Thyroxine administration at the end of the rutting season does not affect testosterone secretion or horn growth in the Iberian ibex (Capra pyrenaica)

Thyroid hormones play a role in the control of seasonal reproduction in wild and domestic ruminants such as red deer and sheep. Horn growth shows an inverse relationship with reproductive activity, but the effect of thyroid function on the regulation of the seasonal pattern of horn growth is unknown. This work examines the seasonal pattern of plasma thyroxine concentration, and its effect on testosterone secretion and seasonal changes in horn growth, in male Iberian ibexes (Capra pyrenaica). Plasma testosterone, plasma thyroxine, and horn growth were measured over a year in four males kept under natural photoperiod conditions. In another four, thyroxine was administered coinciding with the period of physiological reduction in plasma testosterone (January–February). In the first four, seasonal changes in the plasma concentration of thyroxine were detected with the highest concentrations recorded in October, November, and December, coinciding with the greatest testosterone secretion. In the second four, thyroxine treatment affected neither plasma testosterone concentration nor horn growth. The plasma concentrations of testosterone and thyroxine appear to be closely related in the ibex, but the seasonal pattern of horn growth does not seem to be modulated by seasonal changes in thyroxine.

Growth and Plasma Triiodothyronine Concentrations Are Modified by Selenium Deficiency and Repletion in Second-Generation Selenium-Deficient Rats

Classical glutathione peroxidase (GPX) is a useful Se-dependent parameter for determining Se status, but loss of GPX activity alone cannot explain the full effects of Se deficiency. The recent identification of type I thyroxine 5′-deiodinase as a Se-dependent enzyme provides a new potentially critical role for Se. To develop a model of impaired growth due to Se deficiency, second generation deficient weanling rats were fed a Se-deficient amino acid diet with adequate vitamin E and methionine. Initial growth rates of deficient males and females were 53 and 63%, respectively, of rats fed 0.1 µg Se/g diet. In short-term experiments with deficient males, liver Se and GPX activity were reduced 99%, liver glutathione-s-transferase activity was increased 114%, plasma thyroxine concentrations were increased 67%, plasma triiodothyronine was decreased 23% and the plasma triiodothyronine:thyroxine ratio was decreased 55%, compared with rats fed 0.2 µg Se/g diet. When deficient rats were injected on d 14 with 0, 1, 5 or 10 µg Se/100 g, rats grew 4.45, 7.62, 7.17 and 9.05 g/d, respectively, over the next 7 d. Injection with 10 µg Se/100 g restored plasma thyroxine and triiodothyronine concentrations 7 d later. Rats injected with 1 µg Se/100 g rat had significantly altered plasma thyroxine and triiodothyronine concentrations 1 but not 7 d after injection. Infusion of Se-deficient rats with 438 ng triiodothyronine/d for 7 d restored plasma triiodothyronine concentrations but did not increase growth rate compared with rats infused with saline. This model produced a significant growth depression that was significantly reversed by as little as 1 µg Se/100 g rat, but not by triiodothyronine infusion, suggesting that other Se-dependent roles in addition to 5′-deiodinase and GPX are necessary for adequate growth.

Hypothyroidism impairs development of the gastrointestinal tract in the ovine fetus.

Growth and maturation of the fetal gastrointestinal tract near term prepares the offspring for the onset of enteral nutrition at birth. Structural and functional changes are regulated by the prepartum rise in cortisol in the fetal circulation, although the role of the coincident rise in plasma tri-iodothyronine (T3) is unknown. This study examined the effect of hypothyroidism on the structural development of the gastrointestinal tract and the activity of brush-border digestive enzymes in the ovine fetus near term. In intact fetuses studied between 100 and 144days of gestation (dGA; term ∼145days), plasma concentrations of T3, cortisol and gastrin; the mucosal thickness in the abomasum, duodenum, jejunum and ileum; and intestinal villus height and crypt depth increased with gestational age. Removal of the fetal thyroid gland at 105-110 dGA suppressed plasma thyroxine (T4) and T3 concentrations to the limit of assay detection in fetuses studied at 130 and 144 dGA, and decreased plasma cortisol and gastrin near term, compared to age-matched intact fetuses. Hypothyroidism was associated with reductions in the relative weights of the stomach compartments and small intestines, the outer perimeter of the intestines, the thickness of the gastric and intestinal mucosa, villus height and width, and crypt depth. The thickness of the mucosal epithelial cell layer and muscularis propria in the small intestines were not affected by gestational age or treatment. Activities of the brush border enzymes varied with gestational age in a manner that depended on the enzyme and region of the small intestines studied. In the ileum, maltase and dipeptidyl peptidase IV (DPPIV) activities were lower, and aminopeptidase N (ApN) were higher, in the hypothyroid compared to intact fetuses near term. These findings highlight the importance of thyroid hormones in the structural and functional development of the gastrointestinal tract near term, and indicate how hypothyroidism in utero may impair the transition to enteral nutrition and increase the risk of gastrointestinal disorders in the neonate.

Short-term physiological responses to moderate heat stress in grazing dairy cows in temperate climate.

Even in temperate climate regions, an increase in ambient temperature and exposure to solar radiation can cause heat stress in lactating dairy cows. We hypothesised that grazing dairy cows exhibit short-term physiological changes due to increasing heat load under moderate climate conditions. Over two consecutive summers, 38 lactating Holstein dairy cows were studied in a full-time grazing system. Data were collected in 10 experimental periods of up to three consecutive days with a moderate comprehensive climate index (CCI). The individual animals' vaginal temperature (VT), heart rate, and locomotor activity data were automatically monitored with sensors. Blood samples and proportional whole milk samples were collected at afternoon milking. The concentrations of beta-hydroxybutyrate, glucose, non-esterified fatty acids, urea nitrogen, plasma thyroxine and triiodothyronine were analysed in blood plasma, and fat, protein, lactose, urea nitrogen, cortisol, Na+, K+, and Cl- concentrations were analysed in milk. The daily distribution of VT recordings greater than 39°C showed a circadian rhythm with a proportion of recordings of 2% and lower during the night and a percentage of 10% or higher in the afternoon. The cows' maximal daily vaginal temperature (VTMAX) between 0830 and 1430h was positively related to the mean daily CCI in the same time period (CCIMEAN; mean and SD 23.6±5.4°C). Cows with greater VTMAX had an increased mean heart rate, plasma glucose and milk cortisol concentrations and decreased concentrations of plasma thyroxine and triiodothyronine. The concentration of Na+ in milk was lower, and the concentration of K+ in milk tended to be higher in cows with increased VTMAX. For beta-hydroxybutyrate, non-esterified fatty acids and urea nitrogen concentrations in plasma and fat and lactose concentrations in milk no relationships were found in terms of increasing VT. For milk urea nitrogen and protein concentrations, the proportion of total variance explained by inter-individual or -period variance was high. In conclusion, changes observed in milk and blood likely reflected short-term physiological responses to moderate heat stress. In particular, milk cortisol and Na+ may be useful traits for timely monitoring of heat stress in individual cows because their inter-individual variances were relatively small and samples can be collected non-invasively.

Modulatory effects of lycopene and vitamin E on cloacal temperature, thyroid hormonal and reproductive performance responses in laying hens during the hot-dry season.

The aim of the study was to evaluate the effects of lycopene and vitamin E on cloacal temperature (CT), thyroid hormones and performance indices in laying hens (Gallus domesticus) during the hot-dry season. The dry-bulb temperature and temperature-humidity index in the pen and CT were measured in all hens twice weekly and thyroid hormones for five consecutive weeks. Ovarian and follicular activities were assessed at the end of the study after slaughter. The CT values in control hens at 09:00h, 12:00h and 15:00h (41.20±0.07°C, 41.84±1.8°C and 42.1±1.1°C, respectively) were higher (P<0.05), compared to the corresponding values recorded in lycopene (41.50±0.07°C, 41.50±0.07°C and 41.73±0.08°C, respectively), and lycopene+vitamin E (41.31±0.07°C, 41.40±0.05°C and 41.63±0.09°C, respectively). In lycopene+vitamin E laying hens, plasma thyroxine concentration (15.22±1.74nmol/L) was greater (P<0.05) than in lycopene (7.64±0.8nmol/L), vitamin E hens (6.80±1.3nmol/L) and controls (6.5±0.9°C nmol/L). Plasma triiodothyronine concentration was highest (P<0.05) in lycopene+vitamin E (4.80±0.37nmol/L), compared to lycopene (3.42±0.4nmol/L), vitamin E (1.96±0.2nmol/L) and control (1.2±0.1nmol/L) laying hens. Lycopene+vitamin E hens recorded higher (P<0.05) count of preovulatory follicles (6.0±0.2) than the controls (4.5±0.3). Countable white follicles were higher (P<0.05) in lycopene+vitamin E and lycopene hens (58.0±1.4 and 48.5±0.5, respectively) than controls (33.0±2.5). In conclusion, lycopene and vitamin E, especially their combination, modulated the heat stress-induced responses in the laying hens by decreasing CT values, and increasing thyroid hormone concentrations, the count of hierarchical preovulatory and white ovarian follicles during the hot-dry season.

Comparison of Different Herbal Additives on Immune Response and Growth Performance of Broiler Chickens

This experiment was conducted to compare the effects of four commercial herbal additives (Noviherb®, Bioessence®, Biostrong®, and Novigrow®), and a commercial antibiotic (virginiamycin) on growth performance, carcass yield, visceral organs weight, thyroid hormones, and humoral immune responses of broiler chickens. Nine hundred day-old Ross 308 male broiler chicks were reared in litter-covered floor cages and distributed into five experimental groups with six replicates of 30 birds. Five dietary treatments tested were diets supplemented with Noviherb®, Bioessence®, Biostrong® (each of them 100g/ton of diet), Novigrow® (1000g/ton of diet), and virginiamycin (100g/ton of diet) as a positive control. Feed intake, body weight, mortality, feed conversion ratio, European broiler index, and feed cost per kilogram of body weight were measured during the experiment. Antibody titers against sheep red blood cells (SRBC), immunoglobulin G, immunoglobulin M, and plasma concentrations of thyroxine (T4) and triiodothyronine (T3) were evaluated at the age of 42 days. Then, four birds per replicate were killed to determine the relative weight of carcass and organs. The GLM procedures of SAS software and Duncan’s multiple range test were applied to analyze data in a completely randomized design with five treatments and six replicates of 30 birds per each. The effects of herbal additives on productive traits feed cost per kilogram of body weight and relative weight of organs were not significant in comparison with antibiotic treatment. Dietary inclusion of Noviherb®, Biostrong®, and Novigrow® significantly decreased feed cost per kilogram of carcass compared with Bioessence® or antibiotic treatments (p&lt;0.05). The titers of IgG in Noviherb® and virginiamycin were significantly higher (p&lt;0.05) than in Biostrong® and Novigrow® fed groups. The plasma concentration of triiodothyronine was significantly decreased (p&lt;0.05) by dietary inclusion of Noviherb® and Novigrow® compared to the antibiotic. In conclusion, all the evaluated herbal additives could act as a substitute for the growth-promoting antibiotic.

Seasonal changes in testosterone and thyroxine concentrations in Mediterranean rams and bucks and their relationship with sperm cryoresistance

This study examines the relationship between seasonal changes in plasma testosterone and thyroxine concentrations and the relationship between these changes and the sperm resistance to the freezing-thawing process in Merino x Merino Precoz crossbreed rams and Florida breed bucks. Plasma testosterone and thyroxine concentration, sperm head area, sperm variables and their cryoresistance ratios were analyzed throughout a year. Both species showed a rise in thyroxine preceding testosterone fall. Plasma thyroxine concentrations in rams were higher during autumn than in spring and summer (P<0.05), whereas testosterone concentrations were not influenced by season. Conversely, bucks showed seasonal differences in plasma testosterone levels, with higher concentrations during summer and autumn than winter and spring (P<0.05), whereas thyroxine concentrations were not influenced by season. In bucks, a negative correlation was detected between plasma testosterone concentrations and the cryoresistance ratio for membrane integrity (R=-0.88; P<0.05) from April to September and between plasma thyroxine concentrations and the cryoresistance ratios for total motility (R=-0.66; P<0.05), curvilinear velocity (R=-0.61; P<0.05), the amplitude of lateral head displacement (R=-0.77; P<0.05) and the beat-cross frequency (R=-0.74; P<0.05), throughout the year. The results suggest that the annual variation in sperm cryoresistance may be related to seasonal endocrine changes, at least in goat. Winter and spring are the most appropriate seasons to collect and freeze sperm from Florida goat.

Determinants of placental iodine concentrations in a mild-to-moderate iodine-deficient population: an ENVIRONAGE cohort study

BackgroundIodine is an essential trace element for the production of thyroid hormones, and plays a key role during the gestational period for optimal foetal growth and (neuro-)development. To this day, iodine deficiency remains a global burden. Previous studies indicate that the placenta can store iodine in a concentration-dependent manner and serve as a long-term storage supply, but studies on the determinants of long-term placental iodine load are limited.MethodsThe placental iodine concentrations were determined for 462 mother-neonate pairs from the ENVIRONAGE birth cohort (Limburg, Belgium). Sociodemographic and clinical variables were obtained from questionnaires and medical files. Determinants of placental iodine concentration were identified using stepwise multiple regression procedures (p value < 0.15). The biological significance of our findings was investigated by measuring the plasma thyroid hormones in maternal and cord blood of 378 participants.ResultsA higher pre-pregnancy BMI, higher gestational weight gain, and alcohol consumption during pregnancy were linked with lower placental iodine storage. Multi-vitamin supplementation during pregnancy and longer gestation were associated with higher levels of placental iodine. Children born during the winter period had on average higher placental iodine levels. Besides, we found a significant positive time trend for placental iodine load over the study period 2013 to 2017. Lastly, we observed positive associations of both the maternal and cord plasma thyroxine concentrations with placental iodine load, emphasizing their biological link.ConclusionsThis study identified some determinants likely presenting a risk of reduced iodine storage during the gestational period of life. Future studies should elucidate the effects of lower placental iodine load on neonatal health, and health later in life.

Obesity treatment effect in Danish children and adolescents carrying Melanocortin-4 Receptor mutations

ObjectivesTo determine the prevalence of Melanocortin-4 Receptor (MC4R) mutations in a cohort of children and adolescents with overweight or obesity and to determine whether treatment responses differed between carriers and noncarriers.MethodsUsing target region capture sequencing, an MC4R mutation screen was performed in 1261 Danish children and adolescents enrolled at a tertiary multidisciplinary childhood obesity treatment center. Measurements of anthropometrics, blood pressure, fasting blood biochemistry including lipid and hormone levels, and dual-energy X-ray absorptiometry were performed at baseline and throughout treatment.ResultsOf 1209 children and adolescents that met all criteria to be included in the described analyses, 30 (2.5%) carried damaging or unresolved MC4R mutations. At baseline, mutation carriers exhibited higher concentrations of plasma thyroid-stimulating hormone (p = 0.003), and lower concentrations of plasma thyroxine (p = 0.010) compared to noncarriers. After a median of 1 year of treatment (range 0.5–4.0 years), body mass index (BMI) standard deviation score (SDS) was reduced in noncarriers but not in carriers, and this difference in treatment response was statistically significant (p = 0.005). Furthermore, HDL cholesterol was reduced in carriers, a response significantly different from that of noncarriers (p = 0.017).ConclusionAmong Danish children and adolescents with overweight or obesity entering a tertiary lifestyle intervention, 2.5% carried damaging or unresolved MC4R mutations. In contrast to noncarriers, carriers of damaging or unresolved MC4R mutations failed to reduce their BMI SDS during obesity treatment, indicating a need for personalized treatment based on the MC4R genotype.

The concentration of triiodothyronine and thyroxine in colostrum, milk and plasma of cows

Milk contains key nutrients in optimal proportions, which makes it to be an important component of the human diet. Despite the fact that consumers receive with milk a significant amount of readily available nutrients, milk can be a source of dangerous factors of chemical and biological origin. The level of physiological hormones found in milk is not a limiting factor and is studied solely for scientific purposes. However, milk hormones can have a negative effect on the health of consumers. Based on this, the aim of this work was to study the concentration of triiodothyronine and thyroxine in colostrum, milk and blood plasma of cows at different stages of lactation. The secretion of the mammary gland and blood plasma of cows of Ukrainian black-and-white dairy breed, in their 2nd–5th lactations with milk yield 5100–5700 kg of milk per previous lactation was the object of the study. The concentration of triiodothyronine and thyroxine using enzyme-linked immunosorbent assay was investigated in withdrawn samples. The obtained data indicates that the highest level of secretion of iodine-containing thyroid hormones with milk is observed on 1st–4th days of lactation. Thereafter, the concentration of hormones in milk statistically significant decreases by 2.8–3-fold (P ˂ 0.001) till 30–40th day of lactation. The high concentration of iodine-containing hormones in colostrum allows to enhance metabolic processes in calves. Studies of the concentration of thyroid hormones in the blood plasma of cows showed a gradual increase from the beginning of lactation till 10th–14th and then till 30th–40th days of lactation. By 10th–14th days we observed 2.2-fold growth of triiodothyronine level (P ˂ 0.001), and till 30th–40th days this parameter has increased by another 12.5 %. The level of thyroxine on 10th–14th days of lactation was higher, compared to such in the beginning of lactation by 62.2 % (P ˂ 0.001), and by 30th–40th day increase was by 40.9 %. The main reasons for such changes are the formation of lactation dominant, the discharge of hormones secreted by the breast and the stimulation of metabolic processes during lactation. The prospects of further research lie in investigation of the dependence of triiodothyronine and thyroxine levels in the secretion of the mammary gland on productivity, breed, age and diet of animals.

Effect of short-term intermittent exposure to waterborne estradiol on the reproductive physiology of the round goby (Neogobius melanostomus)

The objective of this study was to determine how the short-term exposure to a supraphysiological concentration of waterborne 17β-estradiol (E2) influences on melatonin (Mel) and thyroxine (T4) concentrations in plasma and E2 and 11-ketotestosterone (11-KT) concentrations in plasma and gonads in both sexes of round goby (Neogobius melanostomus) during the pre-spawning, spawning, late spawning and non-spawning phases. The experimental protocol was based on short-term, repeated exposures of fish to a supraphysiological dose of waterborne E2. Mel level was unchanged on exposure to E2 during the investigated phases, and its role in determining a time frame for spawning in both sexes of round goby seems to be stable in those conditions. T4 and sex steroids (E2 and 11-KT) were sensitive to the exposure of E2, and those changes influence gonads by accelerating oocyte development, ovulation and regression and inhibiting spermatogenesis in this species. The results demonstrate that the physiological responses of fish in all investigated phases were altered over a short window of exposure, indicating that short-term exposure to a supraphysiological dose of E2 may impact fish in the wild. Furthermore, round goby can be recommended as a very suitable model for studying endocrine disruptors, which is sensitive to even short exposure to E2.

Effects of treatment with haloperidol and clozapine on the plasma concentrations of thyroid hormones in rats.

Psychoactive drugs are group of compounds used to treat severe mental problems, including psychosis, as well as other conditions. This study assessed clinically relevant side effects of haloperidol and clozapine on the thyroid hormones. Haloperidol (0.05 and 2 mg/kg) or clozapine (0.5 and 20 mg/kg) was intraperitoneally injected to male Wistar rats for 28 days. The control group received 2 ml of physiological saline. A chemiluminescent immunoassay was used to measure the plasma levels of thyroid hormones. Plasma concentrations of thyroxine (T4) in rats treated with high-dose (2 mg/kg) of haloperidol decreased significantly compared to the control group (p=0.001). However, both low (0.5 mg/kg) and high clozapine (20 mg/kg) doses did not have a significant effect on the plasma concentrations of T4 and triiodothyronine (T3) (p>0.05). Neither of the compound had a significant effect on T3 plasma concentration levels (p>0.05). Haloperidol and clozapine act via different mechanisms and may have dissociable effects on thyroid hormones. Following treatment with haloperidol, significant changes in T4, but not in T3, serum levels were observed. Haloperidol and clozapine had different effects on the thyroid hormone levels. These results indicate that antipsychotic treatment can contribute to the thyroid dysfunction. Therefore, greater caution should be applied to the antipsychotics use. The thyroid function of the patients should be closely monitored, while using these drugs.

Targeted Pathway-based In Vivo Testing Using Thyroperoxidase Inhibition to Evaluate Plasma Thyroxine as a Surrogate Metric of Metamorphic Success in Model Amphibian Xenopus laevis.

Chemical safety evaluation is in the midst of a transition from traditional whole-animal toxicity testing to molecular pathway-based in vitro assays and in silico modeling. However, to facilitate the shift in reliance on apical effects for risk assessment to predictive surrogate metrics having characterized linkages to chemical mechanisms of action, targeted in vivo testing is necessary to establish these predictive relationships. In this study, we demonstrate a means to predict thyroid-related metamorphic success in the model amphibian Xenopus laevis using relevant biochemical measurements during early prometamorphosis. The adverse outcome pathway for thyroperoxidase inhibition leading to altered amphibian metamorphosis was used to inform a pathway-based in vivo study design that generated response-response relationships. These causal relationships were used to develop Bayesian probabilistic network models that mathematically determine conditional dependencies between biochemical nodes and support the predictive capability of the biochemical profiles. Plasma thyroxine concentrations were the most predictive of metamorphic success with improved predictivity when thyroid gland sodium-iodide symporter gene expression levels (a compensatory response) were used in conjunction with plasma thyroxine as an additional regressor. Although thyroid-mediated amphibian metamorphosis has been studied for decades, this is the first time a predictive relationship has been characterized between plasma thyroxine and metamorphic success. Linking these types of biochemical surrogate metrics to apical outcomes is vital to facilitate the transition to the new paradigm of chemical safety assessments.

Evaluation of Antioxidant Supplementation on Redox Unbalance in Hyperthyroid Cats Treated with Methimazole: A Blinded Randomized Controlled Trial.

Methimazole (MMI) is often the selected medical treatment for feline hyperthyroidism. However, the onset of MMI-related side effects (MMI-SE) is likely caused by oxidative stress. This study evaluated the dietary supplementation of selected antioxidants in hyperthyroid cats receiving MMI, to reduce MMI-SE. Thirty hyperthyroid client-owned cats were randomly allocated in group M (MMI + placebo) or group M+A (MMI + antioxidants). At different time-points from the enrolment (ET) to the end of the trial (FT), the following information was recorded: clinical findings, complete blood count, serum biochemical parameters, urinalysis, total plasma thyroxine concentrations, determinable reactive oxygen metabolites (dROMs), OXY-adsorbent test values, and oxidative stress index (OSi) values, and MMI-SE. dROMs and OSi values significantly increased from ET to FT in group M and were significantly higher in group M than in group M+A at FT. Likewise, OXY-absorbent test values were significantly higher in group M+A than in group M at FT. Moreover, the occurrence rate of MMI-SE in group M+A was lower than in group M. In conclusion, our results show that the dietary supplementation of antioxidants in hyperthyroid cats receiving MMI exerts a protective effect against oxidative stress, likely contributing to the reduction of MMI-SE.

Effect of drinking earthen pot water on physiological response and behavior of sheep under heat stress.

Heat stress is a major limiting factor for animal welfare and sheep production. Traditionally in India, the villagers used to keep their drinking water in the earthen pot to make it cold during summer. The cold drinking water (24-28°C) during summer gives a feeling of relief from the heat. Therefore, the present study was carried out to assess the effect of drinking earthen pot water on physiological response and behavior of sheep under heat stress for one month. For this purpose, eighteen Avishaan rams were selected from the experimental animal flock and they were equally divided into three groups; viz., control (CON), heat stress (HS) and heat stress with earthen pot water (HSC). The animals of HS and HSC were exposed to higher ambient temperatures to induce heat stress inside the psychometric chamber. The animals of CON and HS were provided with ad-libitum water of their ambient temperature whereas; HSC groups were provided with ad-libitum cold water (24-28°C) earthen pot water. All the animals were offered with 400gm concentrate mixture and ad-libitum Cenchrus hay. The bodyweight of HS rams was significantly reduced (P<0.05) at the end of the experimental period as compared to their initial body weight. The total roughage and dry matter intake was significantly higher (P<0.01) in HSC rams as compared to HS rams. The plasma thyroxine concentration was significantly lower (P<0.05) in HSC as compared with HS group. The rumination time significantly reduced (P<0.05) in HSC group. However, The blood biochemical did not differ among the groups. Therefore, it may be concluded that Avishaan rams have the ability to adapt to heat stress. Nevertheless, the availability of earthen pot cold drinking water under heat stress reduced their body weight loss, improves their metabolic activity and ultimately improves their welfare.

Mineral status, metabolism and performance of dairy heifers receiving a combined trace element bolus and out-wintered on perennial ryegrass, kale or fodder beet

The effects of a cobalt (Co), copper (Cu), selenium (Se), and iodine (I) trace-mineral ruminal bolus on the mineral status and performance of out-wintered, pregnant dairy heifers was investigated. Nine commercial farms grazing pasture (G), kale (K), or fodder beet (F) were used (n = 3 per forage), with forty heifers on each farm randomly allocated to not receive (B-) or receive (B+) two combined mineral boluses. Mean plasma Co concentrations were 0.021 and 0.041 µmol/L in B- and B+ respectively (p < 0.001), with serum vitamin B12 also higher in heifers receiving B+ than B- (p < 0.001). Mean plasma Se concentration was 0.50 and 0.82 µmol/L in B- and B+ respectively, with heifers that received B+ also having a higher (p < 0.05) mean blood GSH-Px concentration (30 and 76 U/mL haematocrit in B- and B+ respectively). Providing a mineral bolus did not affect plasma Cu concentration in heifers receiving G or F (p < 0.05), but was higher in KB+ compared to KB- (p < 0.05) at the middle and end of the out-wintering period. Heifers receiving KB- also had a lower haemoglobin and red blood cell count, but a higher mean corpuscular volume than KB+ at the end of the out-wintering period. Animals receiving B- had a higher plasma thyroxine concentration (p < 0.05). Neither the bolus nor forage type affected body weight (p > 0.05), however condition score was higher (p < 0.05) in B+ at the end of the study. It is concluded that the provision of a trace mineral bolus increased plasma concentrations of the minerals supplied, with the greatest benefits in animals grazing kale, but these increases were not translated into improved performance.

Risk factors for low plasma thyroxine and high plasma thyroid-stimulating hormone concentrations in dogs with non-thyroidal diseases.

The aim of the current study was to identify independent risk factors for thyroid axis alterations in dogs with non-thyroidal diseases. In this retrospective cross-sectional study, data and plasma samples from 207 dogs with non-thyroidal diseases was used. The involvement of various factors (disease severity, sex, age, breed, category and duration of disease, and medication) in the alteration of plasma thyroxine (T4) or thyroid-stimulation hormone (TSH) concentrations was analyzed using multivariate logistic regression. Among the 207 dogs analyzed, 99 (47.8%) had low plasma T4 concentrations, while 45 (21.7%) had high TSH concentrations. Intact male sex [odds ratio (OR), 3.25; 1.67–6.35; P<0.001], Labrador Retrievers (OR, 18.70; 2.32–151.00; P=0.006), moderate (OR, 2.39; 1.21–4.74; P=0.012) and severe diseases (OR, 6.84; 2.27–20.70; P<0.001) were associated with increased risk for low plasma T4 concentrations. Meanwhile, intact male (OR, 3.93; 1.51–10.30; P=0.005), spayed female (OR, 4.22; 1.59–11.20; P=0.004), older age (OR, 2.73; 1.28–5.84; P=0.009), and Miniature Dachshunds (OR, 5.39; 2.38–12.20; P<0.001) had increased risk for high plasma TSH concentrations. Disease severity had been determined as an independent risk factor for canine NTIS. In addition, sex, age and breed were also associated with thyroid axis alterations in dogs with non-thyroidal diseases.

Parental co-exposure to bisphenol A and nano-TiO2 causes thyroid endocrine disruption and developmental neurotoxicity in zebrafish offspring

The coexistence of organic toxicants and nanoparticles in the environment influences pollutant bioavailability and toxicity. Using chronic co-exposure to an adult zebrafish model, this study investigated the transfer kinetics and transgenerational effects of bisphenol A (BPA) and titanium dioxide nanoparticles (n-TiO2) exposure in F1 offspring. When single and combined exposure to BPA (0, 2, and 20 μg/L) and n-TiO2 (100 μg/L) were compared, combined exposure was found to reciprocally facilitate bioaccumulation in adult fish while enhancing maternal transfer to offspring. Thyroid endocrine disruption and developmental neurotoxicity were observed in larval offspring by parental exposure to BPA alone or in combination with n-TiO2. Exposure to 20 μg/L BPA significantly decreased the thyroxine (T4) concentration in adult plasma, leading to less transfer into the eggs. The presence of 20 μg/L BPA with n-TiO2 further decreased the level of T4 compared to BPA exposure alone. Additionally, offspring larvae derived from exposed parents exhibited lethargic swimming behavior. Overall, this study examined the interactions of BPA and n-TiO2 with regard to their bioaccumulation, maternal transfer, and developmental effects, which highlighted that co-exposure dynamics are important and need to be considered for accurate environmental risk assessment.

Expression of two glycoprotein hormone receptors in larval, parasitic phase, and adult sea lampreys

All jawed vertebrates have three canonical glycoprotein hormones (GpHs: luteinizing hormone, LH; follicle stimulating hormone, FSH; and thyroid stimulating hormone, TSH) with three corresponding GpH receptors (GpH-Rs: LH-R, FSH-R, and TSH-R). In contrast, we propose that the jawless vertebrate, the sea lamprey (Petromyzon marinus), only has two pituitary glycoprotein hormones, lamprey (l)GpH and l-thyrostimulin, and two functional glycoprotein receptors, lGpH-R I and II. It is not known at this time whether there is a specific receptor for lGpH and l-thyrostimulin, or if both GpHs can differentially activate the lGpH-Rs. In this report, we determined the RNA expression of lGpH-R I and II in the gonads and thyroids of larval, parasitic phase, and adult lampreys. A highly sensitive dual-label fluorescent in situ hybridization technique (RNAScope™) showed lGpH-R I expression in the ovaries of larval lamprey, and co-localization and co-expression of lGpH-R I and II in the ovaries of parasitic phase and adult lampreys. Both receptors were also highly co-localized and co-expressed in the endostyle of larval lamprey and thyroid follicles of parasitic and adult lampreys. In addition, we performed in vivo studies to determine the actions of lamprey gonadotropin releasing hormones (lGnRHs) on lGpH-R I and II expression by real time PCR, and determined plasma concentrations of estradiol and thyroxine. Administration of lGnRH-III significantly (p ≤ 0.01) increased lGpHR II expression in the thyroid follicles of adult female lampreys but did not cause a significant increase in RNA expression of lGpH-R I and II in ovaries. Concomitantly, there was a significant increase (p ≤ 0.01) of plasma estradiol without any significant changes of plasma thyroxine concentrations in response to treatment to lGnRH-I, -II, or -III. In summary, our results provide supporting evidence that the lamprey pituitary glycoprotein hormones may differentially activate the lamprey GpH-Rs in regulating both thyroid and gonadal activities during each of the three life stages of the sea lamprey.

Association of residual feed intake with growth and slaughtering performance, blood metabolism, and body composition in growing lambs

The aim of this study was to determine the association of residual feed intake (RFI) with growth performance, blood metabolic parameters, and body composition factors in growing lambs. Individual body weight (BW) and dry matter intake (DMI) were determined in 137 male Hu lambs that were given a pellet feed four times a day for 50 d. RFI did not show a correlation with metabolic BW (MBW) or average daily gain (ADG), but it showed a positive correlation with DMI and feed conversation ratio (FCR). Organ weight and intestine length had a large influence on RFI in lambs. The low-RFI lambs have smaller rumen and longer duodenum indicating the less feed intake and more sufficient absorption rate of low-RFI lambs. The smaller organs like liver, lung and kidney in low-RFI lambs may be related to lower energy consumption and slower metabolic rate. The observed bigger testis was in low-RFI lambs was another cause of the improved feed efficiency. Finally, the plasma concentrations of thyroxine (T4) and adrenocorticotropic hormone (ACTH) were lower in the ELow-RFI group than in the EHigh-RFI group. This study provides new insight into the biological processes underlying variations in feed efficiency in growing lambs.
