi Region: A Review

In an ecosystem such as Zambezi strip with characteristic patchwork of different land uses, land tenure types and boundaries, including country borders, veterinary fences, national parks, communal lands, government land, and forest reserves; the role played by free-ranging wildlife is a key point in the occurrence of endemic diseases such as those transmitted by tick vectors to livestock, human and wildlife hosts. Wildlife roams among different countries due to the porous nature of the borders, which makes trans-boundary management of such vector-borne diseases and natural resources a crucial issue in the southern African region. The large populations of wildlife in the tropics provide a reservoir of ticks and infective microbes that spread to domestic animals during grazing in the same environment. Ticks infesting livestock transmit numerous kinds of viruses, fungi, bacteria and protozoa between livestock, wildlife and human hosts causing poor health and socio-economic losses to their respective hosts. Since studies have indicated that there is no single, ideal solution to the control of ticks and tick-borne diseases, it becomes pertinent for stakeholders to use an integrated control, prevention and management approaches involving a comprehensive knowledge of all aspects of the vectors (ticks), hosts and aetiologic agents, particularly focusing on their biology, epidemiology and ecological issues. This review work focused on the current situation of ticks infesting livestock in Zambezi region, typical ticks of livestock, common genera of ticks infesting livestock in Zambezi region, common tick-borne diseases in Zambezi region, habitats and geographical prevalence of ticks as well as epidemiology of ticks and tick-borne diseases.

Ehrlichia ruminantium: A Promiscuous Genome

The intracellular bacterium, Ehrlichia ruminantium, is the causative agent of heartwater, a tick-borne disease of livestock. Because vaccines need to incorporate components from several virulent isolates, it is essential to have information on the extent of genetic variation among isolates. We therefore amplified and sequenced a panel of seven core function genes.

Bos taurus and Bos indicus (Sahiwal) calves respond differently to infection with Theileria annulata and produce markedly different levels of acute phase proteins

Disease-resistant livestock could provide a potentially sustainable and environmentally sound method of controlling tick and tick-borne diseases of livestock in the developing world. Advances in the knowledge and science of genomics open up opportunities to identify selectable genes controlling disease resistance but first, breeds and individuals with distinguishable phenotypes need to be identified. The Bos indicus breed, Sahiwal, has been exploited in dairy breeding programmes, because it is resistant to ticks and has relatively good performance characteristics compared to other indigenous cattle breeds of tropical regions. The analyses reported here show that Sahiwal calves were also more resistant than European Bos taurus (Holstein) dairy breed calves to tick-borne tropical theileriosis ( Theileria annulata infection). Following experimental infection with T. annulata sporozoites, a group of Sahiwal calves all survived without treatment, with significantly lower maximum temperatures ( P<0.01) and lower rates of parasite multiplication ( P<0.05) than a group of Holstein calves, which all had severe responses. Although the Sahiwals became as anaemic as the Holsteins, other measures of pathology, including enlargement of the draining lymph node and the acute phase proteins, α 1 acid glycoprotein and haptoglobin, were significantly less in the Sahiwals than in the Holsteins ( P<0.05). Additionally, the Sahiwals had significantly lower resting levels of α 1 acid glycoprotein than the Holsteins ( P<0.05). Production of a third acute phase proteins, serum amyloid A, had very similar kinetics in both breeds. Acute phase proteins are produced in response to systemic release of the kinds of pro-inflammatory cytokines that are thought to be responsible for the pyrexic, cachectic and anorexic responses characteristic of tropical theileriosis. The prolonged production of α 1 acid glycoprotein in the Holsteins is indicative of chronic production of circulating pro-inflammatory cytokines. In contrast, Sahiwals appear able to overcome infection with T. annulata as well as limit pathology by preventing the over-stimulation of pathways involving these cytokines.

Vector-borne diseases in humans and animals: Activities of the Swiss Tropical Institute and risks for Switzerland

The recent outbreak of anaplasmosis in a Swiss cattle herd triggered a discussion of the risk of vector-borne diseases in animals and humans in relation to climate changes and other factors. This overview presents the Swiss Tropical Institute's (STI) activities on vector-borne diseases (malaria, trypanosomosis, and leishmaniosis in humans and tick-borne diseases in livestock), describes the possible risks for humans and animals in Switzerland, and discusses options for action in the domains of public health, livestock production and companion animals. Switzerland is increasingly confronted with vector-borne diseases in humans and animals, but this is mainly due to an increase in imported cases. The emergence of a disease in one sector (human or veterinary medicine) may predict future trends in the other. A stronger intersectoral collaboration between public health and veterinary institutions at the federal and cantonal level is needed.
