Identification of differentially expressed genes in the growth plate of broiler chickens with thiram-induced tibial dyschondroplasia

Tibial dyschondroplasia (TD) is characterized by expansion of the proximal growth plates of the tibiotarsus that fail to form bone, lack blood vessels, and contain non-viable cells. Thiram (a carbamate pesticide), when fed to young broiler chicks, induces TD with high regularity and precision. We used this experimental model to understand the cause of the defects associated with TD by selecting and identifying the genes differentially expressed in the TD growth plate of broiler chickens. Broiler chicks at 7 days of age were randomly divided into two groups. After fasting overnight, they were fed with regular diet (control) or the same diet containing 100 mg/kg thiram for 96 h to induce TD (thiram-fed). mRNA was purified from the growth plates of control and thiram-fed broilers. Forward and reverse-subtracted cDNA libraries were generated by suppression subtractive hybridization technology. Ten selected genes from cDNA libraries were identified by real-time quantitative polymerase chain reaction. All were differentially expressed in TD growth plates (P<0.05 or P<0.01). The levels of collagen type X (Col X), pro-alpha-1 collagen type I (Col I α1), collagen type IX (Col IX), NADH dehydrogenase (NADH DH), cytochrome C oxidase subunit III (COX III), enolase 1, alpha (ENO1), carbonic anhydrase II (CA2) and heat shock protein 90 (Hsp90) mRNA transcripts were up-regulated, while the expression levels of Matrilin 3 (MATN3) and chondromodulin-I (ChM-I) were down-regulated. Col I and Hsp90 were detected by immunohistochemistry at different stages. Given that these genes are involved in matrix formation, endochondral ossification, developmental regulation, electron transport in the mitochondrial respiratory chain and vascularization, our findings may provide new insights into understanding the pathogenesis of TD.

Hypoxia, Hypoxia-Inducible Factor-1α (HIF-1α), and Heat-Shock Proteins in Tibial Dyschondroplasia

Tibial dyschondroplasia (TD) is one of the most prevalent skeletal abnormalities in avian species; it causes economic losses and is an animal welfare problem. It has been hypothesized that the absence of vasculature in the lesion of the TD growth plates at the ends of the long bones is involved in the etiology of the disease. We evaluated the hypoxia status of normal and thiram-induced TD growth plates by immunostaining the protein adducts after pimonidazole hydrochloride administration. In addition, we evaluated the expression of hypoxia-inducible factor-1α (HIF-1α), the major regulator of the hypoxic response that is essential for chondrogenesis, and that of heat-shock proteins (Hsp) downstream from HIF-1α. We demonstrated that, in contrast to the normal growth plates, those afflicted by TD were hypoxic. A major increase in hypoxia was observed in the proliferative, hypertrophic, and calcified zones. In the normal growth plate, HIF-1α was expressed in chondrocytes of the articular cartilage and of the maturation zone, whereas in cases of TD, HIF-1α was also expressed in chondrocytes below the lesion. The expression level of HIF-1α was related to the severity of the disease, but was independent of its cause; the same pattern of expression was observed in growth plates of chicks selected for a high incidence of TD. No differentiation-dependent expression of HIF-1α was observed in response to hypoxia, as demonstrated by the use of primary cultures of growth plate chondrocytes. In the normal growth plates, Hsp90 and Hsp70 were localized to the maturation zone. More cells expressed both Hsp in the TD lesion. In conclusion, we demonstrated that the TD growth plate, in contrast to the normal one, is hypoxic, probably because of the lack of vascularization. Hypoxia leads to an increase in the transcription factor HIF-1α, causing increases in the levels of Hsp90 and Hsp70.

Expression of type X collagen, Indian hedgehog and parathyroid hormone related-protein in normal and tibial dyschondroplastic chick growth plates

Tibial dyschondroplasia (TD) is a form of aberrant endochondral ossification in chickens, in that a plug of avascular cartilage (TD lesion) is formed within the growth plate. Histologically, the lesion is filled with apparently transitional chondrocytes that have been unable to differentiate to hypertrophic chondrocytes. We have examined the spatial expression of mRNAs for type X collagen, Indian hedgehog (Ihh) and Parathyroid Hormone-related protein (PTHrP) in the TD growth plate by in situ hybridization in order to ascertain at which stage chondrocyte differentiation is arrested in TD. In the normal growth plate, type X collagen mRNA was expressed by both prehypertrophic and hypertrophic chondrocytes. Indian Hedgehog mRNA was detected in a band of prehypertrophic chondrocytes and PTHrP expression was localized to a narrow band of prehypertrophic chondrocytes and in osteoblasts within the diaphysis. In TD sections, collagen X expression was seen within differentiating cells, within a small number of lesion cells, and within hypertrophic chondrocytes on the diaphyseal side of the lesion. Ihh expression was also seen within the differentiating cells and throughout the lesion. These data indicate that chondrocyte differentiation is arrested at the transitional stage just prior to hypertrophy. Contrary to the previously reported PTHrP expression patterns in TD chicks by immunohistochemistry, PTHrP mRNA was not detected in the TD lesion. This observation probably reflects the cessation of PTHrP gene expression by chondrocytes in the more severe TD lesions. The results from the present study also imply that the arrest of cell differentiation in TD is independent of PTHrP and that endochondral ossification in the post-hatch avian growth plate may involve additional regulatory pathways.

The biochemical characterization of aggrecan from normal and tibial-dyschondroplastic chicken growth-plate cartilage

Tibial dyschondroplasia (TD) is a disorder of endochondral ossification characterized by the presence of an avascular, non-mineralized cartilage lesion extending from the growth plate into the metaphysis. Cells within the TD growth plate fail to differentiate to full hypertrophy, and instead appear to maintain a ‘pre-hypertrophic’or ‘transitional’status. Studies of the expression and distribution of cartilage matrix macromolecules in the TD growth plate have shown a marked decrease in the levels of aggrecan in the TD matrix. In the present study we compared the biochemical characteristics of the aggrecan molecules extracted from normal epiphyseal and TD cartilage. We have shown three major differences between normal and TD cartilage aggrecan. These are: (1) increase in molecular mass; (2) increase in the number of keratan sulphate chains; and (3) difference in the pattern of sulphation in TD aggrecan. Such changes in biochemical characteristics of the aggrecan monomers in TD cartilage may be associated with the lack of mineralization of the diseased cartilage. The present study provides a basis for further investigations into the importance of proteoglycans in normal and pathological bone development.

Chondrocytes and Longitudinal Bone Growth: The Development of Tibial Dyschondroplasia

Growth plate cartilage is central to the process of bone elongation. Chondrocytes originating within the resting zone of the growth plate proceed through a series of intermediate phenotypes: proliferating, prehypertrophic and hypertrophic, before reaching a terminally differentiated state. Disruption of this chondrocyte maturational sequence causes many skeletal abnormalities in poultry such as tibial dyschondroplasia (TD), which is a common cause of deformity and lameness in the broiler chicken. Cell and matrix components of the growth plate have been studied in order to determine the cause(s) of the premature arrest of chondrocyte differentiation and retention of prehypertrophic chondrocytes observed in TD. Chondrocyte proliferation proceeds normally in TD, but markers of the differentiated phenotype, local growth factors, and the vitamin D receptor are abnormally expressed within the prehypertrophic chondrocytes above, and within, the lesion. Tibial dyschondroplasia is also associated with a reduced incidence of apoptosis, suggesting that the lesion contains an accumulation of immature cells that have outlived their normal life span. Immunolocalization studies of matrix components suggest an abnormal distribution within the TD growth plate that is consistent with a failure of the chondrocytes to fully hypertrophy. In addition, the collagen matrix of the TD lesion is highly crosslinked, which may make the formed lesion more impervious to vascular invasion and osteoclastic resorption. Recent studies have applied the techniques of differential display and semiquantitative reverse transcriptase-polymerase chain reaction to RNA obtained from discrete populations of growth plate chondrocytes of different maturational phenotypes. This strategy has allowed us to compare phenotypically identical cell fractions from normal and TD growth plates in an attempt to identify possible candidate genes for TD.

Expression patterns of chondrocyte genes cloned by differential display in tibial dyschondroplasia

Tibial dyschondroplasia (TD) appears to involve a failure of the growth plate chondrocytes within growing long bones to differentiate fully to the hypertrophic stage, resulting in a mass of prehypertrophic chondrocytes which form the avascular TD lesion. Many biochemical and molecular markers of chondrocyte hypertrophy are absent from the lesion, or show reduced expression, but the cause of the disorder remains to be identified. As differentiation to the hypertrophic state is impaired in TD, we hypothesised that chondrocyte genes that are differentially expressed in the growth plate should show altered expression in TD. Using differential display, four genes, B-cadherin, EF2, HT7 and Ex-FABP were cloned from chondrocytes stimulated to differentiate to the hypertrophic stage in vitro, and their differential expression confirmed in vivo. Using semi-quantitative RT-PCR, the expression patterns of these genes were compared in chondrocytes from normal and TD growth plates. Surprisingly, none of these genes showed the pattern of expression that might be expected in TD lesion chondrocytes, and two of them, B-cadherin and Ex-FABP, were upregulated in the lesion. This indicates that the TD phenotype does not merely reflect the absence of hypertrophic marker genes, but may be influenced by more complex developmental mechanisms/defects than previously thought.

Development of avian tibial dyschondroplasia: gene expression and protein synthesis.

Age-dependent gene expression and protein synthesis associated with chondrocyte differentiation were evaluated in the epiphyseal growth plates of normal and tibial dyschondroplasia (TD)-afflicted chickens. In the normal growth plate, collagen type II gene is expressed mainly by chondrocytes at the upper zone of the growth plate and by the chondrocytes in the articular cartilage. Collagen type X and osteopontin (OPN) genes are expressed in the lower zone of the growth plate and in the zone of cartilage-to-bone transition. No age-dependent changes in the pattern of OPN and collagen type II or X gene expression were observed up to 20 days of age. In the TD-afflicted growth plates, the lesion is enlarged with age, and chondrocytes expressing the collagen type II gene were observed in the hypertrophic zone as early as 8 days posthatching. Abnormal expression of OPN and collagen type X genes was also observed starting at 13 days of age. At day 20, the entire TD lesion-which was significantly enlarged-was surrounded by collagen type II, collagen type X, and OPN expressing cells. The level of OPN in TD was reduced with increasing age, and at 20 days almost no OPN could be detected in either the upper or the lower hypertrophic zones. The level of bone sialoprotein (BSP) also diminished with increasing age in the TD growth plates. In contrast to OPN, the age-dependent reduction in BSP levels was mainly in the lower hypertrophic zone (LHZ), and at 20 days of age, BSP was barely detected in the LHZ, whereas in the upper hypertrophic zone, the levels of BSP were similar to those in normal growth plate. In summary, our results suggest that the primary event of the TD lesion occurs in cells of proliferative phenotype within the hypertrophic zone. These cells divide and form the TD lesion, which consists of cells that do not express the genes associated with hypertrophy.

Expression of growth factors in chicken growth plate with special reference to tibial dyschondroplasia

Immunoreactive growth factors were identified in chick embryonic cartilage and bone, and in the growth plate of normal tibiotarsi and tibiotarsi affected with tibial dyschondroplasia (TD). A specific pattern of temporal and spatial expression was observed for each growth factor. Transforming growth factor beta and alpha (TGF beta and TGF alpha) and epidermal growth factor (EGF) were briefly expressed in chondrocytes of early chick embryos. Immunolabelling for TGF beta then gradually shifted into cartilaginous matrix and was not observed in cytoplasm of hypertrophic chondrocytes until the late embryonic and post-hatch stages. The distribution and intensity of TGF beta labelling was the same in chondrocytes of the TD and normal growth plate. Insulin-like growth factor I (IGF-I) labelling persisted from the early embryonic stage to the end of the mid-stage and then disappeared from chondrocytes. IGF-I appeared again in chondrocytes 1-2 days before hatching. After hatching, the labelling intensified in prehypertrophic and hypertrophic chondrocytes. TD lesions displayed IGF-I in the distal region, mainly in chondrocytes around small blood vessels. EGF reappeared in proliferative and hypertrophic chondrocytes of the mid-embryonic stage. By day 18 after hatching, EGF was present mainly in prehypertrophic and hypertrophic chondrocytes. EGF was demonstrated only in distal proliferative and early prehypertrophic chondrocytes of the dyschondroplastic growth plate. TGF alpha was identified in hypertrophic chondrocytes adjacent to the periosteum and in the distal tip of the mid-embryonic growth plate. With progressing ossification, TGF alpha labeling intensified in the embryonic hypertrophic chondrocytes. In the TD growth plate at day 18 after hatching, TGF alpha expression was limited to 1-3 concentric layers of chondrocytes surrounding blood vessels.

Matrix metalloproteinase activities in avian tibial dyschondroplasia

Tibial dyschondroplasia (TD) in poultry is a disorder of growth plate cartilage that fails to resorb and consequently prevents bone formation. Matrix metalloproteinases (MMP) contribute to the process of resorption through the degradation of extracellular matrices and facilitating vascularization, growth plate remodeling, and maturation. In order to understand the cause of the failure of cartilage degradation in TD, the gelatinase and collagenase activities, and the levels of collagen and glycosaminoglycans of conditioned media derived from cartilage-explant cultures of normal and TD growth plates were measured. Substrate zymography exhibited two prominent gelatinolytic and collagenolytic bands corresponding to MW 63, 59, and a broad but fuzzy band of activity between 100 and 200 kDa. On treatment with 4-aminophenylmercuric acetate, a compound that converts proenzyme forms of MMP, the 63 kDa MW gelatinolytic band migrated as a approximately 60 kDa band and contributed to the broadening of the 59 kDa band. The TD-growth plate-conditioned media had significantly lower collagenolytic-gelatinolytic activities. The sulfated glycosaminoglycans, but not the collagen contents of the conditioned media from TD-explant cultures, were also reduced significantly. It is likely that the decreased matrix metalloproteinase activities of growth plate chondrocyte may contribute to a reduced turnover of extracellular matrices (ECM), leading to the retention of cartilage and its lack of vascularity in TD-affected growth plates.

Growth plate chondrocyte vitamin D receptor number and affinity are reduced in avian tibial dyschondroplastic lesions.

Tibial dyschondroplasia (TD) is a condition of rapidly growing poultry in which a mass of unmineralized cartilage extends distally from the tibiotarsal growth plate, leading to deformity and lameness. The lesion is characterized by the accumulation of prehypertrophic chondrocytes, probably because the maturing chondrocytes are unable to differentiate fully. The condition can be prevented by feeding 1,25-(OH)2D3. We have investigated the possibility that vitamin D receptors (VDR), through which 1,25-(OH)2D3 exerts its differentiating effects on chondrocytes, may be defective in TD birds. Chondrocytes were isolated from the proliferating and hypertrophic zones of normal tibiotarsi and from the proliferating zone and lesion of affected birds and receptors were characterized by Scatchard analysis. Results showed that, while cells from the proliferating zone in TD birds had normal receptors, those from the TD lesion had significantly lower numbers and affinity for 1,25-(OH)2D3 compared to all other zones. Lesion VDR had low affinity; Kd 83.9 +/- 20.6 pM compared to 30.0 +/- 2.8, 37.8 +/- 3.1, and 33.0 +/- 4.0 pM (p < 0.001), and low receptor number per cell, 920 +/- 74, compared to 1329 +/- 151, 1664 +/- 167, and 1360 +/- 104 (p < 0.01) in the normal proliferating, normal hypertrophic, and TD proliferating cells, respectively. These findings were confirmed by immunohistochemical localization of VDR in sections of normal and TD growth plates using monoclonal antibody 9A7 gamma. In normal growth plate, most cells were VDR positive with intense staining in the mature hypertrophic chondrocytes; in TD growth plates, proliferating zone cells stained well but signal was largely absent from chondrocytes in the lesion. Image analysis showed integrated nuclear staining density per cell of 168.2 +/- 36.9 arbitrary units in normal hypertrophic cartilage compared to 98.8 +/- 60.2 units in the top of the lesion and 2.2 +/- 2.0 units in the midlesion. We conclude that both numbers and affinity of VDR are reduced in TD and this may explain the failure of chondrocytes to differentiate to the mature form. The adverse consequences of defective receptors may be partly overcome by treatment with 1,25-(OH)2D3.

Expression and distribution of cartilage matrix macromolecules in Avian tibial dyschondroplasia

Tibial dyschondroplasia (TD) is a disorder of endochondral ossification characterized by the presence of an avascular, non-mineralized cartilage lesion extending from the growth plate into the metaphysis. Cells within the TD growth plate fail to differentiate to full hypertrophy, and instead appear to maintain a 'pre-hypertrophic' or 'transitional' status. The synthesis and distribution of aggrecan, biglycan, decorin, and collagen types II and X in the growth plates of normal and tibial dyschondroplastic chickens have been investigated using in situ hybridization and immunolocalization. Marked reductions in the amount of aggrecan and biglycan core protein mRNAs were observed in the tibial dyschondroplastic lesion by in situ hybridization. Reduction in mRNA production seemed to be specific to the extracellular matrix components since total mRNA expression showed no significant difference between normal and dyschondroplastic cartilage. In addition, expression of collagen type II and decorin did not differ significantly between normal and TD cartilage. Distribution of aggrecan biglycan, decorin, type II and X collagens were examined using immunohistochemistry. Normal hypertrophic cartilage showed a strong matrix labelling for aggrecan and biglycan. Type X collagen in the normal hypertrophic cartilage showed strong pericellular and matrix distribution, whereas in TD cartilage labelling for aggrecan, biglycan and collagen X was located intracellularly with a very low level of signal in the matrix. In contrast, collagen type II was found to be present throughout the extracellular matrix of both the normal growth plate and the TD lesion, suggesting that the differences observed in aggrecan, biglycan and type X collagen distribution are specific to these proteins and not a general disturbance of matrix macromolecular metabolism. The reduced deposition of these macromolecules may have implications in normal and pathological bone development.
